Cyanophycin synthesis is catalyzed by cyanophycin synthetase (CphA). It was believed that CphA requires l-aspartic acid (Asp), l-arginine (Arg), ATP, Mg2+, and a primer (low-molecular mass cyanophycin) for cyanophycin synthesis and catalyzes the elongation of a low-molecular mass cyanophycin. Despite extensive studies of cyanophycin, the mechanism of primer supply is still unclear, and already-known CphAs were primer-dependent enzymes. In the present study, we found that recombinant CphA from Thermosynechococcus elongatus BP-1 (Tlr2170 protein) catalyzed in vitro cyanophycin synthesis in the absence of a primer. The Tlr2170 protein showed strict substrate specificity toward Asp and Arg. The optimum pH was 9.0, and Mg2+ or Mn2+ was essential for cyanophycin synthesis. KCl enhanced the cyanophycin synthesis activity of the Tlr2170 protein; in contrast, dithiothreitol did not. The Tlr2170 protein appeared to be a 400 ± 9 kDa homo-tetramer. The Tlr2170 protein showed thermal stability and retained its 80% activity after a 60-min incubation at 50°C. In addition, we examined cyanophycin synthesis at 30°C, 40°C, 50°C, and 60°C. SDS-PAGE analysis showed that the molecular mass of cyanophycin increased with increased reaction temperature.  相似文献

Abstract

In this context, carboxymethyl cellulase (CMCase) production from Glutamicibacter arilaitensis strain ALA4 was initially optimized by one factor at a time (OFAT) method using goat dung as proficient feedstock. Two-level full factorial design (25 factorial matrix) using first-order polynomial model revealed the significant (p?<?0.05) influence of pH, moisture, and peptone on CMCase activity. Central composite design at N?=?20 was further taken into account using a second-order polynomial equation, and thereby liberated maximum CMCase activity of 4925.56?±?31.61?U/g in the goat dung medium of pH 8.0 and 100% moisture containing 1% (w/w) peptone, which was approximately two fold increment with respect to OFAT method. Furthermore, the partially purified CMCase exhibited stability not only at high pH and temperature but also in the presence of varied metal ions, organic solvents, surfactants, and inhibitors with pronounced residual activities. The enzymatic hydrolysis using partially purified CMCase depicted the maximum liberation of fermentable sugars from alkali pretreated lignocellulosic wastes biomass in the order of paddy straw (13.8?±?0.15?mg/g)?>?pomegranate peel (9.1?±?0.18?mg/g)?>?sweet lime peel (8.37?±?0.16?mg/g), with saccharification efficiency of 62.1?±?0.8, 40.95?±?0.4, and 37.66?±?0.4%, respectively after 72?hr of treatment.  相似文献

Abstract

Sequential optimization of propionate production using apple pomace was studied. All experiments were performed in a static flask in anaerobic conditions. Effect of apple pomace as nitrogen source against conventional N sources (yeast extract, peptone) was studied. The double increase was observed in propionic acid production while using yeast extract and peptone (0.29?±?0.01?g/g), as against the use of only apple pomace extract (APE) (0.14?±?0.01?g/g). Intensification of propionic acid fermentation was also achieved by increasing the pH control frequency of the culture medium from 24-(0.29?±?0.01?g/g) to 12-hour intervals (30?°C) (0.30?±?0.02?g/g) and by increasing the temperature of the culture from 30 to 37?°C (12-hour intervals of pH control) (0.32?±?0.01?g/g). An important factor in improving the parameters of fermentation was the addition of biotin to the medium. The 0.2?mg/L dose of biotin allowed to attain 7.66?g/L propionate with a yield of 0.38?±?0.03?g/g (12-hour intervals of pH control, 37?°C).  相似文献

1. The release of total phosphorus (TP) and nitrogen (N in ammonium) was measured for the five most abundant fish species (>85% of biomass) in Mouse and Ranger Lakes, two biomanipulated, oligotrophic lakes in Ontario. 2. The specific release rate of both nutrients was significantly related to fish mass; log10 TP release rate (μg h?1) = 0.793 (±0.109) [log10 wet mass (g)] + 0.7817 (±0.145), and log10 N release rate (μg h?1) = 0.6946 (±0.079) [log10wet mass (g)] + 1.7481 (±0.108). 3. When fish nutrient release was standardized for abundance (all populations, 1993–95) and epilimnetic volume, fish were estimated to contribute 0.083 (±0.061) μg TP L?1 day?1, and 0.41 (±0.17) μg N L?1 day?1 in Mouse L., and 0.062 (±0.020) μg TP L?1 day?1 and 0.31 (±0.08) μg N L?1 day?1 in Ranger L. 4. In comparison, concurrent rates of total planktonic P regeneration were 1.02 (±0.45) μg L?1 day?1 (Mouse L.) and 0.85 (±0.19) μg L?1 day?1 (Ranger L.). Fish represented 8% of planktonic P release in Mouse L. and 7% in Ranger L. 5. Fish dry mass had mean elemental body compositions of 39.3% carbon, 10.9% nitrogen, and 4.0% phosphorus (all fish combined), with a mean molar C : N : P ratio of 27 : 6 : 1. This comprised about 55% and 23% of the total epilimnetic particulate P and N respectively. 6. Turnover times of P and N in fish were approximately 103 and 48 days respectively. In comparison, planktonic turnover times of particulate P in Mouse and Ranger Lakes were 4.3 and 4.4 days respectively. The ability to synthesize Indole-3-acetic acid (IAA) is widely associated with the plant growth promoting rhizobacteria (PGPR). The present work deals with isolation and characterization of such bacteria from the rhizosphere of medicinal plant Stevia rebaudiana and optimization of IAA production from its isolates. The optimization of IAA production was carried out at different pH and temperature with varied carbon and nitrogen sources of culture media. Out of different isolates obtained, three of them were screened as efficient PGPRs on the basis of different plant growth promoting attributes. Isolates CA1001 and CA2004 showed better production of IAA at pH 9 (91.7?µg?ml?1) and at temperature 37?°C (81.7?µg?ml?1). Dextrose (1%) was found to be the best carbon source for isolate CA1001 with 104?µg?ml?1 IAA production. Isolate CA 2004 showed best production of IAA 36?µg?ml?1 and 34?µg?ml?1 at 1.5% and 1% Beef extract as nitrogen source respectively. Isolate CA 1001 showed 32?µg?ml?1 IAA production at 0.5% nicotinic acid concentration. From the current study, CA1001 and CA2004 emerged as noble alternatives for IAA production further which also resulted in root and shoot biomass generation in crop plants, hence can be further used as bio-inoculants for plant growth promotion.  相似文献

Aims: Peptones are one of the most expensive constituents of microbial media. This study was undertaken to prepare the peptone from waste chicken feathers through a new process. Methods and Results: The chemical analysis of chicken feather peptone (CFP) was performed. The ability of CFP to support the growth of the three test bacteria in liquid and agar media was comparable to those of three commercial peptones [tryptone peptone (TP), fish peptone and protease peptone (PP)]. Conclusions: CFP was found to be rich in ash (42·1 g 100 g?1), protein (55·8 g 100 g?1) and mineral contents. The maximum biomass yield (3·13 g l?1) and colony number (83 × 108 CFU ml?1) for bacterium Bacillus subtilis were attained with CFP. The maximum biomass yields and colony numbers for Lactobacillus delbrueckii ssp. bulgaricus and Escherichia coli were reached in TP medium. Second high biomass yield (2·64 g l?1) and colony number (75 × 108 CFU ml?1) for E. coli were achieved using CFP. Third high biomass yield (1·29 g l?1) and colony number (90 × 107 CFU ml?1) for Lact. delbrueckii ssp. bulgaricus were obtained in CFP medium. Significance and Impact of the Study: Usability of waste chicken feathers as substrate for bacteria was investigated for the first time in the present study. The peptone may be used in industrial fermentations for production of antibiotics, organic acids, enzymes and biopolymer. It may be also used in clinical microbiology. A new chemical process was developed for peptone preparation. This process may be also employed for peptone preparation from other organic materials, especially fibrose protein‐containing materials.  相似文献

Switchgrass (Panicum virgatum L.) has gained importance as feedstock for bioenergy over the last decades due to its high productivity for up to 20 years, low input requirements, and potential for carbon sequestration. However, data on the dynamics of CO2 exchange of mature switchgrass stands (>5 years) are limited. The objective of this study was to determine net ecosystem exchange (NEE), ecosystem respiration (Re), and gross primary production (GPP) for a commercially managed switchgrass field in its sixth (2012) and seventh (2013) year in southern Ontario, Canada, using the eddy covariance method. Average NEE flux over two growing seasons (emergence to harvest) was ?10.4 μmol m?2 s?1 and reached a maximum uptake of ?42.4 μmol m?2 s?1. Total annual NEE was ?380 ± 25 and ?430 ± 30 g C m?2 in 2012 and 2013, respectively. GPP reached ?1354 ± 23 g C m?2 in 2012 and ?1430 ± 50g C m?2 in 2013. Annual Re in 2012 was 974 ± 20 g C m?2 and 1000 ± 35 g C m?2 in 2013. GPP during the dry year of 2012 was significantly lower than that during the normal year of 2013, but yield was significantly higher in 2012 with 1090 g  m?2, compared to 790 g m?2 in 2013. If considering the carbon removed at harvest, the net ecosystem carbon balance came to 106 ± 45 g C  m?2 in 2012, indicating a source of carbon, and to ?59 ± 45 g C m?2 in 2013, indicating a sink of carbon. Our results confirm that switchgrass can switch between being a sink and a source of carbon on an annual basis. More studies are needed which investigate this interannual variability of the carbon budget of mature switchgrass stands.  相似文献

Gaeolaelaps aculeifer (Canestrini, 1883) is a soil-dwelling predatory mite with potential for use as a biological control agent of fungus gnats (Diptera: Sciaridae) in mushroom production. The life table, predation rate and population growth rate of G. aculeifer on a diet of larvae of the sciarid fly, Lycoriella auripila, at 23?±?1°C, 60?±?5% RH and a photoperiod of 0:24 (L:D)?h was investigated. The results revealed that the duration of egg, larva, protonymph, deutonymph, females and males of G. aculeifer were 3.8?±?0.1, 1.4?±?0.1, 3.9?±?0.1, 4.1?±?0.1, 67.7?±?2.8 and 60.3?±?3.1 days, respectively. Net reproductive rate (R0) was 54.8?±?7.1 offspring, intrinsic rate of increase (r) was 0.12?±?0.01 offspring day?1, finite rate of increase (λ) was 1.13?±?0.01 day?1and mean generation time (T) was 32.3?±?0.6 days. The predator consumed a mean of 0.08?±?0.05, 1.73?±?0.18, 3.16?±?0.28 and 75.9?±?7.1 third instar L. auripila larvae during the larval (1.3?±?0.1 days), protonymph (3.9?±?0.1 days), deutonymph (4.1?±?0.1 days) and adult (52.6?±?2.2 days) stages. Population parameters and consumption rates suggest that G. aculeifer has good potential as a biological control agent of L. auripila in mushroom production.  相似文献

The kinetic study of Arthrospira platensis extracellular polymeric substances (EPS) production under different trophic modes??photoautotrophy (100???mol photons m?2?s?1), heterotrophy (1.5?g/L glucose), and mixotrophy (100???mol photons m?2?s?1 and 1.5?g/L glucose)??was investigated. Under photoautotrophic and heterotrophic conditions, the maximum EPS production 219.61?±?4.73 and 30.30?±?1.97?mg/L, respectively, occurred during the stationary phase. Under a mixotrophic condition, the maximum EPS production (290.50?±?2.21?mg/L) was observed during the early stationary phase. The highest specific EPS productivity (433.62?mg/g per day) was obtained under a photoautotrophic culture. The lowest specific EPS productivity (38.33?mg/g per day) was observed for the heterotrophic culture. The effects of glucose concentration, light intensity, and their interaction in mixotrophic culture on A. platensis EPS production were evaluated by means of 32 factorial design and response surface methodology. This design was carried out with a glucose concentration of 0.5, 1.5, and 2.5?g/L and at light levels of 50, 100, and 150???mol photons m?2?s?1. Statistical analysis of the model demonstrated that EPS concentration and EPS yield were mainly influenced by glucose concentration and that conditions optimizing EPS concentration were dissimilar from those optimizing EPS yield. The highest maximum predicted EPS concentration (369.3?mg/L) was found at 150???mol photons m?2?s?1 light intensity and 2.4?g/L glucose concentration, while the highest maximum predicted EPS yield (364.3?mg/g) was recorded at 115???mol photons m?2?s?1 light intensity and 1.8?g/L glucose concentration.  相似文献

We investigated whether rates of net primary production (NPP) and biomass turnover of floating grasses in a central Amazon floodplain lake (Lake Calado) are consistent with published evidence that CO2 emissions from Amazon rivers and floodplains are largely supplied by carbon from C4 plants. Ground‐based measurements of species composition, plant growth rates, plant densities, and areal biomass were combined with low altitude videography to estimate community NPP and compare expected versus observed biomass at monthly intervals during the aquatic growth phase (January–August). Principal species at the site were Oryza perennis (a C3 grass), Echinochloa polystachya, and Paspalum repens (both C4 grasses). Monthly mean daily NPP of the mixed species community varied from 50 to 96 g dry mass m?2 day?1, with a seasonal average (±1SD) of 64±12 g dry mass m?2 day?1. Mean daily NPP (±1SE) for P. repens and E. polystachya was 77±3 and 34±2 g dry mass m?2 day?1, respectively. Monthly loss rates of combined above‐ and below‐water biomass ranged from 31% to 75%, and averaged 49%. Organic carbon losses from aquatic grasses ranged from 30 to 34 g C m?2 day?1 from February to August. A regional extrapolation indicated that respiration of this carbon potentially accounts for about half (46%) of annual CO2 emissions from surface waters in the central Amazon, or about 44% of gaseous carbon emissions, if methane flux is included.  相似文献

The spore productivity and insecticidal activity of two opportunistic insect pathogenic Aspergillus species (namely: Aspergillus clavatus Desmazieres and Aspergillus flavus Link (Ascomycota: Eurotiales, Trichocomaceae)) were compared to Metarhizium anisopliae sensu lato (Metchnikoff) Sorokin (Ascomycota: Hypocreales, Clavicipitaceae) for mosquito (Diptera: Culicidae) control. The production of aerial spores on wheat bran and white rice was investigated in solid-, semi-solid-, and liquid-state media supplemented with a nutritive solution. Wheat bran-based media increased the spore yield in solid-state from three to sevenfold: A. clavatus produced 48.4?±?5.2 and 15.7?±?1.6?×?108 spores/g, A. flavus produced 22.3?±?4.1 and 3.1?±?2.5?×?108 spores/g, and M. anisopliae produced 39.6?±?6.5 and 13.1?±?2.6?×?108 spores/g of wheat bran or white rice, respectively. A. clavatus, A. flavus and M. anisopliae spores harvested from wheat bran-based solid-state media showed lethal concentrations (LC50) of 1.1, 1.8, and 1.3?×?108 spores/ml against Culex quinquefasciatus Say larvae in 72?h. Because A. clavatus and M. anisopliae displayed similar features when cultured under these conditions, our results suggest that insect pathogenic Aspergillus species may be as productive and virulent against mosquito larvae as a well-recognised entomopathogenic fungus.  相似文献

Abstract

Pullulan is an extracellular water-soluble polysaccharide with wide applications. In this study, we screened strains that could selectively produce high molecular weight pullulan for application in industrial pullulan production. A new fungus strain A4 was isolated from soil and identified as Aureobasidium melanogenum based on colony characteristics, morphology, and internally transcribed spacer analysis. Thin-layer chromatography, Fourier-transform infrared spectroscopy, and nuclear magnetic resonance analysis suggested that the dominant exopolysaccharide produced by this strain, which presented a molecular weight of 1.384?×?106 Dalton in in-gel permeation chromatography, was pullulan. The culture conditions for A. melanogenum A4 were optimized at 30?°C and 180?rpm: carbon source, 50?g/L maltose; initial pH 7; and 8?g/L Tween 80. Subsequently, batch fermentation was performed under the optimized conditions in a 5-L stirred-tank fermentor with a working volume of 3?L. The fermentation broth contained 303?g/L maltose, which produced 122.34?g/L pullulan with an average productivity of 1.0195?g/L/h and 82.32?g/L dry biomass within 120?h. The conversion efficiency of maltose to pullulan (Y%) and specific production rate (g/h/g dry cells) (Qs) reached 40.3% and 0.0251?g/L/g dry cells, respectively. The results showed strain A4 could be a good candidate for industrial production.  相似文献

Cyanophycin is non-ribosomally synthesized protein-like copolymer. Synthesis of cyanophycin is catalyzed by cyanophycin synthetase (CphA). In this study, a novel cyanophycin synthetase CphA49 belonging to NOR5 clade of Gammaproteobacteria was identified with primer-based screening from a deep-sea sediment metagenomic library. The cphA49 gene contained an open reading frame of 2,637 bp and encoded a protein with a predicted molecular mass of 100 kDa. A recombinant CphA49 was obtained by the functional expression of cphA49 in Escherichia coli BL21 (DE3). The biochemical properties of the purified CphA49 were determined. The optimum pH and temperature of the recombinant CphA49 were 9.0 and 40 °C, respectively. The enzyme was stable at temperatures below 40 °C. The recombinant CphA49 exhibited strict primer dependency and broad substrate specificities. Cyanophycin catalyzed by CphA49 exhibited homogenous molecular mass. The amino acid composition of cyanophycin was determined and constitutes arginine, aspartic acid, and lysine.  相似文献

Bioconversion of biodiesel-derived crude glycerol into carotenoids and lipids was investigated by a microbial conversion of an oleaginous red yeast Sporidiobolus pararoseus KM281507. The methanol content in crude glycerol (0.5%, w/v) did not show a significant effect on biomass production by strain KM281507. However, demethanolized crude glycerol significantly supported the production of biomass (8.64?±?0.13?g/L), lipids (2.92?±?0.03?g/L), β-carotene (15.76?±?0.85?mg/L), and total carotenoids (33.67?±?1.28?mg/L). The optimal conditions suggested by central composite design were crude glycerol concentration (55.04?g/L), initial pH of medium (pH 5.63) and cultivation temperature (24.01°C). Under these conditions, the production of biomass, lipids, β-carotene, and total carotenoids were elevated up to 8.83?±?0.05, 4.00?±?0.06?g/L, 27.41?±?0.20, and 53.70?±?0.48?mg/L, respectively. Moreover, an addition of olive oil (0.5???2.0%) dramatically increased the production of biomass (14.47?±?0.15?g/L), lipids (6.40?±?0.09?g/L), β-carotene (54.43?±?0.95?mg/L), and total carotenoids (70.92?±?0.51?mg/L). The oleic acid content in lipids was also increased to 75.1% (w/w) of total fatty acids, indicating a good potential to be an alternative biodiesel feedstock. Meanwhile, the β-carotene content in total carotenoids was increased to 76.7% (w/w). Hence, strain KM281507 could be a good potential source of renewable biodiesel feedstock and natural carotenoids.  相似文献

This randomized and controlled trial investigated whether the increase in elite training at different altitudes altered the oxidative stress biomarkers of the nervous system. This is the first study to investigate four F4-neuroprostanes (F4-NeuroPs) and four F2-dihomo-isoprostanes (F2-dihomo-IsoPs) quantified in 24-h urine. The quantification was carried out by ultra high pressure liquid chromatography-triple quadrupole-tandem mass spectrometry (UHPLC-QqQ-MS/MS). Sixteen elite triathletes agreed to participate in the project. They were randomized in two groups, a group submitted to altitude training (AT, n?=?8) and a group submitted to sea level training (SLT) (n?=?8), with a control group (Cg) of non-athletes (n?=?8). After the experimental period, the AT group triathletes gave significant data: 17-epi-17-F2t-dihomo-IsoP (from 5.2?±?1.4?μg/mL 24?h?1 to 6.6?±?0.6?μg/mL 24?h?1), ent-7(RS)-7-F2t-dihomo-IsoP (from 6.6?±?1.7?μg/mL 24?h?1 to 8.6?±?0.9?μg/mL 24?h?1), and ent-7-epi-7-F2t-dihomo-IsoP (from 8.4?±?2.2?μg/mL 24?h?1 to 11.3?±?1.8?μg/mL 24?h?1) increased, while, of the neuronal degeneration-related compounds, only 10-epi-10-F4t-NeuroP (8.4?±?1.7?μg/mL 24?h?1) and 10-F4t-NeuroP (5.2?±?2.9?μg/mL 24?h?1) were detected in this group. For the Cg and SLT groups, no significant changes had occurred at the end of the two-week experimental period. Therefore, and as the main conclusion, the training at moderate altitude increased the F4-NeuroPs- and F2-dihomo-isoPs-related oxidative damage of the central nervous system compared to similar training at sea level.  相似文献

In this study we maximized biomass production by the basidiomycete Ganoderma australe ATHUM 4345, a species of pharmaceutical interest as it is a valuable source of nutraceuticals, including dietary fibers and glucans. We used the Biolog FF MicroPlate to screen 95 different carbon sources for growth monitoring. The pattern of substrate catabolism forms a substrate assimilation fingerprint, which is useful in selecting components for media optimization of maximum biomass production. Response surface methodology, based on the central composite design was applied to explore the optimum concentrations of carbon and nitrogen sources of culture medium in shake flask cultures. When the improved culture medium was tested in a 20‐L stirred tank bioreactor, using 13.7 g/L glucose and 30.0 g/L yeast extract, high biomass yields (10.1±0.4 g/L) and productivity of 0.09 g L?1 h?1 were obtained. The yield coefficients for total glucan and dietary fibers on biomass formed were 94.82±6 and 341.15±12.3 mg/g mycelium dry weight, respectively.  相似文献

Abstract

The production of polyhydroxyalkanoates (PHA) using digestate of chicken manure combined with waste sunflower oil as no-cost feedstocks in a multi-stage process was investigated. Using Cupriavidus necator H16 in combined culture media, a maximum PHA accumulation of 4.6?±?0.2?g/L at 75.1?±?1.4% of cell dry matter and a residual cell matter yield of 1.5?±?0.1?g/L were obtained after 96?hr of cultivation (30?°C, 160?rpm, pH 7.0) in flask-based experiments. Manure was acidogenically fermented in a continuous stirring tank reactor in fed-batch mode. The bioreactor was operated at varying organic loading rates (OLR) and hydraulic retention times (HRT) ranging from 1–4?g volatile solids (VS)/L/d and 4–8?days, respectively. Optimal operation was observed at an OLR of 4?g VS/L/d and an HRT of 4?days. Analysis showed the presence of significant amounts of ammonia, potassium, magnesium, calcium, and trace elements, i.e. Fe, Cu, Ni, Mn, Co, Zn, Cr in the digestate. The micro-filtered digestate was utilized as a complex culture media base while waste oil served as an additional carbon source and supplemented for effective PHA production. The total volatile fatty acid content of digestate greatly affected the growth performance of the PHA-producing microorganism Cupriavidus necator H16.  相似文献

Xanthan gum is an important commercial polysaccharide produced by Xanthomonas species. In this study, xanthan production was investigated using a local isolate of Xanthomonas campestris MO-03 in medium containing various concentrations of chicken feather peptone (CFP) as an enhancer substrate. CFP was produced with a chemical process and its chemical composition was determined. The addition of CFP (1–8?g/l) increased the conversion of sugar to xanthan gum in comparison with the control medium, which did not contain additional supplements. The highest xanthan production (24.45?g/l) was found at the 6?g/l CFP containing control medium in 54?h. This value was 1.73 fold higher than that of control medium (14.12?g/l). Moreover, addition of CFP improved the composition of xanthan gum; the pyruvate content of xanthan was 3.86% (w/w), higher than that of the control (2.2%, w/w). The xanthan gum yield was also influenced by the type of organic nitrogen sources. As a conclusion, CFP was found to be a suitable substrate for xanthan gum production.  相似文献

Summary Effects of nutritional factors on exopolysaccharide production by submerged cultivation of the medicinal mushroom Oudemansiella radicata were investigated in shake flasks. Sucrose and peptone were optimal carbon and nitrogen sources for cell growth and exopolysaccharide production. The exopolysaccharide production was increased with an increase in initial sucrose concentration within the range of 10–40 g l−1 and initial peptone concentration within the range of 1–3 g l−1. To enhance further exopolysaccharide production, the effect of carbon/nitrogen ratios was studied using central composite design (CCD) and response surface analysis. The maximum exopolysaccharide production of 2.67 ± 0.15 g l−1 was achieved in medium with optimized carbon and nitrogen sources, i.e. 39.3 g sucrose l−1 and 3.16 g peptone l−1 in the same cultivation conditions. The information obtained is helpful for the hyperproduction of exopolysaccharide by submerged cultivation of O. radicata on a large scale.  相似文献

Lipid homeostasis is well-known in oleaginous yeasts, but there are few non-oleaginous yeast models apart from Saccharomyces cerevisiae. We are proposing the non-oleaginous yeast Candida zeylanoides QU 33 as model. The aim of this study was to investigate the influence of the carbon/nitrogen ratio and the type of nitrogen source upon oil accumulation by this yeast grown on shake flask cultures. The maximum biomass was obtained in yeast extract (2.39?±?0.19 g/l), followed by peptone (2.24?±?0.05 g/l), while the highest content of microbial oil (0.35?±?0.01 g/l) and the maximum lipid yield (15.63 %) were achieved with peptone. Oleic acid was the predominant cellular fatty acid in all culture media (>32.23 %), followed by linoleic (>15.79 %) and palmitic acids (>13.47 %). The highest lipid yield using glucose and peptone was obtained at the C/N ratio of 200:1.  相似文献
