放免法测定自发性高血压大鼠(SHR)血浆及心肌血管紧张素Ⅱ(AngⅡ)含量,凝胶内磷酸化法测定心肌丝裂素活化蛋白激酶活性(MAPK),以心脏重/体重表示心肌肥厚程度。结果表明:与4个月的WKY大鼠比较,4个月的SHR血浆和心肌组织AngⅡ及心肌MAPK活性分别增加了218.6%、101.2%和107.0%,心肌肥大程度严重,其中MAPK活性与心肌肥大程度呈明显正相关。提示4个月SHR心肌肥厚可能是  相似文献

目的: 研究有氧运动训练对自发性高血压大鼠(SHR)肾脏纤维化影响, 探讨有氧运动对高血压肾损害的保护作用。方法: 8周龄雄性SHR和同龄Wistar京都大鼠(WKY)随机分为4组(n=6):安静WKY对照组(WKY-S)、安静SHR对照组(SHR-S)、低强度运动组(SHR-L)和中强度运动组(SHR-M)。SHR-L组、SHR-M组分别以14 m/min(最大有氧速度的35%)、20 m/min(最大有氧速度的50%)在0°坡度的运动跑步机上跑步,共运动14周,每周5次,每次60 min,WKY-S和SHR-S组安静饲养。14周后,运动训练结束72 h后检测大鼠血压;之后取血和肾脏检测血清肌酐SCr和尿素氮BUN含量,苏木精与伊红(HE)染色观察肾组织形态,Masson染色观察肾组织胶原沉积情况,计算肾脏胶原容积分数(CVF),检测肾脏 AngⅡ、AT1R、TGF-β、α-SMA、CTGF蛋白表达。结果: 与WKY-S组相比,SHR-S组的血压和血清SCr、BUN含量、肾脏CVF水平和AngⅡ、AT1R、TGF-β、α-SMA、CTGF蛋白表达均明显升高(P<0.05);与SHR-S组相比,SHR-L组和SHR-M组血压和血清SCr、BUN含量、肾脏CVF水平和AngⅡ、AT1R、TGF-β、α-SMA、CTGF蛋白表达均明显下降(P<0.05)且SHR-M组下降趋势更明显(P<0.05)。结论: 有氧运动可通过抑制肾脏AngⅡ-AT1R-TGF-β通路,改善自发性高血压大鼠的肾纤维化与肾功能。  相似文献

目的观察不同年龄自发性高血压大鼠(SHR)心脏AT2R的表达水平及心肌胶原含量,探讨AT2R在高血压发生、发展过程中的作用。方法 1月龄组(S1)、2月龄组(S2)、3月龄组(S3)、6月龄组(S6)和9月龄组(S9)雄性SHR共五组,每组各6只,各组均有相应月龄的Wistar-Kyoto大鼠(WKY)作对照。采用RBP-I型大鼠血压心率测定仪测量大鼠动脉收缩压(SBP);放免法(RIA)测定血浆血管紧张素Ⅱ(AngⅡ);免疫组化染色结合计算机图像分析方法测定心脏AT2R的表达水平,天狼星红胶原染色大鼠的心脏切片。结果 1.SHR SBP随着月龄的增加呈持续上升(P〈0.05),SHR的SBP均高于相应配对的WKY组(P〈0.05)。2.一个月后SHR血浆AngⅡ浓度均高于S1(P〈0.05),一个月后SHR血浆AngⅡ浓度均高于相应配对的WKY组(P〈0.05)。3.SHR心脏AT2R免疫染色阳性面积比随着月份的增加而降低,SHR心脏AT2R免疫染色阳性面积比均低于相应配对的WKY组(P﹤0.05)4.SHR心肌中的胶原含量随着月龄的增加而增加。结论 SHR心脏AT2R表达水平比WKY低,并随着年龄的增加而降低。SHR心肌中的胶原含量随着月龄的增加而增加,而WKY无类似趋势。  相似文献

为了探索血小板源生长因子 (platelet derivedgrowthfactor,PDGF)受体介导的信号转导在自发性高血压大鼠(spontaneouslyhypertensiverats,SHR)心肌肥大中的作用 ,实验采用Westernblot法检测SHR及其对照WKY大鼠心肌PDGF受体β和细胞外信号调节激酶 (extracellularsignal regulatedkinase1/ 2 ,ERK 1/ 2 )的蛋白表达和ERK 1/ 2磷酸化水平的变化。结果显示 :4周龄SHR的收缩压、舒张压、±dp/dtmax和心肌肥大指数与同龄WKY大鼠相比均无明显差异 ,而 12周龄SHR上述指标与同龄WKY大鼠相比均明显升高 ,表明 12周SHR已发生高血压 ,心脏收缩功能代偿性增强 ,并出现心肌肥大。 4周龄SHR心肌PDGF受体 β和ERK1/ 2的磷酸化水平以及ERK 1/ 2的蛋白表达水平与同龄对照相比均无明显变化 ,12周时SHR心肌PDGF受体 β的蛋白表达较同龄WKY增加 32 77% (P <          0 0 5 ) ,PDGF受体介导的信号转导通路的下游信号分子ERK 1/ 2的磷酸化水平较同龄WKY升高 19 6 % (P =0 0 1) ,表明ERK 1/ 2的活化增加 ,但ERK 1/ 2的蛋白表达水平尚无变化。为进一步明确PDGF受体 β在心肌细胞生长中的作用及其与ERK 1/ 2活性的关系 ,采用PDGF BB刺激培养的乳鼠心肌细胞 ,发现 [3 H]亮氨酸掺入量明显增加 ,ERK 1/ 2的磷酸化水平明  相似文献

为了明确开搏通对运动性心肌肥大的作用。本实验采用游泳训练引起的大鼠心肌肥大模型,应用放射免疫及生化等方法对心肌肥大及心脏肾素-血管紧张素系统(R-A-S)的变化进行了初步研究。观察到游泳五周时,大鼠心重/体重比值(H/Bwt)显著升高,同时心肌血管紧张素Ⅱ(AngⅡ)及心肌血管紧张素转换酶(ACE)也明显升高。而服用开搏通的游泳大鼠心肌AngⅡ,ACE较单纯游泳组低,但心重/体重比值却升高。上述结果提示开搏通能抑制运动所致心肌AngⅡ升高,但不能防止运动引起的心肌肥大。  相似文献

Renovascular hypertension was induced in rats by left renal artery constriction. Force-velocity relation, end systolic tension-length relation (ESTLR) and responses to high extracellular calcium were investigated in hypertrophied myocardium with 4-week hypertension. The results showed that: (1) The myocardial hypertrophy was accompanied by increased peak active tension, decreased maximal velocity of shortening and prolonged contraction duration (P less than 0.01). (2) The ESTLR in hypertrophied myocardium was similar to that in the control, fitted well by an exponential curve and did not show significant alterations in all its regression parameters (P greater than 0.05). (3) No significant difference about the responses to high extracellular calcium (4 mmol/L) was observed between the control and the hypertrophied myocardium (P greater than 0.05). It is concluded that the mechanical properties of hypertrophied myocardium were characterized by a dissociation between force development and velocity of shortening and possibly these contractile abnormalities at the early stage of cardiac hypertrophy are not related to ability of calcium transport in cardiac plasma membrane. The indexes of myocardial mechanics are more sensitive to changes in contractility of hypertrophied myocardium as compared with the parameters of ESTLR.  相似文献

在自发性高血压大鼠(SHR)的心肌和主动脉血管组织上观察牛磺酸(taurine)转运和牛磺酸转运体(taurine transporter,TAUT) mRNA 的改变,结果显示,与对照组WKY大鼠相比,SHR组血浆牛磺酸水平和牛磺酸释放量增加,而心肌和血管组织牛磺酸水平和TAUT mRNA含量均降低,牛磺酸最大转运速率(Vmax)分别低24%和35%(P<0.05),米氏常数(Km)值分别高16%和39%(P<0.05),这些结果提示,SHR的心肌和血管组织牛磺酸转运障碍可能与TAUT活性和亲和力降低及TAUT基因水平的下调有关。  相似文献

The content of norepinephrine (NE) and epinephrine (E) in the brain of spontaneously hypertensive rats has proved abnormal, but the cause remained unknown. It was shown in the recent work that NE content in pons, posterior hypothalamus, nucleus caudatus and E concentration in medulla oblongata, anterior and posterior hypothalamus of 12-week old stroke-prone spontaneously hypertensive rats (SHRSP) were much higher than those of age-matched Wister-Kyoto rats (WKY). SHRSP also showed higher levels of systolic blood pressure (SBP) and brain angiotensin II (A II) than WKY. Intracerebroventricular (icv) perfusion of angiotensin-converting enzyme inhibitor captopril (20 micrograms for each time and three times for each day for four weeks) inhibited the synthesis of brain A II and reduced SBP and NE, E contents in all examined brain areas in SHRSP and WKY. However, the effects of chronically perfused captopril on SBP and brain NE, E levels in SHRSP were much more significant than in WKY. The results indicate that the modulatory effects of central renin-angiotensin system (RAS) on central adrenergic and noradrenergic system might be overactivated in SHRSP, which might partially responsible for the abnormally high levels of NE, E in some of the brain areas of SHRSP.  相似文献

工作分析了不同年龄易卒中型自发性高血压大鼠(SHRSP)主动脉中血管紧张素Ⅱ(AⅡ)含量与收缩压(SBP)间的关系。SHRSP的SBP在12及16周龄时均持续上升,20周龄时不再继续上升但维持在高水平;三个年龄组的SHRSP的主动脉AⅡ含量均明显高于同年龄WKY对照鼠,向SHRSP侧脑室灌注巯甲丙脯氨酸四周不仅降低脑区中AⅡ含量,而且具有明显降压效应,同时显著降低主动脉AⅡ含量及血浆、主动脉中去甲肾上腺素和肾上腺素水平,上述结果证实了SHRSP血管中肾素-血管紧张素系统活动的异常与高血压发病学间的密切关系,提示中枢AⅡ可能通过易化外周交感-肾上腺系统活动调节血管中AⅡ水平。  相似文献

Effects of continuous oral administration of captopril were investigated on acute phase in two-kidney Goldblatt hypertensive (2 KGH) rats and spontaneously hypertensive rats (SHR). Systolic blood pressure gradually rose throughout the experimental period of 7, 14, 21 and 28 days in both 2 KGH rats and SHR. These gradual increases of systolic blood pressure were reduced by administration of captopril in both rats. Plasma renin activity were markedly increased throughout the experimental period in both rats treated with captopril, and were modestly increased in 2 KGH rats. In contrast, those changes in plasma renin activity were not obvious in SHR. In 2 KGH rats, juxtaglomerular index (JGI) and juxtaglomerular cell count (JGCC) of the clipped kidneys increased whereas JGI of the opposite kidneys decreased. In contrast, those changes in JGI and JGCC were not obvius in SHR. On the other hand, JGI and JGCC of the clipped kidneys increased in 2 KGH rats treated with captopril and those of the both kidneys increased in SHR treated with captopril. These results suggested that juxtaglomerular cells were related to the development of hypertension in 2 KGH rats, but were not clear in SHR. And these results were found that captopril showed antihypertensive effects, in spite of rises in JGI and JGCC of both 2 KGH rats and SHR.  相似文献

目的 比较两肾两夹和自发性高血压大鼠左心室肥厚的异同.方法 采用不同周龄两肾两夹高血压和自发性高血压大鼠模型,测定动脉血压和心肌肥大指数, 超声心动图观察左心室结构和功能,左心室插管检测血流动力学指标.结果 不同周龄两肾两夹高血压和自发性高血压大鼠的血压及心肌肥大指数均显著高于周龄匹配的对照大鼠.与对照组相比,手术后4周的两肾两夹大鼠及8、12、16周龄的自发性高血压大鼠,其室间隔及左室后壁厚度均明显增加,左室内径显著减小,呈现出明显的向心性肥厚.而手术后8周及12周的两肾两夹大鼠,其室间隔及左室后壁厚度均明显减小,左室内径显著增大,呈明显的离心性肥厚.同时,手术后8周、12周的两肾两夹大鼠以及16周龄的自发性高血压大鼠,其射血分数、缩短分数、室内压最大上升和下降速率均较对照组显著降低,左室舒张末压及等容舒张期压力下降的时间常数明显升高.表明随时间延长,二者心功能都有不同程度的下降.结论 两肾两夹高血压和自发性高血压大鼠均发生了不同程度的左心室肥厚,但其肥厚表型不同.  相似文献

In an attempt to investigate the role of nucleoside transporter function in the hypertensive state, we have compared the binding of [3H]nitrobenzylthioinosine ([3H]NBMPR), a nucleoside transporter probe, in membranes prepared from platelet, renal, pulmonary, cardiac and brain tissues of spontaneously hypertensive rats (SHR) to those of age-matched Wistar-Kyoto (WKY) controls. At 4 weeks of age, [( 3H]NBMPR) binding sites (Bmax) increased in the kidney of SHR but decreased in platelets, whereas no changes were found in the heart, lung or brain. At 18 weeks of age, [3H]NBMPR binding sites (Bmax) remained increased in the kidney and decreased in platelets with no changes in the other tissues. The only change in apparent binding affinity (KD) was an increase in the heart of SHR at 4 weeks. Age-dependent decreases were also observed in the heart and platelets of both SHR and WKY at 18 weeks. The results indicate that the changes in binding characteristics may be due to a combination of the pharmacodynamic differences between the strains, age, as well as to the pathogenesis of hypertension. Consequently, it cannot be concluded that the altered binding characteristics are the result of the elevated blood pressure.  相似文献

Fifteen adult male spontaneously hypertensive rats (one year old) (SHR) were separated into three groups (n=5 each) during 15 weeks as follows: initial control group (IC); final control group (FC); and telmisartan group (T) (1.2 mg/kg/day of telmisartan). Serum and urinary creatinine and proteinuria were not different comparing untreated and telmisartan-treated SHRs. FC rats showed a continuous BP increase during the study while T rats reached the 15th week with a significantly low BP. The LV mass index was significantly smaller in the T group than in the FC group, as was the glomerular hypertrophy. The cardiomyocyte nuclei density per area and the cardiomyocyte mean cross-sectional area were smaller in the T group than in both the IC and FC groups. Intramyocardial artery densities (per area and per volume) were greater in the T group than in untreated SHRs, but myocardial fibrosis was reduced. In conclusion, telmisartan monotherapy effects on BP and also on the hypertension target organs, heart and kidney, are favorable. Telmisartan is able to attenuate SHR cardiomyocyte and glomerular hypertrophies, and myocardial reactive fibrosis as well. It also is favorable to the intramyocardial microcirculation.  相似文献

Hypertension is one of the leading causes of health morbidity and mortality which are linked to many life threatening diseases such as stroke heart problems and renal dysfunction.The integrity of renal microcirculation is crucial to maintaining the clearance and the excretory function in the normotensive and hypertensive conditions. Furthermore, any alteration in the renal function is involved in the pathophysiology of hypertension.The aim of this review is to provide a brief discussion of some factors that regulate renal haemodynamics in spontaneously hypertensive rats, an animal model of hypertension, and how these factors are linked to the disease.  相似文献

Rats made severely hypertensive by renal arterial clipping were treated for 24 days with the arterial vasodilator minoxidil (40, 80, and 120 mg/L drinking water). In all three treated groups of animals, blood pressure initially decreased markedly and to a similar extent. Subsequently partial tolerance developed to the antihypertensive effects of minoxidil. All three doses induced hypertrophy of the right ventricle to a similar degree. In contrast, the hypertension-induced hypertrophy of the left ventricle was further increased in a dose-dependent fashion by minoxidil.  相似文献

The spontaneous tone of vascular smooth muscle is augmented in hypertension. The present study examined the role of nitric oxide (NO), cyclooxygenase (COX), thromboxane A(2)/prostanoid (TP) and PGE(2)/prostanoid (EP-1) receptors, reactive oxygen species, and large-conductance Ca(2+)-activated K(+) (BK(Ca)) channels in the regulation of spontaneous tone in renal arteries of young and mature Wistar-Kyoto (WKY) and spontaneously hypertensive rats (SHR). Rings of arteries, with and without endothelium, were suspended in a myograph for isometric force recording. Spontaneous tone (increase above initial tension) was observed only in arteries of mature SHR and was greater in arteries without endothelium. N(omega)-nitro-L-arginine methyl ester (L-NAME, an inhibitor of NO synthases) induced larger contractions in arteries of SHR than WKY. Indomethacin (a COX inhibitor), SC-19220 (an EP-1 receptor antagonist), and terutroban (a TP receptor antagonist) reduced the L-NAME-evoked contractions. Tiron (a superoxide anion scavenger), catalase (an enzyme that degrades H(2)O(2)), and deferoxamine (a hydroxyl radical scavenger) augmented the L-NAME-induced contractions in arteries of mature SHR. Charybdotoxin (a BK(Ca) channel blocker) caused contractions in arteries of mature SHR without endothelium and in arteries with endothelium incubated with L-NAME. A decreased protein level of endothelial NO synthase, an increased release of prostacyclin, and an increased expression of EP-1 receptors were observed in arteries of mature SHR. The present study suggests that spontaneous tone is precipitated by age and hypertension. The reduced production of NO, leading to decreased activation of BK(Ca) channels, may leave the actions of endogenous vasoconstrictors unopposed. COX products that activate EP-1 and TP receptors are involved in the development of spontaneous tone.  相似文献

The differences in the levels of cytochrome P-450s in hepatic and renal microsomes between spontaneously hypertensive rats (SHR) and normotensive control rats (Wistar Kyoto rats, WKY) were investigated by Western blotting with a specific antibody. Differences in the metabolic activity of the microsomes were also studied. In hepatic microsomes, the content of P450 PB-1 (IIIA2) was 140% higher in SHR than in WKY and the content of P450 IF-3 (IIA1) in SHR was one-seventh that in WKY. The differences reflected the increase in testosterone 6 beta-hydroxylation activity and decrease in testosterone 7 alpha-hydroxylation activity in hepatic microsomes of SHR. The level of P450 K-5 (IVA2) in hepatic microsomes of SHR was 4-times that in microsomes of WKY. The levels of other cytochrome P-450s in SHR were not very different from those in WKY. In renal microsomes, the levels of three renal cytochrome P-450s, P450 K-2, K-4, and K-5, were measured. The level of P450 K-5 (fatty acid omega-hydroxylase) in SHR was 50% higher than that in WKY and the difference reflected the increase in lauric acid omega- and (omega-1)-hydroxylation activities of the renal microsomes of SHR. The levels of P450 K-2 and K-4 did not differ in both rats.  相似文献

Since the identification of the alternative angiotensin converting enzyme (ACE)2/Ang‐(1‐7)/Mas receptor axis, renin‐angiotensin system (RAS) is a new complex target for a pharmacological intervention. We investigated the expression of RAS components in the heart and kidney during the development of hypertension and its perinatal treatment with losartan in young spontaneously hypertensive rats (SHR). Expressions of RAS genes were studied by the RT‐PCR in the left ventricle and kidney of rats: normotensive Wistar, untreated SHR, SHR treated with losartan since perinatal period until week 9 of age (20 mg/kg/day) and SHR treated with losartan only until week 4 of age and discontinued until week 9. In the hypertrophied left ventricle of SHR, cardiac expressions of Ace and Mas were decreased while those of AT1 receptor (Agtr1a) and Ace2 were unchanged. Continuous losartan administration reduced LV weight (0.43 ± 0.02; P < 0.05 versus SHR) but did not influence altered cardiac RAS expression. Increased blood pressure in SHR (149 ± 2 in SHR versus 109 ± 2 mmHg in Wistar; P < 0.05) was associated with a lower renal expressions of renin, Agtr1a and Mas and with an increase in ACE2. Continuous losartan administration lowered blood pressure to control levels (105 ± 3 mmHg; P < 0.05 versus SHR), however, only renal renin and ACE2 were significantly up‐regulated (for both P < 0.05 versus SHR). Conclusively, prevention of hypertension and LV hypertrophy development by losartan was unrelated to cardiac or renal expression of Mas. Increased renal Ace2, and its further increase by losartan suggests the influence of locally generated Ang‐(1‐7) in organ response to the developing hypertension in SHRs.  相似文献
