We have developed a simple and rapid technique for measuring the action spectra for phototaxis of populations of microorganisms and applied it to halobacteria. A microscope with a dark-field condenser was used to illuminate the cell suspension in a sealed chamber with light of wavelength greater than 750 nm; in this region of the spectrum, the halobacteria show no phototactic response. A 150-micron spot of light from a xenon arc lamp, whose wavelength and intensity can be varied, was projected through the objective lens into the center of the dark field. The objective lens imaged this measuring spot through a 780-nm cut-off filter on an aperture in front of a photomultiplier. The intensity of the scattered 750-nm light, and therefore the photomultiplier current, is proportional to the number of cells in the measuring spot. A third lamp provided background light of variable wavelength and intensity through the dark-field condenser. To minimize secondary effects due to large changes in cell density, we recorded the initial changes in the photomultiplier current over 1 min after the actinic light had been switched on. By plotting the rate of change against wavelength, we obtained action spectra after the proper corrections for changes in light intensity with wavelength were applied and saturation effects were avoided.  相似文献

The changes in skin morphology over time are a consequence of both chronologic aging and the accumulation of environmental exposure. Through observation, we know that actinic radiation intensifies the apparent aging of skin. We have investigated the effects of aging and actinic radiation on the ability of fibroblasts to contract collagen-fibroblast lattices. Preauricular and postauricular skin samples were obtained from eight patients aged 49 to 74 undergoing rhytidectomy. The samples were kept separate, and the fibroblasts were grown in culture. Lattices constructed with preauricular fibroblasts consistently contracted more than lattices containing postauricular fibroblasts. The difference in amount of contraction in 7 days between sites was greatest for the younger patients and decreased linearly as donor age increased (r = -0.96). This difference may be due to preauricular fibroblasts losing their ability to contract a lattice as aging skin is exposed to more actinic radiation.  相似文献

It has been shown on a photomultiplier (PM) that glycine solution excited with quartz lamp of low intensity acquires the ability to produce ultra-weak ultraviolet radiation--mitogenetic radiation. The appearance of radiation is connected with the process of oxidative deamination of glycine molecules. These data confirm the formerly established facts of glycine mitogenetic radiation determined by biological detection.  相似文献

The earlier developed double-modulation chlorphyll (Chl) fluorometer was modified for measurements with intact leaves of higher plants. The Chl fluorometer is based on a non-periodic modulation of both actinic and measuring flashes. In addition, continuous orange actinic and far-red radiation were produced by separate arrays of light-emitting diodes (LEDs). Programmable timing of the flashes allows to cover a wide dynamic range from microseconds to minutes. We have demonstrated that the LEDs can produce single-turnover flashes that saturate QA reduction of intact leaves of Glyceria maxima and shoots of Picea abies. A periodic shading mechanism (about 0.5 s period) is believed to be used by flagellated algae to detect light direction. Time changes in the orientation of a population ofHaematococcus pluvialis under positive photoaxis conditions (cell samples were stimulated alternatively by two diametrically opposed beams of actinic light), have been investigated by analysing the Doppler shifts of laser light scattered by the cells by means of heterodyne detection techniques. This technique allows us to measure the mean value of the component of the cells' swimming velocities along the light stimulus axis. Preliminary data indicate that the time taken by a cell population to change orientation is about 1 s.  相似文献

应用建立的人工同龄林辐射场模型对杉木 (Cunninghamialanceolata)人工林冠层辐射场进行了模拟 ,以实测的冠层辐射数据对模型进行了验证 ,对模型的不确定性以及叶面积密度和叶片散射的影响进行了计算和分析。结果表明 ,模拟和实测辐射场及日总量无显著差异 ,所建模型适于对杉木人工林冠层辐射的模拟。模型可能的误差来源包括树冠形状的不规则性、树冠大小和高低的不一致性、叶片在小范围内的簇生性等。考虑二维叶面积密度 (LAD)分布的模拟结果优于平均LAD。叶片的散射辐射约占总辐射的 3%~ 11%,且越靠近树冠里层和下部 ,散射作用越大。  相似文献

The effect of molecular oxygen on the photochemical activity of the Rhodobacter sphaeroides reaction centers frozen to 160 K under actinic illumination was investigated by the ESR method. About 90% of initially photochemically active bacteriochlorophyll (P) were fixed at 160 K for a long time in aerobic samples in an inactive form. In anaerobic samples, not more than 65% were fixed in an inactive form under the same conditions. In aerobic preparations, a small portion of photochemically active bacteriochlorophyll (about 10%) that retains its photochemical activity at 160 K after freezing under illumination has dark reduction kinetics similar to that of samples at room temperature after several seconds of actinic illumination. In anaerobic samples frozen under illumination, the remaining photochemically active reaction centers (35%) have the same dark reduction kinetics as samples illuminated at 295 K for 1-2 min. The conclusion is that the irreversible stabilization of bacteriochlorophyll P in the oxidized inactive state formed in the reaction centers frozen under illumination is brought about by light-induced conformational changes fixed under low temperatures.  相似文献

The kinetics of dark reduction of chlorophyll P700 oxidized by steady-state illumination in photosystem I reaction center preparations of cyanobacterium Synechocystis sp. coolled in the dark to 160 K is greatly nonexponential. The characteristic times for the components of the reaction are from fractions of a second to minutes and more. During cooling reaction center preparations on actinic light, a great part of chlorophyll P700 is fixed at 160 K in oxidized state. The kinetics of dark reduction of P700+ in the fraction of reaction centers that retain the photochemical activity in these conditions is faster than the kinetics in samples cooled in the dark. A theoretical analysis of the substantial deceleration of the P700+ dark recovery kinetics was done for photosystem I reaction center preparations oxidized by steady-state illumination to 160 K in contrast with situation that arises after the oxidation of reaction centers by single short light pulses. The deceleration of the kinetics in samples activated by steady-state illumination can be explained by processes of microconformational relaxation, connected with proton shifts in the reaction center structure.  相似文献

The kinetics of dark reduction of chlorophyll P700 oxidized by continuous light in preparations of photosystem I reaction centers from cyanobacterium Synechosystis spharoides cooled in the dark to 160 K is essentially nonexponential. The characteristic times of the components range from fractions of a second to minutes or more. During the cooling of reaction center preparations under illumination with actinic light, most of the chlorophyll P700 molecules are fixed in the oxidized state at 160 K. The kinetics of dark reduction of P700+ in the fraction of reaction centers that retain photochemical activity under these conditions is somewhat faster compared to the samples cooled in the dark. A theoretical analysis of substantial deceleration of P700+ dark recovery kinetics was done for preparations of photosystem I reaction centers oxidized by continuous light at 160 K in comparison to the experiments where reaction centers were oxidized by short single light flashes. This slowing down of the kinetics in samples excited by continuous illumination can be explained by microconformational relaxation processes related to proton shifts in the reaction center.  相似文献

A PAM fluorometer was used for measuring induction curves in the range of actinic radiation of 3-35 W m-2, with an ordinary value of about 15 W m-2. The effects of various mediators, i.e., ascorbate, methyl viologen (MV), dithiothreitol (DTT) and nigericin, on the quenching process were tested. Simultaneously, the absorbance was measured during a 15-20 min period of irradiation and after the actinic radiation was turned off, i.e., in the recovery period. The pH values of chloroplast suspensions were 5.5, 6.5 and 8.0, the largest non-photochemical quenching was observed at pH of 6.5. The irradiation of chloroplasts led to an absorption decrease within the entire photosynthetically active range, attaining saturation when the fluorescence reached Fs level, and to an absorption increase during the recovery period. Absorbance changes at the maximum of red band were 10-20 %. A decrease in Chl concentration (10 %) after irradiation was found only at pH of 5.5, when the recovery time was the longest, i.e., about 60 min.  相似文献

An apparatus is described by means of which the power versus frequency spectrum of the photomultiplier current can be obtained for laser light scattered by streaming cytoplasm in the algal cell Chara corallina. A Doppler peak is noted in the spectrum which is abolished when cytoplasmic streaming is arrested by electrical stimulation. For 5 cells of Chara, this simple laser-Doppler velocimeter gave streaming velocities (46-7 mum s-1, S.D. +/- 4-8 at 20 degrees C) similar to those obtained for the same cells using the light microscope (44-3 mum s-1, S.D. +/- 5-3 at 20 degrees C). A narrow distribution of streaming velocities is indicated. The technique described provides a rapid, quantitative assay of the in vivo rheological properties of cytoplasm.  相似文献

Integrating spheres are widely used with UV-Vis and occasionally with infrared spectrophotometers to measure different types of samples, either in transmission mode (scattered transmission accessories) or in total/diffuse reflectance mode. We built a prototype sphere of the demountable type, which fits easily the sample compartment of a commercial CD spectropolarimeter, requiring neither any alignment nor the use of a dedicated photomultiplier. Samples can be inserted either at the sphere entrance (for scattered transmission mode) or in the center of the sphere (for total reflectance experiments). Crude cholera exotoxin (filtrate toxin) was irradiated with increasing doses of gamma radiation. A significant drop in enterotoxicity, in the activity of the permeation factor and a decrease in toxicity were shown to occur as radiation doses increased. Radiation doses of 50-70 kGy were found to completely inactivate enterotoxicity in liquid toxic preparations. A higher radioresistance of dried preparations in comparison with liquid ones was registered: inactivation occurred at 150-200 kGy. Different batches of the initial filtrate toxin had varying radiosensitivity. The sterilizing effect of gamma radiation was achieved at doses of 20 kGy for liquid preparations and 30 kGy for dried preparations. During the prolonged storage of the irradiated preparations of crude toxin (the term of observation being 1.5 years) at different temperatures no reversion of toxicity was found to occur, while their immunogenic properties remained unchanged.  相似文献

Dried preparations with Streptococcus faecium, strain A(2)1, and spores of Bacillus sphaericus, strain C(I)A, normally used for control of the microbiological efficiency of radiation sterilization plants and preparations with spores of Bacillus subtilis, normally used for control of sterilization by dry heat, formalin, and ethylene oxide, as well as similar preparations with Micrococcus radiodurans, strain R(1), and spores of Bacillus globigii (B. subtilis, var. niger) were gamma irradiated with dose rates from 16 to 70 krad/h at temperatures from 60 to 100 C. At 80 C the radiation response of the spore preparations was the same as at room temperature, whereas the radiation resistance of the preparations with the two vegetative strains was reduced. At 100 C the radiation response of preparations with spores of B. subtilis was unaffected by the high temperature, whereas at 16 and and 25 krad/h the radiation resistance of the radiation-resistant sporeformer B. sphaericus, strain C(I)A, was reduced to the level of radiation resistance of preparations with spores of B. subtilis. It is concluded that combinations of heat and gamma irradiation at the temperatures and dose rates tested may have very few practical applications in sterilization of medical equipment.  相似文献

为探究森林冠层结构与林下光照的变化规律及其相关性,在河南省白云山国家森林公园选取人工林、择伐林、皆伐林和老龄林建立四块面积为1 hm2(100 m×100 m)的固定监测样地,利用半球面影像技术获取冠层结构及林下光照数据。研究发现:1)随着人为干扰强度降低,冠层覆盖度与叶面积指数呈增加趋势,林下散射辐射与直接辐射呈减少趋势;2)择伐林冠层覆盖度与叶面积指数最大,平均叶倾角与透光比最小,林下光照(直接辐射、散射辐射)与冠层覆盖度和叶面积指数都呈显著负相关,林下散射辐射与冠层覆盖度和叶面积指数的负相关关系最强;3)处于不同干扰强度的群落由于冠层结构的差异,形成了不同群落内特定的光照环境。研究结果丰富了暖温带-亚热带生态过渡区森林冠层结构与林下光照动态变化研究资料,同时也为该区域森林的恢复与重建等提供科学的依据。  相似文献

A comparative study was carried out of temperature dependence of kinetics of dark reduction of bacteriochlorophyll P870 oxidized both by pulsed laser and continuous actinic light in preparations of photosynthetic reaction centres-bacteriochlorophyll-protein complexes from Rhodopseudomonas spheroides, strain 1760-1. Half-time of the recombination of primary products--P870+ and reduced primary electron acceptor A1, which decreases with temperature lowering from 180-240 ms at 120K, is determined. Values of the rate constant of electron transfer from A1 to secondary acceptors at 29,K (2.7.10-1s-1) and the activation energy of this reaction in the range of 298-255K which is 11.8 kcal/mole are calculated.  相似文献

A computational procedure is described for the analysis of fluorescence decay data convolved with a lamp flash of finite width. The computer program calculates the ratio of the Laplace transforms of the decay and the lamp flash for different values of s to give the transforms of the impulse response for each value of s. These are set equal to the analytical Laplace transforms of the decay law involved. Solution of the nonlinear simultaneous equations yields the desired decay parameters. The method can be modified to analyze data that contains a component due to scattered light and can also provide essential information regarding transit time changes of the photomultiplier with changes in emission wavelength. The method was tested by the analysis of real and simulated data. The accuracy of the analysis depends on the degree of correlation among the parameters.  相似文献

Previously untanned buttock skin of 4 volunteers (skin type II; tan with difficulty as they sunburn easily) was treated with various sunscreen preparations and solar--simulated radiation (SSR) or SSR alone for 2 weeks. One week later, the treatment sites were challenged with a DNA-damaging dose of SSR--twice the minimal erythema dose (2 MED). Skin biopsy samples were assayed for the levels of unscheduled DNA synthesis (a measure of DNA damage), melanin distribution, and skin thickening. 5-Methoxypsoralen-containing sunscreen preparations plus SSR or SSR alone induced melanogenesis and increased the stratum corneum thickness, but only the former regimen afforded a high degree of protection against subsequent SSR-induced DNA damage. 5-Methoxypsoralen-free sunscreen preparations plus SSR induced negligible tanning, skin thickening, and photoprotection. These findings are relevant to the risk-benefit analysis of sunscreen preparations, especially in skin type II, as they provide evidence that a 5-methoxypsoralen-induced tan is protective against the DNA-damaging effects of solar UV radiation, and thus has the potential to reduce the carcinogenic risk of exposure to such radiation.  相似文献
