This paper reports autecological field-studies in Singaporeon Drymoglossum piloselloides (L.) Presl., an epiphytic fernof the humid tropics which is capable of performing Crassulaceanacid metabolism (CAM). As indicated by the gas exchange patternsand by the occurrence of a diurnal malic acid rhythm, the plantalso features CAM in situ at its natural sites. Both in well-wateredand in naturally droughted plants external CO2 was taken upsolely during the night. Water stress decreased nocturnal CO2uptake,but left the synthesis and storage of malic acid unaffected.This indicates that CO2 recycling of respiratory CO2 by CAMis ecophysiologically important at the high night temperaturestypical of the tropical habitats of the fern. The plants showeda diel fluctuation of cell-sap osmotic pressure which paralleledthat of malic acid, while the fluctuation of the xylem tensionfollowed the curve of transpiration more closely than it followedthat of the malic acid content. CAM in D. piloselloides wasclearly not limited by natural access to mineral ions and nitrogen.It is concluded that the ecophysiological advantage of CAM forD. piloselloides lies in a better water use efficiency as comparedwith C3 ferns and in the salvaging of carbon by CO2 recycling. Gas exchange and organic acid accumulation of the C3-CAM intermediateClusia minor L. were investigated in response to various day/nighttemperatures and two light regimes (low and high PAR). For bothlight levels equal day/night temperatures between 20°C and30°C caused a typical C3 gas exchange pattern with all CO2uptake occurring during daylight hours. A day/ night temperatureof 15°C caused a negative CO2 balance over a 24 h periodfor low-PAR-grown plants while high-PAR-grown plants showeda CAM gas exchange pattern with most CO2 uptake taking placeduring the dark period. However, there was always a considerablenight-time accumulation of malic acid which increased when thenight-time temperature was lowered and had its maximum (54 mmolm–2) at day/night temperature of 30/15°C. A significantamount of malic acid accumulation (23 mmol m–2) in low-PAR-grownplants was observed only at 30/15°C. Recycling of respiratoryCO2 in terms of malic acid accumulation reached between 2·0and 21·5 mmol m–2 for high-PAR-grown plants whilethere was no significant recycling for low-PAR-grown plants.Both low and high-PAR-grown plants showed considerable night-timeaccumulation of citric acid. Indeed under several temperatureregimes low-PAR-grown plants showed day/night changes in citricacid levels whereas malic acid levels remained approximatelyconstant or slightly decreased. It is hypothesized that lowand high-PAR-grown plants have different requirements for citrate.In high-PAR-grown plants, the breakdown of citrate preventsphotoinhibition by increasing internal CO2 levels, whereas inlow-PAR-grown plants the night-time accumulation of citric acidmay function as an energy and carbon saving mechanism. The activity of photosynthesis and mitochondrial respiration,nocturnal organic acid accumulation and water relations wereinvestigated in Prenia sladeniana L. Bol. [malic enzyme (ME)-type]andCrassula lycopodioides Lam. [phosphoenolpyruvate carboxykinase(PEPCK)-type] to compare the physiological responses to waterdeficit in crassulacean acid metabolism (CAM) plants differingin their decarboxylating enzyme systems. Withholding water inhibiteddaytime gas exchange within 2 d while night time CO2gain andmalic acid accumulation remained relatively unchanged in bothspecies. In P. sladeniana, maximum photochemical efficiency(Fv/Fm) and photosynthetic electron transport declined to nearlythe same degree as CO2supply was restricted during drought.Despite limited CO2availability, photosynthetic activity waslargely unaffected in C. lycopodioides, as were mitochondrialproperties. There is no indication of a drought-induced increasein the capability to totally oxidize malate, yielding 4 CO2, in either species. Nevertheless, the enhanced ratio of malateto glycine oxidation may have increased the in vivo capabilityfor malate oxidation in P. sladeniana. Although pressure potentialwas maintained throughout the experiment in both species, activeosmotic adaptation occurred only inP. sladeniana. The observeddecrease in photosynthetic and mitochondrial activity may haveresulted from the large increase in osmotic concentration inthis species. We have been successful in building an energy-level model thatdescribes seed germination. We used the autocatalytic reactionrate equation to fit the germination rate for seed germination.The two parameters [A]0 and [F]0 were found by fitting the integratedgermination rate equation to the data. The values of [A]0 and[F]0 obey the Arrhenius equation and give activation energiesfor the second and third stage of the four-compartmental modelof seed germination. The thermodynamics of isothermal seed germinationis proposed and the enthalpy, entropy, and free energy are calculatedfor the transition from state A to state F. The time delay isa function of temperature and it leads to a rate constant thatcan be used to get the activation energy for the total germinationprocess. We believe the model is universal. Changes in respiration rate and in the contents of various constituentsduring the early period of germination of Phaseolus mungo seedswere studied. The course of the respiration developed in threephases. A sharp rise was observed in the first phase (Phasea), followed by the second phase (Phase b) of fairly constantrespiration rate. The respiration rate increases again in thethird phase (Phase c). The O2 uptake in Phases a and b was notinhibited by iodoacetate and fluoroacetate, while that in Phasec was inhibited. The contents of aspartic and glutamic acidsand amides were very high. A decrease of aspartic acid contentwas notable during the first few hours of germination. Citricand malic acids were the major organic acid constituents. Citricacid content in the seeds decreased, while that of malic acidremained unchanged. The leaching of malic acid into the soakingmedium was remarkable during the first 6 h of imbibition  相似文献

Organic acid metabolism and ethylene formation during controlledatmosphere storage (CA-storage) of apples (Mallus pumila MILLER,cv Rolls) were studied. A higher titratable acidity was observedin apples during CA-storage as compared to those in air control.The incorporation of atmospheric 14CO2 into malic acid was greaterin apples stored in the higher CO2 concentration. The conversionof succinic acid-14C into fumaric acid-14C was slightly lessin the apple in modified high carbon dioxide atmosphere thanthose in air. O2 uptake and CO2 output by apple slices weremarkedly inhibited by the addition of succinic and malic acidsat a concentration higher than 25 mM. These factors seem to be the possible cause of a higher acidityof fruits stored in CA-condition. Ethylene production from wholefruits or tissue slices was markedly inhibited under CA-condition. The retardation of acid metabolism and the inhibition of ethyleneproduction of apples during CA-storage seem to be the importantfactors which help to maintain their storage quality. Gibberellin A3 (GA3), kinetin and zeatin promoted germinationof conditioned witchweed (Striga asiatica [L.] Kuntze) seedssimilar to dl-strigol and a natural stimulant from corn rootexudates. GA3 promoted germination only with scarified seeds.However, these regulators, as well as dl-strigol and naturalstimulants, inhibited the conditioning of seeds and their subsequentgermination. On the other hand, abscisic acid induced 74% germinationof conditioned seeds but had no effect on seed conditioning.Other chemicals such as sucrose, NaNO3, vitamin B1, L-methionine,nicotinamide, indoleacetic acid and colchicine had no apparenteffect on seed conditioning, while sucrose, L-methionine andinositol induced about 30 % germination of conditioned seeds.Seeds generally required either a longer conditioning time ora higher terminal dl-strigol concentration to eliminate anyinhibition of seed conditioning induced by a compound. dl-Strigol, germination stimulation, parasitic plants, seed conditioning, seed germination, Striga asiatica (L.) The carbon balance of shade-grown Ananas comosus was investigatedwith regard to nitrogen supply and responses to high PAR. Netdark CO2 uptake was reduced from 61.2 to 38.5 mmol CO2 m–2in N limited (–N) plants grown under low PAR (60 µmolm–2 s–1) and apparent photon yield declined from0.066 to 0.034 (mol 02.mol–1 photon), although photosyntheticcapacities (measured under 5% CO2) were similar. Following transferfor 7 d to high PAR (600. µmol m–2 s–1), netCO2 uptake at night increased by 14% in +N plants, and daytimephotosynthetic capacity was higher, with a maximum value of7.8 µmol m–2 s–1. The magnitude of dark CO2 fixation during CAM was measured asdawn—dusk variations in leaf-sap titratable acidity (H+)and as the proportion of malic and citric acids. The contributionfrom re-fixation of respiratory CO2 recycling (measured as thedifference between net CO2 uptake and malic acid accumulation)varied with growth conditions, although it was generally lower(30%) than reported for other bromeliads. Assuming a stoichiometryof 2H+: malate and 3H+: citrate, there was a good agreementbetween titratable protons and enzymatically determined organicacids. The accumulation of citric acid was related to nitrogensupply and PAR regime, increasing from 7.0 mol m–3 (+Nplants) to 18 mol m–3 (–N plants) when plants weretransferred to high PAR; malate: citrate ratios decreased from13.1 to 2.5 under these conditions. Under the low PAR regime, leaf-sap osmotic pressure increasedat night in proportion to malic acid accumulation. However,following the transfer to high PAR for 7 d, there was a muchgreater depletion of soluble sugars at night which correspondedto a decrease in leaf-sap osmotic pressure. Although a rolefor citric acid in CAM has not been properly defined, it appearsthat the accepted stoichiometry for CAM in terms of gas exchange,titratable acidity, malic acid and osmotic pressure may nothold for plants which accumulate citric acid. Positive linear relationships were shown between constant temperaturesand the rates of progress of germination to different percentiles,G, for single populations of each of five genotypes of chickpea(Cicer anetinum L.). The base temperature, Tb, at which therate of germination is zero, was 0·0°C for all germinationpercentiles of all genotypes. The optimum temperature, To(G),at which rate of germination is most rapid, varied between thefive genotypes and also between percentiles within at leastone population. Over the sub-optimal temperature range, i.e.from Tb to To(G), the distribution of thermal times within eachpopulation was normal. Consequently a single equation was appliedto describe the influence of sub-optimal temperatures on rateof germination of all seeds within each population of each genotype.The precision with which optimum temperature, Tb(G), could bedefined varied between populations. In each of three genotypesthere was a negative linear relationship between temperatureabove Tb(G) and rate of germination. For all seeds within anyof these three populations thermal time at supra-optimal temperatureswas constant. Variation in the time taken to germinate at supra-optimaltemperatures was a consequence of normal variation in the ceilingtemperature, To(G)—the temperature at or above which rateof progress to germination percentile G is zero. A new approachto defining the response of seed germination rate to temperatureis proposed for use in germplasm screening programmes. In two populations final percentage germination was influencedby temperature. The optimum constant temperature for maximumfinal germination was between 10°C and 15°C in thesepopulations; approximately 15°C cooler than the optimumtemperature for rate of germination. A hybrid Phalaenopsis plant was grown hydroponically on a nutrientsolution of Hyponex in the greenhouse. Typical daily patternsof CO2 fixation, production of malic acid and citric acid andpH of the leaf and root were determined. The rate of primaryfixation of CO2 in the leaf increased markedly in the evening,remained high until well into the afternoon, then decreasedsharply. The pattern of production of organic acids resultedfrom the fact that the rate of uptake of CO2 gas was highestat night and fell during the day. A high correlation betweenpH value and level of malic acid in the leaves was observed,and an exponential relationship appeared to exist between theseparameters. These rhythmic sequences were not observed in theroot. The results suggest that metabolism in the leaf had characteristicsof CAM (Crassulacean Acid Metabolism), while the root did not. The role of ethylene during in vitro maturation of Nicotianatabacum pollen from the mld-binucleate (MB) stage was analysedby the addition of aminooxyacetic acid (AOA), aminoethoxyvinylglycine(AVG), CoCl2 and AgNO3 to the maturation medium (AMGLu). Anincrease in ethylene production was obtained in both isolatedpollen and pollen surrounded by sporophytic tissue during insitu maturation. in vitro maturation of pollen was inhibitedby AOA and AVG; ACC and ethrel were able to overcome this inhibitoryeffect. Cyclohexylamine (CHA) reverted the inhibition provokedby both Ag+ and Co2+ The results reported in this paper indicatethat ethylene is one of the factors implicated in in vitro maturationof MB pollen of Nicotiana tabacum. Factors concerning the chloroplast disposition in bundle sheathcells were investigated in finger millet (Eleusine coracanaGaertn.), and NAD malic enzyme type C4 plant with the centripetalarrangement of bundle sheath chloroplasts. Segments were cutfrom immature regions of emerging leaves in which the centripetalarrangement of bundle sheath chloroplasts had not yet been established.The leaf segments were floated on solutions with or withoutreagents. Sections were made of the segments at time intervalsand the distribution of bundle sheath chloroplasts was observedby light microscopy. The bundle sheath chloroplasts migratedto the vascular bundle and established a centripetal arrangementby 12-16 h in control solutions. Auxins, cycloheximide and cytochalasinB inhibited the disposition of bundle sheath chloroplasts whilechloramphenicol and colchicine had no effect. The inhibitoryeffect of auxins appeared only at early stages of chloroplastmigration while cycloheximide and cytochalasin B were effectiveeven at later stages. Dry lettuce seeds (Lactuca sativa L. cv. Grand Rapids), whichreceived 5 min far-red light (FR) 0.5 h after the onset of waterimbibition, showed 17% and 50% germination without and withacid immersion treatment (pH 0.1) for 1 h and rinsing with water,respectively. The acid treatment caused only 6% germinationor less in FR-treated seeds held for 10 to 30 d in dark storage.The 10 to 30 d skotodormant seeds did not respond to red light(R) or gibberellin A3 (GA3) singly, but showed 84% or higherpercentage germination if 1 h acid immersion was given beforeR or GA3. The 20 d skotodormant seeds, which received R treatmentat day 10 but remained dormant showed 89% germination with onlyacid treatment. Similar values were obtained with 30 d skotodormantseeds which received one or two R treatments at day 10 or 20,i.e. the only requirement for these R-treated dormant seedswas an acid immersion. This releases the skotodormancy and rendersthe seeds more sensitive to R or GA3, but the skotodormancywas initiated again if no light or hormone treatments were givenimmediately. The repetitive R or GA3 treatments, which did notcause skotodormant seeds to germinate, lessened the degree ofskotodormancy. The germination of these skotodormant seeds canonly be induced by the synergistic action of R and GA3. In thisstudy, GA3 caused higher germination percentages in R-treatedskotodormant seeds than R stimulated in GA3-treated seeds. Itis suggested that (i) repetitive R or Ga3 treatments maintaina high endogenous level of the far-red-absorbing form of phytochrome(Pfr) and GA activity, respectively, (ii) the accumulated stableintermediates of phytochrome persist in fully-imbibed skotodormantseeds for up to 20 d, without phytochrome expressing its functionuntil the seeds are acidified and (iii) a model is formulatedto interpret the results of acidification, growth promotersand R effects on germination of light-sensitive lettuce seeds. NADP malic enzyme (EC 1.1.1.40 [EC] ) from leaves of two C4 speciesof Cyperus (C. rotundus and C. brevifolius var leiolepis) exihibiteda low level of activity in an assay mixture that contained lowconcentrations of Cl. This low level of activity wasmarkedly enhanced by increases in the concentration of NaClup to 200 mM. Since the activity of NADP malic enzyme was inhibitedby Na2SO4 and stimulated by relatively high concentration ofTris-HCl (50–100 mM, pH 7–8), the activation ofthe enzyme by NaCl appears to be due to Cl. Variationsin the concentration of Mg2+ affected the KA (the concentrationof activator giving half-maximal activation) for Cl,which decreased from 500 mM to 80 mM with increasing concentrationsof Mg2+ from 0.5 mM to 7 mM. The Km for Mg2+ was decreased from7.7 mM to 1.3 mM with increases in the concentration of NaClfrom zero to 200 mM, although the increase of Vmax was not remarkable.NADP malic enzyme from Cyperus, being similar to that from otherC4 species, was able to utilize Mn2+. The Km for Mn2+ was 5mM, a value similar to that for Mg2+. The addition of 91 mMNaCl markedly decreased the Km for Mn2+ to 20 +M. NADP malicenzyme from Setaria glauca, which contains rather less Clthan other C4 species, was inactivated by concentrations ofNaCl above 20 mM, although slight activation of the enzyme wasobserved at low concentrations of NaCl at pH7.6. Use of the Weibull Function to Calculate Cardinal Temperatures in Faba Bean   总被引:1,自引:0,他引:1  
The onset of germination of faba bean seeds at constant temperaturewas progressively delayed as that temperature diverged froman optimum of 25.5 ?C. At temperatures below 10 ?C, or above28 ?C, the maximum germination percentage fell to below 90%.There was no germination at 39 ?C. Positive and negative linearrelationships were established between the constant temperaturesand the rates of progress of germination to different percentiles,at sub-optimal and supra-optimal temperatures, respectively.Like germination rates, base temperature (Tb) declined from3.71 to –0.83 ?C as the percentile value increased from10% to 80%. Caution was urged in extrapolating beyond the experimentaldata set. Differences in the ceiling temperature (Tc) with percentilecould not be discerned. Cumulative germination progress curves at each temperature weremodelled by the Weibull, logistic, and cumulative normal distributionfunctions. Cardinal temperatures (Tb and Tc) calculated fromthese data reasonably approximated the actual data. The Weibullfunction demonstrated a good approximation at all percentilelevels, while the logistic and cumulative normal distributionfunctions, as a result of their inherent symmetry, deviatedat the extreme percentiles. It was concluded that the Weibullfunction not only accurately modelled cumulative germinationbut could also be used in the calculation of cardinal temperatures. Distribution of iron-containing oxidases in aging nodal rootsof rice and wheat was studied. Activities of cytochrome c oxidase(1.9.3.1 [EC] , cytochrome c : O2 oxidoreductase), catalase (1.11.1.6 [EC] ,H2O2: H2O2 oxidoreductase) and peroxidase (1.11.1.7 [EC] , donor:H2O2 oxidoreductase) in wheat roots were comparatively higherthan were those in rice roots at corresponding stages. Cytochromec oxidase in roots remained active throughout the lives of therice and wheat crops. In rice roots, catalase seemed to playa distinct role around the panicle formation stage. Decay ofcatalase activity took place earlier than did that of peroxidaseand cytochrome c oxidase activities. In wheat roots similarenzyme activity changes were not observed. The effects of red light, far-red light, Gibberellin A3, andethephon were studied on the germination of lettuce seeds cv.Grand Rapids harvested at different stages of development. Seeds did not become capable of germination until 8 days afteranthesis. Red light promoted seed germination from the age of8–9 days following anthesis up to the newly mature stage.Ten or 11 days following anthesis, a large percentage of seedsbecame capable of germination in the dark and therefore couldbe considered not dormant. They were affected by far-red light,but less so than the mature seeds. The effect of light on the germination of developing seeds appearedto be similar to the known light effect on mature lettuce seedgermination. The effects of CO2 on dormancy and germination were examinedusing seeds of cocklebur (Xanthium pennsylvanicum Wallr.) andgiant foxtail (Setaria faberi Herrm.). The rate of germinationof the giant foxtail seeds as well as cocklebur was promotedby exogenously applied CO2 at a concentration of 30 mmol mol-1regardless of the sowing conditions. However, seeds which failedto germinate in the presence of CO2, entered a secondary phaseof dormancy under unfavourable germination conditions. If CO2was applied to seeds under conditions such as water stress imposedwith a 200 mol m-3 mannitol solution, a hypoxic atmosphere of100 mmol mol-1 O2 or a treatment of 0·1 mol m-3 ABA,development of secondary dormancy was accelerated. These contrastedeffects of CO2 were observed in ecological studies. Under naturalfield conditions germination of buried giant foxtail seeds respondedpositively to CO2 during a period of release from primary dormancyfrom Feb. to May, but CO2 accelerated secondary dormancy commencingin early Jun. In other words, in the presence of CO2, both theenvironmental conditions and the germination states of the seedsclearly showed secondary dormancy-inducing effects. The germination of pearl millet (Pennisetum typhoides S. &H.) seeds was investigated at constant temperatures between12 ?C and 47 ?C on a thermal gradient plate. The rate of germination increased linearly with temperaturefrom a base Tb to a sharply defined optimum To beyond whichthe rate decreased linearly with temperature, reaching zeroat Tm. The linearity of the response both above and below Toallowed time and temperature to be combined in a thermal timeat which a specified fraction of the seeds germinated. Cicer arietinum L. cv. G 62404 was seen to have a very low fruit-setpercentage at low temperatures and as temperatures increased,either in nature or in culture conditions, the fruit-set alsoincreased. This effect of temperature could also be seen inpercent pollen germination and pollen tube growth. M 450, amutant of G 62404, was seen to have a better fruit-set percentageat low temperature and better pollen germination. This may bedue to differences in malic acid concentration, which increasedpollen germination, and pollen tube growth under in vitro cultureconditions.  相似文献
