Controlled cultivation of marine macroalgal biomass such as Ulva species, notably Ulva lactuca, is currently studied for production of biofuels or functional food ingredients. In a eutrophic environment, this macrophyte is exposed to varying types of nutrient supply, including different and fluctuating levels of nitrogen sources. Our understanding of the influences of this varying condition on the uptake and growth responses of U. lactuca is limited. In this present work, we examined the growth response of U. lactuca exposed to different sources of nitrogen (NH4+; NO3; and the combination NH4NO3) by using photo-scanning technology for monitoring the growth kinetics of U. lactuca. The images revealed differential increases of the surface area of U. lactuca disks with time in response to different N-nutrient enrichments. The results showed a favorable growth response to ammonium as the nitrogen source. The NH4Cl and NaNO3 rich media (50 μM of N) accelerated U. lactuca growth to a maximum specific growth rate of 16.4 ± 0.18% day−1 and 9.4 ± 0.72% day−1, respectively. The highest biomass production rate obtained was 22.5 ± 0.24 mg DW m−2·day−1. The presence of ammonium apparently discriminated the nitrate uptake by U. lactuca when exposed to NH4NO3. Apart from showing the significant differential growth response of U. lactuca to different nitrogen sources, the work exhibits the applicability of a photo-scanning approach for acquiring precise quantitative growth data for U. lactuca as exemplified by assessment of the growth response to two different N-sources.  相似文献

Biotreatment of bagasse effluent using Phanerochaete chrysosporium (white rot fungus) is investigated. This study confirmed that lignin is the major pollutant component in this effluent followed by different carbohydrates. The treatment conditions must be very proper, especially in terms of biomass culture to achieve a successful treatment. The best conditions of temperature, biomass concentration, pH and duration for biotreatment of this effluent were 35°C, 552 mg l−1, 6 and 5 to 9 days, respectively. Under these conditions, a 9 days long treatment reduced by 98.7% the original biochemical oxygen demand (of 2,780 mg l−1) and by 98.5% the dissolved chemical oxygen demand (initial 4,200 mg l−1). Moreover, fungal treatment reduced total dissolved solids from 3,950 to 575 mg l−1 and color from 560 mg l−1 PtCo to 111 mg l−1 PtCo.  相似文献

The seaweed Ulva lactuca L. was spray cultured by mariculture effluents in a mattress‐like layer, held in air on slanted boards by plastic netting. Air‐agitated seaweed suspension tanks were the reference. Growth rate, yield, and ammonia‐N removal rate were 11.8% · d?1, 171 g fresh weight (fwt) · m?2 · d?1, and 5 g N · m?2 · d?1, respectively, by the spray‐cultured U. lactuca, and 16.9% · d?1, 283 g fwt · m?2 · d?1, and 7 g N · m?2 · d?1, respectively, by the tank U. lactuca. Biomass protein content was similar in both treatments. Dissolved oxygen in the fishpond effluent water was raised by >3 mg · L?1 and pH by up to half a unit, upon passage through both culture systems. The data suggest that spray‐irrigation culture of U. lactuca in this simple green‐mattress‐like system supplies the seaweed all it needs to grow and biofilter at rates close to those in standard air‐agitated tank culture.  相似文献

In this study, salt tolerance was investigated in Odyssea paucinervis Staph, an ecologically important C4 grass that is widely distributed in saline and arid areas of southern Africa. Plants were subjected to 0.2%, 10%, 20%, 40%, 60% and 80% sea water dilutions (or 0.076, 3.8, 7.6, 15.2, 22.8, and 30.4 parts per thousand) for 11 weeks. Increase in salinity from 0.2% to 20% sea water had no effect on total dry biomass accumulation, while further increase in salinity to 80% sea water significantly decreased biomass by over 50%. Morphological changes induced by salinity included reductions in the number of culms, leaves and internodes as well as decreases in internode length and leaf length:leaf width ratios. Carbon dioxide exchange, leaf conductance and transpiration decreased at salinities of 40% and higher, while quantum yield of photosystem II (PSII), electron transport rate (ETR) through PSII and intrinsic photosynthetic efficiency generally decreased at salinities of 60% and higher compared to 0.2% sea water. Concentrations of Na+ and Cl increased significantly with salinity increase in both roots and shoots. Na+/K+ ratios in the roots and shoots ranged from 0.66 to 3.28 and increased with increase in substrate salinity. The maximal rate of secretion at 80% sea water was 415 nmol cm−2 d−1 for Na+ and 763 nmol m−2 d−1 for Cl with high selectivity for these two ions. Predawn and midday ψ decreased with increase in salinity and were more negative than those of the treatment solutions. The concentration of proline increased with increase in salinity in both roots and shoots. The data clearly indicated that O. paucinervis is a highly salt-tolerant species that is morphologically and physiologically adapted to a saline environment.  相似文献

A facultatively aerobic deep brown coccoid to ovoid bacterium, strain EG17T, was isolated from a saline effluent stream in the NaCl-dominated brine spring system known as East German Creek in the province of Manitoba, Canada. The strain produced BChl a incorporated into a functional reaction center and two light-harvesting complexes with absorption peaks at 802, 850, and 879 nm. EG17T is the first reported anoxygenic phototroph capable of photoheterotrophic growth under both oxic and anoxic conditions. It yielded proportionally the greatest aerobic photosynthetic biomass under oligotrophic conditions. The results of 16S rRNA gene sequence comparisons revealed that EG17T was related most closely to the aerobic anoxygenic phototrophs Roseibacterium elongatum (98.3%) and quite distantly to both Dinoroseobacter shibae (95.2%) and Roseicyclus mahoneyensis (94.7%). The DNA G + C content was 65.6 mol%. On the basis of the unique dual aerobic/anaerobic photosynthetic capability, the distinctive spectrophotometric absorption of the photosynthetic apparatus, diagnostic physiological and biochemical traits, and the moderate phylogenetic separation between EG17T and its nearest relatives, it is concluded that this microorganism should be classified as a novel genus and species, Charonomicrobium ambiphototrophicum gen. nov., sp. nov., with EG17T as the type strain.  相似文献

A mesocosm study was conducted to determine the effects of variable salinity and light on Vallisneria americana Michx. (wild celery) and associated algal community components in the lower St. Johns River, Florida. Fifteen centimeter diameter intact plant plugs were collected from the LSJR in March 2001 and transported to mesocosm facilities in Lafayette, Louisiana. A factorial experimental design was used consisting of three salinity levels (1, 8, and 18 ppt), three light levels (0, 50, and 90% shading), and three replicate mesocosms of each for a total of 27 mesocosms. The experiment consisted of a 4-week acclimation period followed by a 5-month treatment period. V. americana responded negatively to increased salinity. Although V. americana survived 8 ppt salinity, growth was limited. At 18 ppt, almost all V. americana aboveground biomass had perished within 10 weeks, but when salinity was lowered back to 1 ppt, approximately 20% of the aboveground biomass recovered within the following 10 weeks. At midtreatment harvest, light did not affect V. americana biomass directly (P = 0.8240), but by final harvest (20 weeks) light affected belowground biomass (P < 0.0014). Both salinity and light affected algal growth. Macroalgae dominated 1 ppt salinity treatments in ambient light, but phytoplankton dominated 8 and 18 ppt salinity treatments in ambient light. Algal communities were greatly inhibited by 90% shading. While salinity directly impacted V. americana growth and survival, light effects were less direct and involved algal community associations.  相似文献

Drought and salinity are major abiotic stresses affecting rice production. To improve plant tolerance to salinity and drought, we overexpressed rice Na+/H+ exchangers (OsNHX1) and H+-pyrophosphatase in tonoplasts (OsVP1) in a japonica elite rice cultivar, Zhonghua 11. Compared with our wild-type control, transgenic plants overexpressing both genes incurred less damage when exposed to long-term treatment with 100 mM NaCl or water deprivation. Under high-saline conditions, the transformants accumulated less Na+ and malondialdehyde in the leaves, thereby allowing the plants to maintain a low level of leaf water potential and reduce stress-induced damage. Those transgenics also had higher photosynthetic activity during the stress period. Under those conditions, they also showed an increase in root biomass, which enabled more water uptake. These results suggest that OsVP1 and OsNHX1 improve the tolerance of rice crops against drought and salt by employing multiple strategies in addition to osmotic regulation.  相似文献

The effects of 16 different combinations of nutrient load and agitation on yield, nutrient uptake and proximate chemical composition of the seaweed Ulva lactuca cultured in tanks were evaluated. Intensive fishpond outflow passed through seaweed tanks at four nutrient loading levels and four water agitation combinations of water exchange, bottom aeration and frequently changing water levels (an accelerated tide regime). Specific results from these outdoor experiments were examined further under controlled conditions in laboratory experiments. Agitation treatments affected the performance of U. lactuca only under TAN () load levels below 4 g N m−2 day−1; biofiltration of TAN was the parameter most affected. Biomass yields at each of the four nutrient loading levels were not significantly different between the agitation treatments. Protein content increased significantly with increasing nutrient loading. The agitation treatments had a slight effect on seaweed protein content only at the lowest nutrient loading levels. There were no significant differences in dissolved oxygen concentration, pH, and temperature among the agitation treatments at all nutrient loading levels. Under laboratory conditions, growth rates, protein content, and photosynthetic and biomass yield of the seaweed were affected by water velocity under low nutrient concentrations. It is concluded that the effect of air agitation under the conditions of these experiments was not directly related to photosynthesis, excess dissolved oxygen, or carbon limitation, but to the diffusion of macro nutrients from the water to the seaweed. Therefore, once nutrient concentrations are high enough (above about 4 μM of TAN with the other nutrients in their corresponding proportions), aeration per se is not essential for effective growth and biofiltration by seaweeds.  相似文献

We examined responses of batch cultures of the marine microalga Nannochloropsis sp. to combined alterations in salinity (13, 27, and 40 g/l NaCl) and light intensity (170 and 700 μmol photons/m2·s). Major growth parameters and lipid productivity (based on total fatty acid determination) were determined in nitrogen-replete and nitrogen-depleted cultures of an initial biomass of 0.8 and 1.4 g/l, respectively. On the nitrogen-replete medium, increases in light intensity and salinity increased the cellular content of dry weight and lipids due to enhanced formation of triacylglycerols (TAG). Maximum average productivity of ca. 410 mg TFA/l/d were obtained at 700 μmol photons/m2·s and 40 g/l NaCl within 7 days. Under stressful conditions, content of the major LC-PUFA, eicosapentaenoic acid (EPA), was significantly reduced while TAG reached 25% of biomass. In contrast, lower salinity tended to improve major growth parameters, consistent with less variation in EPA contents. Combined higher salinity and light intensity was detrimental to lipid productivity under nitrogen starvation; biomass TFA content, and lipid productivity amounted for only 33% of DW and ca. 200 mg TFA/l/day, respectively. The highest biomass TFA content (ca. 47% DW) and average lipid productivity of ca. 360 mg TFA/l/day were achieved at 13 g/l NaCl and 700 μmol photons/m2·s. Our data further support selecting Nannochloropsis as promising microalgae for biodiesel production. Moreover, appropriate cultivation regimes may render Nannochloropsis microalgae to produce simultaneously major valuable components, EPA, and TAG, while sustaining relatively high biomass growth rates.  相似文献

The growth, biofiltering efficiency and uptake rates of Ulva clathrata were studied in a series of outdoor tanks, receiving waste water directly from a shrimp (Litopenaeus vannamei) aquaculture pond, under constant aeration and two different water regimes: (1) continuous flow, with 1 volume exchange a day (VE day-1) and (2) static regime, with 1 VE after 4 days. Water temperature, salinity, pH, dissolved inorganic nitrogen (DIN), phosphate (PO4), chlorophyll-a (chl-a), total suspended solids (TSS), macroalgal biomass (fresh weight) and tissue nutrient assimilation were monitored over 12 days. Ulva clathrata was highly efficient in removing the main inorganic nutrients from effluent water, stripping 70–82% of the total ammonium nitrogen (TAN) and 50% PO4 within 15 h. Reductions in control tanks were much lower (Tukey HSD, P < 0.05). After 3 days, the mean uptake rates by the seaweed biomass under continuous flow were 3.09 mg DIN g DW day−1 (383 mg DIN m−2 day−1) and 0.13 mg PO4 g DW day−1 (99 mg PO4 m−2 day−1), being significantly higher than in the static regime (Tukey HSD, P < 0.05). The chl-a decreased in seaweed tanks, suggesting that U. clathrata inhibited phytoplankton growth. Correlations between the cumulative values of DIN removed from the water and total nitrogen assimilated into the seaweed biomass (r = 0.7 and 0.8, P < 0.05), suggest that nutrient removal by U. clathrata dominated over other processes such as phytoplankton and bacterial assimilation, ammonia volatilization and nutrient precipitation.  相似文献

The objective of this paper was to study the potential for bioremoval of a textile dye, Reactive Red 198 (RR198), by a fungus isolated from soil collected from an effluent disposal area near a textile company. The fungus was identified as Aspergillus flavus, and its use as a low-cost live-cell biomass for the biodegradation of RR198 from contaminated water was investigated using batch studies. The effects of time, dye concentration, and pH as variable factors were examined in the process. Results showed that bioremoval of RR198 by A. flavus increased to over 84.96% with increasing time until equilibrium was reached after a period of 24 h. A low pH was the most effective, as were lower levels of dye concentration. The decolorization was determined by the decrease in the absorption maximums of this dye by UV–visible spectroscopy. A. flavus was shown to be an efficient fungus for removal of RR198 from wastewater.  相似文献

Enteromorpha linza (L.) J. Ag. was grown in full strength sewage effluent, various combinations of sewage effluent and seawater, and in natural seawater. It was found that full strength sewage effluent with a salinity of 14 supported best growth of the alga. After a 12 day cultivation period, growth ofE. linza in full strength sewage effluent and 75% sewage effluent- seawater mixture showed 3.5-fold and 2-fold increase in fresh weight over that grown in natural seawater; respectively. Uptake of PO inf4 sup3– -P, NH3-N and NO inf3 sup– -N by cells ofE. linza was extremely efficient in all tested media. Data obtained from the experiments indicated that inorganic nitrogen rather than phosphorus was the limiting nutrient factor for growth ofE. linza in full strength sewage effluent and in other sewage effluent- seawater mixtures. NH3-N at concentrations above 4.5 ppm was found to inhibit uptake of NO inf3 sup– -N in the same culture medium by the algal cells. The fact that sewage grownE. linza contained comparatively much higher protein content (30.2% dry weight) than that grown in natural seawater (12.5% dry weight) leads to the conclusion that sewage grownE. linza could serve as an economically feasible feed for livestock in Hong Kong where the sewage is characterized by having a salinity of approximately 14. It is proposed that this multicellular green alga is a suitable algal species to serve the dual function of wastewater purification through the production of algal protein from sewage effluent having high salinities.  相似文献

Reaumuria vermiculata (L.), a perennial dwarf shrub in the family of Tamaricaceae, is a salt-secreting xero-halophyte found widely in arid areas of Tunisia. In the present study, physiological attributes of R. vermiculata were investigated under salt stress. Four-month-old plants were subjected to various salinity levels (0, 100, 200, 300, 400 or 600 mM NaCl) for 30 days under greenhouse conditions. Results showed that plants grew optimally when treated with standard nutrient solution without NaCl supply. However, increasing osmolality of nutrient solutions caused a significant reduction in biomass production and relative growth rate. This reduction was more pronounced in roots than in shoots. In addition, this species was able to maintain its shoot water content at 30% of the control even when subjected to the highest salt level, whereas root water content seemed to be unaffected by salt. Shoot water potential declined significantly as osmotic potential of watering solutions was lowered and the more negative values were reached at 600 mM NaCl (−3.4 MPa). Concentrations of Na+ and Cl in the shoots of R. vermiculata were markedly increased with increasing osmolality of nutrient solutions, whereas concentration of K+ was not affected by NaCl supply. Salt excretion is an efficient mechanism of Na+ exclusion from the shoots of this species exhibiting high K+/Na+ selectivity ratio over a wide range of NaCl salinity. Proline accumulation in shoots was significantly increased with increase in salt level and may play a role in osmoregulation.  相似文献

Macrobenthos of the shallow (<10 m) nearshore marine waters of northern Kotzebue Sound was examined in 2002–2004 to (1) determine nearshore community structure and (2) assess the influence of sewage disposal. A variable number of benthic stations were sampled during three summers, with extensive effort at the disposal zone in 2003. The benthic community structure is similar to other nearshore Arctic locations, and was similar to a previous benthic study done in 1986–1987. The potential of sewage impact was assessed at the request of the community, because sewage is occasionally discharged into the Sound, in volumes of up to 38 million liters, typically through the ice in early spring. Only minimal effects of disposal on the benthos were evident and the effects could not be separated from the impacts of low salinity and relatively high water pigments. Low diversity (H′) and species richness (d) and high biomass characterized stations in the sewage area. Parameters often associated with extreme sewage pollution, particularly hypoxic and/or anoxic conditions and high abundance of opportunistic taxa, were not observed. Local traditional ecological knowledge was solicited throughout the study, and was used to help define the area potentially affected by sewage disposal.  相似文献

Exposure of coral reefs to river plumes carrying increasing loads of nutrients and sediments is a pressing issue for coral reefs around the world including the Great Barrier Reef (GBR). Laboratory experiments were conducted to investigate the effects of changes in inorganic nutrients (nitrate, ammonium and phosphate), salinity and various types of suspended sediments in isolation and in combination on rates of fertilisation and early embryonic development of the scleractinian coral Acropora millepora. Dose–response experiments showed that fertilisation declined significantly with increasing sediments and decreasing salinity, while inorganic nutrients at up to 20 μM nitrate or ammonium and 4 μM phosphate had no significant effect on fertilisation. Suspended sediments of ≥100 mg l−1 and salinity of 30 ppt reduced fertilisation by >50%. Developmental abnormality occurred in 100% of embryos at 30 ppt salinity, and no fertilisation occurred at ≤28 ppt. Another experiment tested interactions between sediment, salinity and nutrients and showed that fertilisation was significantly reduced when nutrients and low concentrations of sediments co-occurred, although both on their own had no effect on fertilisation rates. Similarly, while slightly reduced salinity on its own had no effect, fertilisation was reduced when it coincided with elevated levels of sediments or nutrients. Both these interactions were synergistic. A third experiment showed that sediments with different geophysical and nutrient properties had differential effects on fertilisation, possibly related to sediment and nutrient properties. The findings highlight the complex nature of the effects of changing water quality on coral health, particularly stressing the significance of water quality during coral spawning time. In the past decade, algal waste has been used as useful natural resource for production of enormous range of products that have wide economical and commercial importance. Pectinases are group of enzymes that have wide commercial applications. Hence, current study was designed to utilize algal biomass for the production of pectinases using submerged (SmF) and solid state fermentation (SSF) techniques. Different algal sources including brown (Dictyopteris polypodioides, Sargassum wightii and Dictyopteris divaricata) and green algae (Ulva lactuca and Codium tomentosum) were used and U. lactuca was found to be the most suitable substrate. Several bacterial and fungal strains were screened and among them Bacillus licheniformis KIBGE-IB4 was selected based on maximum pectinase production. SmF and SSF were studied utilizing U. lactuca as a substrate and results revealed that enzyme production was favoured by SmF (2457?±?3.31?U?mg?1) as compared to SSF (1432?±?1.46?U?mg?1). Parametric optimization of pectinase production indicated that B. licheniformis KIBGE-IB4 requires 10.0?g L–1 U. lactuca as a biomass in the medium with a pH 7.0 when incubated at 37?°C for 24 hours. Likewise, production of pectinase using algal resource was also compared with that of the conventional agricultural biomass and it was observed that when U. lactuca was used, the selected bacterial isolate produced a higher yield of enzyme than sugarcane bagasse and rice husk. Hence, it is anticipated that algal biomass can be efficiently utilized as an environmental friendly bioresource for the production of industrially important hydrolytic enzymes.  相似文献

We investigated the ability of the ichthyotoxic haptophyte Prymnesium parvum to use sewage-originated nutrients applying stable carbon (C) and nitrogen (N) isotope techniques. P. parvum was cultured under N and phosphorus (P) sufficient and deficient conditions in either sewage effluent-based medium or in a nitrate- and phosphate-based control. Cell densities and toxicities were monitored and stable carbon N isotopes signatures (δ13C and δ15N) of P. parvum and the sewage effluent analysed. Nitrogen and P sufficient cultures achieved the highest biomass followed by P and N deficient cultures, regardless of sewage effluent additions. The P deficient cultures with sewage effluent had higher toxicity, estimated as haemolytic activity (9.4 ± 0 × 10?5 mg Saponin equiv. cell?1) compared to the P deficient control and to all N deficient and NP sufficient cultures. Nutrient deficient conditions had no effect on the cell δ15N, but a decreasing effect on δ13C in the inorganic N deficient treatment. Growth in sewage-based media was followed by a substantial increase in the cell δ15N (10.4–16.1‰) compared to the control treatments (2.4–4.9‰), showing that P. parvum is capable of direct use of sewage-originated N, inorganic as well as organic. Uptake of terrestrial derived C in the sewage treatments was confirmed by a decrease in cell δ13C, implying that P. parvum is able to utilize organic nutrients in sewage effluent.  相似文献

Lake Vesijärvi, southern Finland, suffered sewere eutrophication by sewage effluent from the city of Lahti during the 1960's and the early 1970's. The municipal sewage loading was diverted from the lake in 1976 and the lake started to recover. However, in the 1980's blue-green algal blooms increased again and the recovery of the lake faded. Enclosure experiments demonstrated that high roach (Rutilus rutilus) biomass is one of the key factors in the fading recovery of the lake. In this study, the influence of roach and another cyprinid fish species (bleak, Alburnus alburnus) to planktonic algal productivity and biomass in Lake Vesijärvi was examined. Enclosure experiments in the field showed the impacts of planktivorous bleak on water quality; in an enclosure with a density of 1 fish m–2 average daily algal production (1370 mg C m–2) and chlorophyll-a concentration (50–90 µg 1–1) were more than twice that in an enclosure without fish. Laboratory experiments showed that the availability of planktonic food affects the foraging behaviour of roach and consequently the internal nutrient loading from the sediment into the water. Roach caused the highest phosphorus loading and turbidity when there was no zooplanktonic food available in the water. The possible interactions between planktivorous and omnivorous fish species are discussed.  相似文献

Members of the genus Ulva are widespread and abundant in intertidal and shallow subtidal areas but there are conflicting data regarding susceptibility to herbivory. While some studies have documented that Ulva spp. were favored by a diversity of marine herbivores, other work has revealed herbivore deterrence. We investigated grazing and growth rates of the littorinid species, Littorina littorea and L. obtusata, when offered Fucus vesiculosus, Ascophyllum nodosum, Ulva lactuca, and Chondrus crispus, highlighting distinctive vulnerabilities to toxic effects of U. lactuca. Ulva lactuca was the preferred food of L. littorea, while L. obtusata showed no grazing on this ephemeral algal species. In contrast, F. vesiculosus was highly preferred by L. obtusata. Although L. littorea demonstrated a grazing preference for U. lactuca, growth rate of this gastropod species was nearly 3× greater when fed F. vesiculosus, suggesting a non‐lethal, negative effect of U. lactuca on L. littorea with long‐term exposure. Mortality of L. obtusata ranged from 0% to 100% when held in the presence of various Ulva densities for 1 week, and Ulva exudate depressed herbivory of this gastropod. We conclude that the water‐soluble, toxic exudate produced by U. lactuca in response to herbivory had allelochemical properties, and may contain a cleavage product (acrylic acid) of dimethylsulfoniopropionate or reactive oxygen species (i.e., H2O2). Observed differences in susceptibility to Ulva toxicity by the littorinid species may be related to generalist versus specialist feeding and habitat strategies.  相似文献

We examined changes in the expression of Na+/K+-ATPase mRNA in the gills of the cinnamon clownfish using quantitative real-time PCR in an osmotically changing environment [seawater (35 psu; practical salinity unit, 1 psu ≈ 1‰) → brackish water (17.5 psu) and brackish water with prolactin]. The expression of Na+/K+-ATPase mRNA in gills was increased after the transfer to brackish water, and the expression was repressed by prolactin treatment. Also, activities of gill Na+/K+-ATPase and plasma cortisol levels increased after the transfer to brackish water and were repressed in brackish water with prolactin treatment. Na+/K+-ATPase-immunoreactive cells were almost consistently observed in the gill filaments, but absent from the lamella epithelia. The plasma osmolality level decreased in brackish water, but the level of this parameter increased in brackish water with prolactin treatment during salinity change. These results suggest that the Na+/K+-ATPase gene plays an important role in osmoregulation in gills, and prolactin improves the hyperosmoregulatory ability of cinnamon clownfish in a brackish water (hypoosmotic) environment.  相似文献
