We recorded from single units of individual sensilla of the thoracic infrared (IR) pit organs of Melanophila acuminata. When the organ was stimulated with a thermal radiator whose emission spectrum was similar to that of a typical forest fire, units responded phasically with up to seven spikes within 30–40 ms at a radiation power of 24 mW cm−2. In the experiments all wavelengths shorter than 1.6 μm were excluded by a longpass IR filter. Response latencies were about 4 ms and initial impulse frequencies were up to 250 impulses per second (ips). A single spike could be generated even when stimulus duration was only 2 ms. Reduction of total radiation power from 24 mW cm−2 to 5 mW cm−2 resulted in increased response latencies of 5–6 ms and the occurrence of only two to three spikes. Initial impulse frequencies decreased to 125 ips. According to our physiological results and calculations, Melanophila should be able to detect a 10-hectare fire from a distance of 12 km. Mechanical stimuli also evoked responses of the IR sensilla. All present morphological and physiological findings lead to the conclusion that the IR receptors of Melanophila must function by means of a hitherto undescribed photomechanic mechanism. During recording of impulse activity from single nerve fibers of electroreceptors of the ampullae of Lorenzini of skates, we studied the responses to electromagnetic radiation (EMR) at a frequency of 37–55 GHz and an intensity of 1–100 mW/cm2.Exposure of the ampullar canal pore to EMR at an intensity of 1–5 mW/cm2 and a distance of 1–10 mm evoked a transient increase in the frequency of low-threshold receptor activity (current threshold was 0.04–0.2 µA). An increase in EMR intensity by more than 8–10 mW/cm2 produced, together with elevation of receptor activity, an inhibition due to a rise in temperature of 1–3°C in the region exposed. The phase of increase in frequency of activity was absent in high current-threshold receptors (0.3–2.0 µA) when exposed to EMR. The receptors responded to irradiation of the ampullar canal pore at a distance of 15–20 mm by an increase in discharge frequency for 20 min. Direct irradiation of the ampullae of Lorenzini induced only inhibitory responses in receptor cells regardless of their excitability.The results obtained indicate that the sensory receptors of vertebrates are sensitive to EMR. Summary The relationship between lipid composition and phase transition was investigated by differential scanning calorimetry for intact and membrane phospholipid extracts of wild-type (w/t) and thecel (Tw 40) mutant ofNeurospora crassa. Thecel (Tw 40) mutant (grown on minimal, sucrose medium supplemented with Tween 40 at 34 °C) had approximately twice the saturated fatty acid content ofw/t organisms grown at 22 °C. The gel-liquid crystal phase transitions of ergosterol-free extracts derived fromw/t andcel (Tw 40) occur at –31 and –11 °C, respectively. The heats of transition (H) of these extracts were 1 and 13 cal/g, respectively. The addition of ergosterol (the predominant sterol inNeurospora) to the phospholipid extracts decreased the observed heats of transition, but did not alter the transition temperature. IntactNeurospora, whetherw/t orcel (Tw 40) did not manifest similar gel-liquid crystal phase transitions in the differential scanning calorimeter. However, an endothermic peak at approximately 30 °C was observed in intact cells and extracted phospholipids of bothw/t andcel (Tw 40) organisms. This peak was insensitive to the addition of ergosterol, had a low heat content (H1 cal/g), and was reversible.  相似文献

Summary Since the reported alterations of permeability of the blood-brain barrier by microwave radiation have implications for safety considerations in man, studies were conducted to replicate some of the initial investigations. No transfer of parenterally-administered fluorescein across the blood-brain barrier of rats after 30 min of 1.2-GHz radiation at power densities from 2–75 mW/cm2 was noted. Increased fluorescein uptake was seen only when the rats were made hyperthermic in a warm-air environment. Similarly, no increase of brain uptake of14C-mannitol using the Oldendorf dual isotope technique was seen as a result of exposure to pulsed 1.3-GHz radiation at peak power densities up to 20 mW/cm2, or in the continuous wave mode from 0.1–50 mW/cm2. An attempt to alter the permeability of the blood-brain barrier for serotonin with microwave radiation was unsuccessful. From these studies it would appear that the brain must be made hyperthermic for changes in permeability of the barrier induced by microwave radiation to occur.The research reported in this paper was conducted by personnel of the Radiation Sciences Division, USAF School of Aerospace Medicine, Brooks AFB, Tex. 78235. Summary In conventional two microelectrode experiments, acetylcholine had qualitatively the same effect as GABA and glutamate on membrane potential and input resistance of muscle fibres of the opener and intrinsic stomach muscles of crayfish (Austropotamobius torrentium). In patch-clamp experiments, acetylcholine occasionally elicited single channel openings in cell-attached patches on these muscles. If outside-out patches were excised and the Cl concentration was high on both sides of the membrane, acetylcholine at concentrations of 1 nM regularly elicited single channel currents. The amplitude of single channel currents depended strongly on the intracellular concentration of Cl. The reversal potential of the channel, determined after replacing intracellular K+ with Cs+, corresponded to the Nernst potential for Cl. The voltage dependence and the reversal potential of single channel current amplitudes elicited by ACh, glutamate and GABA were identical. The distribution of life times of openings (>1 ms) elicited by ACh and glutamate could be fitted by a single exponential with a time constant of about 2.5 ms, corresponding to the mean open time. ACh and glutamate applied to the same outside-out patch showed cross-desensitization, and thus ACh and glutamate activate the same channels. An excitatory, cationic ACh-activated channel could not be identified. Permeabilities of the chloride channel were calculated according to the Goldman-Hodgkin-Katz equation at different membrane potentials. Negative single channel current amplitudes (inward currents) could be fitted with a permeability of 2= 3.9×10–14 cm3s–1. For positive currents (outward) the channel had a permeability of 1= 1.4× 10–14 cm3s–1. The permeability of the channel declined from 16×10–14 cm3s–1 to 2.3×10–14 cm3s–1 if the intracellular Cl-concentration was raised from 6 to 257 mM. The activation elicited by acetylcholine was inhibited by extracellular Ca++. The mean current activated by ACh was reduced by a factor of 50 if the extracellular concentration of Ca++ was raised from 0.1 mM to the physiological concentration of 13.5 mM.  相似文献

In 2003, 50 game carcasses (ungulates) originating from one Austrian hunting ground were subject to visual examination for (fecal) contamination of the body cavities and microbiological testing of the body cavities in order to assess variations in microbial surface contamination in the season June–August compared to October–December. No carcass tested positive for the bacterial pathogens Salmonella or Listeria. Bacterial surface counts in October–December (median values: total aerobic count: 4.12 log10 colony-forming-units (cfu)/cm2; Enterobacteriaceae: 2.48 log10 cfu/cm2) were significantly lower than those in June–August (median values: total aerobic count: 5.65 log10 cfu/cm2; Enterobacteriaceae: 3.45 log10 cfu/cm2). The cooling regime (0.4 °C, 62% relative humidity) allowed no microbial growth for 96 h but was associated with weight loss of the carcasses. All carcasses had undergone a precooling phase of 8–12 h, with temperatures of 17.8±1.2 °C in the season June–August and 9.8±1.2 °C in October–December. This temperature difference was identified as the most probable effector for the observed seasonal variation. The results demonstrate the need for a continuous cool chain after evisceration of game carcasses.  相似文献

Summary Changing the temperature from 10–40 °C modifies the transmission at an established monosynaptic connection between the fast extensor tibiae (FETi) and flexor tibiae motor neurons in the metathoracic ganglion of the locustSchistocerca gregaria (Forskål). Striking changes occur to the shape of the spikes, to membrane resistance, to the synaptic delay, and to the evoked synaptic potentials.In the presynaptic FETi motor neuron, raising the temperature reduces the amplitude of an antidromic spike recorded in the soma by a factor of 10 (40 mV to 4 mV), reduces the time taken to reach peak amplitude by 5 (3.5 to 0.7 ms) and decreases the duration at half maximum amplitude by 0.5. The conduction velocity of the spike in the axon is increased by 50% from 10 °C to 40 °C. Orthodromic spikes are affected by temperature in a similar way to the antidromic spikes.The membrane resistance of both pre- and postsynaptic motor neurons falls as the temperature is raised. The membrane resistance of FETi falls by a factor of 4 (about 4 M at 10 °C to 1 M at 40 °C). A contributory component to this fall could be the increase in the frequency of synaptic potentials generated as a result of inputs from other neurons. No temperature dependence could be demonstrated on the voltage threshold relative to resting potential for evoking orthodromic spikes, but because the resistance changes, the current needed to achieve this voltage must be increased at higher temperatures.The latency measured from the peak of the spike in the soma of FETi to the start of the EPSP in the soma of a flexor motor neuron decreases by a factor of 20 (10 ms at 10 °C to 0.5 ms at 40 °C).In a postsynaptic flexor tibiae motor neuron, the amplitude of the evoked synaptic potential increases by a factor of 3.4 (5 mV to 17 mV), its duration at half maximum amplitude decreases by 3 (7 ms at 12 °C to 2.3 ms at 32 °C) and its rate of rise increases by 3. Surface phytoplankton assemblages were studied in January/February 1999 in the Crozet Basin (43°50S–45°20S; 61°E–64°30E) between the northern Polar Zone and the Agulhas Front. Cell concentrations increased several fold northwards from the SubAntarctic Zone (SAZ) and reached peak numbers (average 2×106 cells l–l ) in the central and western Subtropical Zone (STZ). The most spectacular increase in cell numbers occurred at the Subtropical Front (STF) and was attributed to dinoflagellates and diatoms. Nanoflagellates and picoplankton were dominant in the entire area (average 2.8×105–1.6×106 cells l–l). In the SAZ they were followed by coccolithophorids, dinoflagellates and diatoms. In the STZ coccolithophorids were often outnumbered by dinoflagellates. Diatoms were dominated by Pseudonitzschia delicatissima and were generally the least abundant algae, but reached peak densities of 1.2–4×105 cells l–l at, and north of the STF. Coccolithophorids contained mainly Emiliania huxleyi, but in the SAZ and STF Gephyrocapsa oceanica was a co-dominant species. Dinoflagellates were dominated by nano-sized species of Gymnodinium, Gyrodinium and Prorocentrum. The numbers of dinoflagellate and coccolithophorid species increased considerably in the convergence zone (STZ), which suggests their in-situ development. Heterotrophic dinoflagellates and ciliates were mainly present in the subtropics. Cell carbon biomass was attributed chiefly to auto- and heterotrophic dinoflagellates (av. 23–72 g C l–l; 68–87%), showing their important contribution to the carbon flow. Variations in cell concentrations across the fronts and water masses, and the distribution of major species were most likely controlled by the combined effect of such factors as nutrient renewal in the convergence zone, availability of iron, increased water-column stability at fronts, and high horizontal gradients in surface-water temperature.  相似文献

Isolate 1051 of Trichoderma harzianum, a mycoparasitic fungus, was found to impair development of the phytopathogen, Crinipellis perniciosa, in the field. This Trichoderma strain growing in liquid medium containing chitin produced substantial amounts of chitinases. The N-acetylglucosaminidase present in the culture-supernatant was purified to homogeneity by gel filtration and hydrophobic interaction chromatography, as demonstrated by SDS-PAGE analysis. The enzyme had a molecular mass of 36 kDa and hydrolyzed the synthetic substrate -nitrophenyl-N-acetylglucosaminide (NGlcNAc) with Michaelis–Menten kinetics. Maximal activities were determined at pH 4.0 and a temperature range of 50–60°C. K m and V max values for NGlcNAc hydrolysis were 8.06 moles ml–1 and 3.36 moles ml–1 min–1, respectively, at pH 6.0 and 37°C. The enzyme was very sensitive to Fe3+, Mn2+ and Co2+ ions, but less sensitive to Zn2+, Al3+, Cu2+ and Ca2+. Glucose at a final concentration of 1 mM inhibited 65% of the original activity of the purified enzyme. Determination of the product (reducing sugar) of hydrolysis of C. perniciosa mycelium and scanning electron microscopic analysis revealed that the N-acetylglucosaminidase hydrolyses the C. perniciosa cell wall.  相似文献

The expression of a methanol-soluble, heat-stable enterotoxin (STA) fused to the B subunit of the heat-labile enterotoxin (LTB) at 35°C or higher temperatures caused strains ofEscherichia coli deficient in lactose permease to behave on indicator media as Lac+; however, at 33°C or lower temperatures the original Lac phenotype of the host strains was maintained. The apparent phenotypic suppression oflacY was shown to be due to lysis of a fraction of the bacteria and the consequent release of active-galactosidase to the culture supernatant. After incubation at 37°C for 1 h, the cultures were committed to lyse. Plasmid and chromosomal mutants that do not show this phenotype were isolated by selecting Lac colonies at the unpermissive temperature. The mutations on the plasmids were localized in both the heat-stable and the heat-labile enterotoxin genes. Chromosomal mutants that show normal levels of-galactosidase and fused toxins have also been isolated.  相似文献

Summary The high membrane potential ofAcetabularia (E m=–170 mV) is due to an electrogenic pump in parallel with the passive diffusion system (E d=–80 mV) which could be studied separately in the cold, when the pump is blocked. Electrical measurements under normal conditions show that the pump pathway consists of its electromotive forceE p with two elementsP 1 andP 2 in series;P 2 is shunted by a large capacitance (C p=3 mF cm–2). The nonlinear current-voltage relationship ofP 1 (light- and temperature-sensitive) could be determined separately; it reflects the properties of a carrier-mediated electrogenic pump. The value ofE p (–190 mV) indicates a stoichiometry of 21 between electrogenically transported charges and ATP. The electrical energy, normally stored inC p, compares well with the metabolic energy, stored in the ATP pool. The nonlinear current-voltage relationship ofP 2 (attributed to phosphorylating reactions) is also sensitive to light and temperature and is responsible for the region of negative conductance of the overall current-voltage relationship. The power of the pump (1 W cm–2) amounts to some percent of the total energy turnover. The high Cl fluxes (1 nmol cm–2 sec–1) and the electrical properties of the plasmalemma are not as closely related as assumed previously. For kinetic reasons, a direct and specific Cl pathway between the vacuole and outside is postulated to exist.  相似文献

Young sporophytes of short-stipe ecotype ofEcklonia cavafrom a warmer locality (Tei, Kochi Pref., southern Japan) and those of long-stipe ecotype from a cooler locality (Nabeta, Shizuoka Pref., central Japan) were transplanted in 1995 to artificial reefs immersed at the habitat of long-stipe ecotype in Nabeta Bay, Shizuoka Pref., central Japan. The characteristics of photosynthesis and respiration of bladelets of the transplanted sporophytes of the two ecotypes were compared in winter and summer 1997; the results were assessed per unit area, per unit chlorophyllacontent and per unit dry weight. In photosynthesis-light curves at 10–29 °C, light saturation occurred at 200–400 mol photon m–2s–1in sporophytes from both Tei and Nabeta. The maximum photosynthetic rate (P max) at 10–29 °C and the light-saturation index (I k) at 25–29 °C in sporophytes from both localities were generally higher in winter than in summer.P maxat 25–29 °C (per unit area and chlorophylla) were higher in sporophytes from Tei than those from Nabeta in both seasons. The optimum temperature for photosynthesis was 25 °C in winter and 27 °C in summer at high light intensities of 100–400 mol photon m–2s–1. However, at lower light intensities of 12.5–50 mol photon m–2s–1, it was 20 °C in winter and 25–27 °C in summer for sporophytes from both locations. Dark respiration increased with temperature rise in the range of 10–29 °C in sporophytes from both locations in summer and winter. The sporophytes transplanted from Tei (warmer area) showed higher photosynthetic activities than those from Nabeta (cooler area) at warmer temperatures even under the same environmental conditions. This indicates that these physiological ecotypes have arisen from genetic differentiation.  相似文献

Summary Body temperature (T b), oxygen consumption , thermal conductance (C) and evaporative water loss (EWL) were measured at various air temperatures (T a) in two starlings which evolved in the tropics: a migratory species from a temperate climate,Sturnus vulgaris, and a resident, desert species,Onychognathus tristrami (Aves, Passeriformes, Sturnidae).AtT a's of 4–35°C both birds hadT b of 40.6°C. At 44°C,T b ofSturnus was 45.8°C and that ofOnychognathus 43.3°C.T a of 44°C was tolerated only byOnychognathus. The thermoneutral zone (TNZ) ofSturnus was in theT a range of 29.5°C–36.5°C, that ofOnychognathus 21.5–36.5°C. ofSturnus within its TNZ (BMR) was 2.37 ml O2 g–1 h–1, which is close to the expected BMR; that ofOnychognathus, 1.67 ml O2 g–1 h–1, is only 74% of the expected. AtT a'sNZ,C ofSturnus was twice as high as that ofOnychognathus and 1.68 times the expected value, whereasC ofOnychognathus was only 94% of the expected. At highT a'sOnychognathus had higherC thanSturnus. At either low or highT a's EWL ofSturnus was greater than ofOnychognathus.The responses shown bySturnus are typical of a tropical bird living in a moderate environment. Biofilm formation on metallic surfaces in marine and freshwater environments often precedes corrosion and other biofouling conditions. Attachment is mediated by such environmental factors as the presence of surface conditioning films, fluid dynamics, bulk-phase nutrient levels, and surface chemistry. In this study, we utilized a Fowler Cell Adhesion Measurement Module to demonstrate that the changes in cellular concentration and composition of a monoculture ofPseudomonas atlantica biofilms on stainless steel were a function of the applied shear force. At shear forces in the range of 3–10 dynes cm–2 (1.0 liter min–1), attachment as measured by acridine orange direct microscopic counts was greatest at the higher shear forces.14C-Acetate uptake activity on the stainless steel surfaces increased with shear stress. Acetate incorporation ranged from 1×10–5 to 19×10–5 mol cm–2 between 0.15 and 30 dynes cm–2 for 30 min uptake periods. On a per cell basis, however, activity decreased with shear, indicating a shift in metabolism. Fourier transform infrared spectroscopy revealed that protein and carbohydrate concentrations also increased with the applied shear. Increased biofilm CN ratios and total fatty acids were associated with the higher shear stresses. Neither radius of interaction nor biofilm age appeared to significantly influence the relationship between fluid shear and attachment and cellular composition ofP. atlantica biofilms in the range of 1–10 dynes cm–2.  相似文献

Inactivation of urease (25 nM) in aqueous solutions (pH 5.0–6.0) treated with low-frequency ultrasound (LFUS; 27 kHz, 60 W/cm2, 36–56°C) or high-frequency ultrasound (HFUS; 2.64 MHz, 1 W/cm2, 36 or 56°C) has been characterized quantitatively, using first-order rate constants: k in, total inactivation; k in *, thermal inactivation; and k in(us), ultrasonic inactivation. Within the range from 1 nM to 10 M, propyl gallate (PG) decreases by approximately threefold the rate of LFUS-induced inactivation of urease (56°C), whereas resorcinol poly-2-disulfide stops this process at 1 nM or higher concentrations. PG completely inhibits HFUS-induced inactivation of urease at 1 nM (36°C) or 10 nM (56°C). At 0.2–1.0 M, human serum albumin (HSA) increases the resistance of urease treated with HFUS to temperature- and cavitation-induced inactivation. Complexes of gallic acid polydisulfide (GAPDS) with HSA (GAPDS–HSA), formed by conjugation of 1.0 nM GAPDS with 0.33 nM HSA, prevent HFUS-induced urease inactivation (56°C).  相似文献

Summary The flux from the biomedical channel at TRIUMF increases with increasing channel momentum, while the contaminating electron flux decreases. Since the electrons appear to result from conversion of the high energy-rays produced by 0 decay in the production target, the electron contamination can be reduced further by target configurations which minimize gamma conversion.The attenuation of beams by in-flight interactions was found to decrease from an initial value of 1.67 ± 0.02 % per g/cm2 at zero depth to 1.48 ± 0.02 % per g/cm2 near the stopping peak.The inactivation of cultured CHO cells by an extended-peak dose distribution has been measured using the gel technique. The survival data have been fitted by a model which characterizes the physical quality of the dose profile by means of measured star densities. This model provides a convenient method of analysis for large sets of survival data and may be useful for prediction of the biological effect of new dose distributions.The RBE value for 50% survival measured at the centre of a 7 cm extended peak was found to be approximately 1.4, in reasonable agreement with recent values obtained at LAMPF and SIN.  相似文献

Summary The decreasing effect of -adrenergic blockade on skin resistance to vapor diffusion and the onset of cutaneous water evaporation in the pigeon (Columba livia) was investigated. Oral administration of 1, 2.3 and 5 mg propranolol to pigeons (268±53 g) initiated intensive trans-cutaneous water evaporation (CWE) up to 29.1 mg H2O·cm–2·h–1 in resting birds at 30°C air temperature (Ta), but had only a slight effect on CWE of birds exposed to 50 °C Ta.After 7 h of effective -adrenergic blockade (oral administration of 5 mg propranolol), skin and body temperature stabilized at 39.0±0.5 °C and 41.0±0.7 °C, compared to 40.2±0.8 °C and 41.9±0.6 °C in the control group, respectively. A slight hypothermia was accompanied by feather fluffing.Intradermal injection of 0.001, 0.01 and 0.12 mg propranolol also caused intensive CWE. Local -adrenergic blockade in relatively low blocker doses (0.001 and 0.01 mg propranolol) decreased skin resistance from a high value of 44.5 s·cm–1 to about 6.0 s·cm–1, and caused a sharp increase in CWE from a control value of about 4 to a high of 26.4 mg H2O·cm–2·h–1 during the first two hours of exposure to 30°C Ta.The possible role of -adrenergic blockade in regulation of trans-cutaneous water evaporation of latent heat dissipation is discussed.  相似文献

Summary Respiratory gas exchange and blood respiratory properties have been studied in the East-African tree frogChiromantis petersi. This frog is unusually xerophilous, occupies dry habitats and prefers body temperatures near 40°C and direct solar exposure. Total O2 uptake was low at 81 l O2·g–1·h–1±19.0 (SD) at 25°C increasing to 253.5 l O2·g–1·h–1±94.8 (SD) at 40°C giving aQ 10 value of 2.1. Skin O2 uptake at 25°C was 38.5% of total. The gas exchange ratio was 0.71 for whole body gas exchange, 0.61 for the lungs and 1.02 for the skin at 25°C.Blood O2 affinity was low with aP 50 of 47.5 mmHg at 25°C and pH 7.65. Then H-value at 25°C increased from 2.7 aroundP 50 to 5.0 at O2 saturations exceeding 70–80%. Surprisingly, blood O2 affinity was nearly insensitive to temperature expressed by a H value of ±1.0 kcal·mole between 25 and 40°C.The adaptive significance of the low O2 affinity, the increase ofn H with O2 saturation and the temperature insensitive O2-Hb binding is discussed in relation to the high and fluctuating body temperatures ofChiromantis.  相似文献

Summary The biological effects of microwaves in the hyperfrequency range, 9.4 GHz, 17 GHz, and 70–75 GHz were investigated in bacteria and yeast. At power densities below 60 mW/cm2 and SAR values not exceeding 28 mW/g no significant effects on survival of repair competent and deficient strains were observed inEscherichia coli andSaccharomyces cerevisiae. In addition, microwaves of 17 GHz did not induce mutations inE. coli B/r WP2trp uvr above the spontaneous level, and the induction of nuclear reversions, cytoplasmic petite mutations and mitotic recombination as well as the efficiency of sporulation was not affected in yeast.  相似文献

Summary Continuous cultures ofSulfolobus BC oxidising tetrathionate were found not to be tetrathionate limited, but molybdenum limited. Incorporation of only 1.3M Na2MoO4 into the medium of autotrophically grown, continuous cultures ofSulfolobus BC oxidising 15mM tetrathionate at a dilution rate of 0.029h–1 increased the steady state biomass from absorbance (660nm) 0.160 to 0.315. The biomass yield gave a corresponding increase from 5.93±0.84 to 11.52±0.68 g dry weight mol–1 tetrathionate oxidised.  相似文献
