In legume plants, the determination of individual seed weight is a complex phenomenon that depends on two main factors. The first one corresponds to the number of cotyledon cells, which determines the potential seed weight as the cotyledon cell number is related to seed growth rate during seed filling. Since cell divisions take place between flowering and the beginning of seed filling, any stress occurring before the beginning of seed filling can affect individual seed growth rate (C and N reserve accumulation in seeds), and thus individual seed weights. The second factor concerns carbon and nitrogen supply to the growing seed to support reserve accumulation. Grain legume species produce protein-rich seeds involving high requirement of nitrogen. Since seed growth rate as determined by cotyledon cell number is hardly affected by photoassimilate availability during the filling period, a reduction of photosynthetic activity caused by nitrogen remobilization in leaves (e.g., remobilization of essential proteins involved in photosynthesis) can lead to shorten the duration of the filling period, and by that can provoke a limitation of individual seed weights. Accordingly, any biotic or abiotic stress during seed filling causing a decrease in photosynthetic activity should lead to a reduction of the duration of seed filling.  相似文献

The seed development and composition of Medicago truncatula Gaertn., the new model plant for grain legumes, was studied using nine genotypes of the species complex: M. truncatula-Medicago littoralis (M. truncatula). The seed development of M. truncatula was very similar to that of other legumes, the only notable exception being the presence, in the mature seed, of an endosperm layer that is absent in grain legumes. During early embryogenesis and until mid-maturation, transient storage of starch occurred in the seed coat and embryo. This temporary storage probably contributed to the early development of the embryo and reserve synthesis. During maturation the synthesis and accumulation of proteins and oil took place at quasi-constant rates. Conversely oligosaccharides, mainly stachyose, were synthesised only during late maturation and at the beginning of desiccation. Proteins represented the major class of storage compounds and their average amino acid composition was found to be very close to that of pea and robust in various environmental conditions. Similar compositions between the two species and other grain legumes were also found for the fatty acids and the soluble sugars; most of these characters varied depending on the various environmental conditions used for seed production. All these similarities fully justify the use of M. truncatula as a model plant for genomic approaches to grain legume improvement. The major difference between M. truncatula seeds and European grain legume seeds resides in the nature of their carbon storage namely triacylglycerides for M. truncatula and starch for pea and faba bean.  相似文献

A series of galactomannans with varying degrees of galactose substitution have been extracted from the endosperms of legume seeds with water and alkali and the amount of substitution required for water solubility has been determined. Some were heterogeneous with respect to the degree of galactose substitution. The structural requirements for hydrolysis by plant β-mannanase have been studied using the relative rates and extents of hydrolysis of these galactomannans. A more detailed examination of the products of hydrolysis of carob galactomannan has been made. At least two contiguous anhydromannose units appear to be needed for scission. This is similar to the requirement for hydrolysis by microbial enzymes. Judas tree (Cercis siliquastrum) endosperm contained a polysaccharide with a unique composition for a legume seed reserve. Gel chromatography and electrophoresis on cellulose acetate indicated homogeneity. Hydrolysis with a mixture of β-mannanase and α-galactosidase gave a glucose-mannose disaccharide and acetolysis gave a galactose-mannose. These results, as well as the pattern of hydrolysis by β-mannanase were consistent with a galactoglucomannan structure.  相似文献

We utilized a proteomic approach to investigate seed development in Medicago truncatula, cv Jemalong, line J5 at specific stages of seed filling corresponding to the acquisition of germination capacity and protein deposition. One hundred twenty proteins differing in kinetics of appearance were subjected to matrix-assisted laser desorption ionization time of flight mass spectrometry. These analyses provided peptide mass fingerprint data that identified 84 of them. Some of these proteins had previously been shown to accumulate during seed development in legumes (e.g. legumins, vicilins, convicilins, and lipoxygenases), confirming the validity of M. truncatula as a model for analysis of legume seed filling. The study also revealed proteins presumably involved in cell division during embryogenesis (beta-tubulin and annexin). Their abundance decreased before the accumulation of the major storage protein families, which itself occurs in a specific temporal order: vicilins (14 d after pollination [DAP]), legumins (16 DAP), and convicilins (18 DAP). Furthermore, the study showed an accumulation of enzymes of carbon metabolism (e.g. sucrose synthase, starch synthase) and of proteins involved in embryonic photosynthesis (e.g. chlorophyll a/b binding), which may play a role in providing cofactors for protein/lipid synthesis or for CO2 refixation during seed filling. Correlated with the reserve deposition phase was the accumulation of proteins associated with cell expansion (actin 7 and reversibly glycosylated polypeptide) and of components of the precursor accumulating vesicles, which give rise to a trypsin inhibitor on maturation. Finally, we revealed a differential accumulation of enzymes involved in methionine metabolism (S-adenosyl-methionine synthetase and S-adenosylhomo-cysteine hydrolase) and propose a role for these enzymes in the transition from a highly active to a quiescent state during seed development.  相似文献

The major carbohydrate of the legume seed is starch, which represents up to 45% of the total seed weight. In recent years, substantial progress has been made in understanding the relationship between starch structure and functionality. However, these studies have been mainly on cereal and tuber starches. The present status of knowledge on the composition, structure, functionality, digestibility, and chemical modification of legume starches is reviewed. In addition present concepts of granule structure, gelatinization, retrogradation, and rheology are also reviewed. Future research needs in the area of legume starch chemistry are discussed.  相似文献

Abstract Germinable seed stores were estimated for 12–13 year old rehabilitation sites in the jarrah forest over two seasons (autumn and spring). Collected soils were subjected to combinations of smoking and heating treatments before the germination procedure was commenced. The mean topsoil seed reserve to a depth of 10 cm was 1938 seeds m?2. Of this total reserve, more than 80% was found in the upper 5 cm of soil. The topsoil seed reserve of these rehabilitation areas was almost seven times that of the adjacent native jarrah forest (292 seeds m?2; Ward et al. 1997). There was a seasonal effect with autumn-collected soils having significantly more germinable seeds (2723 seeds m?2) than soils collected in spring (1153 seeds m?2). More than half (53%) of the topsoil seed reserve in rehabilitated areas was composed of annual weed species, dominated by Aira caryophyllea, Centaurium erythraea and the native Levenhookia pusilla. Of the total of 70 species identified, 13 species showed significantly higher germination in smoked trays while 11 species exhibited significantly higher germination in heated trays. Species responding to the smoking treatment tended to be annuals while those responding to the heating treatment were typically legume shrub species. Examination of the topsoil seed reserve and the vegetation present in these areas showed that while the species composition was similar between the seed store and the vegetation, there was a large difference in densities, with species occurring at much higher densities in the topsoil than in the vegetation. However, the rankings of species were significantly correlated between the topsoil seed reserve and the vegetation present at the site. The implications of these results to prescribed burning of these rehabilitated areas is discussed.  相似文献

The low sulfur amino acid content of legume seeds restricts their nutritive value for animals. We have investigated the limitations to the accumulation of sulfur amino acids in the storage proteins of narrow leaf lupin (Lupinus angustifolius) seeds. Variation in sulfur supply to lupin plants affected the sulfur amino acid accumulation in the mature seed. However, when sulfur was in abundant supply, it accumulated to a large extent in oxidized form, rather than reduced form, in the seeds. At all but severely limiting sulfur supply, addition of a transgenic (Tg) sink for organic sulfur resulted in an increase in seed sulfur amino acid content. We hypothesize that demand, or sink strength for organic sulfur, which is itself responsive to environmental sulfur supply, was the first limit to the methionine (Met) and cysteine (Cys) content of wild-type lupin seed protein under most growing conditions. In Tg, soil-grown seeds expressing a foreign Met- and Cys-rich protein, decreased pools of free Met, free Cys, and glutathione indicated that the rate of synthesis of sulfur amino acids in the cotyledon had become limiting. Homeostatic mechanisms similar to those mediating the responses of plants to environmental sulfur stress resulted in an adjustment of endogenous protein composition in Tg seeds, even when grown at adequate sulfur supply. Uptake of sulfur by lupin cotyledons, as indicated by total seed sulfur at maturity, responded positively to increased sulfur supply, but not to increased demand in the Tg seeds.  相似文献

Nitrogen (N) accumulation in legumes is one of the main determinants of crop yield. Although N accumulation from symbiotic nitrogen fixation or N absorption from the soil has been widely investigated, there is no clear consensus on timing of the beginning of N accumulation and the termination of N accumulation and the physiological events that may be associated with these two events. The analyses conducted in this study aimed at identifying the determinant of N accumulation in two grain legume species. Nitrogen accumulation dynamics and mass accumulation and development stages were recorded in the field for several genotypes of common bean (Phaseolus vulgaris) and faba bean (Vicia faba) under different growing conditions. This study showed that during the vegetative stages, N accumulation rate was correlated with mass accumulation rate. However, the maximum accumulation of N did not correspond to the time of the maximum mass accumulation. In fact, for both species, N accumulation was found to persist in seed growth. This challenges a common hypothesis that seed growth causes a decrease in N accumulation because of a shift of the photosynthate supply to support the seed growth. Even more surprising was the shift of the active accumulation of N in faba bean to late in the growing season as compared with common bean. N accumulation by faba bean only was initiated at high rates very late in vegetative growth and persisted at high rates well into seed fill.  相似文献

The structure, location in the seed and distribution of the storage protein of legume seeds are described. Methods which have been employed for the extraction, purification and characterisation of seed globulins are reviewed in relation to modern biochemical practice. The physical, chemical and immunological characteristics of the classical legumin and vicilin preparations from Pisum sativum are summarised and the distributions of proteins with sedimentation coefficients and/or immunological determinants similar to those of legumin and vicilin, are tabulated. High-resolution 13C MAS NMR spectroscopy was used to profile a range of primary and secondary metabolites in vivo in intact whole seeds of eight different conifer species native to North America, including six of the Pinaceae family and two of the Cupressaceae family. In vivo 13C NMR provided information on the total seed oil content and fatty acid composition of the major storage lipids in a non-destructive manner. In addition, a number of monoterpenes were identified in the 13C NMR spectra of conifer seeds containing oleoresin; these compounds showed marked variability in individual seeds of Pacific silver fir within the same seed lot. In imbibed conifer seeds, the 13C NMR spectra showed the presence of considerable amounts of dissolved sucrose presumed to play a protective role in the desiccation-tolerance of seeds. The free amino acids arginine and asparagine, generated as a result of storage protein mobilization, were detected in vivo during seed germination and early seedling growth. The potential for NMR to profile metabolites in a non-destructive manner in single conifer seeds and seed populations is discussed. It is a powerful tool to evaluate seed quality because of its ability to assess reserve accumulation during seed development or at seed maturity; it can also be used to monitor reserve mobilization, which is critical for seedling emergence.  相似文献

Seed vigour is important for successful establishment and high yield, especially under suboptimal environmental conditions. In legumes, raffinose oligosaccharide family (RFO) sugars have been proposed as an easily available energy reserve for seedling establishment. In this study, we investigated whether the composition or amount of soluble sugars (sucrose and RFO) is part of the genetic determinants of seed vigour of Medicago truncatula using two recombinant inbred line (RIL) populations. Quantitative trait loci (QTL) mapping for germination rate, hypocotyl and radicle growth under water deficit and nutritional stress, seed weight and soluble sugar content was performed using RIL populations LR1 and LR4. Seven of the 12 chromosomal regions containing QTL for germination rate or post-germinative radicle growth under optimal or stress conditions co-located with Suc/RFO QTL. A significant negative correlation was also found between seed vigour traits and Suc/RFO. In addition, one QTL that explained 80% of the variation in the ratio stachyose/verbascose co-located with a stachyose synthase gene whose expression profile in the parental lines could explain the variation in oligosaccharide composition. The correlation and co-location of Suc/RFO ratio with germination and radicle growth QTL suggest that an increased Suc/RFO ratio in seeds of M. truncatula might negatively affect seed vigour.  相似文献

Seed physiology of wild species has not been studied as deeply as that of domesticated crop species. Trichocline catharinensis (Asteraceae) is an endemic wildflower species from the high-altitude fields of southern Brazil. This species is of interest as a source of genes to improve cultivated Asteraceae because of its ornamental features, disease resistance and ability to tolerate drought and poor soil conditions. We studied the effects of abscisic acid (ABA) and gibberellic acid (GA3) and their inhibitors, fluridone (FLU) and paclobutrazol (PAC), on seed germination. We individually assessed ultrastructural changes and differential protein accumulation. The principal component analysis explained 69.66% of differential accumulation for 32 proteins at phase II of seed germination in response to hormone and inhibitor treatment. GA3-imbibed seed germination (98.75%) resulted in increased protein accumulation to meet energy demand, redox regulation, and reserve metabolism activation. FLU-imbibed seeds showed a higher germination speed index as a consequence of metabolism activation. ABA-imbibed seeds (58.75%) showed osmotolerance and flattened cells in the hypocotyl-radicular axis, suggesting that ABA inhibits cell expansion. PAC-imbibed seeds remained at phase II for 300 h, and germination was suppressed (7.5%) because of the increased signaling proteins and halted reserve mobilization. Therefore, our findings provide insight into the behavior of Asteraceae non-dormant seed germination, which broadens our knowledge of seed germination in a wild and endemic plant species from a threatened ecosystem.  相似文献

Plant regenerations were achieved from tissue cultures of 31 species of 15 genus of legume plants on A and B media. Various factors including medium composition (major elements, minor element:s, organic components and phytohormones), seed germination rate, illumination and temperature conditions were tested for their effects on callus differentiation in tissue cultures. Combinations of major elements of A or B medium, minor elements and organic components of B5 medium, illumination of 1500–2500 lx, and temperature of 18–27℃ were found suitable for callus differentiation of most legume plants. The calli induced from freshly collected seeds had higher differentiation capacity than from seeds sored for three years. Four types(a,b,c and d) of callus were morphologically distinguished during the differentiation in legume plant tissue cultures. Generally, calli from plants of the same genus belonged to the same type.  相似文献

The seeds of Spartium junceum contained a large quantity of lectin-like protein that did not appear to be either a hemagglutinin or active lectin. The cross-reactive material (CRM), like most legume seed lectins, was a tetrameric glycoprotein of about 130,000 Mr. The singlesized subunits of about 33,000 Mr were not covalently associated. The amino acid composition was typical of legume lectins and was rich in hydroxy-amino acids and poor in sulfur-containing amino acids. The Spartium CRM contained about 3.5% covalently associated carbohydrate, most likely of the high-mannose type, since the CRM was precipitated by concanavalin A. The CRM was localized by electron-microscopic immunocytochemistry and found to be exclusively in protein-filled vacuoles (protein bodies). Because this protein was so similar immunologically, structurally, and in its physiology, to classic legume seed lectins, it is most likely a lectin homolog. Similar seed lectin CRMs appear to be both common and widespread in the Leguminosae.  相似文献

Genes for vicilin, a component of legume seed storage proteins, have been identified in the cultivated lentil (Lens culinaris ssp. culinaris) and in wild species of the genus Lens. Five different types of vicilin sequences (designated A-E) have been identified in each lentil individual. The different types of sequences, and some possible variants of them (also present in each individual) are part of the vicilin family of genes. Type D sequences have the characteristics of nonprocessed pseudogenes. Comparison of nucleotide sequences indicates that lentil vicilin sequences are similar to vicilin sequences of other legume species, in particular to those of the tribe Vicieae, in which the genes Lens is included. Sequence comparison and distance and parsimony trees indicated that two groups or subfamilies of sequences, including, respectively, types A, B, and E (47 kDa vicilins) and types C and D (50 kDa), can be distinguished in lentil and other Vicieae species, and that in the Vicieae species there is no evidence of concerted evolution among the vicilin sequences of different gene subfamilies or sequences groups, as has been suggested for other legume species.   相似文献

The dry weight of harvested grain legume seeds is strongly related to their growth rate during the period of storage accumulation in the cotyledons, which begins approximately at the end of embryo cell division. Depodding, defoliation, shading or changes in air CO2 concentration were applied during seed filling (i.e. during the decrease in seed water concentration) to field and glasshouse-grown plants, in order to affect the source-sink ratio. The experiments involved three legume species, namely pea (Pisum sativum L.), soybean (Glycine max L. Merr.) and white lupin (Lupinus albus L.). Some treatments affected the number of abortions of less developed seeds from younger pods, but they did not significantly affect the number or the growth rate of filling seeds, demonstrating the priority of carbohydrate partitioning to filling seeds. The maximum growth rate of seeds was achieved regardless of the intra-plant competition level, and the duration of seed growth was shortened if the photosynthetic activity was not sufficient to fulfil the assimilate demand of filling seeds.  相似文献

We have isolated a water-soluble polysaccharide from the Jack bean [Canavalia ensiformis (L) DC] seed coat that was shown to be highly detrimental to larval development of the cowpea weevil (Callosobruchus maculatus) (Coleoptera: Bruchidae). Determination of the composition and structure of this polysaccharide showed that it is a galactorhamnan with an Mw of 883.0, containing 92% rhamnose and 8% galactose. The polymer is formed by a main chain of rhamnose (1to2) substituted at O-4 by galactose nonreducing end-units. Immunolocalisation by light and electron microscopy showed that this polysaccharide is localised in the innermost cell layer of the seed coat and also in cotyledon tissues in the cytoplasm space. The presence of this toxic polysaccharide in the testa of a non-host seed may have been important for the evolutionary discrimination of legume seeds by bruchids.  相似文献

Several legume seed proteins that are potentially allergenic, poorly digested by farm animals, and/or have undesirable functional properties, have been described. One of these is the albumin protein in pea (Pisum sativum) called PA2. A naturally occurring mutant line that lacks PA2 has been exploited in studies to determine the biological function of this nonstorage protein in seed development. The mutant, which has a small seed, a tall plant phenotype, and lacks most of the PA2-encoding genes, has been crossed with a standard cultivar, 'Birte,' which contains PA2 to give rise to a recombinant inbred (RI) population. An F(3) line carrying the mutation and having a short plant phenotype has been used to generate backcross (BC) lines with 'Birte.' Despite having a lower albumin content, seeds from the mutant parent and RI lines lacking PA2 have an equivalent or higher seed nitrogen content. Metabolite profiling of seeds revealed major differences in amino acid composition and polyamine content in the two parent lines. This was investigated further in BC lines, where the effects of differences in seed size and plant height between the two parents were eliminated. Here, differences in polyamine synthesis were maintained as was a difference in total seed protein between the BC line lacking PA2 and 'Birte.' Analysis of enzyme activities in the pathways of polyamine synthesis revealed that the differences in spermidine content were attributable to changes in the overall activities of spermidine synthase and arginine decarboxylase. Although the genes encoding spermidine synthase and PA2 both localized to the pea linkage group I, the two loci were shown not to be closely linked and to have recombined in the BC lines. A distinct locus on linkage group III contains a gene that is related to PA2 but expressed predominantly in flowers. The results provide evidence for a role of PA2 in regulating polyamine metabolism, which has important functions in development, metabolism, and stress responses in plants.  相似文献
