Morphological, reproductive and karyological variability were investigated in fifteen populations of Allium oleraceum transferred from natural populations in Lithuania to the field collection of medicinal and aromatic plants of the Institute of Botany of the Nature Research Centre in Vilnius. Nine populations were tetraploid (2n = 4x = 32), four pentaploid (2n = 5x = 40) and two consisted of both cytotypes. The greatest differences among the populations and between the ploidy levels were observed in stem height and mass of aerial bulbils per plant. The seed production was very low and did not differ significantly between the ploidy levels, while neither tetraploids nor pentaploids did produce any seed if insects were prevented from visiting their flowers. The means of the morphological characters were higher in pentaploids than in tetraploids. However, none of them provided the possibility of determining ploidy level without chromosome counting.  相似文献

Analysis of 265 plants derived from 13 sites in the Rokko Mountains shows that the widely distributed eastern AsianEupatorium chinense var.simplicifolium consists of seven cytotypes. These polymorphic karyotypes comprise four levels of ploidy based on X=10 and partial deficiency, occurring either alone or in combination. The polyploid cytotypes exhibit anomalous meiosis and highly variable pollen stainability in contrast to the nearly-normal behaviour of the diploid. The polyploid cytotypes were revealed as being agamospermous. They are readily distinguishable from the diploid cytotypes by several morphological characters and by growth habits closely related to their respective niches. Two or more cytotypes occurred in all sites examined, the most common being the coexistence of triploid and tetraploid cytotypes. The frequencies of occurrence of the pentaploid plants and the cytotypes involving a deficient chromosome were relatively low. While the diploid cytotype is restricted to fragile gravitational slopes and rocky areas which are poor in species and lack tall competitors, the polyploids occur widely in the grasslands, the roadsides or the forest-margins, closely associated with tall grasses and forbs such asMiscanthus sinensis. Based on these data, theE. chinense var.simplicifolium complex is divided into two distinct groups: the diploid cytotype and the polyploid cytotypes.  相似文献

The genus Cynodon comprises ten species. The objective of this study was to evaluate the genetic diversity of Korean bermudagrasses at the morphological, cytological and molecular levels. Morphological parameters, the nuclear DNA content and ploidy levels were observed in 43 bermudagrass ecotypes. AFLP markers were evaluated to define the genetic diversity, and chromosome counts were made to confirm the inferred cytotypes. Nuclear DNA contents were in the ranges 1.42-1.56, 1.94-2.19, 2.54, and 2.77-2.85 pg/2C for the triploid, tetraploid, pentaploid, and hexaploid accessions, respectively. The inferred cytotypes were triploid (2n = 3x = 27), tetraploid (2n = 4x = 36), pentaploid (2n = 5x = 45), and hexaploid (2n = 6x = 54), but the majority of the collections were tetraploid (81%). Mitotic chromosome counts verified the corresponding ploidy levels. The fast growing fine-textured ecotypes had lower ploidy levels, while the pentaploids and hexaploids were coarse types. The genetic similarity ranged from 0.42 to 0.94 with an average of 0.64. UPGMA cluster analysis and principle coordinate analysis separated the ecotypes into 6 distinct groups. The genetic similarity suggests natural hybridization between the different cytotypes, which could be useful resources for future breeding and genetic studies.  相似文献

Belonging to the genus Cenchrus with 16–22 species, Cenchrus ciliaris L. (syn. Pennisetum ciliare (L.) Link, buffelgrass) is a perennial, common in warmer regions of both hemispheres, growing as a C4 grass in a wide range of habitats. In the present study we determined chromosome number and nuclear DNA content (2C DNA) for 28 natural populations collected from northern to southern Tunisia. Three ploidy levels were found: one tetraploid population (2n?=?4x?=?36), three pentaploid (2n?=?5x?=?45), and 24 hexaploid populations (2n?=?6x?=?54). The hexaploid chromosome number has already been reported for Tunisian populations of C. ciliaris but tetraploid and pentaploid (2n?=?45) are new for this area. The tetraploid population was found in the semi-arid north; pentaploids were mostly on the northern side of the arid region, while the hexaploids were located mainly in the arid southern Tunisian and Saharan region. 2C DNA values, assessed using flow cytometry, correlated with chromosome counts. Nuclear DNA content ranged from 2C?=?3.03 to 4.61 pg, revealing three ploidy levels corresponding to 4x, 5x, 6x, and mean 2C DNA amounts were of 3.03, 3.7 and 4.48 pg, respectively. Each cytotype produced viable pollen. Flow cytometric seed screening neither proved nor disproved apomixis. The most frequent hexaploid populations seem best adapted to arid conditions in southern Tunisia. The monoploid value, 1Cx, was constant. The existence of pentaploid cytotype suggests hybridization ability between tetraploids and hexaploids. It appears that polyploidization is the major evolutionary mechanism in the speciation of C. ciliaris.  相似文献

It has been proposed that plant species cytotypes commonly exhibit altered morphology, reproduction, geographic and ecological distributions. We studied phenotypic variation in height, sexual (flowers, seeds) and asexual (aerial bulbils) reproduction in natural populations and in the conditions of a common garden of three cytotypes (2n = 4x, 5x, 6x) of the bulbous geophyte Allium oleraceum in the Czech Republic. Additionally, we compared the germination and dormancy of seeds and bulbils to determine whether propagules have different ecological roles. The pattern of morphological differentiation observed between cytotypes under natural conditions was similar to that observed under common garden conditions, suggesting that variability in morphological characteristics appears to be associated with ploidy levels. We revealed differences in size and sexual and asexual production between A. oleraceum cytotypes, but with wide overlap among cytotypes, suggesting a limited possibility of the studied traits to reliably distinguish between cytotypes. Tetraploids and pentaploids were rather similar; they were taller and produced more flowers than hexaploids, which were mostly flowerless (mean <0.7 flower/plant). All cytotypes were able to produce viable seeds, but their numbers were extremely low, usually less than 3 seeds per inflorescence; clonal reproduction via aerial bulbils dominates in all cytotypes (flower:bulbil ratio <0.5), with tetraploids producing more but lighter bulbils than other cytotypes. The seed: ovule ratio was low (<0.1) in all cytotypes, although hexaploids reached higher values than other cytotypes. Bulbils germinated better (means >80%) than seeds in all cytotypes, with pentaploid bulbils showing the highest germination (mean 90.5%). The cytotypes did not differ in seed germination (range of means 73.4%–76.3%). About 6% of seeds did not germinate but were still viable at the end of the experiment, while all non-germinated bulbils of all cytotypes had rotten away. Seeds, but not bulbils, can likely form a short-term persistent propagule soil bank. We found no evidence of a phenotypic trade-off between the production of flowers (seeds) and the production of bulbils within the inflorescence of all studied cytotypes. Together, inter-cytotypic differences in fitness-related traits cannot completely explain the different geographic and ecological distributions of cytotypes in the Czech Republic found by previous research.  相似文献

为探讨国产毛茛科(Ranunculaceae)驴蹄草属(Caltha L.)植物的细胞学特征,对驴蹄草(C.palustris L.)3个居群和花葶驴蹄草(C.scaposa Hook.f.&Thoms.)5个居群进行了细胞学研究。驴蹄草贵州纳雍居群的染色体数目为2n=32(四倍体),两个云南中甸居群的染色体数目均为2n=64(八倍体)。花葶驴蹄草四川红原、康定、石渠居群的染色体数目均为2n=32(四倍体),该数目为首次报道;西藏林芝和云南德钦居群的染色体数目均为2n=64(八倍体)。驴蹄草的染色体比花葶驴蹄草大。这两种植物的32或64条染色体分别以4条或8条为单位大致能够排列为8组同源染色体,但同一组内的染色体经常具有明显的异形性(heteromorphy),不同居群的核型组成多少具有差异。同时,还分析了驴蹄草和花葶驴蹄草的不同倍性细胞型在我国的地理分布式样。  相似文献

Areas of immediate contact of different cytotypes offer a unique opportunity to study evolutionary dynamics within heteroploid species and to assess isolation mechanisms governing coexistence of cytotypes of different ploidy. The degree of reproductive isolation of cytotypes, that is, the frequency of heteroploid crosses and subsequent formation of viable and (partly) fertile hybrids, plays a crucial role for the long‐term integrity of lineages in contact zones. Here, we assessed fine‐scale distribution, spatial clustering, and ecological niches as well as patterns of gene flow in parental and hybrid cytotypes in zones of immediate contact of di‐, tetra‐, and hexaploid Senecio carniolicus (Asteraceae) in the Eastern Alps. Cytotypes were spatially separated also at the investigated microscale; the strongest spatial separation was observed for the fully interfertile tetra‐ and hexaploids. The three main cytotypes showed highly significant niche differences, which were, however, weaker than across their entire distribution ranges in the Eastern Alps. Individuals with intermediate ploidy levels were found neither in the diploid/tetraploid nor in the diploid/hexaploid contact zones indicating strong reproductive barriers. In contrast, pentaploid individuals were frequent in the tetraploid/hexaploid contact zone, albeit limited to a narrow strip in the immediate contact zone of their parental cytotypes. AFLP fingerprinting data revealed introgressive gene flow mediated by pentaploid hybrids from tetra‐ to hexaploid individuals, but not vice versa. The ecological niche of pentaploids differed significantly from that of tetraploids but not from hexaploids.  相似文献

The taxomically critical species Centaurea stoebe is represented in Central Europe by a diploid (2n?=?18) and a tetraploid (2n?=?36) cytotype. Their morphological differentiation and taxonomic treatment is still controversial. Karyological (chromosome numbers and flow cytometric measurements) and multivariate morphometric analyses were used here to address cytotype distribution patterns and their morphological differentiation. Material from 38 localities (771 individuals) in Slovakia, Hungary and Austria, including type localities of the names traditionally applied to the different cytotypes, was sampled and evaluated using both morphometric and karyological approaches. The morphological tendency towards cytotype differentiation is evident only at a population level, and is blurred at the level of individual plants. Diploid populations prevail in the area studied, as well as throughout Europe; mixed-cytotype populations were also found. The present data, namely the weak morphological distinction, largely sympatric occurrence of the cytotypes, and the existence of mixed-cytotype populations, favour taxonomic treatment as a single species, without recognition of infraspecific units.  相似文献

To elucidate the mechanisms of the adaptive advantages of polyploidy, there is a need to identify physiological traits that participate in the success of polyploids. We studied selected photosynthetic characteristics, stomatal density, and specific leaf area of three ploidy levels (2n = 4x, 5x, 6x) of the geophyte Allium oleraceum that partially differ in their ecological niches. Although the cytotypes were on average similar with regard to most of the measured photosynthetic traits, the hexaploids showed more rapid initial photosynthetic induction and a tendency for a higher maximum photosynthetic rate per unit area. The stomatal density was not affected by ploidy, though the specific leaf area was reduced for the hexaploids compared to the other cytotypes. A lower intracytotype variation was found for most of the studied photosynthetic and anatomical traits for the hexaploids compared to the large variation found within other cytotypes. A comparison of the photosynthetic traits between the cytotypes showed that the ecological differentiation between cytotypes is only weakly related to the characteristics of their photosynthetic apparatus. However, contrasting ranges of variability in the measured traits between the cytotypes can be related to previously observed differences between cytotypes with regard to the ranges of intracytotype genetic variation, genome size variation, and niche breadth. A higher variability of photosynthetic traits in tetraploids and pentaploids may be related to the existence of a spectrum of types adapted to different environmental conditions. Hexaploids may represent a recently formed cytotype adapted to open environmental conditions.  相似文献

Many plant species contain populations with more than one polyploid cytotype, but little is known of the mechanisms maintaining several cytotypes in a population. Andropogon gerardii cytotypes were compared to evaluate different models of autopolyploid cytotype coexistence. The enneaploid (90 chromosome, 9x) cytotype was found to be larger and taller than the hexaploid (60 chromosome, 6x) cytotype. Seed production is significantly more efficient in hexaploids, but seed production per area was not significantly different. The two cytotypes are not exomorphologically separable in the field because of great plasticity in response to environmental variation and wide variation within each cytotype. These data suggest cytotypic variation is maintained by natural selection.  相似文献

Jacobaea vulgaris subsp. vulgaris (syn. Senecio jacobaea subsp. jacobaea) constitutes an intricate polyploid complex distributed in Europe. Four cytotypes have been reported in this species, three with euploid (diploid, tetraploid and octoploid; 2n=20, 40 and 80) and one with aneuploid (2n=32) chromosome numbers. Here we report that the diploid chromosome number (2n=20) reported from Bulgaria is due to misidentification with Jacobaea aquatica. On the other hand, we have discovered a new, hexaploid (2n=6x=60) cytotype within J. vulgaris subsp. vulgaris using flow cytometry. The new cytotype occurs within four sympatric populations of otherwise tetraploid and octoploid plants in Pannonia (one locality in the eastern Czech Republic and two localities in southwestern Slovakia) and in Podillya (one locality in western Ukraine). The frequency of hexaploid individuals within 76 studied populations is very low (only 10 of 693 analysed plants), and hexaploids probably represent hybrids between tetraploid and octoploid plants. Three mixed populations with hexaploid plants were subjected to detailed morphological and pollen fertility analyses. Multivariate morphometric analysis reveals partial separation of tetraploid and octoploid plants, whereas hexaploid individuals are similar in morphology to octoploids. In comparison with tetraploids, octoploids and hexaploids exhibit slightly longer ray florets, involucral bracts and tubular florets and more hairy outer achenes. Hexaploid plants display larger pollen grains and lower pollen fertility compared to tetraploids and octoploids.  相似文献

The use of local, native plant materials is now common in restoration but testing for polyploidy in seed sources is not. Diversity in cytotypes across a landscape can pose special seed transfer challenges, because the methods used to determine genetically appropriate materials for seed transfer do not account for cytotypic variation. This lack of consideration may result in mixing cytotypes through revegetation, which could reduce long‐term population viability. We surveyed nine populations of a native bunchgrass, Pseudoroegneria spicata, in three EPA Level III Ecoregions in the western United States to determine the frequency of polyploidy, whether there are differences in traits (phenotype, fecundity, and mortality) among plants of different cytotypes, and whether cytotype frequency varies among ecoregions. We assessed trait variation over 2 years in a common garden and determined ploidy using flow cytometry. Polyploidy and mixed cytotype populations were common, and polyploids occurred in all ecoregions. Four of the nine populations were diploid. The other five had tetraploids present: three had only tetraploid individuals whereas two had mixed diploid/tetraploid cytotypes. There was significant variation in traits among cytotypes: plants from tetraploid populations were larger than diploid or mixed populations. The frequency and distribution of cytotypes make it likely that seed transfer in the study area will inadvertently mix diploid and polyploid cytotypes in this species. The increasing availability of flow cytometry may allow ploidy to be incorporated into native plant materials sourcing and seed transfer.  相似文献

We studied hybridization between the diploid Centaurea pseudophrygia and the tetraploid C. jacea by performing crossing experiments and screening natural populations using flow cytometry. The experiments confirm that the studied species exhibit strong reproductive isolation. Interspecific hybrids were formed at a low frequency, including triploids (originating from reduced gametes) and tetraploids (involving unreduced gametes of the diploids). In contrast, hybrids were almost absent among seeds and adult plants of natural mixed populations and among the offspring from experimental pollinations with a mixture of pollen of both ploidy levels. We found that mixed pollination is an important mechanism for preventing hybridization between plants of different ploidy levels and sustaining the reproduction of the tetraploids. A mentor effect (induced selfing in the presence of pollen of different ploidy levels) was observed in both diploids and tetraploids, reinforcing the reproductive isolation between cytotypes. Higher ploidy levels (pentaploid, hexaploid) involving unreduced gametes of the tetraploid species were identified. Notably, pentaploids were discovered for the first time in Centaurea sect. Morphological, cytological and ecological observations were made onLycopodium clavatum. Among 121 individuals from 26 populations studied 31 were of the diploid cytotype with 2n=68, 45 were of the triploid cytotype with 2n=102 and 45 were of the tetraploid cytotype with 2n=136. The populations were divided into three types: (1) the sole type—characterized by a single cytotype occupying an entire population, (2) the mixed type A—characterized by two or three cytotypes occupying together the same population, but each cytotype choosing different habitats, (3) the mixed type B—characterized by two or three cytotypes occupying together the same population and sharing the same habitat with each other. The mixed type B populations were found in the places of the early phase of secondary succession in a plant community. In the sole type and the mixed type A, the habitat preference for each cytotype was described as follows: the habitat for the diploid was in humid and shady places, that for the tetraploid was in open and sunny, unstable places, and that for the triploid was intermediate between the other two. Each cytotype can progress towards occupying their optimum habitat successively from the mixed type B population to the mixed type A population and finally reaching the sole type population.  相似文献

Two cytotypes (2n = 36 and 38) of blind mole rats, Nannospalax xanthodon (Nordmann, 1840), from the Ayd?n and Manisa provinces in Turkey were investigated. Conventional chromosome staining, Ag-NOR staining and C-banding analysis were carried out. From the cytogenetic point of view, the particular phylogenetic position of these populations is supported by their low diploid numbers only, and the Cbanding pattern and the NORs distribution seem generally similar to populations with higher chromosome numbers. Several autosomal pairs with centromeric dark Cbands were observed in the 2n=36 cytotype. One autosomal pair possessed an interstitial dark C-band on the short arm; another pair possessed an interstitial dark Cband on the long arm. Whole C-heterochromatic short arms were observed in three subtelocentric autosomal pairs in the 2n=38 cytotype. Most of the other autosomal pairs possessed centromeric dark C-bands. Distinct dark C-bands were observed also in the presumed X chromosomes of both the cytotypes. The Ag-NOR regions were found on three autosomal pairs of both the cytotypes. These sites were located in telomeric areas of the short arms of two subtelocentric and one submetacentric pair.  相似文献

In many North American prairies, populations of Andropogon gerardii Vitman (Poaceae) are composed of hexaploid and enneaploid cytotypes (2 n  = 60, 90), with intermediates occurring occasionally. Under controlled pollination, the two common cytotypes can be crossed, producing progeny with a range of chromosome numbers. In an investigation of fertility and compatibilities of intermediate cytotypes, individuals with chromosome numbers between 60 and 90 were crossed with each other, with the 2 n  = 60 and 90 cytotypes, and with South American Andropogon species having 60 chromosomes. Regardless of cytotype, all A. gerardii plants had some fertility and virtually all crosses produced seeds. Cytotype is only partially predictive of fertility. Inter-specific hybrids between A. gerardii and South American hexaploid species were vigorous but sterile. Qualitative and quantitative characteristics of mega- and microspores from all the cytotypes of JapaneseIsoetes are described based on the voucher specimens whose chromosome numbers were known. InI. japonica, the hexaploid possessed reticulate megaspores and levigate microspores, while the octaploid and the heptaploid had echinate microspores. Mega- and microspores of the hexaploid and the octaploid were of normal appearance, while those of the heptaploid displayed polymorphism. The tetraploid and the hexaploid ofI. sinensis resembled each other, since they both possessed cristate megaspores and echinate microspores. Echinate megaspores and levigate microspores characterized the diploidI. asiatica. The spore size was largely variable within each cytotype, while the size of the megaspores varied more than that of the microspores. The microspore length was closely correlated with polyploid level. InI. sinensis, the mean microspore length of the tetraploid was 27.6 μm while that of the hexaploid was 31.9 μm, hence these two cytotypes were easily distinguishable. In the hexaploidI. japonica, variations in mega-and microspore size displayed geoclinal variation showing a positive correlation (r=0.43–0.55) with the longitude and the latitude of the populations. Despite the presence of intraspecific polyploidy (2x, 4x, 5x and 6x) in Agrimonia eupatoria, origin of these cytotypes has never been addressed adequately. The aim of the present study was to record the original chromosome counts and characterize chromosomal pairing during meiosis and microsporogenesis in the 5x cytotype, and discussing the hypothesis regarding the possible origin of polyploid cytotypes (4x, 5x and 6x) in the species. The geographical distribution pattern of cytotypes in the Indian Himalayas and elsewhere has also been analyzed. The present meiotic analysis revealed three chromosomes counts, the tetraploid (2n?=?4x?=?56), the pentaploid (2n?=?5x?=?70) and the hexaploid (2n?=?6x?=?84) cytotypes based on x?=?14. Meiotic course was perfectly normal in the 4x and 6x cytotypes resulting into high pollen fertility (94–100 %). Meiotic course in the imbalanced 5x cytotype has been found to be irregular characterized by the presence of high frequency of univalents at diakinesis and metaphase-I. Abnormal meiotic course contributed towards high pollen sterility (74–88 %). Even the apparently fertile/stained pollen grains were of irregular shape and of heterogeneous sizes. Meiotic behaviour of the 5x cytotype is like typical of allopolyploid. Individuals of 5x cytotype did not produce seeds and propagate vegetatively (root suckers) while 4x and 6x cytotypes exploited sexual (seeds) as well as vegetative means for propagation. Chromosomal pairing in pentaploid cytotype is like typical of an allopolyploid and we assume that it might have originated owing to natural inter-cytotype hybridization between 4x and 6x cytotypes in a mixed population. Analysis of geographical distribution pattern of cytotypes shows that Indian Himalayas represent the most cytotype-diverse region for A. eupatoria with the existence of all the four cytotypes (2x, 4x, 5x, 6x). This shows the dynamic nature of the species at chromosomal level in this part of the world.  相似文献

Phenotypic differentiation of two tetraploid (2n = 4x = 36, 36+1B, 36+2B) populations of Santolina rosmarinifolia geographically isolated from diploid populations was investigated. The karyotype was relatively homogeneous, meiosis was regular and pollen was fertile in both cytotypes. An autopolyploid or allopolyploid origin for tetraploid cytotypes is discussed. Overall, 80.82% of all variance in achene weight, time t0, t50 and t90 of germination and accumulated germination rate was due to achene age at each ploidy level. Partition of the total phenotypic variance showed that there was extensive variation between ploidy levels. The mean of morphological characters was generally higher in polyploids. For diploid cytotypes, flower number, achene production and fruiting percentage were significantly higher than for tetraploid cytotypes. Cluster analysis indicated that the patterns of seedling morphology and development were similar in three diploid individuals and several tetraploids; the same analysis showed high similarity between diploid individuals of the natural populations, whereas tetraploid individuals showed high dissimilarity among themselves and with diploid individuals. Multiple correspondence analysis and logistic regression analysis indicated that qualitative characters contribute strongly to cytotype differentiation. In the northeastern part of Belgium, the Centaurea jacea complex shows extensive morphological variation and is represented by a diploid (2 n = 22) and a tetraploid (2 n = 44) cytotype. Polysomic inheritance of allozyme markers in the tetraploids, suggesting autopolyploidy, is here demonstrated for the first time. In order to test whether the two cytotypes occupy distinct habitats and possess different gene pools, patterns of allozymic and morphological variation were investigated in relation to ploidy level and site characteristics in 26 populations from the Belgian Ardennes. The two cytotypes showed a parapatric distribution, the diploids occurring at higher elevations (mostly above 500 m) than the tetraploids (mostly below 500 m). Three mixed populations were found near the contact zone of the two cytotypes. Within the mixed populations no triploid plant and no evidence for gene flow between cytotypes were found, despite widely overlapping flowering periods. The two cytotypes can be distinguished on the basis of morphological traits and enzymatic gene pools. The congruence of morphological and allozymic variation with chromosome numbers suggests a secondary contact between the two cytotypes with limited gene flow between them. The origin and persistence of the parapatric distribution are discussed.  相似文献
