该研究采用脂类组学方法,系统地研究了烟草种子成熟过程中膜脂含量及组成比例的变化规律。结果表明:(1)构成叶绿体和类囊体膜的重要脂类质体膜脂的含量及其在总膜脂中的组成比例,在种子成熟的整个过程中保持下降趋势;而构成细胞膜的重要脂类质外体膜脂含量在种子成熟前期则下降显著,在授粉21 d后基本保持不变。(2)总膜脂含量的变化规律与质体膜脂类似,但在授粉后第29天后含量却达到稳定状态。(3)因油脂在种子成熟过程中不断积累,且化学结构与膜脂相似,质体膜脂含量的降低可能与种子成熟过程中种子对油脂累积的持续需求以及对叶绿体及类囊体的需求降低有关。(4)质外体膜脂含量在授粉21 d后基本保持不变的原因,可能是由于脂质外体膜脂是细胞膜组成的主要膜脂,细胞膜在种子成熟以及成熟种子萌发过程中均发挥重要作用,因此质外体膜脂只在种子成熟的前期有部分转化为油脂。  相似文献

The C‐4 salt marsh grass, Spartina patens , thrives in the upper portion of the marsh where soil salinities may be equal to coastal seawater. Spartina patens was grown in hydroponic culture in a greenhouse at 0, 340, and 510 m M NaCl, and measured for growth, tissue cation content, and root plasma membrane (PM) lipid composition. From 0 to 340 and 510 m M , the shoot growth decreased, but root growth was not affected. The Na+ content increased in both shoots and roots when plants were grown in salt, while the shoots had a decreased K+ content and the roots had a decreased Ca2+ content. Spartina patens root plasma membrane was isolated with an aqueous polymer two‐phase system. The purity of the plasma membrane was verified with cytochemical tests on membrane enzyme markers. Plasma membrane lipids were stable relative to the membrane protein content. Molar percentages of sterols (including free sterols) and phospholipid decreased with increasing salinity. However, glycolipid showed a statistically significant increase in the total lipid as salinity in the medium was increased from 0 to 510 m M . Even at a salinity of 510 m M , the plasma membrane sterol/phospholipid ratio was unaffected by NaCl. When the plants were grown in NaCl media, the plasma membrane had a decreased phosphatidylcholine (PC) and phosphatidylethanolamine (PE) content, but the PC/PE ratios were not affected. The plasma membrane molar percentage of sitosterol in total free sterol increased when plants were grown in salt media. The predominant membrane fatty acids were C11 and C14, and the major unsaturated one was C14:1. An increase in growth medium salinity resulted in a decreased root plasma membrane fluidity.  相似文献

从不同干旱胁迫时间下的小麦(Triticum aestivum L.)叶片中提取微粒体膜,测定活性氧自由基变化和膜伤害程度。结果表明,随着干旱胁迫时间的延长发生以下变化:(1)O_2~-和H_2O_2的产生速率在一定时间内逐渐加快然后下降;(2)膜脂过氧化产物丙二醛(MDA)的含量逐渐增加;(3)微粒体膜上的SOD活性逐渐下降;(4)—SH基的含量在一定时间内逐渐增多以后下降;(5)与膜伤害程度有关的生理指标如根系电导率、叶片水势及其含水量等均有变化。  相似文献

The microsome membrane of wheat ( Triticum aestivum L. ) leaves under different time of drought stress was purified to determine the active oxygen free radicals generated and their severity of injury to the membrane. During the drought stress the rate of O2- and H2O2 generation increased for a certain time and then decreased. The concentration of MDA increased continuously. The activity of SOD decreased gradually. The concentration of —SH group and MDA increased from the beginning until the 36th hour under drought stress, thereafter MDA content continued to increase while the —SH content decreased. Several physiological indexes related to membrane injury, such as root electroconductivity, leaf water potential and water content were all affected accordingly.  相似文献

以人工栽培的栀子为材料,研究了其叶片低温半致死温度及膜保护系统随气温下降的变化。结果表明,低温半致死温度降低的同时,SOD、POX活性增强,Vc含量增加,使膜保护系统清除自由基的能力加强,O2-水平降低,MDA含量下降。膜保护系统的适应性变化导致低温半致死温度的下降是抗寒性提高的基础。  相似文献

以尖果沙枣1年生实生苗为材料,研究了自然干旱时不同土壤相对含水量对幼苗叶片细胞质膜相对透性、叶片相对含水量、根系活力、光合色素含量和光合参数等指标的影响.结果表明:土壤相对含水量从70%(CK)降到40%时,幼苗根系活力和净光合速率均逐渐上升并达到最大值,分别为1178μg.g-1.h-1和21.9μmol.m-2.s-1;光合色素含量稳步上升;蒸腾速率和水分利用效率均保持稳定;叶片细胞质膜相对透性保持较低水平.土壤相对含水量从40%降到20%时,幼苗叶片相对含水量仍在50%以上,叶片细胞质膜相对透性仍保持较低水平;根系活力和光合色素含量仍较高;但其他光合参数开始缓慢下降.土壤相对含水量从10%降到5%时,幼苗叶片细胞质膜相对透性急剧上升;叶片相对含水量、根系活力、总叶绿素含量、光合参数均极显著下降;而土壤相对含水量为10%时幼苗表现出最高的水分利用效率.尖果沙枣土壤相对含水量最好控制在40%~50%,其1年生实生苗的永久萎蔫系数为4.3%(土壤相对含水量).  相似文献

Multiple methods have been developed to isolate the intestinal microvillus membrane and facilitate the study of its composition and function. Variations in membrane composition and fluidity may result from different preparative techniques. This study shows that the use of MgCl2 and/or KSCN in vesicle preparation alters phospholipid and protein composition of the membrane compared to CaCl2 precipitation. The use of MgCl2 in membrane preparation increased phosphatidylethanolamine and decreased phosphatidylinositol content. The use of KSCN in membrane preparation decreased the protein content. The structural changes seen with the use MgCl2 alone are accompanied by an increase in both static and dynamic membrane fluidity. These results suggest that different methods of membrane vesicle preparation affect membrane phospholipid and protein content as well as membrane fluidity.  相似文献

Dietary control of diacylphosphatidylethanolamine species in brain   总被引:2,自引:0,他引:2  
The content and composition of the brain diacylphosphatidylethanolamine species was examined in response to dietary fat intake. Synaptic plasma membrane and microsomal membrane subcellular fractions contain phosphatidylethanolamine species profiles that respond differently to modulation by diet fat. The microsomal content of individual phosphatidylethanolamine species was most responsive to diet treatment and to addition of cholesterol to the diet. Feeding fish oil or linseed oil diets resulted in an increased membrane content of phosphatidylethanolamine species containing six double bonds for both microsomal and synaptic plasma membranes, compared with soya-bean oil- or sunflower oil-fed animals. The 22:5(n - 6) content present in phosphatidylethanolamine species of linseed oil and fish oil-fed animals was also reduced. For microsomal membranes, increase in dietary 18:3(n - 3) resulted in an increased content of phosphatidylethanolamine species containing one double bond. Addition of cholesterol to linseed oil or fish oil diets decreased the microsomal membrane content of phosphatidylethanolamine species containing six double bonds and increased the membrane content of species containing one double bond. For synaptic plasma membrane, addition of cholesterol to linseed oil and fish oil diets increased membrane content of species containing six double bonds. Fish oil-fed animals exhibited a decreased content of species containing a single double bond. The implications of the diet-induced changes in phospholipid species content and composition are discussed.  相似文献

The (Na+,K+)-ATPase ATP hydrolyzing activity from rabbit kidney medulla basolateral membrane vesicles was studied as a function of the cholesterol content of the basolateral membranes. The cholesterol content of the membranes was modified by incubation with phospholipid vesicles. When the cholesterol content was increased above that found in the native membrane, the (Na+,K+)-ATPase ATP hydrolyzing activity was inhibited. When the cholesterol content was decreased from that found in the native membranes, the (Na+,K+)-ATPase ATP hydrolyzing activity was inhibited. Analogous effects were found with the K+-activated phosphatase activity of the same membrane vesicles. Therefore, at low cholesterol contents, cholesterol was stimulatory, and at high cholesterol contents, cholesterol was inhibitory. The structural specificity of this effect was tested by introducing lanosterol and ergosterol as 50% of the membrane sterol. Ergosterol was the least effective at supporting (Na+,K+)-ATPase ATP hydrolyzing activity, while lanosterol was more effective, but still not as effective as cholesterol.  相似文献

采用盆栽控水法,研究了珍稀濒危植物堇叶紫金牛(Ardisia violacea)在持续干旱条件下的生理响应。随着持续干旱时间的延长,堇叶紫金牛应对持续干旱的阶段可分为适应期、轻度干旱期、中度干旱期和重度干旱期。在适应期和轻度干旱期,堇叶紫金牛叶片游离脯氨酸和可溶性糖含量稳定在一个较低水平,可溶性蛋白质含量先下降后快速上升,细胞膜系统和抗氧化酶系统能主动进行生理调节;中度干旱期,丙二醛(MDA)含量和质膜相对透性迅速升高,细胞膜系统受损加剧,游离脯氨酸、可溶性糖含量均急剧增加,对抵御干旱起到重要的渗透调节作用。在轻度干旱期和中度干旱期,光合色素中叶绿素a和叶绿素b含量显著提高,以抵抗干旱胁迫。重度干旱期,细胞膜系统、抗氧化酶SOD、游离脯氨酸和可溶性糖含量上升,但MDA略微下降,这时可能达到植物耐受干旱的极限,不再发生膜脂过氧化作用。综上表明,堇叶紫金牛具有较强的耐旱性,RWC为49.94%是细胞膜系统、抗氧化酶系统和渗透调节物质含量变化的拐点,渗透调节和抗氧化酶系统的主动适应是其耐旱的主要机制。  相似文献

杂交稻幼苗经聚乙二醇(PGE4000)渗透胁迫(-0.95MPa)处理,幼苗含水量及相对含水量下降,游离脯氨酸和膜脂过氧化产物丙二醛(MDA)含量上升,质膜透性增大。随PEG渗透胁迫时间延长,幼苗膜脂饱和脂肪酸含量逐渐增加,不饱和脂肪酸含量降低,不饱和脂肪酸指数(IUFA)减少。脯氨酸累积与MDA增长及膜透性加大呈正相关性,与膜脂脂肪酸不饱和度呈负相关性。讨论了游离脯氨酸累积与细胞透性的相关性,以  相似文献

本文分析了菠菜叶片失水萎蔫时叶绿索、相对含水量,膜透性及膜脂过氧化产物丙二醛的变化结果表明:叶片萎蔫衰老时随着相对含水量的降低,叶绿索含量减少,膜透性和丙二醛含量增大;并且随萎蔫程度加探,膜透性与丙二醛含量也明显增加。这说明菠菜叶片失水萎蔫与膜结构完整性的破坏有密切的关系。  相似文献

以不同浓度的铝处理水稻幼苗 ,研究铝对水稻幼苗的生理生化效应。结果表明 ,5 0 μmol·L 1的铝对水稻幼苗生长有促进作用 ,高于 5 0 μmol·L 1后 ,随着铝浓度的增加 ,植株生长明显受到抑制 ,株高、根长、鲜重和干重均下降 ;叶片叶绿素含量和SOD活性下降 ,POD活性先上升后下降 ,细胞膜透性增大。表明高浓度铝的毒害 ,导致体内保护酶活性受到抑制 ,膜系统受到伤害 ,从而影响水稻幼苗的生长。  相似文献

1. The effect of different dietary fat intake on the lipid composition and fluidity of microsomal membranes as well as in the enzymatic activity of the Ca2+-ATPase from chick breast muscle was investigated. 2. When a standard diet was supplemented with 10% sunflower seed oil, an increase in the relative amounts of unsaturated fatty acids and membrane fluidity and a decrease in the cholesterol content was observed. 3. The presence of 6% cholesterol in the diet does not modify the fatty acid composition and the fluidity of the membrane but increased, in a low extension, the cholesterol content. 4. The provision of the sunflower seed oil-rich diet supplemented with cholesterol just 48 hr before death promoted an increase in the relative amounts of unsaturated fatty acids and cholesterol content whereas the membrane fluidity decreased in a significant extent. 5. Despite that dietary lipids gave rise in some cases to changes in lipid composition and in the physical state of the microsomal membrane, neither the Ca2+ uptake capacity nor the ATPase activity were significantly affected.  相似文献

以蔗糖梯度法纯化的菊花 (Dendranthemamorifolium (Ramat.)Tzvel.)愈伤组织质膜微囊为材料 ,研究了声波刺激下质膜膜脂代谢和物理状态的变化。结果表明 ,一定强度 (10 0dB)和频率 (10 0 0Hz)的声波刺激使质膜磷酸二脂含量和二苯己三烯 (DPH)荧光偏振值降低 ,质膜光散射值、MC5 40荧光强度和磷酸单脂含量增加。表明一定强度和频率的声波刺激使质膜变疏松 ,膜的流动性增加 ,膜表面电荷密度和疏水性降低 ,膜脂合成代谢增加 ,分解代谢减弱。由此可见 ,膜脂物理状态和膜脂代谢对声波刺激极为敏感。  相似文献

Considering membranes and membrane components as possible pacemakers of the main processes taking place inside mitochondria, changes in phospholipids or fatty acids could play a central role linking different mechanisms involved in cumulative damage to cell molecules and dysfunction during periods of high stress, such as rapid growth and aging. Changes affecting either lipid class or fatty acid compositions could affect phospholipid and membrane properties and alter mitochondrial function and cell viability. In the present study, mitochondrial oxidative status and mitochondrial membrane phospholipid compositions were analyzed throughout the life-cycle of zebrafish. TBARS content significantly increased in 18-month-old fish while aconitase activity decreased in 24-month-old fish, which have been related with oxidative damage to molecules. Mitochondria-specific superoxide dismutase decreased in 24-month-old animals although this change was not statistically significant. Age affected both mitochondrial phospholipid content and the peroxidation index of most phospholipid classes suggesting that oxidative damage to mitochondrial lipids was occurring.  相似文献

用荧光探剂ANS对抗旱性不同的甘蔗品种在水分胁迫下叶片线粒体膜流动性的变化进行的研究表明,水分胁迫降低了线粒体膜的流动性,抗旱性强的甘蔗品种Co 617和F.Y.79-9的下降幅度分别小于抗旱性弱的Co 740和M.T.77-208;水分胁迫下线粒体膜流动性的下降与膜脂过氧化产物丙二醛含量的增加有密切关系。外源自由基处理试验也表明,甘蔗叶片线粒体膜流动性的下降与膜脂过氧化作用有关。  相似文献

Liposomes prepared with 25-hydroxycholesterol and egg phosphatidylcholine (PC) were incubated with bovine arterial smooth muscle cells for 8 h at 37 degrees C. Cells incubated in the absence of liposomes or with liposomes containing cholesterol and PC were used as controls. The results indicated that calcium accumulated in the smooth muscle cells incubated in the presence of 25-hydroxycholesterol containing liposomes in an amount proportional to the time of incubation. The calcium accumulation, as indicated by kinetic analysis, resulted from an increased compartment size. (Ca(2+)+Mg2+)-ATPase exhibited decreased activity after pretreatment with 25-hydroxycholesterol containing liposomes and the increased intracellular calcium content was directly proportional to the decreased (Ca(2+) + Mg2+)-ATPase activity. When lipids in the cell membrane were examined, a failure to change the cholesterol/phospholipids ratio in the membrane was noted. The 25-hydroxycholesterol content in the membrane determined by HPLC did not increase. An increase in sphingomyelin and a decrease in phosphatidylethanolamine and acidic phospholipids in the membrane was noted. We suggest that the accumulation of intracellular calcium comes from both an increase of calcium influx and a decrease of (Ca(2+) + Mg2+)-ATPase activity, which may be the consequence of changes in membrane phospholipid composition.  相似文献

The modulation of immunoglobulin on the surface of rabbit B lymphocytes by goat antibodies with specificity for rabbit surface membrane immunoglobulin or by such goat antibodies covalently linked to Sepharose was studied in relation to the proliferative response to these agents. Although the induction of DNA synthesis was greater in the presence of Sepharose-linked antibody than in the presence of free antibody, modulation of surface membrane immunoglobulin was induced with free but not with Sepharose-linked antibody. Thus, in the presence of free antibody the surface membrane immunoglobulin content of cells was rapidly decreased and remained at a low level throughout the culture period, whereas the surface immunoglobulin content of cells incubated with Sepharose antibody was essentially unaltered. The surface immunoglobulin lost from cells incubated with free goat antibodies reappeared slowly upon further incubation in culture medium devoid of antibody, and such reappearance of rabbit surface membrane immunoglobulin was inhibited by puromycin. Upon culture with Sepharose-linked antibody the surface membrane immunoglobulin content of B cells was unaffected by puromycin. This result was interpreted as indicating that surface membrane immunoglobulin loss followed by reappearance does not occur. Lastly, the linkage of surface membrane immunoglobulin to cytoskeletal elements induced by free antibody was not induced by Sepharose-linked antibody as judged from differences in detergent solubilization characteristics. Possible mechanisms to account for these differences in surface membrane immunoglobulin modulation as they relate to the proliferative response are considered.  相似文献
