Molecular biology, combined with Mendelian and quantitative genetics in quantitative trait locus (QTL) mapping and marker-assisted selection (MAS), provides powerful new tools to facilitate efficient genetic manipulation by plant breeders of complex traits such as drought tolerance and phosphorus (P) acquisition ability. This paper examines current opportunities for genetically manipulating the ability of crop plants to more efficiently acquire (i.e. access and take up) essential soil nutrients, using as examples P and several of the crops in the genetic improvement mandate of the International Crops Research Institute for the Semi-Arid Tropics (ICRISAT) – chickpea (Cicer arietinum L.), groundnut (Arachis hypogaea L.), pearl millet [Pennisetum glaucum (L.) R. Br.], pigeonpea [Cajanus cajan(L.) Millsp.], and sorghum [Sorghum bicolor (L.) Moench]. It is concluded that for at least some of these important, but often academically and economically orphaned tropical food, forage and feed-grain crops, the genetic variation and molecular tools that we will need already exist or can be expected to become available in the very near future. With appropriate, targeted research, these tools can permit empirical exploration of the potential for marker-facilitated mapping and manipulation of major genes that can contribute to enhanced ability of these crops to acquire P from sources with limited availability. With these tools, delivery of new versions of currently popular high-yielding, high quality, disease resistant crop cultivars, having genetically improved ability to acquire P currently in soils but unavailable for crop growth, could take as little as five to seven years. Sustainable use of such improved cultivars would require their utilization as components of integrated soil fertility management systems.  相似文献

This paper reports the effects under green-house and field conditions of four arbuscular-mycorrhizal (AM) fungi on the growth and nutrition of some tropical and subtropical crops of economic importance in the Canary Islands (Spain) whose mycorrhizal dependency was unknown. Avocado (Persea americana Mill.), papaya (Carica papaya L.), pineapple (Ananas comosus (Merr.) L.) and banana (Musa acuminata Colla) were highly responsive to the mycorrhizal condition when the AM inoculum was Glomus sp. endophytes. Under our experimental conditions, Glomus fasciculatum was the most effective fungus at improving plantlet growth and nutrition. Acaulospora sp. was ineffective and Scutellospora was only effective with banana. The data clearly show the advantage of mycorrhization during the first phase of development of the plant system.  相似文献

Diapause and cold tolerance are essential for temperate insects to pass the winter, with the mechanisms controlling these two traits varying considerably among insects. In the present study, diapause and cold tolerance are compared among three Leptopilina species: Leptopilina japonica Novkovi? & Kimura, Leptopilina victoriae Nordlander and Leptopilina ryukyuensis Novkovi? & Kimura, all larval parasitoids of frugivorous drosophilid flies, with the aim of understanding their climatic adaptations. The first species is divided into the temperate (Leptopilina japonica japonica) and subtropical subspecies (Leptopilina japonica formosana), and the latter two species are distributed in the tropical and subtropical regions. The temperate subspecies of L. japonica enters prepupal diapause at low temperatures (15 or 18 °C), irrespective of photoperiod, and some individuals enter diapause when exposed to 0 °C for 1 or 2 day(s) or when placed at low humidity. Leptopilina victoriae also shows signs of diapause initiation at 15 °C, although L. ryukyuensis and L. j. formosana from the subtropical regions do not. Preimaginal viability at low temperature (13, 14 or 15 °C) is usually lower in L. victoriae from the tropical regions compared with L. japonica or L. ryukyuensis from the temperate or subtropical regions. Diapausing prepupae of the temperate subspecies appear to be cold tolerant. However, the cold tolerance of nondiapausing prepupae, pupae and adult females varies little among the tropical, subtropical and temperate species or subspecies, and adult males of the temperate subspecies of L. japonica are less cold tolerant than those of the tropical or subtropical species or subspecies. Cold tolerance may be unnecessary, except for diapausing individuals of the temperate species, because nondiapausing individuals appear in warmer seasons.  相似文献

The molecular genetic mechanisms underlying fruit size remain poorly understood in perennial crops, despite size being an important agronomic trait. Here we show that the expression level of a microRNA gene (miRNA172) influences fruit size in apple. A transposon insertional allele of miRNA172 showing reduced expression associates with large fruit in an apple breeding population, whereas over‐expression of miRNA172 in transgenic apple significantly reduces fruit size. The transposon insertional allele was found to be co‐located with a major fruit size quantitative trait locus, fixed in cultivated apples and their wild progenitor species with relatively large fruit. This finding supports the view that the selection for large size in apple fruit was initiated prior to apple domestication, likely by large mammals, before being subsequently strengthened by humans, and also helps to explain why signatures of genetic bottlenecks and selective sweeps are normally weaker in perennial crops than in annual crops.  相似文献

Fruit rot of chillies (Capsicum annuum L.), caused by Colletotrichum capsici under tropical and subtropical conditions, results in qualitative and quantitative yield losses. Based on variation in cultural and morphological traits of C. capsici populations, 37 isolates were categorized into five groups designated, respectively, as Cc‐I, Cc‐II, Cc‐III, Cc‐IV and Cc‐V. In culture, most of the isolates produced cottony, fluffy or suppressed colonies. However, no significant differences were noticed in shape and size of conidia. The reaction of the 37 isolates on an indigenously developed differential set of Capsicum cultivars indicated the existence of different virulences in Himachal Pradesh (HP) chilli populations. Fifteen pathotypes of the pathogen were characterized from various chilli‐growing regions of HP. Pathotype CCP‐1 was most virulent and attacked all the differential cultivars. The genetic relationship between five morphological groups recognized within C. capsici was investigated using random amplified polymorphic DNA (RAPD) analysis. Molecular polymorphism generated by RAPD confirmed the variation in virulences of C. capsici and different isolates were grouped into five clusters. However, four isolates (Cc‐5, Cc‐33, Cc‐29 and Cc‐37) exhibited identical RAPD haplotypes. The pathological and RAPD grouping of isolates suggested no correlation among the test isolates.  相似文献

Banana is one of the most important subtropical crops. The genetic system, however, is relatively unknown and is complicated by specific interhybridization, heterozygosity, and polyploidy, which are common in most clones. These factors make identification of closely related banana cultivars difficult, particularly when sterile. Amplified fragment length polymorphism (AFLP) analysis using eight primer combinations was carried out on 16 banana cultivars. Results showed that AFLP could be used to distinguish the different cultivars by their unique banding patterns. Unique AFLP molecular markers were detected for 12 banana cultivars, which can be used to develop specific probes for identification purposes. The cluster analysis also revealed the need for a link between genotype studies using molecular techniques and the current system of classification of Musa cultivars based purely on morphological traits.  相似文献

Populations of many perennial plants intermittently produce large seed crops—a phenomenon referred to as mast seeding or masting. Masting may be a response to spatially correlated environmental conditions (the Moran effect), an adaptive reproductive strategy reflecting economies of scale, or a consequence of the internal resource budgets of individual plants. Fruit production by endozoochorous plants representing eight genera varied synchronously over much of central Ontario, Canada, 1998–2009. We tested for effects of weather and prior reproduction on fruit production by comparing AICc values among regression models fit to time series of fruit production scores and partitioning contributions by different predictors to multiple R 2 into independent and joint contributions. Fruit production by mountain ash (Sorbus spp.), juneberry (Amelanchier spp.), dogwoods (Cornus spp.), nannyberry (Viburnum lentago), and possibly cherries (Prunus spp.) was inversely related to production in the previous year. These effects were independent of weather conditions, suggesting that intrinsic factors such as internal resource budgets or an adaptive strategy of variable reproductive output influenced fruit production. To our knowledge, this is the first evidence of masting in members of the genera Cornus, Viburnum, and Amelanchier, and in members of Prunus and Sorbus in North America. All species produced fewer fruits when weather conditions were dry, so the Moran effect could have synchronized fruit production both within and among species. Patterns and causes of variation in berry crops have implications for ecosystem dynamics, particularly in boreal and subArctic environments where berry crops are important wildlife foods.  相似文献

An improved protocol for genetic transformation of juvenile explants of Carrizo (Citrus sinensis Osb. × Poncirus trifoliata L. Raf.), Duncan (Citrus paradisi Macf.), Hamlin (Citrus sinensis (L.) Osbeck) and Mexican Lime (Citrus aurantifolia Swingle) cultivars using a vector containing a bifunctional egfp-nptII fusion gene is described. Several parameters were investigated to optimize genetic transformation of these four cultivars. It was determined that a short preincubation in hormone rich liquid medium and subculture of Agrobacterium for 3 h in YEP medium containing 100 μM acetosyringone were required for improvement of transformation efficiency. Co-cultivation duration as well as addition of acetosyringone to co-cultivation medium also played an important role in transformation efficiency as did OD600 value of the Agrobacterium suspension used for transformation. We regenerated numerous EGFP expressing transgenic lines from all four cultivars. Based on these results, we conclude that genetic transformation of citrus is cultivar specific and optimization of conditions for maximum transgenic production are required for each individual cultivar.  相似文献

Pollination ecology of many crops is not fully known, especially in tropical and subtropical regions. Non-bee pollinators may contribute substantially to crop yield, even if they do not receive much attention. Although moth pollination has fascinated ecologists and evolutionary biologists since Darwin, crop pollination by moths has not been well investigated and experimentally examined. Hence, we explored the pollination ecology of four cucurbit crops with crepuscular or nocturnal flowers. Lagenaria siceraria (Molina) Standl., Luffa acutangula (L.) Roxb., Trichosanthes anguina L., and Trichosanthes kirilowii Maxim. all display floral traits suggestive of moth pollination, such as opening around dusk or in evenings and secreting ample dilute nectar. We demonstrated that these crops’ flowers attracted a wide range of moth species, especially hawkmoths. The assemblage of flower-visiting moths varied according to location and season. Pollination treatments and pollen load analysis confirmed the pollination of the four crops by moths, especially hawkmoths. Our results provide evidence for the value as wild pollinators for the four crops, for which domesticated bees cannot provide reliable pollination services in practice. This study lends support to the proposal to pay more attention to the value of non-bee pollinators and to leave some areas unexploited in rural landscapes for the conservation of wild pollinators, including moths.  相似文献

As the human population has increased, so too has the demand for biotically pollinated crops. Bees (Apoidea) are essential for pollen transfer and fruit production in many crops, and their visit patterns can be influenced by floral morphology. Here, we considered the role of floral morphology on visit rates and behaviour of managed honey bees (Apis mellifera) and wild bumble bees (genus Bombus), for four highbush blueberry cultivars (Vaccinium corymbosum L.). We measured five floral traits for each cultivar, finding significant variation among cultivars. Corolla throat diameter may be the main morphological determinant of visit rates of honey bees, which is significantly higher on the wider flowers of cv. ‘Duke’ than on ‘Bluecrop’ or ‘Draper’. Honey bees also visited cv. ‘Duke’ legitimately but were frequent nectar robbers on the long, narrow flowers of cv. ‘Bluecrop’. Bumble bees were infrequent (and absent on cv. ‘Draper’) but all observed visits were legitimate. Crop yield was highest for the cultivar with the highest combined (honey bee + bumble bee) visit rate, suggesting that aspects of floral morphology that affect pollinator visit patterns should be considered in crop breeding initiatives.  相似文献

In Costa Rica, dioecious Carica papaya has been observed growing in disturbed areas and within secondary lowland forests. Such populations can serve as a reservoir of genetic and morphological diversity for this tropical fruit crop. We quantify the levels and patterns of the diversity of naturally occurring populations of C. papaya and address the demographic history of these populations. We measured 29 vegetative and reproductive morphological traits in situ from 252 plants and found significant heterogeneity among regional populations in the majority of these traits. Significant variation was found among regional populations with respect to fruit size and shape, with plants in the Nicoya Peninsula possessing smaller, less fleshy fruit, a characteristic of previously described wild populations of papaya. We then assessed the levels and patterns of genetic diversity in 164 plants from natural populations and 20 cultivars. Natural populations exhibit a deficiency of heterozygotes; however, this is much more pronounced within the cultivars. Although there is little genetic differentiation among natural populations, we did find evidence of cryptic genetic population structure. Analyses of population demography indicate that these natural populations have undergone a recent genetic bottleneck, followed by recent population expansion, possibly promoted by the transformation of the Costa Rican landscape for agricultural use. Lonicera caeruleabrix is a perennial shrub native to North America, Europe and Asia. It produces dark blue berries known as honeyberries or haskap berries which are produced commercially in several territories including Canada, Japan, Russia and Poland. Plants are suited to UK environments, but it is yet to be widely commercially developed in the UK. In the present work, quality and nutritional traits of six honeyberry cultivars grown in Scotland were compared with other commonly grown berry fruits (strawberry, raspberry, blueberry, blackberry, blackcurrant) to aid the identification of environmentally stable, high-quality honeyberry cultivars suitable for UK cultivation. Differences were observed in fruit quality variables (soluble solids, titratable acidity and Brix/acid ratios) between honeyberry cultivars. Three of six cultivars examined exhibited notable variation in soluble solids dependent on harvest year with ‘Aurora’ and ‘Strawberry Sensation’ having consistently high oBrix values. Titratable acidity exhibited cultivar differences and there was limited variation over harvest years. ‘Aurora’ exhibited consistently high oBrix/titratable acidity ratio reflected by high glucose and fructose content. Honeyberry fruit had good nutritional profile relative to other soft fruits with higher polyphenol and anthocyanin content than strawberry, blueberry, blackberry and raspberry, manifested in greater antioxidant capacity. The major anthocyanins in aqueous honeyberry fruit extracts were cyanidin, pelargonidin and peonidin glycosides. These findings indicate that L. caerulea represents a crop suitable for UK cultivation capable of producing high quality fruit with a valuable nutritional profile relative to other soft fruits. Cultivars exhibit significant differences in fruit quality and nutritional profile as well as harvest consistency and growers should consider this when establishing new plantations.  相似文献

Strawberry (Fragaria × ananassa Duch.), a member of the Rosaceae family, is one of the most important fruit crops cultivated worldwide. Strawberry is unique within the Rosaceae because it is a rapidly growing herbaceous perennial with a small genome, short reproductive cycle, and facile vegetative and generative propagation for genetic transformation. For these reasons, strawberry has been recognized as excellent germplasm for genetic and molecular studies for the Rosaceae family. Although traditional breeding methods have achieved steady improvement in agronomic traits, the lack of useful economic characters still remains a major challenge. Genetic transformation has opened a new era for greater creativity in strawberry breeding and germplasm by offering an effective method for creating new varieties that selectively targets a specific interested gene or a few heterologous traits. Enormous advances have been made in strawberry genetic transformation since the first transgenic strawberry plant was obtained in 1990. This paper reviews recent progress in genetic transformation of strawberry on increasing resistance to viruses, fungi, insects, herbicides, stress, and achieving better quality. Problems and prospects for future applications of genetic transformation in strawberry are also discussed.  相似文献

【背景】芒果蛎蚧属于盾蚧类昆虫,食性杂,分布广,是危害热带和亚热带水果、蔬菜和园林植物的重要害虫,主要随水果、苗木和交通工具等介质进行远距离传播。该虫被列入中华人民共和国进境植物检疫性有害生物名录。惠州口岸于2014年12月从芬兰进境的货物中截获该虫,属于我国口岸首次截获。【方法】在收集整理芒果蛎蚧的生物学、地理学等信息的基础上,介绍了芒果蛎蚧的主要形态特征、地理分布、寄主植物和生物学特性,并分析了该有害生物入侵我国的风险。【结果】芒果蛎蚧雌成虫为椭圆形,长约1.2 mm,触角每侧有1根长毛,每侧前气门有3个盘状腺,形态特征与近似种Lepidosaphes camelliae和Lepidosaphes pallida非常相似。风险分析表明,我国广东、广西、海南、云南、福建、台湾以及四川等省非常适合芒果蛎蚧生存和危害。芒果蛎蚧入侵我国会给芒果等果树造成危害,给相关产业带来损失,影响从业人员收入,还影响生态环境,可能导致生态灾难。【结论与意义】芒果蛎蚧有入侵我国的可能,加强检疫是防范该虫入侵的主要手段,生物防治是治理该虫的重要措施。  相似文献

Greengram [Vigna radiata (L.) Wilczek], also known as mung bean, widely cultivated in a large number of countries, is an important pulse crop of Asia and is considered one of the ancestral species of the genus Vigna. Since yields of greengram have remained low across subtropical and tropical Asia, it is important to estimate genetic diversity in existing cultivars in order to see if the lack of genetic variability might be a constraining factor. In this study, 32 Indian cultivars of greengram were subjected to random amplified polymorphic DNA (RAPD) analysis using 21 decamer primers. A total of 267 amplification products were formed at an average of 12.71 per primer with an overall polymorphism of 64%. The extent of polymorphism was moderate to low. Jaccard similarity coefficient values ranged from 0.65 to 0.92. The cluster analysis resulted in mainly three clusters revealing greater homology between cultivars released from the same source. The results of principal components analysis also substantiated this conclusion. The close genetic similarity between the cultivars could be explained due to the high degree of commonness in their pedigrees. The narrow genetic base of the greengram cultivars revealed in the present analysis emphasises the need to exploit the large germplasm collections having diverse morphoagronomic traits in cultivar improvement programs. Brevicoryne brassicae (L), Lipaphis erysimi (Kalt) and Myzus persicae (Sultzer) (Homoptera: Aphididae) form the aphid complex that causes great losses in Brassicaceae in tropical and subtropical regions. Knowledge of their population dynamics is important for the development of integrated pest management programmes. This study aimed to investigate the effects of cabbage phenology, climatic factors and natural enemies populations on the dynamics of these organisms, and the factors regulating their predators’ occurrence. The densities of aphids and their natural enemies and the climate were monitored for two years in 16 cabbage crops. The highest densities of the aphids occurred during periods of relative humidity (RH) drop, a condition that affects them positively. Regarding the predators, the factors affecting their abundance varied but RH was positively related to most of them. This study provides relevant insights into the factors that regulate the aphids in cabbage and for the decision‐making process of control of these severe pests.  相似文献

The ability to transform chloroplasts in multiple species is important for improving agricultural traits. Chloroplast transformation of alfalfa (Medicago sativa L.), a useful forage plant with high market value, was achieved using a vector carrying aadA and gfp genes being introduced into the chloroplasts of alfalfa via particle bombardment using leaves and calli as explants. Resistant somatic embryos were generated and developed into plantlets from explants. The transformation efficiency was 1.3% for callus explants and 2.7% for leaf explants. PCR and Southern blotting analyses revealed that the foreign genes were integrated into the transformed chloroplast genome. The occurrence of GFP was further confirmed by fluorescence microscopy. Expression of foreign genes in alfalfa chloroplasts is therefore possible, and provides a novel means for genetic improvement of agronomically important traits and production of value-added proteins.  相似文献

Anaerobic conditions developing under ice cover affect winter survival and spring regrowth of economically important perennial crops. Our objective was to assess interspecific differences in the resistance to anaerobic conditions at low temperature, and to relate those differences to plant metabolism. Four perennial forage species, alfalfa (Medicago sativa L.), red clover (Trifolium pratense L.), timothy (Phleum pratense L.) and orchardgrass (Dactylis glomerata L.), were subjected to a progressively developing anoxic stress by enclosing potted plants in gas‐tight bags in late autumn and exposing them to simulated winter conditions in an unheated greenhouse. Near‐anaerobic conditions were reached after 60 d of enclosure for orchardgrass, alfalfa and red clover, and after 80 d for timothy. The sensitivity of the species to anaerobic conditions, based on plant regrowth, was: red clover and orchardgrass > alfalfa > timothy. The concentration of ethanol increased in response to oxygen deprivation, and reached the highest value in the sensitive red clover, whereas its concentration was the lowest in timothy. The expression of the alcohol dehydrogenase (ADH) gene was markedly lower in timothy than in the other three species for which the expression was equivalent. We conclude that the greater resistance of timothy to anaerobic conditions at low temperature is associated with a slower glycolytic metabolism.  相似文献

Black raspberry (Rubus occidentalis) is an important specialty fruit crop in the US Pacific Northwest that can hybridize with the globally commercialized red raspberry (R. idaeus). Here we report a 243 Mb draft genome of black raspberry that will serve as a useful reference for the Rosaceae and Rubus fruit crops (raspberry, blackberry, and their hybrids). The black raspberry genome is largely collinear to the diploid woodland strawberry (Fragaria vesca) with a conserved karyotype and few notable structural rearrangements. Centromeric satellite repeats are widely dispersed across the black raspberry genome, in contrast to the tight association with the centromere observed in most plants. Among the 28 005 predicted protein‐coding genes, we identified 290 very recent small‐scale gene duplicates enriched for sugar metabolism, fruit development, and anthocyanin related genes which may be related to key agronomic traits during black raspberry domestication. This contrasts patterns of recent duplications in the wild woodland strawberry F. vesca, which show no patterns of enrichment, suggesting gene duplications contributed to domestication traits. Expression profiles from a fruit ripening series and roots exposed to Verticillium dahliae shed insight into fruit development and disease response, respectively. The resources presented here will expedite the development of improved black and red raspberry, blackberry and other Rubus cultivars.  相似文献

Domestication Syndrome in Caimito ( Chrysophyllum cainito L.): Fruit and Seed Characteristics: The process of domestication is understudied and poorly known for many tropical fruit tree crops. The star apple or caimito tree (Chrysophyllum cainito L., Sapotaceae) is cultivated throughout the New World tropics for its edible fruits. We studied this species in central Panama, where it grows wild in tropical moist forests and is also commonly cultivated in backyard gardens. Using fruits collected over two harvest seasons, we tested the hypothesis that cultivated individuals of C. cainito show distinctive fruit and seed characteristics associated with domestication relative to wild types. We found that cultivated fruits were significantly and substantially larger and allocated more to pulp and less to exocarp than wild fruits. The pulp of cultivated fruits was less acidic; also, the pulp had lower concentrations of phenolics and higher concentrations of sugar. The seeds were larger and more numerous and were less defended with phenolics in cultivated than in wild fruits. Discriminant Analysis showed that, among the many significant differences, fruit size and sugar concentration drove the great majority of the variance distinguishing wild from cultivated classes. Variance of pulp phenolics among individuals was significantly higher among wild trees than among cultivated trees, while variance of fruit mass and seed number was significantly higher among cultivated trees. Most traits showed strong correlations between years. Overall, we found a clear signature of a domestication syndrome in the fruits of cultivated caimito in Panama.  相似文献
