In order to discover whethor the production of aroma volatilesby apple fruits is dependent on the synthesis of appropriateenzymes during ripening, excised peel, excised cortical tissue,and whole apples were treated with cycloheximide (CH). Volatilerelease, ethylene production, respiration, flesh softening,and peel chlorophyll degradation were measured. The ethylene and volatile compounds produced by excised peelapparently resulted from wounding rather than processes analogousto fruit ripening. Excised cortical tissue was capable of autonomousripening with ethylene production, respiration, and softeningcomparable to that in intact fruits. After infiltration withsucrose solution the same changes occurred, but they were delayedby up to 4 d. Cycloheximide inhibited respiration although theextent of this inhibition decreased after 3 d. Cycloheximideprevented the onset of rapid ethylene production but stimulatedproduction of ethanol, ethyl acetate, and other volatiles. Softeningof CH-treated cortical discs was associated with progressivenecrosis. When whole apples were infiltrated with CH through hypodermicneedles inserted into the core, [14C]valine incorporation wasinhibited from the core to the mid-cortex but not in the peeland outer cortex. Infiltration with sucrose solution delayedmany ripening changes although the time of maximum [14C]valineincorporation was unaffected. Early effects of CH on respirationwere masked by the effects of infiltration, but after 5 d CH-infiltratedfruit contained higher CO2 concentrations and respired morerapidly than controls. Internal ethylene concentrations wereusually lower in CH-treated apples than in controls. CH stimulated release of ethanol and ethyl acetate but inhibitedrelease of higher molecular weight esters such as propyl andbutyl acetates. Cycloheximide-treated fruit softened, but thiswas apparently due to internal necrosis. Peel chlorophyll degradationwas inhibited by CH treatment of whole apples although the tissuehad apparently received no inhibitor.  相似文献

Rapid ripening of mango fruit limits its distribution to distant markets. To better understand and perhaps manipulate this process, we investigated the role of plant hormones in modulating climacteric ripening of ??Kensington Pride?? mango fruits. Changes in endogenous levels of brassinosteroids (BRs), abscisic acid (ABA), indole-3-acetic acid (IAA), and ethylene and the respiration rate, pulp firmness, and skin color were determined at 2-day intervals during an 8-day ripening period at ambient temperature (21?±?1°C). We also investigated the effects of exogenously applied epibrassinolide (Epi-BL), (+)-cis, trans-abscisic acid (ABA), and an inhibitor of ABA biosynthesis, nordihydroguaiaretic acid (NDGA), on fruit-ripening parameters such as respiration, ethylene production, fruit softening, and color. Climacteric ethylene production and the respiration peak occurred on the fourth day of ripening. Castasterone and brassinolide were present in only trace amounts in fruit pulp throughout the ripening period. However, the exogenous application of Epi-BL (45 and 60?ng?g?1 FW) advanced the onset of the climacteric peaks of ethylene production and respiration rate by 2 and 1?day, respectively, and accelerated fruit color development and softening during the fruit-ripening period. The endogenous level of ABA rose during the climacteric rise stage on the second day of ripening and peaked on the fourth day of ripening. Exogenous ABA promoted fruit color development and softening during ripening compared with the control and the trend was reversed in NDGA-treated fruit. The endogenous IAA level in the fruit pulp was higher during the preclimacteric minimum stage and declined during the climacteric and postclimacteric stages. We speculate that higher levels of endogenous IAA in fruit pulp during the preclimacteric stage and the accumulation of ABA prior to the climacteric stage might switch on ethylene production that triggers fruit ripening. Whilst exogenous Epi-BL promoted fruit ripening, endogenous measurements suggest that changes in BRs levels are unlikely to modulate mango fruit ripening.  相似文献

The dependence of ethylene production and respiration rate on the concentration of gibberellin (GA) applied in water emulsion of lanoline to young fruits was studied in Yellow transparent apples. Ethylene production and respiration rate were slightly increased by GA treatment. The increased level was maintained throughout the entire ripening period, which was also apparent from the total amount of both released gases. A gas-chromatographic method, using a flame ionization detector for simultaneous determination of ethylene and CO2 in the atmosphere of experimental biological containers, is described in this paper. CO2 is hydrogenated to methan on Ni — ThO2 catalyst by hydrogen which is used as carrier gas. The gases pass the hydrogenating reactor after the separation in the Chromatographic column before entering the detector.  相似文献

Continuous application of propylene to 40 to 80% mature fruits of normal tomato strains (Lycopersicon esculentum Mill.) advanced ripening in fruits of all ages by at least 50%. Although preclimacteric respiration was stimulated by propylene treatment, there was no concomitant increase in ethylene production. Once ripening commenced, the rates of endogenous ethylene production were similar in both propylene-treated and untreated fruits. Continuous exposure to propylene also stimulated respiration in immature fruits of rin, a nonripening mutant. Although respiration reached rates similar to those during the climacteric of comparable normal fruits there was no change in endogenous ethylene production which remained at a low level. Internal ethylene concentrations in attached 45 to 75% mature fruits of rin and a normal strain were similar. It is suggested that the onset of ripening in normal tomato fruit is not controlled by endogenous ethylene, although increased ethylene production is probably an integral part of the ripening processes.  相似文献

Coffee quality is strongly influenced by a great number of factors, among which the fruit ripening stage at harvest time has a major influence on this feature. Studies comprising ethylene production and the regulation of ethylene biosynthesis genes during the ripening process indicate that ethylene plays an important role on coffee fruit ripening. Coffee early cultivars usually show a more uniform ripening process although little is known about the genetic factors that promote the earliness of ripening. Thus, in order to better understand the physiological and genetic factors involved in the regulation of ripening time, and consequently ripening uniformity, this study aimed to analyze ethylene and respiration patterns during coffee ripening, as well as to analyze ACC oxidase, an ethylene biosynthesis enzyme, gene expression, in fruits of early (Catucaí 785-15) and late (Acauã) coffee cultivars. Coffee fruits were harvested monthly from 124 days after flowering (end of February), until complete maturation (end of June). Dry matter, moisture content, color, respiratory rate and ethylene production analysis were performed. In silico analysis identified a coffee ACC oxidase gene (CaACO-like) and its expression was analyzed by real-time PCR. Dry matter and relative water content constantly increased and gradually decreased, respectively, during fruit ripening, and the color analysis enabled the observation of the earliness in the ripening process displayed by Catucaí 785-15 and its higher fruit ripening uniformity. The results obtained from the CaACO-like expression analysis and respiration and ethylene analysis suggest that the differences in ripening behavior between the two coffee cultivars analyzed in this study may be related to the differences in their capacity to produce ethylene, with fruits of Catucaí 785-15 and Acauã showing a typical and an attenuated climacteric phase, respectively, which may have lead to differences in their ripening time and uniformity.  相似文献

Apples (Malus sylvestris Mill, cv Golden Delicious) were treated before harvest with aminoethoxyvinylglycine (AVG). AVG is presumed to reversibly inhibit 1-aminocyclopropane-1-carboxylic acid (ACC) activity, but not the formation of ACC synthase. AVG treatment effectively blocked initiation of autocatalytic ethylene production and ripening of harvested apples. Exogenous ethylene induced extractable ACC synthase activity and ripening in AVG-treated apples. Removal of exogenous ethylene caused a rapid decline in ACC synthase activity and in CO2 production. The results with ripened, AVG-treated apples indicate (a) a dose-response relationship between ethylene and enhancement of ACC synthase activity with a half-maximal response at approximately 0.8 μl/l ethylene; (b) reversal of ethylene-enhanced ACC synthase activity by CO2; (c) enhancement of ACC synthase activity by the ethylene-activity analog propylene.

Induction of ACC synthase activity, autocatalytic ethylene production, and ripening of preclimacteric apples not treated with AVG were delayed by 6 and 10% CO2, but not by 1.25% CO2. However, each of these CO2 concentrations reduced the rate of increase of ACC synthase activity.

Ripening of climacteric fruit is accompanied by an increasein respiration and autocatalytic ethylene synthesis. In harvestedmelons, there is variation in the magnitude and duration ofthe respiratory climacteric depending on the cultivar. It hasrecently been reported that, while the ripening-associated increasein ethylene production is present, the respiratory climactericis absent in ripening melon fruit attached to the plant, leadingto the suggestion that climacteric respiration is an artifactof harvest. To address the universality of this phenomenon,ripening behaviour in the melon cultivar Charentais (Cucumismelo cv. Reticulatus F1 Alpha), was investigated and the resultsshow that the respiratory climacteric occurs in fruit ripenedboth on and off the plant. Apples are predominantly stored in controlled atmosphere (CA) storage to delay ripening and prolong their storage life. Profiling the dynamics of metabolic changes during ripening and CA storage is vital for understanding the governing molecular mechanism. In this study, the dynamics of the primary metabolism of ‘Jonagold’ apples during ripening in regular air (RA) storage and initiation of CA storage was profiled. 1‐Methylcyclopropene (1‐MCP) was exploited to block ethylene receptors and to get insight into ethylene mediated metabolic changes during ripening of the fruit and in response to hypoxic stress. Metabolic changes were quantified in glycolysis, the tricarboxylic acid (TCA) cycle, the Yang cycle and synthesis of the main amino acids branching from these metabolic pathways. Partial least square discriminant analysis of the metabolic profiles of 1‐MCP treated and control apples revealed a metabolic divergence in ethylene, organic acid, sugar and amino acid metabolism. During RA storage at 18°C, most amino acids were higher in 1‐MCP treated apples, whereas 1‐aminocyclopropane‐1‐carboxylic acid (ACC) was higher in the control apples. The initial response of the fruit to CA initiation was accompanied by an increase of alanine, succinate and glutamate, but a decline in aspartate. Furthermore, alanine and succinate accumulated to higher levels in control apples than 1‐MCP treated apples. The observed metabolic changes in these interlinked metabolites may indicate a coordinated adaptive strategy to maximize energy production.  相似文献

When treated with acetic or propionic acid vapours, intact pre-climacteric Golden Delicious apples started ripening prematurely. Experiments with [2?14C]propionic acid showed that a small part of the added acid is transformed into [14C]ethylene, and that ripening probably begins in these circumstances as a result of the artificial increase in the endogenous ethylene level. It may be that in unripe apples the small amount of evolved ethylene is mostly derived from simple organic acids, so that the moment at which its triggering concentration is reached depends on the available supply of acids. As this, in turn, is a function of the state of maturity of the fruit, there must be a direct relationship between the start of ripening and the degree of maturity. Thus, even in very unripe apples, catabolic processes are at work or potentially present because of the modified β–oxidation of [1?14C]propionic acid observed.  相似文献

The ability of ethylene to stimulate respiration and advance the onset of rapid ethylene production was investigated at different times during storage of 'Gloster 69' apples in 2 kPa O2 at 1.5–3.5°C. Ethylene stimulated respiration in apples at 15°C immediately after harvest; maximal rates were recorded at 10–1000 μl I−1 but attainment of these rates was delayed after low O2 storage until day 3 of treatment at 15°C. The onset of rapid ethylene production at 15°C occurred later in non-ethylene-treated apples after storage than after harvest. Ethylene production was induced in some apples during ethylene treatment for 3 or 6 days; in others it was induced about 20 days after treatment, but a proportion of the fruit showed no induction in the 45-day duration of experiments. An ethylene treatment at 10 μl I−1 led to a near maximal increase in the frequency of induction of ethylene production at all times. After storage apples were mainly induced during treatment or not induced, whereas after harvest induction after treatment was more frequent. The presence of 2000 μl l−1 norbornadiene during ethylene treatment inhibited the stimulation of respiration and the induction of ethylene production; this inhibition was only partly reversed by ethylene at 1000 μl l−1 the experiments suggest that receptors for ethylene were present at all stages but that response capacity changed during storage.  相似文献

Avocado (Persea americana Mill. cv Hass) discs (3 mm thick) ripened in approximately 72 hours when maintained in a flow of moist air and resembled ripe fruit in texture and taste. Ethylene evolution by discs of early and midseason fruit was characterized by two distinct components, viz. wound ethylene, peaking at approximately 18 hours, and climacteric ethylene, rising to a peak at approximately 72 hours. A commensurate respiratory stimulation accompanied each ethylene peak. Aminoethoxyvinyl glycine (AVG) given consecutively, at once and at 24 hours following disc preparation, prevented wound and climacteric respiration peaks, virtually all ethylene production, and ripening. When AVG was administered for the first 24 hours only, respiratory stimulation and softening (ripening) were retarded by at least a day. When AVG was added solely after the first 24 hours, ripening proceeded as in untreated discs, although climacteric ethylene and respiration were diminished. Propylene given together with AVG led to ripening under all circumstances. 2,5-Norbornadiene given continuously stimulated wound ethylene production, and it inhibited climacteric ethylene evolution, the augmentation of ethylene-forming enzyme activity normally associated with climacteric ethylene, and ripening. 2,5-Norbornadiene given at 24 hours fully inhibited ripening. When intact fruit were pulsed with ethylene for 24 hours before discs were prepared therefrom, the respiration rate, ethylene-forming enzyme activity buildup, and rate of ethylene production were all subsequently enhanced. The evidence suggests that ethylene is involved in all phases of disc ripening. In this view, wound ethylene in discs accelerates events that normally take place over an extended period throughout the lag phase in intact fruit, and climacteric ethylene serves the same ripening function in discs and intact fruit alike.  相似文献

Observations were made of the effects of several plant regulators, indole-3-acetic acid, kinetin, abscisic acid, and gibberellic acid, as well as of extracts prepared from leaves and fruit stalks on the respiration pattern, ethylene production, and the number of days to ripen of avocado fruits (Persea americana Mill.). These substances were vacuum infiltrated to insure good penetration and distribution. Kinetin, abscisic acid, gibberellic acid, and the extracts had no effect on either ripening time or on the respiration pattern and ethylene production of the fruits. Indoleacetic acid, however, had a marked effect on ripening. At high concentrations (100 and 1000 mum), indoleacetic acid stimulated respiration and induced preclimacteric ethylene production, resulting in accelerated ripening of the fruits. At the low concentrations (1 and 10 mum), it delayed ripening of fruits and suppressed the climacteric respiration and ethylene production. The results reinforce several previous observations with other fruits that auxins may largely constitute ;resistance to ripening' and may be responsible for the lack of ripening shown by unpicked fruits.  相似文献

Avocado (Persea americana Mill.) fruits were harvested at successivedevelopment stages during a period of 10 months. Ethylene productionand respiration were determined during the post-harvest period. Detached immature fruits were found to have a preclimactericincrease in ethylene production and respiration without anysigns of ripening. In fruits larger than 20 g a second phaseof climacteric ethylene production and respiration, associatedwith ripening, ensued. The preclimacteric ethylene was produced mainly by the seedcoat. It is suggested that the high ethylene production potentialof the seed coat may serve as a means for inducing abscissionin young fruits.  相似文献

Ethylene promotes fruit ripening, including softening. The fruit of melting-flesh peach (Prunus persica (L). Batsch) cultivar 'Akatsuki' produces increasing levels of ethylene, and the flesh firmness softens rapidly during the ripening stage. On the other hand, the fruit of stony hard peach cultivars 'Yumyeong', 'Odoroki', and 'Manami' does not soften and produces little ethylene during fruit ripening and storage. To clarify the mechanism of suppression of ethylene production in stony hard peaches, the expression patterns of four ethylene biosynthesis enzymes were examined: ACC synthases (Pp-ACS1, Pp-ACS2, and Pp-ACS3) and ACC oxidase (Pp-ACO1). In the melting-flesh cultivar 'Akatsuki', Pp-ACS1 mRNA was dramatically induced after harvesting, and a large amount of ethylene was produced. On the other hand, in stony hard peaches, Pp-ACS1 mRNA was not induced during the ripening stage, and ethylene production was inhibited. Since Pp-ACS1 mRNA was induced normally in senescing flowers, wounded leaves, and wounded immature fruit of 'Yumyeong', Pp-ACS1 was suppressed only at the ripening stage, and was not a defect in Pp-ACS1. These results indicate that the suppression of fruit softening in stony hard peach cultivars was caused by a low level of ethylene production, which depends on the suppressed expression of Pp-ACS1.  相似文献

The effects of daminozide (butanedioic acid-2,2-dimethylhydrazide) on ethylene synthesis by apple fruits were investigated. The objective was to determine the effects of postharvest applications as compared to the standard application of diaminozide in the orchard. Immersion in a solution containing 4.25 g L?1 active ingredient for 5 min delayed the rise in ethylene production in individual “Cox” apples at 15°C by about 2 days, whereas orchard application of 0.85 g L?1 caused delays of about 3 days. Both modes of application depressed the maximal rate of ethylene production attained by ripe apples by about 30%. Daminozide did not affect the stimulation of respiration by ethylene treatment of “Gloster” apples, but it delayed the increase in ethylene synthesis. Daminozide applied immediately after harvest delayed the rise in ethylene synthesis in “Golden Delicious” held at 15°C, but it was less effective when applied 48 h after harvest or when apples were held at 5°C. Exposure to 1–2 μl L?1 ethylene for 48 h was less effective in promoting the rise in ethylene in daminozidetreated “Cox” and “Gloster” apples than in untreated fruit. High (100–1000 μl L?1) concentrations of ethylene more or less overcame the daminozide effect. Apples absorbed about 40% of surface-applied [14C]daminozide in 48 h, but more than 90% of the radioactivity in the fruit was recovered from the peel and outer 1 cm of the cortex. Daminozide was partly converted to carbon dioxide and other metabolites.  相似文献

Apples ( Malus domestica Borkh.) were heated for 4 days at 38°C immediately after harvest and then placed at 20°C for 7–10 days. Protein synthesis, ethylene production and fruit softening were reversibly inhibited by the heat treatment. Fruit respiration, membrane permeability and chlorophyll degradation in the fruit peel were enhanced during the treatment. The heat-treated apples ripened normally but more slowly than untreated apple We hypothesize that heat treatment differentially affects processes which normally increase simultaneously during fruit ripening, by inhibiting those processes which require tie novo protein synthesis and enhancing those that do not.  相似文献

Papaya ( Carica papaya L.) cell wall matrix polysaccharides are modified as the fruit starts to soften during ripening and an endoxylanase is expressed that may play a role in the softening process. Endoxylanase gene expression, protein amount and activity were determined in papaya cultivars that differ in softening pattern and in one cultivar where softening was modified by the ethylene receptor inhibitor 1-methylcyclopropene (1-MCP). Antibodies to the endoxylanase catalytic domain were used to determine protein accumulation. The three papaya varieties used in the study, 'Line 8', 'Sunset', and 'Line 4-16', differed in softening pattern, respiration rate, ethylene production and showed similar parallel relationships during ripening and softening in endoxylanase expression, protein level and activity. When fruit of the three papaya varieties showed the respiratory climacteric and started to soften, the level of endoxylanase gene expression increased and this increase was related to the amount of endoxylanase protein at 32 kDa and its activity. Fruit when treated at less than 10% skin yellow stage with 1-MCP showed a significant delay in the respiratory climacteric and softening, and reduced ethylene production, and when ripe was firmer and had a 'rubbery' texture. The 1-MCP-treated fruit that had the 'rubbery' texture showed suppressed endoxylanase gene expression, protein and enzymatic activity. Little or no delay occurred between endoxylanase gene expression and the appearance of activity during posttranslational processing from 65 to 32 kDa. The close relationship between endoxylanase gene expression, protein accumulation and activity in different varieties and the failure of the 1-MCP-treated fruit to fully soften, supported de novo synthesis of endoxylanase, rapid posttranslation processing and a role in papaya fruit softening.  相似文献
