Root growth in chickpea (Cicer arietinum) has been studied fromthe early vegetative phase to the reproductive stage in orderto elucidate its growth and maintenance respiration and to quantifythe translocation of assimilates from shoot to root. A carbonbalance has been drawn for this purpose using the growth andrespiration data. The increase in the sieve tube cross-sectionalarea was also followed simultaneously. Plants growing in a nutrient culture medium were studied todetermine the relative growth rate (RGR) 5–60 d aftergermination. RGR declined from 113 to 41 mg d–1 g–1during the measurement period. Simultaneous with the RGR analysis,respiration rate was also measured using an oxygen electrode.The respiration rate declined as the plants aged and a drasticreduction was recorded following anthesis. The relationshipbetween RGR and respiration rate was used to extrapolate themaintenance respiration (m) and growth respiration (1/YEG).The respiration quotient (r.q.) of the roots was 1.2 and theQ10 in the range 20–25 °C was 2·2. A carbon balance for the roots was constructed by subtractingthe carbon lost during respiration from that gained during growth.The roots were found to respire no less than 80% of the carbontranslocated. The increase in the cross-sectional area composed of sieve tubeswas measured near the root-shoot junction as the plants grew.Chickpea has storied sieve plates which simplifies these measurements.Their cross-sectional area increased during growth mainly becauseof an increase in sieve tube number. The diameter of individualsieve tubes remained constant. Specific mass transfer (SMT) values for seive tubes into theroots have been computed during various stages of growth. SMTvalues were relatively constant before anthesis (approx. 6·5g h–1 cm–2), but decreased following anthesis. Wedid not evaluate possible retranslocation from roots: any suchretranslocation would have the effect of increasing our SMTvalues. Plasmolytic disruption of plasmodesmata interconnecting metaphloemsieve element-companion cell complexes with small and largephloem parenchyma cells in the elongating region of internode2 ofPhaseolus vulgarisL. seedlings did not affect accumulationof phloem-imported14C-photosynthates and86rubidium. The membrane-impermeantdye, 5(6) carboxyfluorescein, loaded into leaf phloem as themembrane-permeant diacetate ester, was found not to move radiallyout of the importing sieve elements in the internode elongationregion. In contrast, the apoplasmic tracer, Calcuofluor White,rapidly moved laterally throughout all tissues of the elongationzone. Hexoses, sucrose and potassium were identified as themain osmotica in internode apoplasmic sap. Label asymmetry in[14C](fructosyl)sucrose was retained on accumulation by excisedstem segments. Uptake of [14C]sucrose and86rubidium by stemsegments exhibited saturation kinetics. Sucrose uptake was inhibitedby the slowly penetrating sulphydryl reagent, para-chloromercuribenzenesulphonicacid.In vitrorates of sucrose uptake, at apoplasmic concentrations,corresponded to its predictedin vivorate of delivery to thestem ground tissues from mature sieve elements when respiratorylosses were assumed to be confined to the stem phloem. For potassium,the total delivery rate could be accounted for by itsin vitrorateof uptake. The abscisic acid (ABA) content of wrinkled (rr) pea seed tissueshas been quantified during development using multiple-ion-monitoringcombined gas chromatography-mass spectrometry and a deuteratedinternal standard. The level of ABA in the embryo generallyincreased with increasing cotyledon fresh weight while thatin the testa showed a distinct maximum at the time of maximumendosperm volume and the slowing in the growth of the testa.Pods contained relatively little ABA on a fresh weight basis.The total seed ABA content showed a biphasic distribution, thefirst maximum following the maximum growth rate of the testaand the second that of the embryo. The biphasic distributionof ABA in the pea seed was confirmed using a second pea genotype,near-isogenic to the first except for the r locus, and by theanalysis of individual seeds using a radioimmunoassay for ABA.The first maximum was composed mainly of a testa component andthe second mainly of an embryo component. When plants were grownin different environments, wrinkled seeds were found to containslightly more ABA than round (RR) but this was only significantlate in development. Immature seeds were capable of metabolizing17'-deoxy ABA to ABA, as determined by incorporation of either3H or 2H, and the metabolite was present mainly in the testa.The production of ABA in pea seeds is discussed in relationto the development of the different seed tissues. Mycclia of Neurospora crassa wild type (FE SC no. 853), harvestedduring the exponential phase of growth on defined minimal mediaincorporated glycine-2-14C, serine-3-14C and formate-14C intoproteins, DNA and RNA. Supplementing the growth medium with1 mM glycine increased the flow of glycine and formate carboninto these products. In contrast, this supplement decreasedthe incorporation of serine-14C. When such cultures were preincubatedfor 30 min with adenine, formaldehyde, formate or L-methionine,labelling of the nucleic acids and protein fractions by glycine-2-14Cwas altered. Changes in the activity of adenine and guanine salvage in nucleotideand nucleic acid synthesis during the growth of Catharanthusroseus were investigated. Incorporation of [8-14C]adenine intoATP and ADP and that of [8-14C]guanine into GTP and GDP increasedmarkedly in the lag phase of cell growth and then sharply decreased.The incorporation into RNA from both precursors showed a similarpattern. Biochemical properties, i.e. endogenous abscisic acid, proline,sugars, respiration, adenosine phosphates and adenylate energycharge, and growth and moisture content were measured duringthe development of seeds of Machilus thunbergii. As dry matteraccumulated in the embryo during development, moisture content,ABA, proline, respiration and sugars all declined. At maturity,the dry mass of the seeds failed to attain a plateau beforethe period of natural seed shedding; the axis and cotyledonsreached moisture contents of 58 and 45%, respectively. Dryingof immature seeds at 73% relative humidity and 25 °C for30 d resulted in a complete loss of viability at all developmentalstages tested with the exception of mature seeds that were ableto tolerate a 5% decrease in moisture content before germinationdeclined. ABA was detected in all embryos tested, with a maximum value16.·16 µg g-1 d. wt about midway through development.Although the presence of ABA induced no tolerance to desiccationof mature seeds, it did coincide with decreased content of waterin the developing seeds and decreased respiration. Desiccationdamage of M. thunbergii seeds occurred when moisture contentwas still high (45%) and this damage was not related to theabsence of oligosaccharides in the mature seeds. We concludethat developing embryos and mature seeds of M. thunbergii haveproperties common to many recalcitrant seeds, with seeds beingsensitive to desiccation at all stages, having a prominent ABApeak, little proline, lacking oligosaccharides, and specifically,little dormancy and a moderate rate of respiration of matureseeds (0·9 µmol O2 min-1 g-1 f. wt). Adenosinetriphosphate content and energy charge decreased from stagefour to stage eight of seed development, then increased againto 103 nmol g-1 d. wt and 0·73, respectively, in matureseeds. The effect of environmental salinity on cell growth, and onthe composition and accumulation of compatible solutes, or osmotica,of five yeast strains (Aureobasidium pullulans, Candida sp.,Cryptococcus albidus var. albidus, Debaryomyces hansenii andRhodotorula rubra) was compared. All these yeast were isolatedfrom manne environments, but were able to grow in the absenceof salt and should therefore be considered as halotolerant strains.According to their specific cell growth rates at different saltconcentrations, these strains vary in their capacity to osmoticallyadjust to modifications in external salinity. Candida sp. appearsto be the most sensitive since the maximum salt concentrationat which it can grow is 1.54 mol 1-1 NaCl; however, it showedthe highest specific cell growth in the range of 0 to 1.54 mol1-1 NaCl. Aureobasidium pullulans, on the other hand, showedthe lowest specific growth rate, but the highest halotolerancerange from 0 to 5.13 mol 1-1 NaCl. Debaryomyces hansenii, incontrast, showed higher specific growth at this salinity rangeCryptococcus albidus var. albidus and Rhodotorula rubra showedsimilar specific cell growth rate values and halotolerance between0 and 2.45 mol 1-1 NaCl. The protein and carbohydrate contentof the biomass of the different yeast cells, as a result ofexternal salinity vanation, remained practically constant. Themost important effects of the increase in salt concentrationin the culture medium were the reduction of cell volume andthe accumulation of low-molecular-weight metabolites (LMWM).which appear to act as osmoregulators. Glycerol was found asthe major compatible solute in the different marine yeasts studiedherein with a total contribution of 64–96% of the internalcell osmolarity. Other LMWM, like carbohydrates and amino acids,contributed to a lesser extent to compensate for the rise inosmotic pressure promoted by the salinity of the external environment.  相似文献

Simultaneous observations on extension growth and respirationrate (oxygen consumption) of 2-mm. sections excised from theextension zone of roots of pea (Pisum sativum) growing in distilledwater and 0·5 per cent. sucrose have yielded resultsclosely similar to those of Brown and Sutcliffe (1950). Respirationrate is not obviously correlated with growth rate either inwater or in sucrose, but it is strongly correlated with sectionlength. Respiration rate per unit section length (¬per unitfresh weight) shows a marked downward drift during extensionand is affected little by growth conditions. Tentative suggestionsare advanced to account for the small differences between driftsin o·5 per cent. sucrose and those in distilled water. Medium agitation produces an immediate and sustained stimulationof growth but no stimulation of oxygen uptake until the latergrowth stages. Thus respiration per unit section length is unaffectedby agitation at any stage. A typical growth response to ß-indolylacetic acid(IAA) was obtained, with a maximum stimulation (of about 35per cent.) at 1 part in 1011 and inhibitions increasing progressivelywith concentration beyond the threshold of about i part in 109.Both percentage stimulation and percentage inhibition of growthwere independent of the presence of sucrose. Respiratory responses to ß-indolylacetic acid werecomplex. In water no immediate response could be detected witheither a growth-stimulatory (10–11) or a growth-inhibitory(10–-8) concentration, while in 0·5 per cent. sucrosethe inhibitory concentration prevented the small immediate respiratoryrise due to the sucrose, probably by impeding sugar entry. Duringthe subsequent period of rapid growth (up to 36 hours) the smallrespiratory responses observed closely followed the small growthresponses to both concentrations of IAA, suggesting that theformer are the direct result of the differences in section lengthinduced by the auxin. When growth ceases (at 48 hours) sectionswhich have grown considerably in sucrose show respiratory ratesstill closely correlated with section length, whereas in waterboth concentrations of auxin induce marked depressions in respirationrate. It is concluded that ß-indolylacetic acid in bothgrowth-stimulatory and growth- inhibitory concentrations hasno direct effect on the activity of the respiratory enzyme systemof growing root cells. The small respiratory responses are bestexplained as resulting from differential changes in sectionsize and correlated changes in the enzyme complements of thegrowing cell.  相似文献

Ochna pulchra Hook. is a deciduous broad-leaved tree in theMixed Bushveld vegetation of the Northern Transvaal. The growthand development of leaves taken from trees in the field werestudied from a stage shortly before bud break, in late spring,until they were fully expanded and at the peak of photosyntheticactivity. Leaf area was measured by photographing the leaf against a transparentmm2 grid. Finally a constant relationship between leaf area(A) and the linear dimension of length (L) and breadth (B) wasestablished: A = b x LB, where coefficient b = 0.72. Transverse sections of the lamina of the youngest leaves showeda five-layered plate meristem with a few functional conductingelements in the midrib. During further leaf development, celldivision was followed by means of autoradiography using [3H]thymidine.It was most active during the week after bud break. Leaf cell increment following on cell division made the majorcontribution to leaf growth resulting in a lamina that was atleast 90% expanded 4 weeks after bud break. The histologicalchanges accompanying cell division were observed using lightand electron microscopy. Even in late stages of leaf development mature and differentiatingstomata occurred together, limited to the abaxial epidermisand the midrib. Scanning electron microscopy showed stomataldistribution, their increasing density and gradual opening.The structure of these sunken stomata could reduce the outwarddiffusion of water vapour and increase the diffusion resistanceto carbon dioxide. Carbon assimilation rates of the developing leaves were measuredusing an IRGA (infra-red gas analyser) and their chlorophyllvalues were calculated. Photosynthesis was first measured amonth after bud break when the leaves were fully expanded, over50 % of the stomata exposed and leaf mesophyll tissue differentiatedwith mature chloroplasts. As yam bulbils (Dioscorea batatas Decne.) grow on the motherplants, dormancy of the bulbils develops gradually. This isevidenced by the fact that the sproutability of bulbils decreasedin proportion to their sizes, 2–4 mm to 8–12 mmin diameter when bulbils are detached and sown in Petri dishes.This decreasing sproutability was seen until the stratificationat 5°C for dormancy release was completed. During the growth process of attached bulbils, the contentsof batatasins I and II increased markedly per surface area andper bulbil although on a fresh weight basis they remained constant.The increases of batatasin III and abscisic acid (ABA) weremore striking and were evident in all comparisons. These results corresponded to different localization of batatasinsand ABA in bulbil issues. All of 3 batatasins were localizedin the outermost layers of the bulbils, whereas ABA was distributedevenly throughout their tissues. TA [4-ethoxy-1-(p-tolyl)-s-triazine-2,6 (1H, 3H)-dione] wasprepared chemically by intramolecular cyclization of IU [2-ethyl-3-methoxycarbonyl-1-(p-tolylcarbamoyl)-isourea],which has been reported to be an effective GA-synergist. TAalone slightly stimulated the shoot growth of rice seedlings,and in combination with GA showed a distinctly synergistic effecton the growth of rice shoots. The results suggest that TA andIU are nearly equal in their physiological activity. From theresults of the rice seedling test of these two compounds andtheir analogs, structure-activity relationships of isoureaswere correlated with those of triazinones. The results of applicationof IU, at 100 mg/liter to rice seedlings followed by extractionand silica gel thin-layer chromatography, suggested that IUwas biologically converted into its closed-ring form, TA, whichis a stable and potent GA-synergist. When Phaseolus mungo seeds were allowed to germinate in 5–20mM quinate solution in the dark, a marked increase in the endogenousshikimic acid level occurred in their tissues. The acid levelrose distincdy on the 2nd day of germination and reached a maximumon the 4th day. The quinate-14C fed to germinating seeds waspredominantly converted to shikimic acid, and little radioactivitywas found in 3-dehydroshikimic acid. When quinate-14C was suppliedsimultaneously with relatively high concentrations of 3-dehydroquinicacid or 3-dehydroshikimic acid, its conversionto shikimic acidwas restrained, but hardly any radioactivity was trapped ineither of the dehydro compounds. 3-Dehydroquinic acid-14C or3-dehydroshikimic acid-14C fed to the seeds was metabolizedpreferentially to shikimic acid. The experimental results arediscussed with respect to the metabolic relationship betweenquinic acid and odier alicyclic acids in the aromatic biosynthesisof P. mungo seedlings. Specific growth rates of Limnozhrix redekei, Planktothrix agardhii(cyanobacteria), Synedraacus, Stephanodiscus minutulus (diatoms),Scenedesmus acuminatus and Scenedesmus armatus (Chlorophyceae)were compared under different time structures of illumination,but the same daily light exposure, at 20C. Fluctuating irradiancesimulating a uniform rapid transport of the algal cells acrossthe aquatic light field on a cloudless day with Zeu/Zmix=1 wascompared with constant irradiance throughout the same photoperiodof 12 h length as well as a photoperiod of 6 h length. Fluctuatinglight (30 min for a cycle) resulted in a decrease in specificgrowth rates as compared with constant irradiance at the samephotoperiod length. This decrease amounts to 15–20% fordiatoms, 20–25% for Chlorophyceae and 35–40% forcyanobacteria, respectively. The decrease is somewhat lowerif the fluctuations simulating mixing are slower (60 min fora cycle). The specific growth rate is also decreased by a shorterphotoperiod, but this effect is more species specific. Regardingthe in vivo absorption spectra, fluctuating light or a shorterphotoperiod has little or no effect on the Chlorophyceae anddiatoms studied, whereas cyanobacteria show an increase in lightabsorption by chlorophyll a and phycobilins.  相似文献

We investigated the seasonal occurrence, wet : dry : carbon: nitrogen weight ratios, population biomass, gastric pouchcontents, and rates of feeding, growth and respiration of thescyphomedusa Aurelia aurita in the central part of the InlandSea of Japan. Aurelia aurita medusae began to appear in January/Februaryas ephyrae, reached annual maximum body size in July/August,and disappeared, presumably due to death, by November. Initialslow growth in early spring was followed by a period of exponentialgrowth (mean growth rate: 0.069 d–1) between April andJuly. In the Ondo Strait, which is characterized by strong tidalmixing, the A. aurita population (mean carbon biomass: 66.0mg C m–3) overwhelmingly dominated the zooplankton-communitybiomass (mean biomass of micro- and mesozooplankton: 23.7 mgC m–3) between May and early August The gastric contentanalysis revealed that A. aurita ate almost all micro- and mesozooplankters,of which small copepods were most important. On the basis ofdigestion time for small copepods (60 min) and their abundancein the gastric pouch of field-collected A. aurita, we determinedthe weight specific feeding rates and clearance rates. The formerincreases linearly with increasing copepod abundance, but thelatter was relatively constant irrespective of the food supply.We also measured the respiration rates of A. aurita and expressedthem as functions of body weight and temperature. These physio-ecologicalparameters enabled us to construct the carbon budget of theA. aurita population typical of early summer in the Ondo Strait.Predicted population-feeding rate (6.07 mg C m–3 d–1)was higher than the population-food requirement for both metabolismand growth (4.55 mg C m–3 d–1), indicating thatfood supply was sufficient to sustain the observed growth rate.This feeding rate was equivalent to 26% of micro- and mesozooplanktonbiomass, a significant impact on zooplankton.  相似文献

A long-term, steady-state 13CO2 assimilation system at a constantCO2 concentration with a constant 13C abundance was designedand applied to quantitative investigations on the allocationof photoassimilated carbon in nodulated soya bean (Glycine maxL.) plants. The CO2 concentration in the assimilation chamberand its 13C abundance were maintained constant with relativevariances of less than ±0.5 per cent during an 8-h assimilationperiod. At the termination of 8-h 13CO2 assimilation by plantsat early flowering stage, the currently assimilated carbon relativeto total tissue carbon (measured by the degree of isotopic saturation)were for young leaves (including flower buds), 13.9 per cent;mature leaves, 15.7 per cent; stems+petioles, 5.9 per cent;roots, 5.4 per cent and nodules, 6.9 per cent, 48 h after theend of the 13CO2 assimilation period, they were 12.3, 7.5, 7.4,6.8 and 6.1 per cent, respectively. The treatment with a highconcentration of nitrate in the nutrient media significantlydecreased the allocation of 13C into nodules. Experiments on13CO2 assimilation by plants at the pod-filling stage were alsoconducted. Labelling by 13C was weaker than at the early floweringstage, but an intense accumulation of 13C into reproductiveorgans was observed. Poly(A)+ and poly(A)RNA from wounded potato tuber tissuesand crown gall tumors were separated from total RNA by oligodeoxythymidylicacid-cellulose affinity chromatography. The poly(A)+RNA wascharacterized by sucrose density gradient centrifugation, hybridizationwith 3(H)polyuridylic acid [Poly(U)] and in vitro translationin a rabbit reticulocyte lysate system. The tumor poly(A)+RNAwas a heterodisperse mixture from 3.5S to 35S. Upon poly(U)hybridization of the gradient fractions two major hybridizationpeaks at 7S and 21S and two peaks at 11S and 16S appeared. Inan in vitro translation system the poly(A)+RNA programmed thesynthesis of 23 different polypeptides of 9,000 to 79,800 daltonsmolecular weight as determined by SDS-polyacrylamide gel electrophoresis.The 21S poly(A)+RNA was about 5 times more active in in vitroprotein synthesis than the 7S poly(A)+RNA. The poly(A)+RNA from wounded tissues was also heterodisperse(from 4.5S to 31S) with a modal peak at 18S. This RNA codedfor at least 28 polypeptides, which were different from thoseof crown gall tumor tissues. On a per unit poly(A)+RNA basis the tumor RNA was slightly moreactive in translation than that from wounded tissues. The translationof tumor poly(A)+RNA was completely blocked by 0.5 mM 7-methylguanosine5'-phosphate, but not by 7-methylguanosine, suggesting the presenceof a 5'-cap structure. The growth of Escherichia coli strain B in a liquid medium wasfound to cease at a cell density of 5x109 cells per ml. (Thiscritical concentration is designated as the maximum or M-concentration.)Even cells harvested from the logarithmic growth phase couldnot divide at this or higher cell densities. Investigationson the metabolic activities of such cultures, however, showedthat the synthesis of cellular protein and nucleic acid wastaking place under such circumstances, showing that only someprocess (or processes) particularly related to cell divisionwas suppressed at the critical cell concentration in question. This finding led us to devise a new method of synchronizationof E. coli: cells harvested from a logarithmic phase were preincubatedat the critical concentration of 5x109 cells per ml for 45 minutes,and then diluted 100 times with fresh medium. Yellow prisms of asparagusic acid, with a molecular formulaof C4H6O2S2 were isolated from etiolated asparagus tissues (Asparagusofficinalis L.). This acid inhibits growth in lettuce and otherseedlings when applied in concentrations of 6.67x10–7Mto 6.67xl0–7M. Two approaches to quantifying relationships between nutrientsupply and plant growth were compared with respect to growth,partitioning, uptake and assimilation of NO3 by non-nodulatedpea (Pisum sativum L. cv. Marma). Plants grown in flowing solutionculture were supplied with NO3 at relative addition rates(RAR) of 0·03, 0·06, 0·12, and 0·18d–1, or constant external concentrations ([NO3)of 3, 10, 20, and 100 mmol m–3 over 19 d. Following acclimation,relative growth rates (RGR)approached the corresponding RARbetween 0·03–0.12 d-1, although growth was notlimited by N supply at RAR =0.18 d-1. Growth rates showed littlechange with [NO3–] between 10–100 mmol m–3(RGR=0·15 –0·16 d-1). The absence of growthlimitation over this range was suggested by high unit absorptionrates of NO3, accumulation of NO3 in tissues andprogressive increases in shoot: root ratio. Rates of net uptakeof NO3 from 1 mol m–3 solutions were assessed relativeto the growth-related requirement for NO3, showing thatthe relative uptake capacity increased with RGR between 0·03–0·06d–1 , but decreased thereafter to a theoretical minimumvalue at RGR  相似文献

This work examined three possible explanations of growth rateresponses to leaf area index (LAI) in which growth rate perunit of ground area (crop growth rate, CGR) increased to a plateaurather than decreasing above an optimum LAI at which all lightwas intercepted. Single leaf photosynthetic measurements, andwhole plant 24 h photosynthesis and respiration measurementswere made for isolated plants and plants in stands using Amaranlhushybridus, Chenopodium album, and two cultivars of Glycine maxgrown at 500 and 1000 µimol m–2 S–1 photosyntheticphoton flux density at 25 °C. CGR, relative growth rate(RGR), and LAI were determined from 24 h carbon dioxide exchangeand leaf area and biomass measurements. Respiration increasedrelative to photosynthesis with crowding in A. hybridus andthere was an optimum LAI for CGR. In contrast, the ratio ofrespiration to photosynthesis was constant across plant arrangementin the other species and they had a plateau response of CGRto LAI. Neither increased leaf photosynthetic capacity at highLAI nor a large change in biomass compared to the change inLAI could account for the plateau responses. It was calculatedthat maintenance respiration per unit of biomass decreased withdecreasing RGR in C. album and G. max, but not A. hybridus,and accounted for the plateau response of CGR to LAI. Sincesimilar decreases in maintenance respiration per biomass atlow RGR have been reported for several other species, a constantratio of respiration to photosynthesis may occur in more speciesthan constant maintenance respiration per unit of biomass.
