Aims Forests represent the most important component of the terrestrial biological carbon pool and play an important role in the global carbon cycle. The regional scale estimation of carbon budgets of forest ecosystems, however, have high uncertainties because of the different data sources, estimation methods and so on. Our objective was to accurately estimate the carbon storage, density and sequestration rate in forest vegetation in Jilin Province of China, in order to understand the role of the carbon sink and to better manage forest ecosystems. Methods Vegetation survey data were used to determine forest distribution, size of area and vegetation types regionally. In our study, 561 plots were investigated to build volume-biomass models; 288 plots of shrubs and herbs were harvested to calculate the biomass of understory vegetation, and samples of trees, shrubs and herbs were collected to analyze carbon content. Carbon storage, density and sequestration rate were estimated by two forest inventory data (2009 and 2014), combined with volume-biomass models, the average biomass of understory vegetation and carbon content of vegetation. Finally, the distribution patterns of carbon pools were presented using ArcGIS soft ware. Important findings Understory vegetation biomass overall was less than 3% of the tree layer biomass, varying greatly among different forest types and even among the similar types. The carbon content of trees was between 45.80% 52.97%, and that of the coniferous forests was higher than that of the broadleaf forests. The carbon content of shrub and herb layers was about 39.79% 47.25% and 40%, respectively. Therefore, the vegetation carbon conversion coefficient was 0.47 or 0.48 in Jilin Province, and the conventional use of 0.50 or 0.45 would cause deviation of ±5.26%. The vegetation carbon pool of Jilin Province was at the upper range of regional carbon pool and had higher capacity of carbon sequestration. The value in 2009 and 2014 was 471.29 Tg C and 505.76 Tg C, respectively, and the total increase was 34.47 Tg C with average annual growth of 6.89 Tg C•a1. The corresponding carbon sequestration rate was 0.92 t•hm 2•a1. The carbon density rose from 64.58 t•hm 2 in 2009 to 66.68 t•hm2 in 2014, with an average increase of 2.10 t•hm2. In addition, the carbon storage of the Quercus mongolica forests and broadleaved mixed forests, accounted for 90.34% of that of all forests. The carbon increment followed the order of young > over-mature > near mature > middle-aged > mature forests. The carbon sequestration rate of followed the order of over-mature > young > near mature > middle-aged > mature forests. Both the carbon increment and the carbon sequestration rate of mature forests were negative. Furthermore, spatially the carbon storage and density were higher in the east than in the west of Jilin province, while the carbon increment was higher in northeast and middle east than in the west. The carbon sequestration rate was higher in Tonghua and Baishan in the south, followed by Jinlin in the middle and Yanbian in the east, while Baicheng and Songyuan, etc. in west showed negative values.  相似文献

The sulfate reduction rate was measured for almost four years in the profundal sediments of Lake Kizaki, a mesotrophic lake in central Japan. The rate was generally highest in the surface layer and decreased with depth. Seasonally, sulfate reduction tended to be high in spring and summer, and then to decrease until the end of stratification (December) in spite of a constant in situ temperature of around 6 °C, although fluctuations were found in every year. The rate also fluctuated greatly according to year. The maximum rate of sulfate reduction was 0.33 mmol m−2 d−1 in May, 1990, and the minimum was 0.004 mmol m−2 d−1 in March, 1993. These relatively low rates, compared with those reported for freshwater sediments, seem to be due to low concentrations of sulfate in the sediments (5–23 μmol l−1 in the surface layer). The rate was highly correlated with the concentration of sulfate in the sediments. The addition of sulfate stimulated sulfate reduction in all sediment samples tested, but adding lactate did not. Therefore, sulfate reduction should be limited mainly by the supply of sulfate. Measurements of sulfate reduction rates at different concentrations of added sulfate revealed a low concentration of half-saturation constant as low as 12 μmol l−1.  相似文献

The aim of this study was to investigate the role of β-adrenergic receptors in modulating associative long-term depression (LTD) at CA1 synapses in rat hippocampal slices. Standard extracellular electrophysiological techniques were employed to record field excitatory post-synaptic potential (fEPSP) activity and to induce associative LTD. Two independent Schaffer collateral pathways were elicited in hippocampal CA1 areas. In one (weak) pathway, the stimulating intensity was adjusted to elicit small fEPSP activity (20–30% of the maximum response). In contrast, 80–90% of the maximum response was evoked in the other (strong) pathway. Associative LTD of weak pathway could be induced by paired stimulation of weak and the strong pathways, repeated 100 times at 0.167 Hz. The associative LTD of weak pathway was NMDA receptor- and phophatase 2B dependent, because bath application of 50 μM D, L-AP5 or 10 μM cypermethrin blocked its induction. Bath application of 1 μM isoproterenol inhibited associative LTD, and this effect was blocked by timolol, suggesting the involvement of β-adrenergic receptors. The inhibitory effect of β-adrenergic receptors on LTD induction was blocked in slices pretreated with inhibitors of protein kinase A and mitogen-activated protein kinase, suggesting that these signal cascades are downstream effectors following activation of β-adrenergic receptors. Nevertheless, bath application of timolol or cypermethrin alone did not have significant effect on associative LTD induction, suggesting neither endogenous function of β-adrenergic receptor nor endogenous PKA activity does have a role in associative LTD induction.  相似文献

The change of cholinergic transmission of ?-amyloid protein (β-AP) treated rats was studied by intracerebral microdialysis sampling combined with HPLC analysis. β-AP1—40 was injected into nucleus basalis magnocellularis (NBM). Passive avoidance response test (step-down test) and delayed alternation task were used for memory testing. The impairment of memory after injection of β-AP1—40 into NBM exhibited mainly the deficiency of short-term working memory. One week after injection of β-AP1—40 the release of acetylcholine (ACh) from frontal cortex of freely-moving rats decreased significantly, and the response of cholinergic nerve ending to the action of high [K+] solution was rather weak. In control animals the percentage of increase of ACh-release during behavioral performance was 57%, while in β-AP1—40-treated rats it was 34%. The temporary increase of the ACh-release of the rat put into a new place was also significantly diminished in β-AP1—40-treated rats. The results show that the injection of β-AP1—40 into NBM impairs the cholinergic transmission in frontal cortex, and the impairment of cholinergic transmission may be the main cause of the deficit of working memory.  相似文献

Aims Our objectives were to study the spatial distribution of soil organic carbon (SOC) density and its influencing factors in the main forest ecosystems in Guangxi. Methods A total of 345 sample plots were established in Guangxi, and the size of each plot was 50 m × 20 m. Based on the forest resource inventory data and field investigation, the SOC storage of the main forests in Guangxi was estimated. Geostatistics was applied to analyze the spatial pattern of SOC density and the main influencing factors on SOC density were also explored by principal component analysis and stepwise regression. Important findings The total SOC storage in the main forests in Guangxi was 1 686.88 Tg, and the mean SOC density was 124.70 Mg•hm2, which is lower than that of China. The best fitted semivariogram model of SOC density was exponential model, and the spatial autocorrelation was medium. The contour map based on Kriging indicated that northeastern Guangxi had high SOC density and northwestern Guangxi had low SOC density, which corresponded to high SOC density in non-karst region and low SOC density in karst region. The SOC density followed the sequence of bamboo forest > deciduous broadleaf forest > warm coniferous forest > mixed evergreen and deciduous broadleaf forest > evergreen broadleaf forest, and yellow soil > red soil >lateritic red soil > limestone soil. The dominant environment factors affecting SOC density included soil depth, longitude, latitude, and altitude. Soil depth was the most influential factor, which was mainly attributed to the karst landscape.  相似文献

Summary. The aim of this study was to evaluate the effect of pegylated interferon-alpha (PEG-IFN-α) on the plasma citrulline/arginine ratio, regarded as an index of nitric oxide (NO) synthesis, in patients with high-risk melanoma. Forty patients were randomly assigned to either PEG-IFN-α treatment (n = 22) or to observation only (control group, n = 18). The treatment group received 6 μg PEG-IFN-α/kg once a week during 8 weeks, followed by a maintenance dose of 3 μg/kg/wk. Blood was collected at different time points, plasma concentrations of citrulline and arginine were measured and the ratio of citrulline/arginine was calculated. Patients treated with PEG-IFN-α showed a significant decrease in the concentrations of citrulline and in the citrulline/arginine ratio during the whole study period, both compared to baseline values and to the control group. The data suggest that therapy with PEG-IFN-α results in a marked decrease in the synthesis of NO in melanoma patients.  相似文献

The native Goα was purified from bovine brain cortex and palmitoylated in vitro. The in vitro palmitoylation site was the same as that in vivo. The internal palmitoylation of purified native Goα was found to be largely maintained. The apparent palmitoylation ratio was significantly increased after the Goα was treated with DTT. The GTPγS binding characteristic of Goα was not influenced by palmitoylation, however, the affinity for LUVs was increased dramatically. The in vitro palmitoylation model of Goα provides a better basis for studying the functional role of G protein palmitoylation in signal transduction.  相似文献

In the present study, the relationship between the nutritional status of leaves and the development of symptoms of cotton leaf curl virus (CLCuV) in two cotton (Gossypium hirsutum L.) cuItlvars (I.e. CIM-240 and S-12) was Investigated. The incidence of disease attack was found to be 100% In the S-12 cuItlvar and 16% in the CIM-240 cuItivar. Geminivirus particles in infected leaves were confirmed by transmission electron microscope examination of highly specific geminivirus coat protein antlsera-treated cell sap. The CLCuV Impaired the accumulation of different nutrients in both cuItivars. A marked decrease in the accumulation of Ca^2+ and K^+ was observed in infected leaves. However, the disease had no effect on leaf concentrations of Na^+, N, and P. It was observed that the curling of leaf margins in CLCuV-Infected plants was associated with the leaf Ca^2+ content; leaf curling was severe in plants with a significant reduction In Ca^2+ content. Moreover, leaf K&+ content was found to be associated with resistance/susceptibility to CLCuV infection.  相似文献

The viral protein 22 (VP22) in the tegument of Marek’s disease virus serotype 1 (MDV-1) plays an im-portant role in cell-to-cell spread and viral propagation. Antiserum against the carboxyl terminus of VP22 was prepared by immunizing mice with recombinant VP22 expressed in E. coli, and used to in-vestigate its expression in chicken embryo fibroblast (CEF) cells infected with different MDV-1 strains. At an infection dose of PFU=50, intercellular trafficking of the VP22 into the nuclei of the surrounding receipt cells was detected as early as 3 hours post infection. By 6 hours after infection (before viral plague formation), the protein was detected in the whole nuclei of the recipient cells with no difference among MDV-1 strains CVI988/Rispens, GA and RB1B. Intra-nuclear accumulation of the VP22 protein was further increased when the viral plagues started to form. These results indicate that, albeit the ex-istence of the 201TKSERT206 deletion, the VP22 of the CVI988/Rispens vaccine strain has also intercel-lular-trafficking function, which might serve as a potential alternative delivering protein instead of virulent strains VP22.  相似文献

The pre-calling behavior of female adults of the common cutworm, Spodoptera litura (Lepidoptera: Noctuidae) collected in southern Japan and southeastern Asian countries was investigated. Most females started calling on days 1-4 after adult emergence. In three populations obtained in Saga prefecture in Kyushu mainland, Japan, two different patterns of pre-calling period (PCP) were observed: one was the above-mentioned pattern shown by most individuals, and the other was a pattern in which females sexually matured within several hours after emergence. Diel periodicity was shown in the time of calling activity, and its pattern varied among the populations. Pre-calling period was stable over successive generations in the laboratory.  相似文献

Here we present the optimization of PCR conditions for microsatellite analysis of coniferous trees. The use of touchdown protocol for annealing resulted in a high success rate for optimization using fewer temperature profiles. The use of SYBR Green gel stain to detect PCR products in agarose gels was more sensitive than ethidium bromide. This is valuable for determining the success of PCR reactions and estimating the amount of PCR products formed—which is crucial in determining the dilution required to produce bands of similar intensity upon silver staining of the polyacrylamide gels. The use of SYBR Gold for staining polyacrylamide gels was not satisfactory in terms of the image quality produced. However, it was comparable to silver staining in terms of sensitivity, and could possibly be used in cases where the products are present as sharp single bands. In those cases, the use of SYBR Gold gel stain would save time and money for staining polyacrylamide gels.  相似文献

The effects of development states on the artemisinin content of clone S1 of Artemisia anuua L. grown in a greenhouse were investigated in the present study. The artemisinin content increased gradually during the phase of vegetative growth and reached its highest level at 8-9 mg/g dry weight (DW) when the S1 was 6 months old on a long day (LD) photoperiod. Treatment with 9-18 d of short day (SD) photoperiod resulted in the artemisinin content reaching and being maintained at a higher level (2.059-2.289 mg/g DW), twofold that of control plants and plants of S1 presented at the pro-flower budding and flower-budding stages. The artemisinin content varied in different parts of the plant. The artemisinin content of leaves was higher than that of florets and branches. The artemisinin content in middle leaves was higher than that of bottom leaves, and then top leaves. Different densities of capitate glands (the storage organ of artemisinin) located on the surface of leaves, florets, and branches explained the variations in artemisinin content in these parts of the plant. The correlation coefficient between artemisinin content and density of capitate glands on the surface of different organs was 0.987. The genetic marker for artemisinin content was screened using random amplified polymorphic DNA (RAPD) and sequence characterized amplified region (SCAR) techniques. The random primer OPAl5 (5'-TTCCGAACCC-3') could amplify a specific band of approximately 1 000 bp that was present in all high-artemisinin yielding strains, but absent in all low-yielding strains in three independent replications. This specific band was cloned and its sequence was analyzed. This RAPD marker was converted into a SCAR marker to obtain a more stable marker.  相似文献

To explore the necessity of electronic crossmatching applied to irregular antibodies from blood donors, to ensure blood transfusion safety. Irregular antibody screening was performed on blood samples collected in the Dongguan Blood Center from Apr 17, 2014 to Dec 31, 2017. Primary screening was performed on the Sanquin automatic blood group analyzer by the microcolumn gel method (Sanquin). The positive samples were analyzed again using the salt medium method, polybrene method and micro-column gel method (Diana) for the second screening. If the second screening was positive, it was used to determine irregular antibody specificity (using panel cells) and any irregular antibody titer was detected. A total of 208,004 samples were detected, of which 316 were positive (316/208,004; 0.152%). Among them, 120 alloantibodies (120/135,139; 0.088%) were detected in male donors, which was much lower than in female donors (173/72,865; 0.237%) (P<0.01). A total of 16 kinds of known irregular antibodies and 40 cases of unknown antibody specificity were detected, with 119 cases of IgG type and 197 cases of IgM type, at the ratio of 1.65:1. In female donors, the frequencies of anti-D, anti-E, anti-M and anti-Lea were significantly higher than in male donors (P<0.01). In married couples, the frequencies of anti-D and anti-E were significantly higher than those unmarried (P<0.05). In minority nationalities, the frequency of anti-M was significantly higher than in the Han nationality (P<0.05). In non-Guangdong donors, the frequencies of anti-D and anti-Lea were significantly higher than in Guangdong donors. 87.02% of irregular antibody positive donors’ antibody titers were lower than “++”, which was deemed as no serious hazard for clinical transfusion. The study suggests that it is unnecessary to screen for irregular antibodies in blood donors. Male donors from Guangdong may not be required to undergo screening for irregular antibodies, and anti-D and anti-E only identification is also not required to be detected in female donors.  相似文献

Summary Migration and tissue distribution of immunocompetent cells may be critical prerequisites for efficient immune surveillance. The effect of various concentrations of the mistletoe extract Iscador? QuFrF on the locomotory behavior and viability of immunomagnetically isolated human CD4+ and CD8+ T lymphocytes within three-dimensional collagen gels was investigated. Although variation in baseline activities of spontaneously migrating T cells was donor-dependent, a dose-dependent stimulation of the locomotory activity in both CD4+ and CD8+ T cells for noncytotoxic concentrations of Iscador QuFrF (0.25–1.25 μg/ml) was detected. The optimal concentration of mistletoe extract and time of maximal response were specific for each donor. As shown by cell tracking and subsequent data analysis, CD4+ T cells exposed to the mistletoe extract displayed a significant increase in mean velocity and time locomoting; total distance migrated was nearly doubled. In contrast, CD8+ T cells showed less pronounced changes in these critical parameters. Cytotoxic effects of the mistletoe preparation on T lymphocytes, which could at least partially be attributed to the induction of apoptosis, were drastically reduced in the presence of fetal calf serum in the culture system. Our data suggest that the direct stimulation of T-cell migration in the presence of mistletoe components may modulate in a dose-dependent manner the system of immune surveillance and recognition in patients under mistletoe therapy.  相似文献

 It was found that the atmospheric carbon dioxide (CO2) concentration in an urban canyon in Fukuoka city, Japan during August 1997 was about 30 μmol mol−1 higher than that in the suburbs. When fully exposed to sunlight, in situ the rate of photosynthesis in single leaves of Ilex rotunda planted in the urban canyon was higher when the atmospheric CO2 concentration was elevated. A biochemically based model was able to predict the in situ rate of photosynthesis well. The model also predicted an increase in the daily CO2 exchange rate for leaves in the urban canyon with an increase in atmospheric CO2 concentration. However, in situ such an increase in the daily CO2 exchange rate may be offset by diminished sunlight, a higher air temperature and a lower relative humidity. Thus, the daily CO2 exchange rate predicted using the model based soleley on the environmental conditions prevailing in the urban canyon was lower than that predicted based only on environmental factors found in the suburbs.
