Oxygen uptake and tetrazolium reduction occurred at higher rates in discs from potato tubers (Solanum tuberosum L.) stored at 0° than in discs from tubers stored at 12.8°. Tetrazolium reduction was at a higher rate in mitochondrial fractions from tubers stored at 0° than in mitochondrial fractions from tubers stored at 12.8°. These physiological activities were more resistant to hypertonic KCl treatments in tissue and mitochondrial fractions from tubers stored at 0° than in tissue and mitochondrial fractions from tubers stored at 12.8°. Inhibition of O2 uptake and tetrazolium reduction progressively increased with increasing concentrations of KCl for tissue and mitochondrial fractions from tubers stored at 0 and 12.8°, but inhibition was more severe and occurred at lower concentrations of KCl for the material from tubers stored at 12.8°. Tissue from tubers stored at 0° was at the same time more sensitive to hypotonic solutions and more resistant to hypertonic solutions than corresponding tissue from tubers stored at 12.8°. Adaptive changes brought on in the tubers by the stress of cold storage were demonstrated in the discs and mitochondrial fractions prepared from cold-stored tubers.  相似文献

The influence of temperature on total glycoalkaloid (TGA) synthesis in tubers exposed to light (250 jumol m“2 s”2 PAR, Photosynthetically Active Radiation) or dark environments for 96 h was examined in three potato cultivars. Following 96 h light or dark the tubers were stored without light at 5°C or 24°C and TGA concentrations monitored over the subsequent 30 and 90 days. Exposure to light and cultivar were found to be major factors influencing TGA concentrations; temperature had no significant effect. TGA content in illuminated tubers of cvs ‘Pentland Hawk’ and ‘Kerrs Pink’ were significantly higher (P < 0.01) compared with tubers placed in the dark. TGA concentrations in cv. ‘Desiree’ increased significantly only following exposure to light at low temperatures (P < 0.05). Removal of tubers from storage at 5°C and immediate illumination at 24°C altered the ratio of glycoalkaloids in cvs ‘Pentland Hawk’ and ‘Kerrs Pink’. Regardless of cultivar and storage temperature TGA concentrations were higher at the end of the storage period compared with initial TGA concentrations. During storage TGA concentrations fluctuated widely and gradual accumulation of glycoalkaloids with time was rarely demonstrated except in cv. ‘Desiree’. Tubers stored at 24°C accumulated higher TGA concentrations than those stored at 5°C in cv. ‘Kerrs Pink’ but not in cvs ‘Pentland Hawk’ and ‘Desiree’. Tubers of cv. ‘Kerrs Pink’ exposed to light prior to storage accumulated glycoalkaloids more rapidly than unexposed tubers during storage at 24°C and occasionally at 5°C. Light enhanced glycoalkaloids are not degraded over time.  相似文献

The respiration of potato tubers (Solanum tuberosum var. Russet Burbank) which have been kept at room temperature for 10 days is stimulated upon subsequent treatment with C2H4 (10 microliters per liter) and O2. The respiratory rise reaches a peak in 24 to 30 hours and thereafter declines. Coincident with the rise in tuber respiration is an increase in the respiratory rates of fresh slices and isolated mitochondria. Slices and mitochondria from C2H4- and O2-treated tubers also display substantial resistance to CN, and the resistant respiration is inhibited by hydroxamates.

The longer the tubers are stored after harvest, the less effective is C2H4 in causing CN resistance in slices and mitochondria from treated tubers. Addition of 10% CO2 to the C2H4-O2 mixture, however, causes extensive CN resistance to develop, even in slices and mitochondria from old tubers. The results show that C2H4, O2, and CO2 act synergistically to induce alternative path development in potatoes.

The changes in the components of the ascorbateglutathione systemduring the storage of potato (Solanum tuberosum L. cv. Spunta)tubers for 40 weeks at both 3C and 9C were studied in relationto lipid peroxidation. The level of malondialdehyde was foundto be higher at 3C than at 9C throughout storage. Thus, lipidperoxidation, which is the main cause of membrane deterioration,was accelerated at the lower temperature. Catalase activityincreased throughout storage independently of temperature. Theascorbate content of tubers decreased during storage both at3C and at 9C, as in other ageing processes. However, ascorbateperoxidase activity reached a maximum after about 8 weeks ofstorage, then declined at 9C, but held a higher level at 3C.The dehydroascorbic content also reached a maximum after about8 weeks and was significantly higher in tubers stored at 3C.These findings indicate a greater utilization of ascorbate byascorbate peroxidase at the lower temperature. Ascorbate freeradical reductase, dehydroascorbate reductase and glutathionereductase, the enzymes involved in the regeneration of ascorbate,were not affected by temperature and remained quite unchangedthroughout storage. It can be concluded that the ascorbate systemis involved in the scavenging of the free-radicals responsiblefor lipid peroxidation in stored potato tubers, at least atlow temperatures and in the first period of storage. Photosynthesis and cell composition of Porphyraleucosticta discs grown at low (< 0.0001% in air), current (control) and high (1% CO2 in air)inorganic carbon (Ci) concentrations were analyzed. Carbohydrate content in discs grown at high Ci increased (15.1 mg g-1 FW) with respect to the control (6.4 mg g FW-1), whereas soluble protein content decreased to one-third (5.6 to2.1 mg g-1 FW). Carbohydrate content was unaffected and soluble protein slightly increased in discs grown at low Ci. As a consequence of these changes, a lower C/N molar ratio (8.6) was found in the discs grown at low compared to high Ci(12.4). Nitrate reductase activity increased at high Ci from 0.3 ± 0.2 to 1.7 ± 0.4 μmolNO2 - g-1 FW h-1indicating that reduction and assimilation of nitrate were uncoupled. The response of photosynthesis to increasing irradiance, estimated from O2evolution vs. irradiance curves, was affected by the treatments. Maximum quantum yield (Φ O2°) and effective quantum yield (Φ O2) at 150 μmol photon m-2s-1 decreased by 20% and 50%, respectively, at low Ci. These differences could be due to changes in photosynthetic electron flow between PSII and PSI. Treatments also produced changes in maximal (Fv/Fm) and effective (ΔF/Fm′)quantum yield for photosystem II charge separation. Potato titbers infested with Phoma exigua var. foveata were uniformly wounded and sprayed or dipped in fungicide suspensions either immediately or after periods of up to 21 days' storage at 5, 10, 15 or 20 °C. Tubers were then stored at 5 °C and gangrene assessed after 12 wk. Incidence of gangrene on untreated tubers was progressively decreased by increasing the length of storage at 15 or 20° (curing) but was not affected by 3 days' storage at any temperature. Fungicide treatment immediately after wounding gave best control of the disease; treatment after 3 days' delay was less effective and after 14 or 21 days was usually ineffective. Gangrene was decreased by fungicides more on tubers stored for 3 or 7 days at 5 °C than at higher temperatures. Control of gangrene by curing or fungicides diminished when the amount of inoculum on tubers was increased. Increasing the amount of fungicide applied improved control and fungicides were more effective in decreasing gangrene on cut and crush wounds than on cut wounds. At the arbitrary concentrations used in these experiments imazalil gave better disease control than thiabendazole, prochloraz, carbendazim plus quinolin 8-ol or triadimefon.  相似文献

A large increase in the activity of hydroxycinnamyl CoA:quinate hydroxycinnamyl transferase (CQT) occurred in potatoes stored at 0 and 2° and such an increase was prevented by storage at either 5 or 10°. The increase was most rapid in potatoes stored at 0° where it reached a maximum after 28 days and then declined slowly during storage for up to 6 months. Accompanying these changes in CQT were transitory increases in p-coumarate CoA ligase and PAL which occured during the first few weeks of storage at 0° and during this period there was nearly a two fold increase in the chlorogenic acid content of the tissue. The increase in chlorogenic acid did not occur at 10° when the increases in PAL, ligase and CQT were also prevented. The increase in CQT was reversed when tubers stored at 0° for 14 days were returned to 10° and this warming up period prevented further increase in CQT on return to 0°. The increase in CQT at 0° was prevented if the air in the storageatmosphere was replaced by N2, 1 % O2 or 10–15% CO2. Similar increases in CQT, ligase and chlorogenic acid occurred in sweet potatoes stored at 7.5° but were prevented by storage at 15°. The role of PAL, ligase and CQT in the control of chlorogenic acid accumulation in these commodities and the significance of changes in their activities in relation to physiological changes at low temperatures are discussed.  相似文献

A sharp temperature optimum is observed at 21.5°C when the incorporation of [14C]sucrose into starch is measured with discs cut from developing tubers of potato (Solanum tuberosum L. cv Desirée). By contrast, increasing temperatures over the range 9 to 31°C only enhance release of 14C to respiratory CO2 and incorporation of 14C into the ethanolsoluble fraction. By comparison, starch synthesis in discs from developing corms of cocoyam (Colocasia esculenta L. Schott) is increased by raising the temperature from 15 to 35°C. The significance of a relatively low temperature optimum for starch synthesis in potato is discussed in relation to the yield limitations imposed by continuously high soil temperatures. Amyloplasts isolated from protoplasts prepared from developing potato tubers contain activities of alkaline pyrophosphatase, NAD-dependent glyceraldehyde-3-phosphate dehydrogenase, fructose-1,6-bisphosphatase, and phosphoglucomutase in addition to ADP-glucose-pyrophosphorylase, starch phosphorylase and starch synthase. Cell-free amyloplasts released by thinly slicing developing potato tubers synthesize starch from [14C]triose-phosphate generated from [14C]fructose-1,6-bisphosphate in the reaction medium. This starch synthesis is inhibited by addition of 10 millimolar inorganic phosphate and requires amyloplast integrity, suggesting the operation of a triose-phosphate/inorganic phosphate exchange carrier at the amyloplast membrane. The temperature optimum at 21.5°C observed with tissue discs is not observed with amyloplasts.  相似文献

Storage temperatures greater than 4 °C (that is, heat-unit accumulation) increase respiration and accelerate physiological aging of seed tubers. The degree of apical dominance is a good indicator of physiological age (PAGE). As seed age advances, apical dominance decreases, resulting in more stems, greater tuber set, and shifts in tuber size distribution. Herein we provide evidence that tuber respiration rate may constitute the “pacemaker” of aging. Tubers exposed to a brief high-temperature age-priming treatment initially in storage, followed by holding at 4 °C for the remainder of a 190–200-day storage period, maintained a higher basal metabolic (respiration) rate throughout storage compared with tubers stored the entire season at 4 °C. Tubers thus “remembered” the age-priming treatment as reflected by their elevated respiration rate. Moreover, reducing the respiration rate of age-primed seed by subsequently storing it at 3.5 % O2 (4 °C) until planting significantly attenuated the effects of the aging treatment on apical dominance, tuber set, and size distribution. The effect of the age-priming treatment on the magnitude of the respiratory response was the same whether given at the beginning or toward the end of storage. However, moving the age-priming treatment progressively later in the storage season effectively decreased its impact on plant growth and development. These results underscore the importance of time in the aging process. Exposure of seed to a high-temperature age-priming treatment at the beginning or end of storage elevated respiration (the pacemaker) to the same extent; however, the timing of these treatments resulted in vastly different physiological ages. The longer the respiration rate of tubers remains at an elevated level, the greater their PAGE at planting. Thus, an accurate but impractical measure of PAGE may be the respiratory output from vine kill to subsequent planting. Respiration appears to be the pacemaker of PAGE and production, and storage conditions that affect respiration may “set the clock speed” that will ultimately determine the PAGE at planting.  相似文献

The relationships of potato (Solanum tuberosum L.) tuber membrane permeability and membrane lipid composition to sugar accumulation were examined. Tubers from four potato cultivars were stored for 40 weeks at 3°C and 9°C. Rates of tuber membrane electrolyte leakage, total fatty acid composition, free fatty acid composition, and sugar content were measured throughout the storage period. Storage of tubers at 3°C caused dramatic increases in total fatty acid unsaturation, membrane permeability, and sugar content compared to tubers stored at 9°C. Cultivars with higher levels of fatty acid unsaturation had lower rates of membrane electrolyte leakage and lower sugar contents. We propose that high initial levels or high induced levels of membrane lipid unsaturation mitigate increases in tuber membrane permeability during storage, thus positively influencing the processing quality of stored potato tubers.  相似文献

To clarify the effects of storage temperature on potato components and acrylamide in chips, tubers from five cultivars were stored at various temperatures (2, 6, 8, 10, and 18 degrees C) for 18 weeks, and the contents of sugars, free amino acids in tubers, and acrylamide in chips after frying were analyzed. At temperatures lower than 8 degrees C, the contents of reducing sugars increased markedly in all cultivars, with similar increases in the acrylamide level and dark brown chip color. Free amino acids showed little change at the storage temperatures tested and varied within certain ranges characteristic of each cultivar. The contents of reducing sugars correlated well with the acrylamide level when the fructose/asparagine molar ratio in the tubers was <2. When the fructose/asparagine ratio was >2 by low-temperature storage, the asparagine content, rather than the reducing sugar content, was found to be the limiting factor for acrylamide formation.  相似文献

The effect of low concentrations of O2 (1%) with or without the application of exogenous ethylene (10 l/l) on the production of endogenous ethylene, the activity of polygalacturonase (PG), and the ripening of tomato fruits during storage for three weeks at 20°C and four weeks at 10°C, followed by one week under ambient conditions (25°C) was studied. The internal ethylene concentration in the fruits stored under low O2 at 10 or 20°C was low during storage and increased only when fruits were transferred to ambient conditions. The application of exogenous ethylene to fruits stored under low O2 at 10 or 20°C did not induce autocatalytic ethylene synthesis. By contrast, the internal ethylene concentration of fruits stored in air was high at 20°C and somewhat lower at 10°C. Under low O2 conditions, PG activity was low and the fruits remained firm and green throughout storage, whereas, during storage in the air, PG activity increased and the fruits softened and developed their characteristic red color.  相似文献

The acceptability of potatoes for processing chips and French fries is largely dependent on the color of the finished product. Most potato cultivars and varieties stored at temperatures below 9–10 °C are subjected to low temperature sweetening (LTS) which result in the production of bitter-tasting, dark colored chips and French fries which are unacceptable to consumers. However, storing tubers at low temperatures (i.e., <10 °C) has many advantages such as lowered weight loss during storage, natural control of sprouting, and reduction/elimination of chemical sprout inhibitors. Our earlier research results on LTS suggested a role for pyruvate decarboxylase (PDC) in LTS-tolerance. In the present study, the role of PDC was examined whereby the potato variety Snowden was transformed with Arabidopsis cold-inducible pyruvate decarboxylase gene 1 (AtPDC1) under the control of promoter rd29A. Two transgenic plants were selected and storage studies were conducted on tubers harvested from one of the transgenic lines grown under green house conditions. Transgenic tubers showed higher Agtron chip color score indicating lighter chip and lower reducing sugar and sucrose concentrations compared to the untransformed tubers during the storage periods studied at 12 °C and 5 °C. These results suggest that overexpression of pyruvate decarboxylase gene resulted in low temperature sweetening tolerance in the transgenic Snowden.  相似文献

Germination of Beauveria bassiana (Bb) and Metarhizium anisopliae (Ma) conidia determined from a fast-rehydration (FR) protocol were compared to those obtained when dry conidia were subjected to slow rehydration (SR) by holding under high humidity conditions prior to aqueous suspension. Differences in viability estimates obtained using the FR vs. SR protocols increased markedly after conidia were exposed to various stress factors in storage (high aw, temperature, and O2 concentrations), with the SR protocol producing higher estimates of viability in all cases. After Bb conidia were stored under moist conditions for 21 days at 25 °C, the SR estimate of viability was >21% greater than the FR estimate. In jars flushed with different O2 concentrations and stored at 50 °C for 34 days, proportional differences between protocols varied, depending on water activity, from 18-44% in jars flushed with 0% O2 (100% N2) to as high as 63-93% when treated with 21-22% O2. For conidia stored over a broad range of moderate to high temperatures in the absence of O2, SR-FR differences were ?9% at 25-40 °C but 30% at 50 °C. Germination of stressed Bb and Ma conidia increased substantially when incubation time on the germination substrate was increased from 24 to 72 h, whereas germination of non-stressed conidia showed little change. Conidia debilitated by stress were characterized by hypersensitivity to lethal imbibitional damage (damage that is mitigated by slow rehydration) and slow germination. Viability protocols that may provide more reliable assessments of overall mycopesticide quality are discussed.  相似文献

Storage of mature or developing potato tubers (Solanum tuberosum “Up-to-Date” variety) at 4 C causes a reduction in the starch content and the elevation in the level of free sugars. This phenomenon is not observed when the tubers are stored at 25 C. Changes in the morphology of cells from developing or mature tubers after storage at 4 or 25 C have been followed by electron microscopy. During all stages of the tuber development the starch granules are surrounded by a membrane derived from the plastid envelope. Storage in the cold induces disintegration of this membrane. A membrane fraction isolated from starch granules of tubers stored at 4 C has a lower buoyant density, and the electrophoretic pattern of its proteins is different from that of a similar membrane fraction obtained from tubers stored at 25 C. It is suggested that the cold-induced changes in the starch and sugar content during storage of potato tubers might be correlated with damage to the membranes surrounding the starch granules and changes in their permeability to degradative enzymes and substrates.  相似文献

In this study a Brazilian granulovirus strain, PhopGV, isolated from the potato tuber moth (PTM) Phthorimaea operculella, was investigated regarding its potential for biological control and in vivo production. The relationship between mortality of P. operculella larvae and virus concentration was determined at different temperatures on potato tubers and susceptibility of P. operculella to PhopGV was also determined on potato leaves. Virulence of PhopGV to P. operculella was not affected by temperatures from 18 to 30°C. The median lethal concentration (LC50) of larvae fed on potato foliage treated with PhopGV was not higher than that verified with larvae fed on treated tubers. Optimal conditions for production of virus-infected larvae were obtained by using the virus suspensions of 41 × 105, 6.3 × 105 and 62 × 105 OBs ml−1 at 18, 24 and 30°C, which resulted in 32.0, 31.4 and 34.8% of infected larvae collected, respectively. The maximum percentage of infected larvae recovered from tubers was not affected by temperature. However, time for production of virus-infected larvae was longer at 18°C and shorter at 30°C. Persistence of PhopGV was determined on stored tubers and we observed that the virus remained effective for at least two months, causing up to 84.2% mortality of P. operculella at 1 × 107 OBs ml−1. The pathogen was also highly virulent to tomato pinworm, Tuta absoluta, inflicting high percentage of mortality, delaying larval growth and inhibiting pupation. This Brazilian PhopGV strain has potential to control PTM larvae on potato tubers at a broad range of temperature and can be produced in vivo using virus-treated tubers.  相似文献
