The extent of gene flow during the range expansion of non-native species influences the amount of genetic diversity retained in expanding populations. Here, we analyse the population genetic structure of the raccoon dog (Nyctereutes procyonoides) in north-eastern and central Europe. This invasive species is of management concern because it is highly susceptible to fox rabies and an important secondary host of the virus. We hypothesized that the large number of introduced animals and the species’ dispersal capabilities led to high population connectivity and maintenance of genetic diversity throughout the invaded range. We genotyped 332 tissue samples from seven European countries using 16 microsatellite loci. Different algorithms identified three genetic clusters corresponding to Finland, Denmark and a large ‘central’ population that reached from introduction areas in western Russia to northern Germany. Cluster assignments provided evidence of long-distance dispersal. The results of an Approximate Bayesian Computation analysis supported a scenario of equal effective population sizes among different pre-defined populations in the large central cluster. Our results are in line with strong gene flow and secondary admixture between neighbouring demes leading to reduced genetic structuring, probably a result of its fairly rapid population expansion after introduction. The results presented here are remarkable in the sense that we identified a homogenous genetic cluster inhabiting an area stretching over more than 1500km. They are also relevant for disease management, as in the event of a significant rabies outbreak, there is a great risk of a rapid virus spread among raccoon dog populations.  相似文献

The population of raccoon dogs (Nyctereutes procyonoides) in Denmark has increased rapidly from 1995 when the first was recorded until today where 3291 raccoon dogs are trapped, shot by hunters or road killed. The aims of this study are to present the first data on reproduction and life tables of raccoon dogs in Denmark and to compare mortality from modelled life tables with game bag records and sampled raccoon dogs in different age groups. In this study, the uteri of 89 adult females (> 10 months) were examined for placental scars (PSC), and 561 individuals (289 males, 272 females) were aged using pulp cavity width and dental lines in canine teeth. The litter size of raccoon dogs in Denmark is to date the largest litter size recorded in the wild (mean ± SE) 10.8 ± 0.4, range 1–16 pubs and fecundity 8.4 ± 0.6 pubs. The percent-reproducing females are 78–83%, based on dark and all PSC, respectively. A significant difference was found between the proportion of individuals composing the different age groups based on age determination of individuals collected (Ntage) and the modelled number of individuals in age groups based on fecundity and different mortality rate (Ntmodel), X2 = 8, p < 0.05. The discrepancy between the relatively high reproduction and lifetables may be due to older and more experienced animals that avoid culling. A low population density in a newly founded Danish population of raccoon dogs, together with a milder climate where raccoon dogs can forage during the winter, may cause an exceptionally high reproduction in Danish raccoon dogs.  相似文献

Animal parasitic nematodes can cause serious diseases and their emergence in new areas can be an issue of major concern for biodiversity conservation and human health. Their ability to adapt to new environments and hosts is likely to be affected by their degree of genetic diversity, with gene flow between distinct populations counteracting genetic drift and increasing effective population size. The raccoon roundworm (Baylisascaris procyonis), a gastrointestinal parasite of the raccoon (Procyon lotor), has increased its global geographic range after being translocated with its host. The raccoon has been introduced multiple times to Germany, but not all its populations are infected with the parasite. While fewer introduced individuals may have led to reduced diversity in the parasite, admixture between different founder populations may have counteracted genetic drift and bottlenecks. Here, we analyse the population genetic structure of the roundworm and its raccoon host at the intersection of distinct raccoon populations infected with B. procyonis. We found evidence for two parasite clusters resulting from independent introductions. Both clusters exhibited an extremely low genetic diversity, suggesting small founding populations subjected to inbreeding and genetic drift with no, or very limited, genetic influx from population admixture. Comparison of the population genetic structures of both host and parasite suggested that the parasite spread to an uninfected raccoon founder population. On the other hand, an almost perfect match between cluster boundaries also suggested that the population genetic structure of B. procyonis has remained stable since its introduction, mirroring that of its raccoon host.  相似文献

The raccoon (Procyon lotor) has successfully invaded central Europe, despite the population apparently having been founded by a small number of individuals in two distinct populations in Germany. The ecological success of the invasion has been explained by raccoons being an adaptable, truly omnivorous species. However, the German raccoon population might have a larger number of founders and be more genetically diverse than assumed, as accidental or deliberate releases of household pets or individuals from zoos are relatively common. In the present study, we sequenced a 550-base-pair long fragment of the mitochondrial control region in 193 raccoons from Germany and neighbouring countries. We only identified six different haplotypes; of which, five were limited to Germany. Our results support the notions that the population was founded by a small number of females and that the German raccoons originate from two separate release events in central and eastern Germany. Additionally, however, we provide evidence for the presence of a distinct population in Saxony, eastern Germany. Further studies using different genetic markers are necessary to gain additional information on genetic diversity and population genetic structure.  相似文献

Honey bees are the most important managed pollinators as they provide key ecosystem services for crop production worldwide. Recent losses of honey bee colonies in North America and Europe have demonstrated a need to develop strategies to improve their health and conserve their populations. Previously, we showed that feral honey bees—colonies that live in the wild without human assistance—exhibit higher levels of immunocompetence than managed colonies in North Carolina (USA). In a first attempt to investigate the underlying mechanisms of this difference in immune response, here we characterize the genetic composition of feral and managed honey bees using microsatellite markers. Our results reveal significant but small genetic differentiation between feral and managed honey bee colonies (?CT = 0.047, P?=?0.03) indicating admixture between these two groups. Higher genetic diversity was correlated with higher immune response in feral (P MANOVA = 0.011) but not managed bees, despite the fact that the latter group showed significantly higher average genetic diversity (P ANCOVA < 0.001). These findings suggest that genetic diversity is positively associated with immunocompetence in feral honey bee colonies, and that the benefits of genetic diversity are obscured in managed bees, perhaps as a result of artificial selection. We hypothesize that high genetic variability provides the raw material upon which natural selection acts and generates adaptive genotypes in unmanaged populations. Feral populations could be useful sources of genetic variation to use in breeding programs that aim to improve honey bee health.  相似文献

Phylogeographic studies of highly mobile large carnivores suggest that intra-specific genetic differentiation of modern species might be the consequence of the most recent Pleistocene glaciation. However, the relative influence of biogeographical processes and subsequent human-induced population fragmentation requires a better understanding. Poland represents the western edge of relatively continuous distributions of many wide-ranging species, e.g. lynx (Lynx lynx), wolves (Canis lupus), moose (Alces alces) and, therefore, a key area for understanding historic and contemporary patterns of gene flow in central Europe. We examined wolf genetic structure in Poland and in a recently recolonized area in eastern Germany using microsatellite profiles (n = 457) and mitochondrial DNA sequencing (mtDNA, n = 333) from faecal samples. We found significant genetic structure and high levels of differentiation between wolves in the Carpathian Mountains and the Polish lowlands. Our findings are consistent with previously reported mtDNA subdivision between northern lowlands and southern mountains, and add new and concordant findings based on autosomal marker variation. Wolves in western Poland and eastern Germany showed limited differentiation from northeastern Poland. Although the presence of private alleles suggests immigration also from areas not sampled in this study, most individuals seem to be immigrants from northeastern Poland or their descendants. We observed moderate genetic differentiation between certain northeastern lowland regions separated by less than 50 km. Moreover, mtDNA results indicated a southeastern subpopulation near the border with Ukraine. The observed structure might reflect landscape fragmentation and/or ecological differences resulting in natal habitat-biased dispersal.  相似文献

Genetic admixture between captive-bred and wild individuals has been demonstrated to affect many individual traits, although little is known about its potential influence on dispersal, an important trait governing the eco-evolutionary dynamics of populations. Here, we quantified and described the spatial distribution of genetic admixture in a brown trout (Salmo trutta) population from a small watershed that was stocked until 1999, and then tested whether or not individual dispersal parameters were related to admixture between wild and captive-bred fish. We genotyped 715 fish at 17 microsatellite loci sampled from both the mainstream and all populated tributaries, as well as 48 fish from the hatchery used to stock the study area. First, we used Bayesian clustering to infer local genetic structure and to quantify genetic admixture. We inferred first generation migrants to identify dispersal events and test which features (genetic admixture, sex and body length) affected dispersal parameters (i.e. probability to disperse, distance of dispersal and direction of the dispersal event). We identified two genetic clusters in the river basin, corresponding to wild fish on the one hand and to fish derived from the captive strain on the other hand, allowing us to define an individual gradient of admixture. Individuals with a strong assignment to the captive strain occurred almost exclusively in some tributaries, and were more likely to disperse towards a tributary than towards a site of the mainstream. Furthermore, dispersal probability increased as the probability of assignment to the captive strain increased, and individuals with an intermediate level of admixture exhibited the lowest dispersal distances. These findings show that various dispersal parameters may be biased by admixture with captive-bred genotypes, and that management policies should take into account the differential spread of captive-bred individuals in wild populations.  相似文献

Studies on the genetic diversity and relatedness of zoo populations are crucial for implementing successful breeding programmes. The European wildcat, Felis s. silvestris, is subject to intensive conservation measures, including captive breeding and reintroduction. We here present the first systematic genetic analysis of the captive population of Felis s. silvestris in comparison with a natural wild population. We used microsatellites and mtDNA sequencing to assess genetic diversity, structure and integrity of the ex situ population. Our results show that the ex situ population of the European wildcat is highly structured and that it has a higher genetic diversity than the studied wild population. Some genetic clusters matched the breeding lines of certain zoos or groups of zoos that often exchanged individuals. Two mitochondrial haplotype groups were detected in the in situ populations, one of which was closely related to the most common haplotype found in domestic cats, suggesting past introgression in the wild. Although native haplotypes were also found in the captive population, the majority (68%) of captive individuals shared a common mtDNA haplotype with the domestic cat (Felis s. catus). Only six captive individuals (7.7%) were assigned as wildcats in the STRUCTURE analysis (at K = 2), two of which had domestic cat mtDNA haplotypes and only two captive individuals were assigned as purebred wildcats by NewHybrids. These results suggest that the high genetic diversity of the captive population has been caused by admixture with domestic cats. Therefore, the captive population cannot be recommended for further breeding and reintroduction.  相似文献

Aim Invasive alien species usually exhibit very high adaptation and rapid evolution in a new environment, but they often have low levels of genetic diversity (invasive species paradox). Genetic variation and population genetic structure of feral American mink, Neovison vison, in Poland was investigated to explain the invasion paradox and to assess current gene flow. Furthermore, the influence of mink farming on adaptation of the feral population was evaluated by comparing the genetic structure of feral and ranch mink. Location Samples from feral mink were collected in 11 study areas in northern and central Poland and from ranch mink at 10 farms distributed throughout the country. Methods A 373‐bp‐long mtDNA control region fragment was amplified from 276 feral and 166 ranch mink. Results Overall, 31 haplotypes, belonging to two groups from genetically diverse sources, were detected: 11 only in feral mink, 12 only in ranch mink and eight in both. The genetic differentiation of feral mink from the trapping sites was high, while that among ranch mink from various farms was moderate. There was no significant relationship between genetic and geographic distance. The number of trapping sites where given haplotypes occurred correlated with the number of farms with these haplotypes. The mink from two sites were the most divergent, both from all other feral mink and from ranch mink. Comparison of mtDNA and microsatellite differentiation suggests male‐biased dispersal in this species. Main conclusions American mink in Poland exhibit high genetic diversity and originate from different source populations of their native range. The process of colonization was triggered by numerous escapees from various farms and by immigrants from Belarus. The genetic structure of local feral mink populations was shaped by the founder effect and multiple introductions. The genomic admixture that occurred during mixing of different populations might have increased the fitness of individuals and accelerated the invasiveness of this species.  相似文献

The diversity of 11 microsatellite loci was examined to estimate the genetic variability of ranch and feral American minkNeovison vison (Schreber, 1777) in Poland. Samples were collected from 10 mink farms (182 individuals) and from 5 areas in the north-eastern part of the country (87 individuals). At each examined locus the observed heterozygosity (H o) was lower than the expected heterozygosity (H e). Feral mink showed lower genetic variability than ranch mink; however, in the former group the mean value of the inbreeding coefficient (F IS=0.306) was higher than in the latter (0.242). These results demonstrated that feral and ranch mink belong to two genetically close but separate groups. Genetic differences were identified between mink colour breeds but not between animals from particular farms. The height of the modal values of ΔK indicated the presence of four genetic clusters: (1) farmed mink sapphire, (2) farmed mink standard and pastel, (3) farmed mink pearl and (4) feral mink. Assignment of mink individuals using assignment test, STRUCTURE and GeneClass 2.0. revealed that 12–16% of the feral mink group are likely to be ranch mink escapees. It may be concluded that approximately 30 years after the start of the expansion of feral mink in north-eastern Poland, this wild-living population exists without a major input of individuals bred on fur farms.  相似文献

Invasions of non-native species are of great concern as they have a devastating impact on native biodiversity and can also affect the economy of a region. Multiple introductions in several locations of a new range greatly promote the success of non-native species. The raccoon (Procyon lotor) is an omnivore whose native distribution extends from southern Canada to Panama. It has been successfully introduced in many European countries. We examined the microsatellite and mitochondrial diversity of raccoon populations in Central Europe (Germany, Poland, and Czech Republic) in order to determine their introduction sources and pathways as well as the factors affecting genetic structure in this invasive species. We found low diversity of the mtDNA control region and moderate diversity of microsatellite markers. Raccoon showed three hierarchic levels of genetic structure which separate at different levels sampled from Czech Republic, Germany and raccoon inhabiting two different habitats in Poland. In Poland the raccoon population was established through migration from Germany to Czech Republic. Analysis of the intensity of migration between two different habitat types indicated source-sink dynamics in the Polish populations of raccoons. Our results confirm the high intensity of the raccoon invasion in Central Europe and point to specific measures needed as part of an effective management strategy.  相似文献

Tuberculosis is a significant problem globally for domestic animals as well as captive and free ranging wild life. Rapid point of care (POC) serology kits are well suited for the diagnosis of TB in wild animals. However, wild animals are invariably exposed to environmental non-pathogenic mycobacterium species with the development of cross reacting antibodies. In the present study, POC TB diagnosis kit was developed using a combination of pathogenic Mycobacteria specific recombinant antigens and purified protein derivatives of pathogenic and non-pathogenic Mycobacteria. To benchmark the TB antibody detection kit, particularly in respect to specificity which could not be determined in wildlife due to the lack of samples from confirmed uninfected animals, we first tested well-characterized sera from 100 M. bovis infected and 100 uninfected cattle. Then we investigated the kit’s performance using sera samples from wildlife, namely Sloth Bears (n = 74), Elephants (n = 9), Cervidae (n = 14), Felidae (n = 21), Cape buffalo (n = 2), Wild bear (n = 1) and Wild dog (n = 1).In cattle, a sensitivity of 81% and a specificity of 90% were obtained. The diagnostic sensitivity of the kit was 94% when the kit was tested using known TB positive sloth bear sera samples. 47.4% of the in-contact sloth bears turned seropositive using the rapid POC TB diagnostic kit. Seropositivity in other wild animals was 25% when the sera samples were tested using the kit. A point of care TB sero-diagnostic kit with the combination of proteins was developed and the kit was validated using the sera samples of wild animals.  相似文献

Estimates of genetic diversity and phylogenetic affiliation represent an important resource for biodiversity assessment and a valuable guide to conservation and management. We have found a new population (Jawor—JW) of the common hamster Cricetus cricetus in western Poland that is remote from the nearest populations by 235–300 km. With the objective of genetically characterizing of this population, we compared it with other populations from Poland and Germany by taking into account sequences of four mitochondrial DNA genes and variation at 10 microsatellite loci. The JW population exhibited low levels of genetic diversity and allelic and haplotype richness, which likely reflects its extreme isolation. This factor, coupled with inbreeding and genetic drift, are major threats to JW. A neighbor-joining tree based on mtDNA haplotypes shows that JW clusters among samples representing the Central subgroup that is known from central Germany but that has not yet been identified in Poland. Findings presented here improve our understanding of the spread and diversification of the common hamster. We offer the following hypotheses to explain the observed pattern of mtDNA haplotype distribution: JW could be a byproduct of postglacial migrations or back-migrations from eastern refugia to the western part of Europe, or/and be a result of population and habitat fragmentation. We recommend translocation of individuals as an effective management strategy, both at the level of Central phylogeographic group and at the species level, to overcome the negative consequences of inbreeding and geographical isolation of the JW population.  相似文献

The main objective was to discover extent of interference and/or exploitative competition between the native red fox (Vulpes vulpes) and the introduced, invasive raccoon dog (Nyctereues proconoides) in the intensively used, agricultural landscape of northeast Germany (Mecklenburg-Western Pomerania) using very high frequency (VHF) radio telemetry. We recorded location data for 12 foxes and 16 raccoon dogs between July 2004 and December 2006. Species had similar average home range sizes estimated in each season (K95). Home ranges of adjacent raccoon dogs and foxes overlapped from 0.5 to 74.5 % with a mean of 26.4 %. We found a significantly different home range overlap index between the species showing that raccoon dog ranges shifted between seasons to a greater extent than red fox ranges. The raccoon dog differed significantly from the red fox in its use of habitat types, preferring dense vegetation cover and avoiding open areas. The red fox displayed less preference for or avoidance of specific habitat types. Moreover, an almost neutral inter-specific interaction index ranging from ?0.12 to 0.12 indicates that raccoon dogs and red foxes ignored each other. It is concluded that widespread and available resources and differences in spatial use patterns prevent competition between red foxes and raccoon dogs in the agricultural landscape of northeast Germany.  相似文献

In total, 721 specimens from eight populations of the raccoon dog Nyctereutes procyonoides across native and colonized areas have been studied. The most polymorphic populations inhabit the European part of Russia and a part of the native area. The raccoon dog from Eastern Poland and the Primorie Far East is characterized by low intrapopulational variability. Quantitative genetic variations in the studied populations indicate a relatively higher level in the population from the Primorie Far East compared to the Southern Vologda and Eastern Poland. The pattern of morphological variability in the raccoon dog is shaped by such factors as the latest phylogenetic history of the species and the structure of the morphotype frequency in the time of the divergence of the populations on both micro-and macro-geographic levels. The influence of geographic and climatic factors forms a well-pronounced trend towards diversification in tooth morphology, which is most probably the consequence of adaptations to changes in the food niche.  相似文献

The raccoon dog, a medium-sized carnivore, has long been recognised as a prominent example of an invasive alien species in Europe with a wide distribution, significant ecological impact and remarkable dynamics of spread at both national and continental scales. We conducted a study of genetic diversity of 73 individuals collected at 20 sites across North and Central Europe to (1) identify major phylogenetic lineages and (2) elucidate spatial patterns of population genetic structure. Reconstructed phylogenies reveal two major clades differing on average by Tamura–Nei corrected distance of 3.4% for a 599-bp segment of the mitochondrial control region corresponding to a coalescence time of approximately 457,800 years ago (95% CI, 223,300–773,900). Many expectations based on introduction history, such as the presence of signatures of repeated founder effects and subsequently rapid population expansion, were not confirmed by our demographic analyses, probably due to an insufficient amount of time since translocations. Nevertheless, global F ST = 13.9% and landscape approaches provided evidence for weak population genetic structure that followed a pattern of isolation by distance. Finally, we found no congruence between previously reported morphological differentiation and the sorting of mtDNA variation. We therefore conclude that an exceptional combination of factors including multiple translocations, secondary contact and admixture of divergent matrilineages, as well as natural processes of colonisation associated with a wide ecological tolerance, promoted the successful spread of the raccoon dog into Europe.  相似文献

African descended populations exhibit an increased prevalence of asthma and allergies compared to Europeans. One approach to distinguish between environmental and genetic explanations for this difference is to study relationships of asthma risk to individual admixture. We aimed to determine the admixture proportions of a case-control sample from the Caribbean Coast of Colombia currently participating in genetic studies for asthma, and to test for population stratification and association between African ancestry and asthma and total serum IgE levels (tIgE). We genotyped 368 asthmatics and 365 non-asthmatics for 52 autosomal ancestry informative markers, six mtDNA haplogroups and nine haplogroups and five microsatellites in Y chromosome. Autosomal admixture proportions, population stratification, and associations between ancestry and the phenotypes were estimated by ADMIXMAP. The average admixture proportions among asthmatics were 42.8% European, 39.9% African and 17.2% Native American and among non-asthmatics they were 44.2% (P = 0.068), 37.6% (P = 0.007) and 18.1% (P = 0.050), respectively. In the total sample, the paternal contributions were 71% European, 25% African and 4.0% Native American and the maternal lineages were 56.8% Native American, and 20.2% African; 22.9% of the individuals carried other non-Native American mtDNA haplogroups. African ancestry was significantly associated with asthma (OR: 2.97; 95% CI: 1.08–8.08), high tIgE (OR: 1.9; 95% CI: 1.17–3.12) and socioeconomic status (OR = 0.64; 95% CI: 0.47–0.87). Significant population stratification was observed in this sample. Our findings indicate that genetic factors can explain the association between asthma and African ancestry and suggest that this sample is a useful resource for performing admixture mapping for asthma.  相似文献

对来自陕西、云南、越南、安徽和广西等地的8只中国貉(Nyctereutesprocyonides)进行随机扩增多态DNA分析。应用28个10bp的随机引物,平均每只貉获得的RAPD标记数约为130条。遗传距离计算结果显示,中国貉个体间的平均遗传距离指数值为11.20%,最大值为14.93%,最小值为2.94%。以赤狐(Vulpesvulpes)为外群,应用PHYLIP3.0计算软件包中的UPGMA和NJ聚类方法构建分子系统树。结果表明,不同地理群体间的中国貉存在遗传分化;中国貉可分为4组:(1)广西貉,(2)安徽貉,(3)陕西貉,(4)云南貉和越南貉。其中安徽貉和广西貉间的关系稍近,陕西貉则与云南貉-越南貉稍近。对合中国貉的形态分类、地理分布、mtDNA多态分析以及进化遗传学的观点,认为陕西貉、广西貉和安徽貉可能与云南貉-越南貉具有等同的分类地位。  相似文献

In Southampton Water, UK, the recent (c. 150 years ago) interspecific hybridisation between Spartina alterniflora (2n = 6x = 62; A-genome) and S. maritima (2n = 6x = 60; M-genome) gave rise to the homoploid hybrid (S. × townsendii, 2n = 6x = 62), and subsequently to the invasive allododecaploid species S. anglica (2n = 12x = 120–124) that has since spread worldwide. To address the question of dynamics of mixed ploidy populations involving these plants, we analysed several Spartina populations (fifty one individuals) in Southern England, UK, one of which was the presumed place of origin of the homoploid hybrid (Hythe). Using a combination of flow cytometry and ribosomal DNA (rDNA) genotyping we were able to identify the genomic composition and ploidy level of each individual analysed. The data show that the homoploid hybrid still dominates the population at Hythe (82 % of individuals collected in that locality) since its origin in the nineteenth century. We also identified S. × townsendii for the first time on Hayling Island (66 % individuals), indicating dispersal beyond its likely origin. The fertile allododecaploid S. anglica was mainly found in populations outside the initial hybridisation site, on Hayling Island and at Eling Marchwood. Quantification of the rDNA contributions from each parental genome showed that the ratios were mostly balanced in S. × townsendii. However, two (3 %) S. anglica individuals analysed have lost nearly all M-genome homeologs, indicating extensive repeat loss. Such variation indicates that despite the presumed single allopolyploid origin of S. anglica and genetic uniformity at other loci, it has undergone substantial changes at the rDNA loci following genome duplication.  相似文献

To establish a management plan for endangered and rare species, genetic assessment must first be conducted. The genetic characteristics of plant species are affected by demographic history, reproductive strategy, and distributional range as well as anthropological effects. Abies koreana E. H. Wilson (Pinaceae), Korean fir, is endemic to Korea and found only in sub-alpine areas of the southern Korean Peninsula and Jejudo Island. This species has been designated as critically endangered by the International Union for Conservation of Nature due to a continuous decline in its range and population fragmentation. We genotyped 176 individuals from seven natural populations and two afforested populations on the Korean Peninsula using 19 microsatellite loci. STRUCTURE analysis revealed two genetic clusters in natural populations (F st  = 0.040 and R st  = 0.040) despite low differentiation. We did not detect a significant reduction in genetic diversity or the signature of a genetic bottleneck despite population fragmentation and small population size. We deduced that this species exhibits a metapopulation structure, with the population on Jirisan Mountain acting as a source of genetic diversity for other local small populations on the Korean Peninsula, through contemporary asymmetric gene flow. However, the majority of afforested individuals on the Korean Peninsula originated from a different gene cluster. Thus, we recommend a conservation strategy that maintains two genetically unique clusters.  相似文献
