Artemia has a remarkable genetic variability that can be expressed in various phenotypic characteristics, such as morphometry, growth rate, reproductive isolation or molecular composition. This study presents reproduction characteristics, survival rate and sex-ratio of four Artemia salina populations from Tunisia cultured under standard conditions. Results show that both low (17.8% and 30.6% in Megrine saltwork and Sabkhet Sijoumi, respectively) and high (83.9% for Sahline saltwork) values were recorded for the percentage of total oviparous offspring. The offspring per brood varied from 70.3?±?41.9 (Megrine saltwork) to 73.6?±?51.4 (Sabkhet El Adhibet). Broods per female varied between 3.1?±?0.9 (Sahline saltwork) and 4.5?±?1.2 (Megrine saltwork), and time between broods ranged from 5.7?±?1.1 to 6.5?±?2.2 days for Megrine and Sahline, respectively. Statistical analysis (one-way analysis of variance, Tukey's HSD test, p?<?0.05) revealed no significant differences between reproductive traits of the four studied populations except for mean of oviparous and oviviparous offspring per female (F?=?9.158, p?<?0.05), and brood per female (F?=?4.779, p?<?0.05). The survival rate of the four studied A. salina populations fluctuated between 31.4% and 64.5% for Megrine and Sahline saltwork, respectively. However, the sex-ratio showed that for Megrine saltwork and Sabkhet El Adhibet, males predominated with 1?:?0.90 and 1?:?0.97 (males?:?females), respectively. The comparison between our results and those reported of other Artemia populations showed that the offspring per brood of Tunisian Artemia are comparable to A. persimilis but different to A. salina from Abu Kammash (Libya) and A. urmiana (Urmia Lake, Iran), and that days between brood, are similar to A. salina from Abu Kammash, A. sinica and polyploid A. parthenogenetica, but different from A. persimilis and A. franciscana.  相似文献

Artemia cysts produced in inoculated salt ponds in the Philippines, Thailand and India, were analyzed in comparison with the parental strains used for the inoculations. Cyst hatching efficiency, hatching rate and color changed significantly after inoculation, whereas the cyst's diameters, nauplius survival and nauplius growth remained fairly constant. The nutritional value of theArtemia nauplii was determined in a standard culture test withMysidopsis bahia juveniles as test animals. Production results were compared with the fatty acid profiles of theArtemia nauplii. Low levels of the essential fatty acid 20:53 in one of the produced cyst batches were probably caused by inadequate food conditions in the pond and resulted in poorer growth of theMysidopsis juveniles. Based on the results of this study a distinction is made between essential and nonessential strain selection-criteria forArtemia inoculations and transplantations.  相似文献

The influence of magnetic fields on hatching and chromatin state of brine shrimp, Artemia sp., was investigated. Dry Artemia cysts were exposed to a magnetic field of intensity 25 mT for 10 min. The magnetic field was applied in different variants: constant field, rotating field of different directions (right-handed and left-handed) and different magnet polarization. The effect of ultra wideband pulse radiation and microwave radiation was also investigated. The energy density on the surface of object exposed to ultra wideband pulse radiation was 10−2, 10−3, 10−4, 10−5 and 10−6 W/cm2, the power of microwave radiation was 10−4 and 10−5 W/cm2, exposure time - 10 s. After incubation of the cysts for 48 hours in sea water the hatching percentage of Artemia from exposed cysts was higher than in controls. The number of heterochromatin granules was significantly higher in the nauplia (newborn larvae of Artemia) developed from cysts that had been exposed to magnetic and electromagnetic fields. The data obtained demonstrate an increase in percentage hatching of Artemia cysts after treatment with magnetic and electromagnetic fields and chromatin condensation in nauplia. We have also shown different effects of right-handed and left-handed rotating magnetic fields on these processes.  相似文献

Although the crustacean Artemia has been commonly used as an experimental organism and served as a live bait feed for aquaculture, gene transfer system on Artemia sp. to generate stable lines is not well developed. In this study, we optimized a condition for cyst-eletroporation and generated stable lines of transgenic A. sinica. Two expression plasmids directed by the hybrid promoters of cytomegalovirus (CMV) and medaka β-actin (Mβ) were co-electroporated on decapsulated cysts: pCMV-Mβ-GFP contained GFP reporter gene and pCMV-Mβ-ypGH contained yellowfin porgy GH (ypGH) cDNA. We examined the GFP shown in the Artemia larvae and found that the expression rate was 13.3% (3,219 out of 24,054 examined). We then chose 200 G0 founders which strongly expressed GFP to generate transgenic lines. Homozygotic strains derived from F4 generation of each transgenic line, A3 and A8, were obtained. We proved that transgenic lines A3 and A8 also harbored pCMV-Mβ-ypGH and produced recombinant ypGH with a concentration of 0.089 and 0.032 μg per 50 homozygotic nauplii, respectively. Ten live Artemia nauplii were fed daily to zebrafish larvae during 25 to 35 days of post-fertilization. The average body length gain rates of zebrafish larvae fed transgenic Artemia were 16–20% greater than those of control group, indicating the exogenous ypGH produced by transgenic Artemia is functional. Therefore, we concluded that the transgenesis on Artemia is developed, and transgenic Artemia might be highly potentially useful as a new bioreactor material for application in aquaculture and biological researches.  相似文献

Artemia cysts collected from inoculation experiments in Cam Ranh salterns are evaluated for their potential use in aquaculture. Cyst biometrics, hatching quality, naupliar fatty acid profile and naupliar growth were measured and compared to reference Artemia strains. Cyst characteristics reveal that the parthenogenetic strain (PR China) used in inoculations, was eliminated from the environment and that the remaining brine shrimp are likely to be composed of Macau and Great Salt Lake Artemia strains, and of their cross-breds. Differences in cyst diapause deactivation characteristics between Macau and Great Salt Lake Artemia may have resulted in the disappearance of Macau Artemia during the rainy season and the persistence of Great Salt Lake Artemia during the following dry season.  相似文献

Naturally spawned eggs of the Pacific saury,Cololabis saira, were collected in the field and reared in a tank to examine daily periodicity of growth increment formation in the otolith. Larvae were 6.9 mm in knob length at hatching. Their otoliths (sagittae) were 31 μm in radius and had 3–6 faint concentric rings. They started feeding within two days and grew at a rate of 1.1 mm/day on average through larval and juvenile stages feeding on rotifers,Artemia nauplii, and artificial diets. Otolith growth increments showed a concentric pattern with a distance of 3.5–5.0 μm between two adjacent increments. The number of growth increments was almost equal to a known age in days plus 4 or 5. A regression line of number of increments (N) on known age in days (D) between 0–30 days after hatching was N = 4.81 + 1.01D, which shows that one increment was deposited per day.  相似文献

Cysts of parthenogenetic Artemia strains collected from the Citros and M. Embolon saltworks in Northern Greece are evaluated for their potential use in aquaculture. The following characterizations were performed: cyst and naupliar biometrics, cyst hatching characteristics, fatty acid profile of the nauplii, caloric content of nauplii stored at 25 °C and in a refrigerator (4–8 °C). The above evaluation reveals that the two Artemia strains studied exhibit good qualities for use in aquaculture, especially in culturing fresh-water species. The biometrical analysis of cysts, nauplii and adults shows a high degree of similarity with other parthenogenetic strains from various geographical sources, but especially with tetraploid Artemia from Spain. The Greek Artemia strains cannot be considered as sources for aquacultural uses unless proper management of the saltworks is assured.  相似文献

The recent exponential growth in industrial aquaculture has led to a huge increase in Artemia biomass production in order to meet increased fish production needs. The present study explores the potential use of salt gradient solar ponds (SGSPs) for production of Artemia nauplii. An SGSP is a basin of water where solar energy is trapped and collected via an artificially imposed gradient. Three zones can be identified in an SGSP: upper and lower zones, which are both convective, and a middle zone, which is intended to be non-convective. The latter acts as a transparent insulation layer and allows for storage of solar energy at the bottom, where it is available for use. The combination of salt, temperature and high transparency could make SGSPs promising bioreactors for the production of Artemia nauplii. Using particle image velocymetry (PIV) and Shadowgraph visualisation techniques, the behaviour of Artemia nauplii under critical cultivation parameters (namely, salinity, temperature and light) was monitored to determine movement velocity, and how movement of Artemia affects the salt gradient. It was observed that Artemia nauplii constantly follow light, irrespective of adverse salinity and/or temperature conditions. However, despite the substantial displacement of Artemia following the light source, the salt gradient is not disrupted. The suitability of SGSPs as bioreactors for Artemia biomass production was then tested. The results were disappointing, probably due to the lack of sufficient O2 for Artemia survival and growth. Follow-up trials were conducted aimed at using the SGSP as a green and economically attractive energy source to induce faster hatching of cysts and improved Artemia nauplii growth. The results of these trials, and a case study of Artemia nauplii production using an SGSP, are presented. The authors constructed a Solar Pond device, which they suggest as a novel way of supplying thermal energy for Artemia biomass production in an aquaculture enterprise. Finally, the authors suggest a method of producing and collecting Artemia biomass, and of heating a fish larval tank, in an ‘ideal’ Solar Pond device, profiting from the low investment costs of using a decommissioned salt works.  相似文献

Nine types of live foods viz. L, S and SS morphotypes of Brachionus plicatilis sp. complex, first instar Artemia franciscana, Fabrea salina, Acartia tsuensis, Tigriopus japonicus, Diaphanosoma celebensis, Moina mongolica and a formulated feed of two sizes (400 and 700 μm) were used to observe feeding behaviour and growth of mangrove killifish, Kryptolebias marmoratus. Behavioural observations were made for one hour on days 0, 1, 5, and 10 after hatching. Focus, unsuccessful and successful attacks and vomit were noted. With rotifers L, S and SS types and newly hatched Artemia nauplii as food, all the larvae showed maximum feeding success throughout the experimental period. Larvae did not consume any of the 700 μm artificial diet. Vomiting was noticed on capturing the ciliate Fabrea and 400 μm artificial diets. Rotifers were ingested in greater numbers. SS-type rotifers were consumed in largest number (209.2/h per individual) on day 10. Significantly greater growth was observed after 10 days rearing with L type rotifer, Artemia nauplii, T. japonicus, A. tsuensis, M. mongolica, D. celebensis, and a mixture of L type rotifer and F. salina (Tukey–Kramer post hoc test, P < 0.05). Feed selectivity experiments on days 0, 1, 5 and 10 revealed that killifish larvae feed preferentially on Artemia nauplii and rotifers from a mixture of Artemia nauplii, rotifers, A. tsuensis, T. japonicus, D. celebensis and M. mongolica. Techniques for culturing various zooplankton at small-scales are also described.  相似文献

Aim This paper reports the presence of Artemia franciscana ( Kellogg, 1906 ) in Mar Chiquita (CHI) salt lake and in Las Tunas (TUN) lagoon, as well as at a shallow lake in Salinas Grandes (Province of Córdoba) in Argentina. To date, this species has been considered absent from Argentina. This study also provides further data on the characterization of the A. franciscana populations from this area. Location Province of Córdoba (Argentina). Methods The cyst samples collected at the three hypersaline environments were measured to assess its mean diameter with a dissecting microscope. The length of the nauplii hatched from the cysts was also determined with the microscope. The adults raised from these nauplii under strictly controlled conditions were analysed for 12 morphological parameters and compared through multivariate discriminant analysis with other American populations. Fatty acids from the total lipids were analysed by gas chromatography. Reproductive compatibility was evaluated from single‐pair intraspecific and interspecific crosses of adult specimens. Results The results endorse the morphometric assimilation of the Artemia populations of this area to other American populations of the A. franciscana‘super‐species’, together with its morphometric differentiation from the species A. persimilis ( Piccinelli & Prosdocimi, 1968 ). The more conspicuous population from CHI shows cross‐fertility with A. franciscana original from San Francisco Bay (California, USA). Both populations from CHI and TUN show cross‐fertility between them and reproductive isolation with A. persimilis from Hidalgo (Province of La Pampa), which to date is considered to be endemic or exclusive of hypersaline ecosystems in Argentina. The reproductive characteristics displayed by the population from TUN lagoon suggest the existence of an occasional hybridization between A. franciscana and A. persimilis ( Papeschi et al., 2000 ). Artemia cysts from CHI and TUN show a fatty acid profile rich in eicosapentaenoic acid differing markedly from the fatty acid profiles found in A. persimilis cysts. Main conclusions: Artemia franciscana is present in Argentina at 36° S and north of this latitude. Artemia persimilis is confined to the south of latitude 37°10′ S. There are different pieces of evidence that point to a certain level of hybridization of the two species taking place in the land belt between these parallels.  相似文献

Oxygen consumption rates of nauplii of the brine shrimp Artemia franciscana Kellogg 1906 were determined over a range of salinities from 10 to 110 ppm, in temperatures from 0 to 30°C, using a multi-factorial design. The oxygen micro-sensors employed have a fast response time and are capable of accurately measuring oxygen concentrations at temperatures well below 0°C. Oxygen uptake rate ranged from 0.03 to 0.66 μmol O2 mg−1 h−1 and was sensitive to changes in both salinity and temperature. Temperature was the dominant factor affecting oxygen consumption rates, which showed a significant increase with increasing temperature. A slight decrease was measured in oxygen consumption with increasing salinity related to differential solubility of oxygen in waters of different salinities. Thermal sensitivity of oxygen consumption determined from calculations of Q 10, indicated physiological adaptation of Artemia nauplii to the ranges of temperatures tested. Little is known about the larviculture of the chub, Leuciscus cephalus (L.), an endangered cyprinid species endemic to European flowing waters. The use of decapsulated Artemia cysts as food for chub larviculture was investigated. After 3‐day feeding with the rotifer Brachionus calyciflorus, the larvae were fed on different diets: (i) dried decapsulated Artemia cysts, (ii) Artemia nauplii, (iii) rotifers for seven more days and then Daphnia collected from a pond, and (iv) an artificial diet. After a 24‐day rearing period, the highest survival rate was obtained with the larvae receiving decapsulated Artemia cysts. Feeding of the larvae with an artificial diet resulted in a significantly lower survival rate compared with the other groups. At the end of the experiment, the larvae fed on Artemia nauplii yielded a significantly higher mean length compared with the other groups. Feeding an artificial diet resulted in a significantly lower average weight and mean length gain compared with the other groups.  相似文献

In the Great Salt Lake of Utah, the brine shrimp Artemia franciscanaKellogg is an important food resource for birds and they produce dormant cysts that are harvested and used extensively in the aquaculture industry. We analyzed the limnological factors controlling Artemia growth and cyst production over 12 months in 1994 and 1995. Laboratory experiments showed that inter-brood intervals were highly dependent on temperature and slightly on food level. At optimal temperatures and nutritious food, juveniles reached reproductive size within 7 d in the laboratory. In winter when temperatures were less than 3 °C, Artemia were absent from the lake, phytoplankton abundance was high (13 Chl a g l–1), and the dominant grazers were ciliated protozoans. In the spring, cysts hatched when phytoplankton was abundant (15–30 g Chl a l–1), and the Artemia grew and produced large clutches of ovoviviparous eggs. Estimated naupliar production from these eggs was 80 l–1 from April to May. Despite the high production of nauplii, Artemia densities declined to 8 l–1by June and the growing shrimp population grazed down the phytoplankton resource to <1 g Chl a l–1. With the depleted phytoplankton food resource during the summer, Artemia growth slowed, lipid indices decreased, clutch sizes declined, and females switched primarily to oviparous cyst production. During the summer, there was limited production of ovoviviparous eggs, and limited recruitment of juveniles, probably due to low food. Although oviparous reproduction began in June, more than 90% of the cysts were produced after July when female densities had declined to 1.5 l–1, but nearly all of them were producing cysts. Estimated cyst production was 650000 m–2, or 4.54 × 106 kg dry weight for the entire lake. The reported commercial harvest took 21% of the 1994 cyst production. That harvest had little impact on the subsequent year's population, as Artemia densities were ultimately controlled by algal production in the lake.  相似文献

The zooxanthellate octocoral Sinularia flexibilis is a producer of potential pharmaceutically important metabolites such as antimicrobial and cytotoxic substances. Controlled rearing of the coral, as an alternative for commercial exploitation of these compounds, requires the study of species-specific growth requirements. In this study, phototrophic vs. heterotrophic daily energy demands of S. flexibilis was investigated through light and Artemia feeding trials in the laboratory. Rate of photosynthetic oxygen by zooxanthellae in light (≈200 μmol quanta m−2 s−1) was measured for the coral colonies with and without feeding on Artemia nauplii. Respiratory oxygen was measured in the dark, again with and without Artemia nauplii. Photosynthesis–irradiance curve at light intensities of 0, 50, 100, 200, and 400 μmol quanta m−2 s−1 showed an increase in photosynthetic oxygen production up to a light intensity between 100 and 200 μmol quanta m−2 s−1. The photosynthesis to respiration ratio (P/R > 1) confirmed phototrophy of S. flexibilis. Both fed and non-fed colonies in the light showed high carbon contribution by zooxanthellae to animal (host) respiration values of 111–127%. Carbon energy equivalents allocated to the coral growth averaged 6–12% of total photosynthesis energy (mg C g 1 buoyant weight day 1) and about 0.02% of the total daily radiant energy. “Light utilization efficiency (ε)” estimated an average ε value of 75% 12 h 1 for coral practical energetics. This study shows that besides a fundamental role of phototrophy vs. heterotrophy in daily energy budget of S. flexibilis, an efficient fraction of irradiance is converted to useable energy.  相似文献

A first attempt to introduce tench (Tinca tinca (L.)) to Israel, followed by comparative growth experiments, was carried out in 1947. A batch of 200 fingerlings, which were supposed to serve later as brood fish, was imported from Switzerland to Fish Culture Experimental Station at Sdeh Nahum. However, after 2 years tench were considered unsuitable for culturing in Israel and the trials were abandoned. Two factors affected negatively these historically early trials: (1) Tench was just second exotic species introduced to Israel some years after introduction of the common carp, and the Israeli fish farming expertise at that time was low. (2) The Independence War temporarily delayed advancement of aquaculture. A second introduction of tench was performed in 1992, when a golden mutant of tench was imported to Gan Shmuel Fish Breeding Center (GS-FBC) from the Research Institute of Fish Culture and Hydrobiology at Vodňany, Czech Republic. Currently, GS-FBC is the only Israeli fish hatchery that maintains golden tench as a part of breeding program as well as a number of other varieties of ornamental fish and species, all of them produced mostly for export. Tench (800–1,000 g) females are maintained separately from males, in order to prevent uncontrolled reproduction. Routinely, in April, the fish are transferred indoors and induced to spawn artificially with a single injection of GnRH synthetic analog—DAGIN. Dry method of stripping is used to obtain eggs and sperm. After fertilization the sticky eggs are spread in a single layer on walls of 230 dm3 cylindrical incubators provided with conical bottom, or they are degummed using a solution of milk/water. At a water temperature of 24°C, hatching (70% hatching rate) occurs 36–40 h after fertilization. The larvae absorb their yolk sac 2–3 days after fertilization and they start feeding on small size (200 μm) Artemia nauplii. At the age of 7–10 days, if fry are retained indoors, they are fed with regular size Artemia and starter feed, or are released into a nursing 0.5–1.0 ha pond. Usually, the fish are grown to the marketing size (5–15 cm) and exported abroad, mainly to Europe. About 100,000 golden tench fingerlings, with a total value of € 40,000 are produced and marketed annually.  相似文献

We studied the Artemia populations existing in Lake Urmia (north‐western Iran), one of the largest habitats of Artemia in the world, in order to settle the long‐standing controversy over the sexual status of the endemic Artemia populations. Experiments were carried out in the laboratory and in the field. Cysts, collected from different sites of the lake and peripheral lagoons, were hatched and cultured to adults in the laboratory. Adult sexual and parthenogenetic animals were isolated and newly hatched nauplii from them were cultured to maturity in different salinities, ranging from 15–80 ppt. Survival levels and percentage of animals attaining adulthood were measured over a period of 30 days. In the field experiment, cysts taken from Lake Urmia were hatched and the resulting nauplii were inoculated into six earthen ponds (80–140 ppt) constructed in the vicinity of the lake. Population composition in each pond was determined over a period of two years. Results indicated that both sexual and parthenogenetic Artemia coexist in Lake Urmia. While the lake itself is dominated by sexual Artemia, the asexual populations were found to be restricted to particular areas in or near the lake. Artemia appearing seasonally in the lagoons adjacent to the lake were exclusively parthenogenetic. Parthenogens could grow, mature and reproduce at very low salinities (15–33 ppt), whereas higher salinities (above 50 ppt) were required for A. urmiana to attain sexual maturity. The combined effect of temperature, food level and the presence of an invertebrate predator on the body size of the rotifer Brachionus havanaensis were tested in this study. B. havanaensis was cultured at 15, 20, and 25°C under three different Chlorella vulgaris levels (0.5 × 106, 1.0 × 106 and 2.0 × 106 cells ml−1) in the presence and in the absence of Asplanchna girodi. For each treatment we maintained three replicates and constant (0.4 ind ml−1) population density of B. havanaensis. In treatments containing A. girodi, the predator was separated from the prey by a mesh (pore size 50 μm). On the last day of the experiment, a portion of the B. havanaensis population was sampled for several morphometric measurements (adult lorica length, width, posterior spine length, body volume, and the egg volume). Size measurements were done by drawing the specimens using a calibrated camera lucida. Statistically significant impact of temperature as well as the predator’s presence was observed on the lorica length, posterior spine, and egg volume of B. havanaensis. The interactions of food × temperature, or predator′s presence × food × temperature were non-significant (P > 0.05) for lorica length, spine length, body volume, and egg volume. Regardless of the type of treatment, there was a direct positive correlation between lorica length and width. Egg volume was linearly related to the adult size. The predatory bug Macrolophus caliginosus, which is widely used in greenhouse crops, is limited in its application by its high price. An important factor in the cost is the high price of Ephestia kuehniella eggs, the prey used in their mass rearing. In order to reduce their price, alternatives to moth eggs are currently being investigated. The brine shrimp Artemia sp. is produced in large quantities in saline lakes and is fed as live food source to the larvae of a variety of marine and freshwater organisms. In this study, we tested Artemia sp. as prey for rearing M. caliginosus from two strains. We evaluated developmental and reproduction parameters of the predator when fed nauplii, enriched nauplii with a fatty acid, dry cysts and hydrated cysts, and were compared with those obtained when the predator was fed with E. kuehniella eggs. Nauplii had a significant reduction in survivorship, a delay in development of nymphs and a low reproduction of adults. Nauplii enriched with docosahexaenoic acid (DHA, 22:6n − 3), a common practice for larviculture of some marine fish species, resulted toxic to M. caliginosus nymphs and survival was quite low. On the contrary, either dry or hydrated cysts from the two strains tested of the brine shrimp produced the same nymphal survivorship, nymphal development time and weight and fecundity of adults as those obtained with E. kuehniella eggs. Demographic parameters of the eighth generation of the predator reared with cysts of the two strains, either dry or hydrated, were as good as those of moth eggs. We concluded that Artemia sp. cysts were a good substitution prey for the mass rearing of M. caliginosus.  相似文献
