Abstract.  1. When first instar nymphs and adults of the grain aphid Sitobion avenae (Fabricius) (Hemiptera: Aphidiae) were maintained in long-term cultures (>6 months) at 20 °C and 10 °C, the LT50 decreased from −8 and −8.8 °C to −16.0 and −13.5 °C, respectively.
2. When aphids from the 20 °C culture were transferred to 10 °C, there was a progressive increase in cold tolerance through three successive generations. Transfer of newly moulted pre-reproductive adults reared at 10 °C for three generations back to 20 °C resulted in a rapid loss of cold hardiness in their nymphal offspring.
3. In all generations reared at 10 °C, first born nymphs were more cold hardy than those born later in the birth sequence. The LT50 of nymphs produced on the first day of reproduction in the first, second and third generations maintained at 10 °C were −14.8, −17.0 and −16.6 °C, respectively. Thereafter, nymphal cold hardiness decreased over the subsequent 14 days of reproduction in each generation at 10 °C with mean LT50 values of −10.3, −12.6 and −14.8 °C, respectively. By contrast, the cold tolerance of first born nymphs of aphids reared continuously at 20 °C did not differ in comparison with later born siblings. The LT50 of adult aphids was also unaffected by ageing.
4. The ecological relevance of these findings is discussed in relation to the overwintering survival of aphids such as S. avenae .  相似文献

The LT50 (lethal temperature) of first instar and adult stages of the peach-potato aphid Myzus persicae was lowered following long term acclimation at low temperatures.
First instars consistently showed greater cold hardiness than adult stages at each acclimation temperature, with the differential increasing as the temperature was lowered. When maintained at 5°C (the lowest acclimation regime) nymphs and adults had dLT508.3°C and 4.7°C respectively lower than those for non-acclimated individuals.
When 10°C acclimated adults were returned to 20°C, the acclimation effect was retained in full for 6 days but complete deacclimation occurred by day 10. In contrast the LT50 of their progeny increased gradually from the first day of adult deacclimation towards the level of the unacclimated control over a period of 10 days.
A change in cold hardiness was observed in first instars according to their position in the birth sequence. The LT50 of first-born nymphs (day 1 of reproduction) from 20°C parents was - 15.9°C rising to - 8.3°C by day 4 and remaining at this level until the end of the reproductive period.
The differential mortality between nymphs and adults observed in the laboratory was supported by the results of a field experiment. Adult aphids kept in clip-cages on a crop of oilseed rape showed greater mortality compared with those introduced as nymphs when the minimum temperature fell below -4°C for the first time in winter. At - 10°C mortality of aphids introduced as adults approached 100% whereas more than 50% of those introduced as nymphs were still alive at this temperature.  相似文献

During cold acclimation of potato plantlets ( Solanum commersonii Dun, PI 458317), there are two transitory increases in free ABA content corresponding to a three-fold increase on the 2nd day and a five-fold increase on the 6th day (Ryu and Li 1993). During this period, plantlets increased in cold hardiness from −5°C (killing temperature, control grown at 22/18°C, day/night) to −10°C by the 7th day of exposure to 4/2°C (day/night). This increase in free ABA was not found when cycloheximide (CHI), an inhibitor of cytoplasmic protein synthesis, was added to the culture medium 6 h before exposure to low temperatures. Plantlets treated with CHI did not acclimate to cold, maintaining a hardiness level (−5°C) similar to that of the 22/18°C-grown plantlets. When the CHI-treated plantlets were exposed to low temperatures for 3 days, transferred to CHI-free culture medium and grown at low temperatures, the plantlets showed a transitory increase in free ABA 2 days later. This increase was followed by the development of cold hardiness (−8°C). Application of CHI to the culture medium after 3 days of cold acclimation, when the first ABA peak and a partial development of cold hardiness (−8°C) had occurred, blocked the second transitory increase in free ABA and resulted in no further development of cold hardiness. These results suggest that de novo synthesis of proteins is required for these transitory increases in free ABA during cold acclimation of potato plantlets.  相似文献

The fall webworm Hyphantria cunea Drury, which was accidentally introduced to Japan in 1945, overwinters on the ground in pupal diapause. Diapause termination, as indicated by the respiration rate and the period required for adult emergence, began in March and ended in April. Cold hardiness (the ability to survive exposure to -15 degrees C) decreased linearly with diapause development from November to the following April under field conditions. Cold hardiness of diapause pupae (DP) decreased as the acclimation temperature decreased from 15 to -10 degrees C, whereas cold hardiness of non-diapause pupae (NDP) remained high as the acclimation temperature decreased from 5 to -5 degrees C. However, H. cunea in Japan can survive exposure to -5 degrees C for two weeks, whether it is in a diapause or non-diapause state. Trehalose was the main sugar detected in the body, but its level was less than 0.8%. Trehalose levels increased in field-collected pupae from January to March. DP accumulated less trehalose than NDP, as the acclimation temperature was decreased from 5 to -5 degrees C. The alanine content in field-collected pupae increased from November to February. Both diapause and low temperature caused an accumulation of alanine. These results suggest that under field conditions, overwintering pupae of H. cunea in Japan do not accumulate high levels of sugars and polyols and do not develop a high level of cold hardiness. Furthermore, DP do not accumulate high levels of sugars and polyols and their ability to survive exposure to -15 degrees C is not greater than that of NDP. The physiological and biochemical bases of diapause in H. cunea from Japan are discussed.  相似文献

The cold hardiness of arthropods is an important characteristic associated with overwintering success. Cold‐tolerant stages affect overwintering strategy, especially in arthropods that continuously feed on evergreen host plants in temperate regions. However, cold hardiness to mildly low temperature is rarely investigated. In the present study, we estimate the stage‐specific cold hardiness of a population of the spider mite Stigmaeopsis longus (Saito) (Acari: Tetranychidae) occurring on evergreen Sasa bamboo in a temperate area (Kochi City, southwestern Japan). Individuals of each stage developing under diapause‐inducing conditions (LD 10 : 14 h at 20 °C) are maintained at 4 °C (approximating the mean daily minimum temperature for December in Kochi City) for 30 days (eggs are cooled immediately, without short‐day development first). They are then returned to the initial conditions to assess survival rates. The survival rate of adult females (i.e. the diapausing stage) is high (99.5%), as is that of adult males and deutonymphs of both sexes (84.2–98.7% and 89.7–89.8%, respectively). The survival rate of mobile immature stages tends to increase toward adulthood in both sexes, whereas the survival rate of the eggs and quiescent stages is extremely low (16–23% and 0–20%, respectively). The poor cold hardiness of the eggs and quiescent stages presumably prevents the normal development of immature individuals during winter.  相似文献

【目的】滞育诱导期进行短光照处理可影响昆虫耐寒性。为明确光周期对中华通草蛉Chrysoperla sinica (Tjeder)耐寒性的影响, 针对中华通草蛉滞育解除过程及非滞育虫态的耐寒性进行了一系列研究。【方法】测定了中华通草蛉自然越冬成虫的过冷却点(supercooling point, SCP)以及长光周期(15L∶9D)和短光周期(9L∶15D)条件下自然越冬成虫在滞育解除过程中在-12℃下的死亡率, 并测定了室内长、 短两种光周期下实验种群2龄和3龄幼虫的过冷却点(SCP)、 结冰点(freezing point, FP)以及-7℃下的死亡率。【结果】中华通草蛉12月份的自然越冬成虫SCP集中在-10~-14℃之间。SCP低于-12℃的个体占43.70%, 且-12℃处理1 d死亡率为62.00%。-12℃处理1 d条件下的长、 短光周期处理自然越冬成虫, 除处理0 d外, 长光周期处理死亡率均高于短光周期处理的, 且在处理15 (P=0.012), 20 (P=0.01)和25 d (P=0.001)差异显著。中华通草蛉试验种群相同龄期幼虫在短光周期下的SCP和FP均高于长光周期下, 但差异不显著(P>0.05); 但在-7℃下, 2龄幼虫短光周期下的低温死亡率为67.00%±4.04%, 显著低于长光周期下的低温死亡率(78.00%±1.33%)(P=0.011), 3龄幼虫短光周期条件下低温死亡率为24.33%±1.33%, 显著低于长光周期下的低温死亡率(53.00%±3.46%)(P=0.002)。【结论】中华通草蛉为结冰敏感型, 诱导滞育的短光照处理可提高其幼虫期及滞育解除过程中成虫的耐寒能力。  相似文献

Larvae of Syrphus ribesii collected from overwintering sites in the U.K. are strongly freeze tolerant with 70% survival at -35 degrees C. The cold tolerance of laboratory reared insects increased with increasing periods of acclimation at 0 degrees C, with a concurrent rise in the supercooling point (SCP) from -6.8+/-0.1 to -5.1+/-0.3 degrees C. There was 50% survival in the most cold-hardy group 72h after brief exposures to -30 degrees C. The retention of gut contents caused a decrease in cold hardiness, with only 13% of larvae surviving 72h after exposure to -15 degrees C, with no subsequent pupation or emergence. Wet larvae had a significantly higher SCP (-5.0+/-0.2 degrees C) compared to dry larvae (-7.8+/-0.4 degrees C), although survival of larvae was similar in both groups. There was no nucleator activity in the haemolymph of field collected larvae. The importance of these findings are discussed in relation to the freeze tolerance strategy of S. ribesii.  相似文献

Abstract The apple leaf miner Phyllonorycter ringoniella (Matsumura) (Lepidoptera: Gracillariidae) overwinters as a diapausing pupa. The diapause rate reaches 100% in early October. Diapause intensity decreases gradually from early October and diapause terminates in early February. The fresh body weight of diapausing pupae is 1.6 times that of non-diapausing pupae. The main cryoprotectant in P. ringoniella pupae is trehalose. Three stages are distinguishable as indicated by the correlations between diapause intensity, levels of cold hardiness and the trehalose content: diapause induction occurred in October, diapause development from November to December, and post-diapause quiescence from January to April. During diapause induction, the pupae accumulate low levels of trehalose and do not survive exposure to −15 °C. During diapause development, the pupae gradually accumulate more trehalose and show some ability to survive exposure to −15 °C, but not to −20 °C. During post-diapause quiescence, the pupae accumulate relatively more trehalose and cold hardiness fully develops, but decreases quickly in April. The trehalose content in pupae sampled in December is unaffected by acclimation temperatures in the range 0–30 °C, but decreases in pupae sampled in March after acclimation at temperatures from 5 to 15 °C. These results suggest that overwintering pupae of P. ringoniella have the ability to accumulate trehalose and develop a high level of cold hardiness during diapause development.  相似文献

中华通草蛉(Chrysoperla sinica)成虫在自然条件下抗寒能力的季节性变化研究表明,雌、雄成虫的抗寒能力均呈现出季节性的变化趋势,即随着冬季低温的到来,其抗寒能力逐渐增强,冬季过后又随气温的回升,其抗寒力逐渐减弱.雌、雄成虫的体内含水量、过冷却点(SCP)和结冰点(FP)均随气温的降低而降低,升高而升高,但体内总脂肪的含量却随气温的降低而升高,升高而降低.越冬代在越冬的前期和中期,雌、雄成虫体内的含水量和SCP均显著低于生长季节的其它各代,越冬代后期的含水量和SCP与生长季节其它各代没有显著差异.经回归分析发现,雌、雄成虫体内含水量与SCP之间均存在极显著负相关关系(p<0.01);越冬代成虫(特别是越冬前期)的体内总脂肪含量显著高于其它时期,并且雌成虫体内的总脂肪含量与SCP之间呈显著正相关(p<0.05),雄成虫体内总脂肪含量与SCP之间没有显著相关性(p>0.05).在一年的不同世代中,雌、雄成虫体内含水量只是在越冬前期有显著差异,SCP和体内总脂肪含量却在越冬后期有显著差异.  相似文献

Amounts of several metabolites were measured in overwintering larvae of Enosima leucotaeniella acclimated to temperatures between -5 and 15 degrees C for 30days. In the diapausing stage, cold hardiness, as shown by the survival rate, began rising below 15 degrees C. Glycogen content decreased as the temperature decreased from 10 to 0 degrees C. Trehalose content rose as the temperature decreased from 15 to 5 degrees C, but remained unchanged as the temperature decreased from 5 and 0 degrees C. Twenty-eight free amino acids were detected in the haemolymph; levels of proline, glutamine and glutamic acid increased at high temperatures, but alanine increased at low temperatures, especially as temperature decreased from 5 to 0 degrees C. Lipid content was unchanged by the different acclimation temperatures. The effects of temperature, diapause and aerobic conditions on the levels of carbohydrates and amino acids in overwintering larvae were analyzed. Alanine levels rose at low temperature only when the larvae were in the diapausing stage. The level of trehalose rose at low temperature in both the diapausing and post-diapausing stages, although it was higher at aerobic conditions in the post-diapausing stage. These results suggest that efficient trehalose synthesis occurs under the combination of low temperature and aerobic conditions of the post-diapausing stage, so that cold hardiness in overwintering E. leucotaeniella larvae may rise to a high level in winter.  相似文献

Cicadulina bipunctata was originally distributed in tropical and subtropical regions of the Old World. This leafhopper recently expanded its distribution area to southern parts of temperate Japan. In this study, factors affecting the overwintering ability of C. bipunctata were examined. A series of laboratory experiments revealed that cold acclimation at 15 °C for 7 days enhanced the cold tolerance of C. bipunctata to the same level as an overwintering population, adult females were more tolerant of cold temperature than adult males, and survival of acclimated adult females was highly dependent on temperature from −5 to 5 °C and exposure duration to the temperature. The temperature of crystallization of adult females was approximately −19 °C but temperatures in southern temperate Japan rarely dropped below −10 °C in the winter, indicating that overwintering C. bipunctata adults in temperate Japan are not killed by freezing injury but by indirect chilling injury caused by long-term exposure to moderately low temperatures. An overwintering generation of C. bipunctata had extremely low overwinter survival (<1%) in temperate Japan; however, based on winter temperature ranges, there are additional areas amenable to expansion of C. bipunctata in temperate Japan.  相似文献

Cold hardiness of Helicoverpa zea (Lepidoptera: Noctuidae) pupae   总被引:1,自引:0,他引:1  
An insect's cold hardiness affects its potential to overwinter and outbreak in different geographic regions. In this study, we characterized the response of Helicoverpa zea (Boddie) pupae to low temperatures by using controlled laboratory measurements of supercooling point (SCP), lower lethal temperature (LT(50)), and lower lethal time (LLTime). The impact of diapause, acclimation, and sex on the cold hardiness of the pupae also were evaluated. Sex did not significantly affect the SCP, LT(50), or LLTime. However, the mean SCP of diapausing pupae (-19.3°C) was significantly lower than nondiapausing pupae (-16.4°C). Acclimation of nondiapausing pupae to constant temperatures from 10 to 20°C before supercooling also produced a significantly lower SCP than nondiapausing pupae held at 25°C. The LT(50)s of nondiapausing and diapausing were not significantly different, but confirmed that H. zea pupae are chill-intolerant because these lethal temperatures are warmer than the corresponding mean SCPs. Diapausing pupae survived longer than nondiapausing pupae at the same, constant, cold temperatures, a finding consistent with the SCP results. Both of these results suggest enhanced cold hardiness in diapausing pupae. When laboratory results were compared with field temperatures and observed distributions of H. zea in the contiguous United States, the laboratory results corroborated what is currently perceived to be the northern overwintering limit of H. zea; approximately the 40(th) parallel. Moreover, our research showed that areas north of this limit are lethal to overwintering pupae not because of low temperature extremes, but rather the length of time spent at near-zero temperatures.  相似文献

Plantlets of Solanum commersonii stem-culture were acclimated at 5°C day/night temperature for 14 days. Cold hardiness increased from – 3.5°C to – 8.6°C. During the course of acclimation, the synthesis of polypeptides was investigated and poly (A+) RNA was isolated. Translation products of poly(A+) RNA in a rabbit rcticulocyte lysate system were then analyzed. During the 14 days of acclimation, 23 cold-induced polypeptides were identified. Most of them disappeared following 1 day of de-acclimation at a 20/15°C day/night regime. The synthesis of one group of polypeptides is prominent and stable throughout the acclimation period. The other group is transient. The most prominent and stable polypeptides have molecular weights of 21, 22, 31 and 83 kDa.
Acclimation alters translatable mRNA population during the development of cold hardiness. Two mRNAs encoding in vitro translation products at 26 and 27 kDa were identified during the course of acclimation. These proteins may play important roles in the overall programming for the development of cold hardiness in tuber-bearing S. commersonii.  相似文献

Abstract. 1. For many species of insect, cold hardiness is an important trait that enables a population to develop in the next season and to extend its range. To elucidate the role of cold hardiness of the migratory locust Locusta migratoria L. in its outbreak and distribution areas, egg cold hardiness was examined in locusts derived from four locations from latitude 18°23'N to latitude 41°10'N in eastern China.
2. The supercooling points of eggs from different geographic populations did not differ significantly for the first development stage, with an average ± SE of −24.5 ± 0.51 °C, or for the second stage, −22.06 ± 0.68 °C, however there was a significant difference for the embryonic development phase among the four geographical populations. The egg supercooling point increased gradually from neonatal egg to old egg; eggs prior to hatching always had a much higher supercooling point.
3. Comparisons of the cold hardiness of four populations were carried out by validating the close correlation between latitude and the effects of cold on hatching, low lethal temperature (Ltemp50), and low lethal time (Ltime50). There were significant differences among the four populations; the northern population was more cold hardy than the southern population, and the two mid-latitude populations were intermediately cold hardy.
4. The cold hardiness of all populations was enhanced to various degrees by short-term cold acclimation at 0 °C and 5 °C. For most populations, a 2-day acclimation period seemed to be optimal.  相似文献

A plasmalemma fraction was isolated from homogenized apple tree (Mains domestica Borkh 'Golden Delicious') hark tissues using aqueous phase partitioning and ultra-centrifugation. Results of marker enzyme assays indicated that a membrane preparation highly enriched in plasma membranes was obtained. ATPase activity in this preparation possessed a high specificity for ATP as substrate, was inhibited by vanadate, diethylstilbcsterol and dicyclohexylcarbocHimide, and was insensitive to inhibitors of mitochondria! and tonoplasl ATPases. Specific activity of the plasma-lemma ATPase increased during cold acclimation prior to the attainment of vegetative maturity. Kinetic parameters (Km, Vln) determined from assays performed at different temperatures (10, 30°C) indicated a differential effect of cold acclimation on enzyme activity. Vm increased during cold acclimation, whereas Km increased when determined at 30°C but declined at 10°C. Acclimation treatments during April and May resulted in alteration of ATPase kinetics in the absence of any increase in bark frost hardiness. Changes in ATPase kinetics may be related more to enhanced low temperature metabolism than to frost hardiness per se.  相似文献

Supercooling point studies were used to investigate the factors influencing the cold hardiness of the peach-potato aphid Myzus persicae, a freezing-susceptible insect. Overwintering adults lost cold hardiness as winter progressed, with a variable proportion showing a marked reduction in supercooling ability. Cold hardiness increased in spring so that all individuals demonstrated extensive supercooling ability typical of aphids reared in the laboratory at 20°C with a long photoperiod; these levels of cold hardiness were maintained in the field during summer and early autumn. First instar nymphs demonstrated considerable cold hardiness all year. Surface moisture caused inoculative freezing in some first instar nymphs and adults when supercooled, but the majority were unaffected. In the laboratory, adults starved for 7 days at 5°C showed distinct losses of supercooling potential equivalent to those observed in the field during mid to late winter. No loss of cold hardiness was found in first instar nymphs starved under the same conditions. The results demonstrate that the cold hardiness characteristics of M. persicae are atypical of those observed in other freezing-susceptible insects and it is suggested that continued feeding during mild winter conditions allows maintenance of cold hardiness particularly in adult aphids, and provides a possible explanation for the successful anholocyclic overwintering of M. persicae during such winters.  相似文献

Abstract.  The effect of moulting on the cold hardiness of the oribatid mite Alaskozetes antarcticus (Michael) is investigated. Non moulting animals show clear seasonal patterns of cold hardiness with high supercooling points (SCPs) at the peak of summer and an increasing proportion of low SCPs with declining environmental temperatures. By contrast, both field-fresh and laboratory acclimated (5 °C) mites in the moult state are consistently found to have low SCPs regardless of environmental temperature.  相似文献

Leptopilina heterotoma is a Drosophila parasitoid mainly occurring in temperate regions, and females of this species are pro‐ovigenic (i.e. all or nearly all of their lifetime eggs are already mature at emergence). Here I investigated how L. heterotoma overwinters in Sapporo, Japan, a cool temperate region, by outdoor and laboratory experiments. Females of this species had mature eggs at emergence as expected, and they did not resorb eggs even if they were exposed to cold or starvation. Nevertheless, females were able to overwinter, and post‐overwintering females retained parasitization capacity. In this species, thus, adult overwintering is not associated with reproductive diapause. However, females grown in late autumn or at low temperatures (e.g. 5°C) had a fewer number of mature eggs and higher winter survival. At low temperatures, female larvae could save energy by reducing egg production and allocate the saved energy to expenditure for overwintering. In contrast to females, males and pre‐adult individuals were not able to overwinter.  相似文献

Mortality of adult Tingis ampliata H.-S. increases with decreasing temperatures from ?2 to ?13° in the laboratory. There is no differential susceptibility of the sexes to low temperatures in the above range, but a higher proportion (5%) of females tolerated longer periods of exposure to these temperatures than males. There is a great deal of individual variation in cold hardiness of bugs of both sexes. Seasonal variation in cold hardiness is associated with variations in mean air temperatures during winter. Cold hardiness increases from October to December and then decreases progressively afterwards till the following April. During the coldest month (December), undercooling point ranged from ?22.0 to ?31.5° for females and ?22.2 to ?31.4° for males. Presence of food in the gut and contact with wet organic substrate such as dead Holcus leaf, both reduce cold hardiness of bugs by about 5° compared with unfed bugs or those tested on dry, dead or living green Holcus leaf. The difference between cold hardiness of control and fed bugs is more marked in individuals that have access to food for 48 hours than in those that have access for only 24 hours.  相似文献

This investigation examined the influence of soil moisture and associated parameters on the cold hardiness of the Colorado potato beetle (Leptinotarsa decemlineata Say), a temperate-zone species that overwinters in terrestrial burrows. The body mass and water content of adult beetles kept in sand at 4 °C varied over a 16-week period of diapause according to the substratum's moisture content. Changes in body water content, in turn, influenced the crystallization temperature (range −3.3 to −18.4 °C; n = 417), indicating that environmental moisture indirectly determined supercooling capacity, a measure of physiological cold hardiness. Beetles held in dry sand readily tolerated a 24-h exposure to temperatures ranging from 0° to −5 °C, but those chilled in sand containing as little as 1.7% water (dry mass) had elevated mortality. Thus, burrowing in dry soils not only promotes supercooling via its effect on water balance, but may also inhibit inoculative freezing. Mortality of beetles exposed to −5 °C for 24 h was lower in substrates composed of sand, clay and/or peat (36–52%) than in pure silica sand (78%) having an identical water content (17.0% dry mass). In addition to moisture, the texture, structure, water potential, and other physico-chemical attributes of soil may strongly influence the cold hardiness and overwintering survival of burrowing insects.
