Germination and growth of six xerophilic fungi, Aspergillus flavus, Aspergillus ochraceus, Eurotium chevalieri, Chrysosphorium fastidium, Wallemia sebi and Xeromyces bisporus were examined on media of a wide range of water activities (aw). The influence of three solutes, NaCl, glycerol and a glucose/fructose mixture, was studied at pH 4-0 and pH 6-5 using a plate-slide technique. Germination times and growth rates were affected by solute type, but the influence of pH was less marked. Except for Wallemia sebi, the fungi grew most strongly on glucose/fructose and were partially or completely inhibited by NaCl. The results showed that a universal isolation medium for xerophilic fungi could be based on glycerol or glucose/fructose but not on NaCl as aw-limiting solute.  相似文献

An improved viable counting technique was developed to facilitate study of the heat resistance of fungal spores. Spores were heated and subsequently incubated in the same medium. After germination, hyphae and germ tubes were stained with lactofuchsin, and the germinated spores were counted with the aid of a microscope. A number of xerophilic strains were examined, mostly isolates from spoiled highmoisture prunes. Of these, ascospores of Aspergillus chevalieri, A. mangini, and Xeromyces bisporus were the most heat-resistant. A decimal reduction curve obtained for A. chevalieri was specified by a z value of 23 F and an F(180) of 2.2 min.  相似文献

We provide a phylogenetic diversity assessment study in genus Agaricus as part of our ongoing work to saturate ITS and LSU rDNA sequence diversity of soil-dwelling fungi in Alaska. Pairwise sequence similarity-based groupings and statistical parsimony analyses were applied to delimit operational taxonomic unit (OTU) and were compared to results of full phylogenetic analyses. Our results show that the proportion of section Arvenses taxa is particularly high in the boreal forest and hypo-arctic (low arctic) regions, whereas the genus is represented by section Agaricus in high arctic habitats. Furthermore our findings suggest that the commercially important A. bisporus occurs naturally in the boreal region of interior Alaska, substantially expanding the known northern limit of the species. Delimitations of OTU varied greatly with different methods. In general 95% similarity-based grouping proved to be the least sensitive method, often resulting in section- and subsection-level groups. The 95% connection-limit statistical parsimony separated far more groups. The 98% similarity-based groups and the 98% connection limit networks recognized respectively 11 and 13 OTU containing our specimens. The 98% connection limit statistical parsimony was the only method in which all recognized OTU consisted of members grouped by branches with significant (> .95) posterior probabilities, providing an independent support for the groups. Our results also point out that considerable additional efforts will be needed to elucidate the evolution of this diverse genus and to assess its phylogenetic diversity, given that most taxa in our analyses could not be placed convincingly within well characterized species using ITS/LSU data.  相似文献

Wallemia (Wallemiales, Wallemiomycetes) is a genus of xerophilic Fungi of uncertain phylogenetic position within Basidiomycota. Most commonly found as food contaminants, species of Wallemia have also been isolated from hypersaline environments. The ability to tolerate environments with reduced water activity is rare in Basidiomycota. We sequenced the genome of W. sebi in order to understand its adaptations for surviving in osmotically challenging environments, and we performed phylogenomic and ultrastructural analyses to address its systematic placement and reproductive biology. W. sebi has a compact genome (9.8 Mb), with few repeats and the largest fraction of genes with functional domains compared with other Basidiomycota. We applied several approaches to searching for osmotic stress-related proteins. In silico analyses identified 93 putative osmotic stress proteins; homology searches showed the HOG (High Osmolarity Glycerol) pathway to be mostly conserved. Despite the seemingly reduced genome, several gene family expansions and a high number of transporters (549) were found that also provide clues to the ability of W. sebi to colonize harsh environments. Phylogenetic analyses of a 71-protein dataset support the position of Wallemia as the earliest diverging lineage of Agaricomycotina, which is confirmed by septal pore ultrastructure that shows the septal pore apparatus as a variant of the Tremella-type. Mating type gene homologs were identified although we found no evidence of meiosis during conidiogenesis, suggesting there may be aspects of the life cycle of W. sebi that remain cryptic.  相似文献

Twelve isolates from the genus Agaricus (Fungi, Basidiomycota) were investigated for their ability to support development of the phorid fly, Megaselia halterata (Wood), which is an important pest of the commercial white button mushroom Agaricus bisporus. Combined effects of oviposition of adult female M. halterata and larval development in mushroom compost inoculated with Agaricus mycelium were determined using bioassays. The numbers of M. halterata offspring that developed were affected by the Agaricus isolate used, and there was a significant separation between resistant and susceptible isolates. In a bioassay where the female phorids had a choice of all 12 isolates for oviposition, three isolates produced >200 adults per 100 g compost pot while the remaining nine isolates had <20 adults per pot. Where there was no choice of Agaricus isolate for oviposition, five isolates resulted in >100 adults per 100 g compost pot while the remainder resulted in <4 adults per pot. With the susceptible isolates, there was a positive correlation between increasing concentration of mycelium in the substrate and phorid development until the concentration exceeded 40% after which numbers of emerging phorids declined. Genetic identity of Agaricus isolates was determined using ITS sequencing and phylogenetic methods, which revealed two major cluster groups. Isolates supporting the development of large populations of M. halterata were located in one of these clusters (group I), and were either Agaricus bisporus or other species from the same Agaricus section Duploannulatae. Isolates that did not support the development of M. halterata populations were located in a different cluster (group II) and were more genetically distant from A. bisporus, e.g. Agaricus sections Arvenses, Minores and Xanthodermatei. Species of Agaricus with resistance to M. halterata could have significant potential for the breeding and cultivation of phorid-free mushrooms.  相似文献

红曲菌(Monascus spp.)是我国重要的药食两用微生物资源之一,能够产生天然食品添加剂红曲色素、降血酯活性物质Monacolin K等有益次生代谢产物,但也能分泌真菌毒素桔霉素(Citrinin),红曲菌及其相关产品的安全性由此受到质疑.因此,如何促进红曲菌有益代谢产物的产生,减少或抑制桔霉素的产生成为广大科研工作者研究的重点方向.近年来,红曲菌的分子生物学研究有了较快的发展,红曲菌次生代谢产物生物合成及其调控的研究是热点.本文重点介绍红曲色素、Monacolin K和桔霉素生物合成途径及相关基因的研究进展,以期为有效调控红曲菌次生代谢产物的产生、提高红曲产品的安全性提供参考和借鉴.  相似文献

The predominant spoilage fungi of dried and high-moisture prunes were members of the Aspergillus glaucus group and Xeromyces bisporus. Chrysosporium spp. were also important. At the mean pH of prune flesh (3.8) and at 25 C, X. bisporus grew at water activities (aw) down to 0.605, and Chrysosporium fastidium grew to 0.686. Germination was always followed by growth, but within the 120-day incubation period, the minimum aw permitting asexual sporulation was usually higher than that permitting germination. Sexual sporulation often required an even higher aw. The water requirements of aspergilli were appreciably greater at this pH than near neutrality, no species germinating below 0.738 aw. This was probably a consequence of a high spore-death rate during incubation at low aw and pH.  相似文献

Melanized yeast-like meristematic fungi are characteristic inhabitants of highly stressed environments and are rare eukaryotic extremophiles. Therefore, they are attractive organisms for studies of adaptations. In this study we compared two meristematic species of the genus Trimmatostroma on media of differing water potentials isolated from distinct water-stressed environments: T. salinum from the hypersaline water of a solar saltern, and T. abietis from a marble monument in Crimea. The morphology and melanization of both isolates in response to sodium chloride-induced water stress were investigated by means of light and electron microscopy. We describe and compare the colony form and structure, ultrastructure, and degree of cell-wall melanization of both species in reaction to salinity and to inhibited melanin synthesis. The halophilic T. salinum responded to changed salinity conditions on the level of individual cell ultrastructure and degree of cell wall melanization, whereas the xerophilic rock-inhabiting T. abietis responded with modification of its colony structure. Surprisingly, both the halophilic and the xerophilic Trimmatostroma species were able to adapt to hypersaline growth conditions, although their growth patterns show distinct adaptation of each species to their natural ecological niches.  相似文献

We obtained four isolates of the xerophilic genus Wallemia from the rooftop of a house made of red brick and cement in an agronomic field planted with common beans and maize in Pachacamac, Lima, Peru. Bayesian phylogenetic analysis with rDNA gene sequences showed these Wallemia isolates form a distinct and strongly supported clade closely related to W. hederae. We examined the macro and micromorphology, growth rate and production of exudates of isolates on media containing different amounts of glucose and NaCl (water activity from 0.9993 to 0.8480). Their chaotropic and kosmotropic tolerance were tested on media with multiple molar concentrations of MgCl2 and MgSO4 (water activity from 0.9880 to 0.7877). Isolates are xerophilic and halotolerant, growing on 17% NaCl-supplemented media (water activity = 0.8480). Maximum concentrations of MgCl2 and MgSO4 at which growth was observed were 1.7 and 3.5 M, respectively. Isolates were shown to represent a novel species, described as Wallemia peruviensis sp. nov. In contrast to W. hederae, W. peruviensis does not produce exudates on malt extract agar + 17% NaCl media. An updated dichotomous key to Wallemia species is provided. This is the first new species of Wallemia described from South America and the first association of a Wallemia species with an agricultural environment on this continent.

A non-radioactive PCR coupled ligase detection reaction was developed to discriminate the food spoilage yeasts Zygosaccharomyces bailii and Z. bisporus from each other and from other members of the genus. A short region of the 18S rRNA gene was amplified from boiled cell lysates and polymerase chain reaction (PCR) products used as target in the template directed ligation of two adjacent oligonucleotides. Ligated products were captured using biotin-streptavidin chemistry and detected using digoxigenin
immuno-chemiluminescence. The ligase detection reaction was able to discriminate to the species level, targeting a single base deletion. The specificity of the reaction was assessed using seven species of the genus Zygosaccharomyces . Only strains of Z. bailii and Z. bisporus gave positive results with their respective primer sets. The lower detection limit of the strategy was 10pg (3 times 107 targets) of amplified product.  相似文献

1980—1981年自北京、福州、三明、泉州、漳州、厦门和昆明等地双孢蘑菇[Agaricusbisporus(Lange)Sing]病菇上和不出蘑菇的培养料内,分离到一种真菌。在 PDA 培养基上的培养性状,肉眼观察与蘑菇轮枝菌(Verticillium psalliotae Tresch.)极难区别。根据其分生孢子和瓶梗的形态及其在匐匍气丝上的排列情况等,鉴定为珠网丝枝霉。因具短棒状原瓶梗,认为是一新变种,定名为珠网丝枝霉中国变种 Aphanocladium aranearum(Petch)W.Gams var.sinense J.D.Chen var.nov.它是双孢蘑菇的重要害菌之一。这个“属”和“种”都是我国的新记录。在猴头(Hericium erinaceus)、香菇(Lentinus edo-des),糙皮侧耳(Pleurotus ostreatus)上和水稻土中也有少量分布。  相似文献

The taxonomic validity of the genus Hydropuntia Montagne (1843) (including Polycavernosa) within the Gracilariaceae (Gracilariales, Rhodophyta) is controversial. Morphological characters that define species of Hydropuntia are said to be variable and to overlap with those of Gracilaria. Here we present a global phylogenetic study of the family based on a Bayesian analysis of a large rbc L DNA sequence dataset indicating that the genus Hydropuntia forms a well supported monophyletic clade within the family, and recognize Hydropuntia as a genus distinct from Gracilaria. We also conducted smaller phylogenetic analyses in which thirty four Hydropuntia rbc L sequences resulted in two major clades within the genus, comprising a Caribbean clade and an Indo-Pacific clade. Diagnostic reproductive stages that separate these two clades will be illustrated.  相似文献

The evolution of breeding systems was studied in the genus Acer, with special attention to the origin of androdioecy and dioecy, using a phylogenetic approach. Parsimony and maximum-likelihood techniques were used to infer the ancestral character state and trends in the evolution of breeding systems. Information on breeding systems was obtained from the literature, and phylogenetic relationships were taken from three published phylogenies. Although a general trend from duodichogamy to dioecy through heterodichogamy has been proposed for the genus Acer, our results show that a general trend is not detected when phylogenetic relationships are taken into account. Dioecy appeared as a derived state that evolved at least three times and never reversed towards other states. Three different paths to dioecy have been followed in the genus Acer: from heterodichogamous androdioecy; from heterodichogamous trioecy; and from dichogamous subdioecy. Therefore, although the best documented cases of evolution of androdioecy indicate that this breeding system evolves from dioecy, in the genus Acer the opposite situation occurs (androdioecy leading to dioecy). Here we discuss the role of inbreeding avoidance and sexual specialization as selective forces driving the evolution of dioecy in the genus Acer.  相似文献

A phylogenetic tree for fowl including chicken in the genus Gallus and based on mitochondrial D-loop analysis further supports the hypothesis developed from morphology and progeny production that red junglefowl (RJF) is the direct ancestor of the chicken. The phylogenetic positions of the chicken and the other fowl species in the genus Gallus are of great importance when considering maintenance and improvement of chicken breeds through introgression of genetic variation from wild-type genomes. However, because the phylogenetic analysis based on the DNA sequences is not sufficient to conclude the phylogenetic positions of the fowls in the genus, in the present study, we have determined sequences of whole mitochondrial DNA (mtDNA) and two segments of the nuclear genome (intron 9 of ornithine carbamoyltransferase, and four chicken repeat 1 elements) for the species in the genus Gallus. The phylogenetic analyses based on mtDNA sequences revealed that two grey junglefowls (GyJF) were clustered in a clade with RJFs and chicken, and that one GyJF was located in a remote position close to Ceylon junglefowl (CJF). The analyses based on the nuclear sequences revealed that alleles of GyJFs were alternatively clustered with those of CJF and with those of RJFs and chicken. Alternative clustering of RJF and chicken alleles were also observed. These findings taken together strongly indicate that inter-species hybridizations have occurred between GyJF and RJF/chicken and between GyJF and CJF.  相似文献

以中国产熊野藻属Kumanoa的两个种, 绞扭熊野藻K. intorta (=绞扭串珠藻Batrachospermum intortum), 弯形熊野藻K. curvata (=弯形串珠藻B. curvatum)和其他6种淡水红藻为实验材料, 对其psaA和psbA基因进行扩增和测序, 并与GenBank中相近序列进行比对分析, 以贝叶斯法、最大似然法和邻接法分别构建了单基因和联合基因系统发育树. 结果表明, 3种方法构建的系统树具有相似的拓扑结构, 反映的系统发育关系基本一致, 熊野藻属中的两个种聚为一支, 与串珠藻属相分离, 支持该属的建立; 中国产的熊野藻属分子学研究结果与来自南美洲及澳洲的该属植物结果一致, 说明该属的建立具有广泛的地理适用性. 系统发育树聚类结果也明确反映了熊野藻属与串珠藻属较近的亲缘关系, 根据果胞枝形态特点, 推测熊野藻属进化地位晚于串珠藻属植物, 而早于顶丝藻目和红索藻目. 此外, 胶串珠藻与其他串珠藻组植物分离, 支持将其单独分组, 红索藻目植物与串珠藻目植物分离, 支持红索藻目的建立. 同时也表明psaA和psbA基因用于淡水红藻分析, 能够较好地反映其系统发育关系.    相似文献

Citrinin is a mycotoxin, which is produced by fungi belonging to the genus Monascus, known in biotechnology as producers of azaphilone pigments. The relation between biosynthesis of these secondary metabolites was investigated in different species of the genus Monascus in batch-culture at the following cultivation conditions: T = 28 degrees C, agitation 220 rpm, and a medium, which induce citrinin production, containing ethanol as a carbon source. The screening was carried out with 16 fungal strains and the biosynthesis of citrinin and pigments was monitored quantitatively at the standard conditions mentioned above. Some kinetic parameters of the process have been determined. The values of the growth yield coefficient Y(X/C) were between 0.32 and 0.57. The amount of the extracellular red and orange pigments at the end of cultivation varied for the different strains between 0.09 and 1.33 OU/ mg dry weight, and 0.15 and 0.96 OU/mg dry weight, respectively. The amount of the total pigments measured was between 0.16 and 3.6 OU/mg dry weight, and between 0.21 and 3.39 OU/mg dry weight. The determined ratio 500 nm/400 nm, characterizing the pigment production, ranged between 0.60 and 1.06. Twelve of the investigated strains produced citrinin and pigments, two of them produced only pigments. Two strains were not able to produce neither pigments nor citrinin. Thus, the biosynthesis of citrinin appeared to be strain-specific and does not correlate with the pigments' biosynthesis by the fungal strains belonging to the genus Monascus.  相似文献

The spectacular, virtually endemic radiation of Philippine semi-fossorial skinks of the genus Brachymeles represent one of the few radiations of scincid lizards to possess both fully limbed and limbless species. And yet, nothing is known of the phylogenetic relationships of this exceptional group. Morphologically similar body plans have made it difficult to assess species-level diversity, and the genus has long been recognized as one of the more modest radiations of southeast Asian lizards. However, recent large-scale survey efforts have resulted in the discovery of numerous new species, and taxonomic studies indicate that the diversity within the genus Brachymeles is grossly underestimated. In this study we provide the first robust estimate of phylogenetic relationships within the genus Brachymeles using a multi-locus dataset and nearly complete taxonomic sampling. We provide statistical tests of monophyly for all polytypic species and two widespread limb-reduced species and our results indicate wholesale deviations from past summaries and taxonomic evaluations of the genus. With few exceptions, we are able to reject the monophyly of all polytypic and widespread species, thereby validating the need for large-scale taxonomic revisions. Our results reveal that the limbless, monotypic, genus Davewakeum is nested within Brachymeles. Mapping of body form on our preferred phylogenetic tree suggests that limb-reduction and digit loss has occurred on multiple occasions in the history of the genus. A Bayesian reconstruction of ancestral areas indicates strong statistical support for a minimum of five major dispersal events that have given rise to a major component of the observed species diversity on separate Pleistocene aggregate island platforms of the archipelago.  相似文献

The diverse group of Asian wolf snakes of the genus Lycodon represents one of many poorly understood radiations of advanced snakes in the superfamily Colubroidea. Outside of three species having previously been represented in higher‐level phylogenetic analyses, nothing is known of the relationships among species in this unique, moderately diverse, group. The genus occurs widely from central to Southeast Asia, and contains both widespread species to forms that are endemic to small islands. One‐third of the diversity is found in the Philippine archipelago. Both morphological similarity and highly variable diagnostic characters have contributed to confusion over species‐level diversity. Additionally, the placement of the genus among genera in the subfamily Colubrinae remains uncertain, although previous studies have supported a close relationship with the genus Dinodon. In this study, we provide the first estimate of phylogenetic relationships within the genus Lycodon using a new multi‐locus data set. We provide statistical tests of monophyly based on biogeographic, morphological and taxonomic hypotheses. With few exceptions, we are able to reject many of these hypotheses, indicating a need for taxonomic revisions and a reconsideration of the group's biogeography. Mapping of color patterns on our preferred phylogenetic tree suggests that banded and blotched types have evolved on multiple occasions in the history of the genus, whereas the solid‐color (and possibly speckled) morphotype color patterns evolved only once. Our results reveal that the colubrid genus Dinodon is nested within Lycodon—a clear finding that necessitates the placing of the former genus in synonymy with the latter.  相似文献
