Diphenylmethylation of carbohydrate hydroxyl groups may be effected by the thermal reaction with diazo(diphenyl)methane in the absence of catalysts. Migration of the labile ester groups of methyl 2,3,4-tri-O-acetyl-α-d-glucopyranoside and 3-O-benzoyl-1,2-O-isopropylidene-α-d-glucofuranose does not occur during diphenylmethylation by this procedure. The diphenylmethyl group may be readily removed by catalytic hydrogenolysis, and is sufficiently acid-stable to enable the selective hydrolysis of acetal groups. Its use as an O-4 protecting-group and as a non-participating O-2 protecting-group in α-glycoside synthesis has been demonstrated in syntheses of methyl 2,3,6-tri-O-methyl-α-d-glucopyranoside and kojibiose octa-acetate, respectively.  相似文献

2-Methyl-[3,6-di-O-acetyl-2-deoxy-4-O-(2,3,4,6-tetra-O-acetyl-β-d-galactopyranosyl)-α-d-glucopyrano]-[2,1-d]-2-oxazoline (4) was prepared from 2-acetamido-3,6-di-O-acetyl-2-deoxy-4-O-(2,3,4,6-tetra-O-acetyl-β-d-galactopyranosyl)-α-d- glucopyranosyl chloride. Condensation of 3,4:5,6-di-O-isopropylidene-d-mannose dimethyl acetal with 4 in the presence of a catalytic amount of p-toluenesulfonic acid afforded O-(2,3,4,6-tetra-O-acetyl-β-d-galactopyranosyl)-(1 → 4)-O-(2-acetamido-3,6-di-O-acetyl-2-deoxy-β-d-glucopyranosyl)-(1 → 2)-3,4:5,6-di-O-isopropylidene-d-mannose dimethyl acetal (6) in 8.6% yield. Catalytic deacetylation of 6 with sodium methoxide, followed by hydrolysis with dilute sulfuric acid, gave O-β-d-galactopyranosyl-(1 → 4)-O-(2-acetamido-2-deoxy-β-d-glucopyranosyl)-(1 → 2)-d-mannose (7). The inhibitory activities of 7 and related sugars against the hemagglutinating activities of various lectins were assayed, and 7 was found to be a good inhibitor against Phaseolus vulgaris hemagglutinin.  相似文献

Oxidation with the dimethyl sulfoxide-acetic anhydride reagent of methyl 2-O-acetyl-4,6-O-benzylidene-α-d-mannopyranoside, obtained in quantitative yield from the corresponding 4,6-benzylidene acetal by stereoselective opening of a 2,3-orthoester, led in good yield to methyl 2-O-acetyl-4,6-O-benzylidene-α-d-arabino-hexopyranosid-3-ulose, which was reduced with either sodium borohydride or sodium borodeuteride into a methyl 4,6-O-benzylidene-α-d-altropyranoside or its 3-2H derivative. A sequence involving a C-6 halogenation-dehydrohalogenation followed by catalytic hydrogenation of the resulting methyl 6-deoxy-α-d-arabino-hex-5-enopyranoside gave methyl 6-deoxy-β-l-galactopyranoside (methyl β-l-fucopyranoside) and then α-l-fucose, with an overall yield of 24% with respect to the starting methyl α-d-mannopyranoside.  相似文献

Treatment of methyl 2,3-anhydro-5-deoxy-α-d-ribofuranoside with lithium dimethyl cuprate gave methyl 2,5-dideoxy-2-C-methyl-α-d-arabinofuranoside (54% yield) and methyl 3,5-dideoxy-3-C-methyl-α-d-xylofuranoside (10%). The former was converted into its 3-O-acetyl and 3-O-benzyl derivatives, which, upon acid hydrolysis, afforded 3-O-acetyl- and 3-O-benzyl-2,5-dideoxy-2-C-methyl-d-arabinofuranose in 60–75% overall yield. Treatment of the 3-O-benzyl compound with ethanethiol in the presence of trifluoromethanesulfonic acid afforded 3-O-benzyl-2,5-dideoxy-2-C-methyl-d-arabinose diethyl dithioacetal (20%) and ethyl 3-O-benzyl-2,5-dideoxy-2-C-methyl-1-thio-α-d-arabinoside (73%). The former, which was also available from the latter by equilibration in acidic ethanethiol, was acetylated at O-4 and the product converted into the corresponding dimethyl acetal (85% overall yield). This compound was, after debenzylation, hydrolyzed with acid, to provide 4-O-acetyl-2,5-dideoxy-2-C-methyl-d-arabinose in 70% overall yield.  相似文献

Conversion of benzyl αβ-D-galactofuranoside into the 5,6-O-[α-(dimethyl-amino)benzylidene] derivative, followed by acetylation of HO-2 and HO-3, and selective ring opening or the acetal, gave benzyl 2,3-di-O-acetyl-6-O-benzoyl-αβ-D-galactofuranoside(4). Condensation of 3,4:5,6-di-O-isopropylidene-D-mannose dimethyl acetal with 2-methyl-(3,4,6-tri-O-acetyl- 1,2-dideoxy-α-D-glucopyrano)-[2′, 1′:4,5]-2-oxazoline in the presence of a catalytic amount of p-toluenesulfonic acid afforded crystalline 2-O-(2-acetamido-3,4,6-tri-O-acetyl-2-deoxy-β-D-glucopyranosyl)-3,4:5,6-di-O-isopropylidene-D-mannose dimethyl acetal (3) in 25% yield. Catalytic deacetylation of 3 with sodium methoxide, followed by hydrolysis with dilute sulfuric acid, gave 2-O-(2-acetamido-2-deoxy-α-D-glucopyranosyl)-D-mannose (4). Treatment of 3 with boiling 0.5% methanolic hydrogen chloride under reflux gave methyl 2-O-(2-acetamido-2-deoxy-β-D-glucopyranosyl)-α-D-mannopyranoside (5) and methyl 2-O-(2-acetamido-2-deoxy-β-D-glucopyranosyl)-α-D-mannofuranoside (6). The inhibitory activities of 4, 5, and 6 against the hemagglutinating and mitogenic activities of Lens culinaris and Pisum sativum lectins and concanavalin A were assayed. From the results of these hapten inhibition studies, subtle differences of specificity between these D-mannose-specific lectins were confirmed.  相似文献

Benzyl 2-O-acetyl-4,6-O-benzylidene-3-O-(2,3,4-tri-O-acetyl-α-l-rhamnopyranosyl)-β-d-galactopyranoside (11) has been synthesised by two routes. Partial deacetylation of 11 and then acid hydrolysis yielded benzyl 2-O-acetyl-3-O-α-l-rhamnopyranosyl-β-d-galactopyranoside, catalytic hydrogenolysis of which gave the first title compound in excellent yield. Benzyl 4,6-O-benzylidene-3-O-α-l-rhamnopyranosyl-β-d-galactopyranoside was benzylated, and hydrogenolysis (LiAlH4-AlCl3) of the product gave the disaccharide derivative 16 with only HO-6 unsubstituted. Acetylation of 16 followed by catalytic hydrogenolysis gave the crystalline, second title compound. As model compounds for comparative n.m.r. studies, 2-O-, 3-O-, and 6-O-acetyl-d-galactose were also synthesised.  相似文献

Condensation of dimeric 3,4,6-tri-O-acetyl-2-deoxy-2-nitroso-α-D-glucopyranosyl chloride (1) with 1,2-O-isopropylidene-α-D-glucofuranurono-6,3-lactone (2) gave 1,2-O-isopropylidene-5-O-(3,4,6-tri-O-acetyl-2-deoxy-2-hydroxyimino-α-D-arabino-hexopyranosyl)-α-D-glucofuranurono-6,3-lactone (3). Benzoylation of the hydroxyimino group with benzoyl cyanide in acetonitrile gave 1,2-O-isopropylidene-5-O-(3,4,6-tri-O-acetyl-2-benzoyloxyimino-2-deoxy-α-D-arabino-hexopyranosyl)-α-D-glucofuranurono-6,3-lactone (4). Compound 4 was reduced with borane in tetrahydrofuran, yielding 5-O-(2-amino-2-deoxy-α-D-glucopyranosyl)-1,2-O-isopropylidene-α-D-glucofuranose (5), which was isolated as the crystalline N-acetyl derivative (6). After removal of the isopropylidene acetal, the pure, crystalline title compound (10) was obtained.  相似文献

The koenigs-Knorr glycosylation of 4,6-O-ethylidene-1,2-O-isopropylidene-3-O-(2,3-O-isopropylidene-α-l-rhamnopyranosyl)-α-d-galactopyranose (3) by 4,6-di-O-acetyl-2,3-O-carbonyl-α-d-mannopyranosyl bromide (10), as well as Helferich glycosylations of 3 by tetra-O-acetyl-α-d-mannopyranosyl and -α-d-glucopyranosyl bromides, proceeded smoothly to give high yields of trisaccharide derivatives (12, 16, and 17). An efficient procedure for the transformation of 12, 16, and 17 into the α-deca-acetates of the respective trisaccharides has been developed. Zemplén de-acetylation then afforded the title trisaccharides in yields of 53, 52, and 62 %, respectively, from 3. A new route to 1,4,6-tri-O-acetyl-2,3-O-carbonyl-α-d-mannopyranose is suggested.  相似文献

Methyl 2-acetamido-5,6-di-O-benzyl-2-deoxy-β-d-glucofuranoside (11) was obtained in six steps from the known methyl 3-O-allyl-2-benzamido-2-deoxy-5,6-O-isopropylidene-β-d-glucofuranoside. Mild acid hydrolysis, followed by benzylation gave the 5,6-dibenzyl ether. The benzamido group was exchanged for an acetamido group by strong alkaline hydrolysis, followed by N-acetylation, and the allyl group was isomerized into a 1-propenyl group that was hydrolyzed with mercuric chloride. Treatment of 11 with l-α-chloropropionic acid and with diazomethabe gave methyl 2-acetamido-5,6-di-O-benzyl-2-deoxy-3-O-[d-1-(methoxycarbonyl)ethyl]-β-d-glucofuranoside which formed on mercaptolysis the internal ester 16, further converted into 2-acetamido-4-O-acetyl-5,6-di-O-benzyl-2-deoxy-3-O-[d-1-(methoxycarbonyl)ethyl]-d-glucose diethyl dithioacetal (18) by alkaline treatment followed by esterification with diazomethane and acetylation. Attempts to remove the O-acetyl group of the corresponding dimethyl acetal 20 with sodium methoxide in mild conditions were not successful.  相似文献

Condensation of 3-O-benzyl-D-allose with acetaldehyde forms a complex mixture from which potentially useful mono- and di-O-ethylidene derivatives were isolated and identified. Compounds isolated and identified after conversion of unsubstituted hydroxyl groups into the corresponding acetates included 1,2-di-O-acetyl-3-O-benzyl-4,6-O-ethylidene-β-D-allopyranose; 5,6-di-O-acetyl-3-O-benzyl-1,2-O-(R)-ethylidene-α-D-allofuranose; and two 3-O-benzyl-1,2:5,6-di-O-ethylidene-α-D-allofuranoses, both having the R configuration in the 1,2-O-ethylidene ring. Furanose and pyranose conformations were determined by n.m.r. analysis, and the location and configuration of each acetal ring was established. The benzyl ether group in the furanose derivatives was removed by catalytic hydrogenation with subsequent formation of 3-O-acetyl analogs.  相似文献

3,4-Di-O-acetyl-2-O-benzyl-α-d-xylopyranosyl bromide (1) reacts with methyl 2,3-anhydro-α-d-ribopyranoside (2) to afford, in high yield, methyl 2,3-anhydro-4-O- (3,4-di-O-acetyl-2-O-benzyl-β-d-xylopyranosyl)-β-d-ribopyranoside (3). Deacetylation of 3 gave 4, which reacted with 2,3,4-tri-O-acetyl-α-d-xylopyranosyl bromide to give the branched tetrasaccharide derivative 5, which, in turn, was converted by a series or conventional reactions into methyl 4-O-[3,4-di-O-(β-d-xylopyranosyl)-β-d- xylopyranosyl]-β-d-xylopyranoside. The reaction of 1 with its hydrolysis product gave 3,4-di-O-acetyl-2-O-benzyl-α-d-xylopyranosyl 3,4-di-O-acetyl-2-O-benzyl-β-d-xylopyranoside, which was also isolated after the reaction of 1 with 2.  相似文献

Two new acylated triterpenoid saponins named pendulaosides A and B as well as the known phenolic compounds methyl gallate, gallic acid, 1,2,3,6-tera-O-galloyl-β-d-glucose and 1,2,3,4,6-penta-O-galloyl-β-d-glucose, were isolated from the seeds of Harpullia pendula. The structures of pendulaosides A and B were determined using extensive 1D and 2D NMR analysis and mass spectrometry as well as acid hydrolysis, as 3-O-β-d-glucopyranosyl-(1→2)-[α-L-arabinofuranosyl-(1→3)]-β-d-glucuronopyranosyl-22-O-angeloyl-3β,16α,22α,24β,28-pentahydroxylolean-12-ene and 3-O-β-d-glucopyranosyl-(1→2)-[α-L-arabinofuranosyl-(1→3)]-β-d-glucuronopyranosyl-16-O-(2-methylbutyroyl)-3β,16α,22α,24β,28-pentahydroxylolean-12-ene, respectively. To the best of our knowledge the two triterpene parts 22-O-angeloyl-3β,16α,22α,24β,28-pentahydroxylolean-12-ene and16-O-(2-methylbutyroyl)-3β,16α,22α,24β,28-pentahydroxylolean-12-ene have never been characterized before. The two isolated saponins were assayed for their in-vitro cytotoxic activity against the three human tumor cell lines HepG2, MCF7 and PC3. The results showed that pendulaoside A exhibited moderate activity on PC3 cell line with IC50value equal to 13.0 μM and weak activity on HepG2 cell line with IC50 value equal to 41.0 μM. Pendulaoside B proved to be inactive against the three used cell lines.  相似文献

Solutions of O-acetyl-α-glycosyl bromide derivatives of d-glucose, cellobiose, and lactose in hexamethylphosphoramide were converted into corresponding β-chlorides at room temperature by the action of lithium chloride. At 3:1 mM ratios of chloride ion to glycose, 5–10% w/v solutions of glycosyl bromide formed α- and β-chlorides in ratios of (or greater than) 1:19 within 2–13 min and produced crystalline β-chlorides in 70–80% yields. Anomeric compositions were determined by n.m.r. spectroscopy in hexamethylphosphoramide. Older methods of preparing 1,2-trans-O-acetylgIycosyl chlorides, with aluminum chloride or titanium tetrachloride, gave the α- and β-cellobiosyl and -Iactosyl chlorides in ratios that varied from 2:3 to 1:4 and reached 85–95% levels of β-chloride only with β-d-glucose pentaacetate. When hydrolyzed under conditions that controlled solution acidity, the β-cellobiosyl and -Iactosyl chlorides each gave 2-hydroxy derivatives in yields that could be varied from 16 to 60%. Hepta-O-acetyl-2-O-methyl-α-cellobiose was prepared to demonstrate how these hydrolysis mixtures can be used to synthesize a 2-O-substituted derivative.  相似文献

The major product obtained on acetonation of d-mannose with a 2-molar excess of isopropenyl methyl (or ethyl) ether is 4,6-O-isopropylidene-α-d-mannopyranose (3a), the product of kinetic acetonation: a larger excess of the reagent leads, to the 2,3:4,6-diisopropylidene acetal (6). The course of the reaction and side-products formed were examined in detail. The 1,2,3-triacetate of 3a was deacetonated to give α-d-mannopyranose 1,2,3-triacetate; similar reactions were performed on the β anomers. The 1-acetate of the diacetal 6 could be selectively deacetonated to give 1-O-acetyl-2,3-O-isopropylidene-α-d-mannopyranose. The reactions provide access to protected derivatives of d-mannose, and partially acylated derivatives, having modes of substitution different from those obtainable by classical acetonation procedures conducted under conditions of thermodynamic control.  相似文献

During the chromatographic separation of 3-S-acetyl-1,2-O-isopropylidene-3-thio-α-d-allofuranose on silica gel, a migration of the acetyl group from S to O was observed to give 6-O-acetyl-1,2-O-isopropylidene-3-thio-α-d-allofuranose, whereas 3-S-acetyl-6-O-benzoyl-1,2-O-isopropylidene-3-thio-α-d-allofuranose gave 5-O-acetyl-6-O-benzoyl-1,2-O-isopropylidene-3-thio-α-d-allofuranose. No acetyl migration was observed, however, in the case of 3-O-acetyl-1,2-O-isopropylidene-α-d-allofuranose.  相似文献

The two purple-membrane glycolipids O-β-d-glucopyranosyl- and O-β-d-galactopyranosyl-(1→6)-O-α-d-mannopyranosyl-(1→2)-O-α-d-glucopyranosyl-(1→1)-2, 3-di-O-phytanyl-sn-glycerol were prepared by coupling O-(2,3,4-tri-O-acetyl-α-d-mannopyranosyl)-(1→2)-O-(3,4,6-tri-O-acetyl-α-d-glucopyranosyl)-(1→1)-2, 3-di-O-phytanyl-sn-glycerol (9) with 2,3,4,6-tetra-O-acetyl-α-d-glucopyranosyl bromide or 2,3,4,6-tetra-O-acetyl-α-d-mannopyranosyl bromide, respectively, followed by deacetylation. The glycolipid sulfate O-(β-d-glucopyranosyl 3-sulfate)-(1→6)-O-α-d-mannopyranosyl-(1→2)-O-α-d-glucopyranosyl-(1→1)-2,3-di-O-phytanyl-sn-glycerol was prepared by coupling of 9 with 2,4,6-tri-O-acetyl-3-O-trichloroethyloxycarbonyl-α-d-glucopyranosyl bromide in the presence of Hg(CN)2/HgBr2 followed by selective removal of the 3?-trichloroethyloxycarbonyl group, sulfation of HO-3?, and deacetylation. The suitably protected key-intermediate 9 could be prepared by two distinct approaches.  相似文献

N-[2-O-(2-Acetamido-2,3-dideoxy-5-thio-d-glucopyranose-3-yl)-d-lactoyl]-l-alanyl-d-isoglutamine, in which the ring-oxygen atom of the sugar moiety in N-acetylmuramoyl-l-alanyl-d-isoglutamine (MDP) has been replaced by sulfur, was synthesized from 2-acetamido-2-deoxy-5-thio-α-d-glucopyranose (1). O-Deacetylation of the acetylated acetal, derived from the methyl α-glycoside of 1 by 4,6-O-isopropylidenation and subsequent acetylation, gave methyl 2-acetamido-2-deoxy-4,6-O-isopropylidene-5-thio-α-d-glucopyranoside (4). Condensation of 4 with l-2-chloropropanoic acid, and subsequent esterification, afforded the corresponding ester, which was converted, viaO-deisopropylidenation, acetylation, and acetolysis, into 2-acetamido-1,4,6-tri-O-acetyl-2-deoxy-3-O-[d-1-(methoxycarbonyl)ethyl]-5-thio-α-d-glucopyranose (12). Coupling of the acid, formed from 12 by hydrolysis, with the methyl ester of l-alanyl-d-isoglutamine, and de-esterification, yielded the title compound.  相似文献

6-O-β-D-Galactofuranosyl-D-galactose was obtained in crystalline form by condensation of 3,5,6-tri-O-acetyl-1,2-O-(1-methoxyethylidene)-α-D-galactofuranose with benzyl 2,3,4-tri-O-benzyl-α-D-galactopyranoside, followed by O-deacetylation and catalytic hydrogenation. The aminocyclitol antibiotic neamine has been chemically modified at the hydroxyl group on C-6 of the 2-deoxystreptamine moiety. The partially acetylated neamine derivatives, 6,3′,4′-tri-O-acetyl- (3) and 5,3′,4′-tri-O-acetyl-1,3,2′,6′-tetra-N-(ethoxycarbonyl)neamine (4), were prepared by random hydrolysis of the 5,6-O-ethoxyethylidene derivative (2), followed by chromatographic purification. Condensation of 4 and 2,3,5-tri-O-benzoyl-d-ribofuranosyl chloride led to 6-O-(β-d-ribofuranosyl)neamine (7). Analogous condensation of 4 with 2,3,4,6-tetra-O-acetyl-α-d-glucopyranosyl bromide or 2,3,4,6-tetra-O-acetyl-α-d-galactopyranosyl bromide afforded the corresponding 6-O-(d-hexopyranosyl)neamines.  相似文献
