1. In minesoil reclamation, the establishment of a sustainable plant cover requires the improvement of limiting conditions and the re-initiation of carbon (C) and nutrient cycling.
2. The approach used in this study for reclaiming an abandoned sandpit in Quebec, Canada, was based on a heavy organic amendment as an attempt to accelerate the reconstruction of a functional ecosystem.
3. The one-time intervention consisted of incorporating paper de-inking sludge into soil at two rates (0 and 105 dry t ha–1), supplemented with nitrogen (N) at three rates (3, 6 and 9 g kg–1 sludge) and phosphorus (P) at two rates (0·5 and 1·0 g kg–1 sludge) followed by seeding (mid-summer) of Agropyron elongatum (Host) Beauv. (tall wheatgrass).
4. Standing biomass increased in the presence of sludge after both the first and second full growing seasons. High N application rates further increased yield, more importantly in the second season. The high P rate improved grass establishment in all cases. Ground cover increased with time and doubled in the presence of sludge whereas it declined in the absence of sludge. Phosphorus and N uptake was improved consistently in the presence of sludge.
5. Sludge application resulted in improved water retention and cation exchange capacities, and an increase in pH and bulk density of sandpit minesoil, all of which may have accounted for the significant improvement in plant responses. Levels of soil C and N suggest that this reconstructed system is approaching sustainability.
6. Adequate N and P supplements will accentuate the positive influence of sludge on revegetation.  相似文献

1. It is generally agreed that the blackening of osmium tetroxide by unsaturated lipid is too unpredictable to demonstrate lipid in tissues.
2. At neutral pH osmium tetroxide combines with the double bonds in the lipoproteins of cellular membranes (mitochondria, etc.) and the deep colour reaction of ethyl gallate with this osmium provides good staining of lipid for the light microscope.
3. Osmium taken up by tissue proteins at neutral pH is only a small fraction of that taken up by the lipid. (After acid fixatives osmium tetroxide is a general protein stain.)
4. The uptake of Sudan black B by partition from dilute solution is a specific test for lipid, but in normally fixed tissue most of the structural lipid is 'bound' and is not accessible to the dye.
5. Cautious treatment of fixed tissue with dilute sodium hypochlorite will unmask this lipid for viewing by the light microscope.
6. Direct fixation with neutral osmium tetroxide is an effective method for visualizing lipid for the electron microscope (as in the ethyl gallate method for the light microscope). But the poor penetration of osmium limits its use in this way.
7. After formol/glutaraldehyde fixation much of the lipid in the tissues is 'bound' and does not take up osmium. It can be unmasked by a saturated aqueous solution of thymol.
8. The unmasked lipid can then be rendered more osmiophil by partition in a solution of the highly unsaturated terpene farnesol, thus increasing the uptake of osmium in a renewed application.
9. Some of the novel observations on tissue lipids made by these methods are reviewed.  相似文献

PROPERTIES OF MEMBRANE-BOUND HEXOKINASE IN RAT BRAIN   总被引:2,自引:0,他引:2  
Abstract— —-(1) The total hexokinase activity present in the mitochondrial fraction can be solubilized completely by incubation with salt and Triton X-100. This activity cannot be entirely released by washing with sucrose or by freezing and thawing.
(2) A part of the particle bound hexokinase exists in a latent form. The latent form is apparent after incubation with high salt concentrations, detergents or by freezing and thawing.
(3) Solubilization of membrane bound hexokinase is pH-dependent. Incubation in salt solutions increases the specific activity ten-fold. The salt concentration and pH are con-current. At pH 7.0 part of the hexokinase is solubilized. The lower the pH the less salt is required to release the same amount of activity.
(4) Triton X-100 solubilizes particle-bound hexokinase, but to a less extent than salts. The activation of hexokinase is greater with Triton X-100 than with salt.
(5) The possible nature of the bonds between hexokinase and mitochondrial membranes is discussed.  相似文献

The effect of pH on the efficiency of an SBR processing piggery wastewater   总被引:1,自引:0,他引:1  
To treat piggery wastewater efficiently, the hydrolysis of urea (mainly derived from swine urine) in piggery wastewater with the change of sewage pH must be considered. Using activated sludge, piggery wastewater was treated in a sequencing batch reactor (SBR), and the effects of influent pH on SBR processing efficiency, sludge settle ability, and sludge activity were investigated. The results showed that a high influent pH value contributed to the improvement of the removal rate of ammonia nitrogen and reduction of the chemical oxygen demand (COD). When the influent pH was between 9.0 and 9.5, the removal rate of ammonia nitrogen was higher than 90%, and the reduction of COD from its original value was 80%. The influent pH had a greater influence on sludge concentration and sludge activity. When the influent pH increased from 7.0 to 9.5, the sludge concentration increased from 2,350 to 3,947 mg/L in the reactor, and the activities of ammonium oxidizing bacteria (AOB) and nitrite oxidizing bacteria (NOB) first increased and then decreased. When the influent pH was 9.0 and 8.0, the maximum values (0.48 g O2/(g MLSS/day) and 0.080 g O2/(g MLSS/day)) were reached, and the sludge settling ratio was nearly steady between 20 and 35% in each reactor.  相似文献

THE EFFECT OF ADDED NITROGEN ON THE RATE OF DECOMPOSITION OF ORGANIC MATTER   总被引:17,自引:0,他引:17  
(1) N added to decomposing organic matter often has no effect or a negative effect on microbial activity, at least in the long term. More than 60 papers are cited in support of this statement.
(2) The negative effect of N is mainly found with recalcitrant organic matter with a high C/N ratio (straw, wood, etc.), whereas a positive effect of N is common for easily degradable organic material with low C/N ratio.
(3) The negative effect of N could be explained by: (i) N disturbs the outcome of competition between potent and less potent decomposers; (ii) through 'ammonia metabolite repression', N blocks production of certain enzymes, at least in basidiomycetes, and enhances breakdown of the most available cellulose, whereby recalcitrant lignocellulose accumulates; (iii) amino compounds condense with polyphenols and other decomposition products, forming 'browning precursors' which are toxic or inhibitory.
(4) The effect of adding N may depend on the microflora present.
(5) There are indications that some microorganisms have a 'luxury uptake' of N when it is present in sufficient amounts, thereby delaying N mineralization.
(6) The addition of N seems to increase the formation of water-soluble , brown, recalcitrant compounds, but to decrease the amount of humus formed.  相似文献

The objective of this study was to assess the effects of soil pH adjustment and treatment with sludge on plant growth and cadmium (Cd) accumulation in Chinese cabbage (pak choi) grown in Cd-contaminated soils. Soil was artificially spiked to raise its final Cd concentration to 10 mg/kg, adjusted to different pH values, and then supplemented with 5% sludge: either biosolids (BS) from wastewater treatment or sludge from drinking water treatment (WTS). Pak choi seeds were sown and the seedlings potted in the treated soil, then grown in a greenhouse for 45 days. The added Cd was present mainly in the exchangeable fraction and its concentration correlated to Cd accumulation in the roots and shoots. Adjustment of pH and application of sludge affected the Cd concentration in different fractions. Addition of WTS and adjustment to pH 7.0 inhibited growth and Cd accumulation in the pak choi. Addition of BS increased the soil nutrient content and thus enhanced growth and Cd accumulation. If pak choi were the only form of vegetable intake, consumption of pak choi grown in the Cd-spiked soils used in this study would exceed the maximum acceptable intake (60 μg/day) proposed by the World Health Organization.  相似文献

1. The hydrolysis of gelatin at a constant hydrogen ion concentration follows the course of a monomolecular reaction for about one-third of the reaction. 2. If the hydrogen ion concentration is not kept constant the amount of hydrolysis in certain ranges of acidity is proportional to the square root of the time (Schütz''s rule). 3. The velocity of hydrolysis in strongly acid solution (pH less than 2.0) is directly proportional to the hydrogen ion concentration as determined by the hydrogen electrode i.e., the "activity;" it is not proportional to the hydrogen ion concentration as determined by the conductivity ratio. 4. The addition of neutral salts increases the velocity of hydrolysis and the hydrogen ion concentration (as determined by the hydrogen electrode) to approximately the same extent. 5. The velocity in strongly alkaline solutions (pH greater than 10) is directly proportional to the hydroxyl ion concentration. 6. Between pH 2.0 and pH 10.0 the rate of hydrolysis is approximately constant and very much greater than would be calculated from the hydrogen and hydroxyl ion concentration. This may be roughly accounted for by the assumption that the uncombined gelatin hydrolyzes much more rapidly than the gelatin salt.  相似文献

1. Comparisons have been made between the action of rubidium sulphate and palladium chloride on the growth of barley, wheat, oats peas and beans in complete nutrient solutions.
2. Over a wide range of concentrations rubidium sulphate was not found to exercise either a beneficial or a harmful action on the growth of any of the species tested. The germination of the seeds was likewise not affected.
3. No benefit was derived from palladium chloride , but at a comparatively low concentration a harmful action occurred which became more intense with increasing concentration. Stunting of the main root and laterals was a characteristic feature of this toxicity. With the lower concentrations the check was temporary, and the roots eventually made normal growth, as good as that in the control plants. With increasing amounts of pallahum chloride the poisoning effect became more persistent, until a concentration was reached which did not allow of any root or shoot recovery.
4. The tolerance of palladium varies with the species, as was indicated b. the measure of recovery. Barley appeared to be the least, and oats the most sensitive of the three cereals tested. Peas responded at much the same concentrations as barley, but broad beans made so complete a recovery from the initial checking that the dry weights were ultimately not reduced even by the strongest concentration tested, the plants being indistinguishable from the controls.
5. The effect of palladium poisoning was similar whether the seeds were germinated in the presence of palladium or whether the seedlings were not introduced to it until they were about a week old.  相似文献

Abstract— (1) Swelling of synaptosomes was measured spectrophotometrically by recording changes in extinction at 520 nm.
(2) Synaptosomes behaved as osmometers in NaCl solutions. When the tonicity of the medium was changed, synaptosome volume changed in accordance with Boyle and van't Hoff's Law. These changes were reversed on restoring the tonicity of the medium.
(3) The rate at which a solute entered the synaptosome was determined from the rate of swelling in the presence of that solute. Permeability of synaptosomes to non-electrolytes was in the order glucose ≪ glycerol < thiourea = formamide < propylene glycol = dimethylsulphoxide.
(4) Synaptosomes were freely permeable to ammonium and acetate ions and impermeable to Ca2+, Mg2+, PO42−, SO42− and oxalate ions.  相似文献

Glucocorticoids induce an alteration of the surface of hepatoma tissue culture (HTC) cells as expressed by changes in cell electrophoretic, antigenic, and adhesive properties. The alteration is assayed by the increased adhesiveness of induced cells for a glass surface. The induction process has a lag period of about 3 hr and attains a plateau level after 24–30 hr when 50–80% of the steroid-treated cells are firmly adhered. Less than 10% of untreated cells adhere under the same conditions. Induction is inhibited by actinomycin D and cycloheximide, demonstrates both pH and temperature dependence, and responds to changes in steroid concentration and structure. By contrast, the attachment per se of preinduced cells is not affected by inhibitors of RNA and protein synthesis, fluctuations of temperature and pH, and the presence or absence of the hormone. When the induction process is reversed by removal of steroid or addition of actinomycin D, preinduced adhesiveness is lost with a half-life of 13–24 hr, but in the presence of cycloheximide the loss is accelerated (t1/2 3–5.5 hr). These results suggest that glucocorticoids induce the biosynthesis of a protein which either modifies the cell surface (an enzyme) or is incorporated into surface structures (structural protein).  相似文献

Abstract— The effect of excess leucine in the diet on serotonin metabolism in the brain was investigated in experimental animals. It was found that:
(1) Animals receiving diets containing 3 % and 8 % leucine and those receiving jowar diets had significantly lower levels of serotonin in the brain.
(2) Intraperitoneal administration of the precursor amino acid 5-HTP increased the serotonin concentration in brain in both control and leucine-fed animals. However, the serotonin concentration in leucine-fed animals was significantly lower than that of pairfed controls. Larger amounts of the synthesized serotonin were found to be catabolized in 3 hr in leucine-fed animals than in control animals.
(3) The in vitro uptake of [14C]5-HTP by brain slices of animals fed leucine was found to be similar to that of control animals.
(4) The basal concentration of 5-HIAA in brain was higher in leucine-fed animals, suggesting a higher rate of catabolism of serotonin.
(5) Administration of nicotinic acid resulted in a further fall of serotonin concentration in the brains of leucine-fed animals but not in control animals.  相似文献

The toxicity of solutions of ammonia to fish in different dilution waters showed a variation which was not entirely related to the concentration of the un-ionized ammonia molecule. Evidence is presented which shows that this variation can be attributed to the increase in the concentration of free carbon dioxide at the gill surface, which causes a decrease in both the pH value and the concentration of unionized ammonia present; the extent of the decrease depends on the initial concentration of free carbon dioxide in the bulk of the solution. Quantitative measurements of the relation between the free carbon dioxide concentration of the water and the toxicity of un-ionized ammonia are correlated with a theoretical estimation of the pH value of the water at the gill surface.  相似文献

SUMMARY. 1. Filamentous algae in flowing freshwaters can represent a spatially and temporally distinct sub-habitat for epiphytic diatom communities. This sub-habitat is described in a low discharge, spring-fed stream with extensive filamentous green algal mats, and in a tuft of Cladophora glomerata (L.) Kützing from a large river.
2. Oxygen micro-electrodes, a thermistor current velocity probe, a standard pH probe and water chemistry were employed to assess spatial heterogeneity. Temporal patterns of epiphyte colonization were evaluated on filamentous artificial substrates.
3. There were steep spatial gradients in the low discharge stream. At mid-day, O2 ranged from 0–1.5 times air-saturated O2 concentrations, pH varied from 7.25–8.0, and current velocity spanned 0–0.5 m s−1. Areas near the surface of algal mats had high O2, pH and current velocity. These patterns were correlated with epiphyte community structure.
4. In the interior of C. glomerata tufts O2 concentration was raised and current velocity depressed compared to the surrounding water, even when external current velocity was as high as 0.4 m s−1.
5. After thirty-five days of colonization of artificial substrate in the low discharge environment, epiphyte communities were similar to those on filamentous atgae. Epiphyte diversity on artificial substrates subsequently decreased compared to natural substrates as did the similarity between the types on substrates, suggesting that microscale renewal of epiphyte habitat (growth of filamentous algae) maintains high epiphytic diversity.  相似文献

林地利用被认为是污泥资源化利用的重要方式,但施用污泥对林木根系生长的影响报道较少。本研究通过根箱试验,分析表施和混施10%污泥对速生树种团花不同土层根系形态、土壤pH值和电导率动态变化及根系重金属含量的影响,并拟合土壤pH值、电导率和根系重金属含量与根长的关系。结果表明: 与不施污泥(对照)相比,混施污泥显著抑制了团花根长、根表面积和根体积增长,混施污泥120和240 d后,0~20 cm土层总根长分别为不施污泥的76.9%和67.4%;表施污泥对团花根长和根表面积的影响不显著,但显著提高了根体积。混施污泥显著提高了土壤pH值和电导率及根系重金属含量,混施污泥0~20和20~40 cm土层根系镉含量分别是不施污泥的11.5和10.0倍。线性回归拟合分析显示,不同处理0~20 cm土层的电导率与根长均呈显著负相关;表施和混施污泥根系镉含量与根长呈极显著负相关。上述结果表明,混施污泥抑制了团花根系生长,这可能是由于混施污泥提高了土壤电导率和根系镉含量所致,而表施污泥对团花根系生长的作用不明显。  相似文献

Wastewater sludges have been proposed as an effective media for the production of rhizobia. The effect of total suspended solid (TSS) concentrations and pretreatments of sludge on the growth of Sinorhizobium meliloti were investigated. Acid (pH 2.0-6.0 obtained with H2SO4) and alkaline (50-200 mequiv.wt./L of NaOH) treatments were applied to enhance the biodegradability of primary (0.325%-3.2% TSS obtained by dilution of original sample) and secondary (0.2%-0.4% TSS obtained by concentration of original sample) sludges. In primary sludge without pretreatment, the highest cell count (11.10 x 10(9) cfu/mL) was obtained with 1.3% TSS. However, a maximum cell count of 13.00 x 10(9) cfu/mL was reached using an acid treatment of pH 2.0 and a 0.325% TSS concentration. Moreover, the alkaline treatment with 100 mequiv.wt./L of NaOH and 0.65% TSS increased the cell yield to 21.00 x 10(9) cfu/mL. For secondary sludge without pretreatment, no enhancement of growth was observed while increasing TSS concentration. This may be due to the increase of inhibitory substances, such as heavy metals, and of the Ca and Mg concentrations. As in primary sludge, some acid and alkaline treatments of secondary sludge tend to improve the cell count of S. meliloti. However, the highest value of 9.80 x 10(9) cfu/mL obtained with 0.4% TSS at pH 2.0 was lower than that obtained with primary sludge. It was also observed that S. meliloti grown in treated sludges maintained its capacity to nodulate alfalfa.  相似文献

Inhibition of the fermentation of acetate to methane and carbon dioxide by acetate was analyzed with an acetate-acclimatized sludge and with Methanosarcina barkeri Fusaro under mesophilic conditions. A second-order substrate inhibition model, q(ch(4) ) = q(m)S/[K(s) + S + (S/K(i))], where S was the concentration of undissociated acetic acid, not ionized acetic acid, could be applicable in both cases. The analysis resulted in substrate saturation constants, K(s), of 4.0 muM for the acclimatized sludge and 104 muM for M. barkeri. The threshold concentrations of undissociated acetic acid when no further acetate utilization was observed were 0.078 muM (pH 7.50) for the acclimatized sludge and 4.43 muM (pH 7.45) for M. barkeri. These kinetic results suggested that the concentration of undissociated acetic acid became a key factor governing the actual threshold acetate concentration for acetate utilization and that the acclimatized sludge in which Methanothrix spp. appeared dominant could utilize acetate better and survive at a lower concentration of undissociated acetic acid than could M. barkeri.  相似文献

Dehydrogenase activity assay of activated sludge using the redox dye 2-(p-iodophenyl)-3-(p-nitrophenyl)-5-phenyltetrazolium chloride (INT) was investigated. INT-dehydrogenase activity (INT-DHA) was directly proportional to INT dosage and inversely proportional to bio-mass concentration over limited ranges. INT dosages exceeding 2.5m/M were toxic to dilute activated sludge suspensions. INT-DHA was greatest near pH 9, whereas the peak oxygen uptake rate (OUR) occurred at pH 8. Both INT-DHA and OUR varied inversely with sludge age, but INT-DHA was the more sensitive of the two parameters to this variable. Consistently good and highly significant correlations between INT-DHA and OUR of chlorine stressed activated sludge were found at sludge ages ranging 2.2-7.0 days.  相似文献

.1. A study of individually marked Fulmars during three seasons has shown that both sexes can breed in successive years. The evidence for the theory of intermittent breeding is examined and definitions are given of the various categories of birds which compose the population during the breeding season. It is suggested that the supposed non-breeding adults which are seen, often in moult, in July are failed-breeders which have lost their egg or nestling.
2. No marked individual changed its nest-site between years, although the number and identity of sites occupied each July varied considerably.
3. No changes of mate occurred among the six marked pairs which nested in two consecutive years.
4. There is some evidence that both parents remain near the colony during the week before and after hatching.
5. Analysis of the 90 known sites of a colony subject to avian but not wild mammalian predation has revealed a wide range of habitat selection and a strong tendency toward close concentration of nests.
6. There is some evidence that the breeding season, as measured by date of hatching, varies from year to year.  相似文献

1. From 1954 to 1956 inclusive, the biology of individual marked Kittiwakes was studied at North Shields, Northumberland.
2. It was concluded that older Kittiwakes reacted to the breeding stimulus earlier, more intensively and with greater success than younger breeding birds.
3. Birds with previous breeding experience returned to the colony before birds breeding for the first time and these before non-breeders.
4. Before breeding started, birds which had bred previously spent more of their time at the colony than those about to breed for the first time.
5. Birds breeding for at least the second time laid the first egg 7·5 days earlier than those breeding for the first time.
6. Breeding started one day later for every four days the return to the colony was postponed.
7. Older breeding birds showed greater nest-site tenacity, laid larger clutches and had greater breeding success than younger birds.
8. The chicks in broods of two (but not of one) increased in weight more rapidly where the parents had previous experience.
9. Breeding Kittiwakes showed strong colony tenacity, but 24% of the marked non-breeding birds were subsequently seen in other colonies.
