Brachiaria, a genus of forage grasses of African origin, is gaining considerable importance because of both its nutritional value and its high stress resistance. An extraordinary resistance to Al toxicity has been reported in B. decumbens. The mechanisms of this hyperresistance are still unknown. This study explores the localization of Al in two contrasting Brachiaria species, the hyperresistant B. decumbens and the less resistant B. brizantha. Scanning Electron Microscope/Energy Dispersive Spectrometry, confocal fluorescence microscopy and optical microscopy of lumogallion or morin-stained roots was performed. Species differences in Al resistance were evident at 32 μM Al3+ activity in low ionic strength full nutrient solution containing Si. Roots of B. decumbens accumulated less Al than those of B. brizantha. Moreover, location and Al form seemed different. In B. decumbens Al accumulation was localized in hot spots of high Al concentrations. These sites with high Al accumulation mainly correspond to root hairs. B. brizantha exhibited a more even distribution of Al in cell walls of the root tip. Analysis of soluble phenolic substances in roots revealed species differences in response to Al. An Al-induced increase of chlorogenic acid concentrations was found in B. decumbens but not in B. brizantha. Taken together the results suggest a possible role for chlorogenic acid as a primer for changes in root epidermal cell patterning that may contribute to the Al hyperresistance in B. decumbens.  相似文献

The genus Urochloa P. Beauv. [syn. Brachiaria (Trin.) Griseb.] comprises species of great economic relevance as forages. The genomic constitution for the allotetraploid species Urochloa brizantha (cv. Marandu) and Urochloa decumbens (cv. Basilisk) and the diploid Urochloa ruziziensis was previously proposed as BBB1B1, B1B1B2B2 and B2B2, respectively. Evidence indicates U. ruziziensis as the ancestral donor of genome B2 in U. decumbens allotetraploidy, but the origin of the genomes B and B1 is still unknown. There are diploid genotypes of U. brizantha and U. decumbens that may be potential ancestors of the tetraploids. The aim of this study was to determine the genomic constitution and relationships between genotypes of U. brizantha (2x and 4x), U. decumbens (2x and 4x) and U. ruziziensis (2x) via genomic in situ hybridization (GISH). Additionally, chromosome number and genome size were verified for the diploid genotypes. The diploids U. brizantha and U. decumbens presented 2n?=?2x?=?18 chromosomes and DNA content of 1.79 and 1.44 pg, respectively. The GISH analysis revealed high homology between the diploids U. brizantha and U. decumbens, which suggests relatively short divergence time. The GISH using genomic probes from the diploid accessions on the tetraploid accessions’ chromosomes presented similar patterns, highlighting the genome B1 present in both of the tetraploids. Based on GISH results, the genomic constitution was proposed for the diploid genotypes of U. brizantha (B1B1) and U. decumbens (B1′B1′) and both were pointed as donors of genome B1 (or B1′), present in the allotetraploid genotypes.

Abstract

Histone post-translational modifications and DNA methylation are epigenetic processes that are involved in structural and functional organization of the genome. This study compared the chromosomal distribution of modified histones and 5-methylcytosine (5-mCyt) in species and interspecific hybrids of Brachiaria with different ploidy levels and reproduction modes. The relation between H3K9me2 and 5-mCyt was observed in the nucleolus organizer region, centromeric central domain and pericentromeric region. H3K4me2 was detected in euchromatic domains, mainly in the terminal chromosomal regions. Comparison of chromosomal distribution among species and hybrids showed greater variation of chromosomal types for the H3K9me2 in B. decumbens (tetraploid and apomictic species) and the 963 hybrid, while, for the H3K4me2, the variation was higher in B. brizantha and B. decumbens (tetraploid and apomictic species) and 963 hybrid. The chromosome distribution of 5-mCyt was similar between B. brizantha and B. decumbens, which differ from the distribution observed in B. ruziziensis (diploid and sexual species). Significant alterations in DNA methylation were observed in the artificially tetraploidized B. ruziziensis and in the interspecific hybrids, possibly as result of hybridization and polyploidization processes. The monitoring of histone modifications and DNA methylation allowed categorizing nuclear and chromosomal distribution of these epigenetic marks, thus contributing to the knowledge of composition and structure of the genome/epigenome of Brachiaria species and hybrids. These data can be useful for speciation and genome evolution studies in genus Brachiaria, and represent important markers to explore relationships between genomes.

Five emergent plant species were compared for their effectiveness in treating contaminants in a wetland system constructed on a military base in El Salvador. The system consisted of the subsurface flow (SSF), open water (OW) and free surface flow (SF) wetlands with a combined flow capacity of up to 151.4 m3 d?1. Reliability and consistent performance in extreme conditions, such as those occurring during the tropical dry or wet seasons were important evaluation criteria. The discontinuous flow patterns typical of tropical climates necessitated the use of water balance calculations using climatic data such as rainfall and evapotranspiration. System characterization was achieved by computation of daily input and output mass loading rates for each individual constituent. Results suggest that Phragmites and Brachiaria were the most effective plants in SSF wetland. Brachiaria provided the added benefit of serving as a source of fodder and proved proficient, with N and P uptakes of 1.5–3.14% and 0.17–0.25% per dry plants’ biomass, respectively. Typha yielded the highest dry season removal efficiency within the SF (BOD5: 80.78 ± 9.35%, COD: 65.18 ± 19.6%, TN: 58.59 ± 19.3%, oil and grease: 78.34 ± 10.55%, total dissolved phosphorus: 66.5 ± 20.7%). PhragmitesTypha treatment subset performed better year-round than either ThaliaThalia or BrachiariaCyperus. Evaluated plants were capable of surviving and proliferating in extreme tropical climates.  相似文献

Background Urochloa (syn. Brachiaria) is a genus of tropical grasses sown as forage feedstock, particularly in marginal soils. Here we aimed to clarify the genetic diversity and population structure in Urochloa species to understand better how population evolution relates to ploidy level and occurrence of apomictic reproduction.MethodsWe explored the genetic diversity of 111 accessions from the five Urochloa species used to develop commercial cultivars. These accessions were conserved from wild materials collected at their centre of origin in Africa, and they tentatively represent the complete Urochloa gene pool used in breeding programmes. We used RNA-sequencing to generate 1.1 million single nucleotide polymorphism loci. We employed genetic admixture, principal component and phylogenetic analyses to define subpopulations.ResultsWe observed three highly differentiated subpopulations in U. brizantha, which were unrelated to ploidy: one intermixed with U. decumbens, and two diverged from the former and the other species in the complex. We also observed two subpopulations in U. humidicola, unrelated to ploidy; one subpopulation had fewer accessions but included the only characterized sexual accession in the species. Our results also supported a division of U. decumbens between diploids and polyploids, and no subpopulations within U. ruziziensis and U. maxima.ConclusionsPolyploid U. decumbens are more closely related to polyploid U. brizantha than to diploid U. decumbens, which supports the divergence of both polyploid groups from a common tetraploid ancestor and provides evidence for the hybridization barrier of ploidy. The three differentiated subpopulations of apomictic polyploid U. brizantha accessions constitute diverged ecotypes, which can probably be utilized in hybrid breeding. Subpopulations were not observed in non-apomictic U. ruziziensis. Sexual Urochloa polyploids were not found (U. brizantha, U. decumbens) or were limited to small subpopulations (U. humidicola). The subpopulation structure observed in the Urochloa sexual–apomictic multiploidy complexes supports geographical parthenogenesis, where the polyploid genotypes exploit the evolutionary advantage of apomixis, i.e. uniparental reproduction and clonality, to occupy extensive geographical areas.  相似文献

The purpose of this study was to investigate the aluminum (Al) concentration in Lycopodium clavatum, Dicranopteris flexuosa, Sticherus nudus, Anemia villosa, Cyathea gibbosa, Pteridium arachnoideum, Pteris vittata, Thelypteris dentata, Blechnum occidentale, Elaphoglossum sporadolepis, Nephrolepis cordifolia and Polypodium pseudoaureum, species from 11 families with different phylogenetic position, found on soils with a high concentration of Al (up to 13 g kg?1 dry mass (DM)). When Al concentration and mineral nutrients in aerial organs were considered, pteridophytes were classified into three groups: group one included pteridophytes with Al concentrations over 1000 mg kg?1 DM in their aerial organs, a ratio between Al and essential plant nutrients such as Ca, Mg and P higher than one and a K/Al ratio between 0.68 and 2.56 mol mol?1. In group 1 was the well known Al-accumulator L. clavatum (Lycophyte) as well as the Neotropical ferns D. flexuosa, S. nudus (both basal leptosporangiate ferns), and C. gibbosa (core leptosporangiate tree fern). Group 2, ferns which accumulate Al over 1000 mg kg?1 DM in their fronds, and had an Al/Ca and Al/Mg ratio <1. Species in this group included E. sporadolepis and N. cordifolia (derived polypod ferns). Group 3, ferns classified as Al-excluders, showing Al concentration <782 mg kg?1 DM in the fronds, had Al/Ca and Al/Mg ratios <1, Al/P ratio ≤1 and a K/Al ratio between 18.10 and 80.36 mol mol?1. In group 3, were A. villosa (basal leptosporangiate fern) and the derived polypod ferns P. arachnoideum, P. vittata, T. dentata, B. occidentale and P. pseudoaureum. The translocation factor of Al from subterranean to aerial organs was up to 4 in S. nudus, and subterranean organs from E. sporadolepis showed the highest concentration of Al (12 g kg?1 DM). We coincide with early literature in that other criteria in addition to the Al concentration should be considered to define the Al accumulation, such as its relationship with macronutrients. For example, we propose the inclusion of K/Al ratio. We conclude that out of six Al-excluders five belonged to the derived polypods while two species from Polypodiales showed high Al concentrations. We reconfirm accumulation of Al in L. clavatum and C. gibbosa and discover two new Al-accumulating species in the more ancient ferns: S. nudus and D. flexuosa.  相似文献

Aluminum (Al) is the most inhibiting factor for plant root elongation in acidic soils. Boron (B) is an essential micronutrient whose deficiency occurs in acid soils because of high leaching. Lisianthus (Eustoma grandiflorum L.) is a plant to which Al seems to be beneficial, but the range of B requirement for its growth is not documented yet. Both Al toxicity and B deficiency result in oxidative stress in certain plants. The present study was aimed to evaluate the effects of B and Al on the activity of antioxidant system of rooted cuttings of lisianthus. The plants were grown in nutrient solution and treated with 0, 0.05 and 0.1 mM of boron and 0.88 mM of aluminum for 24 h. Activity of certain antioxidant enzymes and the contents of non-enzymatic antioxidants were evaluated. The expression of CAT gene was quantified by semi-quantitative RT-PCR technique. Increase activities of superoxide dismutase (SOD), catalase (CAT), and ascorbate peroxidase (APX) and increase of flavonoids and anthocyanin contents was observed in the plants which treated with 0.1 mM B and 0.88 mM Al, compared to those treated with 0 and 0.05 mM B. Increase of CAT activity was the most pronounced among antioxidant enzymes and was parallel with increased expression of its gene. The results showed that Al and B (in higher concentration) provide lisianthus plant with reinforced antioxidant system.  相似文献

Twelve forage grass accessions including 11 accessions of Brachiaria Griseb, were evaluated in a glasshouse for host plant resistance to nymphs and tolerance to feeding damage caused by adults of Zulia colombiana (Lallemand) (Homoptera: Cercopidae). Resistance to nymphs was evaluated with a technique that provided uniform environmental conditions and abundant feeding sites. B. brizantha Stapf (cv. Marandú) was the most resistant of the accessions tested based on nymphal mortality, duration of nymphal stadia, and weight of adult females. Andropogon gayanus Kunth, resistant to spittlebug attack in the field, was susceptible under the conditions of this study. While growth habit and rooting characteristics may contribute to field resistance, other resistance factors are present within the genus Brachiaria, particularly in the case of B. brizantha cv. Marandú. The number of insect-days causing severe damage in the most tolerant species (B. dictyoneura Stapf and B. humidicola Schweick) was approximately six times greater than that necessary to cause the same level of damage to the most susceptible species (B. ruziziensis Germain & Evrard and B. decumbens Stapf). No difference was found in regrowth capacity between infested and noninfested plants within accessions. There was a significant positive correlation between number of insect-days causing severe damage (tolerance) and regrowth of infested plants.
Résumé Brachiaria est une graminée fourragère prometteuse pour les sols tropicaux acides, saturés d'aluminium. Z. colombiana est un Cercopidae très répandu, limitant l'utilisation de Brachiaria en Amérique Latine. La résistance (antibiose et tolérance) à Z. colombiana, de Brachiaria d'origines diverses a été examinée. B. brizantha cv. Marandù s'est révélé le plus résistant d'après la forte mortalité larvaire, la prolongation du développement larvaire, et le poids réduit des femelles adultes de Z. colombiana. Andropogon gayanus, résistant dans la nature, s'est révélé sensible. Ces résultats suggèrent que cette résistance de A. gayanus dans la nature pourrait être due à la structure du végétal et à son mode de croissance. Dans le cas de B. brizantha cv. Marandù, des facteurs supplémentaires de résistance, mis en évidence à partir de différents modes de croissance, ont été éliminés, de façon à identifier les mécanismes de l'antibiose présents chez Brachiaria. Une grande gamme de résistance aux attaques alimentaires a été observée chez Brachiaria. Les plus résistants ont besoin de 6 fois plus de jours d'attaque par Z. colombiana pour provoquer les dégâts observés sur individus sensibles.

A 100 d experiment was conducted to determine the effects of aluminum (Al) source and concentration on mineral status, emphasizing phosphorus (P), of 50 feeder lambs. Six treatments, fed at 10% of the total diet, were formulated using two sources of Al, AlCl3 and an Al-based water treatment residual (WTR, 11.1% Al), with varying levels of Al and P: (1) control (10% sand, C), (2) low WTR (2.5% WTR and 7.5% sand, L-WTR), (3) AlCl3 with added P (1% AlCl3, 9% sand, and 0.4% P, AlCl3 + P), (4) high WTR (10% WTR, H-WTR), (5) AlCl3 (1% AlCl3 and 9% sand, AlCl3), and (6) high WTR with added P (10% WTR and 0.4% P, H-WTR + P). The total Al varied from 0.037 to 1.2% among diets. Only lambs fed the high WTR diet without P supplementation (H-WTR) decreased feed intakes. These lambs consumed about half as much feed as lambs on all the other treatments, and had lower (P < 0.05) BW from d 84 on. Lambs receiving the H-WTR had the lowest bone Ca, P and Mg concentrations (fresh basis, mg/cm3) and lowest bone mineral content (BMC) as determined by radiographs (mm of Al). Results for the lambs on H-WTR were confounded by the greatly reduced feed intake of animals on this treatment. Plasma P decreased in all lambs consuming Al, regardless of Al source, but the effects were less severe in animals provided additional P supplementation (AlCl3 + P and H-WTR + P). Apparent absorption of P was affected by concentration and source of Al in two metabolism trials (n = 42) beginning on d 34 and d 70, respectively. In the first trial, d 34, lambs receiving AlCl3 treatment had reduced apparent P absorption, −17.7% (P < 0.05), when compared to all other treatments. In the d 70 trial, lambs receiving both AlCl3 and H-WTR treatments were negatively impacted (P < 0.05) compared to the control, −20.9 and −2.5% apparent P absorption, respectively, but were no longer different from one another (P > 0.05). Diets containing 1.2% Al as WTR without P supplementation depressed feed intakes, weight gains, plasma P concentrations (P < 0.05), and BMC. However, given adequate P supplementation, even lambs consuming this amount of Al did not suffer detrimental effects, as lambs on H-WTR + P did not differ from the control (P > 0.05) in feed intakes, weight gains, or BMC.  相似文献

The effects of metformin and pioglitazone on ghrelin, a physiologic regulator of appetite and food intake, have not been clearly established. In a randomized clinical trial, we randomly assigned 60 type 2 diabetic patients to either metformin (Group A; n = 30) or pioglitazone (Group B; n = 30) treatment groups. The groups were similar in their baseline characteristics. A standard fasting 75 g oral glucose tolerance test was performed at time zero before starting metformin or pioglitazone, and 3 months later. After 3 months of treatment, pioglitazone, but not metformin, was significantly associated with weight gain. Both groups experienced a significant reduction in fasting plasma glucose (p < 0.01), hemoglobin A1c (p < 0.01 in Group A and p < 0.05 in Group B), and insulin resistance (p < 0.01). The effect of metformin on preprandial ghrelin and its response to glucose challenge was not significant, while the pioglitazone group had a significant reduction in preprandial ghrelin levels after treatment (p < 0.05). The effect of pioglitazone on ghrelin was independent of changes in body weight, body mass index, glucose control, insulin resistance, and plasma insulin. In conclusion, treatment with pioglitazone is associated with a decrease in preprandial ghrelin levels and therefore, the weight gain and increased food intake related to pioglitazone use cannot be explained by its effects on ghrelin. The effect of pioglitazone on ghrelin is independent of changes in body weight, body mass index, plasma insulin, insulin resistance, or glucose control.  相似文献

Nerolidol is a sesquiterpene present in the essential oils of many plants, approved by the U.S. FDA as a food flavoring agent. Nerolidol interferes with the isoprenoid biosynthetic pathway in the apicoplast of P. falciparum. In the present study, the in vitro growth of four Babesia species was significantly (P < 0.05) inhibited in the presence of nerolidol (IC50s values = 21 ± 1, 29.6 ± 3, 26.9 ± 2, and 23.1 ± 1 µM for B. bovis, B. bigemina, B. ovata, and B. caballi, respectively). Parasites from treated cultures failed to grow in the subsequent viability test at a concentration of 50 µM. Nerolidol significantly (P < 0.05) inhibited the growth of B. microti at the dosage of 10 and 100 mg/kg BW, while the inhibition was low compared with the high doses used. Therefore, nerolidol could not be used as a chemotherapeutic drug for babesiosis.  相似文献

In order to detect the response of trees to environmental pollution in Shenyang during the last century, tree rings of two 100+ year old Chinese pine (Pinus tabulaeformis Carr.) were analyzed for nine trace element concentrations (K, Ca, Mg, Mn, Cu, Zn, Al, Pb and Cd) and compared to local environmental change. The concentrations of Cu, Zn and Al had significant (p < 0.01) increasing trends after 1940s and correlated significantly (p < 0.01), with increasing production of nonferrous metal from local industrial processing. Concentrations of nutrient element (K, Ca and Mg) showed significant (p < 0.01) increasing trend from heartwood to sapwood with declines in the last three decades. Significant correlation between climatic factors and element concentration in tree rings were also found, in which the temperature (November to May) and relative humidity in April and May showed significant positive correlation with Ca, Zn and Cu (p < 0.05) as well as Al (p < 0.01) concentrations in tree rings. April rainfall showed significant positive correlation with K and Zn (p < 0.05) as well as Al (p < 0.01) concentration in tree rings. The results suggest that tree rings from Chinese pine in Shenyang are sensitive to environmental change and have the potential to be used as a biomonitor for the environmental pollution. The results also demonstrate the feasibility of applying dendrochemical techniques to areas with limited samples.  相似文献

The purpose of this study was to investigate a possible relation between resistance to bile salts and low pH with exopolysaccharide (EPS) producing of Bifidobacterium spp. In this study, a total of 31 Bifidobacterium spp. were isolated from breast fed infants feces and breast milk samples. As a result of the identification tests, isolates were identified as Bifidobacterium breve (15 strains), B. bifidum (11 strains), B. pseudocatenulatum (3 strains) and B. longum (2 strains). Bifidobacterium spp. were determined exopolysaccharide (EPS) production. EPS productions observed at chance rations (38.00–97.64 mg/l) among of Bifidobacterium spp. Furthermore, Bifidobacterium spp. were determined resistance to bile salts and low pH. Positive correlations between production of exopolysaccharide and resistance to bile salts (p < 0.01) or low pH (p < 0.01) were found Bifidobacterium spp. This investigation showed that high EPS production of Bifidobacteria may be important in the selection of probiotic strains for resistance to bile salts and low pH.  相似文献

We are reporting the stability constants of the different complexes between phosphonoacetic acid (PAA), phosphonoformic acid (PFA), aminomethylphosphonic acid (AMPA), aminoethylphosphonic acid (AEPA) and methylenediphosphonic acid (MDP) with the aluminum metal ion in aqueous solutions. (In this study the term aluminum is reserved for the 3+ ion.) The affinity of the phosphonic acid derivatives to chelate aluminum has been tested by potentiometric titrations with I = 0.10 M KNO3 at 25 ± 0.1 °C. The proposed aluminum–ligand complex structures have been confirmed by 31P NMR, 13C NMR, and 27Al NMR experiments. Both PAA and PFA formed simple one to one complexes. The respective values for PAA are log β111 = 13.57, log β110 = 10.58, and log β11−1 = 5.84. The respective values for PFA are log β112 = 15.24, log β111 = 13.11, and log β110 = 6.88. In contrast to PAA and PFA, the major species formed with AMPA and AEPA consist of a series of dimeric complexes. The 31P NMR spectra of these complexes indicate that the amine groups do not co-ordinate to aluminum and remain protonated. In addition to these dimeric complexes, a monoprotonated monomer of Al–AMPA also has been detected. The 27Al NMR experiments indicated that the aluminum is hexacoordinated in all the complexes in this study and the hydroxide ion did not remove aluminium from its complexes. Among the ligands studied, PAA and PFA were able to solubilize aluminum at physiological pH. A comparison between the acidities and the chelating properties of the ligands used is presented.  相似文献

BackgroundBacillus anthracis is the causative agent of anthrax, a disease associated with a very high mortality rate in its invasive forms.MethodsWe studied a number of ebselen analogs as inhibitors of B. anthracis thioredoxin reductase and their antibacterial activity on Bacillus subtilis, Staphylococcus aureus, Bacillus cereus and Mycobacterium tuberculosis.ResultsThe most potent compounds in the series gave IC50 values down to 70 nM for the pure enzyme and minimal inhibitory concentrations (MICs) down to 0.4 μM (0.12 μg/ml) for B. subtilis, 1.5 μM (0.64 μg/ml) for S. aureus, 2 μM (0.86 μg/ml) for B. cereus and 10 μg/ml for M. tuberculosis. Minimal bactericidal concentrations (MBCs) were found at 1–1.5 times the MIC, indicating a general, class-dependent, bactericidal mode of action. The combined bacteriological and enzymological data were used to construct a preliminary structure–activity-relationship for the benzoisoselenazol class of compounds. When S. aureus and B. subtilis were exposed to ebselen, we were unable to isolate resistant mutants on both solid and in liquid medium suggesting a high resistance barrier.ConclusionsThese results suggest that ebselen and analogs thereof could be developed into a novel antibiotic class, useful for the treatment of infections caused by B. anthracis, S. aureus, M. tuberculosis and other clinically important bacteria. Furthermore, the high barrier against resistance development is encouraging for further drug development.General significanceWe have characterized the thioredoxin system from B. anthracis as a novel drug target and ebselen and analogs thereof as a potential new class of antibiotics targeting several important human pathogens.  相似文献

Duckweeds, as a group, are important early warning indicators for the assessment of contaminated ecosystems due to their propensity to accumulate pollutants. In the present study, we investigated the potential use of Lemna gibba L. (Lemnaceae) as an ecological indicator for boron (B) mine effluent containing B concentration above 10 mg l−1. For this purpose, L. gibba fronds were grown for 7 days in simulated water contaminated with B mine effluent. The important note is that this study was carried out in Kırka (Eskişehir, Turkey) B reserve area, which is the largest borax reserve in all over the world, under natural climatic conditions in the field. The results demonstrated that accumulations of B by L. gibba gradually increased based on the initial B concentrations (10, 25, 50, 100, and 150 mg l−1) of the mine effluent. B concentration in the dry weight of the plant reached 639 mg kg−1 when the minimum initial dosage (10 mg l−1) was applied and 2711 mg kg−1 when the maximum initial dosage (150 mg l−1) was applied during the study. However, significant reductions in their relative growth rates occurred in 50, 100 and 150 mg l−1 initial B concentrations. Results suggest that 25 mg l−1 B concentration in water seemed to be a sensitive endpoint for L. gibba that could be used as a critical bioindicator level of B contaminated water. Following our data, we also constructed a simple growth model under the climatic conditions in this region of Turkey, but in instructive as a worldwide model. Variations in the soil carbon sequestration capability of different types of salt marsh soils at Chongming Dongtan and its influencing factors were studied by analyzing the soil organic carbon (SOC) content, organic matter input and microbial activities. The results indicated that the total SOC content at Area A (southeast of Dongtan, sandy soil with Phragmites communis) was only 46.11% of that of Area B (northeast of Dongtan, clay soil with mixed P. communis and Spartina alterniflora) (P = 0.000 < 0.05), but their organic matter input per year was almost identical. These findings implied that Area B had a lower output of SOC. The microbial biomass at Area A was 3.83 times greater than that at Area B (P = 0.049 < 0.05); the soil catalase and invertase activities at Area A, which were related to carbon metabolism, were 60.31% (P = 0.006 < 0.05) and 34.33% (P = 0.021 < 0.05) higher than at Area B, respectively; and the soil respiration at Area A was also higher than at Area B. These findings implied that the microbial activities at Area A were greater than those at Area B, and therefore the carbon metabolism was rapid, resulting in increased SOC output at Area A. Increased water content and salinity in the clay soil at Area B may inhibit the microbial activities, thereby reducing the decomposition of the organic matter and enhancing carbon sequestration. In addition, some artificial measures for controlling spread of S. alterniflora at Area B (mowing/digging and tillage (M + D); mowing/digging and tillage/waterlogging (M + D + W)) were found to generally improve the microbial activity of soil, thereby increasing SOC output. However, when the two different physical controlling modes were compared, the SOC and microbial activities of the soil subjected to the M + D + W treatment were relatively high and low, respectively, due to waterlogging restraining the microbial metabolism. These findings indicated that the difference in microbial activities was the important factor leading to variability in the SOC sequestration capability between Areas A and B. Additionally, with the exception of soil texture and vegetation types, environmental conditions and artificial turbulence also influenced microbial activities of soil, and hence SOC output and organic carbon sequestration capability.  相似文献

In the present study, we identified, cloned and expressed a 40-kDa heat shock protein, DnaJ, from Bacillus halodurans. The open reading frame of the cloned gene contained 1116 bp and encoded 371 amino acid residues. The purified recombinant DnaJ contained a His-tag at the C-terminus and showed a single band at approximately 41-kDa on SDS-PAGE gel. The 3D structures of DnaJ obtained by I-TASSER showed that the overall structures of DnaJ from B. halodurans Guj1 and E. coli are very similar, with 45% sequence similarity. The present study revealed that the DnaJ protein from B. halodurans inhibits the heat-induced aggregation of insulin in a concentration-dependent manner as aggregation of the insulin B-chain was reduced by approximately 50% at 40 °C in the presence of 0.1 mg/ml of purified recombinant DnaJ. The overexpression of DnaJ improved thermotolerance properties in E. coli transformed with pET-28a + DnaJ. Salt resistance experiments indicated that the survival of E. coli transformed with DnaJ was enhanced 1.85-fold compared to that of the control cells in the presence of 0.5 M NaCl for 72 h. According to the results obtained, DnaJ from B. halodurans can potentially be used for improving the functional properties of enzymes and proteins in various applications.  相似文献

Two triterpenoid saponins have been isolated from the seed kernels of Entada rheedii. Their structures have been established using 1D- and 2D-NMR and mass spectrometry as 3-O-β-d-xylopyranosyl-(1  3)-O-α-l-arabinopyranosyl-(1  6)-2-acetylamino-2-deoxy-β-d-glucopyranosylentagenic acid 28-O-β-apiofuranosyl-(1  3)-β-d-xylopyranosyl-(1  2)-β-d-glucopyranoside (Rheediinoside A, 1) and 3-O-β-d-glucopyranosyl-(1  3)-O-[β-d-xylopyranosyl-(1  3)-α-l-arabinopyranosyl-(1  6)]-2-acetylamino-2-deoxy-β-d-glucopyranosylentagenic acid 28-O-β-apiofuranosyl-(1  3)-β-d-xylopyranosyl-(1  2)-β-d-glucopyranoside (Rheediinoside B, 2). Compounds 1 and 2 were tested for their antiproliferative activity against T98G, A431, PC3 and B16-F1 cell lines, and further for their antioxidant properties. Moderate cytotoxic potency and antioxidant properties were found for these compounds whereas Rheediinoside B was in all assays more active than Rheediinoside A.  相似文献

Mutations in the second EF-hand (D61N, D63N, D65N, and E72A) of S100B were used to study its Ca2 + binding and dynamic properties in the absence and presence of a bound target, TRTK-12. With D63NS100B as an exception (D63NKD = 50 ± 9 μM), Ca2 + binding to EF2-hand mutants were reduced by more than 8-fold in the absence of TRTK-12 (D61NKD = 412 ± 67 μM, D65NKD = 968 ± 171 μM, and E72AKD = 471 ± 133 μM), when compared to wild-type protein (WTKD = 56 ± 9 μM). For the TRTK-12 complexes, the Ca2 +-binding affinity to wild type (WT + TRTKKD = 12 ± 10 μM) and the EF2 mutants was increased by 5- to 14-fold versus in the absence of target (D61N + TRTKKD = 29 ± 1.2 μM, D63N + TRTKKD = 10 ± 2.2 μM, D65N + TRTKKD = 73 ± 4.4 μM, and E72A + TRTKKD = 18 ± 3.7 μM). In addition, Rex, as measured using relaxation dispersion for side‐chain 15N resonances of Asn63 (D63NS100B), was reduced upon TRTK-12 binding when measured by NMR. Likewise, backbone motions on multiple timescales (picoseconds to milliseconds) throughout wild type, D61NS100B, D63NS100B, and D65NS100B were lowered upon binding TRTK-12. However, the X-ray structures of Ca2 +-bound (2.0 Å) and TRTK-bound (1.2 Å) D63NS100B showed no change in Ca2 + coordination; thus, these and analogous structural data for the wild-type protein could not be used to explain how target binding increased Ca2 +-binding affinity in solution. Therefore, a model for how S100B–TRTK‐12 complex formation increases Ca2 + binding is discussed, which considers changes in protein dynamics upon binding the target TRTK-12.  相似文献
