Three new triterpenes from the lichen Xanthoria resendei

The three new migrated hopene type triterpenes 12α-acetoxy-3β-hydroxyfern-9(11)-ene, 3β,12α-dihydroxyfern-9(11)-ene and 3,12-diketofern-9(11)-ene have been isolated from the ether extracts of the lichen Xanthoria resendei, together with peroxyergosterol and the anthraquinone pigments physcion, fallacinal and fallacinol.     

Biotransformation of the diterpene 15β-hydroxy-18(4→3)-abeo-ent-kaur-4(19),16-diene by the fungus Fusarium fujikuroi

15β-Hydroxy-18(4→3)-abeo-ent-kaur-4(19),16-diene (4) was biotransformed by the fungus Fusarium fujikuroi into 3α,11β,15β-trihydroxy-18(4→3)-abeo-ent-kaur-4(19),16-diene (5). The hydroxylation at C-3(α) in this diterpene reminds a similar reaction that occurs at C-13 in the biosynthesis of gibberellic acid in this fungus. The presence of the 15β-alcohol in the substrate directs the second hydroxylation at C-11(β), which had been observed in the incubation of ent-kaur-16-ene derivatives with this fungus when the C-19 hydroxylation was inhibited by the existence in the molecule of a 3α-OH or 3-oxo group. We also show that the angelate of the substrate is an undescribed natural product now identified as a component of the plant Distichoselinum tenuifolium.     

Sesquiterpenes and diterpenes from Ambrosia arborescens

Six compounds, eudesm-11(13)-en-4β,9β-diol, 15R,16-dihydroxy-3-oxoisopimar-9(11)-ene, 15S,16-dihydroxy-3-oxoisopimar-9(11)-ene, 1α-hydroxy-7-oxo-iso-anhydrooplopanone, 10α-hydroxy-11,13-dihydro-5-epi-psilostachyin, and 4β-hydroxypseudoguaian-12,6-olide 4-O-β-d-glucopyranoside, together with 12 known sesquiterpenes, were isolated from the leaves of Ambrosia arborescens. Structures were elucidated by 1D and 2D NMR spectroscopy including 1D-TOCSY, DQF-COSY, 2D-ROESY, HSQC, and HMBC experiments, as well as by ESI mass spectrometry. The absolute configuration of the 15,16-diol moiety in 15R,16-dihydroxy-3-oxoisopimar-9(11)-ene and 15S,16-dihydroxy-3-oxoisopimar-9(11)-ene was determined using Snatzke’s method. All compounds were evaluated for antiproliferative activity.     

New NF-κB Inhibitory Steroids from the Marine Sponge Dysidea avara Collected from the South China Sea

Chemical investigation of the marine sponge Dysidea avara, collected from the South China Sea, yielded 13 steroids, including nine new ( 1 – 9 ) and four known ( 10 – 13 ) ones. The new structures were elucidated as (3S,14R)-3,14-dihydroxycholesta-5,8-dien-7-one ( 1 ), (22E,24R)-7α-ethoxy-5α,6α-epoxyergosta-8(14),22-dien-3β-ol ( 2 ), 3β-hydroxy-7α-ethoxy-5α,6α-epoxy-8(14)-cholestene ( 3 ), 3β,5α-dihydroxy-6α-ethoxychofesta-7,9(11)-diene ( 4 ), 3β,5α-dihydroxy-6β-ethoxycholest-7-ene ( 5 ), (22E,24R)-24-ethoxy-3β,5α-dihydroxy-6β-ethoxyergosta-7,22-diene ( 6 ), (22E)-3β,5α-dihydroxy-6β-ethoxycholesta-7,22-diene ( 7 ), 24-ethoxy-3β,5α-dihydroxy-6β-ethoxycholest-7-ene ( 8 and 9 ), by extensive spectroscopic analyses, such as HR-ESI-MS, 1D and 2D NMR data. The absolute configuration of 1 was assigned by comparison the experimental ECD spectra with the calculated ones. Among the 13 metabolites, compounds 1 , 4 , 11 , 12 , and 13 showed NF-κB inhibitory activities in human HER-293 cells with IC₅₀ values of 6.4, 18.7, 8.1, 9.6, and 7.5 μM, respectively. Preliminary structure−activity relationship analysis unveiled that the conjugated ketones or unsaturated double bonds might be the functional groups for the five active steroids.     

Studies on Triterpenoid Constituents Isolated from the Roots of Sabia schumanniana

Five triterpenoids were isolated from the roots of Sabia schurnanniana Diels. Based on their physical constants, UV. IR, 1H-NMR, 13C-NMR, DEPT spectroscopic analysis and preparation of their derivatives, they were identified as 3-oxo-olean-ll, 13 (18)-diene (1), 3, 1 1-dioxo-olean- 12-ene (2), 3β -hydroxy-olean- 11, 13 (1 8) -diene (3), 3-oxo, 11 α-hydroxy-olean-12-ene (4), 3, 11α-dihydroxy-olean-12-ene (5) . Compound 4 is a new compound. Their 13C-NMR spectra have been signified by means of DEPT analysis.     

Biotransformation of 5α-hydroxycaryophylla-4(12),8(13)-diene with Cunninghamella elegans and Rhizopus stolonifer

Abstract

Biotransformation of 5α-hydroxycaryophylla-4(12),8(13)-diene (1) was studied with Cunninghamella elegans and Rhizopus stolonifer. Incubation of 1 with C. elegans gave regioselective oxidative addition (hydration) and isomerization at the C-4(12) exocyclic double bond and hydroxylation at C-3 and C-15, and thus provided two polar metabolites, (3Z),8(14)-caryophylladiene-5α,(11R)-15-diol (2) and 3β,4β,5α-trihydroxycaryophylla-8(13)-ene (3). Incubation of 1 with R. stolonifer gave a transannular cyclization reaction and afforded 2β-methoxyclovan-9-one (4), clovan-2β-ol-9-one (5) and 8-methoxycaryolane-5α,13β-diol (6). Compounds 3 and 6 are new compounds described here for the first time; their structures were deduced with the help of different spectroscopic techniques.     

Orgogenin,a pregnane derivative from orthenthera viminea

A new pregnane genin designated as orgogenin has been isolated from Orthenthera viminea and characterized to be 20-oxo-pregn-5-ene, 3β,8β,11α,12β,14β,15α-hexol.     

苋科植物的丛枝菌根

有些种子植物如莎草科、十字花科、灯心草科、藜科、石竹科等20余科,以往曾被认为不能或不易形成丛枝菌根(郭秀珍等,1989;刘润进等,2000).随着对菌根的深入研究,曾被认为是不易与菌根菌组合的湿地生植物、寄生性植物、或一年生植物都被发现是可以形成内生菌根的(Trappe等,1992).此外,Allen等(1989)研究证实,Salsola kali,Atriplex roseum等生长于沙漠、海滨的藜科植物,进行接种处理后,也能形成丛枝菌根.我们在西双版纳调查热带雨林植物的丛枝菌根状况时,偶然发现刺苋(Amaranthus spinosus Linn.)的根系受到了丛枝菌根真菌的侵染,因此,对苋科植物作了扩大采样调查.本文主要报道从热带采集的5属6种苋科植物的根受丛枝菌根真菌感染形成丛枝菌根(arbuscular mycorrhiza,AM)和这些植物根际士壤中的丛枝菌根真菌(arbuscular mycorrhizal fungi,AMF)的状况.     

高等真菌黑虎掌子实体的化学成分

黑虎掌 (Ｓａｒｃｏｄｏｎａｓｐｒａｔｕｍ (Ｂｅｒｋ .)Ｓ .Ｉｔｏ) ,又名香茸 ,是一种美味食用菌。近年来发现该属Ｓ .ｓｃａｂｒｏ ｓｕｓ (Ｆｒ.)Ｐ .Ｋａｒｓｔ.中含有对神经生长因子 (ＮＧＦ)的合成具有诱导作用的生物活性二萜 (Ｏｈｔ等 ,1998)。作为“高等真菌生物活性代谢产物研究”的一部分 ,我们对采自云南武定的样品进行了化学分析。从黑虎掌的新鲜子实体中分得 15个化合物。它们分别为ｃｅｒｅｂｒｏｓｉｄｅＢ (1) (12 0ｍｇ) ,阿洛酮糖腺苷(2 ) (12ｍｇ) ,三磷酸尿苷 (3) (7ｍｇ) ,尿嘧啶 (4 ) (12ｍｇ) ,腺嘌呤 (5 ) (8ｍ…     

The synthesis of 13α-androsta-5,16-diene derivatives with carboxylic acid,ester and carboxamido functionalities at position-17 via palladium-catalyzed carbonylation

17-Alkoxycarbonyl- and 17-carboxamido-3β-hydroxy-13α-androsta-5,16-diene derivatives were synthetized in high yields in the palladium-catalyzed carbonylation reactions of the corresponding 3β-hydroxy-17-iodo-13α-androsta-5,16-diene. This substrate with a 17-iodo-16-ene functionality was obtained from the 17-keto derivative via its 17-hydrazone, which was treated with iodine in the presence of a base (1,1,3,3-tetramethylguanidine). 17-Carboxamides were obtained by chemoselective aminocarbonylation through the use of amines, including amino acid esters, as N-nucleophiles. The 17-methoxycarbonyl-16-ene derivative was synthetized by using methanol as O-nucleophile. The parent compound of this series, the 17-carboxylic acid derivative, was formed in the presence of water via hydroxycarbonylation.     

Zierane sesquiterpene lactone,cembrane and fusicoccane diterpenoids,from the Tahitian liverwort Chandonanthus hirtellus

Cembrane-type diterpenoids, 13,18,20-epi-iso-chandonanthone (1) and (8E)-4α-acetoxy-12α,13α-epoxycembra-1(15),8-diene (2), two fusicoccane-type diterpenoids, fusicoauritone 6α-methyl ether (3) and 6β,10β-epoxy-5β-hydroxyfusicocc-2-ene (4) and a zierane sesquiterpene γ-lactone, chandolide (5) were isolated from the Tahitian liverwort Chandonanthus hirtellus (Web.) Mitt., together with eight known diterpenoids, chandonanthine (6), fusicogigantone A (7), fusicogigantone B (8), fusicogigantepoxide (9), anadensin (10), fusicoauritone (11), ent-verticillol (12) and ent-epi-verticillol (13). Their structures were established by a combination of extensive NMR spectroscopy and/or X-ray crystallographic analyses. Compounds 1, 5 and 10 showed weak cytotoxic activity against HL-60. Compound 3 also indicated weak cytotoxic activity against KB cell lines.     

海莲内生真菌Pestalotiopsis clavispora代谢产物研究

从海莲内生真菌Pestalotiopsis clavispora发酵液乙酸乙酯萃取物中分离得到9个化合物。根据波谱数据,化合物1－9结构分别鉴定为：3β,22β,24-三羟基齐墩果-12-烯,乌苏酸,3-羟基-4-甲氧基苯乙烯,邻苯二甲酸二异丁酯,3,4-二羟基苯乙醇,对羟基苯乙醇,3β-羟基-5α,8α-过氧化麦角甾-6,22-二烯,麦角甾-4,6,8(14),22-四烯-3-酮,胸苷。9个化合物均为首次从该内生真菌中分离得到。     

Trichothecene mycotoxins from Fusarium sulphureum

Four mycotoxins isolated from moulded maize cultures of Fusarium sulphureum have been characterized as 3α,4β,15-triacetoxy-12,13-epoxytrichothec-9-ene, 4β,15-diacetoxy-3α-hydroxy-12,13-epoxytrichothec-9-ene, 15-acetoxy-3α,4β-dihydroxy-12,13-epoxytrichothec-9-ene and 4β-acetoxy-3α,15-dihydroxy-12,13-epoxytrichothec-9-ene.     

Cissogenin,a pregnane genin from Marsdenia tenacissima

A new polyhydroxy pregnane designated as cissogenin has been isolated from the seeds of Marsdenia tenacissima. Its structure has been established as 3β,11α,12β,14β,20 S-pentahydroxy-pregn-5-ene.     

Three ent-kaurene diterpenes from Vellozia caput-ardeae

Three new ent-kaurene diterpenes have been isolated from the roots and stem of Vellozia caput-ardeae. Their structures were elucidated by spectroscopic methods as ent-9β-hydroxy kaur-16-ene, ent-11α-hydroxy kaur-16-ene and ent-9β,11α-dihydroxy kaur-16-ene.     

Hirsudiol a triterpenoid from cocculus hirsutus

A new triterpenoid alcohol hirsudiol, has been isolated from Cocculus hirsutus. Its structure was determined as 2α,3α-dihydroxyolean-13(18)-ene through spectroscopic studies. Nonacosan-10-ol was also isolated.     

绿茎还阳参化学成分的研究

采用柱层析和重结晶从药用植物绿茎还阳参(Crepis lignea)丙酮提取物中分离得到7个单体化合物,并根据化合物的理化性质和波谱分析,分别鉴定为:3β-acetoxyurs-13( 18)-ene (1),lupeol acetate (2),4-hydroxy-3 -methoxycinnamaldehyde (3),(Z)-2-ethylidene-3-methylsuccinic acid (4),olean- 12 -ene-11 α-methoxy-3β-acetate (5),α-amyrin acetate (6)和lupeol-9( 11) en-3β-acetate(7).上述化合物均为首次从绿茎还阳参中分离得到.     

21beta-Hydroxy-oleanane-type triterpenes from Hippocratea excelsa

Stem bark of Hippocratea excelsa afforded six pentacyclic triterpenes, five oleanane and one ursane types. They were identified as 11beta,21beta-dihydroxy-olean-12-ene-3-one (2), 3alpha,11alpha,21beta-trihydroxy-olean-12-ene (3), 3alpha,21beta-dihydroxy-11alpha-methoxy-olean-12-ene (4), 3alpha,21beta-dihydroxy-olean-9(11),12-diene (5), 3alpha,21beta-dihydroxy-olean-12-ene (6) and 3alpha,21beta-dihydroxy-11alpha-methoxy-urs-12-ene, isolated as its diacetate derivative (7), as well as 3alpha,21beta-dihydroxy-olean-12-ene (1) previously isolated from the root bark. The known alpha- and beta-amyrin, oleanoic and ursolic acids, trans-polyisoprene, and the ubiquitous beta-sitosterol were also isolated. Structures were elucidated on the basis of spectroscopic analyses, including homo- and heteronuclear correlation NMR experiments (COSY, ROESY, HSQC and HMBC) and comparison with literature data. The antigiardial activity of compounds 2-5 was not significant.     

Volatile components from European liverworts Marsupella emarginata,M. aquatica and M. alpina

The hydrodistillation products of the liverworts Marsupella emarginata, M. aquatica and M. alpina were investigated by spectroscopic methods. A number of new compounds could be isolated by preparative gas chromatography (GC) and identified by spectroscopic techniques including GC-mass spectrometry, NMR and chemical correlations in conjunction with enantioselective GC. From M. emarginata, in addition to many known compounds, the sesquiterpene hydrocarbon (-)-7-epi-eremophila-1(10),8,11-triene (1) and the sesquiterpene derivatives (-)-4-epi-marsupellol (2), (-)-marsupellol acetate (18), (-)-4-epi-marsupellol acetate (4), (+)-5-hydroxymarsupellol acetate (5) and (-)-9-acetoxygymnomitr-8(12)-ene (24) could be identified. In M. aquatica the sesquiterpene hydrocarbons (-)-myltayl-8(12)-ene (7), ent-(+)-amorpha-4,11-diene (8), (-)-amorpha-4,7(11)-diene (9), the sesquiterpene alcohol (+)-9-hydroxyselina-4,11-diene (10) and (-)-2-acetoxyamorpha-4,7(11)-diene (11) were identified. In M. alpina (-)-trans-selina-4(15),11-dien-5-ol (12), (+)-8,9-epoxyselina-4,11-diene (13) and (+)-cis-selina-4(15),11-dien-5-ol (14) were found as new natural products.     

Tenasogenin,a pregnane ester from Marsdenia tenacissima

A new polyhydroxy pregnane ester named tenasogenin was isolated from the seeds of Marsdenia tenacissima. On the basis of chemical and spectroscopic evidence and identification of its hydrolysis products, its structure has been established as 11α-O-β,β-dimethylacryloyl-3β,12β,14β,20R-tetrahydroxypregn-5-ene.
