The melon fly, Bactrocera cucurbitae Coquillett, is a species of fruit flies of significant agricultural interest. Of supposed Indian origin, the melon fly is
now widely distributed throughout South East Asia up to China, while it has been recently eradicated from Japan. The population
structure of seven geographic populations from coastal China, as well as samples from other regions of South East Asia and
Japan, including lab colonies, have been studied using a 782 bp fragment of mitochondrial cytochrome oxidase I (COI) gene
sequence. The observed genetic diversity was exceedingly low, considering the geographic scale of the sampling, and one single
haplotype was found to be predominant from Sri Lanka to China. We confirm that Bactrocera cucurbitae exists in South East Asia as a single phyletic lineage, that Chinese populations are genetically uniform, and that no apparent
genetic differentiation exists between these and three available Japanese melon fly sequences. 相似文献

Population genetics is a powerful tool for invasion biology and pest management, and useful for a range of questions from tracing invasion pathways to informing management decisions with inference of population demographics. Genomics greatly increases the resolution of population-scale analyses, yet outside of model species with extensive genomic resources, few studies have used population genomics in invasion biology. We use genome-wide single nucleotide polymorphisms (SNPs) to investigate population genomic structure with samples from across the range of melon fly, Bactrocera cucurbitae (Coquillett, 1849), a highly polyphagous pest of commercial produce. We then make use of a chromosome-scale genome assembly and gene set to compare signatures of selection across the melon fly’s genome, both across our sampling as a whole and in the context of two independent, established introductions. Using multiple approaches, we find support for six genetic clusters across melon fly’s distribution. Some of these agree with previously identified genetic clusters using microsatellites, but consensus of clusters in mainland and oceanic southeast Asia is confounded by variable sampling between studies. We find few adaptive signatures across the genome, and virtually no unique signatures when comparing the two independent introductions, which suggests that similar management strategies are appropriate across melon fly’s range. This is the first use of genome-wide data to characterize population structure in tephritid fruit fly pests, and our SNP dataset provides a foundation for objective and cost-effective genotyping of previously collected melon fly specimens. Future research needs to focus on truly comprehensive sampling across melon fly’s range to overcome the historic variability of range-wide estimates of population structure for this pest. 相似文献

Rhagoletis completa Cresson (Diptera: Tephritidae) is native to North America and invaded Western Europe in the late 1980s, causing important damage to its principal host, walnut (Juglans spp.). In this review, we summarize the important elements of R. completa’s biology, phytosanitary status and methods used in Europe for its control, and then present the main conclusions associated with a completed risk analysis performed in 2014 to evaluate the dispersion and establishment potential of R. completa in Europe. The walnut husk fly was initially identified in Switzerland (1988) and Italy (1991), from where it spread to at least seven additional countries: France, Spain, Germany, Austria, Croatia, Slovenia and Hungary. R. completa has not reached the limits of its potential distribution. The main dissemination pathways within Europe include: (1) natural adult dissemination; (2) adult hitchhiker behaviour; and, to a lesser extent, (3) transportation of larvae in fresh fruits. R. completa host plants are widely distributed in Europe, either as isolated wild trees or in orchards, favouring the probability of fly establishment in currently fly-free areas. In addition, the European territories where Juglans species are present share biogeographic similarities. In orchards where R. completa is present and uncontrolled, 100% of walnut trees can be infested, causing losses in walnut yields of up to 80%. The negative effect is low (<10% yield loss) under phytosanitary control, although additional costs must also be considered to support specific monitoring for R. completa. The information presented here underlines a strong need for better walnut husk fly monitoring across European countries, as well as for increasing efforts to develop biological methods to control this emerging pest. 相似文献

The Ethiopian fruit fly, Dacus ciliatus, is an important pest of cucurbits, which recently invaded the Middle East. The genetics and cytogenetics of D. ciliatus have been scarcely studied. Such information is, however, an essential basis for understanding the biology of insect pests,
as well as for the design of modern control strategies. We report here the mitotic karyotype and detailed photographic maps
of the salivary gland polytene chromosomes of this species. The mitotic metaphase complement consists of six pairs of chromosomes,
including one pair of heteromorphic sex (XX/XY) chromosomes. The heterogametic sex is ascribed to the male. The analysis of
the salivary gland polytene complement shows a total number of five long chromosomes (10 polytene arms), which correspond
to the five autosomes of the mitotic nuclei, and a heterochromatic mass corresponding to the sex chromosomes. Banding patterns,
as well as the most characteristic features and prominent landmarks of each polytene chromosome are presented and discussed.
Chromosomal homologies between D. ciliatus and Bactrocera oleae are proposed by comparing chromosome banding patterns and by in situ hybridization of the hsp70 gene. 相似文献

Most studies on the sexual behaviour of tephritid fruit flies are conducted in laboratory or field cage settings. Here, we present field data on the mating behaviour of the melon fly, Bactrocera cucurbitae (Coquillett). A number of Tephritidae are lekking species, forming aggregations in which males fight to defend a small territory where they court females and mate. Our results confirm that males of the melon flies congregated and engaged in wing vibration in the late afternoon on non-host plants, such as Ficus carica, Cydonia oblonga and Prunus persica which bordered cucurbit fields. The nature and outcome of male-male aggressive encounters are described, and a high degree of female mate selectivity is documented. 相似文献

female flies. Herein we demonstrate that female D. alloeum wasps reared from either apple or hawthorn race R. pomenella larvae also deposit an OMP that reduces oviposition by conspecific female wasps. Significantly fewer wasps accepted fruit
that had received prior wasp oviposition and OMP or OMP alone without oviposition compared with control fruit for a minimum
of 7 days on both fruit types. Rinsing fruit with a 50% ethanol solution appeared to remove the OMP rendering fruit more acceptable
for oviposition than marked fruit that was not rinsed. Wasps of each host race were able to detect and avoid the OMP of the
sister race and fruit substrate type did not affect wasp response to the pheromone. The possibility of an internal marker

In many insect species including fruit flies (Diptera: Tephritidae), the behavioral responses to volatiles emitted by their host plants can be modulated by environmental conditions and by the physiological state of the insect. Here, we quantified (1) the effects of female age on the attraction of female melon fly, Zeugodacus cucurbitae (Diptera: Tephritidae) to volatiles emitted by intact and mechanically damaged tomato (Solanum lycopersicum), eggplant (S. melongena), zucchini (Cucurbita pepo), bitter melon (Momordica charantia), and cucumber (Cucumis sativus) fruit, and (2) the influence of time elapsed since fruit damage on the outcome. The investigations were conducted under semi-natural conditions in Hawai’i. Results from the first experiment revealed that, for freshly damaged tomato, eggplant, and zucchini, the level of female response was comparatively low and was not affected by female age. For bitter melon and cucumber, higher levels of response to freshly damaged fruit were documented, and the response levels gradually increased as female age increased from 1 to 4 weeks, reaching 70% for 4-week-old females exposed to cucumber odor. Results from our second experiment indicated that, on average, 56% of the females released responded in 20 min when cucumbers were freshly sliced, and the level of response was reduced 6-fold within a couple of hours. Females did not respond to tomato and cucumber odor when fruits were damaged >8 h before testing. Fruit volatiles involved in female Z. cucurbitae attraction seem to be released shortly upon mechanical damage and they are short-lived. The plasticity of the olfaction-driven behavior observed in Z. cucurbitae depending on female age and on the strength of the olfactory cues associated with preferred and less preferred hosts, adds another dimension to our understanding of the host-seeking behavior of this invasive species. 相似文献

In this contribution on the genus Cantharellus in Asia, C. subvaginatus is described from the Republic of Korea as a close relative to the Chinese C. vaginatus, which is here reported for the first time from India. Both species are here placed in Cantharellus subg. Cantharellus sect. Amethystini, together with the Indian C. pseudoformosus (syn.: C. umbonatus) and the Malayan C. subamethysteus. As such, Asia has suddenly become the continent with the highest diversity for Amethystini. Species delimitation in sect. Amethystini is molecularly supported by a combined phylogenetic analysis of rDNA sequences obtained for LSU and ITS and additionally suggests the existence of a still undescribed species in North America. Character variability is discussed for all known members of Amethystini, including atypical specimens of the North American C. lewisii that are morphologically more reminiscent of the South Korean C. subvaginatus. 相似文献

The Asian papaya fruit fly, Bactrocera papayae Drew and Hancock, was treated with hot-water immersion and forced hot air to develop a phytosanitary heat treatment schedule. Hot-water immersion tests were conducted with 12- and 24-h-old eggs and with first and third instar larvae to compare the relative thermotolerances of this fruit fly among these life stages. The 24-h-old eggs, the most thermotolerant among the four life stages tested, were subjected to time and temperature tests using cage-infested papaya fruits in a forced hot-air chamber. Heating the papayas to a minimum core temperature of 47.7 °C (95% confidence interval 47.2–48.3 °C) was estimated to induce probit-nine mortality based on a probit analysis of the data. Confirmatory tests in which papayas infested with 24-h-old eggs were heated to a minimum fruit core temperature of 47.2 °C that was maintained for 0–30 min followed by hydrocooling to a fruit core temperature of ≈25 °C resulted in the complete mortality of an estimated treated population of 43,425 eggs aged 24 h (99.9931% mortality at the 95% confidence level). Therefore, heating the papaya fruits to a core temperature of 47.2 °C for a minimum dwell time of 30 min, which was the longest dwell time in the confirmatory tests, may serve as a phytosanitary heat treatment schedule for the control of B. papayae in papaya fruits. 相似文献

The natural history and mating behavior of a species of tephritid fruit fly in the poorly studied genus Haywardina are described for the first time. Haywardina cuculi Hendel larvae were recovered over four field seasons from infested fruit of Vassobia breviflora (Sendtn.) Hunz, which constitutes a new host plant record for this species. Recovered pupae emerged as adults over 183 days on average, suggesting that most individuals became dormant. Adult flies engaged in sexual activity as soon as two days after emergence, were highly promiscuous, and displayed large variability in copulation duration. As for most tephritid species in the subtribe Carpomyini, H. cuculi exhibited a resource defense mating system. Fly activity peaked around noon. Copulation could last from 9 min to 17 h, with most copulations beginning in the afternoon and lasting until the following day. We discuss the potential significance of copulation duration variability in light of mate guarding and sperm competition hypothesis and outline future research to understand the evolution of life history and these behavioral strategies. 相似文献

A likely symbiotic association between tephritid fruit flies and gut bacteria has been recognized since the beginning of the last century. However, direct evidence for a link between gut bacteria and fruit fly fitness is still limited or absent for many species. Similar to other tephritids, the gut of Bactrocera minax (Enderlein) (Diptera: Tephritidae) is known to contain bacteria throughout the life stage, but what, if any, impact these bacteria have on B. minax fitness is entirely unknown. In order to elucidate the effects of bacteria on the fitness of B. minax, resident bacteria were isolated from the adult gut using culture-dependent techniques. Adult fly diets were subsequently supplemented with three bacterial isolates (Klebsiella pneumonia, Citrobacter braakii and Pantoea dispersa), or bacteria were removed from flies by antibiotics treatment: untreated adults provided a control. Adult fitness parameters (male and female longevity, female fecundity, male copulation number) were measured for the two treatments and one control group. Results were complex depending on the fitness parameter measured and the bacterial species. Compared to the controls, antibiotic treated B. minax had significantly decreased fecundity, but male and female longevity was increased. When flies were fed diets supplemented with any of the three bacterial isolates, female fecundity was significantly enhanced. However, only Citrobacter braakii significantly increased male mating frequency than control males. The results show that gut bacteria directly influence fitness of B. minax, but impacts are dependent on the bacterial species and the fitness parameters measured. 相似文献

Anastrepha fraterculus is an important pest of commercial fruits in South America. The variability observed for morphological and behavioural traits
as well as genetic markers suggests that A. fraterculus represents a complex of synmorphic species rather than a single biological species. We studied the correlation between geographical
distribution and genetic variation in natural populations from Argentina and south Brazil. Fragments of the mitochondrial
gene COII were sequenced in 28 individuals. The matrix of aligned sequences was phylogenetically analysed by parsimony, yielding
a cladogram of haplotypes. Based on Templeton’s nested method, no clade showed any geographic pattern for the gene COII, indicating
lack of significant association between haplotypic variability and geographic distribution. The analysis of nucleotide substitution
distances by Neighbour-Joining algorithm showed that geographically distant populations exhibit low genetic distances. The
corresponding trees clustered the populations without showing any geographic pattern. This result suggests that the populations

Five larval diets for laboratory rearing of Ceratitis capitata were tested. These diets were based on wheat bran, microcellulose, potato starch and agar. To evaluate the quality of diet,
pupal rearing efficacy and pupal weight were checked. The best results were obtained with an agar based diet used for Manduca sexta laboratory rearing. To simplify the preparation and to reduce the cost of this diet, a new formulation was developed. Larvae
reared on the new agar-based diet achieved higher pupal rearing efficacy than larvae reared on bran diet recently used in
medfly mass rearing facilities. Heat treated medflies reared on the new agar-based diet achieved similar pupal rearing efficacy
with heat treated medflies reared on bran diet. When testing population density, higher pupal rearing efficacy was again achieved
on new diet. The highest pupal rearing efficacy was achieved with 100 eggs per 25 g of diet, lowest with 500 eggs per 25 g
of diet. Concerning pupal weight, there was no difference in results achieved on Petri dishes with different larval population
densities. Larvae reared on new agar-based diet reached better results than larvae reared on bran diet. The preparation of
the new diet is simple and the cost is low, so it is good for laboratory tests and rearing. 相似文献

Two western Pacific triplefins, Enneapterygius fuscoventer Fricke 1997 and E. howensis Fricke 1997 (Perciformes: Tripterygiidae), are similar to each other in sharing 15–19 (usually 17) notched lateral-line scales and the mandibular pore formula 3–5 + 1 + 3–5 (usually 4 + 1 + 4), in addition to similar coloration, viz. body with four vertical bands, the first and second forked ventrally, dorsal-fin membrane semi-transparent, anal fin entirely blackish, and caudal fin blackish with a semi-transparent margin. These species have previously been known only from preserved specimens. Examination of additional specimens plus color photographs of males and females of both species when fresh, and comparisons with type specimens resulted in several features, including coloration and counts of second dorsal-fin spines, anal-fin soft rays, pored lateral-line scales and longitudinal scale rows, being regarded as new diagnostic characters. Enneapterygius fuscoventer and E. howensis have been newly recorded from southern Japan and coastal eastern Australia, respectively. 相似文献

Two repeated DNA sequences isolated from a partial genomic DNA library of Helianthus annuus, p HaS13 and p HaS211, were shown to represent portions of the int gene of a Ty3 /gypsy retroelement and of the RNase-Hgene of a Ty1 /copia retroelement, respectively. Southern blotting patterns obtained by hybridizing the two probes to BglII- or DraI-digested genomic DNA from different Helianthus species showed p HaS13 and p HaS211 were parts of dispersed repeats at least 8 and 7 kb in length, respectively, that were conserved in all species studied. Comparable hybridization patterns were obtained in all species with p HaS13. By contrast, the patterns obtained by hybridizing p HaS211 clearly differentiated annual species from perennials. The frequencies of p HaS13- and p HaS211-related sequences in different species were 4.3x10(4)-1.3x10(5) copies and 9.9x10(2)-8.1x10(3) copies per picogram of DNA, respectively. The frequency of p HaS13-related sequences varied widely within annual species, while no significant difference was observed among perennial species. Conversely, the frequency variation of p HaS211-related sequences was as large within annual species as within perennials. Sequences of both families were found to be dispersed along the length of all chromosomes in all species studied. However, Ty3 /gypsy-like sequences were localized preferentially at the centromeric regions, whereas Ty1/ copia-like sequences were less represented or absent around the centromeres and plentiful at the chromosome ends. These findings suggest that the two sequence families played a role in Helianthusgenome evolution and species divergence, evolved independently in the same genomic backgrounds and in annual or perennial species, and acquired different possible functions in the host genomes. 相似文献

A genetic transformation system has been developed for callus cells of Crataegus
aronia using Agrobacterium
tumefaciens. Callus culture was established from internodal stem segments incubated on Murashige and Skoog (MS) medium supplemented with
5 mg l−1 Indole-3-butyric acid (IBA) and 0.5 mg l−1 6-benzyladenine (BA). In order to optimize the callus culture system with respect to callus growth and coloration, different
types and concentrations of plant growth regulators were tested. Results indicated that the best average fresh weight of red
colored callus was obtained on MS medium supplemented with 2 mg l−1 2,4-dichlorophenoxyacetic acid (2,4-D) and 1.5 mg l−1 kinetin (Kin) (callus maintenance medium). Callus cells were co-cultivated with Agrobacterium harboring the binary plasmid pCAMBIA1302 carrying the mgfp5 and hygromycin phosphotransferase (hptII) genes conferring green fluorescent protein (GFP) activity and hygromycin resistance, respectively. Putative transgenic calli
were obtained 4 weeks after incubation of the co-cultivated explants onto maintenance medium supplemented with 50 mg l−1 hygromycin. Molecular analysis confirmed the integration of the transgenes in transformed callus. To our knowledge, this
is the first time to report an Agrobacterium-mediated transformation system in Crataegus

The study of several fungal pathogens has been aided by the use of invertebrate model hosts. Our efforts to infect the invertebrate
larvae Galleria
