Genus Mucuna which is native to China and Eastern India comprises of perennial climbing legume with long slender branches, trifoliate leaves and bear green or brown pod covered with soft or rigid hairs that cause intense irritation. The plants of this genus are agronomically and economically important and commercially cultivated in India, China and other regions of the world. The high degrees of taxonomical confusions exist in Mucuna species that make authentic identification and classification difficult. In the present study, the genetic diversity among the 59 accessions of six species and three varieties of M. pruriens has been assessed using DNA fingerprinting based molecular markers techniques namely randomly amplified polymorphic DNA (RAPD), inter simple sequence repeats (ISSR) and combined dataset of RAPD and ISSR. Also, genetic relationship among two endemic species of Mucuna namely M. imbricata and M. macrocarpa and two varieties namely IIHR hybrid (MHR) and Dhanwantari (MD) with other species under study was investigated by using cluster analysis and principal coordinate analysis. The cluster analysis of RAPD, ISSR and combined dataset of RAPD and ISSR clearly demonstrated the existence of high interspecific variation than intra-specific variation in genus Mucuna. The utility and efficacy of RAPD and ISSR for the study of intra species and interspecies genetic diversity was evident from AMOVA and PCoA analysis. This study demonstrates the genetic diversity in Mucuna species and indicates that these markers could be successfully used to assess genetic variation among the accessions of Mucuna species.  相似文献

Trichoderma species are widespread phytostimulant fungi that act through biocontrol of root pathogens, modulation of root architecture, and improving plant adaptation to biotic and abiotic stress. With the major challenge to better understand the contribution of Trichoderma symbionts to plant adaptation to climate changes and confer stress tolerance, we investigated the potential of Trichoderma virens and Trichoderma atroviride in modulating stomatal aperture and plant transpiration. Arabidopsis wild-type (WT) seedlings and ABA-insensitive mutants, abi1-1 and abi2-1, were co-cultivated with either T. virens or T. atroviride, and stomatal aperture and water loss were determined in leaves. Arabidopsis WT seedlings inoculated with these fungal species showed both decreased stomatal aperture and reduced water loss when compared with uninoculated seedlings. This effect was absent in abi1-1 and abi2-1 mutants. T. virens and T. atroviride induced the abscisic acid (ABA) inducible marker abi4:uidA and produced ABA under standard or saline growth conditions. These results show a novel facet of Trichoderma-produced metabolites in stomatic aperture and water-use efficiency of plants.  相似文献

Trichoderma species form endophytic associations with plant roots and may provide a range of benefits to their hosts. However, few studies have systematically examined the diversity of Trichoderma species associated with plant roots in tropical regions. During the evaluation of Trichoderma isolates for use as biocontrol agents, root samples were collected from more than 58 genera in 35 plant families from a range of habitats in Malaysian Borneo. Trichoderma species were isolated from surface-sterilised roots and identified following analysis of partial translation elongation factor-1α (tef1) sequences. Species present included Trichoderma afroharzianum, Trichoderma asperelloides, Trichoderma asperellum, Trichoderma guizhouense, Trichoderma reesei, Trichoderma strigosum and Trichoderma virens. Trichoderma asperellum/T. asperelloides, Trichoderma harzianum s.l. and T. virens were the most frequently isolated taxa. tef1 sequence data supported the recognition of undescribed species related to the T. harzianum complex. The results suggest that tropical plants may be a useful source of novel root-associated Trichoderma for biotechnological applications.  相似文献

Trichoderma species are widely used in agriculture as biofungicides. These fungi are rich source of secondary metabolites and the mycoparasitic species are enriched in genes for biosynthesis of secondary metabolites. Most often, genes for secondary metabolism are clustered in fungal genomes. Previously, no systematic study was undertaken to identify the secondary-metabolism related gene clusters in Trichoderma genomes. In the present study, a survey of the three Trichoderma genomes viz. T. reesei, T. atroviride and T. virens, was made to identify the putative gene clusters associated with secondary metabolism. In T. reesei genome, we identified one new NRPS and 6 new PKS clusters, which is much less than that found in T. atroviride (4 and 8) and T. virens (8 and 7). This work would pave the way for discovery of novel secondary metabolites and pathways in Trichoderma.  相似文献

The Thinodromus lunatus species group is revised. The following new species are described: Thinodromus (s. str.) cattiensis sp. n. from Vietnam, Thinodromus (s. str.) forsteri sp. n. from southern Thailand, Thinodromus (s. str.) himalayensis sp. n. from Nepal and northern India, Thinodromus (s. str.) inconspicuus sp. n. from southern China, Thailand, and Vietnam, and Thinodromus (s. str.) spotus sp. n. from southern China. The following new synonymy is established: Thinodromus (s. str.) deceptor (Sharp, 1889) = Thinodromus (s. str.) gravelyi (Bernhauer, 1926), syn. n.; = Thinodromus (s. str.) reitterianus (Bernhauer, 1938), syn. n. Lectotypes are designated for Trogophloeus lunatus Motschulsky, 1857, Trogophloeus pustulatus Bernhauer, 1904, Trogophloeus socius Bernhauer, 1904, Trogophloeus sumatrensis Bernhauer, 1915, Trogophloeus lewisi Cameron, 1919, Trogophloeus gravelyi Bernhauer, 1926, Trogophloeus reitterianus Bernhauer, 1938, and Trogophloeus unipustulatus Cameron, 1941. On the basis of morphological and molecular data, taxonomic diversity of the agamid genus Acanthosaura in Vietnam is discussed. This genus is represented in Vietnam by four species of two complexes, A. capra and A. lepidogaster. DNA analysis and the absence of reliable finds suggest to exclude A. crucigera and A. armata from the faunal list of Vietnam. The recently described species A. nataliae Orlov, Nguyen and Nguyen, 2006 of the A. capra complex is considered. Two species are recognized in the A. lepidogaster complex, one of which, A. coronata Günther, 1861, Smith (1935) regarded as a synonym of A. lepidogaster (Cuvier, 1829).  相似文献

Central Himalayan region of India encompasses varied ecological habitats ranging from near tropics to the mid-elevation forests dominated by cool-temperate taxa. In past, we have reported several new records and novel species from Uttarakhand state of India. Here, we assessed genetic variations in three mitochondrial genes, namely, 16S rRNA, cytochrome c oxidase subunit I and cytochrome c oxidase subunit II (COI and COII) in 26 drosophilid species collected along altitudinal transect from 550 to 2700 m above mean sea level. In the present study, overall 543 sequences were generated, 82 for 16S rRNA, 238 for COI, 223 for COII with 21, 47 and 45 mitochondrial haplotypes for 16S rRNA, COI and COII genes, respectively. Almost all species were represented by 2–3 unique mitochondrial haplotypes, depicting a significant impact of environmental heterogeneity along altitudinal gradient on genetic diversity. Also for the first time, molecular data of some rare species like Drosophila mukteshwarensis, Liodrosophila nitida, Lordiphosa parantillaria, Lordiphosa ayarpathaensis, Scaptomyza himalayana, Scaptomyza tistai, Zaprionus grandis and Stegana minuta are provided to public domains through this study.  相似文献

Gobioid fishes collected in Nha Trang and Van Phong bays (Vietnam) represented by 44 species, including 30 species continuously associated with soft bottoms in the open parts of the bays with normal marine conditions are reviewed. A new species, Navigobius khanhoa sp. n., is described. This species differs from other species of the genus in much higher number of rays in the second dorsal fin (26), smaller size of the mouth, teeth on the jaws arranged in a single row, and other characters. Four species from the genera Acentrogobius, Eviota, Favonigobius, and Gnatholepis have not been identified to the species, and, most likely, some of them are new for science. Nine species are recorded in the fauna of Vietnam for the first time: Aulopareia unicolor, Bathygobius hongkongensis, Egglestonichthys bombylios, Glossogobius circumspectus, Oplopomops diacanthus, Oxyurichthys auchenolepis, Tomiyamichthys ex gr. russus, Tryssogobius porosus, and Valenciennea immaculata. An occurrence of Glossogobius olivaceus in the fauna of Vietnam is confirmed. Three species, G. olivaceus, Oligolepis acutipennis, and Periophthalmus kalolo, are recorded in the Cai River delta for the first time. A new synonymy is established: Eviota gurjanovae (Prokofiev, 2007) = E. prasina (Klunzinger, 1871). The current state of knowledge of Gobiidae fauna of Vietnam is discussed, and the updating of the species composition of Gobiodontini of Nha Trang Bay is presented in addition to the species list published earlier (Prokofiev, 2007).  相似文献

In this study, a total of 27 species of dragonfly and damselfly larvae were identified, including 14 species from suborder Anisoptera and 13 from Zygoptera. The dominant species are Libellula quadrimaculata, Enallagma cyathigerum, and Erythromma najas. Frequently occurring species are Coenagrion hastulatum, C. lunulatum, C. pulchellum, Ischnura elegans, Platycnemis pennipes, Lestes virens, and Libellula depressa. Rare species are Lestes sponsa, Libellula fulva, Gomphus flavipes, Gomphus vulgatissimus, Sympetrum flaveolum, Aeshna viridis, Ae. cyanea, Ae. grandis, Ae. mixta, Calopteryx splendens, C. virgo, and Ophiogomphus cecilia.  相似文献

Plastid DNA markers sequencing and DNA fingerprinting approaches were used and compared for resolving molecular phylogeny of closely related, previously unexplored Amorphophallus species of India. The utility of individual plastid markers namely rbcL, matK, trnHpsbA, trnLCtrnLD, their combined dataset and two fingerprinting techniques viz. RAPD and ISSR were tested for their efficacy to resolves Amorphophallus species into three sections specific clades namely Rhaphiophallus, Conophallus and Amorphophallus. In the present study, sequences of these four plastid DNA regions as well as RAPD and ISSR profiles of 16 Amorphophallus species together with six varieties of two species were generated and analyzed. Maximum likelihood and Bayesian Inference based construction of phylogenetic trees indicated that among the four plastid DNA regions tested individually and their combined dataset, rbcL was found best suited for resolving closely related Amorphophallus species into section specific clades. When analyzed individually, rbcL exhibited better discrimination ability than matK, trnHpsbA, trnLCtrnLD and combination of all four tested plastid markers. Among two fingerprinting techniques used, the resolution of Amorphophallus species using RAPD was better than ISSR and combination of RAPD +ISSR and in congruence with resolution based on rbcL.  相似文献

The DNA barcode approach was used to identify and establish association of Colletotrichum species complex with fruit rot disease of chili (Capsicum annuum L.) in North-Western Himalayan region of India. Twenty isolates of five morphologically identified Colletotrichum species collected from commercial chili growing areas were identified using deoxyribonucleic acid (DNA) barcode marker genes, 5.8S ribosomal ribonucleic acid flanking internal transcribed spacers 1 & 2 and β-tubulin gene. Morpho-cultural identification requires expertise to delineate C. gloeosporioides, C. boninense and C. acutatum complexes from each other, as these species possess minute variation in spore shape and size. Ribosomal DNA and β-tubulin sequence analysis along with species-specific marker amplification established the association of seven Colletorichum spp. viz., C. truncatum (syn. Colletotrichum capsici), C. coccodes, C. karstii, C. kahawae, C. nymphaeae, C. fructicola and C. gloeosporioides complex with fruit rot of chili. Phylogenetic analysis of 35 Colletotrichum sequences including authentic type sequences validated the identified sequences with strong bootstrap support. This approach delineated morphologically identified species with great ease into more reliable genotype based speciation of various Colletorichum complexes. The DNA barcode markers have direct implications for plant pathologists in relation to diagnostics in fields and for the purpose of quarantine and disease management.  相似文献

One hundred and four strains of heterotrophic bacteria have been isolated and characterized from two species of bivalve mollusks cultivated in the Gulf of Nha Trang (Vietnam) and from the water of a mariculture farm. The isolates have been identified on the basis of morphological, physiological, biochemical, and chemotaxonomic properties, as well as by the content of G+C bases in DNA. In the microflora of mollusks, Vibrio alginolyticus was predominant; the pathogenic species V. harveyi and V. splendidus were found as well. Staphylococci and bacilli occupied the second place in abundance after vibrios. In addition, coryneforms and enterobacteria, as well as Pseudomonas spp. and Pseudoalteromonas spp., were revealed. The composition of the water microflora was more diverse as compared with the microflora of mollusks. In the water, Bacillus spp., Vibrio spp., and Pseudomonas spp. were predominant. Brevibacterium spp. and other coryneform bacteria, as well as enterobacteria, occurred in significant amounts. In addition, Pseudoalteromonas spp., Marinococcus sp., Halobacillus sp., Shewanella sp., Sulfitobacter sp., and bacteria of the CFB cluster were noticed. The presence of pathogenic and conditionally pathogenic bacterial species in the water and mollusks is probably the reason for the high death rate of cultivated animals at the mariculture farm.  相似文献

Ganoderma boninense is the causal agent of a devastating disease affecting oil palm in Southeast Asian countries. Basal stem rot (BSR) disease slowly rots the base of palms, which radically reduces productive lifespan of this lucrative crop. Previous reports have indicated the successful use of Trichoderma as biological control agent (BCA) against G. boninense and isolate T. virens 7b was selected based on its initial screening. This study attempts to decipher the mechanisms responsible for the inhibition of G. boninense by identifying and characterizing the chemical compounds as well as the physical mechanisms by T. virens 7b. Hexane extract of the isolate gave 62.60% ± 6.41 inhibition against G. boninense and observation under scanning electron microscope (SEM) detected severe mycelial deformation of the pathogen at the region of inhibition. Similar mycelia deformation of G. boninense was observed with a fungicide treatment, Benlate® indicating comparable fungicidal effect by T. virens 7b. Fraction 4 and 5 of hexane active fractions through preparative thin layer chromatography (P-TLC) was identified giving the best inhibition of the pathogen. These fractions comprised of ketones, alcohols, aldehydes, lactones, sesquiterpenes, monoterpenes, sulphides, and free fatty acids profiled through gas chromatography mass spectrometry detector (GC/MSD). A novel antifungal compound discovery of phenylethyl alcohol (PEA) by T. virens 7b is reported through this study. T. virens 7b also proved to be an active siderophore producer through chrome azurol S (CAS) agar assay. The study demonstrated the possible mechanisms involved and responsible in the successful inhibition of G. boninense.  相似文献

Three new species of the gеnus Entoloma s.l. are described on the basis of morphological and molecular evidence. Two of them are morphologically and phylogenetically closely related to the European species E. violaceozonatum but differ sufficiently by their geographic distribution: E. bulakhae is from the Russian Far East and E. bidupense from Vietnam. The more distant E. atricolor from Vietnam is remarkable because of the brownish-black color of the basidiomata, blackish lamellae edge near the stem, and observed dimorphism in cheilocystidia. These three species are characterized by the combination of clamped basidia and a serrulatum-type lamellae edge and grouped together within the /Inocephalus–Cyanula clade in sister position to /Inocephalus and /Griseorubida. It is argued that they may possibly constitute a separate section in future, when more data will be available. For this reason, section Violaceozonata is proposed here “ad interim” to acknowledge the current isolated position.  相似文献

Aim

Malassezia folliculitis is caused by the invasion of hair follicles by large numbers of Malassezia cells. Several Malassezia researches still use cultures, morphology and biochemical techniques. The aim of this study was to identify Malassezia species isolated from patients diagnosed with folliculitis, at the Parasitology and Mycology Laboratory of Sfax University Hospital, and to explore the genetic diversity of Malassezia by using PCR-RFLP and PCR-sequencing targeting the rDNA region of the Malassezia genome.

Patients and Methods

Specimens were taken from 27 patients with Malassezia folliculitis. For the molecular identification, PCR amplification of the 26S rDNAD1/D2 region was carried out using the Malup and Maldown primers and three restriction enzymes (BanI, MspI and HeaII) for RFLP analysis. The nucleotide sequences of each isolate were compared to those in the NCBI GenBank by using BLASTIN algorithm.

Results

Three species of Malassezia yeasts were identified among the 31 Malassezia strains isolated: M. globosa (83.9%), M. sympodialis (12. 9%) and M. furfur (3.2%). The sequence analysis of M. globosa showed six genotypes.

Conclusion

There is a high genotypic variability of M. globosa colonizing patients with folliculitis.

The species composition and distribution of testate amoebae in Bau Sen and Bau Trang lakes (Binh Thuan Province, Vietnam) were investigated. Fifty-three species, varieties and forms have been identified from the water column and sediments. Twenty-seven species, varieties and forms from the genera Arcella, Centropyxis, Cyclopyxis, Difflugia and Pyxidicula were found in Vietnam for the first time. The updated list of testate amoebae in Vietnam now includes 286 taxa. The genera Arcella, Centropyxis and Difflugia are characterized by the highest frequency of occurrence. The species diversity of Arcella, Centropyxis, Difflugia was the highest. Centropyxis aculeata, Arcella discoides, Difflugia schurmanni, Netzelia oviformis and Difflugia limnetica were the most common species. Many species have shown restricted distribution, some species have been found only in one lake and not been found in another lake. The number of observed species, varieties, and forms in each lake is 34. The average number of the species per sample on the shores of the lakes subjected to human agricultural activity was significantly higher than in the samples from the shores of the lakes with less anthropogenic disturbance. The curves of “cumulative species number vs. sampling effort” are well fitted by equations S = 7.60 N 0.69 for Bau Sen lake and S = 12.52 N 0.46 for Bau Trang. The curves are unsaturated, which indicates that more intensive investigations of testate amoebae should be expected to reveal more species.  相似文献

Endelus (Kubaniellus) indicus sp. n. from India, E. (K.) lao sp. n. and E. (K.) khnzoriani sp. n. from Laos, E. (s. str.) sausai sp. n. from China, and E. (s. str.) dembickyi sp. n. from India are described, the two latter species are included in the Endelus bicarinatus Théry, 1932 species-group recently established by the author. E. collinus Obenberger, 1922 is included in this group; lectotype of this species is designated. Keys to species of the subgenus Kubaniellus and of the E. collinus group are provided. E. (K.) kareni Kalashian is for the first time recorded for Shaanxi Prov., E. pacholatkoi Kalashian, E. smaragdinus Desc. et Vill., and E collinus Obenb., for Laos (the latter species, also for Myanmar).  相似文献

The present study focused on identification and genotypic characterization of Lactic acid bacteria (LAB) in the intestine of freshwater fish. 76 strains of LAB were isolated and identified by 16S rRNA gene sequences and hsp60 gene sequences as different strains of Lactobacillus plantarum, Lactobacillus pentosus, Lactobacillus fermentum, Lactobacillus delbrueckii subsp. bulgaricus, Lactobacillus brevis, Lactobacillus reuteri, Lactobacillus salivarius, Pediococcus pentosaceus, Pediococcus acidilactici, Weissella paramesenteroides, Weissella cibaria, Enterococcus faecium, and Enterococcus durans. The hsp60 gene showed a higher level of sequence variation among the isolates examined, with lower interspecies sequence similarity providing more resolutions at the species level than the 16S rRNA gene. Phylogenetic tree derived from hsp60 gene sequences with higher bootstrap values at the nodal branches was more consistent as compared to phylogenetic tree constructed from 16S rRNA gene sequences. Closely related species L. plantarum and L. pentosus as well as species L. delbrueckii subsp. bulgaricus and L. fermentum were segregated in different cluster in hsp60 phylogenetic tree whereas such a distribution was not apparent in 16S rRNA phylogenetic tree. In silico restriction analysis revealed a high level of polymorphism within hsp60 gene sequences. Restriction pattern with enzymes AgsI and MseI in hsp60 gene sequences allowed differentiation of all the species including closely related species L. plantarum and L. pentosus, E. faecium and E. durans. In general, hsp60 gene with higher evolutionary divergence proved to be a better phylogenetic marker for the group LAB.  相似文献

Six new species are described from Vietnam, Apia simplexsp. n., Bisma angulatasp. n., Pitambara trypetoidessp. n., Sarmatoca cathemerinagen. et sp. n., Maracota soulieraegen. et sp. n., and Serida castaneasp. n., and Zeleja thoracalissp. n. from Malaysia. Two new genera, Maracotagen. n. (type species M. soulierae sp. n.) and Sarmatocagen. n. (type species S. cathemerina sp. n.), and a new subgenus Zelomachasubgen. n. (type species Zeleja thoracalis sp. n.) in the genus Zeleja are erected. The genus Binaluana Soulier-Perkins et Stroiński is downgraded to a subgenus of Zeleja Melichar. The genus Silvispina Wang et Soulier-Perkins with an uncertain systematic position is attributed to the subfamily Lophopinae and the tribe Lophopini. The placement of the genus Elasmoscelis Spinola (and, correspondingly, the tribe Elasmoscelini) in the subfamily Menoscinae, the genus Epiptyxis Gerstaecker in the tribe Menoscini, and the genus Jivatma Melichar in the tribe Lophopini is substantiated. New records of 11 species are given from Vietnam, some being first records for this country.  相似文献
