Cryothecomonas longipes Schnepf and Kühn sp. nov. is a colourless biflagellate organism, 9–14 μm long and 7–9 μm wide when not filled with food vacuoles. It was detected in the North Sea, feeding with pseudopodia on diatoms. It penetrates the host shell, while the main body of the flagellate remains outside the frustule. Cells are covered with a multilayered theca. The pseudopodium protrudes through a preformed slit in the theca. Each flagellum also emerges through a pit in which the theca forms a funnel of complex structure that girdles each flagellum. The anterior flagellum is 9–15 μm long and oriented forward; the ventral flagellum, posteriorly directed, is 20–24 μm long and bears fine hairs. The flagellar roots consist of microtubules that emerge at satellites around the basal bodies and run along the flagellar pits. In addition, the ventral flagellum is accompanied by a band of six microtubules. It is proximally attached to a small fibrillar band, which interconnects the basal bodies. Cryothecomonas longipes has two or three types of extrusomes which pierce the theca when discharged. Their mode of discharge is discussed. Microbody-like vesicles containing small tubules are closely associated with older digestion vacuoles. Cryothecomonas longipes is compared with other species of the genus and a diagnosis is given. The ultrastructure of the marine predatory flagellate Metromonas simplex Larsen et Patterson was studied. The cell is surrounded by a low-contrast fibrous layer composed of thin hairs covered by a thin bilayer membrane and an outer layer of thin short fibers. The plasmalemma lies under these layers. The predator captures whole cells of the prey, usually bodonids or chrysomonads. The cytostome as a cell pocket is undetectable. The long flagellum bears very thin mastigonemes (hairs) with lengths of 0.8–1.0 μm; the short flagellum is naked and reduced in length. The transitional zone lacks spirals or other additional elements. The transversal plate is elevated on the cell surface. The flagellar root system is very simple and has one microtubular band which originates near the kinetosomes. The latter are parallel to each other and interconnected by fibrous bridges. The vesicular nucleus, Golgi apparatus, and endoplasmic reticulum are of typical structures. The oval mitochondria of 0.6–2.5 μm contain lamellar cristae. The cylindrical extrusomes (trichocysts) found in the cytoplasm have lengths of 1.0–1.4 μm and diameters of 0.12–0.08 μm. The trichocysts have a wheel-shaped structure with 13 spokes visible in cross-sections. The contractile vacuole is absent. The similarity that M. simplex shares with Metopion fluens Larsen et Patterson, cryothecomonads, and other predatory flagellates is discussed.  相似文献

Detergent-isolated flagellar apparatuses of the flagellate green alga Dunaliella bioculata retain remnants of the nucleus (the karyoskeleton) which are linked to the basal bodies by the centrin-containing nucleus basal body connectors (NBBC). Such complexes were subjected to different calcium concentrations (1 × 10?9 M ? 5 × 10?4 M Ca2+) and the distance between the basal bodies and the karyoskeleton was measured by light microscopy. The threshold concentration of Ca2+ for NBBC contraction was determined to be around 5 × 10?8 M Ca2+. At > 10?7 M Ca2+ NBBC were maximally contracted and the distance between the basal bodies and the karyoskeleton was only about 50% of the initial distance. Using a polyclonal antibody generated against centrin (Salisbury et al., 1984), the NBBC were visualized by indirect immunofluorescence in both the extended and contracted state. Our results demonstrate that in vitro contraction of centrin-containing filaments in green algae is initiated at about the same free Ca2+ concentration in three different centrin-containing basal apparatus components (i.e. the distal connecting fibre, the flagellar transitional region and the NBBC).  相似文献

A new sand‐dwelling dinoflagellate from Palau, Galeidinium rugatum Tamura et Horiguchi gen. et sp. nov., is described. The life cycle of this new alga consists of a dominant nonmotile phase and a brief motile phase. The motile cell transforms itself directly into the nonmotile cell after swimming for a short period, and cell division takes place in the nonmotile phase. The nonmotile cell possesses a dome‐like cell covering, which is wrinkled and equipped with a transverse groove on the surface. The cell has 10–20 chloroplasts and a distinct eyespot. The motile cell is Gymnodinium‐like in shape. The dinoflagellate possesses an endosymbiotic alga to which the chloroplasts belong and which is separated from the host (dinoflagellate) cytoplasm by a unit membrane. The endosymbiont cytoplasm also possesses its own eukaryotic nucleus and mitochondria. The eyespot is surrounded by triple membranes and is located in the host cytoplasm. Photosynthetic pigment analysis, using HPLC, revealed that G. rugatum possesses fucoxanthin as the principal accessory pigment instead of peridinin. The rbcL tree showed that G. rugatum is monophyletic with Durinskia baltica (Levander) Carty et Cox and Kryptoperidinium foliaceum (Stein) Lindemann and that this clade is closely related to the pennate diatom, Cylindrotheca sp. The endosymbiont of G. rugatum is therefore shown to be a diatom. Phylogenetic analysis based on small subunit rDNA sequences demonstrated that G. rugatum, D. baltica, and K. foliaceum, all of which are known to harbor an endosymbiont of diatom origin, are closely related.  相似文献

This paper concentrates on the study of vegetative development and sexual reproduction inLagenisma coscinodisci Drebes. Before infecting a diatom cell (Coscinodiscus granii), the freshly released zoospores pass through two different cyst stages. The primary zoospores are kidney-shaped and laterally biflagellated. They form a primary cyst with a spiny cyst wall which is left by isomorphic secondary zoospores. The latter form a secondary cyst (sometimes perhaps repeatedly), which is smooth-walled. The secondary cyst germinates and infects a new diatom cell by means of an infecting tube which enters the cell through the gap between epi- and hypotheca and develops a new thallus. Sexual reproduction is induced by ageing of cultures. Two kinds of isomorphic, kidney-shaped, laterally biflagellated swarmers are produced as in zoosporogenesis. Female-determined swarmers settle down near the host cell and encyst with a more or less smooth wall (oogonia). Male-determined swarmers are obviously attracted by the oogonia and encyst close to them to form a more or less smooth-walled antheridium which drives a thin fertilization tube to the oogonium. During plasmogamy the oogonium develops a thick, short hypha. The cytoplasm with the two nuclei moves completely into this hypha, concentrates near the hyphal tip, and surrounds itself with a thick wall to become a resting spore (oospore) in which karyogamy takes place. The walls of cysts, tubes, hyphae and spores react positively with zinc-chlor-iodide. In the spines and walls of the primary cysts the network of 2 to 3 nm thick fibrils is more distinct than in the other walls.  相似文献

Functional studies on molecular transport through plasmodesmata in leaf mesophyll and trichome cells revealed significant differences in their basal size-exclusion limits and their response to microinjected tobacco mosaic virus movement protein (E. Waigmann et al., 1994, Proc. Natl. Acad. Sci. USA 91: 1433–1437; E. Waigmann and P. Zambryski, 1995, Plant Cell 7: 2069–2079). To address the basis for these functional differences, Nicotiana clevelandii trichome and mesophyll plasmodesmata were compared ultrastructurally. Trichome plasmodesmata increase in ultrastructural complexity from the tip to the base cell. Their neck regions, thought to control molecular traffic through plasmodesmata, are clearly distinct from necks of mesophyll plasmodesmata. In contrast to the electron-dense desmotubular area in mesophyll plasmodesmata, trichome plasmodesmata contain an electron-translucent circle in their center, surrounded by an electron-dense ring. This latter ring is connected to the inner leaflet of the plasma membrane by multiple spokes or filaments. Two monoclonal antibodies raised against a maize plasmodesmal protein preparation (A. Turner et al., 1994, J Cell Sci. 107: 3351–3361) interact with both trichome and mesophyll N. clevelandii plasmodesmata. Based on the localization pattern and the high degree of cross-reactivity, both antibodies likely recognize a conserved structural component of plasmodesmata, and may be useful to mark plasmodesma in a variety of plants and tissues. A new species, Marsupiomonas pelliculata gen. et sp. nov. (Pedinophyceae), is described. A single flagellum emerges from a deep pit with a distinctive thickened margin. The flagellum has rigid fibrillar hairs which are probably formed in the perinuclear space. A short second flagellar basal body lies within the cell close to the basal body of the emergent flagellum and the flagellar root system consists of striated and microtubular roots. There is a distinctive theca covering all but the anterior end of the cell and also a single large bright green chloroplast with an immersed pyrenoid surrounded by a starch shell. The wide salinity tolerance of the species is discussed in relation to its distribution in estuarine and salt marsh habitats. The salient features of the new species—the insertion of the emergent flagellum into a deep pit and the possession of a theca—are also seen in Pedinomonas tenuis, and it is suggested that P. tenuis could be transferred to the new genus Marsupiomonas. The class Pedinophyceae now includes three genera (Pedinomonas, Resultor and Marsupiomonas) and the distinguishing features are discussed.  相似文献

This is the first report of a Ulva species reproducing asexually solely by quadriflagellate swarmers. Ulva spinulosa Okamura et Segawa specimens were collected from Ukibuchi on the Pacific coast of Kochi Prefecture, southern Japan. Quadriflagellate swarmers were released from these specimens. The swarmers showed negative phototaxis before settlement. Thalli cultured from these swarmers also released quadriflagellate swarmers in culture. Microspectrophotometric studies demonstrated equivalent DNA in nuclei of vegetative cells in thalli of U. spinulosa and in sporo‐phytes of the other Ulva species with sexual life history (U. fasciata Delile). Furthermore, the quadriflagellate swarmers of U. spinulosa had the same DNA value, demonstrating that the quadriflagellate swarmers are produced without meiosis.  相似文献

Pelagomonas calceolata gen. et sp. nov., an ultra-planktonic marine alga, is described using electron microscopy and the cytoplasmic small subunit (18S) ribosomal RNA (rRNA) gene sequence. Cells are uniflagellate, about 1.5 × 3 μm in size. The flagellium has two rows of bipartite hairs, the paraxonemal rod has a dentate appearance, and a two-gyred transitional helix is present between two transitional plates. Microtubular roots, striated roots, and a second basal body are absent. A thin organic theca surrounds most of the cell. There is a single chloroplast with a girdle lamella and a single, dense mitochondrion with tubular cristae. A single Golgi body with swelled cisternae lies beneath the flagellum, and each cell has an ejectile organelle that putatwely releases a cylindrical structure. A vacuole, or cluster of vacuoles, contains the putative carbohydrate storage product. The 18S rRNA gene was sequenced completely in both directions, excluding three primer regions. When compared to the same gene sequence from other organisms, Pelagomonas calceolata gen. et sp. nov. occupies an unresolved position among other chromophyte algae and is distinct from members of any of these classes. Based on morphological, ultrastructural, and molecular data, we describe this alga as a new species, and we place this highly unusual new species in a new genus, family, order, and class.  相似文献

Pirsonia guinardiae gen. et spec. nov. was discovered as a parasite onGuinardia flaccida in the North Sea near List/Sylt during a bloom of this centric planktonic diatom. It is a colourless, small flagellate with an oblique cell apex and two subapically inserting flagella of different length and different orientation. The flagellates attach to a host cell and form an antapical process which pierces the diatom frustule and develops inside into a “trophosome”, consisting of a proximal digestion vacuole and distal pseudopodia which phagocytise host cytoplasm. The main body, the “auxosome”, remains outside the host cell. The trophosome persists for some time after the detachment of the auxosome or its derivatives. There are two types ofPirsonia guinardiae. Type A attaches to the valvae as well as to the girdle region, the auxosome remains flagellated and generally detaches after the feeding process to divide twice (seldom 3 times). Thick-walled (resting?) cysts are formed. Occasionally, a fusion of two sister cells has been observed. Type B attaches only to the valvae; the auxosome lacks flagella; it divides during the feeding process to give rise to a bouquet of 8 to more than 50 daughter cells which become flagellated when they detach. The taxonomical position of the flagellate is discussed. Diagnoses of genus and species are given.  相似文献

The cell structure of the freshwater chrysomonad Spumella sp. has been studied. The cell contains a vesicular nucleus, mitochondria with tubular cristae, Golgi apparatus, flagellar roots, and wide dorsal microtubular band. The flagella bear the spiral of four to five coils in the transitional zone. The rudiments of mastigonemes have been found in the perinuclear space. The compact leucoplast has an amorphous core surrounded by the membrane. No stigma has been detected. The leucosin vacuole, rhizoplast, and swelling of the short flagellum are absent. One to three osmiophilic granules lie near the leucoplast. The contractile vacuole is surrounded by tubules. The resemblance and difference of investigated flagellate with other chrysomonads are discussed.  相似文献

Invasive species trigger biodiversity losses and alter ecosystem functioning, with life history shaping invasiveness (Sakai et al., Annu Rev Ecol Syst 32:305–332, 2001). However, pinpointing the relation of a specific life history to invasion success is difficult. One approach may be comparing congeners. The two Palearctic pavement ants, Tetramorium sp.E (widely known as T. caespitum, Schlick-Steiner et al., Mol Phylogenet Evol 40:259–273, 2006) and T. tsushimae have invaded North America (Steiner et al., Biol Invasions 8:117–123, 2006). Their life histories differ in that T. sp.E has separate single-queened colonies but T. tsushimae multi-queened colonies scattered over large areas (Sanada-Morimura et al., Insect Soc 53:141–148, 2006; Schlick-Steiner et al., Mol Phylogenet Evol 40:259–273, 2006; Steiner et al., Biol Invasions 8:117–123, 2006). Comparison of the genetic diversity in the entire native and non-native ranges will elucidate the invasion histories. Here, we present 13 and 11 microsatellites, developed for T. sp.E and T. tsushimae, respectively, and characterize all for both species. Florian M. Steiner, Wolfgang Arthofer and Birgit C. The contents of fecal ribbons from Mytilus edulis L. fed either the unicellular flagellate Tetraselmis suecica (Kylin) Butch., or the diatom Thalassiosira pseudonana (Hasle and Heimdal) were examined for their particle size spectra. Contents of fecal ribbons were dispersed by shaking in filtered sea water and analyzed by a Coulter Counter. The flagellate can pass through the mussel's gut without any significant volume changes, implying that its digestion is largely intracellular (after phagocytosis). In contrast, the diatom does undergo volume changes, probably a result of disintegration of cells and aggregation of cells and cell fragments. Undigested flagellate cells can become resuspended in significant quantities after their defecation by the mussel.  相似文献

The ultrathin structure of the amoeboid flagellate Thaumatomonas coloniensis Wylezich et al. has been studied. The cell is surrounded by somatic scales forming on the surface of the mitochondria. The heterodynamic flagella emerge from the small flagellar pocket. Both flagella are covered by pineal scales and thin twisted mastigonemes. The kinetosomes lie parallel to each other. The transitional zone of the flagella carries the thin-walled cylinder. The transversal plate of the flagella is above the cell surface. The flagellar root system consists of three microtubular bands and a fibrillar rhizoplast. The vesicular nucleus and Golgi apparatus are of the usual structure. The mitochondria contain tubular cristae. The extrusive organelles (kinetocysts) contain amorphous material and a capsule; they are located in cytoplasm. The capsule consists of a muff and cylinder. Osmiophilic bodies of various shapes contain crystalloid inclusions. The pseudopodia capturing the bacteria emerge from the ventral groove. The groove is armored by the two longitudinal groups of the close situated microtubules. Microbodies and symbiotic bacteria have not been discovered. The resemblance of Th. coloniensis with other thaumatomonads is discussed.  相似文献

To infer the phylogeny of both the host and the endosymbiont of Peridinium quinquecorne Abé, the small subunit (SSU) ribosomal DNA (rDNA) from the host and two genes of endosymbiont origin (plastid‐encoded rbcL and nuclear‐encoded SSU rDNA) were determined. The phylogenetic analysis of the host revealed that the marine dinoflagellate P. quinquecorne formed a clade with other diatom‐harbouring dinoflagellates, including Kryptoperidinium foliaceum (Stein) Lindeman, Durinskia baltica (Levander) Carty et Cox and Galeidinium rugatum Tamura et Horiguchi, indicating a single endosymbiotic event for this lineage. Phylogenetic analyses of the endosymbiont in these organisms revealed that the endosymbiont of P. quinquecorne formed a clade with a centric diatom (SSU data indicated it to be closely related to Chaetoceros), whereas the endosymbionts of other three dinoflagellates formed a clade with a pennate diatom. The discrepancy between the host and the endosymbiont phylogenies suggests a secondary replacement of the endosymbiont from a pennate to a centric diatom in P. quinquecorne.  相似文献

The parasitoid nanoflagellate (PNF) Pirsonia diadema is hostspecific for the marine centric diatom Coscinodiscus spp. Experimentsshowed that flagellates significantly prefer C. wailesii overC.granii as host species (interspecific selectivity). This preferencewas independent of light conditions (dark, irradiance of 10and 70 µmol m–2 s–1) and temperature (10 and15C). Diatoms in the Southern Ocean are limited by iron and light, and therefore produce little biomass. Sufficient biomass for analysis under these conditions requires large sample volumes, and diatom samples are therefore often pre-concentrated on a filter. A method for the digestion of diatom cells on polycarbonate filters, that is also suitable for trace metal analysis, is described here. Additional analysis by inductively coupled plasma-optical emission spectroscopy (ICP-OES) is used for the determination of biogenic silicon. Although several procedures were tested, the method of Hauptkorn et al., which uses tetramethylammonium hydroxide for the destruction of silicon is adapted here [Hauptkorn et al. (2001) Fres J Anal Chem 370:246–250]. Additional nitric acid is added to destroy the polycarbonate filters. The described method results in clear digests and a good correlation between cell numbers and silicon content. Using this procedure, the cellular silicon content for Chaetoceros brevis was determined as 86 ± 4 fmol cell−1. For Thalassiosira sp. a sensitivity effect was observed, and silicon content was determined as . The obtained cellular silicon contents are in good agreement with values presented in the literature.  相似文献

The components of the flagellar apparatuses of the male and female gametes of Derbesia tenuissima (De Not.) Crouan are compared with those in other swarmers of green algae. Both the male and female gametes were found to have a cruciate microtubular root system, a non-striated capping plate which connects basal bodies, two electron dense terminal caps which partially cover the proximal end of the basal bodies, and two small system II fibrous roots. Similarities exist between these components and those suggested to be typical of ulvalean swarmers. Based upon these similarities, it is proposed that the Caulerpales be classified in the Ulvaphyceae rather than in the Charophyceae or Chlorophyceae.  相似文献

ABSTRACT. A new amoeba, isolated from well water in Gambia, West Africa, is described and named Phreatamoeba balamuthi n. g., n. sp. Requiring anaerobic conditions for growth, it is easily cultured monoxenically with Escherichia coli or axenically in complex, undefined organic media. Three phenotypes have been observed in the life cycle: an amoeba, a flagellate, and a cyst. The amoeba moves by monopodia, is predominantly multinucleate, and varies from 11 to 160 μm in length. The flagellate has a single flagellum and is from 6 to 50 μm long. The cyst is surrounded by a resistant wall that lacks pores and ranges from 9 to 18 μm in diameter. The transformation from amoeba to flagellate can be induced nutritionally, the exact inducing factor(s) being unknown. Sexual reproduction has not been observed.  相似文献

特马豆克阶是奥陶系底部第一个阶,笔石是特马豆克阶高分辨率地层划分与对比的重要化石类群。江南斜坡带是我国早奥陶世特马豆克期漂浮笔石分异度和丰度最高的相区之一,位于该区的湖南益阳南坝剖面,发育有完整的上特马豆克阶笔石地层,特马豆克阶-弗洛阶界线附近地层连续,上特马豆克阶笔石地层研究已取得较大进展,但下特马豆克阶地层缺乏系统研究。近年来,通过对该剖面笔石标本的不间断采集,新识别出下特马豆克阶笔石带Rhabdinopora flabelliformis parabola带。到目前为止,湖南益阳南坝剖面特马豆克阶可以识别出5个笔石带,自下而上依次为:Rhabdinopora flabelliformis parabola带、Adelograptus tenellus带、Aorograptus victoriae带、Araneograptus murrayi带以及Hunnegraptus copiosus带。基于目前已识别出的笔石带,参考国内外同期笔石地层资料,本文详细讨论华南特马豆克期笔石带序列,并与国内外同期地层进行精确对比。  相似文献
