Ribosomal RNA (rRNA) in vertebrates is initially transcribed as a single 47S precursor which is modified by the addition of 2'-O-methyl ribose moieties, pseudouridines, and methyl groups, followed by cleavage at several sites to produce the mature 28S, 18S, and 5.8S rRNAs. Cleavage of the rRNA precursor to generate the 18S rRNA is mediated by a ribonucleoprotein (RNP) complex termed the processome containing U3, a box C/D small nucleolar RNA (snoRNA), and at least 28 cellular proteins. We previously identified a novel human RNA binding protein, NF-kappaB binding protein (NFBP), which is the human homolog of Rrp5p, a protein component of the yeast U3 processome. Here, we show that NFBP colocalizes with and coprecipitates U3 in the nucleolus. We also demonstrate that NFBP is essential for the generation of 18S rRNA as maturation of the 18S rRNA is repressed in the absence of NFBP. Using Northern blot analyses, we further show that NFBP is specifically necessary for cleavages at sites A0, 1, and 2, as unprocessed intermediate forms of rRNA accumulated in the absence of NFBP.  相似文献

Summary Isoelectric focussing in alkaline pH gels has permitted the identification of a new homoeoallelic series of genes,Est-6, encoding grain esterases in bread wheat,Triticum aestivum. Nullisomic analysis located these genes to the short arms of the homoeologous group 2 chromosomes. A search for polymorphism withinEst-6 revealed null alleles at each ofEst-A6,Est-B6 andEst-D6. TheLy-6 locus is now regarded as a gene complex consisting of at least five closely linked loci (Ly-6A-Ly-6E) whose polymorphic products are identified by monoclonal antibodies and distinguished by different tissue distributions.Ly-6 has been assigned by other investigators to chromosome (Chr) 9 (linked toThy-1 or to Chr 2. We report that theLy-6 gene complex, together with theXp-14 andGdc -1 loci, is situated on Chr 15 linked toGpt1. These new linkage data are derived from four sources: (1) three separate crosses that failed to demonstrate linkage ofLy-6 to eitherThy-4 on Chr 9 or to any of five genes present on Chr 2; (2) the NXSM recombinant inbred strains, which suggested the linkage ofLy-6 andXp-14 toGpt-1 on Chr 15; (3) severalGpt-1 andGdc-1 congenic strains that confirmed the assignment ofLy-6 andXp-14 to Chr 15; and (4) backcrosses that further confirmed the linkage ofLy-6, Gpt-1, Gdc-4, andXp-14, the probable gene order beingGpt-11/Ly-6 Xp-14-Gdc-1.  相似文献

OSH6 (Oryza sativa Homeobox6) is an ortholog of lg3 (Liguleless3) in maize. We generated a novel allele, termed OSH6-Ds, by inserting a defective Ds element into the third exon of OSH6, which resulted in a truncated OSH6 mRNA. The truncated mRNA was expressed ectopically in leaf tissues and encoded the N-terminal region of OSH6, which includes the KNOX1 and partial KNOX2 subdomains. This recessive mutant showed outgrowth of bracts or produced leaves at the basal node of the panicle. These phenotypes distinguished it from the OSH6 transgene whose ectopic expression led to a “blade to sheath transformation” phenotype at the midrib region of leaves, similar to that seen in dominant Lg3 mutants. Expression of a similar truncated OSH6 cDNA from the 35S promoter (35S::ΔOSH6) confirmed that the ectopic expression of this product was responsible for the aberrant bract development. These data suggest that OSH6-Ds interferes with a developmental mechanism involved in bract differentiation, especially at the basal nodes of panicles. SKU5-Similar 6 (SKS6) is a one of a large gene family of 19 members in Arabidopsis thaliana (L.) Heynh that encode multicopper oxidase-like proteins that are related to ferroxidases, ascorbate oxidases and laccases. Only one member of the family has been previously studied; Skewed5 (SKU5) is involved in the control of root growth. The encoded SKS6 protein, like SKU5 appears to lack a functional copper-binding site and is most closely related to Bp10 from Brassica napus and Ntp303 from Nicotiana tobacum. The SKS6 promoter contains many putative regulatory sites and differential expression of an SKS6::GUS reporter gene revealed selective induction in several seedling tissues including guard cells, root cortex cells, and leaf margin hydathodes. It was also expressed later in flower development in flower primordia, ovules, and the abscission zones of seeds and siliques. Furthermore, SKS6 was upregulated in roots in response to treatment of seedlings with the hormones abscisic acid, indole-3 acetic acid, 2,4-dichlorophenoxyacetic acid and aminocyclopropane-1-carboxylate. A loss-of function sks6-1 T-DNA insertion allele revealed that cotyledon vascular patterning is affected in the mutant, suggesting a role for the protein in metabolism of nutrients or hormones in the hydathodes, the sites of auxin synthesis and chemical recycling.  相似文献

The comparison of results obtained by different separation and staining techniques permits the definition of esterase-6 in comparison with esterase-9 and a new esterase, esterase-20. Alleles of Es-6 affect the product's ability to aggregate. Esterase-20 may be an aggregated product of Es-9. The close linkage of Es-6 and Es-9 is confirmed. Homology of esterase-6 with esterases from other mammalian species is also suggested.HRN was supported by the Medical Research Council.  Sequence analysis of a newly isolated Hirudo medicinalis cDNA containing an Antennapedia (Antp)-class homeobox suggests that the corresponding gene, Lox6, is an ortholog of the Drosophila Deformed (Dfd) gene. In situ hybridization of whole-mounted preparations shows that the major sites of Lox6 expression during embryogenesis are the central nervous system (CNS) and the peripheral sensory system. Lox6 mRNA can be detected in a subset of neurons in each ganglion from the subesophageal ganglion (RG2) to the most posterior ganglion, with the highest level of expression seen in RG3. Peripherally, Lox6 is expressed principally in the primordia of the sensillae and in the eyes. Previous research has identified a relationship between the rate of dissipation of turbulent kinetic energy, , and the mass-transfer-limited rate of uptake by a surface, herein called the 1/4 law, and suggests this law may be applicable to nutrient uptake on coral reefs. To test this suggestion, nitrate uptake rate and gravitational potential energy loss have been measured for a section of Warraber Island reef flat, Torres Strait, northern Australia. The reef flat section is 3 km long, with a 3 m tidal range, and on the days measured, subject to 6 m s–1 tradewinds. The measured nitrate uptake coefficient, S , on two consecutive days during the rising tide was 1.23±0.28 and 1.42±0.52×10–4 m s–1. The measured loss of gravitational potential energy across the reef flat, GPE , on the same rising tides over a 178 m section was 208±24 and 161±20 kg m–1 s–2. Assuming the GPE is dissipated as turbulent kinetic energy in the water column, and using the 1/4 law, the mass-transfer-limited nitrate uptake coefficient, SMTL , on the two days was 1.57±0.03 and 1.45±0.04×10–4 m s–1. Nitrate uptake on Warraber Island reef flat is close to the mass-transfer limit, and is determined by oceanographic nitrate concentrations and energy climate.Communicated by B.C. Hatcher  相似文献

【目的】东方蜜蜂微孢子虫(Nosme ceranae)专性侵染成年蜜蜂导致微孢子虫病,给养蜂生产造成很大损失。目前,东方蜜蜂微孢子虫的N6-腺苷特异性甲基化转移酶(N6-adenine-specific methyltransferase,N6AMT)基因NcN6AMT的研究仍然缺失。本研究对NcN6AMT的编码序列(coding sequence,CDS)区进行克隆,并解析NcN6AMT蛋白的理化性质和分子特性,进而测定东方蜜蜂微孢子虫侵染意大利蜜蜂(Apis mellifera ligustica)和中华蜜蜂(Apis cerana cerana)工蜂过程中NcN6AMT的相对表达量,以期丰富NcN6AMT的信息,并为探究东方蜜蜂微孢子虫侵染过程NcN6AMT的功能及表观调控机制提供基础。【方法】采用Protparam和ProtScale软件对NcN6AMT进行等电点和亲水性分析。通过SignalP 5.0、NetPhos 3.1、TMHMM-2.0、SOPMA和SWISS-MODEL等软件分别预测NcN6AMT的信号肽、磷酸化位点、跨膜结构域、二级结构和三级结构。使用WoLF PSORT II软件预测NcN6AMT的亚细胞定位。根据N6AMT氨基酸序列,通过TBtools软件对智人(Homo sapiens)、小鼠(Mus musculus)、褐飞虱(Nilaparvata lugens)、兔脑炎微孢子虫(Encephalitozoon cuniculi)、肠脑炎微孢子虫(Encephalitozoon intestinalis ATCC 50506)、蚱蜢脑炎微孢子虫(Encephalitozoon romaleae SJ-2008)、美洲思普雷格孢虫(Spraguea lophii 42_110)、家蚕微孢子虫(Nosema bombycis CQ1)、隐生菱形藻(Nitzschia inconspicua)和东方蜜蜂微孢子虫(Nosema ceranae)的N6AMT进行结构域预测和分析。利用MEME软件和MEGA 11.0软件进行东方蜜蜂微孢子虫和其他物种N6AMT的保守基序预测及进化树构建。采用实时荧光定量聚合酶链式反应(real-time fluorescence quantitative polymerase chain reaction,RT-qPCR)检测NcN6AMT在东方蜜蜂微孢子虫侵染意大利蜜蜂和中华蜜蜂工蜂过程的相对表达量。【结果】通过PCR扩增出大小约500 bp的目的片段,克隆测序结果显示其与GenBank数据库收录的预测序列一致;NcN6AMT蛋白的分子量约为18.7 kDa,分子式为C845H1374N214O249S6,理论等电点为5.88,脂溶系数是119.76,不稳定系数为37.47,平均亲水系数为0.025,含166个氨基酸和15个磷酸化位点,不含典型的跨膜结构域和信号肽,可同时定位于细胞质、线粒体、细胞核和液泡膜;NcN6AMT含1个甲基转移酶小结构域(methyltransferase small domain,MTS),该结构域同样存在于家蚕微孢子虫和兔脑炎微孢子虫等8个其他物种的N6AMT;在东方蜜蜂微孢子虫、兔脑炎微孢子虫、肠脑炎微孢子虫和蚱蜢脑炎微孢子虫的N6AMT中均预测到5个相同的保守基序;NcN6AMT与家蚕微孢子虫、肠脑炎微孢子虫、兔脑炎微孢子虫和蚱蜢脑炎微孢子虫的N6AMT序列一致性达到70.92%;东方蜜蜂微孢子虫和家蚕微孢子虫的N6AMT在系统进化树上聚为一支;东方蜜蜂微孢子虫接种后1–4 d,NcN6AMT在意大利蜜蜂和中华蜜蜂工蜂中肠内均呈现先上升后下降的表达趋势。【结论】成功克隆到NcN6AMT基因的CDS区,明确了NcN6AMT蛋白的理化性质和分子特性,并揭示东方蜜蜂微孢子虫和家蚕微孢子虫的N6AMT蛋白具有较高的保守性,NcN6AMT在东方蜜蜂微孢子虫侵染意大利蜜蜂和中华蜜蜂工蜂的第一个增殖周期(1–4 dpi)内动态表达且均呈上升-下降的表达模式。  相似文献

Broccoli (Brassica oleracea var. italica) deteriorates rapidly following harvest. The two plant hormones ethylene and cytokinin are known to act antagonistically on harvest-induced senescence in broccoli: ethylene by accelerating the process, and cytokinin by delaying it. To determine the level at which these hormones influenced senescence, we isolated and monitored the expression of genes normally associated with senescence in broccoli florets treated with exogenous 6-benzyl aminopurine (6-BAP), 1-aminocyclopropane-1-carboxylic acid (ACC), a combination of 6-BAP and ACC, and sucrose, in the five days following harvest. Exogenous 6-BAP caused both a reduction (BoACO) and an increase (BoACS) in ethylene biosynthetic gene expression. The expression of genes used as senescence markers, BoCP5 and BoMT1, was reduced, whereas BoCAB1 levels were maintained after harvest in response to exogenous 6-BAP. In addition, the expression of genes encoding sucrose transporters (BoSUC1 and BoSUC2) and carbohydrate metabolizing enzymes (BoINV1 and BoHK1) was also reduced upon 6-BAP feeding. Interestingly, the addition of ACC prevented the 6-BAP-induced increase in expression of BoACS, but 6-BAP negated the ACC-induced increase in expression of BoACO. The culmination of these results indicates a significant role for cytokinin in the delay of senescence. The implication that cytokinin regulates postharvest senescence in broccoli by inhibiting ethylene perception and/or biosynthesis, thus regulating carbohydrate transport and metabolism, as well as senescence-associated gene expression, is discussed and a model presented.  相似文献
