In this paper, we investigate the role of arbuscular mycorrhizal fungi in plant phosphorus nutrition. We develop a mathematical model which quantitatively assesses the contribution of external fungal hyphae to plant phosphate uptake.We derive an equation for solute uptake by a growing fungal mycelium which we couple with a model for root uptake. We analyse the model using nondimensionalization and numerical simulations.Simulations predict that removal of phosphate from soil is dominated by hyphal uptake as opposed to root uptake. Model analysis shows that the depletion zones around hyphae overlap within 8 h and that the transfer between fungus and root is a critical step for the behaviour of phosphorus within the mycelial phase. We also show that the volume fraction of mycelium is negligibly small in comparison to other soil phases.This is the first model to quantify the contribution of mycorrhizal fungi to plant phosphate uptake. A full data set for model parametrization and validation is not currently available. Therefore, more complete sets of experimental measurements are necessary to make this model more applicable.  相似文献

An experiment was set up to investigate the role of arbuscular mycorrhiza (AM) in utilization of P from organic matter during mineralization in soil. Cucumber (Cucumis sativus L.) inoculated with one of two AM fungi or left uninoculated were grown for 30 days in cross-shaped PVC pots. One of two horizontal compartments contained 100 g soil (quartz sand: clay loam, 1:1) with 0.5 g ground clover leaves labelled with32P. The labelled soil received microbial inoculum without AM fungi to ensure mineralization of the added organic matter. The labelling compartment was separated from a central root compartment by either 37 m or 700 m nylon mesh giving only hyphae or both roots and hyphae, respectively, access to the labelled soil. The recovery of32P from the hyphal compartment was 5.5 and 8.6% for plants colonized withGlomus sp. andG. caledonium, respectively, but only 0.6 % for the non-mycorrhizal controls. Interfungal differences were not related to root colonization or hyphal length densities, which were lowest forG. caledonium. Both fungi depleted the labelled soil of NaHCO3-extractable P and32P compared to controls. A 15–25% recovery of32P by roots was not enhanced in the presence of mycorrhizas, probably due to high root densities in the labelled soil. The experiment confirms that AM fungi differ in P uptake characteristics, and that mycorrhizal hyphae can intercept some P immobilization by other microorganisms and P-sorbing clay minerals.  相似文献

 The influence of the systemic fungicides propiconazole (Tilt 250E) and carbendazim (Bavistin) at field application rates on the functioning of three arbuscular mycorrhizal fungi was studied. Short-term fungal 32P transport and succinate dehydrogenase (SDH) activity in external hyphae of Glomus intraradices Schenck and Smith, G. claroideum Schenck and Smith and G. invermaium Hall in symbiosis with pea (Pisum sativum L.) were measured. In the experimental system used, the hyphae grew into two root-free hyphal compartments (HCs). The fungicides were applied to each HC 24 days after sowing and 32P was added to one HC of each pot. Four days later, the fungicide effect on fungal P transport was measured as the difference in 32P content of treated and untreated plants. SDH activity in fungal hyphae was determined in the HCs given no 32P. Carbendazim severely inhibited 32P transport and SDH activity in external hyphae at an application rate of 0.5 μg g–1 soil. The ergosterol inhibitor propiconazole affected none of these parameters. The fungicides had similar effects on all three fungal species, although P transport efficiency and SDH activity differed markedly between the fungi. Accepted: 12 December 1996  相似文献

The carbohydrate levels in the roots of two soybean cultivars (Century, Woodworth) were determined during the early vegetative growth stage. Based on Michaelis-Menten kinetics Century was characterized as being more efficient at phosphate uptake when compared to Woodworth. The experiments were carried out at normal nutrition (+P treatment) as well as under a condition of moderate phosphate stress (–P treatment). The stress situation was created by withdrawal of phosphate from the nutrient solution for five or nine days. Compared to the +P-treatment the deprivation of phosphate led to a reduction of more than 50% in the phosphate content of the plant tissue.The carbohydrates in the roots exhibited differences between both cultivars with Century generally having higher sucrose levels. During the morning the +P treatment of Century exhibited 10 mg g-1 and in the afternoon 16 mg g-1 sucrose (plant dry weight basis). This compares to 8 mg g-1 sucrose (morning) and 13 mg g-1 sucrose (afternoon) for Woodworth. Phosphate stress increased the levels of hexoses (glucose, fructose) in both varieties significantly. The sucrose levels remained higher for the Century variety even during phosphate stress. We conclude that the shoot of the Century variety supplies its root with an increased amount of energy in form of carbohydrates compared to the Woodworth variety. This may enable the plant to take up phosphate more efficiently. It is also remarkable that the Century variety reduces the pH of the nutrient medium more rapidly. This active proton deposition may create a high membrane potential which is known to be necessary for phosphate uptake.  相似文献

 Two glasshouse experiments were done to assess the development and metabolic activity of mycorrhizas formed by isolates of arbuscular mycorrhizal fungi (AMF) from three different genera, Acaulospora, Gigaspora and Glomus on either Pueraria phaseoloides L. or Desmodium ovalifolium L. plants. The second of the two experiments included three levels of a localised phosphate source in the pots. Alkaline phosphatase (ALP), stained histochemically in the intra-radical mycelium (IRM) of AMF over sequential harvests, did not provide a direct marker for the efficiency of AMF in mobilising phosphorus (P) for plant growth and development. The ability of the extra-radical mycelium (ERM) to scavenge a localised phosphate source, determined by its extraction from buried 35-μm mesh pouches, was dependent on the species of AMF tested. This work indicates that AMF from different genera have unique patterns of mycelial development when forming mycorrhizas with tropical hosts in the presence of a localised phosphate source. AMF also appear to have different mechanisms for the control of P transfer, within the mycelium, to the host. The significance of the architecture of the ERM is discussed as well as the localisation of ALP in the IRM in determining the efficiency of AMF in terms of P accumulation in planta and subsequent growth of plants. The community composition of arbuscular mycorrhizal fungi (AMF) was investigated in roots of four different plant species (Inula salicina, Medicago sativa, Origanum vulgare, and Bromus erectus) sampled in (1) a plant species-rich calcareous grassland, (2) a bait plant bioassay conducted directly in that grassland, and (3) a greenhouse trap experiment using soil and a transplanted whole plant from that grassland as inoculum. Roots were analyzed by AMF-specific nested polymerase chain reaction, restriction fragment length polymorphism screening, and sequence analyses of rDNA small subunit and internal transcribed spacer regions. The AMF sequences were analyzed phylogenetically and used to define monophyletic phylotypes. Overall, 16 phylotypes from several lineages of AMF were detected. The community composition was strongly influenced by the experimental approach, with additional influence of cultivation duration, substrate, and host plant species in some experiments. Some fungal phylotypes, e.g., GLOM-A3 (Glomus mosseae) and several members of Glomus group B, appeared predominantly in the greenhouse experiment or in bait plants. Thus, these phylotypes can be considered r strategists, rapidly colonizing uncolonized ruderal habitats in early successional stages of the fungal community. In the greenhouse experiment, for instance, G. mosseae was abundant after 3 months, but could not be detected anymore after 10 months. In contrast, other phylotypes as GLOM-A17 (G. badium) and GLOM-A16 were detected almost exclusively in roots sampled from plants naturally growing in the grassland or from bait plants exposed in the field, indicating that they preferentially occur in late successional stages of fungal communities and thus represent the K strategy. The only phylotype found with high frequency in all three experimental approaches was GLOM A-1 (G. intraradices), which is known to be a generalist. These results indicate that, in greenhouse trap experiments, it is difficult to establish a root-colonizing AMF community reflecting the diversity of these fungi in the field roots because fungal succession in such artificial systems may bias the results. However, the field bait plant approach might be a convenient way to study the influence of different environmental factors on AMF community composition directly under the field conditions. For a better understanding of the dynamics of AMF communities, it will be necessary to classify AMF phylotypes and species according to their life history strategies.  相似文献

Different calculation methods, based on needle geometry, for estimating both projected area (PLA) and total surface area (TLA) of foliage in Norway spruce [Picea abies (L.) Karst.] were compared. Seventy-eight shoots of four age classes were sampled from both the basal and top thirds of crowns. Three dimensions (the length, minor and major diameters) of needles were taken, and the needle shape was approximated to a parallelepiped or ellipsoid. There was a perfect coincidence of the measured and estimated values of PLA calculated as the width of the needle projection multiplied by needle length, and corrected for needle taper (method III), or when the needle projection was treated as a rectangle joined with half-ellipses at both ends (method IV). The most reliable estimations of TLA resulted from treating the needle sides as faces of the parallelepiped tapering at their ends in the form of half-ellipses. The ratio of TLA to PLA ranged from 2.2 to 4.0 depending on the needle morphology. Needle minor diameter (anatomical width; D 1) was found to be a better morphological index of the spruce foliage than needle flatness, i.e. the ratio of major to minor diameter. Expressing the factor for converting PLA to TLA as a function of D 1 considerably improved the precision of the estimates. Close relationships were established between specific leaf area, expressed on both a projected area (SLAP) and total surface area basis (SLAT), needle dry weight (R 2 was 0.799 and 0.852, respectively) and minor diameter of needles (R 2 was 0.701 and 0.554, respectively). The aim of this randomised double-blind multi-centre parallel group comparative study was to investigate the efficacy and tolerability of a new standardised fresh-plant extract obtained from the shoot tips of St. John's wort (Hypericum perforatum L.) in the treatment of mild to moderate depression. 348 out-patients (259 female, 89 male) with mild to moderate depression were recruted by 12 psychiatrc specialty practices and 26 general practices. The patients took during 6 weeks 3 times a day 1 tablet of a Hypericum preparation standardised to either 0.17 mg (114 patients), 0.33 mg (115 patients), or 1 mg (119 patients) total hypericin per day (Hyperiforce). The main outcome measure was the Hamilton Psychiatric Rating Scale for Depression; additional measures were the Hospital Anxiety and Depression Scale and the Clinical Global Impression. At the end of treatment, a reduction in the average Hamilton Depression score from an initial 16-17 to 8-9, i.e. a relative reduction of about 50%, was observed in all groups (280 patients, par protocol analysis). The response rates were 62%, 65% and 68%, respectively (348 patients, intention to treat analysis). Overall, the intergroup comparison revealed no significant differences. Tolerability was excellent, with mild adverse reactions probably causally related to the treatment occurring in only 7 of the 348 patients (2%). This Hypericum preparation is effective in all three doses and is well tolerated.  相似文献

Whether a species is native or introduced in a given geographic area is of major interest within the framewok of biological conservation. A practical approach combining phytosociological, ecological, phytogeographical and historical data is proposed to explore this status for rare plant species, and applied to Boxtree in northern France. Buxus sempervirens L. is an evergreen sub-mediterranean species whose wild populations in northern France are very rare and threatened. Its status – native or introduced – has long been be controversial. Three types of Box-woodland were found in the study area: 1) a Taxus baccata – Buxus sempervirens community which is strongly linked to post-Renaissance castles, 2) a Fraxinus excelsiorMercurialis perennis community which may be related both to steep chalk slopes where Box was expected to be native and to Medieval castles, 3) a Quercus pubescensBuxus sempervirens community which may be considered as an immature anthropogenic woodland. In all cases Buxus sempervirens was observed close to archaeological sites and together with exotic and/or nitrophilous plant species. Consequently Box is probably originally an introduced species in northern France and should be considered as both an archaeophyte and a feudal plant. This method offers an interesting alternative to determine the indigenity status of a rare plant species in its localities that would provide sufficiently accurate criteria in most of the cases.  相似文献

It has been hypothesized that species occurring in the eastern and the western Qinghai–Tibet Plateau (QTP) responded differently to climate changes during the Pleistocene. Here, we test this hypothesis by phylogeographic analysis of two sister species, Allium cyathophorum and A. spicata. We sequenced two chloroplast DNA (cpDNA) fragments (accD‐psaI and the rpl16 intron) of 150 individuals, and the nuclear (ITS) region of 114 individuals, from 19 populations throughout the distributional ranges of these species. The divergence between the two species was dated at 779 ‐ 714 thousand years before the present and was likely initiated by the most major glaciation in the QTP. Analysis of chlorotype diversity showed that A. spicata, the species occurring in the western QTP, contains much lower genetic diversity (0.25) than A. cyathophorum (0.93), which is distributed in the eastern QTP. Moreover, multiple independent tests suggested that the A. spicata population had expanded recently, while no such expansion was detected in A. cyathophorum, indicating a contrasting pattern of responses to Pleistocene climate changes. These findings highlight the importance of geographical topography in determining how species responded to the climate changes that took place in the QTP during the Pleistocene.  相似文献
