Twenty-six trichothecene mycotoxins produced by Fusarium sporotrichioides (MC-72083) and Fusarium sambucinum were screened for relative cytotoxicity in cultured baby hamster kidney (BHK-21) cells. The relative cytotoxicity was measured as LC100. The most cytotoxic trichothecenes were T-2 toxin (5 ng/ml) and the recently isolated 4-propanoyl HT-2 (5 ng/ml) and 3-hydroxy T-2 toxin (5 ng/ml). T-2 tetraol (1 × 104 ng/ml), 8--hydroxytrichothecene (1 × 104 ng/ml), sporotrichiol (2 × 104 ng/ml), 8-oxodiacetoxyscirpenol (6 × 104 ng/ml) and 8-acetyl T-2 tetraol (1 × 105 ng/ml) were the least toxic of the regular trichothecenes. None of the modified trichothecenes or the apotrichothecene were very cytotoxic: 8--hydroxysambucoin (2 × 103 ng/ml), FS-1 (5 × 103 ng/ml), 8--hydroxysambucoin (8 × 104 ng/ml) and trichotriol (1 × 105 ng/ml). The modified trichothecenes, FS-2 and FS-3, were not toxic even at 1 × 105 ng/ml. The baby hamster kidney cell bioassay proved to be a very sensitive and reproducible means of screening new trichothecene mycotoxins for relative cytotoxicity.  相似文献

Direct regeneration of shoots and roots through juvenile expiants has been achieved inTribulus terrestris. Cotyledonary leaves along with epicotyl segment from young seedlings were cultured on MS medium containing various concentrations of auxin with cytokinin and glutamine. A combination of 0.2 mgL−1 NAA, 0.5 mgL−1 BAP and 50 mgL−1 glutamine induced high frequency of shoot and root differentiation in 10 weeks. The callus also could be induced on the above medium from the cut end of radical segments. Morphogenic response such as per cent shoot and root differentiation was recorded at regular intervals.  相似文献

Eighty-two cultures of Fusarium species isolated in 1986 from moldy maize in Minnesota were each cultured on rice for 4 weeks and found to produce the following mycotoxins: F. graminearum isolates, deoxynivalenol (DON, 4–225 g/g), 3-acetyldeoxynivalenol (3-ADON, 2–4g/g), 15-acetyldeoxynivalenol (15-ADON, 1–35 g/g) and zearalenone (ZEA, 5–4350 g/g); F. moniliforme, fusarin C (detectable amounts to 1000 g/g); F. mòniliforme, F. oxysporum, F. proliferatum and F. subglutinans isolates, moniliformin (15–6775 g/g); F. moniliforme, F. proliferatum, and F. subglutinans isolates, fusaric acid (detectable amounts). Other mycotoxins screened for in each rice sample and not detected were T-2 toxin, HT-2 toxin, neosolaniol, T-2 tetraol, nivalenol, fusarenon-X, scirpenols, alpha and beta trans-zearalenols, wortmannin, and fusarochromanone. The rat feeding bioassay indicated that other, unidentified toxins may be present.  相似文献

In the present study, a simple one medium formulation protocol for callus culture, somatic embryogenesis and in vitro production of β-carboline alkaloids and diosgenin in Tribulus terrestris L. was developed. Extensive callus induction and proliferation was obtained in leaf explant on Murashige and Skoog (MS) medium supplemented with 5.0 μM 6 benzyl adenine (BA) and 2.5 μM α-naphthaleneacetic acid (NAA). The embryogenic callus was maintained on subculture to fresh parental medium at 4-week intervals over a period of 28 months. The frequency of embryo formation was at a maximum (18.1 ± 0.9 per g of callus) on MS medium containing 5.0 μM BA and 2.5 μM NAA together with 75 mg l−1 casein hydrolysate. Globular embryo developed into torpedo stage embryo under the influence of starvation. The accumulation of β-carboline alkaloids (harmaline and harmine) and steroidal saponin (diosgenin) in non-embryogenic and embryogenic callus culture derived from leaf explant was compared with root, leaf, stem, and fruit of the mother plant. The embryogenic callus accumulated equivalent amounts of harmaline (66.4 ± 0.5 μg/g dry weight), harmine (82.7 ± 0.6 μg/g dry weight), and diosgenin (170.7 ± 1.0 μg/g dry weight) to that of the fruit of T. terrestris. The embryogenic callus culture of this species might offer a potential source for production of important pharmaceuticals.  相似文献

A colorimetric MTT (tetrazolium salt) cleavage test was used to evaluate cytotoxicity of twenty-three Fusarium mycotoxins on two cultured human cell lines (K-562 and MIN-GL1) as well as their inhibitory effect on proliferation of phytohemagglutinin-stimulated human peripheral blood lymphocytes. The values of 50% inhibition of lymphocyte blastogenesis were very close to the 50% cytotoxic doses observed with the more sensitive cell line (MIN-GL1). T-2 toxin was the most cytotoxic with CD50 and ID50 values less than 1 ng/ml. Type A trichothecenes were the most cytotoxic followed by the type B trichothecenes; the non-trichothecenes were the least cytotoxic. Wheat for human consumption (140 samples) was collected after harvest from all regions of Bulgaria. The 1995 crop year was characterized by heavy rainfall in the spring and summer months. The internal mycoflora of wheat samples was dominated by Fusarium spp. and Alternaria spp., and storage fungi were rarely present. The samples were analysed for contamination with Fusarium mycotoxins deoxynivalenol (DON), 3-acetyldeoxynivalenol (3-AcDON), 15-acetyldeoxynivalenol (15-AcDON), T-2 Toxin (T-2), diacetoxyscirpenol (DAS), and zearalenone (ZEA), using enzyme immunoassay methods. DON and ZEA were the predominant toxins, with a contamination frequency of 67% and 69%, respectively. The average levels of these toxins in positive samples were 180 g/kg (DON) and 17 g/kg (ZEA), maximum concentrations were 1800 g kg–1 and 120 g kg–1, respectively. Acetyl derivatives of DON, namely 3-AcDON and 15-AcDON, were found in 2.1 % and 0.7% of the samples, at at maximum level of about 100 g kg–1. Only one sample was positive for T-2 (55 g/kg), DAS was not detected. Tribulus terrestris L., commonly known as puncture vine is a plant that grows in temperate climates and is native in the South and East Europe (Bulgaria, Romania, Hungary and Slovakia). The medicinal attributes of puncture vine influence the furostanol saponins, which have a stimulation effect on natural testosterone levels. In regard to this fact the stimulating effect on the immune, sexual and reproductive system with muscle building, tonic action, vigor, vitality, stamina and physical endurance was proved. The occurrence of puncture vine, in Slovakia, and quantity determination of the specific constituents and heavy metal pollution in this plant material are presented. The quantity of furostanol saponins is various in different plant parts and was modified by growing conditions. The heavy metal content in this plant material was not higher than the maximum permissible concentration of these elements in the food (standard tea) by the Slovakian Food Codex.  相似文献

The steroidal saponins of Tribulus terrestris L. (Zygophyllaceae) are considered to be the factor responsible for biological activity of products derived from this plant. The activity depends on the concentration and the composition of active saponins, which in turn is influenced by the geographical origin of plant material. Samples of T. terrestris collected in Bulgaria, Greece, Serbia, Macedonia, Turkey, Georgia, Iran, Vietnam and India were analyzed by LC-ESI/MS/MS for the presence and the concentration of protodioscin (1), prototribestin (2), pseudoprotodioscin (3), dioscin (4), tribestin (5) and tribulosin (6). The flavonoid rutin (7) was also included in the comparison. The results revealed distinct differences in the content of these compounds depending on region of sample collection, plant part studied and stage of plant development. The samples from Bulgaria, Turkey, Greece, Serbia, Macedonia, Georgia and Iran exhibited similar chemical profile and only some quantitative difference in the content of 1-7 with protodioscin (1) and prototribestin (2) as main components. The Vietnamese and Indian samples exhibit totally different chemical profile. They lack 2 and 5, while tribulosin (6) is present in high amounts. Compounds different from 1 to 7 are dominating in these 3 samples. The presented results suggested the existence of one chemotype common to the East South European and West Asian regions. Most probably, the Vietnamese and Indian samples belong to other chemotypes which are still to be studied and characterized. No clear correlation between the burrs morphology and the chemical composition of the samples has been found.  相似文献

Tribulus terrestris is well known for its medicinal importance in curing urino-genital disorders. Amplified fragment length polymorphism (AFLP), selective amplification of microsatellite polymorphic loci (SAMPL), inter-simple sequence repeat (ISSR) and randomly amplified polymorphic DNA (RAPD) markers were used for the first time for the detection of genetic polymorphism in this medicinal herb from samples collected from various geographical regions of India. Six assays each of AFLP and SAMPL markers and 21 each of ISSR and RAPD markers were utilized. AFLP yielded 500 scorable amplified products, of which 82.9% were polymorphic. SAMPL primers amplified 488 bands, 462 being polymorphic (94.7%). The range of amplified bands was 66 [(TC)8G + M-CAG] to 98 [(CA)6AG + M-CAC] and the percentage polymorphism, 89.9 [from (CT)4C (AC)4A + M-CTG] to 100 [from (GACA)4 + M-CTA]. The ISSR primers amplified 239 bands of 0.4–2.5 kb, 73.6% showed polymorphism. The amplified products ranged from 5 to 16 and the percentage polymorphism 40–100. RAPD assays produced 276 bands, of which 163 were polymorphic (59%). Mantel test employed for detection of goodness of fit established cophenetic correlation values above 0.9 for all the four marker systems. The dendrograms and PCA plots derived from the binary data matrices of the four marker systems are highly concordant. High bootstrap values were obtained at major nodes of phenograms through WINBOOT software. The relative efficiency of the four molecular marker systems calculated on the basis of multiplex ratio, marker index and average heterozygosity revealed SAMPL to be the best. Distinct DNA fingerprinting profile, unique to every geographical region could be obtained with all the four molecular marker systems. Clustering can be a good indicator for clear separation of genotypes from different regions in well-defined groups that are supported by high bootstrap values.  相似文献

A highly embryogenic cell suspension of alfalfa derived from a genotype sensitive to Fusarium oxysporum was successfully used for selection in vitro for resistance to culture filtrates of F. oxysporum, F. solani and F. avenaceum. Fifty two stable resistant cell lines were obtained and 500 plants regenerated from them. Among the 167 regenerants tested under glass there were 12–20% more plants with increased resistance to pathogens than in the group of plants regenerated from a control cell line. It was also found that the cell suspension cultures derived from genotypes of alfalfa with increased resistance to Fusarium spp. better tolerated filtrates of the pathogen. The results of a comparison of virulence of individual isolates of several species of Fusarium with toxicity of their filtrates to plants in vivo and in cell cultures were not unequivocal.  相似文献

Liquid cultures of 200 Fusarium isolates selected to represent the most common species found in autumn pasture (70 isolates) and in grain (130 isolates) grown in New Zealand were analysed for trichothecenes and related compounds. Production of butenolide, cyclonerodiol derivatives and culmorins was also measured. The principal trichothecenes produced were derivatives of either nivalenol (NIV), deoxynivalenol (DON) or scirpentriol (Sctol), in order of frequency. The principal trichothecene producing species were F. crookwellense, F. culmorum and F. graminearum. Isolates of the first two species were predominantly NIV-chemotypes with one or two isolates respectively as Sctol-chemotypes. F. graminearum showed equal quantities of NIV- and DON-chemotypes, with the DON-chemotypes producing primarily 15-acetyldeoxynivalenol (15-ADON).  相似文献

The aim of this study was a monitoring of the occurrence ofAlternaria andFusarium mycotoxins in winter wheat from domestic crop in the year 2003. Altenuene was determined in 56 (100%) samples of winter wheat, range 14.5–41 μg/kg, mean 25 μg/kg. Alternariol was determined in 16 (28.6%) samples of winter wheat, range 6.3–22.1 μg/kg, mean 5.7 μ/kg. DON was determined in 42 (100%) samples of winter wheat, range 250–3500 μg/kg, mean 330 μg/kg. T2-toxin was determined in 42 (100%) samples of winter wheat, range 25–337 μg/kg, mean 99 μg/kg. ZEA was not determined in samples of winter wheat. The effects of neurotensin (NT) and cholecystokinin (CCK) were studied on isolated crop-gizzard preparations of Lumbricus terrestris suspended in a smooth muscle organ bath. Changes in the amplitude and frequency of contractions associated with spontaneous motility were observed in response to neurotransmitters known to have an excitatory effect (acetylcholine) and an inhibitory effect (serotonin); and to the hormones NT and CCK, which in vertebrate models have both been shown to inhibit gastric motility. The overall contractile amplitude and frequency of crop-gizzard contractions were decreased in response to increasing concentrations of NT and CCK. In general, both hormone-induced responses were similar when compared at equal molar concentrations. Cholecystokinin, however, did exhibit a greater reduction in contractile frequency than NT. It is speculated that possible desensitization of earthworm NT-receptors to higher hormone concentrations resulted in a depressed maximal response in the concentration–response curve. Despite that possibility, the overall hormonal inhibition was statistically significant. These results infer that NT- and CCK-induced inhibition of crop-gizzard motility may have a modulatory role in the transport of nutrients and overall efficiency of worm metabolism.  相似文献

The contamination of cereals with mycotoxins produced by species ofFusarium is an important risk to human and animal health. The toxigenic profile is different depending on theFusarium species considered and, in some species, differences can also be observed at intraspecific level. Information about the distribution and variability of the mycotoxigenicFusarium species allow prediction of the toxins that may occur and to devise control strategies. In this work, the occurrence of mycotoxigenicFusarium species associated to cereals was analysed in a wide sample of durum wheat fields (Triticum durum Desf.) and maize from the South West of Spain (Andalucía).F. equiseti, F. graminearum andF. culmorum were the most frequentFusarium species detected in wheat fields followed byF. sambucinum andF. avenaceum, whereas in the case of maize,F. verticillioides andF. proliferatum were the onlyFusarium species present. The relationships of the Spanish isolates from theF. equiseti, F. avenaceum andF. sambucinum species were analysed by nucleotide sequence comparison of a partial region of the Elongation Factor 1 alpha (EF-1α) with other sequences available in data bases. The results indicated thatF. avenaceum andF. equiseti showed high variability and that the SpanishF. equiseti isolates seemed to belong toF. equiseti type II. Three human epithelial cell lines (CaCo-2, HEp-2 and HeLa) implicated as potential targets for three Fusarium toxins were tested for the extent of survival on exposure to increasing toxin concentration and incubation periods. Cytotoxicity assay using 3(4,5-dimethylthiazol-2-yl)2,5-diphenyltetrazolium bromide (MTT) was carried out with deoxynivalenol (DON), T-2 toxins and zearalenone (ZON) on CaCo-2, HEp-2 and HeLa cell lines. Of the three cell lines used, HeLa was the most sensitive, eliciting cell death after 2 days exposure at 100 ng ml–1with T-2 toxin. HeLa was the only cell line to exhibit cytotoxicity towards ZON showing cell death at 1000 ng ml–1after 2 days which increased to 4 days, showing substantial cell death at 200 ng ml–1. HEp-2 was sensitive to DON showing cell death after 2 days (100 ng ml–1) with complete cell death occurring at 200 ng ml–1 after 4 days of exposure. Substantial cytoxicity of T-2 towards HEp-2 occurred after 2 days at 1000 ng ml–1 and complete cell death occurred with 100 ng ml–1 at day 4. The CaCo-2 cell line was generally resistant to the mycotoxins tested between 100 and 1000 ng ml–1. This study shows that cytotoxicity of Fusarium toxins to epithelium cell lines is concentration- and time- dependant and results from ZON–HeLa interaction indicate possible cell type-mycotoxin specificity.  相似文献

Gibberella pulicaris (Fusarium sambucinum) is a promising organism for studying the genetics and regulation of trichothecene biosynthesis; conditions for obtaining fertile crosses have been defined (Desjardins & Beremand, 1987) and crosses between natural variants have provided some information about the number, location, arrangement, and role of genes which determine trichothecene production (Desjardins & Beremand, 1987; Beremand & Desjardins, 1988). The development of some additional experimental tools and methodologies required for the further genetic analysis of trichothecene production inG. pulicaris are described in the present study. A highly fertile, isogenic line was constructed forG. pulicaris strain R-6380. The ability to readily generate mutants in this strain was also demonstrated. Both biochemical and morphological mutants were obtained following UV-mutagenesis. The inheritance of some of these mutations through meiosis indicated that they will be useful genetic markers for crosses and mapping studies. Since strain R-6380 is also transformable (Salch & Beremand, 1988), it is an excellent choice for transmission and molecular genetic studies involving trichothecene production.The mention of firm names or trade products does not imply that they are endorsed or recommended by the U.S. Department of Agriculture over other firms or similar products not mentioned.  相似文献

A total of 57 samples of feedstuffs commonly used for animal nutrition in Colombia (corn, soybean, sorghum, cottonseed meal, sunflower seed meal, wheat middlings and rice) were analyzed for Fusarium contamination. Fusarium fungi were identified at species level by means of conventional methods and the ability to produce fumonisins of the most prevailing species was determined. A total of 41 of the feedstuffs analyzed (71.9%) were found to contain Fusarium spp. Most contaminated substrates were corn (100%), cottonseed meal (100%), sorghum (80%), and soybean (80%). Wheat middlings and rice showed lower levels of contamination (40% and 20%, respectively), while no Fusarium spp. could be isolated from sunflower seed meal. The most prevalent species of Fusarium isolated were F. verticilliodes (70.8%), F.␣proliferatum (25.0%), and F. subglutinans (4.2%). All of them correspond to section Liseola.Production of fumonisins on corn by the isolated Fusarium was screened through liquid chromatography. Almost all strains of F. verticilliodes (97.1%) produced FB1 (5.6–25,846.4 mg/kg) and FB2 (3.4–7507.5 mg/kg). Similarly, almost all strains of F.␣proliferatum (91.7%) produced fumonisins but at lower levels than F.␣verticilliodes (FB1 from 6.9 to 3885.0 mg/kg, and FB2 from 34.3 to 373.8 mg/kg), while F. subglutinans did not produce these toxins. This is the first study in Colombia describing toxigenic Fusarium isolates from␣animal feedstuffs.  相似文献

Fusarium sambucinum Fuckel showed antifeedant activity towards larvae of Galleria mellonella L. when incorporated into insect diet. The activity appeared mostly due to the concentration of trichothecenes present in the fungal extracts. Diacetoxyscirpenol and neosolaniol showed similar levels of activity and were significant potent antifeedants against larvae at 50 and 100 ppm. On the contrary, enniatin B showed no activity up to 100 ppm.  相似文献

【目的】筛选出具有抗菌和抗肿瘤活性的蒺藜内生真菌。【方法】采用牛津杯法、稻瘟菌模型及肿瘤细胞模型评价蒺藜内生真菌PDB和察氏培养基发酵产物的抗菌活性和肿瘤细胞毒性。【结果】PDB培养基发酵液和察氏培养基发酵液抑菌圈直径大于10 mm的菌株分别占总菌株数的19.05%和23.81%。PDB培养基发酵液和察氏培养基发酵液对稻瘟菌的最小抑制浓度(MIC)低于10%的菌株分别占总菌株数的19.05%和47.61%。对肿瘤细胞抑制率高于50%的PDB发酵产物占PDB发酵产物总数的52.38%, 而对肿瘤细胞抑制率高于50%的察氏发酵产物占察氏发酵产物总数的28.57%。【结论】部分蒺藜内生真菌的发酵产物具有抗菌和抗肿瘤活性。  相似文献

Summary Thielavia terrestris NRRL 8126 cell free supernatants contained mannanase and -mannosidase when cultured on a complex media containing locust bean gum. Using acetone precipitation, SP-Sephadex C50 ion exchange chromatography and preparative gel electrophoresis, the crude enzyme was resolved into one -d-mannosidase and four -d-mannanase components. -d-mannosidase had a specific activity of 0.02 (U/mg) onp-nitrophenyl--d-mannopyranoside substrate. Mannanase components M1, M2, M3 and M4 had specific activities of 28.2, 38.7, 52.8 and 4.17 (U/mg) respectively on purified locust bean galactomannan substrate. pH optima for the enzymes were in the range 4.5–5.5. Mannanase component M4 manifested the greatest thermostability, retaining full activity for 3 h at 60°C. Molecular weights determined by SDS-PAGE were 72 000 for -mannosidase and 52 000, 30 000, 55 000 and 89 000 for M1, M2, M3 and M4 respectively. Carbohydrate contents of the enzymes ranged from 6–36%. Preliminary studies indicate that enzyme components hydrolyse the mannan substrate in a synergistic manner.  相似文献
