Activities of Carboxylation Enzymes in Freshwater Macrophytes   总被引:2,自引:0,他引:2  
Fifteen species of freshwater macrophytes, mainly from cool,temperate waters, were assayed for ribulose bisphosphate carboxylase-oxygenase(RuBPCase) and phosphoenolpyruvate carboxylase (PEPCase) activities.In extracts from all the species RuBPCase was the most activecarboxylation enzyme, and the RuBPCase/PEPCase ratio was atleast 2·0, even for the submersed species Isoetes lacustrisL. and Littorella unifiora (L.) Aschers. which have been reportedto show Crassulacean Acid Metabolism (CAM) activity. The PEPCaseactivity in I.lacustris was lower than that found in some non-CAM-likespecies. In this respect, I.lacustris and L unifiora differfrom most terrestrial CAM plants. However, these two species,along with Potamogeton praelongus Wulf. and Juncus bulbosusvar.fluitans L., had the lowest RuBPCASE/PEPCase ratios, lowerthan found in terrestrial C3 species; suggesting that the potentialfor substantial photosynthetic metabolism of C4 acids existsin some temperate, submersed plants. In the three amphibiousspecies (Potamogeton polygonifolius Pourr., Mentha aquaticaL., and Hippuris vulgaris L.) examined, the aerial leaves exhibitedhigher RuBPCase activities than the submersed leaves. Dark carbon dioxide efflux rates of recently fully expandedleaves and whole plants of Amaranthus hypochondriacus L., Glycinemax (L.) Merr., and Lycopersicon esculentum Mill. grown in controlledenvironments at 35 and 70 Pa carbon dioxide pressure were measuredat 35 and 70 Pa carbon dioxide pressure. Harvest data and whole-plant24-h carbon dioxide exchange were used to determine relativegrowth rates, net assimilation rates, leaf area ratios, andthe ratio of respiration to photosynthesis under the growthconditions. Biomass at a given time after planting was greaterat the higher carbon dioxide pressure in G. max and L. esculentum,but not the C4 species, A. hypochondriacus. Relative growthrates for the same range of masses were not different betweencarbon dioxide treatments in the two C3 species, because highernet assimilation rates at the higher carbon dioxide pressurewere offset by lower leaf area ratios. Whole plant carbon dioxideefflux rates per unit of mass were lower in plants grown andmeasured at the higher carbon dioxide pressure in both G. maxand L. esculentum, and were also smaller in relation to daytimenet carbon dioxide influx. Short-term responses of respirationrate to carbon dioxide pressure were found in all species, withcarbon dioxide efflux rates of leaves and whole plants lowerwhen measured at higher carbon dioxide pressure in almost allcases. Fatty acids of chain length from C4 to C12 inhibited ethyleneproduction in wounded albedo tissue of Hassaku (Citrus hassakuHort. ex Tanaka) fruit. Of the fatty acids tested, caprylicacid (C8) and capric acid (C10) were the most effective. Lauricacid (C12) was less effective, and caproic acid (C6) and butyricacid (C4) were the least effective. Caprylic acid at 5 mM markedlyinhibited ethylene production in not only wounded albedo tissueof citrus fruit but also apple (Malus sylvestris Mill.) cortex,tomato (Lycopersicon esculentum Mill.) pericarp, cucumber (Cucumissativus L.) cortex, banana (Musa AAA group Cavendish subgroup)pulp, broccoli (Brassica oleracea L.) floret, spinach (Spinaciaoleracea L.) leaf, lettuce (Lactuca sativa L.) leaf and mungbean (Vigna radiata [L.] Wilczek) hypocotyl. Caprylic acid inhibitedethylene production at the step of conversion of l-aminocyclopropane-l-carboxylicacid to ethylene. The solanifolia mutant (sf/sf) of tomato (Lycopersicon esculentum)produces leaves consisting of leaflets with entire margins,unlike the lobed leaflets of normal plants. Normal plants treatedwith gibberellic acid (GA3) produced leaves with entire marginswhereas mutant plants exposed to 2-chloroethyl-trimethyl ammoniumchloride (CCC)—an inhibitor of gibberellin biosynthesis—producedlobing of leaflets. The leaf area of the mutant was significantlygreater than that of the normal, but was not significantly differentfrom GA3-treated normal leaves. Similarly, in CCC-treated mutantleaves the leaf area was not different from that of normal untreatedleaves. These observations suggest that the sf/sf mutation affectsthe leaf shape through its effect on endogenous gibberellinsand/or inhibitory substances. Amounts of the enzyme ribulose-1,5-bisphosphate carboxylasewere estimated in seedling leaves of barley (Hordewn vulgareL.) and flag leaves of wheat (Triticum aestitum L.) by radialimmuno diffusion. A fourfold variation among barley varietiesfor amount of RuBPCase at the seedling stage was observed (c.3.5–15mg g–1 fr. wt). Range in variation for amountof flag leaf RuBPCase among wheat varieties was 6-09-9.39 mgRuBPCase g–1 fr. wt. F1 hybrids from interspecific andintergeneric crosses of crested wheatgrasses (Agropyron andElymus spp.) and their amphidiploid analogues were comparedfor amount of RuBPCase in the most recent fully expanded leavesharvested before seed set. Amount of enzyme varied from 3.4to 77.6 mg g–1 fr. wt among the hybrids. No effect chromosomenumber on enzyme concentration was observed among 13 hybridsand their amphidiploid counterparts. Two models for canopy photosynthesis (modified versions of thoseof Acock et al. , 1978 and of Thornley, 1976) were examinedby comparison with experimental photosynthesis data of cucumber(Cucumis sativus L.), sweet pepper (Capsicum annuum L.) andtomato (Lycopersicon esculentum Mill.). The data were obtainedin six large-scale, long-term, semi-commercial cultivationsin greenhouses (Nederhoff and Vegter, 1994). Measured environmentalconditions and measured LAI were input to the model. The emphasiswas on the models' sensitivity to the prevailing CO2 concentration. The (modified) Acock model with 'standard' (originally published)parameters underestimated the photosynthesis rate. This modeltuned to one of our experimental data sets did not fit verywell to the other data sets. As expected, if the model was tunedto each particular data set, it was fairly in agreement withthe measurements, but the fitted parameter values were sometimesquestionable. With the (modified) Thornley model it was obligatoryto estimate or tune the light extinction. The model performedreasonably if all parameters were tuned and also if only thelight extinction was tuned. The modified models were considered usable for practical applications,after parameter tuning. Bud removal and decapitation (disbudding) of plants of tomato(Lycopersicon esculentum Miller) and tobacco (Nicotiana tabacumL.) resulted in an increase in both the concentration and thetotal flux of cytokinin in bleeding xylem sap. There was a retardationin the rate of chlorophyll loss from leaves of disbudded plantsof both species. Ultrastructural examination of the chloroplastsof disbudded tomato plants revealed a maintenance of chloroplastintegrity, compared with chloroplasts from control plants. Itis suggested that the delaying of leaf senescence observed indisbudded plants is due to an increased availability of cytokininto these leaves.  相似文献

The rate of net photosynthesis (P) of whole plant stands oftomato (Lycopersicon esculentum Mill.), cucumber (Cucumis sativusL.) and sweet pepper (Capsicum annuum L.) was measured in sixlong-term experiments in large greenhouses under normal operatingconditions and CO2-concentrations between 200 and 1200 µmolmol-1. The objective was to quantify the responses to lightand carbon dioxide and to obtain data sets for testing simulationmodels. The method of measuring canopy photosynthesis involvedan accurate estimation of the greenhouse CO2 balance, usingnitrous oxide (N2O) as tracer gas to determine, on-line, theexchange rate between greenhouse and outside air. The estimatedrelative error in the observed P was about ± 10%, exceptthat higher relative errors could occur under particular conditions. A regression equation relating P to the photosynthetically activeradiation, the CO2 concentration and the leaf area index explained83-91% of the variance. The main canopy photosynthesis characteristicscalculated with the fitted regression equations were: canopyPmax 5-9 g m-2 h-1 CO2 uptake; ratio Pmax/LAI 1·5-3 gm-2 h-1; light compensation point 32-86 µmol s-1 m-2;light use efficiency (quantum yield) at low light 0·06-0·10µmol µmol-1 and CO2 compensation point 18-54 µmolmol-1. Effects of external nitrogen supply on quantity and activityof ribulose 1,5-bisphosphate carboxylase (RuBPCase) in mulberryleaves (Morus alba L. cv. Shin-ichinose) were examined. PhotosyntheticCO2 fixation and the contents of chlorophyll and phosphoruswere increased by the increased N supply. RuBPCase-protein concentrationand its proportion to total soluble-protein or to total-N rosecorrespondingly as the N supply increased. Net photosynthetic rates per unit ground area for plant standsof Solanum melongena L. var. esculentum (aubergine) and Amaranthuscaudatus L. var. edulis (grain amaranth) were measured over10 min intervals in an airtight, glass, controlled-environmentcabinet for a range of light flux densities provided by thediurnal variation in daylight. Light response curves for photosynthesisof stands, grown at ambient CO2 concentration, were definedat 400, 800 and 1200 vpm CO2. Light compensation points for these stands were around 20-30J m-2 s-1 and decreased slightly at higher CO2 concentrations.For aubergine, a C3 species, the short-term effects of CO2 enrichmentwere to increase the initial slope as well as the asymptoteof the light response curve, reducing light saturation at moderateto high light flux densities; but for amaranthus, a C4 species,saturation was less apparent and CO2 enrichment scarcely increasedphotosynthesis except at light flux densities above 150 J m-2s-1. The canopies intercepted 93-98% of incident light. The efficiencyof utilization of intercepted light in photosynthesis (µgCO2 J-1) increased from zero at the light compensation pointto a maximum at an optimum light flux density of about 100 Jm-2 s-1 (the optimum rose a little with CO2 enrichment) anddecreased slightly with further increase in light. Maximum utilizationefficiencies at 400 vpm CO2 were 8-9 µg CO2 J-1. Enrichmentto 1200 vpm did not affect the peak utilization efficiency ofthe C4 amaranthus, but increased that aubergine to 12·2µg CO2 J-1 (equivalent to some 14% when using the heatof combustion of plant dry matter to convert to the dimensionlessform). Species that showed marked morphological and physiological responsesby their roots to Fe-deficiency (Strategy I plants) were comparedwith others that do not exhibit these responses (Strategy IIplants). Roots from Fe-deficient cucumber (Cucumis sativusL.‘Ashley’), tomato (Lycopersicon esculentumMill.T3238FER) and pea (Pisum sativumL. ‘Sparkle’) plantsproduced more ethylene than those of Fe-sufficient plants. Thehigher production of ethylene in Fe-deficient cucumber and peaplants occurred before Fe-deficient plants showed chlorosissymptoms and was parallel to the occurrence of Fe-deficiencystress responses. The addition of either the ethylene precursorACC, 1-aminocyclopropane-1-carboxylic acid, or the ethylenereleasing substance, Ethephon, to several Fe-sufficient StrategyI plants [cucumber, tomato, pea, sugar beet (Beta vulgarisL.),Arabidopsis(Arabidopsis thaliana(L.) Heynh ‘Columbia’), plantago(Plantago lanceolataL.)] promoted some of their Fe-deficiencystress responses: enhanced root ferric-reducing capacity andswollen root tips. By contrast, Fe-deficient roots from severalStrategy II plants [maize (Zea maysL. ‘Funo’), wheat(Triticum aestivumL. ‘Yécora’), barley (HordeumvulgareL. ‘Barbarrosa’)] did not produce more ethylenethan the Fe-sufficient ones. Furthermore, ACC had no effecton the reducing capacity of these Strategy II plants and, exceptin barley, did not promote swelling of root tips. Growth and photosynthesis of an alloplasmic tomato (cybrid),i.e. line AH47, containing the nuclear genome of the chilling-sensitivecytoplasmic albino mutant of L. esculentum Mill. ‘LargeRed Cherry’ (LRC) and the plastome of a more chilling-toleranthigh-altitude accession of the related wild species L. hirsutumHumb. & Bonpl. LA 1777, were investigated at an optimal(25/20°C) and suboptimal (16/14°C) day/night temperatureregime and their performance compared with that of both euplasmicparents. The cybrid shoot had a similar biomass and developmentrate to the nuclear tomato (L. esculentum) parent at both temperatureregimes. Compared with the biomass production of shoots grownat optimal temperature, the reduction in shoot biomass at suboptimaltemperature was smaller for L. hirsutum than for L. esculentumand the cybrid. This difference was related to a stronger inhibitionof leaf area expansion in L. esculentum and the cybrid in thesuboptimal temperature regime than in L. hirsutum. Irrespectiveof the temperature regime under which the plants were grown,photosynthetic performance and leaf pigment, carbohydrate andsoluble-protein contents of the cybrid resembled those of thenuclear parent. No advantages of the alien L. hirsutum chloroplastwith respect to growth and photosynthesis-related characteristicswere observed in the cybrid in the suboptimal temperature regime,indicating that the temperature sensitivity of the photosyntheticapparatus is regulated by nuclear genes. An adverse consequenceof interspecific chloroplast transfer was the increased susceptibilityto chill-induced photoinhibition of the cybrid. It is concludedthat cybridization is not a useful tool for improving low-temperaturetolerance of tomato. A quantitative examination of the development of the diseasesyndrome created by as permyvirus in tomato (Lycopersicon esculentumMill., cultivar Potentate) revealed an early reduction in subapicalmitotic activity which led to the development of smaller internodes.Later symptoms were associated with necrosis of young axillarybuds and the failure of a new vegetative apex to develop onceflower initiation had begun in the primary apical bud. Application of gibberellic acid (GA3) to the growing point tendedto reverse virus-induced stunting by increasing cell expansionbut the response was less than that in healthy plants. Thiswas due to a reduction in the number of mitoses associated withvirus infection. No consistent difference was found betweenthe endogenous gibberellin levels of healthy and infected plants.It is suggested that the most important single factor in earlysymptom development is a virus-induced reduction in cell divisionwhich cannot be overcome by applying GA3.  相似文献

The stems of ‘Y’-shaped (double stemmed) tomato(Lycopersicon esculentum Mill.) plants were mechanically perturbed(MP) by stroking for 6 successive days. The treatment reducedelongation of the two stems by 40 per cent. When only one branchof the pair was treated, its length was reduced to the sameextent as the two branches in the previous treatment, whilethe elongation of the untreated branch was increased by 60 percent over that of the control. Withholding irrigation induced stem pithiness due to droughtstress in non-MP-plants. However, in MP-pretreated plants, thenumber of pithy internodes was markedly less and the degreeof severity of the disorder was reduced. Ethrel applicationmimicked the effects of MP on pithiness. Negative linear relations were detected (P < 0·005)between the rate of progress from sowing to panicle initiationand CO2 concentration (210-720 µmol CO2 mol-1 air) fortwo genotypes of sorghum [Sorghum bicolor (L.) Moench]. Relationsbetween CO2 concentration and the rate of progress from sowingto first flowering were also negative in soyabean [Glycine max(L.) Merrill] (P < 0·025), but positive in cowpea[Vigna unguiculata (L.) Walp.] (P < 0·025), albeitthat in both grain legumes sensitivity was much less than insorghum. Thus CO2 elevation does not delay flowering in allshort-day species. The considerable effect of CO2 concentrationon times to panicle initiation resulted in large differencesamong the sorghum plants at this developmental stage; with increasein CO2 concentration, plants were taller with slightly moreleaves and more pronounced apical extension. At the same timeafter sowing however, sorghum plants were heavier (P < 0·05)at 210 than at 360 µmol CO2 mol-1 air. Studies have been made on the time to germination and earlyseedling growth of the tomato Lycopersicon esculentum var. Potentateand Lycopersicon pimpinellifolium var. Red Currant, as wellas on derivative generations from their hybridization. Althoughtime to germination showed a genetic component, the relationshipsbetween different genotypes was much influenced by environmentalfactors. A marked maternal effect on time to germination dueto the varying seed sizes was noted while variation betweendifferent genotypes of similar seeds size was ascribed to anendospermic effect. The F1 hybrid embryo with L. Pimpinellifoliumas maternal parent contained more cells than that of the parentbut the hybrid cmbryo with L. esculentum as maternal parentcontained a similar number of cells to that of the parent itself.It is suggested that the results for embryo size both supportAshby‘s assertion that embryo size may be important indetermining heterosis, and also Hatcher’s findings in1940, that heterosis for hypocotyl extension is found in hybridsfrom parcnts of different sized seeds provided the small seededvariety is the maternal parent.  相似文献

The effects of withdrawing nitrogen (N) from the nutrient solutionof adult tomato plants growing in rockwool in a greenhouse wereinvestigated over a 6 week period during fruit production. Thetreatment reduced total plant growth after a lag period of about2 weeks. The commercial fruit yield after 6 weeks of N deprivationwas 7.7 kg m-2compared to 9.3 kg m-2in control plants. Duringthe experiment, growth of the -N plants was fuelled by N reservescontained in both the substrate (rockwool) and in plant organs.The nitrogen budget calculated for -N plants showed that onlya small amount of organic-N was readily available for internalcycling from organs such as stems. It served mainly to feedgrowing fruits which were the main sinks in the plant. The studyalso established that stores of nitrate-N were fully depletedbut it took 45 d for the -N plants to metabolize completelytheir nitrate reserves. This indicates that internal nitrateis not a readily-accessible store of labile N. An estimationof the critical N concentration (%Nc) in the aerial dry matterwas made from the data. Thus, for a crop yielding about 9.9tons DM ha-1, %Ncwas close to 2.5%. This result is discussedin light of existing models that describe the ontogenic declinein %Ncin dry biomass of C3plants. The study indicates that thecurrent regime of N fertilization practised in soilless culturesnot only leads to ineffective nitrogen use but also to largelosses of N to the environment; N concentrations should be decreasedin feeding recipes. We studied the influence of internal ammonium and nitrate contenton the temperature response of ammonium and nitrate uptake inboth chilling sensitive and chilling resistant tomatoes. Threetaxa were examined: Lycopersicon esculentum Mill. cv. T-5, achilling sensitive cultivar, Lycopersicon hirsutum Humb. andBompl. LA 1264, a wild, chilling sensitive accession from thelowlands of Ecuador, and Lycopersicon hirsutum LA1778, a chillingresistant accession from the highlands of Peru. Short exposures(4 h) of L. esculentum cv. T-5 to chilling temperatures irreversiblyinhibited ammonium absorption for at least 6 h. Nitrate absorptionin this taxon and ammonium and nitrate absorption in the L.hirsutum accessions recovered fully and immediately from suchexposures. The chilling resistant accession, L. hirsutum LA1778,showed a lower Q10 for ammonium absorption (1?54?0?10, mean?s.e.)than its chilling sensitive relatives, L. hirsutum LA1264 (2?37?0?35)and L. esculentum cv. T-5 (1?92?0?11). The temperature responseof nitrate absorption depended on internal nitrate status; plantsgrown at high levels of ammonium and nitrate (200 mmol m–3)showed higher Q10's for nitrate uptake (2?29?0?10) than thosedepleted of internal (1?86?0?12).
