Increasing contamination and higher enrichment ratio of non-essential heavy metal cadmium (Cd) induce various toxic responses in plants when accumulated above the threshold level. These effects and growth responses are genotype and Cd level dependent. An experiment was conducted to analyze the effect of Cd toxicity in Brassica juncea [L] Czern and Coss by selecting its two varieties Varuna and RH-30. Cadmium (0, 25, 50 or 100 mg CdCl2 kg−1 of soil) fed to soil decreased the values of growth characteristics, activity of nitrate reductase and leaf water potential, whereas activities of antioxidant enzymes and proline content increased with the increasing concentration of Cd, observed at 30 and 60 day stages of growth, in both the varieties. Moreover, Cd uptake by the roots was higher in RH-30 than Varuna. Also the activity of antioxidant enzymes and proline accumulation were higher in Varuna with increasing soil level of Cd. Out of the two varieties, Varuna was more tolerant than RH-30 to Cd stress.  相似文献

Cadmium (Cd) is often associated with reproductive disorders of mammals. Edible bird’s nest (EBN) is a natural food product made of swiftlet''s salivary secretion used to make their nests and it has been consumed as a tonic food for decades. This research aimed to study the protective effects of EBN against Cd-induced uterine toxicity in Sprague Dawley rats. Thirty (30) female Sprague Dawley rats were assigned into five groups as follows: group 1- negative control (NC) received distilled water; group 2 - positive control (PC) administered with CdCl2, 5 mg/kg BW; while groups EBN-1, EBN-2, and EBN-3 received CdCl2 (5 mg/kg BW) plus graded concentrations of 60, 90 and 120 mg/kg BW of EBN, respectively. After four weeks of daily oral treatment, rats were euthanized to collect the uterus for evluations of histopathological changes, Cd concentrations and Metallothionein (MT) expressions using H&E stain, inductive coupled plasma mass spectrometry (ICP-MS) and immunohistochemistry, respectively. Blood samples were collected for superoxide dismutase (SOD) analysis using SOD assay kit. Results revealed that the CdCl2 without EBN supplement (PC) group had elevated levels of Cd in the uterus along with increased MT expressions and decreased SOD enzyme activity as compared to the NC group. Moreover, uterine histopathological changes, including glandular cysts and loss of normal structure of luminal epithelium (LE) and glandular epithelium (GE) were found in the PC group. Interestingly, groups treated with CdCl2 along with EBN (EBN1, EBN2, EBN3) showed lower levels of uterine tissue Cd deposition and MT expression, lower degenerative changes with normal histomorphology of glands, and increased SOD activity as compared to the PC group. Overall, the findings revealed that oral exposure to Cd at a dose of 5 mg/kg BW resulted in significant alterations in the rat''s uterus. However, the toxicity effect was averted by EBN treatment in a dose dependant manner; highest protection achieved with EBN 120 mg/kg BW, through a possible detoxification mechanism and prevention of Cd deposition.  相似文献

The effects of different CdCl2 concentrations on the growth and on certain biochemical parameters of almond seedlings (Prunus dulcis) were studied under controlled conditions in the nutrient solutions containing increasing CdCl2 concentrations ranging from 0 to 150 μM CdCl2. Under Cd stress conditions, damage was variable. Cadmium reduced dry matter production in leaves and roots. While chlorophyll content was severely decreased, that of leaf sugars appeared to be increased. Furthermore, leaf nutritional status seemed to be more altered than that of roots. Both in roots and leaves, there was an increase in MDA content as metal concentration increased. It may be suggested from the present study that toxic concentrations of Cd cause oxidative damage as shown by the increase of lipid peroxidation.  相似文献

Cadmium (Cd) in soil–plant system can abridge plant growth by initiating alterations in root zones. Hydroponics and rhizoboxes are useful techniques to monitor plant responses against various natural and/or induced metal stresses. However, soil based studies are considered more appropriate in order to devise efficient food safety and remediation strategies. The present research evaluated the Cd-mediated variations in elemental dynamics of rhizospheric soil together with in planta ionomics and morpho-physio-biochemical traits of two differentially Cd responsive maize cultivars. Cd-sensitive (31P41) and Cd-tolerant (3062) cultivars were grown in pots filled with 0, 20, 40, 60 and 80 µg/kg CdCl2 supplemented soil. The results depicted that the maize cultivars significantly influenced the elemental dynamics of rhizosphere as well as in planta mineral accumulation under applied Cd stress. The uptake and translocation of N, P, K, Ca, Mg, Zn and Fe from rhizosphere and root cell sap was significantly higher in Cd stressed cv. 3062 as compared to cv. 31P41. In sensitive cultivar (31P41), Cd toxicity resulted in significantly prominent reduction of biomass, leaf area, chlorophyll, carotenoids, protein contents as well as catalase activity in comparison to tolerant one (3062). Analysis of tolerance indexes (TIs) validated that cv. 3062 exhibited advantageous growth and efficient Cd tolerance due to elevated proline, phenolics and activity of antioxidative machinery as compared to cv. 31P41. The cv. 3062 exhibited 54% and 37% less Cd bio-concentration (BCF) and translocation factors (TF), respectively in comparison to cv. 31P41 under highest Cd stress regime. Lower BCF and TF designated a higher Cd stabilization by tolerant cultivar (3062) in rhizospheric zone and its potential use in future remediation plans.  相似文献

Cadmium (Cd) is one of the most dangerous environmental pollutants, among other things, affecting plant mineral composition. Thus, in this study, we investigated the changes in potassium (K) concentration in Cd-treated rice (Oryza sativa L.) seedlings of two cultivars. On treatment with 5 μM CdCl2, the Cd concentration increased in the shoot and roots of Cd-sensitive cultivar (cv. Taichung Native 1, TN1) but not or slightly in the Cd-tolerant cultivar (cv. Tainung 67, TNG67). The decrease in K concentration in the shoot and roots of TN1 caused by Cd was more pronounced than that of TNG67. Exogenous addition of KCl decreased Cd concentration and reduced Cd toxicity of TN1 seedlings. Evidence presented in this study suggests that the improvement of K status is able to reduce toxicity of rice seedlings to CdCl2.  相似文献

The synthesis of Cd-binding peptides (CdBPs) was induced upon addition of 20 micromolar CdCl2 (nonphytotoxic level) to the nutrient solution of hydroponically grown tobacco seedlings (Nicotiana rustica var Pavonii). Amino acid analysis showed that the main components were γ-(Glu-Cys)3-Gly and γ-(Glu-Cys)4-Gly. Seedlings exposed to the metal for 1 week contained similar glutathione levels as found in the controls (about 0.18 micromole per gram fresh weight). If, as has been proposed, CdBPs are involved in Cd-detoxification by chelation, both metal and ligand must be localized in the same cellular compartment. To directly determine the localization of Cd and CdBPs, protoplasts and vacuoles were isolated from leaves of Cd-exposed seedlings. Purified vacuoles contained virtually all of the CdBPs and Cd found in protoplasts (104% ± 8 and 110% ± 8, respectively). CdBPs were associated with the vacuolar sap and not with the tonoplast membrane. Glutathione was observed in leaves and protoplasts but not in vacuoles. The probability that CdBPs are synthesized extravacuolarly and our finding that they and Cd are predominantly located in the vacuole suggest that these molecules might be involved in transport of Cd to the vacuole. Our results also suggest that a simple cytoplasmic chelator role for CdBPs in Cd tolerance cannot be assumed.  相似文献

The objective of the present study was to investigate the effects of cadmium-zinc (Cd-Zn) interactions on their uptake, oxidative damage of cell macromolecules (lipids, proteins, DNA) and activities of antioxidative enzymes in tobacco seedlings as well as roots and leaves of adult plants. Seedlings and plants were exposed to Cd (10 µM and 15 µM) and Zn (25 µM and 50 µM) as well as their combinations (10 µM or 15 µM Cd with either 25 µM or 50 µM Zn). Measurement of metal accumulation exhibited that Zn had mostly positive effect on Cd uptake in roots and seedlings, while Cd had antagonistic effect on Zn uptake in leaves and roots. According to examined oxidative stress parameters, in seedlings and roots individual Cd treatments induced oxidative damage, which was less prominent in combined treatments, indicating that the presence of Zn alleviates oxidative stress. However, DNA damage found in seedlings, and lower glutathione reductase (GR) and superoxide dismutase (SOD) activity recorded in both seedlings and roots, after individual Zn treatments, indicate that Zn accumulation could impose toxic effects. In leaves, oxidative stress was found after exposure to Cd either alone or in combination with Zn, thus implying that in this tissue Zn did not have alleviating effects. In conclusion, results obtained in different tobacco tissues suggest tissue-dependent Cd-Zn interactions, which resulted in activation of different mechanisms involved in the protection against metal stress.  相似文献

The protective effect of selenium (Se) on antioxidant defense and methylglyoxal (MG) detoxification systems was investigated in leaves of rapeseed (Brassica napus cv. BINA sharisha 3) seedlings under cadmium (Cd)-induced oxidative stress. Two sets of 11-day-old seedlings were pretreated with both 50 and 100???M Se (Na2SeO4, sodium selenate) for 24?h. Two concentrations of CdCl2 (0.5 and 1.0?mM) were imposed separately or on the Se-pretreated seedlings, which were grown for another 48?h. Cadmium stress at any levels resulted in the substantial increase in malondialdehyde and H2O2 levels. The ascorbate (AsA) content of the seedlings decreased significantly upon exposure to Cd stress. The amount of reduced glutathione (GSH) increased only at 0.5?mM CdCl2, while glutathione disulfide (GSSG) increased at any level of Cd, with concomitant decrease in GSH/GSSG ratio. The activities of ascorbate peroxidase (APX) and glutathione S-transferase (GST) increased significantly with increased concentration of Cd (both at 0.5 and 1.0?mM CdCl2), while the activities of glutathione reductase (GR) and glutathione peroxidase (GPX) increased only at moderate stress (0.5?mM CdCl2) and then decreased at 1.0?mM severe stress (1.0?mM CdCl2). Monodehydroascorbate reductase (MDHAR), dehydroascorbate reductase (DHAR), catalase (CAT), glyoxalase I (Gly I), and glyoxalase II (Gly II) activities decreased upon exposure to any levels of Cd. Selenium pretreatment had little effect on the nonenzymatic and enzymatic components of seedlings grown under normal conditions; i.e., they slightly increased the GSH content and the activities of APX, GR, GST, and GPX. On the other hand, Se pretreatment of seedlings under Cd-induced stress showed a synergistic effect; it increased the AsA and GSH contents, the GSH/GSSG ratio, and the activities of APX, MDHAR, DHAR, GR, GPX, CAT, Gly I, and Gly II which ultimately reduced the MDA and H2O2 levels. However, in most cases, pretreatment with 50???M Se showed better results compared to pretreatment with 100???M Se. The results indicate that the exogenous application of Se at low concentrations increases the tolerance of plants to Cd-induced oxidative damage by enhancing their antioxidant defense and MG detoxification systems.  相似文献

Cadmium (Cd) accumulation in durum wheat grain is a growing concern. Among the factors affecting Cd accumulation in plants, soil chloride (Cl) concentration plays a critical role. The effect of leaf NaCl application on grain Cd was studied in greenhouse-grown durum wheat (Triticum turgidum L. durum, cv. Balcali-2000) by immersing (10 s) intact flag leaves into Cd and/or NaCl-containing solutions for 14 times during heading and dough stages. Immersing flag leaves in solutions containing increasing amount of Cd resulted in substantial increases in grain Cd concentration. Adding NaCl alone or in combination with the Cd-containing immersion solution promoted accumulation of Cd in the grains, by up to 41%. In contrast, Zn concentrations of grains were not affected or even decreased by the NaCl treatments. This is likely due to the effect of Cl complexing Cd and reducing positive charge on the metal ion, an effect that is much smaller for Zn. Charge reduction or removal (CdCl20 species) would increase the diffusivity/lipophilicity of Cd and enhance its capability to penetrate the leaf epidermis and across membranes. Of even more significance to human health was the ability of Cl alone to penetrate leaf tissue and mobilize and enhance shoot Cd transfer to grains, yet reducing or not affecting Zn transfer.  相似文献

Parsley (Petroselinum hortense L.) plants cultivated under controlled conditions were exposed to different doses of cadmium to investigate the antioxidant capacity and cadmium accumulation in the samples. Two-months-old parsley seedlings were treated with four different concentrations of CdCl2 (0, 75, 150, and 300 μM) for fifteen days. Cadmium level in leaves increased significantly by increasing the Cd contamination in the soil. Total chlorophyll and carotenoid content declined considerably with Cd concentration. Cd treatment caused a significant increase lipid peroxidation in tissue of leaf. Superoxide dismutase activity (SOD, EC 1.15.1.1) increased partially at 75 and 150 μM CdCl2 concentrations whereas the activity decreased at 300 μM CdCl2. Catalase (CAT, EC 1.11.1.6) and ascorbate peroxidase (APX, EC 1.11.1.11) activities were reduced by Cd application. Total phenolic compound amount increased significantly with increasing Cd concentration, as ferric reduction power, superoxide anion radical, and DPPH˙ free radical scavenging activities elevated slightly by the concentration. These results suggest that antioxidant enzymes activities were repressed depending on accumulation of cadmium in leaves of parsley while the non-enzymatic antioxidant activities slightly increased.  相似文献

Cadmium (Cd) exerts a detrimental effect on the metabolism of plants, whereas selenium (Se) may protect them against various stressors through its antioxidative activity. In this in vitro study we investigated the impact of Se (2 µM Na2SeO4) on the growth, nutrient (P, S, K, Ca, Mg, B, Mn, Fe and Zn) concentrations and cell integrity of rape (Brassica napus oleifera) and two wheat (Triticum aestivum) genotypes subjected to Cd stress (600 µM CdCl2). Rape accumulated both Cd and Se more than did wheat. In all plants, Cd markedly reduced the biomass, enhanced lipid peroxidation and diminished plasmalemma fluidity. A drop in the K uptake and the reduced plasmalemma permeability diminished the K efflux from the leaf cells. In contrast, Cd elevated S concomitantly with Zn, indicating an activity of detoxifying SH groups and SOD isoenzymes. When added alone, Se promoted the growth of all plants, it enhanced the accumulation of S, but the impact on other nutrients remained minor. In Cd-stressed plants, Se tended to counterbalance the Cd-induced changes in nutrients, it also reduced the lipid peroxidation and exerted positive effects on the cell membrane stability. The Cd stress and the protective role of Se were most evident in rape. The Finnish wheat genotype was less tolerant to Cd than the Polish one.  相似文献

Cadmium (Cd) is highly toxic metal for plant metabolic processes even in low concentration due to its longer half-life and non-biodegradable nature. The current study was designed to assess the bioremediation potential of a Cd-tolerant phytobeneficial bacterial strain Bacillus sp. SDA-4, isolated, characterized and identified from Chakera wastewater reservoir, Faisalabad, Pakistan, together with spinach (as a test plant) under different Cd regimes. Spinach plants were grown with and without Bacillus sp. SDA-4 inoculation in pots filled with 0, 5 or 10 mg kg−1 CdCl2-spiked soil. Without Bacillus sp. SDA-4 inoculation, spinach plants exhibited reduction in biomass accumulation, antioxidative enzymes and nutrient retention. However, plants inoculated with Bacillus sp. SDA-4 revealed significantly augmented growth, biomass accumulation and efficiency of antioxidative machinery with concomitant reduction in proline and MDA contents under Cd stress. Furthermore, application of Bacillus sp. SDA-4 assisted the Cd-stressed plants to sustain optimal levels of essential nutrients (N, P, K, Ca and Mg). It was inferred that the characterized Cd-tolerant PGPR strain, Bacillus sp. SDA-4 has a potential to reduce Cd uptake and lipid peroxidation which in turn maintained the optimum balance of nutrients and augmented the growth of Cd-stressed spinach. Analysis of bioconcentration factor (BCF) and translocation factor (TF) revealed that Bacillus sp. SDA-4 inoculation with spinach sequestered Cd in rhizospheric zone. Research outcomes are important for understanding morpho-physio-biochemical attributes of spinach-Bacillus sp. SDA-4 synergy which might provide efficient strategies to decrease Cd retention in edible plants and/or bioremediation of Cd polluted soil colloids.  相似文献

Seedlings of wheat (Triticum aestivum L.) cultivars Jing 411, Jinmai 30 and Yangmai 10 were exposed to 0, 10, 20, 30, 40 or 50 μM of CdCl2 in a solution culture experiment. The effects of cadmium (Cd) stress on wheat growth, leaf photon energy conversion, gas exchange, and Cd accumulation in wheat seedlings were investigated. Gas exchange was monitored at 3, 9, 24 days after treatment (DAT). Growth parameters, chlorophyll content, leaf chlorophyll fluorescence, and Cd concentration in shoot and root were measured at 24 DAT. Seedling growth, gas exchange, chlorophyll content, chlorophyll fluorescence parameters were generally depressed by Cd stress, especially under the high Cd concentrations. Cd concentration and accumulation in both shoots and roots increased with increasing external Cd concentrations. Relationships between corrected parameters of growth, photosynthesis and fluorescence and corrected Cd concentrations in shoots and roots could be explained by the regression model Y = K/(1 + exp(a + bX)). Jing 411 was found to be Cd tolerant considering parameters of chlorophyll content, photosynthesis and chlorophyll fluorescence in which less Cd translocation was from roots into shoots. The high Cd concentrations were in shoots and roots in Yangmai 10 which has been found to be a relative Cd tolerant cultivar in terms of most growth parameters.  相似文献

Cadmium (Cd) tolerance and accumulation in wheat varieties were investigated at seedling stage under a controlled environmental condition. The 46 leading wheat varieties cultivated from the 1950s to 2000s in China were treated at the three-leaf stage with a 50 μM CdCl2 solution for 24 days. Growth and photosynthesis parameters were measured and the Cd-tolerance index (ratio of a given parameter under Cd treatment to that of the control) was determined. Cd accumulation in shoots and roots and Cd translocation were also determined. It was found that Cd tolerance and accumulation of these wheat varieties varied over the different decades. Principal components analysis (PCA) showed that wheat varieties in 1950s and 1980s were tolerant while varieties from the last decade were sensitive to Cd stress. Wheat varieties in 1960s and 1970s were particularly tolerant to Cd stress for the parameters of shoot height, secondary root numbers, net photosynthesis and transpiration rate while the varieties in the 1990s were sensitive to Cd stress for shoot dry weight and root dry weight. Comparing each decade to the average Cd translocation ratio from the roots to the shoots for the whole period, the varieties from the 1950s and 1960s had a higher translocation ratio, while varieties in the 1970s were below that average. Varieties from the 1980s to 2000s showed an average translocation ratio. Using cluster analysis (CA), Shannongfu 63, Yangmai 1 and Yangmai 158 were the most Cd-tolerant varieties in which Cd translocation ratio were low, and Yumai 18 and Huaimai 20 were the most Cd-sensitive varieties in which Cd translocation ratio were high. The results indicating that wheat varieties from different decades were different in Cd tolerance and accumulation, and could be useful for breeding wheat for Cd stress tolerance.  相似文献

The effect of different external cadmium (Cd) and copper (Cu) regimes on the concentration of Cd and Cu in roots and shoots of birch (Betula pendula Roth.) seedlings was investigated. The seedlings were grown for 12 days in a weak nutrient solution (containing all essential nutrient elements including 0.025 µM Cu) at pH 4.2 with combinations of additional 0–2 µM CdCl2 and 0–2 µM CuCl2. Root and shoot concentrations of Cu were decreased by Cd in all treatments which included 0.1–2 µM of additional Cu in the treatment solution. When no extra Cu was added, only the shoot concentration of Cu was decreased by Cd whereas the root concentration was not affected. The shoot concentration of Cd was decreased by 0.5 and 2 µM of additional Cu in the treatment solution. The root concentration of Cd was decreased by Cu only when the concentration of additional Cu in the treatment solution was equal to or exceeded the concentration of Cd.  相似文献

Atriplex halimus subsp. schweinfurthii is a newly found cadmium (Cd)-hyperaccumulator, but there have been no detailed studies on its physiological responses when Cd is hyperaccumulated. A. halimus was grown in hydroponic conditions to investigate the effect of cadmium chloride (CdCl2) on growth, water status, leaf chlorophyll concentration, proline and Cd accumulation. Treatments were prepared by adding 0, 50, 100, 200 and 400 μM CdCl2 to the nutrient medium. Plant growth was significantly affected at high-Cd treatments. Increased CdCl2 decreased chlorophyll concentration, transpiration and root hydraulic conductivity (L0). Hence water flux had only a little effect on the uptake of Cd in A. halimus seedlings. In contrast, proline content increased with increasing CdCl2 concentration. Plants accumulated substantial amount of Cd in different plant parts (shoot and root). Most of the Cd taken up was retained in roots (606.51 μg g−1DW after 15 d at 400 μM CdCl2). The addition of Cd in the culture medium affected calcium (Ca) and potassium (K) nutrition in both shoot and root. A. halimus provides a new plant resource for exploring the mechanism of Cd hyperaccumulation and has potential for use in the phytostabilization of Cd-contaminated salt soils.  相似文献

Survival and cold hardiness declined gradually when cold-hardened Fredrick winter wheat (Triticum aestivum L.) was maintained at −6°C for several weeks. Moisture content of crown and root tissue did not change significantly during this period. Uptake of O2 and accumulation of 86Rb by root tissue declined abruptly upon exposure to −6°C, whereas a concomitant negative effect of freezing on these metabolic processes was not observed in crown tissue. Electron spin resonance spectroscopic analysis of microsomal membrane preparations from crown tissue revealed no evidence of gross changes in the physical properties of the bulk lipids even when seedlings were killed. The results provide biochemical evidence that seedling damage due to prolonged exposure to a mild freezing stress is due to disruption of key metabolic process in the root while cells within the crown remain viable.  相似文献

Rice (Oryza sativa L.) seedlings stressed with CdCl2 (0.5 mM or 50 μM) showed typical Cd toxicity (leaf chlorosis, decrease in chlorophyll content, or increase in H2O2 and malondialdehyde contents). Rice seedlings pretreated with heat shock at 45°C (HS) for 2 or 3 h were protected against subsequent Cd stress. Rice seedlings pretreated with HS had similar Cd concentration in leaves caused by CdCl2 as those non-HS. The content of H2O2 increased in leaves 1 h after HS exposure. However, APX and GR activities were higher in HS-treated leaves than their respective control, and it occurred after 2 h of HS treatment. Pretreatment of rice seedlings with H2O2 under non-HS conditions resulted in an increase in APX, GR, and CAT activities and protected rice seedlings from subsequent Cd stress. HS-induced H2O2 production and protection against subsequent Cd stress can be counteracted by imidazole, an inhibitor of NADPH oxidase complex. Results of the present study suggest that early accumulation of H2O2 during HS signals the increase in APX and GR activities, which in turn prevents rice seedlings from Cd-caused oxidative damage.  相似文献

In this study, the effects of cadmium chloride (CdCl2) on plant growth, histology of roots, photosynthetic pigments content, δ-aminolevulinic acid dehydratase (ALA-D; E.C. 4.2.1.24) and acid phosphatase activities (AP; E.C. 3.1.3.2), soluble phosphorus (Pi) measurement and mineral nutrients content in cucumber seedlings (Cucumis sativus L.) were investigated. Cucumber seedlings were grown in vitro in an agar-solidified substrate containing four CdCl2 treatments (0, 100, 400, and 1000 μM) for ten days. Cd was readily absorbed by seedlings and its content was greater in the roots than in the shoot. Cd reduced shoot and root length, and fresh and dry biomass of seedlings. Inhibition of root cell elongation in Cd-treated seedlings was observed by the increase of the mean radial size of cells belonging to three zones of the root tip. The highest level of Cd reduced in a similar manner chlorophyll a, chlorophyll b and total chlorophyll contents. Increasing concentrations of Cd resulted in a linear decrease in carotenoids levels of cotyledons. Interestingly, the ALA-D activity in cotyledons was inhibited only at the highest level of Cd. Root and shoot AP activities were, respectively, activated and inhibited at all CdCl2 concentrations. Root Pi concentration was increased in all Cd treatments and it was not altered in the shoot tissues. Moreover, in general, the nutrient contents were increased in the root and decreased in the shoot.
