Electrostatic charging of an electrically insulated metal body streamlined by a plasma flow is investigated theoretically. The physical processes occurring in the vicinity of the body are considered. The electric currents resulting in the charging or discharging of the body are determined. The electric charge Q(t), electric field E(t), and potential φ(t) of a metal sphere are estimated taking into account the adopted conditions and parameters of the problem. The physical phenomena that take place in the vicinity of a charged body after Q(t) reaches a certain threshold value are discussed. The obtained values of Q(t), E(t), and φ(t) are compared with the corresponding values for a body streamlined by an atmospheric air flow.  相似文献

A nested polymerase chain reaction (PCR) technique for amplifying a fragment of the gene (GH) encoding teleost growth hormone has been developed. Using this technique, a fragment of the pufferfish, Fugu rubripes and Arothron maculatus; dwarf gourami, Colisa lalia; guppy, Poecilia reticulata; and goldfish, Carassius auratus GH genes were cloned. The Fugu rubripes (Fugu) gene fragment was used to isolate the GH gene from a Fugu genomic library. The complete nucleotide sequence of a 8.5-kb SacI genomic fragment containing the Fugu GH gene has been determined. The GH gene spans 2.5 kb from the first codon to polyadenylation signal, and contains six exons and five introns similar to the GH genes of salmonids, tilapia, barramundi, flounder and yellowtail. The GH introns contain microsatellite and satellite sequences. The microsatellites found in the fifth intron of the GH gene are also present in the corresponding introns of tilapia, barramundi and flounder GH genes. Southern analysis revealed that the GH gene is a single-copy gene in the Fugu. The promoter region of the Fugu GH gene contains conserved sequences that are likely to be involved in the pituitary-specific expression of the gene. A phylogenetic tree of nucleotide (nt) sequences of all known teleost GH genes has been inferred using the distance matrix method. The topology of this tree reflects the major phylogenetic groupings of teleosts. The intron patterns and repetitive sequences of GH genes can serve as useful natural markers for the classification and phylogenetic studies of teleosts.  相似文献

Molecular typing is an important tool in the investigation of methicillin resistant Staphylococcus aureus (MRSA) outbreaks and in following the evolution of MRSA. The staphylococcal cassette chromosome mec (SCCmec) contains a hypervariable region with a variable number of 40 bp repeats named direct repeat units (dru). The dru region has been suggested as a supplementary typing method for MRSA and an international nomenclature exists. The purpose of this study was to investigate the diversity and variability of the dru region in a diverse collection of MRSA. We studied 302 MRSA isolates harbouring SCCmec types I to VI. The isolates represented a broad genetic background based on Staphylococcal protein A (spa) typing and multilocus sequence typing (MLST) and included 68 isolates (68 patients) from an outbreak with t024-ST8-IVa and 26 isolates from the same patient. Sequencing identified 53 dru types (dt) in 283 isolates, while eighteen isolates contained no dru repeats and one isolate resisted sequencing. The most common dru type, dt10a, was present in 53% of the sequenced isolates and was found in all SCCmec types, except type II. Seven (10%) of the 68 epidemiologically related patients had isolates with dru type variants indicating that dru typing is not useful as a first line epidemiological typing tool. However, MRSA isolates cultured from a single patient over a three year period exhibited a single dru type. The finding of dt10a in most SCCmec types suggests that dru and mecA originate from the same Staphylococcus species.  相似文献

The narA locus required for nitrate reduction in Synechococcus sp. strain PCC 7942 is shown to consist of a cluster of genes, namely, moeA, moaC, moaD, moaE, and moaA, involved in molybdenum cofactor biosynthesis. The product of the moaC gene of strain PCC 7942 shows homology in its N-terminal half to MoaC from Escherichia coli and in its C-terminal half to MoaB or Mog. Overexpression of the Synechococcus moaC gene in E. coli resulted in the synthesis of a polypeptide of 36 kDa, a size that would conform to a protein resembling a fusion of the MoaC and MoaB or Mog polypeptides of E. coli. Insertional inactivation of the moeA, moaC, moaE, and moaA genes showed that the moeA-moa gene cluster is required for growth on nitrate and expression of nitrate reductase activity in strain PCC 7942. The moaCDEA genes constitute an operon which is transcribed divergently from the moeA gene. Expression of the moeA gene and the moa operon was little affected by the nitrogen source present in the culture medium.  相似文献

An unstable component (I-R) of the R-stippled allele interacts with a linked modifier which enhances stippled's expression ( M-st) to delete the intervening segment. The precision of deletion formation suggests a recombinational basis, specifically unequal crossing over between I-R and M-st. Four deletions were selected as losses of R function from plants homozygous for R-st and M-st. Also lost are stippled's near-colorless seed phenotype, its paramutagenicity and a closely linked gene, Inhibitor of striate. The deletion chromosomes, missing a 6-cM segment, are transmitted normally by ovules but in reduced frequency by pollen. Homozygous and heteroallelic combinations of the deletions confer defective seed lethality. The four did not differ detectably in transmissibility or breakpoint termini. The recurrence of deletions that have the same termini is explained by recombination between I-R and M-st. The homology between M-st and I-R, and their presence in the same chromosome arm, favors the view that M-st originated by I-R transposition.  相似文献

The heterosporous water-fern genusAzolla is one of the few symbioses with a cyanobacterium in the genusAnabaena. TheAzolla-Anabaena association includes six extant speciesof Azolla, which are widely distributed in relatively placid tropical and/or temperate freshwater environments. The earliest mention of the plant seems to be in an ancient Chinese dictionary that appeared about 2000 years ago.Azolla was used in about the 11th century in Vietnam. By 1980 renewed interest in this symbiotic association was shown by the demand for a less fossil energy-dependent agricultural technology. The importation of a variety ofA. filiculoides may have been a most significant breakthrough for the improvementof Azolla cultivation in China. The history of research may be divided into three periods and a new biotechnological stageof Azolla research has recently begun. Each mature dorsal leaf lobe has an ellipsoid cavity which containsAnabaena azollae throughout its development. HeterocystousA. azollae from sixAzolla species share identical and highly specific antigens.Azolla and its endophyte exhibit a coordinated pattern of differentiation and development. Epidermal hair cells of the host are probably interactive with the symbiont. The interior surface of a mature leaf cavity is lined with an envelope and covered by a mucilaginous layer.A. azollae shares the cavity with small populations of the bacteriaPseudomonas andAzotobacter. Endophyte-freeAzolla may rarely occur in nature and can be generated by aseptic techniques.Anabaena azollae can be isolated fromAzolla fronds by gentle pressure and by enzymatic digestion. The free living cultures derived from theAnabaena so obtained differ in some respects, however, from the freshly extracted symbiont, and might better be called the presumptive isolate. BothAzolla andAnabaena contain specific photosynthetic pigments. The optimum conditions for photosynthesis have been measured.Azolla is a C3 plant and has high net photosynthesis. PSII activity in the symbiont is low. Nitrogenase is localized in the heterocysts of the symbiont and has some advantages compared with free-living cyanobacteria. SymbioticA. azollae has a high frequency of heterocysts. Unidirectional hydrogenase occurs in the symbiont and recycles electrons and ATP. Simultaneous measurements of N2 fixation and photosynthesis show the dependence of nitrogenase on photosynthetically captured radiation for energy by an indirect dependence on CO2 fixation. The host contains most of the total GS and GDH activities, and the symbiont excretes a substantial portion of its newly fixed nitrogen as ammonium. The two partners in the association exhibit a comparable developmental gradient and a mechanism of cooperative integration for their energy metabolism, thus improving the efficiency of solar energy conversion and presenting a unique model for biotechnology.  相似文献

The study of a sea level fluctuation based on ostracods is carried out for the first time using core sediments of Mbawane and Retba Lakes, north coast of Senegal. A total of 26 species have been identified and most of them are encountered in the Gulf of Guinea shelf and its adjacent lagoons. The Retba Lake core, which is closer to the seashore, has yielded a richer and more diversified ostracod fauna composed of four assemblages. The F1 assemblage is composed of Cyprideis nigeriensis, Neomonoceratina iddoensis, and Aglaiocypris gambiensis, which characterises a slightly open lagoon developing under a humid climate with its edges covered with mangrove swamps. Assemblage F2, composed mainly of C. nigeriensis, N. iddoensis and “marine species”, which is richer and more diversified, points to a marine little gulf bordered by mangrove swamps. The F3 monospecific assemblage composed of C. nigeriensis indicates a drying confined lagoon under an arid climate. The F4 oligospecific association at the top, composed of C. nigeriensis and Pseudoconcha omatsolai, is typical of a slightly open saline lagoon under arid climate. Core sediments from Mbawane Lake have yielded scarce and badly preserved microfaunas which are distributed into three discontinuous levels that have been correlated to those of the Retba Lake core assemblages. The F1 assemblage composed of Leptocythere culpata and Xestoleberis sp. 2 corresponds to a slightly open lagoon covered at its bottom by marine macrophytes and algae. The F2 assemblage composed of N.iddoensis, Hemicytherura sp. A, Aglaiocypris gambiensis and Ruggieria tricostata characterises a more open lagoon bordered by mangrove swamps under a humid climate. The F3 assemblage is absent in the Mbawane Lake. The F4 monospecific association composed of C. nigeriensis characterises a drying lagoon under an arid climate. The overall marine influences were stronger in the Retba Lake while continental effects were dominant in Mbawane Lake during the same period. The two marine intrusions mentioned above may correspond to those of the Dakarian (3000 years B.P.) and the Saint-Louisian eras (2000–680 years B.P.) that existed during the Upper Holocene on the Senegalese coast.  相似文献

Megachile Latreille is a conspicuous genus of solitary bees distributed worldwide. However, the biology of tropical species is still little known. We present data on biology of Megachile brasiliensis Data Torre, Megachile sejuncta Cockerell and Megachile stilbonotaspis Moure found in two remnants of eastern Amazonian forest in northeastern Brazil. The study was conducted using the trap-nest methodology in two different areas during four periods. We collected a total of 24 nests of M. brasiliensis, 26 of M. sejuncta and 28 of M. stilbonotaspis. The differential abundance of collected nests may reflect the population size in each sampled place. The nesting activity was concentrated mainly between July and January and species presented a multivoltine pattern, except for M. sejuncta, which was partly univoltine. Assessed pollen use showed a predominant use of Attalea sp. (Arecaceae) and, for M. stilbonotaspis, Tylesia sp. and Lepidaploa sp. (Asteraceae). Babassu is a very common palm in the studied areas and the studied species seem to have a strong link with it. We also reported change of pollen use by M. sejuncta, probably due to competition with M. brasiliensis, which may have influenced the biased sex ratio observed in M. sejuncta toward males. Parasites reported here were also recorded for other Megachile species, such as Coelioxys, Brachymeria, Meloidae and Pyralidae species. Mites were observed in association with M. stilbonotaspis. The data presented here set up a background that encourages new studies on the ecology of these three Amazonian species, providing tools for proper biodiversity management and conservation.  相似文献

Costaria costata is a commercially and industrially important brown alga. In this study, we used next-generation sequencing to determine the complete plastid genome of C. costata. The genome consists of a 129,947 bp circular DNA molecule with an A+T content of 69.13% encoding a standard set of six ribosomal RNA genes, 27 transfer RNA genes, and 137 protein-coding genes with two conserved open reading frames (ORFs). The overall genome structure of C. costata is nearly the same as those of Saccharina japonica and Undaria pinnatifida. The plastid genomes of these three algal species retain a strong conservation of the GTG start codon while infrequently using TGA as a stop codon. In this regard, they differ substantially from the plastid genomes of Ectocarpus siliculosus and Fucus vesiculosus. Analysis of the nucleic acid substitution rates of the Laminariales plastid genes revealed that the petF gene has the highest substitution rate and the petN gene contains no substitution over its complete length. The variation in plastid genes between C. costata and S. japonica is lower than that between C. costata and U. pinnatifida as well as that between U. pinnatifida and S. japonica. Phylogenetic analyses demonstrated that C. costata and U. pinnatifida have a closer genetic relationship. We also identified two gene length mutations caused by the insertion or deletion of repeated sequences, which suggest a mechanism of gene length mutation that may be one of the key explanations for the genetic variation in plastid genomes.  相似文献

The genetic basis for a striking aleurone color variegation of a maize population from Colombia is described. The variegation is the result of the interaction of a particular r allele, designated r-cu , and a specific factor, Fcu (Factor Cuna), that collectively compose a controlling element system. In the absence of Fcu, a variable dilute (nonvariegated) pigmentation is expressed in the aleurone. The degree of variegation is determined by differences in the dosage of the Fcu element; differences in variegation between kernels with one r-cu vs. two r-cu's cannot be detected. Because of the specificity of the interaction, Fcu is different from the three other well-known controlling elements, Dt, Ac and En(=Spm), and therefore represents a fourth controlling element system in maize. In addition to Fcu, the Cuna tribal maize contains two other controlling elements, Dt and a very weak En. The prevalence of controlling elements in natural maize populations is discussed.  相似文献

Effects of the following PRLamide peptides on some invertebrate muscles were examined: APNFLAYPRLamide (a peptide of the bivalve mollusc Mytilus), AAPLPRLamide (a peptide of the echiuroid Urechis), FTPRLamide (a fragment of a peptide of the insect Pseudaletia), YFSPRLamide (a fragment of a peptide of the insect Heliothis) and pQTSFTPRLamide (a peptide of the insect Leucophaea).2. In the ABRM of Mytilus, the Mytilus and Urechis peptides potentiated phasic contraction by repetitive electrical stimulation. The former was more potent than the latter. The three insect-origin peptides did not show any potentiating effect but showed a weak antagonistic action on the potentiating effect of the Mytilus peptide. Except the Mytilus peptide, the PRLamide peptides showed little or no relaxing effect on catch tension, though they accelerated catch-relaxing response to repetitive electrical stimulation.3. In the inner circular body-wall of muscle of Urechis, the PRLamide peptides showed a potentiating effect on twitch contraction. The Urechis peptide was most potent. In the esophagus of the annelid Perinereis, the PRLamide peptides showed a contractile effect. The Urechis peptide was also most potent.4. In the crop of the cricket Gryllus, the insect-origin peptides markedly potentiated spontaneous rhythmic contractions. The Mytilus peptide showed a weak potentiating effect, but the Urechis peptide showed little or no effect.  相似文献

The holotypes of the Lower Jurassic DipteraMesorhyphus nanus Handlirsch 1920,M. areolatus Handlirsch 1939,M. anomalus Handlirsch 1939,Eoplecia primitiva Handlirsch 1920,Heterorhyphus latus Bode 1953,H. analivarius Bode 1953 andProtorhyphus (?)ovisimilis Bode 1953 were re-examined. The holotype ofEoplecia primitiva is the counterpart ofMesorhyphus areolatus. M. areolatus is a junior synonym ofM. nanus. Mesorrhyphoides Rohdendorf 1962 is a synonym ofHeterorhyphus Bode 1953.Heterorhyphus analivarius is a synonym ofProtorhyphus (?)ovisimilis, being mistakenly described from the counterpart of the holotype of the latter. The genusHeterorhyphus is valid and comprises onlyH. anomalus (Handlirsch 1939) n. comb.;H. latus is a synonym of the latter species. As the wing venation ofHeterorhyphus is conspicuously different from all fossil and Recent Anisopodomorpha, it is placed into a new family, Heterorhyphidae, within the Bibionomorpha.  相似文献

Bombyx mori nucleopolyhedrovirus (BmNPV) is a major viral agent that causes deadly grasserie disease in silkworms, while BmNPV DNA polymerase (BmNPV-pol), encoded by ORF53 gene, plays a central role in viral DNA replication. Efficacy studies of BmNPV-POL are limited because of poor heterologous protein expression in E. coli. Here, we redesigned the BmNPV-pol to preferentially match codon frequencies of E. coli without altering the amino acid sequence. Following de novo synthesis, codon-optimized BmNPV-pol (co-BmNPV-pol) gene was cloned into pET32a and pGEX-4T-2 vector. The expression of co-BmNPV-POL in E. coli was significantly increased when BmNPV-POL was fused with GST protein rather than a His-tag. The co-BmNPV-POL fusion proteins were isolated using GST affinity chromatography and Mono Q iron exchange chromatography. Protein purity and identity were confirmed by western blot and MALDI-TOF analyses. The biological activity of purified proteins was measured on a poly(dA)/oligo(dT) primer/template. The specific polymerasing activity of the recombinant BmNPV-POL was 6,329 units/mg at optimal conditions. Thus, a large amount of purified protein as a soluble form with high activity would provide many benefits for the functional research and application of BmNPV-POL.  相似文献

The Anopheles genus is a member of the Culicidae family and consists of approximately 460 recognized species. The genus is composed of 7 subgenera with diverse geographical distributions. Despite its huge medical importance, a consensus has not been reached on the phylogenetic relationships among Anopheles subgenera. We assembled a comprehensive dataset comprising the COI, COII and 5.8S rRNA genes and used maximum likelihood and Bayesian inference to estimate the phylogeny and divergence times of six out of the seven Anopheles subgenera. Our analysis reveals a monophyletic group composed of the three exclusively Neotropical subgenera, Stethomyia, Kerteszia and Nyssorhynchus, which began to diversify in the Late Cretaceous, at approximately 90 Ma. The inferred age of the last common ancestor of the Anopheles genus was ca. 110 Ma. The monophyly of all Anopheles subgenera was supported, although we failed to recover a significant level of statistical support for the monophyly of the Anopheles genus. The ages of the last common ancestors of the Neotropical clade and the Anopheles and Cellia subgenera were inferred to be at the Late Cretaceous (ca. 90 Ma). Our analysis failed to statistically support the monophyly of the Anopheles genus because of an unresolved polytomy between Bironella and A. squamifemur.  相似文献

The cyanobacterium Nostoc strain ATCC 53789, a known cryptophycin producer, was tested for its potential as a source of natural pesticides. The antibacterial, antifungal, insecticidal, nematocidal, and cytotoxic activities of methanolic extracts of the cyanobacterium were evaluated. Among the target organisms, nine fungi (Armillaria sp., Fusarium oxysporum f. sp. melonis, Penicillium expansum, Phytophthora cambivora, P. cinnamomi, Rhizoctonia solani, Rosellinia, sp., Sclerotinia sclerotiorum, and Verticillium albo-atrum) were growth inhibited and one insect (Helicoverpa armigera) was killed by the extract, as well as the two model organisms for nematocidal (Caenorhabditis elegans) and cytotoxic (Artemia salina) activity. No antibacterial activity was detected. The antifungal activity against S. sclerotiorum was further studied with both extracts and biomass of the cyanobacterium in a system involving tomato as a host plant. Finally, the herbicidal activity of Nostoc strain ATCC 53789 was evaluated against a grass mixture. To fully exploit the potential of this cyanobacterium in agriculture as a source of pesticides, suitable application methods to overcome its toxicity toward plants and nontarget organisms must be developed.  相似文献

Aim

Ground-penetrating radar (GPR) has been applied to detect coarse tree roots. The horizontal angle of a root crossing a scanning line is a factor that affects both root detection and waveform parameter values. The purpose of this study was to quantitatively evaluate the influence of root orientation (x, degree) on two major waveform parameters, amplitude area (A, dB × ns) and time interval between zero crossings (T, ns).

Methods

We scanned four diameter classes of dowels in a sandy bed as simulated roots using a 900 MHz antenna from multiple angles to clarify the relationships between the parameters and x.

Results

Angle x strongly affected reflection images and A values. The variation in A(x) fitted a sinusoidal waveform, whereas T was independent of x. The value of A scanning at 90° was estimated by A values of arbitrary x in two orthogonal transects. The sum of T in all reflected waveforms showed a significant linear correlation with dowel diameter.

Conclusions

We clarified that root orientation dramatically affected root detection and A values. The sum of T of all reflected waveforms was a suitable parameter for estimating root diameter. Applying grid transects can overcome the effects of root orientation.  相似文献
