A model-building program, XELE, for use in protein crystallography has been written in C under UNIX on a graphics workstation. This program makes full use of the X Window system to display the electron density distribution and to manipulate the polypeptide model, and therefore is named XELE. It utilizes a fast three-dimensional rendering package, Dorè, and is portable to other types of graphics workstations. A part of the program for the man-machine interface uses the library of X Window and X Toolkit, and therefore is highly interactive. The structure analysis program package, PROTEIN, is also implemented in an interactive mode using X Window, and has been interfaced with XELE.  相似文献

The dermatophytes are among the most frequently observed organisms in biomedicine, yet there has never been stability in the taxonomy, identification and naming of the approximately 25 pathogenic species involved. Since the identification of these species is often epidemiologically and ethically important, the difficulties in dermatophyte identification are a fruitful topic for modern molecular biological investigation, done in tandem with renewed investigation of phenotypic characters. Molecular phylogenetic analyses such as multilocus sequence typing have had to be tailored to accommodate differing the mechanisms of speciation that have produced the dermatophytes that are commonly seen today. Even so, some biotypes that were unambiguously considered species in the past, based on profound differences in morphology and pattern of infection, appear consistently not to be distinct species in modern molecular analyses. Most notable among these are the cosmopolitan bane of nails and feet, Trichophyton rubrum, and the endemic African agent of childhood tinea capitis, Trichophyton soudanense, which are effectively inseparable in all analyses. The molecular data require some reinterpretation of results seen in conventional phenotypic tests, but in most cases, phylogenetic insight is readily integrated with current laboratory testing procedures.  相似文献

SINCE 6-aminopenicillanic acid became available1, many semisynthetic penicillins carrying an acyl moiety on the 6-amino-group have been prepared. Thereby penicillins with improved oral absorption, resistance to penicillinase and to a lesser degree increased activity towards Gram-negative bacilli have been made available2. Many other N-substitutions are possible, however, but these have not so far resulted in useful compounds2, 3. Phenmetrazine hydrochloride appeared to be a safe anoretic agent and was effective in weight control in 80 per cent of a series of 49 obese patients.Tolerance to the drug did not develop in periods up to 18 weeks of continued treatment.Phenmetrazine was most effective in patients under 45 years of age in this series.Side effects were minimal and easily controlled. A preparation combining piperazine and senna was clinically tested among 31 families in which at least one member was found to be positive for enterobiasis by the customary cellophane-tape anal smear technique. The diagnostic smears were positive for 58 of those tested. Following a single dose of piperazine-senna mixture, all patients were cleared of the infestation as determined by the customary criteria for cure. In six patients in two families, reinfestation occurred in two to three months after the first administration. All were again cleared with one dose of the mixture.Side effects were insignificant and transient. One 4-year-old child vomited, but an hour later ingested a second dose without incident. This report describes a method for quantifying -endotoxin contents in spray-dried preparations ofBacillus thuringiensis strain GC-91. -Endotoxin is solubilized in the pro-toxin form and separated from other soluble compounds by ion exchange chromatography. The method does not discriminate between the different -endotoxins produced by strain GC-91. It is suitable for quality control, since -endotoxin concentrations found in different preparations correlate inversely with the LC 50 in bioassays on artificial diet against freshly hatched larvae ofSpodoptera littoralis. A typical batch of spray-dried material contains 1.22%±0.08% -endotoxin. The method is most accurate with preparations containing 0.5 to 2.0% toxin, for which triplicate estimations give standard errors close to±0.2%.  相似文献

A new antipruritic, trimeprazine, was used in the treatment of itching associated with a variety of dermatologic disorders in 215 patients.Good to excellent relief of itching was experienced by 71 per cent of the patients, fair relief by 15 per cent and poor relief by 14 per cent.Side effects included mild and transient drowsiness or dizziness that cleared spontaneously; six cases of drowsiness that necessitated discontinuance of the drug; three cases of drowsiness that was controlled by reduction of the dosage; and one case of generalized eruption that cleared upon discontinuance of the drug.  相似文献
