Two metallothionein cDNA isoforms (MT-1 and MT-2) were isolated from carp (Cyprinus carpio) by RT-PCR. Sequence analysis of the cDNAs revealed two amino acid differences between the coding regions and markedly different 3'-untranslated ends. Gene-specific primers were selected and used in RT-PCR reactions to measure the basal MT-1 and MT-2 mRNA levels and to follow the inducer-specific expression of MT genes in different tissues during in vivo studies. In the brain and muscle, the uninduced levels of the two MT mRNAs were similar. In the kidney and liver, the MT-1 gene product predominated, while in the heart the relative expression levels of the two genes were opposite. Both the MT-1 and MT-2 mRNA levels increased with Cd concentration in a time- and dose-dependent manner. The expression of MT-2, however, was more responsive to a high Cd concentration. In parallel with the induction of the MTs by Cd, we followed the accumulation of this metal in the kidney and liver. Although the Cd level was always higher in the kidney during treatment, the rate of accumulation was higher in the liver. Cold stress resulted in a significantly higher induction of MT-1 than of MT-2, while heat shock had no effect on the expression of either gene.  相似文献

We previously isolated two distinct Saccharomyces cerevisiae myo-inositol transporter genes, ITR1 and ITR2 (Nikawa et al., 1991). Here, we studied the regulation of their expression by measuring steady-state mRNA levels and β-galactosidase activities of lacZ fusion genes under different conditions. The results show that the expression of the two ITR genes is differently regulated: ITR1 was repressed by inositol and choline whereas ITR2 was constitutive. Deletion analysis of the ITR1 upstream region and comparison with the upstream regions of other genes involved in phospholipid synthesis indicate that the octamer sequence 5′-TTCACATG-3′ is important for the expression and inositol/choline regulation of the ITR1 gene.  相似文献

Regulation of the closely linked endogenous sheep MT-Ia, MT-Ib, MT-Ic and MT-II genes by heavy metals and dexamethasone was studied in cultured sheep fibroblasts. Only MT-II mRNA was detectable before addition of any inducer. Addition of copper, zinc or cadmium salts to the culture medium increased the level of each mRNA; however, the magnitude of this response varied greatly between the four metallothionein genes. Following induction, levels of MT-Ia mRNA were the highest, followed by MT-II and MT-Ic mRNAs. The MT-Ib mRNA was only present at low levels. Zinc and cadmium were found to be the most effective inducers of each gene. The maximal response of the sMT-Ia, Ib and II genes to copper was only 30% of zinc and cadmium. The sMT-Ic gene responded very weakly to copper, less than 5% of the levels achieved with zinc. Only the MT-II mRNA increased in response to dexamethasone. In the liver of sheep on normal diets, the levels of MT-Ia, Ic and II mRNAs were found to be unexpectedly high and comparable to induced levels in fibroblasts.  相似文献

Two hsp90 cDNA isoforms (hsp90alpha and hsp90beta) were isolated from the common carp (Cyprinus carpio). Gene-specific probes and primers were selected and used in Northern blot hybridization and RT-PCR reactions to measure the basal hsp90 mRNA levels and to follow the inducer-specific expression of the hsp90 genes in different tissues during in vivo studies. The hsp90beta gene is largely constitutively expressed at a fairly high level in all the examined tissues (brain, liver and kidney) and is slightly inducible by an elevated temperature. Hsp90alpha mRNA is present in the brain, but is hardly detectable in the kidney and liver of unstressed animals. In the brain, this gene is greatly upregulated following thermal stress, whereas in the liver and kidney heat shock has only minor effects on its expression. Hsp90alpha, but not hsp90beta, responds to an elevated level of Cd in a dose-, time- and tissue-dependent manner.  相似文献

In Craterostigma plantagineum the CDeT-6-19 and CDeT-27-45 genes are expressed following desiccation and/or ABA treatment. Their promoters were fused to the -glucuronidase reporter gene (GUS) and tested in transgenic Arabidopsis. GUS activity was measured in mature Arabidopsis seeds, and the responsiveness to ABA in vegetative tissue was found to be limited to the early developmental stages. When transgenic plants were crossed with plants over-expressing the ABI3 gene, it was observed that ABI3 is not required for ABA induction of the CDeT-6-19 promoter, whereas it is crucial for expression of the CDeT-27-45 promoter.  相似文献

A tandemly arranged multigene family encoding putative hexose transporters in Trypanosoma brucei has been characterized. It is composed of two 80% homologous groups of genes called THT1 (six copies) and THT2 (five copies). When Xenopus oocytes are microinjected with in vitro-transcribed RNA from a THT1 gene, they express a glucose transporter with properties similar to those of the trypanosome bloodstream-form protein(s). This THT1-encoded transport system for glucose differs from the human erythrocyte-type glucose transporter by its moderate sensitivity to cytochalasin B and its capacity to transport D-fructose. These properties suggest that the trypanosomal transporter may be a good target for antitrypanosomal drugs. mRNA analysis revealed that expression of these genes was life cycle stage dependent. Bloodstream forms express 40-fold more THT1 than THT2. In contrast, procyclic trypanosomes express no detectable THT1 but demonstrate glucose-dependent expression of THT2.  相似文献

Interleukin 1 (IL 1) production is stimulated by infection, cellular injury, and inflammation. This cytokine directs a wide spectrum of host responses. Human interleukin 1 alpha (IL 1 alpha) was used to examine the time course of effects on zinc metabolism as part of the acute phase response. IL 1 produced a transient depression in the serum zinc concentration and increased serum ceruloplasmin. Metallothionein levels were increased in liver 14-fold after IL 1. Increased expression of metallothionein-1 and -2 genes following IL 1 were observed in liver, bone marrow, and thymus. Pulse-labeling experiments with i.v.-administered 65Zn showed that IL 1 drastically altered zinc distribution kinetics among tissues. More 65Zn was taken up (and/or retained) by the liver, bone marrow, and thymus 6 h after IL 1, whereas correspondingly less 65Zn was found in bone, skin, and intestine. Uptake by other tissues was not affected by IL 1. Chromatography of cytosol from tissues with increased 65Zn uptake suggests the IL 1-induced redistribution may be driven by enhanced metallothionein synthesis. Collectively, the results show that IL 1 regulates zinc metabolism and may direct its preferential, tissue-specific distribution via elevated metallothionein-1 and -2 gene expression.  相似文献
