The present study was carried out to evaluate the in vitro brain release of prostaglandin F2 alpha (PGF2 alpha), prostaglandin E2 (PGE2), androgens, and 17 beta-estradiol in male and female crested newt, Triturus carnifex, during three different periods of the annual sexual cycle; in addition, the effects of mammalian gonadotropin-releasing hormone (mGnRH), PGF2 alpha, and PGE2 on prostaglandins and steroids release by the brain were evaluated during the same periods. In brain incubations of both sexes, PGF2 alpha and estradiol were higher during postreproduction, while PGE2 and androgens were higher during reproduction. In both sexes, mGnRH increased PGF2 alpha and estradiol during postreproduction, and PGE2 during reproduction; PGF2 alpha increased estradiol secretion during postreproduction. Only in the male, did both mGnRH and PGE2 increase androgens during reproduction. It could be suggested that in Triturus carnifex, the regulation of the reproductive activity in the central nervous system (CNS) depends on the relationships among mGnRH, prostaglandins and steroids. In particular, PGF2 alpha and PGE2 seem to play different roles in the CNS of the newt: PGF2 alpha is involved in the postreproductive processes, through estradiol secretion, while PGE2 in the reproductive ones (through androgens secretion?).  相似文献

Oviductal motility was measured in the isthmus of ovariectomized New Zealand rabbits. The effects of estradiol and progesterone on spontaneous motility and on the response of the oviduct to exogenously administered prostaglandin E1 (PGE1) and PGF were determined. Estradiol treatment significantly increased both the amplitude (P<0.05) and frequency (P<0.01) of spontaneous contractions. The amplitude of spontaneous activity was less following progesterone treatment than following estradiol treatment (P<0.05). Progesterone treatment increased the duration of oviduct response to PGE1 (P<0.05). Estradiol treatment had no effect on the response to PGE1. Increased oviductal activity caused by PGF lasted significantly (P<0.01) longer in ovariectomized, untreated animals than in ovariectomized animals treated with estradiol or progesterone. Progesterone was more effective than estradiol in decreasing the duration of the response to PGF. These effects of steroid hormones on the responsiveness of the oviduct to PGE1 and PGF could contribute to the physiological control of egg transport. The nadir of ovarian hormone influence, as in the recently ovariectomized animals and as occurs immediately after ovulation, is associated with a high responsiveness of the oviduct to PGF. This could effectively increase isthmic occlusion and prevent the eggs from passing through the oviduct prematurely. The gradual increase in ovarian estradiol and progesterone secretion during the 3 days following coitus could result in decreased responsiveness to PGF and increased responsiveness to PGE1. These changes might cause relaxation of isthmic tone and allow movement of eggs through the isthmus into the uterus.  相似文献

Rat pancreas pieces spontaneously released PGE2 (2.3 ng/100 mg × 45 min) and PGF (7.6 ng/100 mg × 45 min). This release corresponds probably to a neo-synthesis since it was abolished by indomethacin. Carbamylcholine (≥ 10 μM), caerulein (≥ 10 nM) and secretin (≥ 10 nM) stimulated the release of PGE2 and PGF : the concentrations of stimulators required to increase PGs release were thus much higher than those which trigger enzyme secretion. Atropine specifically inhibited the cholinergic stimulation, whereas indomethacin blocked the stimulatory effects of all secretagogues. Stimulation of PGE2 and PGF release was reduced in a Ca++-free medium, abolished by EGTA and mimicked by the ionophore A23187, underscoring the crucial role of Ca++ in the regulation of PGs synthesis by the pancreas. Neither PGE2 nor PGF stimulated enzyme secretion in this system and indomethacin did not inhibit the secretory effect of carbamylcholine. Increased synthesis of prostaglandins in response to pancreatic secretagogues does not appear to be involved in the process of enzyme secretion.  相似文献

The presence of an intrauterine device in the pseudopregnant (PSP) rat suppressed decidual cell reaction (DCR). Treatment with PGF or PGE2 also suppressed DCR, but had no effect on IUD horn. Treatment with PGF from days 1–4 before trauma in ovariectomized rats treated with estradiol and progesterone did not have a significant effect on DCR. However, when PGF was given for 4 days after trauma there was significant inhibition of DCR indicating a direct endometrial effect of PGF. Inhibition of DCR was also recorded when indomethacin was administered to intact PSP or to ovariectomized rats maintained on estradiol and progesterone. This indicated that prostaglandins probably are necessary for normal DCR.  相似文献

Three prostaglandins (PGF2α and PGE1, PGE2) have been found in maternal and fetal circulation during labour. Two of these prostaglandins (PGF2α and PGE2) are present in elevated levels in maternal circulation during labour and their presence in fetal vessels has been shown.These three prostaglandins have been tested for their effects on fetal vessels in vitro (umbilical artery and vein, ductus arteriosus, and smaller pulmonary artery). These vessels were selected as being crucial in the conversion from fetal to extra-uterine circulation in mammalian species. Responses of these vessels to the prostaglandins under varying oxygen regimes have been examined as well as their responses to prostaglandin inhibitors. Activity of vessels of varying gestational ages exposed to PGF2α was also examined. The following results were obtained:
1. All vessels, with the exception of pulmonary arteries, contracted in the presence of oxygen over the range 20–100mmHg pO2. At a pO2 of < 20mmHg the ductus arteriosus remained inactive or dilated. Pulmonary arteries dilated at high pO2.
2. All vessels contracted in response to exogenous PGF2α with the exception of the pulmonary arteries which dilated. In the presence of PGF2α, the umbilical veins dilated under low (< 20mmHg) pO2 and contracted at higher levels. Contraction also occurred at lower levels after a period of time.
3. Although PGF2α was capable of causing contraction in the ductus arteriosus at near zero pO2, oxygen, (or possibly the products of oxygenation), appear to be required for continued contraction in the presence of PGF2α. A synergistic relationship between oxygen and PGF2α responses was found as oxygen tensions increased. A synergistic response between PGF2α and oxygen with umbilical arteries which did not increase with increased pO2 was also found. Oxygen tension appeared to have little effect on the response of other vessels to PGF2α.
4. PGE1 caused dilations in all vessels examined. Such dilations appearing to be independent of the oxygen regime prevailing. However, an increase in oxygen during experiments reversed any dilation caused by the prostaglandins.
5. PGE2 caused contractions in umbilical vessels which were independent of oxygen. PGE2 caused contraction of pulmonary arteries. However, in the ductus arteriosus, PGE2 caused an initial contraction followed by a strong dilation. This dilation became weaker as pO2 increased.
6. Additions of prostaglandin inhibitors (Naproxen and Indomethacin) to the bathing solution in which the ductus arteriosus and umbilical arteries were contracting (in response to PGF2α, or oxygen alone) caused a decrease in contractions, and sometimes a slight decrease when the vessel had been pretreated with PGF2α suggesting a possible need for endogenously synthesised prostaglandins for the maintenance of oxygen mediated contractions (in vivo).
7. Vessels responsed to PGF2α at an early gestational age. A role for prostaglandins and oxygen in the closure of fetal vessels is discussed.

Plasma patterns of prostaglandin F2α (PGF2α) and sex hormones (progesterone, androgens and 17ß-estradiol) have been studied in the male crested newt, Triturus carnifex (Laur.), during the sexual cycle. The effects of exogenous PGF2α on sex steroids have also been observed. In addition, effects of one week's captivity are reported. The patterns of plasma sex hormones, during the annual cycle, are consistent with the results previously reported for the same newt species. PGF2α plasma level peaks in April, is low in summer, and progressively increases during autumn to peak again in December. The April PGF2α peak coincides with a plasma estradiol increase and with an adrogens drop. In April-collected newts, moreover, PGF2α treatment induces a significant estradiol increase. These findings lead us to suppose that at the end of the breeding season (April) a PGF2α-dependent estradiol synthesis occurs which could be implied in reproductive peroid termination. In several vertebrates, including some amphibian species, in fact, chronic administration of estradiol results in a strong inhibition of testicular endocrine tissue activity. The putative role of PGF2α-dependent estradiol production in the gonadal regulation in amphibia living in temperate zones is discussed. The autumn PGF2α increase has been tentatively related to the recovery gonadal processes and secondary sexual character development.  相似文献

Prostaglandin biosynthesis was studied in the rat uterus during the oestrous cycle. Uterine homogenates were incubated for 20 minutes in the presence of exogenous substrate (2.10−5M). PGF and PGE2 were measured by R.I.A.. A sharp peak PGF and a smaller peak of PGE2 were observed at prooestrus, 20 h. Another small PGE2 peak occurred at dioestrus II, 15 h. The lowest values of both PGs were found on dioestrus, 15 h. Plasma oestradiol concentration were highest at proestrus, 15 h and 20 h. A sharp progesterone peak occurred at prooestrus, 20 h. The PGF peak is next to the oestradiol peak and is superimposable or lags slightly beyond the progesterone peak.Incubation with 14C arachidonic acid and subsequent analysis of extracts by TLC and scanning showed that the major metabolite is PGI2, identified as 6 keto PGF. The conversion rate of arachidonic acid into 6 keto PGF is 5 times higher than into PGF. 6 keto PGF was further identified by GC/MS. No significant difference was observed between 6 keto PGF production during oestrus and dioestrus.  相似文献

Four antiestrogens (anordiol, tamoxifen, RU 39411, ICI 182780) and the antiprogestin, mifepristone (RU 486), were administered to the following three animal models: (1) ovariectomized rats, (2) mated rats treated post-coitally; and (3) pregnant rats treated post-implantation. The antiestrogens were administered alone or in combination with mifepristone at doses effective in preventing and/or terminating pregnancy in rats. The objective of the study was to determine whether these drugs influenced uterine concentrations of prostaglandins (PGF and PGE2).Antiestrogens administered alone to ovariectomized rats did not effect uterine PGE2 or PGF concentrations; whereas the combination of anordiol/mifepristone increased uterine PGF concentration, resulting in an increase in the PGF/PGE2 ratio.Mated rats were treated post-coitally for three consecutive days with anordiol, tamoxifen, estradiol and mifepristone alone and with the combination of anordiol/mifepristone and tamoxifen/mifepristone. An increase in uterine PGF concentrations and in the PGF/PGE2 ratio occurred only in anordiol/mifepristone treated group. A decrease in uterine PGE2 concentrations occurred in animals treated with anordiol, tamoxifen and estradiol, resulting in an increase in the PGF/PGE2 ratio.Anordiol (5.0 mg/kg/day) and mifepristone (4.0 mg/kg/day) alone and the combination of anordiol/mifepristone (2.5/1.0 mg/kg/day) administered to pregnant rats on days 7, 8 and 9 of pregnancy induced an increase in PGF levels without affecting uterine PGE2 concentration. The changes in uterine PGF concentrations induced by anordiol and the combination of anordiol/mifepristone resulted in an increase in the PGF/PGE2 ratio.The antiestrogens tested except for ICI 182780 possessed agonist activity when assayed by measuring their capacity to increase the uterine weights in ovariectomized rats. Also, ICI 182789 was the only antiestrogen that did not influence uterine PG concentrations. It can be concluded that ICI 182780 is the only “pure” antiestrogen among those tested.The present results show that antiestrogens and the combination of mifepristone plus anordiol at doses preventing implantation and terminating pregnancy increase uterine PGF and/or decrease PGE2 concentrations, resulting in an alteration of PGF/PGE2 ratio. These findings suggest that there exists a critical balance of PGF to PGE2 concentrations in the uterus required for the normal passage of fertilized ova through the oviduct, initiating implantation of the blastocysts, development of embryos, and maintenance of pregnancy.  相似文献

When ovine large luteal cells are placed in culture and exposed to PGF, there is a rapid and sustained increase in the concentration of free intracellular calcium which is believed to play a major role in the luteolytic and cytotoxic effects of PGF. Since administration of exogenous PGE2 can prevent spontaneous and PGF-induced luteolysis in vivo, and the cytotoxic effects of PGF on large luteal cells in vitro, the objective of this study was to determine if one mechanism by which PGE2 acts is to attenuate increases in free intracellular calcium induced by PGF. At concentrations of 10 nM or greater, PGF caused a significant and sustained increase in free intracellular calcium in large luteal cells. Similarly, PGE2 also induced increases in free intracellular calcium but required doses 20-fold greater than PGF. When PGE2 (1, 10 or 100 nM) was incubated with PGF (100 nM) increases in free intracellular calcium induced by PGF were attenuated (P<0.05) when measured 5 min, but not at 30 min, after initiation of treatment. The observed decrease in the concentration of free intracellular calcium at 5 min in response to PGF was the result of fewer cells responding to PGF. In addition, the concentrations of free intracellular calcium attained in the cells that did respond was reduced 25% compared to cells treated with PGF alone. Thus, part of the luteal protective actions of PGE2 appears to involve an inhibition of the early (5 min) increase in free intracellular calcium induced by PGF.  相似文献

The receptors mediating prostanoid-induced contraction of guinea-pig isolated trachea have been characterised in terms of a recently proposed general classification of prostanoid receptors. Results obtained on the trachea were compared with those obtained on guinea-pig fundus, which contains a sub-type of PGE2-sensitive (EP-) receptor termed the EP1-receptor, and guinea-pig lung strip, which contains a thromboxane-sensitive or TP-receptor. The following agonists were studied, PGE2, PGF2α and the thromboxane-like agonists U-46619 and Wy17186. The antagonists studied were SC-19220 which selectivity blocks EP1-receptors, and AH19437 which selectively blocks TP-receptors. On guinea-pig fundus the rank order of agonist potency was PGE2 > PGF2α > Wy17186 U-46619, and responses to all agonists were antagonised by SC-19220 but not by AH19437. On guinea-pig lung strip the rank order of potency was U-46619 > Wy17186 PGF2α > PGE2 and responses to all agonists tested were blocked by AH19437 but not by SC-19220. On the trachea, the rank order was PGE2 = U-46619 > Wy17186 = PGF2α. SC-19220 antagonised responses to PGE2 and PGF2α, but not those to U-46619 or Wy17186. Conversely, AH19437 antagonised responses to U-46619 and Wy17186 but not those to PGE2 or PGF2α. It is concluded that prostanoid-induced contractions of guinea-pig trachea can be mediated by both EP1- and TP-receptors.  相似文献

Purified preparations of ovine large luteal cells were utilized in a series of experiments to test the effects of prostaglandins (PG) E2 abd F2α on cell morphology, viability and secretion of progesterone. Luteal cells were allowed to attach to culture dishes overnight before experiments. In the first series of experiments incubation of large steroidogenic cells with PGF2α for 6 hr resulted in morphological changes including a retraction of the cell cytoplasm and apparent extrusion of cytoplasmic components which became more pronounced after 12 hr. In a second series of experiments, PGF2α decreased and PGE2 increased progesterone accumulation in media after 6 hr when media were not replaced during the incubation period, while progesterone accumulation was not different than that observed in control dishes when both prostaglandins were present. Hourly replacement of the media negated the inhibitory effects of PGF2α but had no effect on the stimulated secretion of progesterone induced by PGE2. Finally, in incubations without media replacement, PGF2α induced a dose-dependent decrease in progesterone accumulation while PGE2 elicited a biphasic response with progesterone secretion increasing from 0.1 ng/ml to maximal levels at 10 ng/ml followed by a dose-dependent decrease at 100 and 1000 ng/ml. These data are compatible with the hypotheses that: 1) luteolysis is initiated, at least in part, by an action of PGF2α on large luteal cells; and 2) the embryonic signal from the pregnant uterus which rescues the ovine corpus luteum may be PGE2.  相似文献

To evaluate the details of the adrenergic stimulation of urinary prostaglandins in man, ten normal volunteers were given various agonists and antagonists. The effect of 4 hour IV infusions of norepinephrine (NE), NE + phentolamine (PHT), NE + phenoxybenzamine (PHB), NE + prazosin (PZ), isoproterenol (ISO), and PHT alone on urinary PGE2 and PGI2 (6 keto PGF) were determined. PGE2 and 6 keto PGF were measured by radioimmunoassay from 4 hour urine samples. NE stimulated both PGE2 (196±40 to 370±84 ng/4 hrs/g creatinine and 6 keto PGF1α(184±30 to 326±36), both p<0.01. In contrast, ISO had no effect on either PGE2 or 6 keto PGF excretion. Alpha blockade with PHT. PHB, or PZ inhibited the NE induced systemic pressor effect. However, the effect of the alpha blockers on the NE induced stimulation of PGE2 and 6 keto PGF varied. PHT did not alter the NE stimulated PGE2 or 6 keto PGF release (370±84 vs. 381±80) PGE2 and (326±50 vs. 315±40) 6 keto PGF, both p>0.2). PHT alone stimulated only 6 keto PGF. PHB and the specific α1 antagonist PZ similarly eliminated the NE induced prostaglandin release. These results suggest that adrenergically mediated urinary prostaglandin release in man is via an alpha receptor with α1 characteristics.  相似文献

Prostaglandin (PG) E2 was the major PG released from the superfused guinea-pig uterus on Day 7, followed by in descending order 6-oxo-PGF, thromboxane (TX) B2 and PGF. However, the outputs of all four substances were low and were very similar. By Day 15, PGF output from the superfused uterus had increased 21.9-fold, whereas the outputs of PGE2, 6-oxo-PGF and TXB2 had increased only 1.8-, 2.9- and 1.2-fold, respectively. A mechanism is apparently “switched on” between Days 7 and 15 which causes a fairly specific increase in the release of PGF from the uterus.Progesterone and/or estradiol had no effect on PG or TX release when superfused over the uterus on Day 7, nor did they have any effect on PG and TX release from the Day 15 uterus when administered separately. When administered together, however, they significantly inhibited PGF, PGE2 and 6-oxo-PGF, but not TXB2, release from the Day 15 uterus. Oxytocin had no effect on PG release from the Day 7 or Day 15 uterus, while A23187 stimulated PGF, 6-oxo-PGF and, to a lesser extent, PGE2 release from the uterus on both Days 7 and 15 Oxytocin is apparently not important for stimulating PGF release from the guinea-pig uterus in relation to luteolysis, whereas increasing intracellular free Ca++ levels may be part of the mechanism for “switching on” uterine PG synthesis. Furthermore, changes in intracellular free Ca++ levels in the endometrium may be responsible for the pulsatile nature of PGF release from the uterus.  相似文献

The results of the present study establish that 1.5 mg PGE2 (lyophilized sodium salt) incorporated in one cm long open-ended Silastic-polyvinylpyrrolidone (PVP) tube when inserted into 10 day pregnant rats induced abortion within 70–72 hours in all the treated rats. A combined treatment of PGE2 and 17β-estradiol failed to increase the abortion inducing effect of a Silastic-PVP-PGE2 tube. It is observed that PGE2 is about 4 times less potent than PGF in inducing midterm abortion in rats. It is suggested that either PGE2 exerts luteolytic effect after being converted to PGF, although how it occurs is not clear; or PGE2 causes expulsion of the fetuses by its uterine stimulating property. 17β- estradiol increases the uterine synthesis of PGF as described earlier but seems not affecting the production of PGE2 by the uterus. Experiments were carried out to assess the influence of prostaglandins (viz. PGE1, PGE2 and PGF) on plasma concentrations of FSH and LH in the female rhesus monkey. Monkeys were ovariectomized and treated with estradiol benzoate to suppress endogenous gonadotropin levels prior to these experiments. Femoral venous blood was taken at intervals following a single carotid arterial injection of the PG in anesthetized monkeys. FSH and LH concentrations, determined by radioimmunoassay, were not significantly altered in 4 control animals receiving saline (2) or ethanol-saline (2), the vehicles for PGF and for the E series PGs, respectively. PGE1 (5mg) effected dramatic elevations of LH within 5 min in 3 animals and the high plasma concentrations were maintained at least for 60 min. Similarly, 5.0 mg of PGE2 effected rapid elevation of LH concentrations, from 2- to 7-fold pre-injection levels in 3 animals. In contrast, FSH levels were not so markedly altered by PGE1 and PGE2, but in general, appeared to be somewhat decreased by these treatments. PGF had no effect on plasma FSH and LH concentrations. These data demonstrate the ability of PGs of the E series to elevate plasma LH concentrations in the rhesus monkey and support studies in other species suggesting a modulating role for PGs on gonadotropin secretion or release.  相似文献

Exogenously administered PGE1 or PGE2, like atropine, markedly decreased both the flow and calcium concentration of parasympathetically evoked rat parotid saliva: PGF was less effective. Despite the fact that prostaglandins greatly reduced the Ca concentration of nerve-evoked saliva, they did not change the glandular Ca concentration of either control or parasympathetically stimulated parotid glands. Prostaglandins (20 μg/kg, i.a.) decreased the Na or K concentration of nerve-evoked parotid saliva, but at lower doses had no significant effect. PGE1, PGE2, PGF or atropine markedly decreased flow rates of similarly evoked rat submandibular saliva. Prostaglandins and atropine, however, decreased the Na concentration and increased the K concentration of parasympathetically evoked submandibular saliva. PGF, like atropine, increased the Ca concentration of such saliva. Drug vehicle, ethanol, slightly decreased the flow of both parotid and submandibular saliva but not the ion secretion. Endogenous prostaglandins themselves may not play a role in a secretory activities during parasympathetic nerve stimulation of rat salivary glands, since administration of indomethacin, an inhibitor of prostaglandins biosynthesis, prior to or during nerve stimulation did not significantly alter nerve-evoked salivary secretion. The mechanisms by which prostaglandins modulate secretory responses of salivary glands during parasympathetic stimulation are not understood.  相似文献

In these experiments we have examined the effects of PGE1, PGE2, PGF and PGF on synovial perfusion in the normal canine synovial microcirculation. The effects of the drugs on synovial perfusion were determined indirectly from the changes produced in the rate of clearance of 133Xenon from the joint by their intra-articular injection. Prostaglandins PGE1 and PGE2 were found to be strongly vasodilator with PGE1 being the more active. PGF appeared to have little or no vasoactive properties in doses up to 1 ugm. (2.8 × 10−5M) in our I preparation while PGF was vasodilator at this high dosage only. Neither SC19920 nor diphloretin phosphate antagonised the effects of PGE1 in these experiments.  相似文献

Thw radioimmunological (RIA) determination of prostaglandin (PG) E2 and of PGF in urine humans and rats is described in detail. After extraction and chromatography PGE2 was determined by using a PGE specific antibody or by using either PGB or PGF specific antibodies after the respective conversion procedures. The three different RIA procedures were compared to each other. PGF was determined by a specific antibody to PGF. Basal excretion of PGE2 and of PGF in healthy women on free diet was 9.3 ng/hour ± 0.96 and 18.3 ng/hour ± 2.5 respectively. Furosemide increased the excretion of PGE2 and of PGF in humans significantly, while PG-excretion rates decreased on indomethacin. In rat urine PGE2 and PGE increased markedly from 46.2 pg/min ± 9.3 and 27 ± 3.4 to 253.8 ± 43.3 and 108 ± 12.6 pg/min (per one kidney) in the anesthetized-laparotomized animal. This increase was abolished after giving two different PG synthetase inhibitors.  相似文献

Prostanoids play an important role throughout all of pregnancy and during the initiation and progress of labor. The human placenta at term produces large quantities of prostanoids, yet little is known of the factors that regulate their biosynthesis. Herein, we report the effect of estradiol or estradiol and progesterone on the basal release of placental prostanoids from fresh human term placental explants using a perifusion system.The basal release of prostaglandin E2 (PGE2, prostaglandin F (PGF), thromboxane (TxB2) and 6-keto-prostaglandin F (6-keto-PGF) increased about 50% from the fifth to the ninth hour in culture, while the release of 13, 14-dihydro-15-keto-PGF (PGFM) remained constant and hCG release decreased. The dose-related effect of estradiol (20–2,000 ng/ml) in the perifusing medium starting at the fifth hour of perifusiOn (i.e., the zero treatment time) effected no change in the release of TxB2, PGF, PGFM or hCG. A biphasic action on the release of 6-keto-PGF,. was observed, i.e. it was significantly decreased when incubated with 20 ng/ml of estradiol, but effected an increase after exposure to 200 ng/ml. The concomitant addition of progesterone (2,000 ng/ml) with estradiol (200 ng/ml) significantly inhibited the stimulatory action of estradiol at this dose. The release of PGE2 was inhibited in a dose-related fashion with increasing dose of estradiol. We report here that intact Gram positive and Gram negative bacteria, in the presence of exogenous arachidonic acid, produce and release PGE2 and PGF into the medium as measured by radioimmunoassay. The seven bacterial strains so far studied release 6.5–50.9 ng PGE2 and less than 0.02–0.51 ng PGF per mg bacterial protein during a 1 hour incubation, quantities of the same order of magnitude as those observed in mammalian systems. PGE2 and PGF formation in bacteria are inhibited by indomethacin.  相似文献
