Effect of substrate-dependent microbial ethylene production on plant growth   总被引:1,自引:0,他引:1  
Various compounds have been identified as precursors/substrates for the synthesis of ethylene (C2H4) in soil. This study was designed to compare the efficiency of four substrates, namely L-methionine (L-MET), 2-keto-4-methylthiobutyric acid (KMBA), 1-aminocyclopropane-1-carboxylic acid (ACC), and calcium carbide (CaC2), for ethylene biosynthesis in a sandy clay loam soil by gas chromatography. The classic “triple” response in etiolated pea seedling was employed as a bioassay to demonstrate the effect of substrate-dependent microbial production of ethylene on plant growth. Results revealed that an amendment with L-MET, KMBA, ACC (up to 0.10 g/kg soil) and CaC2 (0.20 g/kg soil) significantly stimulated ethylene biosynthesis in soil. Overall, ACC proved to be the most effective substrate for ethylene production (1434 nmol/kg soil), followed by KMBA, L-MET, and CaC2 in descending order. Results further revealed that ethylene accumulation in soil from these substrates caused a classic “triple” response in etiolated pea seedlings with different degrees of efficacy. A more obvious classic “triple” response was observed at 0.15, 0.10, and 0.20 g/kg soil of L-MET, KMBA/ACC, and CaC2, respectively. Similarly, direct exposure of etiolated pea seedlings to commercial ethylene gas also modified the growth pattern in the same way. A significant direct correlation (r = 0.86 to 0.97) between substrate-derived C2H4 and the classic triple response in etiolated pea seedlings was observed. This study demonstrated that the presence of substrate(s) in soil may lead to increased ethylene concentration in the air of the soil, which may affect plant growth in a desired direction. Thirty-day-old seedlings of tomato (Lycopersicon esculentum cv. Kwangsoo) were treated with various cadmium (Cd) concentrations (0, 10, 50, 100, and 500 μM) for up to 20 days, and the detailed distribution of absorbed Cd and its phytotoxicity in different plant parts (root, stem, and leaves) were investigated. The accumulation of Cd in plants increased with external Cd concentrations and Cd was strongly retained by roots, with less than 30% of the absorbed Cd being transported to shoots. Among the leaves, the lower positioned older leaves accumulated more Cd than the younger leaves. Furthermore, Cd-exposure not only reduced the dry weight and length of both shoot and root, chlorophyll levels in leaves, and levels of photosynthesis, but also enhanced the concentration of malondialdehyde (a lipid peroxidation product) in all plant parts. Our results indicate that the physiological impairment of tomato seedlings exposed to toxic levels of Cd may be related to the internal distribution of absorbed Cd, prolonged exposure, and oxidative stress in different plant parts.  相似文献

Phytostabilization strategies may be suitable to reduce the dispersion of uranium (U) and the overall environmental risks of U-contaminated soils. The role of Glomus intraradices, an arbuscular mycorrhizal (AM) fungus, in such phytostabilization of U was investigated with a compartmented plant cultivation system facilitating the specific measurement of U uptake by roots, AM roots and extraradical hyphae of AM fungi and the measurement of U partitioning between root and shoot. A soil-filled plastic pot constituted the main root compartment (CA) which contained a plastic vial filled with U-contaminated soil amended with 0, 50 or 200 mg KH2PO4−P kg–1soil (CB). The vial was sealed by coarse or fine nylon mesh, permitting the penetration of both roots and hyphae or of just hyphae. Medicago truncatula plants grown in CA were inoculated with G. intraradices or remained uninoculated. Dry weight of shoots and roots in CA was significantly increased by G. intraradices, but was unaffected by mesh size or by P application in CB. The P amendments decreased root colonization in CB, and increased P content and dry weight of those roots. Glomus intraradices increased root U concentration and content in CA, but decreased shoot U concentrations. Root U concentrations and contents were significantly higher when only hyphae could access U inside CB than when roots could also directly access this U pool. The proportion of plant U content partitioned to shoots was decreased by root exclusion from CB and by mycorrhizas (M) in the order: no M, roots in CB > no M, no roots in CB > M, roots in CB > M, no roots in CB. Such mycorrhiza-induced retention of U in plant roots may contribute to the phytostabilization of U contaminated environments.  相似文献

A pot experiment was conducted on tomato (Solanum lycopersicum cv. Pusa Ruby) to assess the effect of different phosphorus (P) levels (0, 125, 250 and 500 mg/pot) and the plant growth promoting rhizobacterium, Pseudomonas fluorescens, on the growth of tomato and on the reproduction of Meloidogyne incognita. Maximum growth of tomato occurred at P rates of 125 mg/kg soil, irrespective of whether plants were uninoculated or inoculated with P. fluorescens or M. incognita or inoculated with both the agents. Nematodes per gram of roots, egg masses per root, eggs per egg mass and galls per root significantly increased by increasing levels of P. P. fluorescens performed better than other treatments and different P levels in improving tomato growth and reducing galling and multiplication of M. incognita.  相似文献

The effect of high zinc concentrations on the growth and photosynthetic apparatus of Setaria viridis (L.) Beauv. was investigated under laboratory conditions and a vegetation experiment. The experiments showed that zinc concentrations of 10−6 to 10−3 M did not influence seed germination. Moreover, zinc in concentrations of 40 and 80 mg kg substrate had no significant effect on most of the plant growth parameters. However, higher metal concentrations (160 and 320 mg kg substrate) inhibited shoot biomass, leaf area, inflorescence length, and biomass. At the same time, parameters such as the content of chlorophyll (a + b), F 0, Fv, and the Fv/Fm rate were not reduced even in the presence of zinc in the highest concentrations (320 mg kg substrate). S. viridis tolerance of high zinc concentrations, as well as its capacity to accumulate zinc in roots and shoots, suggests the plant can be used for phytoremediation of zinc polluted soils.  相似文献

The roles of gibberellic acid (GA3) and ethylenediaminetetraacetic acid (EDTA) in phytoremediation of cadmium (Cd)-contaminated soil by Parthenium hysterophorus plant was investigated. GA3 (10?9, 10?7, and 10?5M) was applied as a foliar spray. EDTA was added to soil in a single dose (160 mg/kg soil) and split doses (40 mg/kg soil, four split doses). GA3 and EDTA were used separately and in various combinations. P. hysterophorus was selected due to its fast growth and unpalatable nature to herbivores to reduce the entrance of metal into the food chain. The Cd phytoextraction potential of the P. hysterophorus plant was evaluated for the first time. Cd significantly reduced plant growth and dry biomass (DBM). GA3 alone increased the plant growth and biomass in Cd-contaminated soil, whereas EDTA reduced it. GA3 in combination with EDTA significantly increased the growth and biomass. The highest significant DBM was found in treatment T3 (10?5M GA3). All treatments of GA3 or EDTA significantly enhanced the plant Cd uptake and accumulation compared with control (C1). The highest significant root and stem Cd concentrations were found in the combination treatment T11 (GA3 10?5M + EDTA split doses), whereas in leaves it was found in the EDTA treatments. Cd concentration in plant parts increased in the order of stem < leaves < roots. The combination treatment T9 (GA3 10?7M + EDTA split doses) showed the significantly highest total Cd accumulation (8 times greater than control C1, i.e., only Cd used). The GA3 treatments accumulated more than 50% of the total Cd in the roots, whereas the EDTA treatments showed more than 50% in the leaves. Root dry biomass showed a positive and significant correlation with Cd accumulation. GA3 is environment friendly as compared with EDTA. Therefore, further investigation of GA3 is recommended for phytoremediation research for the remediation of metal-contaminated soil.  相似文献

Summary The absorption and transport of Cu were studied in perennial ryegrass grwon on 21 soils under controlled environment conditions. Neither the concentration, nor the total amount, of Cu in the shoots was related to available Cu in the soils as assessed by extraction with 0.05M EDTA, 0.005M DTPA, or 1.95 per cent HNO3. The concentration in the roots and, more especially, absorption per unit weight of root (i.e. μg Cu g dry wt−1) were, however, highly correlated with available soil Cu. This suggests that, unless the extent of exploitation of the soil by roots is taken into account, measurements of available Cu will not be effective in predicting uptake by plants. On average, 63 per cent of the Cu absorbed by the roots was retained in the roots, and variation in the proportion retained was related to the transport of nitrogen from roots to shoots. On some soils the concentrations of N and Cu in the shoots were significantly correlated, and variation in N concentration accounted for a considerable proportion of the variance in the Cu concentration at later harvests. The relative importance of the measured soil (pH, organic matter) and plant (dry weight, N content) factors changed markedly over 6 successive harvests.  相似文献

This study investigates the distribution of carboxylates and acid phosphatases as well as the depletion of different phosphorus (P) fractions in the rhizosphere of three legume crop species and a cereal, grown in a soil with two different levels of residual P. White lupin (Lupinus albus L.), field pea (Pisum sativum L.), faba bean (Vicia faba L.) and spring wheat (Triticum aestivum L.) were grown in small sand-filled PVC tubes to create a dense root mat against a 38-μm mesh nylon cloth at the bottom, where it was in contact with the soil of interest contained in another tube. The soil had either not been fertilised (P0) or fertilised with 15 (P15) kg P ha−1 in previous years. The mesh size did not allow roots to grow into the soil, but penetration of root hairs and diffusion of nutrients and root exudates was possible, and a rhizosphere was established. At harvest, thin (1 mm) slices of this rhizosphere soil were cut, down to a 10-mm distance from the mesh surface. The rhizosphere of white lupin, particularly in the P0 treatment, contained citrate, mostly in the first 3 mm, with concentrations decreasing with distance from the root. Acid phosphatase activity was enhanced in the rhizosphere of all species, as compared with bulk soil, up to a distance of 4 mm. Phosphatase activity was highest in the rhizosphere of white lupin, followed by faba bean, field pea and wheat. Both citrate concentrations and phosphatase activities were higher in P0 compared with P15. The depletion of both inorganic (Pi) and organic (Po) phosphorus fractions was greatest at the root surface, and decreased gradually with distance from the root. The soil P fractions that were most depleted as a result of root activity were the bicarbonate-extractable (0.5 M) and sodium hydroxide-extractable (0.1 M) pools, irrespective of plant species. This study suggests that differences among the studied species in use of different P pools and in the width of the rhizosphere are relatively small.  相似文献

Argemone mexicana L. (Papaveraceae), a tropical annual weed, is phytotoxic to many crop species. This study was designed to examine the allelochemical and nematicidal potential of A. mexicana and to better understand the role of this weed in the ecosystem. A methanol-soluble extract of the leaf material caused greater juvenile mortality of Meloidogyne javanica than did ethyl acetate or hexane extracts indicating the polar nature of the toxins. Decomposing tissues of A. mexicana in soil at 50 g kg–1 were highly deleterious causing 80% mortality of tomato plants. At 10 g kg–1 plant growth was enhanced, while at 30 g kg–1 plant growth was substantially retarded. M. javanica population densities in the rhizosphere and in roots, and gall formation were significantly suppressed when 10, 30 or 50 g kg–1 A. mexicana was allowed to decompose in the soil. To establish whether decomposition was necessary to produce phytotoxic symptoms, or whether the shoot extract alone could interfere with plant growth, an aqueous shoot extract was applied to soil. Whereas a 50% extract promoted plant growth, a 100% (100 g/500 mL distilled water) concentration significantly reduced plant height, and fresh weights of shoot and root. In general, decomposing plant material caused greater phytotoxicity compared to the aqueous extract. Addition of N as NH4NO3 partially alleviated the phytotoxic action of A. mexicana,and also reduced severity of root-knot disease. Adding Pseudomonas aeruginosa to soil amended with A. mexicana resulted in decreased density of M. javanicain the rhizosphere and in tomato roots, suppressed galling rates and enhanced plant growth.  相似文献

Renealmia mexicana (Klotzsch ex. Petersen) is a tropical plant found in southern México with an ornamental value and a potential source of curcuminoids. Its distribution in Chiapas has decreased because of deforestation and low propagation and germination rate, so a protocol for in vitro propagation was developed. An orthogonal experimental design of L9 (34) in triplicate was used to investigate the effect of 6-benzyl adenine (BA), indole butyric acid (IBA), silver nitrate (AgNO3), and sucrose on shoot, root, and leaf development of plantlets grown in vitro. Plantlets with well-developed shoots and roots were transferred to pots containing a mixture of peat moss and agrolite for hardening before transfer to soil. The Murashige and Skoog (Physiol. Plant. 15:473–497, 1962) mineral medium (MS) supplemented with 4.4 μM BA, 2.5 μM IBA, 11.7 μM AgNO3y and 5.5% (w/v) sucrose gave most shoots, 8.9 μM BA, 2.5 μM IBA, 17.7 μM AgNO3 and 5.5% (w/v) sucrose most roots, and 8.9 μM BA, 4.9 μM IBA, 11.7 μM AgNO3 and 3.0% (w/v) sucrose most leaves, although other combinations were statistically equivalent in each case. Sucrose was the factor that most explained the variation in the promotion of shoots, roots, and leaves. The protocol developed resulted in up to 100% survival when plantlets were transferred to soil using AgNO3, confirming that hardening of plantlets in vitro using hormonal stimulation was a suitable strategy to improve acclimatization.  相似文献

Glasshouse experiments were conducted to evaluate the influence of L-methionine (L-MET) and L-ethionine (L-ETH) added to soil on the growth of corn (Zea mays L.) and tomato (Lycopersicon esculentum), respectively. The application of L-MET and L-ETH stimulated C2H4 production in soil by 299- and 313-fold, respectively, over an unamended control. An L-MET treatment of 1.85 mg kg−1 soil was the most effective in increasing shoot height, shoot fresh weight, internodal distance, and stem diameter in two corn cultivars, Kandy Korn and Miracle, while shoot and root dry weights, leaf width, uppermost leaf collar base distance and resistance to stem breaking were increased in the case of Kandy Korn only. A significant epinastic response was observed in the second and third leaves of tomato plants when soil was treated with L-ETH. An L-ETH treatment of 0.2 mg kg−1 soil resulted in the maximum fresh fruit yield, while 0.02 and 2.0 mg kg−1 gave the most fruit and greater average weight of fresh fruit, respectively. Concentrations ranging from 0.002 to 2.0 mg L-ETH kg−1 soil initiated early fruit formation. Early fruit ripening was observed with an application rate of 20 mg L-ETH kg−1 soil. The mechanism of action of these chemicals could either be attributed to i) substrate-dependent C2H4 production in soil by the indigenous microflora, ii) uptake directly by plant roots followed by metabolism within the tissues, and/or iii) a change in the balance of rhizosphere microflora affecting plant growth.  相似文献

Filipendula ulmaria (L.) Maxim (meadowsweet) is a medicinal plant that is claimed to have several biological activities, including anti-tumor, anti-carcinogenic, anti-oxidant, anti-coagulant, anti-ulcerogenic, anti-microbial, anti-arthritic, and immunomodulatory properties. This report describes, for the first time, an efficient plant regeneration system for F. ulmaria via adventitious shoot development from leaf, petiole, and root explants cultured on Murashige and Skoog’s minimal organics medium containing different concentrations of thidiazuron (TDZ), benzyladenine, and kinetin either alone or in combination with different auxins. Relatively extensive/prolific shoot regeneration was observed in all three explant types with TDZ in combination with indole-3-acetic acid (IAA). Gibberellic acid (GA3), TDZ, and IAA combinations were also tested. The best shoot proliferation was observed among root explants cultured on media supplemented with 0.45 μM TDZ + 2.85 μM IAA + 1.44 μM GA3. Regenerated shoots were transferred to rooting media containing different concentrations of either IAA, indole-3-butyric acid (IBA), naphthalene acetic acid, or 2,4-dichlorophenoxyacetic acid. Most shoots developed roots on medium with 2.46 μM IBA. Rooted explants were transferred to vermiculite in Magenta containers for a 2-wk acclimatization period and then finally to plastic pots containing potting soil. The plantlets in soil were kept in growth chambers for 2 wk before transferring to greenhouse conditions.  相似文献

The effects of increase copper concentrations in medium (10–150 μM CuSO4) on growth and viability of the roots of two-week-old soybean seedlings (Glycine max L., cv. Dorintsa) were studied. Copper excess suppressed biomass accumulation and linear plant growth; copper affected root growth much stronger than shoot growth. The presence of 10 μM CuSO4 in medium suppressed accumulation of plant biomass by 40% and the root length by 70%; in the presence of 25 μM CuSO4, these indices were equal to 80 and 90%, respectively. In the presence of 50 μM CuSO4, roots ceased to grow but biomass and shoot length still increased slightly. 150 μM CuSO4 was lethal for plants. The earliest sign of excessive copper toxicity was the accumulation of MDA, indicating activation of membrane lipid peroxidation. A significant increase in MDA content was observed at plant incubation in medium with 10 μM CuSO4 for 1 h; in this case, the content of copper in the roots increased from 36 ±1.8 (in control) to 48 ± 2.4 μg/g dry wt. The number of dead cells (permeable for the dye Evans Blue) was doubled in the presence of 200 μg/g dry wt within the root; this occurred in 72 h of growth in medium with 10 μM CuSO4, in 6 h at 25 μM CuSO4, in 3 h at 50 μM CuSO4, and 1 h at 150 μM CuSO4. Toxicity of copper excess was manifested stronger in dividing and elongation cells of the root apex (root meristem and the zone of elongation) than in more basal root regions. Copper excess resulted in the formation of breaks in the surface cell layers of the root tips and affect root morphology. When plant grew in medium with 10 μM CuSO4, a distance of lateral root formation zone from the root tip decreased markedly, and spherical swellings were formed on the tips of lateral roots. The higher copper concentrations (50 and 150 μM) suppressed completely the development of lateral roots.  相似文献

Trichoderma asperellum strain T34 has been reported to control the disease caused by Fusarium oxysporum f.sp. lycopersici (Fol) on tomato plants. To study the importance of iron concentration in the growth media for the activity and competitiveness of T34 and the pathogen, we tested four iron concentrations in the nutrient solution [1, 10, 100, and 1000 μM provided as EDTA/Fe(III)] in a biological control experiment with T34 and Fol in tomato plants. The reduction of the Fusarium-infected shoot by T34 was only significant at 10 μM Fe. We hypothesized that Fe competition is one of the key factors in the biocontrol activity exerted by T34 against Fol, as an increase in Fe concentration over 10 μM would lead to the suppression of T34 siderophore synthesis and thus inhibition of Fe competition with Fol. T34 significantly reduced the populations of Fol at all the doses of Fe assayed. In contrast, Fol enhanced the populations of T34 at 1 and 10 μM Fe. Nevertheless, several plant physiological parameters like net CO2 assimilation (A), stomatal conductance (g s), relative quantum efficiency of PSII (Φ PSII), and efficiency of excitation energy capture by open PSII reactive centers (Fv′/Fm′) demonstrated the protection against Fol damage by treatment with T34 at 100 μM Fe. The first physiological parameter affected by the disease progression was g s. Plant dry weight was decreased by Fe toxicity at 100 and 1,000 μM. T34-treated plants had significantly greater heights and dry weights than control plants at 1,000 μM Fe, even though T34 did not reduce the Fe content in leaves or stems. Furthermore, T34 enhanced plant height even at the optimal Fe concentration (10 μM) compared to control plants. In conclusion, T. asperellum strain T34 protected tomato plants from both biotic (Fusarium wilt disease) and abiotic stress [Fe(III) toxic effects].  相似文献

Plant roots are known to orient growth through the soil by gravitropism, hydrotropism, and thigmotropism. Recent observations of plant roots that developed in a microgravity environment in space suggested that plant roots may also orient their growth toward oxygen (oxytropism). Using garden pea (Pisum sativum L. cv. Weibul's Apollo) and an agravitropic mutant (cv. Ageotropum), root oxytropism was studied in the controlled environment of a microrhizotron. A series of channels in the microrhizotron allowed establishment of an oxygen gradient of 0.8 mmol · mol−1 · mm−1. Curvature of seedling roots was determined prior to freezing the roots for subsequent spectrophotometric determinations of alcohol dehydrogenase activity. Oxytropic curvature was observed all along the gradient in both cultivars of pea. The normal gravitropic cultivar showed a maximal curvature of 45° after 48 h, while the agravitropic mutant curved to 90°. In each cultivar, the amount of curvature declined as the oxygen concentration decreased, and was linearly related to the root elongation rate. Since oxytropic curvature occurred in roots exposed to oxygen concentrations that were not low enough to induce the hypoxically responsive protein alcohol dehydrogenase, we suspect that the oxygen sensor associated with oxytropism does not control the induction of hypoxic metabolism. Our results indicate that oxygen can play a critical role in determining root orientation as well as impacting root metabolic status. Oxytropism allows roots to avoid oxygen-deprived soil strata and may also be the basis of an auto-avoidance mechanism, decreasing the competition between roots for water and nutrients as well as oxygen. Growth, lipid peroxidation, H2O2 produciton and the response of the antioxidant enzymes and metabolites of the ascorbate glutathione pathway to oxidative stress caused by two concentrations (50 and 100 µM) of Cr(III) and Cr(VI) was studied in 15 day old seedlings of sorghum (Sorghum bicolor (L.) Moench cv CO 27) after 10 days of treatment. Cr accumulation in sorghum plants was concentration and organ dependant. There was no significant growth retardation of plants under 50 µM Cr(III) stress. 100 µM Cr(VI) was most toxic of all the treatments in terms of root and leaf growth and oxidative stress. 50 µM Cr(VI) treated roots exhibited high significant increase in superoxide dismutase (SOD), dehydroascorbate reductase (DHAR) and glutathione reductase (GR) (p < 0.01) and significant increases in catalse (CAT), ascorbate peroxidase (APX) and monodehydroascorbate reductase (MDHAR) (p < 0.05). A high increase in ascorbic acid (AA) level was seen in roots of 50 µM Cr(VI) treated plants in comparison with control. Levels of reduced glutathione (GSH) showed a varied and complex response in all the treatments in both plant parts. GSH/GSSG ratio was not affected by Cr(III) treatment in leaves, in contrast, roots exhibited significant reduction in the ratio. Results indicate that GSH depletion increased sensitivity to oxidative stress (Cr(VI) roots and leaves and Cr(III) 100 µM roots) and AA in tandem with APX compensated for GSH depletion by acting directly on H2O2 and the mechanism of defensive response in roots as well as leaves varied in its degree and effectiveness due to the concentration dependant differences observed in translocation of the element itself, reactive oxygen species (ROS) generation and enzyme inhibition based on the oxidation state supplied to the plants.  相似文献

Virus-induced gene silencing (VIGS) is potentially an attractive reverse-genetics tool for studies of plant gene function, but whether it is effective in silencing mineral nutritional-related genes in roots has not been demonstrated. Here we report on an efficient VIGS system that functions in tomato roots using a modified viral satellite DNA (DNAmβ) associated with Tomato yellow leaf curl China virus (TYLCCNV). A cDNA fragment of the ferric chelate reductase gene (FRO1) from tomato was inserted into the DNAmβ vector. Tomato roots agro-inoculated with DNAmβ carrying both a fragment of FRO1 and TYLCCNV used as a helper virus exhibited a significant reduction at the FRO1 mRNA level. As a consequence, ferric chelate reductase activity, as determined by visualization of the pink FeBPDS3 complex was significantly decreased. Our results clearly demonstrated that VIGS system can be employed to investigate gene function associated with plant nutrient uptake in roots.  相似文献

Study on the Relationship between Soil Selenium and Plant Selenium Uptake   总被引:13,自引:0,他引:13  
Various extraction methods have been used to determine selenium (Se) concentrations in soils and plants in the second seleniferous regions of China. Our results show tea Se contents in the study area range from 1.009 to 2.6 mg/kg, which reveal that the tea areas in Ziyang County are in seleniferous regions. The four extraction methods evaluated in this study provide different information concerning soil and plant Se levels. The quality control/quality assurance program for this project indicated there is excellent agreement between total soil Se and extractable Se. For example, phosphate extractable Se results from the field investigation and greenhouse study were found to be highly correlated (R 2 > 0.91) by linear regression analyses. Results from rye seedling experiments further show phosphate extractable Se has significant correlations with plant Se uptake and that a 0.1 M solution of KH2PO4 can be used as the extractant of soil available Se. In the acid soil, the Brassica campestris yield could be significantly reduced when the content of Se6+–Se ≥ 0.5 mg/kg, and the influence on the yield was not as obvious when the content of Se6+–Se reached up to 2.0 mg/kg. The uptake by Brassica campestris of Se6+–Se is higher than that of Se4+–Se. The main factors influencing the biological availability of soil Se, in order of their importance are CaCO3, the presence of silt grains, organic matter and the presence of clay grains. pH could affect KH2PO4 extractable Se through CaCO3.  相似文献

Thirty-day-old seedlings of tomato (Lycopersicon esculentum Mill.) were treated with various Hg concentrations (0, 10, and 50 μM) for up to 20 days, and the hypothesis that Hg induces oxidative stress leading to the reduction of biomass and chlorophyll content in leaves was examined. The accumulation of Hg in seedlings increased with external Hg concentration and exposure time, and Hg content in roots exposed to 50 μM Hg for 20 days was about 27-fold higher than that in shoots. Furthermore, Hg exposure not only reduced biomass and chlorophyll levels in leaves but also caused an overall increase of endogenous H2O2, lipid peroxidation products (malondialdehyde), and antioxidant emzymes activities such as superoxide dismutase, catalase, and peroxidase in leaves and roots. Our results suggest that the suppression of growth and the reduction of chlorophyll levels in tomato seedlings exposed to toxic Hg levels may be caused by an enhanced production of active oxygen species and subsequent high lipid peroxidation.  相似文献

Effects of zinc (12–180 μM) alone and in mixtures with 12 μM Cd on metal accumulation, dry masses of roots and shoots, root respiration rate, variable to maximum fluorescence ratio (FV/FM), and content of photosynthetic pigments were studied in hydroponically cultivated chamomile (Matricaria recutita) plants. The content of Zn in roots and shoots increased with the increasing external Zn concentration and its accumulation in the roots was higher than that in the shoots. While at lower Zn concentrations (12 and 60 μM) the presence of 12 μM Cd decreased Zn accumulation in the roots, treatment with 120 and 180 μM Zn together with 12 μM Cd caused enhancement of Zn content in the root. Presence of Zn (12–120 μM) decreased Cd accumulation in roots. On the other hand, Cd content in the shoots of plants treated with Zn + Cd exceeded that in the plants treated only with 12 μM Cd. Only higher Zn concentrations (120 and 180 μM) and Zn + Cd mixtures negatively influenced dry mass, chlorophyll (Chl) and carotenoid content, FV/FM and root respiration rate. Chl b was reduced to a higher extent than Chl a.  相似文献
