Summary Excretion of exocellular dd-carboxypeptidases was tested using 128 strains of streptomycetes. Exocellular enzyme activity was shown in 13% of the trains investigated. Streptomyces strains showed low activity of excretion of dd-carboxypeptidases: 2.7–4.8 M of released C-terminal d-alanine (d-Ala) residue/1 culture supernatant per minute. Saccharopolyspora erythraea mutants produced considerably higher levels of exocellular enzymes, the dynamics of excretion depending upon the medium used. The highest activity of exocellular dd-carboxypeptidase production was 44 M d-Ala/1 culture supernatant per minute. The affinity of exocellular dd-carboxypeptidase of S. erythraea 64-575 for -lactam antibiotics was assessed by a statistical computer programme. The enzyme showed the lowest affinity for sodium cefotaxime, ID50(M) = 7.5 × 10–6, and the highest for potassium cephalosporin C, ID50(M) = 5.0 × 10–9, ID50(M) representing the molar concentration of -lactan antibiotics which decreased by 50% the release of d-Ala. Piperidine is actively transported into the synaptosomal fraction of adult mouse brain. The transport mechanism appears to be Na+ independent but is temperature dependent and sensitive to ouabain. Analysis of kinetic experiments indicates only a low-affinity transport system to be present. By contrast the uptake ofD,L-[3H]pipecolic acid at a concentration of 4×10–7 M was temperature and Na+ dependent, ouabain sensitive, and revealed a two-component system with aK m =3.9±0.17×10–6 M,V max=129±6 pmol/mg protein/3 min for the high-affinity system and aK m =90.2±4.3×10–6 M,V max=2.45±0.19 nmol/mg protein/3 min for the low-affinity system. Compounds structurally related to pipecolic acid such as glycine,l-proline, 4-amino-n-butyric acid, and 5-amino-n-valeric acid showed an inhibitory effect on uptake at a concentration of 10–4 M. The demonstration of biosynthesis of pipecolic acid in mouse brain and the presence of a high-affinity sodium-dependent uptake system suggest a physiological role of this substance in the central nervous system.  相似文献

Conditions for maximising transient expression of GUS in leaf mesophyll protoplasts of Arabidopsis thaliana ecotype C24 were investigated. It was found that the factors most influencing expression levels, with optimum levels in parenthesis, were plasmid DNA quantity (100 g per 5 × 105 protoplasts), inclusion of carrier DNA (50 g), PEG pH and amount (pH above 6, and total PEG concentration at least 9% w/w) and the topological form of the DNA. The ribosomal RNA genes from the sea urchin Lytechinus variegatus have been studied with the electron microscope using the technique of denaturation mapping. A repeating pattern of denatured regions was found with an average repeat length of 3.87±0.24m. DNA obtained by a gentle lysis procedure from adult Drosophila melanogaster was analyzed by sucrose gradient sedimentation. The major portion of the DNA has an estimated weight of at least 5–10×109 daltons. All of the ribosomal genes are present in this high molecular weight DNA in adult males with one nucleolus organizer or in adult females with two nucleolus organizers as shown by hybridizing fractions of the gradient with ribosomal RNA. In female adults with one nucleolus organizer instead of the usual two, 68% of the ribosomal genes are found in high molecular weight DNA and 32% are found in DNA of smaller size (3×108 daltons). We propose that these latter genes are not integrated into the DNA of the chromosome.  相似文献

Two cystatins were purified from tissue extract of bovine brain by alkaline treatment, acetone fractionation, gel chromatography on Sephadex G-75, and affinity chromatography on S-carboxymethyl-papain-Sepharose. One of the inhibitors had a relatively high molecular mass, 25 kDa (HMM-cystatin) with pI 4.7, and the other, 11 kDa (LMM-cystatin) with pI 5.23. Both inhibitors showed considerable stability at pH 2 and 80°C. The cystatins inhibited papain, ficin, and cathepsins B and H, but not trypsin, chymotrypsin, thermolysin, nagarse, and cathepsin D. Ki values for the complexes of papain and the inhibitors were estimated to be 2.8×10–10 M for HMM-cystatin and 1.3×10–9 M for LMM-cystatin. Summary Apical membrane vesicles from human term placenta were isolated using a magnesium precipitation technique, and the purity of the vesicles was assessed morphologically using scanning and transmission electron microscopy, and biochemically, using marker enzymes. The vesicles were found to be morphologically intact and significantly enriched in enzymes associated with apical membranes. 36Cl uptake into these vesicles was studied in the presence of an outwardly directed Cl gradient. This uptake was found to be time dependent, with an initial rapid uptake tending to peak between 10 and 20 min and thereafter decline. Uptake was found to be voltage dependent since 5 m valinomycin caused a decrease in uptake. The effects of N-phenylanthranilic acid (NPA) and 4,4-diisothiocyanostilbene-2,2-disulphonic acid (DIDS) and bumetanide on the initial rate of Cl were examined in the presence and absence of 5 m valinomycin. NPA and DIDS inhibited isotope uptake strongly with IC50 values of 0.83±0.35 m and 3.43±0.37 m, respectively, in the absence of valinomycin. Although valinomycin reduced 36Cl uptake by about 80% when added before the isotope, DIDS reduced the uptake which remained in a concentration-dependent fashion with an IC50 of 5.6±2.1 m. Under these conditions, NPA was without effect at concentrations below 100 m. Bumetanide was without effect at the concentrations used in the absence of valinomycin. However, following valinomycin pretreatment, bumetanide reduced 36Cl uptake significantly at 100 m concentration. Vesicle diameter, as assessed by flow cytometry, did not change under the conditions employed.The effects of some fatty acids were also investigated. Arachidonic acid and linoleic acid inhibited Cl uptake with IC50 values of 37.6±14.9 m and 4.59±0.51 m, respectively. Arachidonyl alcohol and elaidic acid were found to be without effect. These studies show that human placental brush border membrane vesicles possess a chloride conductance channel, the activity of which can be measured in the presence of an outwardly directed Cl gradient and this channel is sensitive to Cl channel inhibitors, especially N-phenylanthranilic acid, and can be inhibited by unsaturated fatty acids such as arachidonic acid and linoleic acid.This work was supported in part by the Cystic Fibrosis Association of Ireland and Eolas, The Irish Science and Technology Agency. Summary To study Cl conductive and cotransport mechanisms, primary cultures of canine tracheal cells were grown to confluency on thin glass cover slips and on porous filters. Transepithelial resistance was >100 ·cm2, and short circuit current (I sc=2–20 A/cm2), representing active secretion of Cl, increased >threefold with addition of 10 m isoproterenol to the serosal solution. Cells made transiently permeable in hypotonic solution were loaded with the Cl-sensitive fluorophore 6-methoxy-N-(3-sulfopropyl) quinolinium (SPQ) (5mm, 4 min, 150 mOsm). The electrical properties of the cell monolayers were not altered by the loading procedure. Intracellular SPQ fluorescence was monitored continuously by epifluorescence microscopy (excitation 360±5 nm, emission>410 nm). SPQ leakage from the cells was <10% in 60 min at 37°C. Intracellular calibration of SPQ fluorescencevs. [Cl] (0–90mm) was carried out using high-K buffers containing the ionophores nigericin (5 m) and tributyltin (10 m); SPQ fluorescence was quenched with a Stern-Volmer constant of 13m –1. Intracellular Cl activity was 43±4mm. Cl flux was measured in response to addition and removal of 114mm Cl from the bathing solution. Addition of 10 m isoproterenol increased Cl efflux from 0.10 to 0.27mm/sec. The increase was inhibited by the Cl-channel blocker diphenylamine-2-carboxylic acid (1mm). In the absence of isoproterenol, removal of external Na or addition of 0.5mm furosemide, reduced Cl influx by >fourfold. In ouabain-treated monolayers, removal of external K in the presence of 5mm barium diminished Cl influx by >twofold, suggesting that Cl entry is in part K dependent. These results establish an accurate optical method for the realtime measurement of intracellular Cl activity in tracheal cells that does not require an electrically tight cell monolayer. The data demonstrate the presence of an isoproterenol-regulated Cl channel and a furosemide-sensitive cation-coupled transport mechanism.  相似文献

Summary A new DNA polymorphism assay was developed in 1990 that is based on the amplification by the polymerase chain reaction (PCR) of random DNA segments, using single primers of arbitrary nucleotide sequence. The amplified DNA fragments, referred to as RAPD markers, were shown to be highly useful in the construction of genetic maps (RAPD mapping). We have now adapted the RAPD assay to tomato. Using a set of 11 oligonucleotide decamer primers, each primer directed the amplification of a genome-specific fingerprint of DNA fragments. The potential of the original RAPD assay to generate polymorphic DNA markers with a given set of primers was further increased by combining two primers in a single PCR. By comparing fingerprints of L. esculentum, L. pennellii, and the L. esculentum chromosome 6 substitution line LA1641, which carries chromosome 6 from L. pennellii, three chromosome 6-specific RAPD markers could be directly identified among the set of amplified DNA fragments. Their chromosomal position on the classical genetic map of tomato was subsequently established by restriction fragment length polymorphism (RFLP) linkage analysis. Isolated embryos ofKarwinskia humboldtiana were cultured in vitro. The growth of embryos and development to plantlets on woody plant medium supplemented with indole-3-acetic acid 6.10-2 mol l–1, gibberellic acid (GA3) 3.10-2 mol l–1, and 6-benzylaminopurine (BA) 2 mol l–1 was obtained. Multiplication of shoots and rooting of excised shoots has been achieved. Cr(VI) tolerance was studied in four strains of Rhodosporidium toruloides and compared with that of a fifth strain, DBVPG 6662, isolated from metallurgical wastes and known to be Cr(VI) resistant. Tolerance was studied in relation to different species of sulfur (sulfates, thiosulfates, methionine, cysteine) at different concentrations. Djenkolic acid, a poor source of sulfur and an activator of sulfate transport, was also considered. In synthetic medium all strains except the Cr(VI)-resistant one started to be inhibited by 10 g ml (0.2 mm) Cr(VI) as K2Cr2O7. DBVPG 6662 was inhibited by 100 g ml (2.0 mm) Cr(VI). In Yeast Nitrogen Base without amino acids (minimal medium), supplemented with varying concentrations of chromate, all Cr(VI)-sensitive strains accumulated concentrations of total chromium (from 0.8 to 1.0 g mg cell dry wt) after 18 h of incubation at 28 °C. In minimal medium supplemented with 10 g ml Cr(VI), the addition of sulfate did not significantly improve the yeast growth. Cysteine at m levels increased tolerance up to 10 g ml, whereas methionine only reduced the Cr(VI) toxicity in the strain DBVPG 6739. Additions of djenkolic acid resulted in increased Cr(VI) sensitivity in all strains. The best inorganic sulfur species for conferring high tolerance was thiosulfate at concentrations up to 1 mm. In all cases increased Cr(VI) tolerance was due to a significantly reduced uptake in the oxyanion by the cells and not to the chemical reduction of Cr(VI) to Cr(III) by sulfur compounds.  相似文献

Summary Osmium tetroxide and OTAN histochemical reactions have been reappraised following recent publications in this journal by Ellbder and Lojda. Provided that the standard OTAN reaction is used on frozen sections of conventional thickness, unsaturated hydrophilic polar lipids are stained in an orange or red shade while unsaturated hydrophobic non-polar lipids are stained brown-black or black. We have confirmed the anomaly — pointed out by Elleder and Lojda — that the polar lipid reaction of the atherosclerotic plaque is essentially extinguised by acetone. Solubility and blockading methods provide circumstantial evidence that Elleder and Lojda's assumed OsO4-protein reactions are due to the lipid moiety of a lipoprotein complex.  相似文献

Summary A brief review is presented of the Vester-Ulbricht -decay Bremsstrahlen hypothesis for the origin of optical activity, and of subsequent experiments designed to test it. Certain of our experiments along these lines, begun in 1974 and involving the irradiation of racemic and optically active amino acids in a 61.7 KCi90Sr–90Y Bremsstrahlen source, have now been completed and are described. After 10.89 years of irradiation with a total Bremsstrahlen dose of 2.5×109 rads, crystallinedl-leucine, norleucine, and norvaline suffered 47.2, 33.6, and 27.4% radiolysis, respectively, but showed no evidence whatsoever of asymmetric degradation.d- andl-Leucine underwent about 48% radiolysis and showed 2.4–2.9% radioracemization. Other samples in solution were too severely degraded to analyze. Probable intrinsic reasons for the failure of the Vester-Ulbricht mechanism to afford asymmetric radiolysis in the present and related experiments involving -decay Bremsstrahlen are enumerated.A portion of this material was presented at the 7th International Conference on the Origins of Life, Mainz, FRG, July 10–15, 1983  相似文献

Summary Euglena gracilis, strain Z, was grown in synchronous culture. Carbon source used was either dl-lactic acid (L) or a mixture of l-glutamic and dl-malic acids (GM). Synchronisation was obtained by transfering the cells in the exponential growth phase, at regular intervals—each 3 days—to a fresh medium.Respiration (measured during a whole cell cycle, 12 h) was 20±6 l/H/106 cells on (GM) and 46±7 l/H/106 cells on (L) medium. At the same time as the increased rate of oxygen uptake on lactate medium, we observed the appearance of a giant chondriome in the cells. On glutamate—malate containing medium the size of mitochondria was normal.

Summary The molecular properties of the P-group plasmids R26, R527 and R18-18 (a carbenicillin-sensitive derivative of R18) have been compared with those of RP1. R18-18 and RP1 have a MW about 38×106 daltons, and R26 and R527 of 52×106 daltons (determined from contour lengths). All three plasmids have a buoyant density similar to that of RP1 (1.719 g/cm3, 60% G+C). From their molecular and phenotypic similarities, these plasmids probably represent two pairs of identical or closely similar elements.Resistant bacteria are not recovered following F116L-mediated transduction of R26 (or R527), and this correlates with the plasmids' larger size (phage genome=40×106 daltons). Fragments of R26 are, however, transduced and their resistance determinants may be rescued by recombination if the recipient harbours R18-18. Such events are accompanied by an increase in the size of the recipient plasmid from 38×106 to 52×106 daltons following inheritance of the resistance determinants Sm Su Gm Hg, but not Cb. Thus, Sm Su Gm Hg are encoded in a DNA segment of MW about 14×106 daltons which apparently has no homologous region on R18-18. Since a piece of DNA of this MW also corresponds to the difference in size between R26 and R18-18, it is possible that the former is derived from an RP1-like element which has acquired these additional resistance determinants.  相似文献

Reassociation kinetics of DNA from the macronucleus of the ciliate, Tetrahymena pyriformis GL, has been studied. The genome size determined by the kinetic complexity of DNA was found to be 2.0×108 base pairs (or 1.2×1011 daltons). About 90% of the macronuclear DNA fragments 200–300 nucleotides in length reassociate at a rate corresponding to single-copy nucleotide sequences, and 7–9% at a rate corresponding to moderate repetitive sequences; 3–4% of such DNA fragments reassociate at C0t practically equal to zero. To investigate the linear distribution of repetitive sequences, DNA fragments of high molecular weight were reassociated and reassociation products were treated with Sl-nuclease. DNA double-stranded fragments were then fractionated by size. It has been established that in the Tetrahymena genome long regions containing more than 2000 nucleotides make up about half of the DNA repetitive sequences. Another half of the DNA repetitive sequences (short DNA regions about 200–300 nucleotides long) intersperse with single-copy sequences about 1,000 nucleotides long. Thus, no more than 15% of the Tetrahymena genome is patterned on the principle of interspersing single-copy and short repetitive sequences. Most of the so called zero time binding or foldback DNA seem to be represented by inverted self-complementary (palindromic) nucleotide sequences. The conclusion has been drawn from the analysis of this fraction isolated preparatively by chromatography. About 75% of the foldback DNA is resistant to Sl-nuclease treatment. The Sl-nuclease resistance is independent of the original DNA concentration. Heat denaturation and renaturation are reversible and show both hyper and hypochromic effects. The majority of the inverted sequences are unique and about 20% are repeated tens of times. According to the equilibrium distribution in CsCl density gradients the average nucleotide content of the palindromic fraction does not differ significantly from that of total macronuclear DNA. It was shown that the largest part of this fraction of the Tetrahymena genome are not fragments of ribosomal genes.  相似文献

Microdissected Deiters' neuron plasma membranes have been used for studying the passage of GABA through the membrane both in the inward and outward direction. Working with 0.2 mM GABA in the compartment simulating the outside of the neurone and with 2.0 mM GABA in the one simulating the inside we found a net transport of GABA towards the inside. This mechanism does not require a Na+ ion gradient across the membrane. The nature of the transport process involved was studied by determining the rate of [3H]-GABA inward passage as a function of GABA concentration (1 nM–800 M) on the outward side of the membrane. The results have shown that until 50 M a diffusion process (v=D1×C, where D1=3.1×10–11 1/m2×sec) is the sole mechanism involved. Above 50 M a second diffusion process is activated v=D2×(C–50×10–6), where D2=2.8×10–11 1/m2×sec. Taking in account both inward and outward directed diffusion, one can calculate 16 M as the equilibrium concentration of GABA on the outward side of the membrane. From a kinetic point of view, these diffusion processes are able to reduce GABA concentration in a synaptic cleft from 3 mM to 20 M within 3 sec. These diffusion systems are discussed as extremely efficient in removing the excess of released GABA in the synaptic cleft.  相似文献

Genetic mapping of QTLs controlling horticultural traits in diploid roses   总被引:9,自引:0,他引:9  
A segregating progeny set of 96 F1 diploid hybrids (2n=2x=14) between Blush Noisette (D10), one of the first seedlings from the original Champneys Pink Cluster, and Rosa wichurana (E15), was used to construct a genetic linkage map of the rose genome following a pseudo-testcross mapping strategy. A total of 133 markers (130 RAPD, one morphological and two microsatellites) were located on the 14 linkage groups (LGs) of the D10 and E15 maps, covering total map lengths of 388 and 260 cM, respectively. Due to the presence of common biparental markers the homology of four LGs between parental maps (D10-1/E15-1 to D10-4/E15-4) could be inferred. Four horticulturally interesting quantitative traits, flower size (FS), days to flowering (DF), leaf size (LS), and resistance to powdery mildew (PM) were analysed in the progeny in order to map quantitative trait loci (QTLs) controlling these traits. A total of 13 putative QTLs (LOD>3.0) were identified, four for FS, two for flowering time, five for LS, and two for resistance to PM. Possible homologies between QTLs detected in the D10 and E15 maps could be established between Fs1 and Fs3, Fs2 and Fs4, and Ls1 and Ls3. Screening for pairwise epistatic interactions between loci revealed additional, epistatic QTLs (EQTLs) for DF and LS that were not detected in the original QTL analysis. The genetic maps developed in this study will be useful to add new markers and locate genes for important traits in the genus providing a practical resource for marker-assisted selection programs in roses.  相似文献

Cytological abnormalities are observed in salivary chromosomes of stocks of Drosophila melanogaster possessing DNA-induced, v + transformations mapping either at 10.0 or 133.0. In initial studies, the untransformed control stock and six transformed stocks were assigned code numbers prior to cytological examination. Salivary chromosome regions 1B9 · 10—1C2 · 3 and 10A1 · 2—10B1 · 2 were carefully examined in each of the coded stocks. (Band 1B11 is tentatively identified as the site of the suppressor-of-sable locus at 10.0 and band 10A1 is the site of the vermilion locus at 133.0.) When cytological studies were complete each of the stocks was identified. In every transformed stock examined, anomalies had been scored in association with the chromosome region corresponding to the map position of the DNA-induced alteration. In the control stock, anomalies were observed at neither position. — Approximately 10–15% of the nuclei in transformed stocks exhibit significant departure from normality in the pertinent chromosome region. The perturbations range from minor alterations of banding pattern to apparent pieces of extra chromatin in the most extreme cases. In stocks with v+ transformations mapping at 133.0, apparent extra chromatin is observed with frequencies varying from 0.05 to 0.02. In these cases the abnormal structures are associated with salivary band 10A1 · 2, forming a band-like structure, frequently an almost perfect doublet (open or closed), often lying in an abnormal position, and with fine chromatin threads connecting to the chromosomal doublet. In stocks with v+ transformations mapping at 10.0, apparent extra chromatin is observed with a frequency of about 0.001. These abnormal structures are frequently thread-like, lying to the side or off the tip of the chromosome, with compact regions which sometimes resemble chromomeres, and with fine threads connecting to the chromosomal 1B11 region.  相似文献

Summary An X-ray diffraction analysis ofZ-l-Leu-Aib-Gly-l-Ile-l-Leu-OMe, containing the N-acylated tetrapeptide amide sequence-l-Leu-Aib-Gly-l-Ile-, showed that in the crystal state the carbonyl group preceding thel-Leu1 residue acts as the acceptor of two C=OH–N intramolecular H-bonds, which give rise to an-l-Leu1-Aib2-type-III' -turn and an-l-Leu1-Aib2-Gly3-l-Ile4--turn, respectively. A second (type-I') -turn encompasses the-Aib2-Gly3-sequence. This is the third type of folding motif known for that tetrapeptide sequence, considering also those already published for the C-terminal segment of the lipopeptaibol antibiotics trichodecenin I and trichogin A IV.  相似文献
