通过不同饥饿处理 ,研究了东方田鼠在不同营养状态下 ,对陌生食物尝试和选择的行为模式。结果表明 ,初次遇到陌生食物时 ,各处理组对陌生食物的采食量分别为饱食 6.9± 1 .8g,饥饿组为 4.8± 1 .6g,饥饿 食物短缺组为 3.4± 1 .4g。各处理组动物对陌生食物的摄入量间差异极为显著 ( P=0 .0 0 4 3)。饱食组、饥饿组及饥饿 食物短缺组动物初次遇到陌生食物时 ,所摄入的陌生食物量占总摄入量的百分比分别为72 %、38%与 1 2 %。说明 ,在实验室条件下饱食动物较饥饿和饥饿 食物短缺的动物学习选择摄食陌生食物的倾向较强。  相似文献

Conditions for the optimal production of polyhydroxyalkanoate (PHA) by Pseudomonas mendocina PSU using a biodiesel liquid waste (BLW) were determined by response surface methodology. These were an initial carbon to nitrogen ratio (C/N) of 40 (mole/mole), an initial pH of 7.0, and a temperature of 35 °C. A biomass and PHA concentration of 3.65 g/L and about 2.6 g/L (77% DCW), respectively, were achieved in a growth associated process using 20 g/L glycerol in the BLW after 36 h of exponential growth. The PHA monomer compositions were 3HB (3-hydroxybutyrate), a short-chain-length-PHA, and the medium-chain-length-PHA e.g. 3-hydroxyoctanoate and 3-hydroxydecanoate. Both the phbC and phaC genes were characterized. The phbC enzyme had not been previously detected in a Pseudomonas mendocina species. A 2.15 g/L of an exopolysaccharide, alginate, was also produced with a similar composition to that of other Pseudomonas species.  相似文献

This study analyzed the environmental impacts of packaging‐derived changes in food production and consumer behavior to assist packaging designers in making environmentally conscious decisions. Packaging can be functionalized to prevent food loss and waste (FLW), for example, extending the expiration date and apportioning the package size, but it can generate additional environmental impacts from changes in food and packaging production. Previous studies assessed additional impacts from packaging production; however, the effects of packaging functionalization are yet to be connected with food production and consumer behavior. To examine the effect of functionalization on these aspects, we analyzed packaging‐derived changes in food production for milk and cabbage products. The case study compared products with functionalized packaging that permits a longer expiration date or a smaller portion size to their base‐case products. Our results showed that the packaging‐derived changes increased the global warming potential (GWP) of food production more than other processes did. Thus, changes in food production weakened the effectiveness of the packaging functionalization to decrease the GWP. Moreover, the analysis of consumer behavior scenarios showed that consumers’ perception of the expiration date decisively influences the effectiveness of packaging functionalization. When consumers discarded food after the expiration date, provided they consumed in small quantities, the packaging functionalization reduced FLW. From the scenario analysis, we identified appropriate combinations of packaging functionalization and consumer behaviors to effectively decrease total GWP. With our expanded analysis, packaging designers can understand the effectiveness of their decisions on the product life cycle in reducing FLW and environmental impacts.  相似文献

温带地区小型哺乳动物经常面临着食物资源的波动。食物对动物的免疫功能具有重要影响。将19只成年雄性黑线仓鼠(Cricetulus barabensis)随机分为自由取食组(n=9)和限食组(n=10)。注射植物血球凝集素(PHA)来测定细胞介导的免疫反应,用匙孔血蓝蛋白(KLH)免疫动物,然后测定抗KLH抗体的浓度以反映其体液免疫功能。旨在检验食物限制是否会抑制黑线仓鼠的细胞免疫和体液免疫功能。结果发现,与对照组相比,限食组黑线仓鼠具有较低的体重、体脂、脾脏鲜重、血清瘦素水平、免疫球蛋白Ig G和Ig M浓度。而限食对胸腺鲜重、白细胞数、皮质酮水平以及PHA反应没有显著影响。结果表明黑线仓鼠免疫系统的不同成分对限食反应存在差异,在食物资源短缺时,黑线仓鼠防御细胞外病原体的能力降低,从而导致生存能力的下降。  相似文献

本文以黑水虻Hermetia illucens为研究对象,探索不同废弃物对其幼虫生长、营养组成及生产性能的影响。结果表明,黑水虻幼虫可将餐厨垃圾(FW)、厨余垃圾(KW)、蔬菜废弃物(VW)、猪粪(PM)、牛粪(DM)、鸡粪(CM)等转化成优质蛋白质和脂肪。以FW和KW作为基质养殖10 d后,幼虫体长(19.0 mm、17.1 mm)与单头重(171.54 mg、136.82 mg)显著高于其它处理。虫粉粗蛋白含量受废弃物种类影响相对较小。不同废弃物处理虫粉均显示出良好氨基酸谱。尽管各处理虫粉总必需氨基酸含量低于鱼粉和豆粕,但FW、KW和CM处理虫粉缬氨酸含量高于鱼粉和豆粕,VW处理虫粉异亮氨酸和缬氨酸含量高于鱼粉和豆粕,PM处理虫粉组氨酸和缬氨酸含量高于鱼粉和豆粕。以FW和KW作为基质养殖10 d后,尽管虫粉不饱和脂肪酸和必需脂肪酸比例低于采食PM和DM的处理,但远高于其它处理的虫粉粗脂肪含量(42.13%、37.49%),虫粉不饱和脂肪酸和必需脂肪酸含量也最高;料虫比(6.06、7.90)显著低于其它处理,幼虫转化率(59.40%、52.07%)显著高于其它处理;废弃物减量化率(89.53%、84.46%)仅低于VW处理(94.86%)。利用黑水虻幼虫处理FW和KW,幼虫生长性能和营养组成优于其它废弃物处理,且生产性能最好。  相似文献

Experiments were conducted on14C-sorbitol, fructose, and glucose uptakeinto flesh discs, and sorbitol efflux from thediscs, with and without ABA application toexamine the effect of abscisic acid (ABA) onsugar accumulation in peach fruit flesh at thestart of the maturation stage in relation tomembrane transport. Total uptake of14C-sorbitol, fructose, and glucose intoflesh discs was effectively promoted by ABA ata concentration of 10–5 M. PCMBS(p-chloromercuribenzensulfonicacid)-sensitive uptake, which was considered ascarrier-mediated uptake, of sorbitol into thediscs was clearly stimulated by ABA at10–5 M, compared with glucose andfructose uptake. Sorbitol efflux from the discsacross the tonoplast was restricted by ABA at10–5 M. ABA application todeveloping fruit increased sugar accumulationin the fruit. Estimated ABA concentration inthis fruit was approximately 10–5 M. These results indicate that sugar accumulationin peach fruit flesh is stimulated by ABA at aconcentration of 10–5 M both invitro and in vivo. ABA stimulatesuptake of sugars, especially sorbitol, into theflesh by enhancing carrier-mediated transportpossibly across both tonoplast and plasmamembrane.  相似文献

限食通常会显著影响鼠类的营养及内分泌等生理指标,但限食后重喂食其生理指标是否能得以恢复尚不
清楚。本文采取70% 的限食水平,研究了限食及重喂食对雄性长爪沙鼠生理指标的影响。将雄性长爪沙鼠分为
限食组、重喂食组和对照组。限食组先自由饮食4 周,后70% 限食4 周;重喂食组先70% 限食4 周,后恢复自
由饮食4 周。对照组自由饮食8 周。实验结束时,检测各组肥满度及血清白蛋白和总蛋白含量、血清甲状腺素
T3 和T4 水平、睾酮和皮质醇含量等各项生理指标的变化。研究结果表明,4 周限食显著降低了雄性长爪沙鼠的肥满度和血清甲状腺素T4 含量,显著升高了其血液皮质醇含量;限食后重喂食4 周后可使上述指标恢复或接近正常,但血清白蛋白含量比对照组低,其他指标与对照组无明显差异,长爪沙鼠的一些生理指标在限食重喂食
后能得以恢复,但其内分泌调节可能存在新的变化,值得进一步研究。  相似文献

Sludge in a sequential batch reactor (SBR) system was used to investigate the effect of lead toxicity on metabolisms of polyphosphate accumulating organisms (PAOs) and glycogen accumulating organisms (GAOs) communities fed with acetic acid or glucose as their sole carbon source, respectively. Results showed that the effect of lead on substrate utilization of both PAOs and GAOs was insignificant. However, lead substantially inhibited both of phosphate release and uptake of PAOs. In high concentration of acetic acid trials, an abnormal aerobic phosphate release was observed instead of phosphate uptake and the release rate increased with increasing lead concentration. Results also showed that PAOs could normally synthesize polyhydroxybutyrate (PHB) in the anaerobic phase even though lead concentration was 40 mg L−1. However, they could not aerobically utilize PHB normally in the presence of lead. On the other hand, GAOs could not normally metabolize polyhydroxyvalerate (PHV) in both the anaerobic and aerobic phases.  相似文献

本文研究了硫酸盐还原菌在还原剂的参与下净化含铬电镀废水中间试验的工艺条件,通过净化措施,使废水中Cr(6 )含量由30~40mg/L,下降为0.1mg/L以下,达到了废水排放标准。去除率为99.67~99.97%。分批净化电镀废水103t,试验证明废水净化工艺具有良好的稳定性。而且消除了二次污染。  相似文献

聚3-羟基丁酸乳酸酯[Poly(3-hydroxybutyrate-co-lactate), P(3HB-co-LA)],属于聚羟基脂肪酸酯(Polyhydroxyalkanoates,PHA)家族的一员,是一种具有良好生物相容性和可降解性的天然高分子生物材料。文中通过在大肠杆菌中引入来源于富养罗尔斯通氏菌Ralstonia eutropha的β-酮硫解酶、乙酰乙酰Co A还原酶、来源于丙酸梭菌Clostridium propionicum的丙酰Co A转移酶突变体以及荧光假单胞菌Pseudomonas fluorescens strain 2P24来源的PHA合成酶突变体等异源酶,成功实现了一步法利用葡萄糖合成P(3HB-co-LA),其中乳酸组分的摩尔百分比达到1.6%,聚合物含量为83.9wt%。在此基础上,通过敲除辅酶Q8合成所需的黄素异戊烯基转移酶基因(ubi X)来弱化呼吸链水平,从而增强乳酸积累,并进一步缺失乳酸脱氢酶基因(dld)以减少乳酸在发酵后期转化成丙酮酸,最终将P(3HB-co-LA)中乳酸组分的摩尔百分比提高至14.1%,而聚合物含量为81.7wt%。上述实验结果表明,采用弱化呼吸链水平策略可有效提高聚合物中乳酸组分的摩尔百分比,从而提供了一种改变生物合成聚合物中单体组分含量的新思路。  相似文献

为阐明动物应对食物短缺的能量学对策与其自身的代谢水平的关系,测定了不同限食程度下黑线仓鼠的体重、基础代谢率和非颤抖性产热。结果发现,限食使基础代谢率、非颤抖性产热、褐色脂肪组织细胞色素c氧化酶活性降低。90% 限食驯化4 周后,存活率为80% ,60% 限食驯化4 周后,存活率为30% 。低温驯化使黑线仓鼠基础代谢率和非颤抖性产热显著增加,使80% 限食动物的体重和存活率显著降低。高基础代谢率组的摄食量比低基础代谢率组多23.8% ,80% 限食后两组体重降低的幅度和存活率差异不显著。结果表明:高水平的代谢率使黑线仓鼠对食物资源短缺的敏感性增加;支持“代谢率转换假说”,符合“具有储食习性的动物对食物短缺的生理耐受性较低” 的预测。  相似文献

The seed pathogen Pyrenophora semeniperda has demonstrated potential as a mycoherbicidal biocontrol for eliminating persistent seed banks of annual bromes on western North American rangelands. This pathogen exhibits variation in virulence that is related to mycelial growth rate, but direct laboratory tests of virulence on seeds often have low repeatability. We developed a rapid and sensitive high pressure liquid chromatography method for quantification of the phytotoxin cytochalasin B in complex mixtures in order to evaluate its use in virulence screening. All 10 strains tested produced large quantities of this metabolite in solid wheat seed culture, with production varying over a fourfold range (535–2256 mg kg?1). No cytochalasin B was produced in liquid potato dextrose broth culture, showing that its synthesis is strongly dependent on cultural conditions. In a Bromus tectorum coleoptile bioassay, solid culture extracts showed mild toxicity similar to the cytochalasin B standard at a concentration equivalent to 10?4 M cytochalasin B (72–95% of control), whereas at 10?3 M equivalent, the extracts exhibited significantly higher toxicity (8–18% of control) than the cytochalasin B standard (34% of control). This suggests the possible presence of other phytotoxic metabolites. Cytochalasin B production in solid wheat seed culture exhibited the predicted significant negative correlation with mycelial growth rate on potato dextrose agar, but the correlation was not very strong, possibly because cytochalasin B production and growth rate were measured under different cultural conditions.  相似文献

Stepwise synthetic assembly of polypeptide chains reversibly linked to polyethylene glycol represents a hybrid between traditional solution and solid-phase chemistries and combines the inherent advantages of both approaches. The technical simplicity and scalability of the liquid-phase peptide synthesis method renders it particularly attractive for multiple parallel syntheses, combinatorial approaches and the large-scale preparation of peptides. The versatile protection strategy based on the N alpha-fluorenylmethoxycarbonyl group commonly used in solid-phase peptide synthesis was adapted to the liquid-phase approach. Fluoride ions were used rather than the conventional organic base piperidine for the repetitive amino-deprotection step. Using a range of acid- and base-labile linkers between the polymer and the peptide, it was shown that free and fully side-chain protected peptides can be obtained using our version of the liquid-phase peptide synthesis method. Protocols for simultaneous multiple syntheses requiring a minimum of equipment are presented and the use of polyethylene glycol-bound peptides in biochemical binding and functional assay systems is demonstrated.  相似文献

蛋白质错误折叠循环扩增(protein misfolding cyclic amplification,PMCA)技术,是一种具有感染性的朊蛋白(PrPsc)体外扩增的技术,可用于抑制细胞型朊蛋白(PrPc)向PrPsc转化药物的筛选.本研究在Saborio等提供的方法基础上,成功优化了最佳扩增时间,利用优化的PMCA技术实验了不同剂量氮芥(mechlorethamine,MCT)联合二硫苏糖醇(dithiothreitol,DTT)对PrPsc转化的抑制效应.结果发现,MCT联合DTT能体外抑制PrPc向PrPsc的转化,抑制作用具有明显的量效关系.利用针对人PrPc不同结构域的抗体发现,MCT联用DTT可使抗体6H4的抗原表位隐蔽,该表位位于PrPc第1个α螺旋区内,是构象转化的主要部位.对其机制的深入探讨,将有助于新一类可传播性海绵状脑病(transmissible spongiform encephalopathy,TSE)治疗药物的研发.  相似文献

【背景】根结线虫病害严重制约我国设施蔬菜的生产。丛枝菌根真菌(Arbuscular Mycorrhizal Fungi,AMF)作为土壤中最重要的有益真菌之一,可以促进植物生长,提高植物抗病性,减轻土传真菌和线虫病害的发生。在蔬菜保护地栽培中,AMF对于植物线虫病防治作用的研究受到了广泛关注。【目的】针对番茄生产中危害最严重的南方根结线虫(Meliodogyne incognita)病害问题,研究AMF和番茄品种不同组合的抗线虫效应,以期为菌根真菌作为生物防治剂和生物菌肥应用于实际生产提供技术基础。【方法】在灭菌土壤中,人工接种根结线虫,比较不同菌种Rhizophagus intraradices(Ri)、Acaulosporamellea(Am)及菌种组合Rhizophagusintraradices+Acaulosporamellea(Ri+Am)在不同番茄品种(感病品种蒙特卡罗和抗线虫品种仙客1号)上对南方根结线虫侵染和繁殖的影响,研究AMF对根结线虫的拮抗效应。另外,采用南方根结线虫连作发病的土壤,在感病品种蒙特卡洛上接种AMF混合菌种Ri+Am,番茄苗移栽入连作土壤中,测定各生长指标和调查根结和卵块数量,评价接种AMF处理对根结线虫病的防治效果。【结果】在灭菌土壤中,普通番茄品种蒙特卡罗的菌根效应显著优于抗线虫番茄品种仙客1号,表现为前者单位根重的根结和卵块的数量均比对照显著降低,而后者仅降低了卵块数量;蒙特卡罗上接种Ri+Am混合菌种的效果优于接种单一菌种Am和Ri;而仙客1号上接种Ri的效果更好。接种线虫也显著影响了AMF的侵染,而且对抗性品种仙客1号的影响更为明显。但除了接种Am的处理,大多数处理收获时菌根侵染率仍维持较高的水平(70%以上)。在连作土壤中,感病品种蒙特卡罗接种混合菌种Ri+Am具有较好的抗/耐线虫效应,主要表现为促进植株生长,显著降低根结和卵块数量,但菌根侵染率较灭菌土壤低,约为40%。【结论】综合以上结果,表明菌根化苗能够在一定程度上减轻根结线虫病的危害。土壤灭菌条件下,在感病和抗线虫番茄品种上接种AMF能够减轻线虫的侵染和繁殖,而且在感病品种上的防治效果更加显著。在连作土壤中,在番茄感病品种上接种AMF也表现较好的抗线虫效果。  相似文献

To increase the production of anthraquinone colorants in madder (Rubia akane Nakai) cell culture, the effects of elicitation on the colorant production were investigated. Chitosan was the best biotic elicitor among nine plant derived and microbial derived polysaccharides. When elicited with 25 mg/L chitosan, the total production was increased approximately two times in a seven-day culture as compared to that in the unelicited cells. Anthraquinone production was increased in proportion to the contact period up to day 3. Maximum anthraquinone colorants were obtained with 3-day treatment of chitosan. During chitosan elicitation, the total production was increased 1.3 times in MS medium containing galactose as compared to that containing sucrose. The degree of deacetylation in chitosan and the use of growth regulator or addition of precursor did not affect the production of anthraquinone colorants. When madder cells were elicited at optimum condition, anthraquinone concentration and specific anthraquinone content increased 1.3 times (0.69 g/L) and 2.2 times (0.32 g/g DCW), respectively.  相似文献

To determine circulating angiotensin-(1-7) [Ang-(1,7)] levels in rats with different angiotensin converting enzyme (ACE) genotypes and to evaluate the effect of hypertension on levels of this heptapeptide, plasma levels of angiotensin II (Ang II) and Ang-(1-7) were determined by HPLC and radioimmunoassay in (a) normotensive F0 and F2 homozygous Brown Norway (BN; with high ACE) or Lewis (with low ACE) rats and (b) in hypertensive F2 homozygous male rats (Goldblatt model). Genotypes were characterized by PCR and plasma ACE activity measured by fluorimetry. Plasma ACE activity was 2-fold higher (p < 0.05) in homozygous BN compared to homozygous Lewis groups. In the Goldblatt groups, a similar degree of hypertension and left ventricular hypertrophy was observed in rats with both genotypes. Plasma Ang II levels were between 300-400% higher (p < 0.05) in the BN than in the Lewis rats, without increment in the hypertensive animals. Plasma Ang-(1-7) levels were 75-87% lower in the BN rats (p < 0.05) and they were significantly higher (p < 0.05) in the hypertensive rats from both genotypes. Plasma levels of Ang II and Ang-(1-7) levels were inversely correlated in the normotensive rats (r = -0.64; p < 0.001), but not in the hypertensive animals. We conclude that there is an inverse relationship between circulating levels of Ang II and Ang-(1-7) in rats determined by the ACE gene polymorphism. This inverse relation is due to genetically determined higher ACE activity. Besides, plasma levels of Ang-(1-7) increase in renovascular hypertension.  相似文献

The objective of this study was to investigate the effect of G870A gene polymorphism of CCND1 on the formation and behavioral features of prolactinomas. One hundred and thirteen patients with prolactinoma and 108 age and gender matched control were included in the study. The patients were divided into two groups as noninvasive and invasive tumors. CCND1 G870A gene polymorphism was compared in patients/control and invasive/noninvasive groups. A and G allele frequencies were found as 41.7% and 58.3% in the controls, and 61.1% and 38.9% in the patients (p<0.01). Rates of G/G, G/A and A/A genotypes were found as 11.8%, 55.9% and 32.4% in the noninvasive group, and 15.6%, 44.4% and 40.0% in the invasive group, respectively. Differences between patient and control groups were significant but were not between invasive and noninvasive groups in terms of the allele frequencies and genotype distribution. Mean tumor size and serum levels of prolactin at the time of diagnosis and change in these values after the treatment were not found statistically significant in genotype subgroups. CCND1 G870A gene polymorphism may be an important factor in the early stages of the tumor formation. However, it did not affect the features of the tumor.  相似文献
