A cDNA coding for a fungal amino acid transporter ( AmAAP1 ) was identified from Amanita muscaria ectomycorrhizas. The transporter gene was expressed at a basal level under all conditions investigated, but its expression was enhanced 10-fold in the absence of a N source utilized by the fungus. Nitrate was not a suitable N source for A. muscaria and resulted in maximal AmAAP1 expression. The expression of AmAAP1 in a yeast mutant revealed its function as a high-affinity amino acid transporter with a broad substrate spectrum. AmAAP1 takes up all investigated amino acids with K m values between 22 μM for histidine and up to 100 μM for proline. Gene expression and amino acid uptake data together indicate two main functions for AmAAP1: uptake of amino acids from soil for fungal nutrition, and prevention of an amino acid loss by hyphal leakage in the absence of a suitable N source.  相似文献

以外生菌根菌鹅膏菌属三个种Amanita muscaria,A.pseudoporphyria和A.fritillaria为研究材料,以生长速率为评价指标,对其最适生长温度、pH值、光照、培养基、C及N源的利用等基本培养条件及所产肽类毒素进行了比较研究。研究结果表明,三种菌株最适生长温度有差异,A.pseudoporphyria和A.fritillaria的最适温度为28℃,A.muscaria的最适温度为22℃;A.muscaria菌丝体生长的pH值范围为5-7,另外两个菌株的pH值范围为3-6;24h光照、12h光暗交替和24h黑暗对鹅膏菌的生长速率影响不大;SPDM培养基和MMN培养基都适合三种菌株的生长,但对于A.muscaria来说PDM培养基更适合其生长。鹅膏菌能够利用比较广泛的C、N源,但三个种在利用的C、N源种类上有一定的差别。通过抑芽法实验和HPLC分析分别表明三种鹅膏菌所含肽类毒素在种类和含量上有所不同,但都对绿豆发芽有一定的抑制作用。A.pseudoporphyria和A.fritillaria菌丝体中α-amanitin的含量分别为35.56μg/gDCW(drycellweight细胞干重)和26.02μg/gDCW,不含有phalloidin和β-amanitin;A.muscaria菌丝体中没有检测到α-amanitin、β-amanitin和phalloidin。结果表明供试的三种鹅膏菌在基本培养条件及所产肽类毒素方面存在种水平上的差异。  相似文献

The role of apoplastic invertase (β-d -fructofuranoside — fructohydrolase, EC 3.2.1.26) of the host Picea abies for carbohydrate uptake and growth of two of its natural ectomycorrhiza partners was studied. For that purpose, hyphae of Amanita muscaria (Pers. ex Fries) Hock. and Hebeloma crustuliniforme (Bull. ex Fries) Quell., as well as roots and suspension cultured cells of Picea abies (L.) Karst. were used. Apoplastic invertase activity was demonstrated on roots and suspension cultured cells of spruce (in the latter case with 21.7 nkat (g fresh weight)?1). Inhibition of the root cell wall invertase activity (pH optimum 4.5) by increasing the apoplastic pH allowed determination of the permanent release of sucrose from the root. However, under in vivo conditions at a lower cell wall pH the hydrolysation products glucose and fructose were predominantly found. In contrast to spruce cells and certain fungi, such as Saccharomyces (Novick et al., 1981) or Phycomyces (Ruiz-Herrera et al., 1989) invertase activity of the mycorrhizal fungi Hebeloma and Amanita was negligibly low. Furthermore, sucrose could not be consumed by Amanita and Hebeloma. As a consequence, cultures of these mycorrhizal fungi starved when kept on media with sucrose as sole carbohydrate source. But addition of invertase initiated hyphal growth immediately. Studies on carbohydrate uptake of host and fungal cells confirmed that the monosaccharides glucose and fructose were readily incorporated by spruce and fungal cells, with a clear preference for glucose. From these results it is suggested that apoplastic invertase activity of the host Picea abies is a precondition for the utilization of sucrose by the studied mycorrhizal fungi during the nutritional interaction of the symbiotic partners.  相似文献

In the soils of mature forests, nitrogen availability is mainly the result of litter decomposition. Thus protein degradation is of major interest for nutrition. Two aspartic proteases were excreted in a pH‐dependent manner by the ectomycorrhizal fungus Amanita muscaria grown in liquid culture. AmProt1 with a molecular mass of approximately 45 kDa was mainly present at pH values up to 5·4, whereas the excretion of AmProt2 with a molecular mass of about 90 kDa was only detectable at pH values between pH 5·4 and 6·3. In addition, the pH optima of both enzymes differed significantly. AmProt1 had a narrow pH optimum around 3, whereas AmProt2 had a broad pH optimum between 3 and 5·5 and a higher affinity to the substrate methylcasein. One cDNA‐clone (AmProt1*) that presumably encodes AmProt1 was identified. Like AmProt1, this cDNA was expressed in a pH‐dependent manner. In addition, carbohydrate and to a lesser extent nitrogen depletion significantly enhanced AmProt1* expression. In fully developed Populus hyb./A. muscaria ectomycorrhizas the expression of AmProt1* was significantly higher in hyphae of the Hartig net compared with those of the fungal sheath. The role of AmProt1 and AmProt2 for fungal physiology and competitiveness is discussed.  相似文献

High performance liquid chromatography analysis of immunoaffinity-purified extracts of mycelia of Amanita muscaria, and the Amaranthus bioassay of the eluted fractions, revealed the following seven cytokinins: zeatin, zeatin riboside, zeatin N-9-glucoside, dihydrozeatin, dihydrozeatin riboside, isopentenyl adenine, and isopentenyl adenosine. The decreased growth of aluminum-treated mycelia correlated with a 35% decrease in the total amount of the cytokinins. Among individual cytokinins, zeatin was the most affected, exhibiting a reduction of about 90%. Arbuscular mycorrhizal (AM) fungi mediate interactions between plants and soils, and are important where nutrient or metal concentrations limit plant growth. Variation in fungal response to edaphic conditions may influence the effectiveness of the plant-mycorrhizal association in some soil environments. Andropogon virginicus (broomsedge) colonizes disturbed sites in the eastern United States, including acidic mine soils where aluminum (Al) is phytotoxic, and Al resistance in broomsedge has been associated with colonization by the AM fungus Glomus clarum. In the present study, inter- and intra-specific variation to confer Al resistance to broomsedge was assessed among selected species of AM fungi. Broomsedge seeds were grown in sand culture inoculated with one of five isolates of three species of fungi (G. clarum, Acaulospora morrowiae, and Scutellospora heterogama). Plants were exposed to 0 or 400 µM Al in nutrient solution and harvested after 4 or 9 weeks of growth. Mean infection percentage, plant biomass, and plant tissue Al and phosphorus (P) concentrations were measured. G. clarum conferred the greatest Al resistance to broomsedge, with the lowest variability among isolates for colonization and growth inhibition by Al [tolerance indices (TI) between 22.4 and 92.7%]. Broomsedge plants colonized by A. morrowiae were consistently the most sensitive to Al, with little variation among isolates (TI between 1.6 and 12.1%). Al resistance by S. heterogama isolates was intermediate and wide-ranging (TI between 3.9 and 40.0%). Across all AM fungal isolates, resistance was associated with high rates of colonization and low tissue Al concentrations of broomsedge plants. The functional diversity in Al resistance displayed by these AM fungi reflect variation in acclimation mechanisms operating in the mycorrhizal symbiosis under environmental stress.  相似文献

A cell-wall fraction of the mycorrhizal fungus Amanita muscaria increased the chitinase activity in suspension-cultured cells of spruce (Picea abies (L.) Karst.) which is a frequent host of Amanita muscaria in nature. Chitinase activity was also increased in roots of spruce trees upon incubation with the fungal elicitor. Non-induced levels of chitinase activity in spruce were higher in suspension cells than in roots whereas the elicitorinduced increase of chitinase activity was higher in roots. Treatment of cells with hormones (auxins and cytokinin) resulted in a severalfold depression of enzyme activity. However, the chitinase activity of hormone-treated as well as hormone-free cells showed an elicitor-induced increase. Suspension cells of spruce secreted a large amount of enzyme into the medium. It is postulated that chitinases released from the host cells in an ectomycorrhizal system partly degrade the fungal cell walls, thus possibly facilitating the exchange of metabolites between the symbionts.  相似文献

We describe the cross‐genomic isolation of 13 single nucleotide polymorphisms (SNPs) and one variable microsatellite from five loci for the death cap mushroom Amanita phalloides. Microsatellite repeats were identified by searching the partial Amanita bisporigera genome. Flanking primers were designed for 25 of these microsatellite loci and tested for cross‐amplification in A. phalloides. One locus contained an interrupted, compound microsatellite, and four loci contained one to six SNPs. These results demonstrate the usefulness of even an incomplete genome to identify molecular markers for population studies in nonmodel organisms.  相似文献

Amanita muscaria sensu lato has a wide geographic distribution, occurring in Europe, Asia, Africa, Australia, New Zealand, and North, Central and South America. Previous phylogenetic work by others indicates three geographic clades (i.e. 'Eurasian', 'Eurasian-alpine' and 'North American' groups) within A. muscaria. However, the historical dispersal patterns of A. muscaria remained unclear. In our project, we collected specimens from arctic, boreal and humid temperate regions in Alaska, and generated DNA sequence data from the protein-coding beta-tubulin gene and the internal transcribed spacer (ITS) and large subunit (LSU) regions of the ribosomal DNA repeat. Homologous sequences from additional A. muscaria isolates were downloaded from GenBank. We conducted phylogenetic and nested clade analyses (NCA) to reveal the phylogeographic history of the species complex. Although phylogenetic analyses confirmed the existence of the three above-mentioned clades, representatives of all three groups were found to occur sympatrically in Alaska, suggesting that they represent cryptic phylogenetic species with partially overlapping geographic distributions rather than being allopatric populations. All phylogenetic species share at least two morphological varieties with other species, suggesting ancestral polymorphism in pileus and wart colour pre-dating their speciations. The ancestral population of A. muscaria likely evolved in the Siberian-Beringian region and underwent fragmentation as inferred from NCA and the coalescent analyses. The data suggest that these populations later evolved into species, expanded their range in North America and Eurasia. In addition to range expansions, populations of all three species remained in Beringia and adapted to the cooling climate.  相似文献

Organic phosphorus sources make up a large fraction of the total P in some soils. Vesicular–arbuscular mycorrhizal fungi provide a large surface area for the absorption of inorganic P. The question of whether or not they have direct access to organic P by producing extracellular phosphatases has hitherto been controversial because experiments had not been performed in the absence of other soil microorganisms. We used a split-dish in vitro carrot mycorrhiza system free from contaminating microorganisms. The extraradical hyphae of Glomus intraradices hydrolysed both 5-bromo-4-chloro-3-indolyl phosphate and phenolphthalein diphosphate. Moreover, they transferred significantly more P to roots when they had access to inositol hexaphosphoric acid (phytate) than when they did not. Thus we show unequivocally that extraradical hyphae of G. intraradices can hydrolyse organic P, and, further, that the resultant inorganic P can be taken up and transported to host roots.  相似文献

随着对湿地重要性认识的增加,湿地植物的丛枝菌根(arbuscular mycorrhiza,AM)结构研究受到了关注。研究表明,AM结构在多种湿地类型和湿地植被类型中存在,但AM形成受植被生长地水文状况的影响;从湿地植物上的AM真菌种类看,球囊霉属(Glomus)是湿地环境的常有属。同陆生植物相比,有关湿地植物AM结构的研究尽管已取得了一些进展,但还有很大差距,尤其在我国还几乎处于空白状态。鉴此,从形成AM的湿地植被类型、湿地环境中的AM真菌种类、AM对湿地植物生长的影响,以及影响湿地植物AM形成的因素等方面进行了详细阐述,并对今后的研究方向做了展望,以期为我国开展湿地植物AM结构研究提供参考。  相似文献

Two generations of two aphid species (Myzus ascalonicus and M. persicae) were reared on Plantago lanceolata plants, with and without root colonization by the arbuscular mycorrhizal fungus, Glomus intraradices. Life history traits of the aphids measured were nymphal development time, teneral adult weight, growth rate, total fecundity, adult longevity and duration of post-reproductive life. For both aphids in both generations, mycorrhizal colonization increased aphid weight and fecundity, while other traits were unaffected. The increases were consistent between generations. In a second experiment, M. persicae was reared on plants with and without the fungus, under varying N and P regimes. The results of N addition were inconclusive because there was high aphid mortality. However, under P supplementation, positive effects of the mycorrhiza on aphid growth were seen at low and medium P levels, while at high P levels these effects disappeared. The positive effects of mycorrhizal colonization reported here are contrary to the majority of previous studies with chewing insects, which have reported negative effects.
