We studied the effect of moderate undernutrition on the fatty acid composition of adipose tissues in reindeer calves (<1 year) between early winter and late spring. Calves studied in early winter (December) had grazed on natural pastures and were in good condition, while the calves in spring (April) had been maintained on a negative energy balance since December, had lost approximately 16% of body weight and were in a moderate undernutritional state. The fatty acid composition of total lipids in adipose tissues (perirenal-abdominal, peristernal, scapular, intralumbar, and caudal locations) had a high proportion of oleic acid (35-47%) in both seasons. The proportion of oleic acid was significantly lower (29%), and that of palmitic acid (31%) was higher in the adipose tissue of cardiac groove as compared to other locations. There were only small differences in the fatty acid composition of adipose tissues between early winter and spring. However, the proportions of the principal C18-polyunsaturated fatty acids (PUFAs), linoleic and alpha-linolenic acid, were significantly lower in all adipose tissues in calves in poor than in good condition. The observations suggest that linoleic and alpha-linolenic acids may be selectively mobilized from adipose tissues of undernourished reindeer during winter.  相似文献

We examined serum fatty acid composition in reindeer during the close postnatal period (from <8 h to 3 weeks) by using maternal serum as a reference point and focusing on the proportions of polyunsaturated fatty acids (PUFAs) in serum lipids. A striking dissimilarity was found in the serum PUFAs between the neonatals and their mothers. In particular, the proportions of linoleic acid (18:2) and α-linolenic acid in serum cholesteryl esters and phospholipids of the newborn reindeer were significantly lower than those of the mothers. Furthermore, serum phospholipids of the newborns had lower arachidonic acid and docosapentaenoic acid but higher docosahexaenoic acid proportions than the maternal phospholipids. Although the proportions of the principal C18 PUFAs were low in reindeer milk, they increased sharply in serum cholesteryl esters and phospholipids of the calves during the first few days after birth. In particular, there were significant positive correlations in the proportions of 18:2 between serum and milk lipids. We conclude that the proportions of the serum C18 PUFAs are low in the newborn reindeer, but they are increased during the close perinatal period by a rate which suggests an efficient mechanism for selective retention of these fatty acids from milk lipids. To study whether moderate under-nutrition causes muscle wasting, reindeer (Rangifer tarandus tarandus L.) calves were fed either pelleted reindeer feed ad libitum (n=8) or restricted amounts of lichens (n=8). The restricted amount was 60% of ad libitum intake of lichens, and the feeding period was 6 weeks preceded by a 2-week adjustment period. Biopsy samples from the middle gluteal muscle (M. gluteus medius) for the analysis of fibre composition and area, as well as for the activity of cathepsin B were taken before the restriction period in November and January, and after the restriction period in April. In all calves the muscle fibre composition remained unchanged during the winter. In the lichen group, the fibre size also remained unchanged, whereas in control calves the cross sectional area of type I and type IIA fibres increased significantly from November to April. Cathepsin B activity decreased in all calves from November to January and remained at that low level for the rest of the study period, which suggests an attenuated rate of protein degradation. These results can be taken as an indication that moderate under-nutrition causes no muscle wasting in reindeer calves, and the decreased availability of nitrogen is partially compensated for by adaptive decrease in protein degradation. Interestingly the adaptive changes in protein metabolism are equally well seen in the well-fed controls as in the undernourished lichen-fed reindeer.  相似文献

A pattern of wolfCanis lupus Linnaeus, 1758 predation on red deerCervus elaphus Linnaeus, 1758 was studied in Bieszczady Mountains in 1991–2002. In total 324 remains of red deer > 4 months old, killed by wolves throughout the year, were found. The sex, age and bone marrow fat content of wolf kills were compared with the same characteristics within the free living red deer population. The overall contribution of calves killed by wolves (24%) in October-May was higher than in the population (17%), and decreased from autumn to spring. Adult males were more vulnerable to wolf predation than adult females: stags constituted 62% and hinds 38% of adult red deer killed by wolves, whereas in the population, the percentages were 37 and 63%, respectively. Stags killed by wolves were younger ([`(x)] = 4.1\bar x = 4.1 years old) than hinds ([`(x)] = 8.9\bar x = 8.9 years old). Wolves killed more > 8 years old hinds and < 5 years old stags than available in the population. In wolf kills, the average fat content in femur marrow was higher among hinds (84.9%) than stags (69.3%) and calves (66.1%). Only 8% of hinds had < 70% femur marrow fat content, whereas 40% of calves and 38% of stags had marrow fat values below that level. Marrow fat content showed seasonal variation and was the lowest in March among all sex-age classes. The monthly share of stags in all kills, and hinds in hind-calf part of the sample was negatively correlated with their monthly average bone marrow fat content, and monthly share of calves was positively correlated with monthly average bone marrow fat content of adults. The segregation of social units (hind-calf and stag groups), except during the rutting season, and the low fat reserves of males from midwinter until spring contribute to the high overall incidence of calves and adult males and the relatively low incidence of adult females among wolf kills.  相似文献

Platelets are formed by fragmentation of the cytoplasm and plasma membrane of the megakaryocyte in the bone marrow. This study has compared the lipid composition of guinea pig platelets and megakaryocytes. Phospholipids were quantitated by TLC and measurement of lipid phosphorus. Cholesterol and fatty acids were quantitated by GLC. The cholesterol/phospholipid molar ratio was 0.35 in megakaryocytes and 0.55 in platelets. The phospholipid distribution in megakaryocytes was: 9.8% phosphatidylserine, 6.7% phosphatidylinositol, 14.2% sphingomyelin, 40.0% phosphatidylcholine and 29.3% phosphatidy lethanolamine. Platelets contained 11.2% phosphatidylserine, 5.1% phosphatidylinositol, 16.1% sphingomyelin, 38.6% phosphatidylcholine and 29.0% phosphatidylethanolamine. The major megakaryocyte fatty acids were 20.0% palmitic, 16.4% stearic, 20.6% oleic, 13.2% linoleic and 8.2% arachidonic. The major platelet fatty acids were 17.4% palmitic, 17.5% stearic, 11.6% oleic, 12.4% linoleic and 14.6% arachidonic. The minor fatty acids were found in similar proportions in both cells. The major and minor fatty acid compositions of the individual platelet phospholipids reflected those of the megakaryocyte counterparts. The increased arachidonic acid and decreased oleic acid in platelets relative to megakaryocytes were found in all four glycerophospholipids. The similarity of the phospholipid and fatty acid composition of megakaryocytes and platelets suggests that the lipid composition of the platelet is determined by the megakaryocyte.  相似文献

Electrocardiograms were recorded for 50 Finnish reindeer of age 1 day to 9 years, using standard and unipolar extremity leads in the frontal plane. The range of heart rate was 40-250 beats per minute. Atrial and ventricular depolarization times were 5-10 ms shorter in newborn calves than in other reindeer, and showed only minor variance with respect to heart rate. The duration of depolarization was dependant on heart rate, and was more rapid in young calves. A-V conduction time decreased with increasing heart rate. The mode of the QRS vector was 280-290 degrees, and the mode of the T vector 200-230 degrees in the older animals, but 100-120 degrees in young calves. The main deflection in the QRS complex was S in the leads II, III and aVF.R was dominant in aVL and I.  相似文献

Keratoconjunctivitis outbreaks occur each summer in reindeer (Rangifer tarandus tarandus) herds in western Alaska, USA. This condition has not been well characterized nor has a definitive primary etiologic agent been identified. We evaluated the eyes of 660 calves near Nome, Alaska, between 29 June and 14 July 2005. Clinical signs of keratoconjunctivitis were observed in 26/660 calves (3.9%). Samples were collected from the conjunctival sac of both affected (n=22) and unaffected (n=24) animals for bacterial culture, enzyme-linked immunosorbent assay testing for Chlamydophila psittaci, and for polymerase chain reaction assays for Mycoplasma and Moraxella spp. No primary bacterial or viral etiologic agent(s) were isolated or identified. The cause of keratoconjunctivitis among reindeer calves was not determined, but it could involve an anaerobic bacteria, a difficult-to-isolate viral agent, stress associated with repeated handling, ocular foreign bodies, exposure to corral dust or arthropods, or a combination.  相似文献

This paper reports the results of our analysis of the impact high levels of de novo fatty acids have on the proportions of essential and non-essential fatty acids in human milk lipids. The data for seven fatty acids (linoleic, alpha-linolenic, arachidonic (AA), docosahexaenoic (DHA), palmitic, stearic and oleic) were derived from several studies conducted in Nigeria. The proportion by weight of each of these fatty acids was plotted versus the proportion of C10-14 fatty acids. As the proportion of C10-14 fatty acids increased from 15 to 65%, there was not a proportional decrease in the percentages of all seven fatty acids, but, instead, preferential incorporation of the essential fatty acids, AA and DHA into the triacylglycerol component of the milk. At the same time, the proportions of stearic and oleic acid declined by 69% and 86%, respectively. However, the proportions of linoleic acid, palmitic acid, DHA, AA and alpha-linolenic acid, in milk lipids decreased by only 44%, 40%, 39%, 28% and 2.3%, respectively. These observations indicate that as the contribution of C10-14 fatty acids increases, essential fatty acids are preferentially incorporated into milk triacylglycerols at the expense of oleic acid and stearic acid.  相似文献

Age, sex and legbone marrow fat content of 90 red deer Cervus elaphus killed by wolves Canis lupus in winter in southeastern Poland were examined 1984–1988. The majority of kills were calves (44%) and hinds (40%); stags formed only 16%. The average age of hinds was 7.2 yr compared to 5.3 yr in stags. Animals older than 10 yr comprised only 13% of prey. Adults showed high femoral fat content (> 80%) throughout the winter (76% in early winter and 52% in late winter). Among calves femur marrow fat varied through the winter. In early winter 70% of calves had a high fat content (> 80%), though by late winter only 20% had such a high fat content, whilst 43% had low fat content (< 20%).  相似文献

1. Three groups of female rats (8-12 weeks old) were maintained respectively on a linoleic acid-rich diet, a linoleic acid-poor predominantly saturated-fatty acid diet and a normal diet. Changes in the fatty acid compositions of serum, brain, brain mitochondria-rich fraction and myelin were observed. 2. Of the serum fatty acids, linoleic acid showed the greatest change in the percentage of the total acids in response to diet; the change in the proportion of oleic acid was considerable. The percentages of arachidonic acid in serum fatty acids in the groups on the linoleic acid-rich and linoleic acid-poor diets were similar, but higher than those in the normal group. 3. Changes in the proportions of linoleic acid, arachidonic acid and docosahexaenoic acid occurred in brain fatty acids that to some extent paralleled those occurring in the serum. Changes in the proportions of most other acids in the serum fatty acids were not accompanied by corresponding changes in the brain fatty acids. 4. The percentage fatty acid compositions of a mitochondria-rich fraction and myelin are given, and changes in the relative proportions of linoleic acid, arachidonic acid and possibly some docosapolyenoic acids were demonstrated to occur as a result of diet. 5. The results are discussed in relation to the possible aetiology of multiple sclerosis.  相似文献

The expression of monocarboxylate transporters MCT1, MCT2 and MCT4 in the rumen, small intestine and liver was examined in free-ranging and captive reindeer. In addition, expression of chaperone protein CD147, which is needed for the activity of MCT1 and MCT4, was studied in the rumen of suckling calves. Immunoblotting of cell membrane proteins showed the expression of MCT1 and MCT4, but not that of MCT2 in the rumen of reindeer. In free-ranging reindeer the amount of MCT1 was higher than in the captive ones (P<0.01). Developing rumen of suckling calves expressed MCT1 and MCT4 and positive correlation was found between MCT1 and CD147. Both MCT1 and CD147 correlated also with age in calves less than 10 days. In the small intestine all the isoforms studied were expressed, but the amounts were lower than in the rumen (P<0.05). In the liver MCT1 and MCT2 were found while MCT4 was nearly undetectable. The expression of MCT isoforms in the rumen and small intestine reflects the site of absorption and concentrations of short chain fatty acids (SCFA). In the liver the expression of high affinity transporters, MCT1 and MCT2, is in accordance with almost complete uptake of propionate from portal blood.  相似文献

Reindeer calves (n = 632) were slaughtered in November/December (n = 476) or in January (n = 156). Dressed weights and amount of perirenal fat were recorded, and the reproductive organs were collected. A separate group of 130 reindeer calves were weighed at 7 months of age and were followed up with repeated weighings and pregnancy examinations up to 21 months. The onset of puberty and the pregnancy rate were significantly influenced by body weight and the amount of perirenal fat. Approximately 60 g of perirenal fat and 22 kg dressed weight were found at the lower limits for pregnancy. A total of 222 (35%) animals had reached puberty and 126 (20%) were pregnant when examined after slaughter. Animals which conceived during their first autumn showed only a moderate weight gain the following year, and the calf mortality rate in these animals was 47.4%. It was concluded that calf pregnancies are common among the reindeer of Southern Norway and that measures need to be taken to prevent them.  相似文献

Considerable behavioural evidence supports harassment by insects as the most important causal link between warm summer temperatures and low body condition of reindeer Rangifer tarandus , and that insect activity is influenced by weather condition. However, much less is known about the effect of insect harassment on individual performance, measured as reduced weight gain during summer, and the related consequences on both the reindeer pastoral economy and reindeer as a biological resource. Using climatic data, this paper develops a simple index for the analysis of insect harassment that takes into consideration weather variables known to significantly affect insect activity and/or the level of insect harassment. The insect harassment index, which is based on mid-day ambient temperature ≥13 °C, wind speed <6 m/s and cloud cover <40%, is further used to test the hypothesis that insect harassment has a negative effect on reindeer performance during summer in three Norwegian populations. Results show that harassment by insects negatively affects the autumn weight of reindeer calves, most probably through reduced grazing time and increased energy expenditure, but also indirectly by negatively influencing milk production of the dam. Moreover, female calves were more vulnerable to insect harassment than males. Insect harassment may have consequences on future reproductive performance, calving time, calf birth weight and hence neonatal mortality, and thus affect reindeer productivity. The presented index is easy to estimate and may be used to quantify and compare harassment levels on various reindeer summer grazing areas for management purposes. Our results also suggest that the expected temperature increase in the course of global warming may increase the insect-related stress on reindeer.  相似文献

The fatty acid spectra of 6 periphyton communities developed in laboratory streams at different combinations of light intensity and current velocity were determined by gas-liquid chromatography and silver nitrate thin-layer chromatography. Differences in species composition of the communities apparently had no striking effect on proportions of palmitic and stearic acids, whereas concentrations of myristic, palmitoleic, oleic, linoleic, and linolenic acids and a C20:5 acid were more closely related to taxonomic differences. In general, communities dominated by blue-green algae exhibited relatively high proportions of oleic, linolenic, and linolenic acids and low proportions of palmitoleic acid and a C20:5 acid, as compared to communities consisting primarily of diatoms. The data also indicated an inverse relationship between fatty acid redundancy and species diversity.  相似文献

Perturbation of the fatty acid composition of human lymphocytes in vitro was investigated by addition of linoleic acid complexed to bovine serum albumin (BSA-LA) and by mitogenic stimulation with phytohaemagglutinin (PHA). BSA-LA resulted in a 45% increase in linoleic acid in phosphatidylethanolamine (PE) and over 100% in phosphatidylcholine (PC) in peripheral blood cells. Supplementation with BSA-LA in PHA-stimulated lymphocytes produced even greater changes: 100% increase in linoleic acid content for PE and over 300% for PC. There was a large decrease in oleic acid: 40% for PE and almost 100% in PC. Significant decreases in arachidonic acid occurred in both phospholipid fractions. PHA alone also altered membrane phospholipid fatty acid composition, with reductions in palmitic, stearic and linoleic acid for PE and increases in oleic acid and arachidonic acid (almost 100%). For PC, there were large decreases in stearic (40%), linoleic (30%) and arachidonic (40%) acids, together with an increase in oleic acid (65%). Cells supplemented with linoleic acid grown in the presence of PHA, compared with those grown in linoleic acid-supplemented medium alone, showed a 40% decrease in palmitic acid and a 55% increase in arachidonic acid in PE. For PC, there were large decreases in stearic acid (40%) and arachidonic acid (57%). Antibody-induced redistribution of surface molecules ('capping') was inhibited by some 14% after incubation with BSA-LA. However, no consistent alterations in PHA-induced cell proliferation were observed. These data suggest that profound alterations of membrane fatty acid composition occur spontaneously during the mitotic cycle, and may be further induced by experimental manipulation, without gross perturbation of cell function.  相似文献

The effects of inclusion of different fatty acids in the medium on the rate of esterification of palmitic acid and its stereospecific distribution among the three positions of the triacyl-sn-glycerols by preparations of rat adipocytes in vitro have been determined. Myristic acid, stearic acid, oleic acid and linoleic acid were used as diluents and the concentration of the combined unesterified fatty acids in the medium was held constant; only the proportion of palmitic acid was varied. The amount of palmitic acid esterified was always linearly related to its relative concentration in the medium and was not significantly affected by the nature of the diluent fatty acid chosen. Constant relative proportions were recovered in triacylglycerols and in intermediates in each instance. The amount of palmitic acid esterified to each of the positions of the triacyl-sn-glycerols was linearly dependent on the relative proportion in the medium but the nature of the relationship was markedly influenced by which fatty acid was present. When stearic acid was present, simple relationships were found over the whole range tested. When either myristic acid, oleic acid or linoleic acid was present, abrupt changes in the manner of esterification of palmitic acid were observed in position sn-1 when the relative concentrations of palmitic acid and the diluent reached critical values, which differed with each fatty acid. In position sn-2 when oleic acid or linoleic acid was present, a similar change was observed, and in position sn-3 it was obtained with myristic acid as diluent. The results are discussed in terms of changes in the relative affinities of the acyltransferases for palmitic acid. Palmitic acid was esterified into various molecular species in proportions that indicated acylation with non-correlative specificity at higher relative concentrations but not at lower.  相似文献

Semen collected from reindeer bulls in an artificial vagina was used for the artificial insemination of 16 reindeer cows: two with undiluted semen, five with diluted semen and nine with frozen semen. The two cows which received undiluted semen gave birth to normal calves 217 days later. The other cows returned to oestrus 23.5 days (mean) after insemination and subsequently calved, having been served by a fertile bull.  相似文献

Thermoregulatory capacities of 51 reindeer calves (Rangifer tarandus tarandus L.) aged 1-35 days were studied at -26.5 to +35.0 degrees C ambient temperatures at Kaamanen reindeer research station, Finland (69 degrees 10' N) during calving periods in May 1981 and May-July 1982. The newborn calves aged 1-4 days maintained a high body temperature (Tre) (mean +40.2 degrees C) even at the lowest experimental temperature of -22.5 degrees C by increasing their metabolic rate five-fold above the level at +11.0 degrees C. Heat production of the new-born calves was largely based on the metabolism of brown adipose tissue, stimulated by cold-induced discharge of the sympathetic nervous transmitter, noradrenaline (NA). Sensitivity of the calves to exogenous NA disappeared during the first 3-4 weeks of life. Thermal conductance of the calves was low at low ambient temperatures, but rose strongly as Ta increased above +10 degrees C. The extensive peripheral cooling, especially in the feet, was demonstrated in the calves aged 1-10 days. The lowest foot temperature (+10.5 degrees C) was measured in a 4-day-old calf at -14.5 degrees C. Slight shivering thermogenesis was recorded in the calves aged 1-4 days and occasionally in the older calves at low values of Ta. Shivering appears to be a reserve mechanism against severe cold. At about +20 degrees C and above the calves increased their Tre (approximately 1 degree C), oxygen consumption and heart rate. In the newborn calves oxygen consumption rose four- to five-fold and in 1-month-old calves about two-fold. Fast growing calves (maximum 400 g/day) appear to be more stressed by heat than by cold exposure.  相似文献

Stimulated neutrophils produce hypochlorous acid (HOCl) via the myeloperoxidase-catalyzed reaction of hydrogen peroxide with chloride. The reactions of HOCl with oleic, linoleic, and arachidonic acids both as free fatty acids or bound in phosphatidylcholine have been studied. The products were identified by gas chromatography-mass spectrometry of the methylated and trimethylsilylated derivatives. Oleic acid was converted to the two 9,10-chlorohydrin isomers in near stoichiometric yield. Linoleic acid, at low HOCl:fatty acid ratios, yielded predominantly a mixture of the four possible monochlorohydrin isomers. Bischlorohydrins were also formed, in increasing amounts at higher HOCl concentrations. Arachidonic acid gave a complex mixture of mono- and bischlorohydrins, the relative proportions depending on the amount of HOCl added. Linoleic acid appears to be slightly more reactive than oleic acid with HOCl. Reactions of oleic and linoleic acids with myeloperoxidase, hydrogen peroxide, and chloride gave chlorohydrin products identical to those with HOCl. Lipid chlorohydrins have received little attention as products of reactions of neutrophil oxidants. They are more polar than the parent fatty acids, and if formed in cell membranes could cause disruption to membrane structure. Since cellular targets for HOCl appear to be membrane constituents, chlorohydrin formation from unsaturated lipids could be significant in neutrophil-mediated cytotoxicity.  相似文献

A non-aqueous reverse-phase HPLC method, based on two columns in series, has been used to separate the major triacylglycerols (TAGs) from commercial castor oil and to perform either on-line negative ion atmospheric pressure chemical ionisation (APCI), or off-line positive ion matrix-assisted laser desorption ionisation (MALDI)/MS. The resulting Mass Spectra showed chloride-attached TAG molecules [M + CI]- in the case of negative-ion APCI, and sodium-attached TAG molecules [M + Na]+ in the case of positive-ion MALDI. For MALDI time-of-flight (TOF)/MS, a liquid binary matrix system consisting of sodium ferrocyanide and glycerol was applied, resulting in excellent TAG sensitivity, which was necessary for the determination of trace amounts of TAGs in castor oil. Both techniques allowed unambiguous molecular mass determination of the intact TAG molecules with no thermal degradation. Furthermore, seamless post source decay (PSD) fragment ion analysis by means of a curved field reflector TOF mass spectrometer allowed the determination of the fatty acid composition of each individual TAG. Castor oil contained eight different TAGs which were successfully determined by both APCI and MALDI techniques. In each TAG, at least two units of 12-hydroxy-9-octadecenoic acid (ricinoleic acid) were present. The following fatty acids were determined by seamless PSD fragment ion analysis and APCI/MALDI molecular mass determination as TAG substructures: ricinoleic acid, palmitic acid, stearic acid, oleic acid, linoleic acid, linolenic acid, dihydroxy stearic acid and eicosenoic acid.
