The possible involvement of opioid system in the regulation of hemolymph glucose level in the fresh water crab Oziotelphusa senex senex Fabricius, was investigated. Opioid agonist and antagonist was also used in addition to methionine-enkephalin itself. Injection of the opioid, methionine-enkephalin and FK 33-824 significantly elevated hemolymph glucose level. In contrast, injection of naloxone in to crab resulted in decrease in hemolymph glucose level. Injection of naloxone prior to injection of methionine-enkephalin blocked the hyperglycemic action of methionine-enkephalin. Injection of methionine-enkephalin, FK 33824 and naloxone produced no significant effect on hemolymph glucose level in eyestalk-less crab. The alterations in the intact crab hemolymph glucose level hypothesised to be due to stimulation of release of hyperglycemic hormone during methionine-enkephalin and FK 33824 treatment and blocking of release of hyperglycemic hormone during naloxone treatment from the eyestalks of crab Oziotelphusa senex senex.  相似文献

1. Na(+)-K+ and Mg(2+)-tissue ATPases of the freshwater crab Oziotelphusa senex senex showed increasing inhibition when exposed to a sublethal concentration (1.86 mg/l = 0.1 of LC50) of endosulfan for 1-30 days. 2. Na(+)-K(+)-ATPase activity in all tissues (thoracic nerve mass, gill, hepatopancreas and claw muscle) was higher than Mg(2+)-ATPase activity. 3. After 30 days exposure tissue Mg(2+)-ATPase was less affected than Na(+)-K(+)-ATPase. 4. Crabs exposed to endosulfan and then returned to uncontaminated water showed greater recovery of Mg(2+)-ATPase than Na(+)-K(+)-ATPase with 90-95% recovery after 1 day exposure and 60-65% recovery after 30 days exposure. 5. Changes in behaviour of the crabs were noted after 7 days exposure to endosulfan with progressive loss of coordination, decreased activity and increased exudation of mucus.  相似文献

The objective of the present study was to explore the site of synthesis of vitellogenin (Vtg) in fresh water edible crab, Oziothelphusa senex senex. Vtg cDNA fragments were isolated from the hepatopancreas of female crabs using RT-PCR method, and the deduced amino acid sequence of O. senex senex showed more than 60% identity with other brachyuran Vtg sequences. RT-PCR analysis showed that Vtg mRNA can be detected only in hepatopancreas of female Oziothelphusa but not in other tissues including eyestalks, Y-organs, mandibular organs, thoracic ganglion, hypodermis and ovary. Antibodies were raised against vitellin purified from the ovary of O. senex senex. Immunoprecipitation analysis revealed the presence of Vtg in the hepatopancreas of vitellogenic stage I females and in the hemolymph, hepatopancreas and ovary extracts from vitellogenic stage II females but absent in hemolymph and hepatopancreas extract of males. These results suggest that Vtg is synthesized only in hepatopancreas but not in the ovaries of O. senex senex. In addition, Vtg synthesized in hepatopancreas is transported to ovary through hemolymph.  相似文献

In the present study, the role of eyestalks and involvement of methionine-enkephalin in the regulation of haemolymph sugar level was studied. Bilateral eyestalk ablation significantly decreased the haemolymph sugar levels, whereas injection of eyestalk extract into ablated crabs significantly increased the haemolymph sugar levels. Total carbohydrate (TCHO) and glycogen levels were significantly increased in hepatopancreas and muscle of eyestalk-ablated crabs, with a decrease in phosphorylase activity. Injection of eyestalk extract into ablated crabs resulted in partial/complete reversal of these changes. Injection of methionine-enkephalin into intact crabs significantly increased the haemolymph sugar level in a dose-dependent manner. Total tissue carbohydrate and glycogen levels were significantly decreased, with an increase in phosphorylase activity in hepatopancreas and muscle tissues of intact crabs after methionine-enkephalin injection. Methionine-enkephalin injection did not cause any changes in haemolymph sugar, tissue total carbohydrate and glycogen levels and activity levels of phosphorylase in eyestalk-ablated crabs. These results suggest that the eyestalks are the main source of hyperglycaemic hormone and methionine-enkephalin induces hyperglycaemia through eyestalks.  相似文献

The concentration of haemolymph sugar and the hyperglycaemic activity of eyestalk extract was measured six times (8, 12, 16, 20 and 4 h) over a 24-h period. The concentration of haemolymph sugar and hyperglycaemic activity of eyestalk extract was higher during the night (0 h through 8 h) than that noted in day time (12 h). Cellular theory of aging states that human aging is the result of cellular aging, in which an increasing proportion of cells reach senescence. Senescence, from the Latin word senex, means “growing old,” is an irreversible growth arrest which occurs in response to damaging stimuli, such as DNA damage, telomere shortening, telomere dysfunction and oncogenic stress leading to suppression of potentially dysfunctional, transformed, or aged cells. Cellular senescence is characterized by irreversible cell cycle arrest, flattened and enlarged morphology, resistance to apoptosis, alteration in gene expression and chromatin structure, expression of senescence associated- β-galactosidase (SA-β-gal) and acquisition of senescence associated secretory phenotype (SASP). In this review paper, different types of cellular senescence including replicative senescence (RS) which occurs due to telomere shortening and stress induced premature senescence (SIPS) which occurs in response to different types of stress in cells, are discussed. Biomarkers of cellular senescence and senescent assays including BrdU incorporation assay, senescence associated- β-galactosidase (SA-β-gal) and senescence-associated heterochromatin foci assays to detect senescent cells are also addressed.  相似文献

The centriole pair in animals shows duplication and structural maturation at specific cell cycle points. In G1, a cell has two centrioles. One of the centrioles is mature and was generated at least two cell cycles ago. The other centriole was produced in the previous cell cycle and is immature. Both centrioles then nucleate one procentriole each which subsequently elongate to full-length centrioles, usually in S or G2 phase. However, the point in the cell cycle at which maturation of the immature centriole occurs is open to question. Furthermore, the molecular events underlying this process are entirely unknown. Here, using monoclonal and polyclonal antibody approaches, we describe for the first time a molecular marker which localizes exclusively to one centriole of the centriolar pair and provides biochemical evidence that the two centrioles are different. Moreover, this 96-kD protein, which we name Cenexin (derived from the Latin, senex for "old man," and Cenexin for centriole) defines very precisely the mature centriole of a pair and is acquired by the immature centriole at the G2/M transition in prophase. Thus the acquisition of Cenexin marks the functional maturation of the centriole and may indicate a change in centriolar potential such as its ability to act as a basal body for axoneme development or as a congregating site for microtubule-organizing material.  相似文献

The duration of the cellular cycle and the diurnal rhythm of the amount of mitosis were studied in young rats in normality and under the influence of thyroxin. The parenchymal and connective-tissue cells of the liver and cells of the liver and the cells of the oesophagus epithelium basal layer were studied. It was found that under the influence of thyroxin there occured a shortening of the periods of the cellular cycle and a 3--6 h shift to the left of the diurnal rhythm curve of the amount of mitoses. In thyroxinized animals the 21--95% increase of the amount of mitoses in the period of maximum values of the mitotic index during a day was observed as compared with control animals. A conclusion is made about the diurnal rhythm of sensitivity of G0-phase cells to the synchronizing factor, suggesting the decisive significance of the state of the cell population in the interaction of the tissue and hormone cells. The data obatained in the work show that the thyroid hormones regulate the cellular reproduction in the organism by stimulating the cells in the division cycle, synchronization of greater amount of cells by the moment of beginning of the mitotic cycle at a definite time of day and by shortening the period of the cell mitotic cycle.  相似文献

The structure of the active center of glyceraldehyde-3-phosphate dehydrogenase and the arrangement of subunits in the tetrameric molecule is delineated. The mechanism of cooperative effects in the oligomer is considered, and the involvement of various regions of the active center and of different-subunit contact area in the realization of the cooperative phenomena is discussed. A special attention is paid to the effect of NAD+ bound to one of the subunits of the tetramer on the structure of an adjacent subunit and to the problem of the participation of the coenzyme in the creation of anion-binding sites of the enzyme. The conditions of reversible dissociation of the tetrameric apoenzyme molecule into dimers are depicted, and the role of NAD+ in the organization of the quaternary structure of the dehydrogenase is discussed. The problem of catalytic activity of the dimeric form of the enzyme is argued.  相似文献

In the first part of the paper, the results of the investigation of the rhodopsin arrestin interaction are presented. The results were mainly obtained with the technique of the selective labelling of the rhodopsin and arrestin SH-groups and the rhodopsin limited proteolysis. These results are discussed in the frame of the latest data on the three-dimensional structure of arrestin. In the second part of the paper, results of the antigenic properties of arrestin (S-antigen) and its role in the pathogenesis of the retina diseases are summarized. The data on the role of the autoimmune processes in the pathogenesis of diabetic retinopathy are presented. We have also described the results of the use of the elaborated technique of the immune diagnostics in the prognosis of the diabetic retinopathy and retinopathy of the premature babies.  相似文献

The interferons (IFN's) are natural regulators of the immune response. This quality of IFN depends to a large extent on their capacity to change the expression of the antigens of the second (II) class of the Major Histocompatibility Complex. The most effective modulator of the expression of the antigens of the class II MHC is beta-IFN. The aim of this work is to explore with the aid of enzyme-linked immunosorbent assay (ELISA) peculiarities of an action of the preparations alpha- and beta-IFN's of a man on the expression of the antigens of the second class on macrophages of mice with using monoclonal antibodies to the products of subregions I-A and I-E MHC. As a result of exploration we have the following: both preparations IFN's (leukinterferon and beta-IFN) reinforce the expression of the antigens of the second class on the macrophages of mice spleens. With combined influence preparation alpha-IFN and beta-IFN show interweakened effect on the expression of the antigens of the class MHC. Exposed regularities of the expression have the same type for products of subregions I-A and I-E H-2 of mice complex. ELISA is the sensible method of determination of a level of the expression of the antigens of the class II MHC on the cell's populations.  相似文献
