Tuberculosis vaccine development: recent progress   总被引:45,自引:0,他引:45  
Recent years have seen a renewed effort to develop new vaccines against tuberculosis. As a result, several promising avenues of research have developed, including the production of recombinant vaccines, auxotrophic vaccines, DNA vaccines and subunit vaccines. In this article we briefly review this work, as well as consider the pros and cons of the animal models needed to test these new vaccines. Screening to date has been carried out in mouse and guinea pig models, which have been used to obtain basic information such as the effect of the vaccine on bacterial load, and whether the vaccine can prevent or reduce lung pathology. The results to date lead us to be optimistic that new candidate vaccines could soon be considered for evaluation in clinical trials.  相似文献

The existence of psoriatic arthritis as a distinct clinical entity remains a topic of debate; some authors propose that it is simply the co-occurrence of psoriasis and inflammatory arthritis. However, a distinct entity is likely to have distinct susceptibility factors in addition to those that contribute to psoriasis and inflammatory arthritis alone. These aetiological factors may be genetic and/or environmental, and in this review, the evidence for distinct psoriatic arthritis genetic susceptibility factors is considered.  相似文献

Psoriatic arthritis is a multigenic autoimmune disease that involves synovial tissue, entheseal sites and skin, and that may result in significant joint damage. Although there are no diagnostic tests for psoriatic arthritis, research has identified consistent features that help to distinguish the condition from other common rheumatic diseases. Comparison of HLA-B and HLA-C regions in psoriatic arthritis with those in psoriasis without joint involvement demonstrates significant differences, such that psoriatic arthritis cannot be viewed simply as a subset of genetically homogeneous psoriasis. T-cell receptor phenotypic studies have failed to identify antigen-driven clones, and an alternative hypothesis for CD8 stimulation involving innate immune signals is proposed. Finally, imaging studies have highlighted entheseal involvement in psoriatic arthritis, and it is possible that entheseal-derived antigens may trigger an immune response that is critically involved in disease pathogenesis.  相似文献

Herbaceous energy crop development: recent progress and future prospects   总被引:2,自引:0,他引:2  
Oil prices and government mandates have catalyzed rapid growth of nonfossil transportation fuels in recent years, with a large focus on ethanol from energy crops, but the food crops used as first-generation energy crops today are not optimized for this purpose. We show that the theoretical efficiency of conversion of whole spectrum solar energy into biomass is 4.6-6%, depending on plant type, and the best year-long efficiencies realized are about 3%. The average leaf is as effective as the best PV solar cells in transducing solar energy to charge separation (ca. 37%). In photosynthesis, most of the energy that is lost is dissipated as heat during synthesis of biomass. Unlike photovoltaic (PV) cells this energetic cost supports the construction, maintenance, and replacement of the system, which is achieved autonomously as the plant grows and re-grows. Advances in plant genomics are being applied to plant breeding, thereby enabling rapid development of next-generation energy crops that capitalize on theoretical efficiencies while maintaining environmental and economic integrity.  相似文献

Summary: Of 100 patients admitted to hospital for treatment of psoriasis, 32 had clinical or radiologic evidence of psoriatic arthritis and 17 had both types of evidence. Eight had radiologic evidence of spinal or sarroiliac involvement without symptoms and seven had clinical evidence of peripheral arthritis without radiologic evidence. Patients with psoriatic sacroilitis and spondylitis were most likely to have typical radiograpic changes. It was concluded that psoriatic arthritis is common in patients with severe psoriasis and that is associated with more extensive skin disease than is found in patients without arthritis.  相似文献

Psoriatic arthritis is a greatly underreported seronegative erosive arthropathy, due to the ambiguous lesions it leaves on bone in all but the most severe cases. For a confident diagnosis of psoriatic arthritis to be made, sacroiliac and intervertebral joint fusion must be present together with erosive lesions of the peripheral skeleton including most especially the terminal interphalangeal joints. In modern times it is only a small percentage of cases who experience such debilitating disease, which may explain who so few cases of psoriatic arthritis can confidently be identified from past populations. This report describes this pathological condition as observed in the comingled skeletal remains of nine males and one female from the tomb of Paulus in the Byzantine Monastery of Martyrius, in the Judean Desert. Visual study was complemented using radiographic techniques along with scanning electron microscopy. Two adult males show characteristic lesions of psoriatic arthritis, demonstrating the form known as arthritis mutilans. A third individual shows less widespread erosive lesions which may signify a pauciarticular example of psoriatic arthritis, as is true of most cases in modern times, or the remains may represent Reiter's disease. During the Byzantine period the earlier practise of expelling those with disfiguring diseases (biblical leprosy) evolved into a philanthropic, caring philosophy where the sick were housed and fed out of charity, often within monasteries. The presence of these cases of psoriatic arthritis within such a Judean Desert monastery confirms earlier suggestions that psoriasis was one of the diseases included by those in the ancient eastern Mediterranean under the umbrella term of biblical leprosy. © 1996 Wiley-Liss, Inc.  相似文献

Determination of the crystal structure of the KcsA potassium channel and its subsequent refinement at 2 A resolution have stimulated much interest in modelling of ion channels. The olfactory system of Drosophila resembles that of vertebrates in its overall anatomical organization, but is considerably reduced in terms of cell number, making it an ideal model system to investigate odor processing in a brain [Vosshall LB, Stocker RF: Molecular architecture of smell and taste in Drosophila. Annu Rev Neurosci 2007, 30:505-533]. Recent studies have greatly increased our knowledge about odor representation at different levels of integration, from olfactory receptors to 'higher brain centers'. In addition, Drosophila represents a favourite model system to study the neuronal basis of olfactory learning and memory, and considerable progress during the last years has been made in localizing the structures mediating olfactory learning and memory [Davis RL: Olfactory memory formation in Drosophila: from molecular to systems neuroscience. These highlights of the presentations and discussions at the Second International Workshop on Natural Killer Cells demonstrate the rapid progress that is being made in our understanding of the fundamental nature and mechanism of action of natural killer cells. Although the workshop was purposefully directed to fundamental studies, it is clear that these research efforts will soon have a substantial impact at the clinical level. For example, E. Lotzová (Houston, Tex., USA) noted that several experimental measures of NK function are altered in patients suffering from myelogenous leukemia. NK cells isolated from these patients demonstrate reduced conjugate formation, a substantial reduction in the Vmax for lysis of K-562 cells, could not recycle and were deficient in production of NKCF. She also presented preliminary data that cytotoxic activity could be restored by partially purified alpha interferon and IL-2. A book based on the proceedings of this workshop will soon be published by Academic Press.  相似文献
