The amount of asparaginase II in an Escherichia coli wild-type strain (cya+, crp+) markedly increased upon a shift from aerobic to anaerobic growth. However, no such increase occurred in a mutant (cya) lacking cyclic AMP synthesis unless supplemented with exogenous cyclic AMP. Since a mutant (crp) deficient in cyclic AMP receptor protein also did not support the anaerobic formation of this enzyme, it is concluded that the formation of E. coli asparaginase II depends on both cyclic AMP and cyclic AMP receptor protein.  相似文献

The acute actions of low and moderate concentrations of catecholamines on cyclic AMP metabolism in SV40-transformed human lung fibroblasts (VA13) were seriously distorted by non-enzymatic destruction of the agonist. Catecholamine destruction, as measured directly with an isotopic method, was slowed by a variety of anti-oxidants and chelating agents. A combination of two anti-oxidants, ascorbate and thiourea, was very effective in protecting isoproterenol in the cell culture system. That is, there was a 10-fold increase in the sensitivity of VA13 to isoproterenol and the duration of action of the catecholamine was greatly prolonged. However, the anti-oxidants did not alter the responses of the cells to prostaglandins. We conclude that any quantitative studies of cyclic AMP responses to catecholamines must address the question of agonist destruction if meaningful results are to be expected. The use of anti-oxidants, especially the combination of ascorbate and thiourea, would appear to be advisable, particularly in situations where the catecholamine concentrations are less than supramaximal.  相似文献

Treatment of intact adipocytes with either or both insulin and adrenaline stimulated membrane cyclic AMP phosphodiesterase activity only in the endoplasmic reticulum subfraction. The cyclic GMP-inhibited cyclic AMP phosphodiesterase activity was also found in this fraction. Quantitative Western blotting using a specific polyclonal antibody, raised against the homogeneous 'dense-vesicle' cyclic AMP phosphodiesterase from rat liver, identified a single 63 kDa species which was localized in the adipocyte endoplasmic reticulum fraction. The ability of adrenaline to stimulate adipocyte membrane cyclic AMP phosphodiesterase was shown to be mediated via beta-adrenoceptors and not alpha 1-adrenoceptors. Membrane cyclic AMP phosphodiesterase was stimulated by glucagon but not by vasopressin, A23187 or 12-O-tetradecanoylphorbol 13-acetate (TPA). Treatment of adipocytes with either chloroquine or dansyl cadaverine failed to affect the ability of insulin to stimulate cyclic AMP phosphodiesterase activity. Treatment of an isolated adipocyte endoplasmic reticulum membrane fraction with purified protein kinase A increased its cyclic AMP phosphodiesterase activity some 2-fold. When this fraction was treated with purified protein kinase A and [32P]ATP, label was incorporated into a 63 kDa protein which was specifically immunoprecipitated with the antiserum against the liver 'dense-vesicle' cyclic AMP phosphodiesterase.  相似文献

The ascomyceteSaccobolus platensis Gamundi´& Ranalli requires light to produce apothecia. It has now been found that this light requirement can be satisfied by a 24-h pulse of white light at certain stages of the sexual cycle. The addition of exogenousN6,O2′-dibutyryl adenosine 3′,5′-cyclic monophosphate (db-cyclic AMP) to the dark growing mycelia could replace rather efficiently the inductory effect of light; cyclic AMP,N6-monobutyryl cyclic AMP, andO2′-monobutyryl cyclic AMP were less effective, while guanosine 3′,5′-cyclic monophosphate (cyclic GMP) was a very weak inducer. An inducing effect similar to that of db-cyclic AMP was obtained by the addition of 3-isobutyl-1-methylxanthine (MIX) or theophylline to cultures developing in darkness. In the presence of theophylline, endogenous cyclic AMP levels of dark-grown mycelia were several fold higher than those of control cultures. The cyclic AMP content of mycelia growing under different light regimes was measured and no significant differences were observed. However, cultures submitted to white light showed an increase in adenylate cyclase (ATP pyrophosphate-lyase (cyclizing), EC 4.6.1.1) and a decrease in cyclic AMP phosphodiesterase (3′,5′-cyclic AMP 5′-nucleotidohydrolase, EC 3.1.4.17) specific activities compared with the activities of dark-grown mycelia. The cyclic AMP phosphodiesterase activity was strongly inhibited by theophylline and by MIX. The possible role of cyclic AMP in the induction of apothecia in this species is discussed.  相似文献

The effect of cyclic AMP on asexual maturation and gametocyte formation of Plasmodium falciparum grown in vitro was examined over a wide range of concentrations. Cyclic AMP inhibited both processes in a stage-specific manner. Asexual maturation was inhibited from shortly after parasite entry into the red cell through the ring stage. However, trophozoites and schizonts matured normally in the presence of cyclic AMP and produced infectious merozoites. Gametocyte formation was inhibited by 95% when 1.0 mM cyclic AMP was added to synchronously growing parasites in the ring stage of development but was only inhibited by 15% when added in the trophozoite or schizont stages. Cyclic AMP was not found to increase gametocyte formation over a wide range of concentrations.  相似文献

The design, calibration, and operation of a source of controlled amounts of cyclic AMP (c-AMP) are described. Typically, 1.5 s pulses containing 10(10)-10(-12) molecules of c-AMP can be delivered to a region about 10 mum in diameter on an agar plate. The resulting concentration profiles are given as functions of distance and time. The diffusion coefficient of c-AMP in agar was measured to be 0.97 times 10(-5) cm2-s-1 at 21 degrees C.  相似文献

Previous studies have demonstrated that collagen substrates stimulate in vitro somite chondrogenesis, and that agents that elevate intracellular cyclic AMP levels in hibit the ability of somites to respond to the inductive influence of collagen. In the present investigation, radiommunoassay was utilized to compare the cyclic AMP content of somite explants cultured on purified Type I collagen substrates with control explants cultured on Millipore filters. During the period of culture, the cyclic AMP content of collagen-treated explants is significantly lower than the cyclic AMP content of control explants. The cyclic AMP content of collagen-treated explants is 66% of control values as early as one hour following the initiation of culture, and the cyclic AMP content of collagen-treated explants remains lower than controls throughout the 3-day cultured period. The greatest difference in the cyclic AMP content of collagen-treated and control explants is observed at the seventeenth hour of culture, at which time the cyclic AMP content of collagen-treated explants is 56% of controls. These results combined with previous studies provides support for the hypothesis that collagen elicits a reduction in the cyclic AMP content of embroyic somites and that this reduction is necessary to trigger chondrogenic differentiation.  相似文献

Ovariectomized rats were treated with estrogen, progesterone or a combination of estrogen plus progesterone. Rats were anesthetized with sodium pentobarbital and uteri were frozen in situ, uterine extracts were prepared and assayed for cyclic AMP and phosphorylase. Uterine cyclic AMP levels were highest in estrogen treated uteri and were significantly reduced when estrogen was withdrawn for two days. Addition of progesterone to the estrogen regimen for two days or changing from estrogen to progesterone for two days produced results comparable to those obtained when estrogen was withdrawn. Similar experiments were done except that 30 seconds before tissue freezing epinephrine was injected intravenously. Both cyclic AMP and glycogen phosphorylase increased markedly in response to epinephrine. The magnitude of the responses were greatest in the uteri pretreated with estrogen. The magnitudes of both the cyclic AMP and phosphorylase responses were significantly reduced by withdrawing estrogen for two days, by adding progesterone to the estrogen treatment or by changing to progesterone from estrogen. Isoproterenol-stimulated cyclic AMP responses were affected by the steroid state of the uterus in the same way as the epinephrine responses.  相似文献

The classical view of the molecular actions of estrogen is described by its interaction with the intracellular estrogen receptor (ER), the binding of hormone receptor complex to the estrogen response element (ERE) on the DNA and followed by the alterations of gene expressions. Recently it has been reported that membrane estrogen receptor (mER) exist and it is suggested to be G protein linked receptor. In this report we show that under steroid-free culture conditions supplemented with low percentage of charcoal-stripped serum, differential estrogen treatments of human breast cancer MCF7 cells induce different responses of cyclic AMP (cAMP) productions. Treating [2-(3)H]adenine-labeled MCF7 cells with 1 nM estrogen for 30 min stimulates cAMP production by measuring the ratio of [3H]cAMP:Total [3H]adenine nucleotides (ATP+ADP+cAMP), as determined by column chromatography, when compared with the control. This short-term estrogen treatment also significantly enhanced forskolin stimulated cAMP production when compared with the ratio of cAMP/Total measured in cells stimulated with forskolin alone. Pre-treating MCF7 cells with the same concentration of estrogen for 24h before the assay, on the contrary, significantly decreased the basal cAMP level and it also suppressed cAMP production stimulated with forskolin when compared with its respective value under short-term estrogen treatment. Estrogen receptor antagonist ICI 182780 abolished both the stimulatory and suppressive effect of estrogen on cAMP synthesis indicating both effects were mediated through ER. Pre-treating cells with pertussis toxin relieved the suppression of cAMP synthesis by chronic estrogen treatment. Our data suggest that estrogen exerts differential effects on the cAMP production in MCF7 cells, involving the activations Galpha(i) and Galpha(s) family of G proteins, depending on the length of time of hormone treatment.  相似文献

1. 1. The effect of stimulation of adenylate cyclase by pancreozymin-C-octapeptide on the cyclic AMP level of rat pancreatic fragments has been investigated.
2. 2. In normal Krebs-Ringer bicarbonate medium pancreozymin-C-octapeptide causes a slight increase in pancreatic cyclic AMP level; this increase can be considerably enhanced by incubation in a calcium-free incubation medium.
3. 3. The dose-responce curve for pancreazymin-C-octapeptide in calcium-free medium is shifted to lower peptide concentrations, compared to the curve in normal Krebs-Ringer bicarbonate medium.
4. 4. The maximal stimulatory effect of pancreozymin-C-octapeptide id obtained at a 1-methyl-3-isobutylxanthine concentration of 10 mM.
5. 5. It suffices to lower the Ca2+-concentration of the medium from 2.5 to 1.5 mM to get the maximal increase in cyclic AMP content under influence of pancreozymin-C-octapeptide.
6. 6. It is concluded that extracellular calcium antagonizes the stimulation of adenylate cyclase by pancreozymin-C-octapeptide. This suggest that a low cytoplasmic Ca2+-concentration is required for the maximal response of acinar cell adenylate cyclase to pancreozymin.

We reported earlier that reactive oxygen species are implicated in necrotic injury induced by a transient exposure of cultured renal tubular cells to a high concentration of cisplatin but not in apoptosis occurring after continuous exposure to a low concentration of cisplatin. We report here the protective effect of cyclic AMP against cisplatin-induced necrosis in cultured renal tubular cells as well as cisplatin-induced acute renal failure in rats. Several pharmacological agents that stimulate cyclic AMP signaling, including the nonhydrolyzable cyclic AMP analogue dibutyryl cyclic AMP, forskolin, 3-isobutyl-1-methylxanthine, and a prostacyclin analogue, beraprost, prevented cisplatin-induced cell injury in a protein kinase A-dependent manner. Cisplatin enhanced lipid peroxidation, decreased CuZn superoxide dismutase (SOD) while enhancing MnSOD activity, and increased cellular tumor necrosis factor-alpha (TNF-alpha) content. The elevation of TNF-alpha content and cell injury induced by cisplatin were attenuated by p38 mitogen-activated protein kinase (MAPK) inhibitors including SB203580 and PD169316. Indeed, cisplatin increased the number of phosphorylated p38 MAPK-like immunoreactive cells. These intracellular events were all reversed by antioxidants such as N-acetylcysteine (NAC) and glutathione or cyclic AMP analogues. The in vivo acute renal injury after cisplatin injection was associated with the elevation of renal TNF-alpha content. The cisplatin-induced renal injury and the increase in TNF-alpha content were reversed by NAC or beraprost. These findings suggest that cyclic AMP protects renal tubular cells against cisplatin-induced oxidative injury by obliterating reactive oxygen species and subsequent inhibition of TNF-alpha synthesis through blockade of p38 MAPK activation.  相似文献

The inclusion of both dibutyryl cyclic AMP and theophylline in the culture medium of human malignant trophoblast cells (JAr line) for 72 hours results in an enhanced estrogen secretion through the increased specific activity of estrogen synthetase (aromatase), a cytochrome P-450 mono-oxygenase enzyme system. The data described here suggest that this increased aromatase activity is due to an increase in the concentration of only one component of the mono-oxygenase system, cytochrome P-450.  相似文献

In short-term tissue cultures dibutyryl cAMP inhibits the cortisone-induced degranulation of mast cells. The effect of methylxanthine was found to be similar but developed at a slower rate. The increase of cAMP level also affected the maturation of mast cells, as the ratio of cells of mixed granulation increased, compared to the alcian blue- and safranin-present model inhibited degranulation.  相似文献

Intravenously administered cyclic [8-3H]AMP to rats was quickly eliminated from the circulation. After 2 min 93% of the administered radioactivity disappeared from the plasues was recovered mainly in the form of nucleotides, ATP, ADP, AMP and IMP. In vitro contact of cyclic AMP with perfused liver, isolated liver cells and adipose tissue resulted in a rapid breakdown of the nucleotide, presumably on the outer surface of the cells. The degradation products have been identified mainly as adenosine and inosine. Incubation of adipose tissue and isolated liver cells with [3H] AMP also resulted in the breakdown of the nucleotide in themedium. The rate of AMP degradation by these tissues was faster than that for cyclic AMP degradation. The data suggest that cyclic AMP is readily metabolized on the outer surface of cells to products which may be converted within the cells to nucleotides. These findings seem of importance for the quantitative assessments of cellular cyclic AMP outflow during hormonal stimulation.  相似文献

On the basis of the data reported here, one may conclude that although many agents that act in the central nervous system are modulators of the action of cyclic AMP, it is difficult to establish a direct connection between the pharmacologic activity and the levels of cyclic AMP in the brain. This lack of interrelation applies to the benzodiazepines as well as to the pyrazolopyridines. The data for members of the latter group are somewhat frustrating in this regard, since an excellent correlation has been shown to exist between the potency of inhibition of PDE and activity in the antianxiety test. In measurements of steroidogenesis in the isolated adrenal cell, the correlation between activity in vito and the conflict assay is even better. The data presented here and reported elsewhere (Shimizu et al., 1974; Kelly et al., 1974; Mayer and King, 1974; King and Mayer, 1974) provide evidence that agents that act as inhibitors of PDE in cell-free systems exert their influence on cyclic AMP in tissue slices of the brain of guinea pigs by mechanisms that seem not to be related to an effect on PDE. Papaverine, and possibly chlordiazepoxide, may act by releasing agonists that, in turn, stimulate the accumulation of cyclic AMP. This activity is blocked bo other inhibitors of PDE, such as theophyline. Results obtained by the use of platelets are refreshingly clear. Inhibition of aggregation has been shown to occur when the level of cyclic AMP is raised, and a suggestive exists that the most potent inhibitors of platelet PDE are the best potentiators of the action of PGE1 in blocking aggregation. The study utilizing drugs collected from a large number of therapeutic classes makes clear that it is difficult to attribute the mechanism of action for any of the classes studied to modulation of cyclic AMP. An unexpected finding of this study, however, was the fact that pharmacologic agents include an unusually large number of inhibitors of PDE as compared with agents chosen at random. This finding provides a powerful tool for the biochemical pharmacologist who is examining large numbers of compounds in the search for potential drugs.  相似文献
