The behavior and fate of mercury (Hg) in soil is mainly controlled by adsorption and desorption processes with various adsorbents, particularly dissolved organic matter (DOM). This study was conducted to assess the effect of DOM from wheat straw (DOMw) and swine manure (DOMs) on Hg (II) adsorption of black, red, and fluvo-aquic soils in China. Results showed that the Hg (II) adsorption isotherms fitted well with Langmuir and Freundlich equations. The maximum Hg (II) potential adsorption capacity by the three soils followed this trend: black soil > red soil > fluvo-aquic soil. The amount of Hg (II) adsorbed on the soils significantly decreased when DOMw and DOMs were added to the soil samples. Furthermore, the extent by which DOMw affected the Hg (II) adsorption of the three soils was higher than the effect of equivalent amounts of DOMs. Therefore, DOM is important to determine the fate of Hg (II) and control Hg (II) pollution in the environment.  相似文献

The solution conformations of pyridoxal-5′ -phosphate and pyridoxamine-5′-phosphate have been investigated using Eu(III) as a nuclear magnetic resonance shift probe. Binding of Eu(III) to pyridoxal phosphate results in the formation of two complexes, at the phosphate group and theo-hydroxy-aldehyde moiety, which are in slow exchange on the nuclear magnetic resonance time-scale. The lanthanide-induced pseudo contact shifts calculated using the McConnell-Robertson equation (J. Chem. Soc. (1950), 22, 1561) are in good agreement with the experimentally observed values for both pyridoxal phosphate and pyridoxamine phosphate and lead to a family of closely related conformations. Heat-aggregated IgG, used as a surrogate for immunoglobulin configuration in immune complexes, mobilized a hydrophobic spin probe within bilayers of anionic liposomes, whereas native IgG was ineffective. Hydrocortisone, preincorporated into liposomal bilayers, both prevented the perturbation of nonpolar membrane regions and reduced the accelerated rate of solute diffusion from liposomes provoked by aggregated immunoglobulin. The capacity of aggregated, but not native, IgG to initiate membrane responses of living cells may therefore be due to its ability to alter surface bilayers or similar configurations of plasma membrane Fc receptors, events antagonized by corticosteroids.  相似文献

We recently reported that mitochondria-targeted derivatives of resveratrol are cytotoxic in vitro, selectively inducing mostly necrotic death of fast-growing and tumoral cells when supplied in the low μM range (N. Sassi et al., Curr. Pharm. Des. 2014). Cytotoxicity is due to H2O2 produced upon accumulation of the compounds into mitochondria. We investigate here the mechanisms underlying ROS generation and mitochondrial depolarization caused by these agents. We find that they interact with the respiratory chain, especially complexes I and III, causing superoxide production. “Capping” free hydroxyls with acetyl or methyl groups increases their effectiveness as respiratory chain inhibitors, promoters of ROS generation and cytotoxic agents. Exposure to the compounds also induces an increase in the occurrence of short transient [Ca2 +] “spikes” in the cells. This increase is unrelated to ROS production, and it is not the cause of cell death. These molecules furthermore inhibit the F0F1 ATPase. When added to oligomycin-treated cells, the acetylated/methylated ones cause a recovery of the cellular oxygen consumption rates depressed by oligomycin. Since a protonophoric futile cycle which might account for the uncoupling effect is impossible, we speculate that the compounds may cause the transformation of the ATP synthase and/or respiratory chain complex(es) into a conduit for uncoupled proton translocation. Only in the presence of excess oligomycin the most effective derivatives appear to induce the mitochondrial permeability transition (MPT) within the cells. This may be considered to provide circumstantial support for the idea that the ATP synthase is the molecular substrate for the MPT pore.  相似文献
