Expenses for marker assays are the major costs in marker-assisted backcrossing programs for the transfer of target genes from a donor into the genetic background of a recipient genotype. Our objectives were to (1) investigate the effect of employing sequentially increasing marker densities over backcross generations on the recurrent parent genome (RPG) recovery and the number of marker data points (MDP) required, and (2) determine optimum designs for attaining RPG thresholds of 93–98% with a minimum number of MDP. We simulated the introgression of one dominant target gene for genome models of sugar beet (Beta vulgaris L.) and maize (Zea mays L.) with varying marker distances of 5–80 cM and population sizes of 30–250 plants across BC1 to BC3 generations. Employing less dense maps in early backcross generations resulted in savings of over 50% in the number of required MDP compared with using a constant set of markers and was accompanied only by small reductions in the attained RPG values. The optimum designs were characterized by increasing marker densities and increasing population sizes in advanced generations for both genome models. We conclude that increasing simultaneously the marker density and the population size from early to advanced backcross generations results in gene introgression with a minimum number of required MDP.  相似文献

The efficiency of marker-assisted introgression in backcross populations derived from inbred lines was investigated by simulation. Background genotypes were simulated assuming that a genetic model of many genes of small effects in coupling phase explains the observed breed difference and variance in backcross populations. Markers were efficient in introgression backcross programs for simultaneously introgressing an allele and selecting for the desired genomic background. Using a marker spacing of 10-20 cM gave an advantage of one to two backcross generations selection relative to random or phenotypic selection. When the position of the gene to be introgressed is uncertain, for example because its position was estimated from a trait gene mapping experiment, a chromosome segment should be introgressed that is likely to include the allele of interest. Even for relatively precisely mapped quantitative trait loci, flanking markers or marker haplotypes should cover ~10-20 cM around the estimated position of the gene, to ensure that the allele frequency does not decline in later backcross generations.  相似文献

This study investigated a marker-assisted introgression programme in Australian Merino sheep. The goal was to introgress an allele with a large negative effect on fibre diameter into a Merino flock possessing medium average fibre diameter. The influence of two factors was explored: the strategy used to select animals from the purebred and backcross line for backcrossing purposes and the use of selection on background markers to accelerate the return to the purebred line''s genome. The results were compared to introgression based on EBVs only. Introgression using EBVs only produced almost the same response in the dollar index as marker-based introgression methods. However, this study did not account for some of the costs associated with implementing the programmes, including the costs of phenotyping and genotyping. Given that the cost of measuring fibre diameter is low, it was concluded that introgression on EBVs only would be the preferred method since the marginal profit of marker-assisted introgression would not be large enough to cover the additional cost of genotyping. In marker-assisted introgression, reciprocal crossing of male and female selection candidates from the backcross and the purebred line was the most advantageous strategy from a practical and profit point of view. Selection for background markers was less profitable in this study than recovering the donor genome by selection on phenotype.  相似文献

Marker-assisted backcrossing is routinely applied in breeding programs for gene introgression. While selection theory is the most important tool for the design of breeding programs for improvement of quantitative characters, no general selection theory is available for marker-assisted backcrossing. In this treatise, we develop a theory for marker-assisted selection for the proportion of the genome originating from the recurrent parent in a backcross program, carried out after preselection for the target gene(s). Our objectives were to (i) predict response to selection and (ii) give criteria for selecting the most promising backcross individuals for further backcrossing or selfing. Prediction of response to selection is based on the marker linkage map and the marker genotype of the parent(s) of the backcross population. In comparison to standard normal distribution selection theory, the main advantage of our approach is that it considers the reduction of the variance in the donor genome proportion due to selection. The developed selection criteria take into account the marker genotype of the candidates and consider whether these will be used for selfing or backcrossing. Prediction of response to selection is illustrated for model genomes of maize and sugar beet. Selection of promising individuals is illustrated with experimental data from sugar beet. The presented approach can assist geneticists and breeders in the efficient design of gene introgression programs.  相似文献

Computer simulations can be employed to find optimal procedures for developing introgression libraries in rye with marker-assisted backcrossing. Our objectives were to investigate the effects of the employed (1) breeding scheme, (2) selection strategy, and (3) population sizes on the donor genome coverage of the library, the number of introgression lines carrying additional donor chromosome segments outside the target regions, and the number of required marker data points. With respect to these target criteria, a BC3S2 breeding scheme and increasing population sizes from early to advanced generations were superior to a BC2S3 breeding scheme and constant population sizes. The smallest number of donor segments outside the target regions was reached with a three-stage selection strategy, which consists on selection for the target segment, selection for recombination at flanking markers and selection for recurrent parent alleles across the entire genome. Omitting the selection for flanking markers in generation BC1 reduced considerably the number of required marker data points. A pre-selection of chromosomes consisting completely of donor genome in BC1 was advantageous, if the effort in the breeding nursery should kept minimum. Adopting the described designs can help rye breeders to successfully develop introgression libraries.  相似文献

A marker-assisted background selection (MABS)-based gene introgression approach in wheat (Triticum aestivum L.) was optimized, where 97% or more of a recurrent parent genome (RPG) can be recovered in just two backcross (BC) generations. A four-step MABS method was developed based on ‘Plabsim’ computer simulations and wheat genome structure information. During empirical optimization of this method, double recombinants around the target gene were selected in a step-wise fashion during the two BC cycles followed by selection for recurrent parent genotype on non-carrier chromosomes. The average spacing between carrier chromosome markers was <4 cM. For non-carrier chromosome markers that flanked each of the 48 wheat gene-rich regions, this distance was ∼12 cM. Employed to introgress seedling stripe rust (Puccinia striiformis f. sp. tritici) resistance gene Yr15 into the spring wheat cultivar ‘Zak’, marker analysis of 2,187 backcross-derived progeny resulted in the recovery of a BC2F2∶3 plant with 97% of the recurrent parent genome. In contrast, only 82% of the recurrent parent genome was recovered in phenotypically selected BC4F7 plants developed without MABS. Field evaluation results from 17 locations indicated that the MABS-derived line was either equal or superior to the recurrent parent for the tested agronomic characteristics. Based on these results, MABS is recommended as a strategy for rapidly introgressing a targeted gene into a wheat genotype in just two backcross generations while recovering 97% or more of the recurrent parent genotype.  相似文献

Near-isogenic lines (NILs) differing with regard to disease QTLs provide valuable material for a more detailed study into the genetic basis of quantitative resistance. Previously obtained information on QTLs that show an effect on leaf rust (Puccinia hordei) in barley was used in a marker-assisted backcross programme. The genome origin in backcross plants was controlled through AFLP marker analysis and graphical genotyping. Plants obtained after the third generation of backcrossing sufficiently resembled the recurrent parent. For one QTL, BC3S1 plants were evaluated in a disease test and genotyped. NILs containing the desired QTL in homozygous condition in a recipient background were finally obtained. A disease test and verification of the marker genotype confirmed the identity of the NILs. Simultaneous with the backcross programme a simulation study on efficiency of marker-assisted backcrossing was performed.  相似文献

The advent of molecular markers as a tool to aid selection has provided plant breeders with the opportunity to rapidly deliver superior genetic solutions to problems in agricultural production systems. However, a major constraint to the implementation of marker-assisted selection (MAS) in pragmatic breeding programs in the past has been the perceived high relative cost of MAS compared to conventional phenotypic selection. In this paper, computer simulation was used to design a genetically effective and economically efficient marker-assisted breeding strategy aimed at a specific outcome. Under investigation was a strategy involving the integration of both restricted backcrossing and doubled haploid (DH) technology. The point at which molecular markers are applied in a selection strategy can be critical to the effectiveness and cost efficiency of that strategy. The application of molecular markers was considered at three phases in the strategy: allele enrichment in the BC1F1 population, gene selection at the haploid stage and the selection for recurrent parent background of DHs prior to field testing. Overall, incorporating MAS at all three stages was the most effective, in terms of delivering a high frequency of desired outcomes and at combining the selected favourable rust resistance, end use quality and grain yield alleles. However, when costs were included in the model the combination of MAS at the BC1F1 and haploid stage was identified as the optimal strategy. A detailed economic analysis showed that incorporation of marker selection at these two stages not only increased genetic gain over the phenotypic alternative but actually reduced the over all cost by 40%.  相似文献

本研究利用鲁豆4号回交转育的大豆种子脂氧酶缺失株系为材料,用IEF-PAGE鉴定大豆种子脂氧酶缺失基因,SSR标记进行遗传背景分析,通过遗传背景回复率相关分析,探索分子标记辅助遗传背景选择时所需的适宜的标记数和选择方式,期望获得可进一步回的鲁豆4号脂氧酶缺失株系。研究明确了大豆SSR标记辅助背景选择时适宜标记数目和选择方式,即先用少数标记初筛,选出遗传背景回复率较高的材料,再用适宜标记鉴定,随着世代递增,已恢复为轮回亲本的标记住点不再分析,逐代减少选择标记数目。获得了合有脂氧酶缺失基因,遗传背景与鲁豆4号差异较小的株系,可用鲁玉4号进一步回交,从而加速培育鲁豆4号脂氧酶缺失近等基因系。  相似文献

Boron (B) is an essential micronutrient for higher plant, but toxic levels can seriously diminish grain yield in cereal crops by affecting root growth, and thus restricting water extraction from the subsoil. Amelioration of high concentrations in soils is expensive and not always feasible, so breeding for B tolerance is the most viable alternative. This article reports the marker-assisted (MAS) transfer of favourable alleles from an unadapted six-rowed barley (Hordeum vulgare L.) variety, Sahara 3771, into two-rowed lines adapted to southern Australia. During the backcrossing process, the SSR marker, EBmac679, located on chromosome 4H was used to control the target region in foreground selection, but no background selection was applied. Gene introgression was confirmed with 40 BC6F1-derived doubled haploid lines segregating for the SSR marker EBmac679. We used a combination of molecular and conventional assays to unequivocally classify the 40 BC6F1-derived DH lines as B tolerant or sensitive, and then compared their means for grain yield measured over 2 years and four locations. Results showed modest improvements in grain yield of lines carrying B tolerance genes at some B toxic environments, and negative impact at others. Our results also showed that malting quality profile was not adversely affected through the introgression of the B tolerance allele from Sahara 3771, allowing the newly developed material to be used by breeding programs without risk of a penalty on malt quality.  相似文献

High sucrose concentration in fruit of Lycopersicon chmielewskii is governed by the recessive sucrose accumulator gene (sucr) that is situated in the pericentromeric region of chromosome 3. The sucr gene was introgressed into the genetic background of the hexose-accumulating cultivated tomato (L. esculentum cv Hunt 100) by marker-assisted selection using tightly linked RFLP markers and a tomato acid invertase cDNA as probes for sucr. RFLP mapping indicated that the segment containing sucr comprised over 43.2 cM in the BC1F2 generation, representing over one-third of the total length of chromosome 3. By selecting for crossovers between sucr and the flanking visual marker r (yellow fruit flesh) and RFLP marker TG288, we were able to reduce the size of the sucr introgression fragment to 0.8–7.1 cM by the BC5 generation. Smaller recombinant fragments were not obtained despite screening a large BC6F2 population. The smallest sucr introgression reduced recombination between the flanking visual markers sy (sunny) and bls (baby lea syndrome) by 38%. To facilitate future introgression and recombination experiments, a PCR-based test for the sucr gene was developed using primers specific to the tomato invertase gene. This assay takes advantage of a small deletion that maps to the second intron of the L. chmielewskii nvertase gene. The assay detected significant allelic variation both within and between hexose- and sucrose-accumulating Lycopersicon spp.  相似文献

Transgene flow from engineered Brassica napus to wild weed relatives could potentially have an environmental effect. To evaluate the introgression of transgenic B. napus into wild Brassica juncea, the hybrid F1 and backcross progenies derived from B. juncea (genome constitution AABB) and transgenic B. napus (AACC) crosses were investigated. C-genome-specific simple sequence repeat (SSR) markers corresponding to linkage groups N11–N19 in B. napus were screened and used to estimate the marker frequency in hybrid F1 and backcross progenies. C-genome-specific markers could be stably detected in hybrid F1 and backcross BC1 plants, but were only rarely found in the BC2–BC5 generations. For example, a specific SSR marker for linkage group N12 segregated in BC2 generation but were completely lost in BC3–BC5, while a specific SSR marker of linkage group N15 segregated in BC1, BC2 and BC3 generations and was absent in more advanced backcrossed generations (BC4 and BC5). The results indicate that a certain gene regions in Brassica napus plants are transmitted at a relatively lower frequency to wild relatives, and more rapidly disappeared in subsequent backcross generations. We propose that a foreign gene or transgene that is integrated in the C-chromosome of Brassica napus could reduce the risk of introgression in nature.  相似文献

Recurrent backcrossing is an established procedure to transfer target genes from a donor into the genetic background of a recipient genotype. By assessing the parental origin of alleles at markers flanking the target locus one can select individuals with a short intact donor chromosome segment around the target gene and thus reduce the linkage drag. We investigated the probability distribution of the length of the intact donor chromosome segment around the target gene in recurrent backcrossing with selection for heterozygosity at the target locus and homozygosity for the recurrent parent allele at flanking markers for a diploid species. Assuming no interference in crossover formation, we derived the cumulative density function, probability density function, expected value, and variance of the length of the intact chromosome segment for the following cases: (1) backcross generations prior to detection of a recombinant individual between the target gene and the flanking marker; (2) the backcross generation in which for the first time a recombinant individual is detected, which is selected for further backcrossing; and (3) subsequent backcross generations after selection of a recombinant. Examples are given of how these results can be applied to investigate the efficiency of marker-assisted backcrossing for reducing the length of the intact donor chromosome segment around the target gene under various situations relevant in breeding and genetic research.  相似文献

Backcrossing together with simple sequence repeat marker strategy was adopted to improve popular Malaysian chilli Kulai (Capsicum annuum L.) for heat tolerance. The use of molecular markers in backcross breeding and selection contributes significantly to overcoming the main drawbacks such as increase linkage drag and time consumption, in the ancient manual breeding approach (conventional), and speeds up the genome recovery of the recurrent parent. The strategy was adopted to introgress heat shock protein gene(s) from AVPP0702 (C. annuum L.), which are heat-tolerant, into the genetic profile of Kulai, a popular high-yielding chilli but which is heat sensitive. The parents were grown on seed trays, and parental screening was carried out with 252 simple sequence repeat markers. The selected parents were crossed and backcrossed to generate F1 hybrids and backcross generations. Sixty-eight markers appeared to be polymorphic and were used to assess the backcross generation; BC1F1, BC2F1 and BC3F1. The average recipient allele of the selected four BC1F1 plants was 80.75% which were used to produce the BC2F1 generation. BC1-P7 was the best BC1F1 plant because it had the highest recovery at 83.40% and was positive to Hsp-linked markers (Hsp70-u2 and AGi42). After three successive generations of backcrossing, the average genome recovery of the recurrent parent in the selected plants in BC3F1 was 95.37%. Hsp gene expression analysis was carried out on BC1F1, BC2F1 and BC3F1 selected lines. The Hsp genes were found to be up-regulated when exposed to heat treatment. The pattern of Hsp expression in the backcross generations was similar to that of the donor parent. This confirms the successful introgression of a stress-responsive gene (Hsp) into a Kulai chilli pepper variety. Furthermore, the yield performance viz. plant height, number of fruits, fruit length and weight and total yield of the improved plant were similar with the recurrent parent except that the plant height was significantly lower than the Kulai (recurrent) parent.  相似文献

Hybrid rice has contributed greatly to the self-sufficiency of the food supply in China. However, bacterial blight is a major disease that limits hybrid rice production in China. The study was conducted to develop an efficient breeding technique to improve the bacterial blight resistance in hybrid rice. A marker-assisted backcross breeding technique was adopted to improve HN189, an elite restorer line containing the Pi1 gene. This breeding technique was simplified to foreground selection with only one generation of backcrossing and background selection based on phenotypic selection. A novel bacterial blight resistance gene, Xa23, was introgressed into HN189. Two improved restorer lines, HBH145 (with one generation of backcrossing) and HBH146 (with two generations of backcrossing), were obtained that had a significant bacterial blight resistance advantage over HN189. The corresponding hybrid combination Tianyou H145 (Tianfeng A / HBH145) was certified one year earlier than Qianyou H146 (Qianjiang 1A / HBH146). The use of the marker-assisted backcross breeding technique with one generation of backcrossing and without background selection in rice breeding programs shortened the breeding period of the rice.  相似文献

MTU 1010 is a high-yielding mega-variety of rice grown extensively in India. However, it does not perform well in soils with low phosphorus (P) levels. With an objective to improve MTU 1010 for tolerance to low soil P, we have transferred Pup1, a major quantitative trait locus (QTL) associated with tolerance from another mega-variety, Swarna, through marker-assisted backcross breeding (MABB). Foreground selection of the F1 and backcross plants was performed with the co-dominant, closely linked CAPS marker, K20-2, while two flanking markers RM28011 and RM28157 were utilized for recombinant selection. At each backcross generation, positive plants were also analyzed with a set of 85 parental polymorphic SSR markers to identify the QTL-positive plants possessing maximum introgression of MTU 1010 genome. At BC2F1, the best backcross plant was selfed to generate BC2F2s. Among them, the plants homozygous for Pup1 (n?=?22) were reconfirmed using the functional marker for Pup1, viz., K46-1, and they were advanced through pedigree method of selection until BC2F6 generation. A total of five elite BC2F6 lines, possessing Pup1 and phenotypically similar to MTU 1010, were screened in the low soil P plot and normal plot (with optimum soil P levels) during wet season, 2016. All the selected lines showed better performance under low P soil with more number of productive tillers, better root system architecture, and significantly higher yield (>?390%) as compared to MTU 1010. Further, under normal soil, the lines were observed to be similar to or better than MTU 1010 for most of the agro-morphological traits and yield. This study represents the successful application of marker-assisted selection for improvement of tolerance to low soil P in a high-yielding Indian rice variety.  相似文献

Fusarium wilt (Fusarium oxysporum Schlecht. f. sp. melongenae) is a vascular disease of eggplant (Solanum melongena L.). The objectives of this work were (1) to confirm the monogenic inheritance of fusarium wilt resistance in eggplant, (2) to identify molecular markers linked to this resistance, and (3) to develop SCAR markers from most informative markers. We report the tagging of the gene for resistance to fusarium wilt (FOM) in eggplant using SRAP, RGA, SRAP-RGA and RAPD markers. Analysis of segregation data confirmed the monogenic inheritance of resistance. DNA from F2 and BC1 populations of eggplant segregating for fusarium wilt resistance was screened with 2,316 primer combinations to detect polymorphism. Three markers were linked within 2.6 cM of the gene. The codominant SRAP marker Me8/Em5 and dominant SRAP-RGA marker Em12/GLPL2 were tightly linked to each other and mapped 1.2 cM from the resistance gene, whereas RAPD marker H12 mapped 2.6 cM from the gene and on the same side as the other two markers. The SRAP marker was converted into two dominant SCAR markers that were confirmed to be linked to the resistance gene in the F2, BC1 and F2 of BC3 generations of the same cross. These markers provide a starting point for mapping the eggplant FOM resistance gene in eggplant and for exploring the synteny between solanaceous crops for fusarium wilt resistance genes. The SCAR markers will be useful for identifying fusarium wilt-resistant genotypes in marker-assisted selection breeding programs using segregating progenies of the resistant eggplant progenitor used in this study.  相似文献

Seed glucosinolate content in Brassica juncea is a complex quantitative trait. A recurrent selection backcross (RSB) method with a doubled haploid (DH) generation interspersing backcross generations was used for the introgression of low glucosinolate alleles from an east European gene pool B. juncea line, Heera into an Indian gene pool variety, Varuna. Phenotypic comparisons among the DH populations derived from early to advanced backcrosses revealed a shift in the mean values for various glucosinolates with the advancement of backcrossing, indicating a change in the selective values of the alleles with change in the genetic background due to the existence of epistasis and context dependencies. QTL mapping for various seed glucosinolates from early (F1DH) and advanced generation (BC4DH) populations confirmed the presence of epistasis and context dependency. The common QTL detected in both F1DH and BC4DH changed their R 2 values from the former to the later generation. Some of the QTL detected in the F1DH became irrelevant in the BC4DH population. Further, new QTL were detected in the BC4DH population for various glucosinolates. A validation study on a population of low glucosinolate DH lines derived from all the backcross generations of the RSB breeding programme revealed that the QTL detected in BC4DH were the ‘true’ QTL. Using glucosinolate as an example, the study provides strong evidence for the importance of the RSB method for the identification of the ‘true’ QTL which would be significant for marker assisted introgression of a complex quantitative trait whose expression is influenced by epistatic interactions. 家蚕突变表型无鳞毛翅(non-lepis wing, nlw)由隐性基因nlw控制。由于家蚕雌性不发生交换, 文章采用有鳞毛翅品系P50和无鳞毛翅品系U06两个品系组配F1代及BC1回交群体, (U06×P50)×U06和U06×(U06×P50)分别记作BC1F和BC1M, 根据已经构建的家蚕SSR分子标记连锁图谱及已经发表的有关序列对nlw基因进行了连锁及定位分析。得到8个与nlw基因连锁的SSR(Simple sequence repeat)标记和1个STS(Sequence-tagged sites)标记。BC1F群中的所有正常翅个体均表现出与(U06×P50)F1相同的杂合带型; 而所有无鳞毛个体带型与亲本U06一致, 为纯合型。利用BC1M群体构建了关于nlw基因的遗传连锁图, 连锁图的遗传距离为125.7 cM, 与nlw基因最近的引物为STS标记cash2p, 图距为11.4 cM。  相似文献
