Submerged culture was better than solid culture in the production of proteinase and peptidases from Aspergillus oryzae 460. On the contrary, solid culture was better than submerged culture in the production of α-amylase, carboxymethyl cellulase, and pectinlyase from the same fungus.The soy souce mash (moromi) made with the enzyme preparation from submerged culture was highly viscous and the soy sauce produced was characteristic in low contents of alcohol and reducing sugar, low pH value, and less aroma. Soy sauce made with the enzyme preparation from solid culture was superior on these points to that from submerged culture.Wheat bran was best as the raw material for the enzyme preparation in easy koji making, large amount produced, and low cost.In enzyme production from a solid culture, addition of urea (0.8% to wheat bran) nearly doubled the leucine aminopeptidase for Leu-Gly-Gly. The incubation period was reduced to 30 to 40 h from 50 to 60 h using germinated spores and moisture-controlled culture with forced aeration.  相似文献

Large increases in sanguinarine production were observed in suspension cultured Eschscholtzia californica cells treated with elicitors prepared from yeast extract, Colletotrichum lindemuthianum and Verticillum dahliae. Compounded silicone fluid, an ideal accumulation phase for the two-phase culture of E. californica, accumulated a large amount of sanguinarine produced in a specific manner and increased sanguinarine production. Elicitation in combination with two-phase culture additionally increased net sanguinarine production, as well as the sanguinarine concentration in the accumulation phase.  相似文献

Pigment production by the fungus Monascus sp. was studied to determine why Monascus sp. provides more pigment in solid culture than in submerged culture. Adding a sterilized nonionic polymeric adsorbent resin directly to the growing submerged culture did not enhance the pigment production, thus indicating that pigment extraction is probably not a factor. Monascus cells immobilized in hydrogel were studied and exhibited decreased pigment production as a result of immobilization. This result is thought to be due to diffusional resistance of the pigment through the hydrogel beads. Addition of the adsorbent resin to the immobilized Monascus culture increased both the maximum pigment yield and the production rate above those of the free-cell fermentations. The provision of a support for the mycelium may explain enhanced pigment production by the solid-state culture. These results indicate that product diffusion from immobilized cell systems can be the limiting factor and that in situ extraction is one possible way to circumvent this problem.  相似文献

Twelve fungal cultures isolated from Indian desert soils belonging toAspergillus, Penicillium, Acrophialophora andAlternaria were found to produce both acid and alkaline phosphatases in liquid medium, their amounts varying from culture to culture. Maximum production of these enzymes was observed withA. niger. In general, acid phosphatase activity was much higher as compared to alkaline phosphatase. The optimum incubation period for the production of these enzymes was found to be 14 d and thereafter it started declining. There was a significant and positive correlation between biomass production and acid phosphatase activity but not with alkaline phosphatase.  相似文献

Recombinant Chinese hamster ovary (CHO-K1) cells expressing human soluble thrombomodulin (rsTM) were cultured in a continuous culture system with a fluidized-bed reactor. Cells were grown in a medium containing 1% serum for 10 d, and then cultured in a serum-free medium. The protein production rate increased remarkably in the serum-free culture, with a decrease in the lactate production rate. This suggests that CHO-K1 cells exhibit different physiological characteristics in response to serum removal from the medium, which resulted in a higher rsTM concentration (about 60 mg/l). A procedure for estimating protein productivity was developed using experimental glucose and lactate measurements. In this procedure, cell density was estimated from the glucose consumption rate, and the specific protein (rsTM) production rate was obtained from the ratio of lactate production/glucose consumption (ΔL/ΔG). Since the cell density and protein productivity in repeated batch culture were well estimated, the procedure was applied to continuous culture in a fluidized-bed bioreactor culture. The estimation procedure was also found to be effective in this continuous culture using the models derived from the repeated batch culture.  相似文献

Environmental awareness has led to a serious consideration for biological surfactants and hence non-edible vegetable oils may serve as a substitute carbon source for bio-surfactant production (rhamnolipid) which might be an alternative to complex synthetic surfactants. There are reports of rhamnolipid production from plant based oil giving higher production than that of glucose because of their hydrophobicity and high carbon content. Therefore the contribution of non-edible oil such as Mesua ferrea seed oil could serve as a good carbon source for rhamnolipid production. Moreover the use of rhamnolipid production from non-edible plant based seed oil has not been reported elsewhere. The present work focus on the optimal production of rhamnolipid by considering both micro and macro nutrients and culture conditions using response surface methodology. The study observes that micronutrients play a significant role in rhamnolipid production from Pseudomonas aeruginosa (MTCC 7815). The investigation results with the statistically optimize parameters able to produce a higher rhamnolipid production and this methodology could be used to optimize the nutrients requirements and culture conditions. The present findings would assist in bioremediation of crude oil contaminated ecosystems.  相似文献

A new autoregulator designated as IM2, which induces blue pigment production in Streptomyces sp. MAFF 10-06015, was discovered. The culture conditions developed here for the production of the pigment by the strain did not require the addition of an artificial inducer such as γ-nonalactone or the autoregulator of S. virginiae MAFF 10-06014, IM, which induces the production of virginiamycin by this microorganism. The major improvements in the culture conditions for spontaneous pigment production included the inoculation conditions and the dilution of the medium. The method of IM2 assay was established and the time courses of IM2 production were followed in the cultures using flasks and a jar fermentor. It was confirmed that IM2 released once into the culture filtrate from the cells was taken up into the cells again. The concentration of IM required to induce pigment production in Streptomyces sp. MAFF 10-06015 was 50 u·ml−1. However a concentration of 200 u·ml−1 of IM2 was unable to induce the production of virginiamycin in S. virginiae MAFF 10-06014.  相似文献

To maximize the productivity of tissue plasminogen activator (TPA) by a mammalian cell culture, on-line determination of the optimum time to switch from the cell growth phase to the TPA production phase was investigated. By measuring the TPA production activity of the cells during the cell growth culture, it was shown that this optimum time was not necessarily the same as the time at which the cell concentration was maximized, and that the optimum time varied with growth culture batch. The TPA production activity of the cells during the growth culture could be estimated by on-line regression analysis using physiological data of the current state, including the oxygen consumption rate (Io2) and cell concentration, as well as data from past batches. Applying this on-line estimation, the optimum switching time was determined to be the time at which the TPA production activity of the cells in the growth culture became highest, or higher than a certain value according to determined criteria.  相似文献

Abstract

Dammarenediol-II synthase catalyzes the cyclization of 2,3-oxidosqualene to dammarenediol-II, which is the basic triterpene skeleton in dammarene-type saponin (ginsenosides) in Panax ginseng. Dammarenediol-II is a useful candidate both for pharmacologically active triterpenes and as a defense compound in plants. Dammarenediol-II is present in the roots of P. ginseng in trace amounts because it is an intermediate product in triterpene biosynthesis. In this work, we established the production of dammarenediol-II via cell suspension culture of transgenic tobacco. The dammarenediol-II synthase gene (PgDDS) isolated from P. ginseng was introduced into the Nicotiana tobacum genome under the control of 35S promoter by Agrobacterium-mediated transformation. Accumulation of dammarenediol-II in transgenic tobacco plants occurred in an organ-specific manner (roots > stems > leaves > flower buds), and transgenic line 14 (T14) exhibited a high amount (157.8 μg g?1 DW) of dammarenediol-II in the roots. Dammarenediol-II production in transgenic tobacco plants resulted in reduced phytosterol (β-sitosterol, campesterol, and stigmasterol) contents. A cell suspension culture was established as a shake flask culture of a callus derived from root segments of transgenic (T14) plants. The amount of dammarenediol-II production in the cell suspension reached 573 μg g?1 dry weight after 3 weeks of culture, which is equivalent to a culture volume of 5.2 mg dammarenediol-II per liter. Conclusively, the production of dammarenediol-II in a cell suspension culture of transgenic tobacco can be applied to the large-scale production of this compound and utilized as a source of pharmacologically active medicinal materials.  相似文献

Ellagic acid production from cups extract of valonia acorns by pure and mixed cultures of Aspergillus oryzae and Trichoderma reesei was investigated. Ellagitannin acyl hydrolase and xylanase as well as cellulase during the pure and mixed cultures were also determined. The results revealed that mixed culture could produce higher ellagic acid yield (23%) than either of pure culture. And it was found that the three enzymes from mixed culture appeared synergistic effect on ellagic acid production. Statistical analysis showed that ellagic acid yield was correlated very well with the three enzymes activities, resulting in the model for ellagic acid production with high R2 value of 0.998 and significant level p < 0.004.  相似文献

A Cordyceps militaris NBRC 10352-3 strain that was isolated from C. militaris NBRC10352 produced 68 mg of cordycepin from 100 mL of medium, which was the highest level of cordycepin among 60 isolates from three C. militaris (NBRC 9787, 100741 and 103752) strains. Interestingly, a liquid surface culture of C. militaris NBRC 103752-3 produced 2-fold cordycepin to that in a submerged culture. Cordycepin production was significantly affected by specific surface area (SSA) in the liquid surface culture, and 120.9 mg of cordycepin was produced on SSA of 1.57/cm (from 50 mL medium). The addition of glycine and adenine as an additive to its culture medium was optimized by an experimental design. When 6.75 g/L of adenine was added to the culture, 312.2 mg of cordycepin (apparent concentration: 6.2 g/L) was produced from 50 mL medium, improving the cordycepin production by 4.6-fold. In this study, the production and productivity of cordycepin were significantly improved in C. militaris wild type by a single cell colony isolation and additives without adopting any mutational technologies. This C. militaris NBRC 10352-3 strain can be used as a new cordycepin-hyperproducing one, instead of a cordycepin-hyperproducing mutant.  相似文献

Optimization of bacteriocin production by Lactobacillus plantarum LPCO10 was explored by an integral statistical approach. In a prospective series of experiments, glucose and NaCl concentrations in the culture medium, inoculum size, aeration of the culture, and growth temperature were statistically combined using an experimental 235-2 fractional factorial two-level design and tested for their influence on maximal bacteriocin production by L. plantarum LPCO10. After the values for the less-influential variables were fixed, NaCl concentration, inoculum size, and temperature were selected to study their optimal relationship for maximal bacteriocin production. This was achieved by a new experimental 323-1 fractional factorial three-level design which was subsequently used to build response surfaces and analyzed for both linear and quadratic effects. Results obtained indicated that the best conditions for bacteriocin production were shown with temperatures ranging from 22 to 27°C, salt concentration from 2.3 to 2.5%, and L. plantarum LPCO10 inoculum size ranging from 107.3 to 107.4 CFU/ml, fixing the initial glucose concentration at 2%, with no aeration of the culture. Under these optimal conditions, about 3.2 × 104 times more bacteriocin per liter of culture medium was obtained than that used to initially purify plantaricin S from L. plantarum LPCO10 to homogeneity. These results indicated the importance of this study in obtaining maximal production of bacteriocins from L. plantarum LPCO10 so that bacteriocins can be used as preservatives in canned foods.  相似文献

Light from fluorescent lamps can induce formaldehyde production and iron deficiency in plant nutrient culture medium. Formaldehyde is produced from EDTA when it is oxidized by the photochemical reduction of ferric iron and it can accumulate to inhibitory levels. The medium becomes iron deficient because iron becomes unchelated and forms an unavailable precipitate as EDTA is oxidized. The combination of light-induced formaldehyde production and loss of available iron reduces the ability of the culture media to support growth of Arabidopsis thaliana roots. Removing ultraviolet and blue wavelengths with a yellow acrylic filter is a simple and effective means of preventing Fecatalyzed photooxidation of EDTA in plant culture media.  相似文献

Growth and kefiran production rates of Lactobacillus kefiranofaciens were significantly enhanced in a mixed culture with Saccharomyces cerevisiae as compared with those in a pure culture. Because a positive effect on growth and kefiran production of L. kefiranofaciens in a mixed culture was observed, the elucidation of interaction between L. kefiranofaciens and S. cerevisiae may lead to higher productivity. Hence, microbial performance of each strain was investigated and analyzed by a mathematical model. The mathematical model for kefiran fermentation in a mixed culture of L. kefiranofaciens and S. cerevisiae was established, and the impact of S. cerevisiae on cell growth, kefiran formation, and substrate assimilation of L. kefiranofaciens were considered. The behavior of L. kefiranofaciens in a mixed culture was predicted using a developed mathematical model in this work, and the predictions were compared with the results from mixed culture experiments. The overall mathematical model is capable of describing the behavior of S. cerevisiae in a mixed culture as a lactic acid consumer, nitrogen source competitor and protective function inducer for L. kefiranofaciens. Furthermore, the constructed model described the phenomena in mixed cultures under aerobic and anaerobic conditions. Finally, the optimal inoculation ratios of S. cerevisiae to L. kefiranofaciens at 7-fold and 10-fold under aerobic and anaerobic conditions were obtained by applying the mixed culture model, respectively.  相似文献

Mexican isolates ofPaecilomyces fumosoroseus (Wize) Brown & Smith virulent to nymphs and adults ofBemisia tabaci Gennadius (Homoptera: Aleyrodidae) were screened in terms of spore production in submerged culture. Effects of light, temperature stress and yeast extract on sporulation were studied. Cycles of 12 hours light/12 hours dark increased spore production as well as an incubation for 24 hours at 37°C prior to incubation at 30°C. In absence of organic nitrogen both fungal growth and sporulation were very low. Spore production in fermentors with a culture media of a C:N ratio of 25 was doubled as compared to a media with a C:N ratio of 11. Both conidia and blastospores were produced. Production of conidia directly from blastospores through microcyclic sporulation was observed. The proportion of conidia obtained under optimal conditions was 88.8%. Submerged culture ofP. fumosoroseus seemed advantageous compared to ricefilled plastic bags production method because of shorter fermentation times, higher spore yields and substantially higher volumetric spore productivity. Results indicated that careful manipulation of nutritional and environmental conditions allowed for production of conidia during submerged growth ofP. fumosoroseus, microcyclic sporulation being induced under a set of environmental conditions including temperature stress and nutrients limitation.  相似文献

An attempt was made to use cane molasses as a culture medium for ε-PolyLysine (ε-PL) production by a natural bacterial isolate. The bacterium was identified as Bacillus sp., as confirmed by 16S rDNA sequence analysis. A BLAST result of the sequence indicated that the closest relative of this Bacillus BHU strain was B. thuringiensis, with 97 % homology. The molasses was found to be a better culture medium compared to commonly used culture media comprised of either glucose or glycerol as a carbon source. The various physicochemical parameters were studied for culture growth and polymer production, and were further optimized using response surface methodology (RSM). The correlation coefficient of the resulting model was found to be R 2?=?0.9828. The RSM predicted optimum conditions for ε-PL production (2.46 g/l) by the Bacillus strain was achieved by using molasses, 59.7 g/l; yeast extract, 15.2 mg/l; pH, 6.8 and fermentation time, 42 h at 30 °C. This study represents the first report on the potential application of cane molasses (a byproduct of sugarcane industries) as a culture medium for ε-PL production by Bacillus species. The specific Bacillus strain used in the present study can be exploited for developing a novel technology using inexpensive renewable resources for ε-PL production, a polymer of commercial interest.  相似文献

Salmosin, a snake venom-derived disintegrin, was successfully expressed in the methylotrophic yeast Pichia pastoris and secreted into the culture supernatant, as a 6 kDa protein. High-cell density fermentation of recombinant P. pastoris was optimized for the mass production of salmosin. In a 5 L jar fermentor, recombinant P. pastoris was fermented in growth medium containing 5% (w/v) glycerol at the controlled pH of 5.0. After culturing for 21 h, glycerol feeding medium was fed at one time into the culture broth. After 7 h (a total of 28 h), induction medium that contained methanol was increasingly added until the culture time totaled 75 h. Finally, these optimized culture conditions produced a high cell density of recombinant P. pastoris (dry cell weight of 113.38 g/L) and led to the mass production of salmosin (a total protein concentration of 369.2 mg/L). The culture supernatant containing salmosin inhibited platelet aggregation, resulting in a platelet aggregation of 9% compared to that of 94% in the control experiment, without culture supernatant. These results demonstrate that recombinant salmosin in culture supernatant from high cell density fed-batch fermentation can serve as a platelet aggregation inhibitor.  相似文献
