The intracellular free Na+ concentration ([Na+]i) increases during muscarinic stimulation in salivary acinar cells. The present study examined in rat sublingual acini the role of extracellular Mg2+ in the regulation of the stimulated [Na+]i increase using the fluorescent sodium indicator benzofuran isophthalate (SBFI). The muscarinic induced rise in [Na+]i was approximately 4-fold greater in the absence of extracellular Mg2+. When Na+ efflux was blocked by the Na+,K+-ATPase inhibitor ouabain, the stimulated [Na+]i increase was comparable to that seen in an Mg2+-free medium. Moreover, ouabain did not add further to the stimulated [Na+]i increase in an Mg2+-free medium suggesting that removal of extracellular Mg2+ may inhibit the Na+ pump. In agreement with this assumption, ouabain-sensitive Na+ efflux and rubidium uptake were reduced by extracellular Mg2+ depletion. Our results suggest that extracellular Mg2+ may regulate [Na+]i in sublingual salivary acinar cells by modulating Na+ pump activity.  相似文献

Bongkrekic acid and atractyloside, inhibitors of adenine nucleotide translocase, do not inhibit Ca2+ uptake and H+ production by pig heart mitochondria. However, bongkrekic acid, but not atractyloside, inhibits dinitrophenol-induced Ca2+ efflux and H+ uptake. Conversely, ruthenium red blocks Ca2+ uptake and H+ production but does not prevent dinitrophenol-induced Ca2+ efflux and H+ uptake by mitochondria. These results suggest that mitochondrial Ca2+ uptake and release exist as two independent pathways. The efflux of Ca2+ from mitochondria is mediated by a bongkrekic acid sensitive component which is apparently not identical to the ruthenium red sensitive Ca2+ uptake carrier.  相似文献

We measured intracellular Mg2+ concentration ([Mg2+]i) in rat ventricular myocytes using the fluorescent indicator furaptra (25°C). In normally energized cells loaded with Mg2+, the introduction of extracellular Na+ induced a rapid decrease in [Mg2+]i: the initial rate of decrease in [Mg2+]i (initial Δ[Mg2+]it) is thought to represent the rate of Na+-dependent Mg2+ efflux (putative Na+/Mg2+ exchange). To determine whether Mg2+ efflux depends directly on energy derived from cellular metabolism, in addition to the transmembrane Na+ gradient, we estimated the initial Δ[Mg2+]it after metabolic inhibition. In the absence of extracellular Na+ and Ca2+, treatment of the cells with 1 μM carbonyl cyanide p-(trifluoromethoxy)phenylhydrazone, an uncoupler of mitochondria, caused a large increase in [Mg2+]i from ∼0.9 mM to ∼2.5 mM in a period of 5-8 min (probably because of breakdown of MgATP and release of Mg2+) and cell shortening to ∼50% of the initial length (probably because of formation of rigor cross-bridges). Similar increases in [Mg2+]i and cell shortening were observed after application of 5 mM potassium cyanide (KCN) (an inhibitor of respiration) for ≥90 min. The initial Δ[Mg2+]it was diminished, on average, by 90% in carbonyl cyanide p-(trifluoromethoxy)phenylhydrazone-treated cells and 92% in KCN-treated cells. When the cells were treated with 5 mM KCN for shorter times (59-85 min), a significant decrease in the initial Δ[Mg2+]it (on average by 59%) was observed with only a slight shortening of the cell length. Intracellular Na+ concentration ([Na+]i) estimated with a Na+ indicator sodium-binding benzofuran isophthalate was, on average, 5.0-10.5 mM during the time required for the initial Δ[Mg2+]it measurements, which is well below the [Na+]i level for half inhibition of the Mg2+ efflux (∼40 mM). Normalization of intracellular pH using 10 μM nigericin, a H+ ionophore, did not reverse the inhibition of the Mg2+ efflux. From these results, it seems likely that a decrease in ATP below the threshold of rigor cross-bridge formation (∼0.4 mM estimated indirectly in the this study), rather than elevation of [Na+]i or intracellular acidosis, inhibits the Mg2+ efflux, suggesting the absolute necessity of ATP for the Na+/Mg2+ exchange.  相似文献

Two types of Na+-independent Mg2+ efflux exist in erythrocytes: (1) Mg2+ efflux in sucrose medium and (2) Mg2+ efflux in high Cl media such as KCl-, LiCl- or choline Cl-medium. The mechanism of Na+-independent Mg2+ efflux in choline Cl medium was investigated in this study. Non-selective transport by the following transport mechanisms has been excluded: K+,Cl- and Na+,K+,Cl-symport, Na+/H+-, Na+/Mg2+-, Na+/Ca2+- and K+(Na+)/H+ antiport, Ca2+-activated K+ channel and Mg2+ leak flux. We suggest that, in choline Cl medium, Na+-independent Mg2+ efflux can be performed by non-selective transport via the choline exchanger. This was supported through inhibition of Mg2+ efflux by hemicholinum-3 (HC-3), dodecyltrimethylammonium bromide (DoTMA) and cinchona alkaloids, which are inhibitors of the choline exchanger. Increasing concentrations of HC-3 inhibited the efflux of choline and efflux of Mg2+ to the same degree. The Kd value for inhibition of [14C]choline efflux and for inhibition of Mg2+ efflux by HC-3 were the same within the experimental error. Inhibition of choline efflux and of Mg2+ efflux in choline medium occurred as follows: quinine>cinchonine>HC-3>DoTMA. Mg2+ efflux was reduced to the same degree by these inhibitors as was the [14C]choline efflux.  相似文献

The K+ uptake pathways in yeast mitochondria are still undefined. Nonetheless, the K+-mediated mitochondrial swelling observed in the absence of phosphate (PO4) and in the presence of a respiratory substrate has led to propose that large K+ movements occur in yeast mitochondria. Thus, the uptake of K+ by isolated yeast mitochondria was evaluated. Two parallel experiments were conducted to evaluate K+ transport; these were mitochondrial swelling and the uptake of the radioactive K+ analog 86Rb+. The opening of the yeast mitochondrial unspecific channel (YMUC) was regulated by different PO4 concentrations. The high protein concentrations used to measure 86Rb+ uptake resulted in a slight stabilization of the transmembrane potential at 0.4 mM PO4 but not at 0 or 4 mM PO4. At 4 mM PO4 swelling was inhibited while, in contrast, 86Rb+ uptake was still observed. The results suggest that an energy-dependent K+ uptake mechanism was unmasked when the YMUC was closed. To further analyze the properties of this K+ uptake system, the Mg2+ and quinine sensitivity of both swelling and 86Rb+ uptake were evaluated. Under the conditions where the unspecific pore was closed, K+ transport sensitivity to Mg2+ and quinine increased. In addition, when Zn2+ was added as an antiport inhibitor, uptake of 86Rb+ increased. It is suggested that in yeast mitochondria, the K+ concentration is highly regulated by the equilibrium of uptake and exit of this cation through two specific transporters.  相似文献

Previous work showed that in hamster red cells the amiloride-sensitive (AS) Na+ influx of 0.8 mmol/liter cells/hr is not mediated by Na-H exchange as in other red cells, but depends upon intracellular Mg2+ and can be increased by 40-fold by loading cells with Mg2+ to 10 mm. The purpose of this study was to verify the connection of AS Na+ influx with Na-dependent, amiloride-sensitive Mg2+ efflux and to utilize AS Na+ influx to explore that pathway.Determination of unidirectional influx of Na+ and net loss of Mg2+ in parallel sets of cells showed that activation by extracellular [Na+] follows a simple Michaelis-Menten relationship for both processes with a K m of 105–107 mm and that activation of both processes is sigmoidally dependent upon cytoplasmic [Mg2+] with a [Mg2+]0.5 of 2.1–2.3 mm and a Hill coefficient of 1.8. Comparison of Vmax for both sets of experiments indicated a stoichiometry of 2 Na: l Mg. Amiloride inhibits Na+ influx and Mg2+ extrusion in parallel (K i = 0.3 mm). Like Mg2+ extrusion, amiloride-sensitive Na+ influx shows an absolute requirement for cytoplasmic ATP and is increased by cell swelling. Hence, amiloride-sensitive Na+ influx in hamster red cells appears to be through the Na-Mg exchange pathway.There was no amiloride-sensitive Na+ efflux in hamster red cells loaded with Na+ and incubated with high [Mg2+] in the medium with or without external Na+, nor with ATP depletion. Our earlier studies indicate that stromal alkalinization is essential for light-induced increase in free Mg2+ concentration ([Mg2+]) in chloroplast. Stromal [Mg2+] was increased by dark incubation of chloroplasts in the K+-gluconate medium (pH 8.0), or by NH4Cl. These results indicate that stromal alkalinization can induce an increase in stromal [Mg2+] without illumination. Some inhibitors of envelope proton-translocating ATPase activity involved in H+ efflux inhibited the alkalinization-induced increase in [Mg2+].  相似文献

The trimeric derivative of 16,16-dimethyl-15-dehydroprostaglandin B1 (termed tri-Calciphor), which protects tissues against ischemic damage, induced Ca2+ efflux and swelling in mitochondria in the absence of phosphate, Mg2+ and ATP. When glutamate/malate rather than succinate was the substrate, higher tri-Calciphor concentrations were required for the ionophoretic activity. Ca2+ efflux and mitochondrial swelling induced by tri-Calciphor were completely inhibited by ATP, phopsphate and Mg2+ added together, and partially inhibited with phosphate plus either ATP or Mg2+. Between 0 and 7 μM added Ca2+ and in the presence of phosphate, ATP and Mg2+, tri-Calciphor stimulated the uptake of Ca2+ by mitochondria and increased the efficiency of buffering of extramitochondrial Ca2+. Thus depending on the assay conditions, two different effects involving Ca2+ movements and mitochondria are observed with tri-Calciphor.  相似文献

The components of magnesium efflux in squid axons have been studied under internal dialysis and voltage clamp conditions. The present report rules out the existence of an ATP-dependent, Na0- and Mg0-independent Mg2+ efflux (ATP-dependent Mg2+ pump) leaving the Mg2+---Na+ exchange system as the only mechanism for Mg2+ extrusion. The main features of the Mg2+ efflux are: (1) The efflux is completely dependent on ATP. (2) The efflux can be activated either by external Na+ (forward Mg2+---Na+ exchange) or external Mg2+ (Mg2+---Mg2+ exchange). (3) The mobility of the Mg2+ exchanger in the Na0+-loaded form is greater than that in the Mg2+-loaded one. (4) In variance with the Na+---Ca2+ exchange mechanism, Mg2+---Mg2+ exchange is not activated by external monovalent cations. (5) ATPγS replaces ATP in activating Mg2+---Na+ exchange suggesting that a phosphorylation/dephosphorylation process regulates this transport mechanism.  相似文献

Parallel measurements of Ca2+ uptake, oxygen consumption, endogenous Mg2+ efflux, and swelling in rotenone-poisoned rat liver and rat heart mitochondria showed that heart mitochondria is much more resistant to uncoupling by Ca2+ in the presence of phosphate than rat liver mitochondria. The extent of Mg2+ efflux and swelling induced by Ca2+ accumulation are much less pronounced in heart mitochondria. Uncoupling and swelling in liver mitochondria seem to result from the loss of membrane-bound Mg2+ as a consequence of Ca2+ recycling across the membrane as induced by phosphate. Exogenous Mg2+ protects liver mitochondria against the deleterious effects of Ca2+ by inhibiting a ruthenium red-insensitive Ca2+ efflux induced by phosphate. Phosphate does not induce recycling of Ca2+ in heart mitochondria. On the other hand, heart mitochondria respiring on NAD-linked substrates or with succinate in the absence of rotenone behave like liver mitochondria with respect to the alterations caused by Ca2+ recycling. In heart mitochondria the recycling of Ca2+ is related to the redox state of pyridine nucleotides, which suggests that the ruthenium red-insensitive efflux of Ca2+ is subject to metabolic control. In addition it has been observed that Sr2+does not undergo cyclic movements across the membrane. The data indicate that the efflux pathway is more specific for Ca2+ than the ruthenium red-sensitive influx transporter.  相似文献

Mitochondria modulate cellular Ca2+ signals by accumulating the ion via a uniporter and releasing it via Na+- or H+-exchange. In smooth muscle, inhibition of mitochondrial Ca2+ uptake inhibits Ca2+ release from the sarcoplasmic reticulum (SR) via inositol-1,4,5-trisphosphate-sensitive receptors (IP3R). At least two mechanisms may explain this effect. First, localised uptake of Ca2+ by mitochondria may prevent negative feedback by cytosolic Ca2+ on IP3R activity, or secondly localised provision of Ca2+ by mitochondrial efflux may maintain IP3R function or SR Ca2+ content. To distinguish between these possibilities the role of mitochondrial Ca2+ efflux on IP3R function was examined. IP3 was liberated in freshly isolated single colonic smooth muscle cells and mitochondrial Na+–Ca2+ exchanger inhibited with CGP-37157 (10 μM). Mitochondria accumulated Ca2+ during IP3-evoked [Ca2+]c rises and released the ion back to the cytosol (within 15 s) when mitochondrial Ca2+ efflux was active. When mitochondrial Ca2+ efflux was inhibited by CGP-37157, an extensive and sustained loading of mitochondria with Ca2+ occurred after IP3-evoked Ca2+ release. IP3-evoked [Ca2+]c rises were initially unaffected, then only slowly inhibited by CGP-37157. IP3R activity was required for inhibition to occur; incubation with CGP-37157 for the same duration without IP3 release did not inhibit IP3R. CGP-37157 directly inhibited voltage-gated Ca2+ channel activity, however SR Ca2+ content was unaltered by the drug. Thus, the gradual decline of IP3R function that followed mitochondrial Na+–Ca2+ exchanger inhibition resulted from a gradual overload of mitochondria with Ca2+, leading to a reduced capacity for Ca2+ uptake. Localised uptake of Ca2+ by mitochondria, rather than mitochondrial Ca2+ efflux, appears critical for maintaining IP3R activity.  相似文献

The efflux of Ca2+ from previously Ca2+ -loaded heart, liver or kidney mitochondria is accompanied by an approximately equal loss of endogenous Mg2+ irrespective of the agent applied to stimulate efflux which may be Na+, a mercurial, a long chain fatty derivative or thyroxin. The proportion of Mg2+ (and of the accompanying adenine nucleotide) in relation to the Ca2+ is diminished if Mg2+ is added to the medium. The similarity between data from different mitochondria and with different agents accords with the ion losses involving a common factor such as generation within the membrane of lysophospholipid by Ca2+ in transit. It is shown that lysophospholecithin stimulates Ca2+ efflux with a hyperbolic concentration dependence.  相似文献

Most measurements of nutrient uptake use either hydroponic systems or soil-grown roots that have been disturbed by excavation. The first objective of this study was to test how root excavation affects nitrate uptake. Rates of NO3? uptake by mycorrhizal loblolly pine (Pinus taeda L.) seedlings were measured in intact sand-filled columns, hydroponics, and disturbed sand-filled columns. Total nitrate uptake in intact sand-filled columns was higher than in disturbed columns, indicating that disturbance lowers uptake. Transferring plants from the sand-filled columns to hydroponics had little effect on NO3? uptake beyond delaying uptake for an hour. The second objective of this study was to determine whether NH4+, Ca2+, Mg2+ and K+ uptake could be studied using sand-filled columns, since previous studies had tested this method only for nitrate uptake. Uptake rates of NH4+ and K+ were positive, while Ca2+ and Mg2+ uptake rates were negative in intact sand-filled columns, indicating that net efflux may occur even without physical disturbance to the root system. The sand-filled column approach has some limitations, but holds promise for conducting nutrient uptake studies with minimal disturbance to the root system.  相似文献

Isolated hepatocytes in physiological [Na+] 0 tightly maintain [Mg2+] i . Upon β-adrenergic stimulation or in the presence of permeable cAMP, hepatocytes release 5–10% (1–3 mM Mg2+) of their total Mg2+ content. However, isolated basolateral liver plasma membranes (bLPM), release Mg2+ in the presence of [Na+] o even in the absence of catecholamine stimulation. The data indicate that a physiological brake for Mg2+ efflux is present in the hepatocyte and is removed upon cellular signaling. In contrast, this regulation “brake” is absent in purified bLPM thus rendering them fully active. The present study was carried out to reconstruct the missing regulatory component. Activation of Mg2+ extrusion in intact cells is consistent with cAMP dependent phosphorylation of the transporter or a regulatory protein. Treatment of bLPM with a non-specific phosphatase such as alkaline phosphatase (AP), decreased Mg2+ efflux by 70% compared to untreated bLPM. When AP-treated bLPM were loaded with protein kinase A (PKA), and stimulated with permeable cAMP, Mg2+ transport fully recovered. These data suggest that phosphorylation of the Na+/Mg2+ exchanger or a nearby protein activates the Mg2+ transport mechanism in hepatocytes.  相似文献

Swelling of nonenergized heart mitochondria suspended in acetate salts appears to depend on the activity of an endogenous cation/H+ exchanger. Passive swelling in acetate shows a characteristic cation selectivity sequence of Na+ >Li+ >K+, Rb+, Cs+, or tetramethylammonium, a sharp optimum at pH 7.2–7.3, activation by Ca2+, and loss of activity on aging which can be related to loss of endogenous K+. The reaction is nearly insensitive to either addition of exogenous Mg2+ or removal of membrane Mg2+ with EDTA. Each of these characteristics of passive swelling in acetate salts is duplicated in chloride media when tripropyltin is added to induce Cl?/OH? exchange. In contrast to nonenergized mitochondria, swelling of respiring mitochondria has been postulated to depend on electrophoretic uptake of cations in response to an interior negative membrane potential. Respiration-dependent swelling in acetate shows an indistinct cation selectivity sequence with Li+ and Na+ supporting higher rates of swelling at higher efficiency than K+, Rb+, and Cs+. The high rates of respiration-dependent swelling in Li+ and Na+ are inhibited by low levels of exogenous Mg2+ (Ki of 5–10 μm), but a significant swelling with almost no cation selectivity persists in the presences of 2 mm Mg2+. Removal of membrane Mg2+ by addition of EDTA strongly activates the rate of respiration-dependent swelling and converts a sigmoid dependency of swelling rate on Li+ concentration to a hyperbolic one with a Km of about 14 mm Li+. The cation selectivity and Mg2+ dependence of the reaction induced in chloride salts by tripropyltin are identical to these properties in acetate. Energy-dependent swelling in acetate shows optimum activity at pH 6.5 which appears related to the availability of free acetic acid, since the corresponding reaction induced in chloride shows a broad optimum at about pH 7.5. These studies support the concept that monovalent cations enter nonenergized mitochondria by electroneutral exchange with protons but penetrate respiring mitochondria by electrophoretic movement through one or more uniport pathways. They further suggest that both a Mg2+-sensitive uniport with high activity for Na+ and Li+ and a Mg2+-insensitive pathway with little cation discrimination are available in the membrane.  相似文献

This study investigates the effect of magnesium (Mg2+) on the secretory responses and the mobilization of calcium (Ca2+) and Mg2+ evoked by cholecystokinin-octapeptide (CCK-8) in the exocrine rat pancreas. In the isolated intact perfused pancreas CCK-8 (10–10 M) produced marked increases in juice flow and total protein output in zero and normal (1.1 mM) extracellular Mg2+ [Mg2+]o compared to a much reduced secretory response in elevated (5 mM and 10 mM) [Mg2+]o Similar effects of perturbation of [Mg2+]o on amylase secretion and 45Ca2+ uptake (influx) were obtained in isolated pancreatic segments. In pancreatic acinar cells loaded with the fluorescent bioprobe fura-2 acetomethylester (AM), CCK-8 evoked marked increases in cytosolic free Ca2+ concentration [Ca2+]i in zero and normal [Mg2+]o compared to a much reduced response in elevated [Mg2+]o Pretreatment of acinar cells with either dibutyryl cyclic AMP (DB2 cAMP) or forskolin had no effect on the CCK-8 induced changes in [Ca2+]i. In magfura-2-loaded acinar cells CCK-8 (10–8 M) stimulated an initial transient rise in intracellular free Mg2+ concentration [Mg2+]i followed by a more prolonged and sustained decrease. This response was abolished when sodium Na+ was replaced with N-methyl-D-glucamine (NMDG). Incubation of acinar cells with 10 mM Mg2+ resulted in an elevation in [Mg2+]i. Upon stimulation with CCK-8, [Mg2+]i. decreased only slightly compared with the response obtained in normal [Mg2+]o. CCK-8 caused a net efflux of Mg2+ in pancreatic segments; this effect was abolished when extracellular sodium [Na+]o was replaced with either NMDG or choline. The results indicate that Mg2+ can regulate CCK-8-evoked secretory responses in the exocrine pancreas possibly via Ca2+ mobilization. Moreover, the movement of Mg2+ in pancreatic acinar cells is dependent upon extracellular Na+.  相似文献

In rat erythrocytes, the regulation of Na+/Mg2+ antiport by protein kinases (PKs), protein phosphatases (PPs), intracellular Mg2+, ATP and Cl was investigated. In untreated erythrocytes, Na+/Mg2+ antiport was slightly inhibited by the PK inhibitor staurosporine, slightly stimulated by the PP inhibitor calyculin A and strongly stimulated by vanadate. PMA stimulated Na+/Mg2+ antiport. This effect was completely inhibited by staurosporine and partially inhibited by the PKC inhibitors Ro-31-8425 and BIM I. Participation of other PKs such as PKA, the MAPK cascade, PTK, CK I, CK II, CAM II-K, PI 3-K, and MLCK was excluded by use of inhibitors. Na+/Mg2+ antiport in rat erythrocytes can thus be stimulated by PKCα.In non-Mg2+-loaded erythrocytes, ATP depletion reduced Mg2+ efflux and PMA stimulation in NaCl medium. A drastic activation of Na+/Mg2+ antiport was induced by Mg2+ loading which was not further stimulated by PMA. Staurosporine, Ro-31-8425, BIM I and calyculin A did not inhibit Na+/Mg2+ antiport of Mg2+-loaded cells. Obviously, at high [Mg2+]i Na+/Mg2+ antiport is maximally stimulated. PKCα or PPs are not involved in stimulation by intracellular Mg2+. ATP depletion of Mg2+-loaded erythrocytes reduced Mg2+ efflux and the affinity of Mg2+ binding sites of the Na+/Mg2+ antiporter to Mg2+. In non-Mg2+-loaded erythrocytes Na+/Mg2+ antiport essentially depends on Cl. Mg2+-loaded erythrocytes were less sensitive to the activation of Na+/Mg2+ antiport by [Cl]i.  相似文献

Addition to rat liver mitochondria of 2 mM inorganic phosphate or 0.15 mM diamide, a thiol-oxidizing agent, induced an efflux of endogenous Mg2+ linear with time and dependent on coupled respiration. No net Ca2+ release occurred under these conditions, while a concomitant release of K+ was observed. Mg2+ efflux mediated either by Pi or low concentrations of diamide was completely prevented by EGTA, Ruthenium red, and NEM. These reagents also inhibited the increased rate of state 4 respiration induced both by Pi and diamide. At higher concentrations (0.4 mM), diamide induced an efflux of Mg2+ which was associated also with a release of endogenous Ca2+. Under these conditions EGTA completely prevented Mg2+ and K+ effluxes, while they were only partially inhibited by Ruthenium red and NEM. It is assumed that Mg2+ efflux, occurring at low diamide concentrations or in the presence of phosphate, is dependent on a cyclic in-and-out movement of Ca2+ across the inner mitochondrial membrane, in which the passive efflux is compensated by a continuous energy linked reuptake. This explains the dependence of Mg2+ efflux on coupled respiration, as well as the increased rate of state 4 respiration. On the other hand, Mg2+ deficiency is also known to be a risk factor for the same category of diseases. In the present study, we examined the effect of imipramine on Mg2+ homeostasis in vascular smooth muscle, especially via melastatin‐type transient receptor potential (TRPM)‐like Mg2+‐permeable channels. The intracellular free Mg2+ concentration ([Mg2+]i) was measured using 31P‐nuclear magnetic resonance (NMR) in porcine carotid arteries that express both TRPM6 and TRPM7, the latter being predominant. pHi and intracellular phosphorus compounds were simultaneously monitored. To rule out Na+‐dependent Mg2+ transport, and to facilitate the activity of Mg2+‐permeable channels, experiments were carried out in the absence of Na+ and Ca2+. Changing the extracellular Mg2+ concentration to 0 and 6 mM significantly decreased and increased [Mg2+]i, respectively, in a time‐dependent manner. Imipramine statistically significantly attenuated both of the bi‐directional [Mg2+]i changes under the Na+‐ and Ca2+‐free conditions. This inhibitory effect was comparable in influx, and much more potent in efflux to that of 2‐aminoethoxydiphenyl borate, a well‐known blocker of TRPM7, a channel that plays a major role in cellular Mg2+ homeostasis. Neither [ATP]i nor pHi correlated with changes in [Mg2+]i. The results indicate that imipramine suppresses Mg2+‐permeable channels presumably through a direct effect on the channel domain. This inhibitory effect appears to contribute, at least partially, to the link between antidepressants and the risk of vascular diseases.  相似文献

Abstract: The effect of replacement of extracellular Na+ with N-methyl-d -glucamine (NMG) on P2 receptor signaling pathways was investigated in dibutyryl cyclic AMP-differentiated NG108-15 cells. Benzoylbenzoic ATP (BzATP) dose-dependently increased the cytosolic Ca2+ concentration ([Ca2+]i) with an EC50 value of 230 µM. Replacement of Na+ with NMG as well as removal of Mg2+ from the bathing buffer potentiated ethidium bromide uptake, [Ca2+]i increase, and 45Ca2+ uptake in response to ATP or BzATP. In contrast, in the presence of 5 mM Mg2+ to limit the amount of ATP4?, replacement of Na+ with NMG had no effect on the ATP-induced [Ca2+]i increase but caused a markedly larger [Ca2+]i increase when the calculated concentration of ATP4? was >10 µM. The calculated EC50 value for ATP4? stimulation of the [Ca2+]i increase was 23 µM in NG108-15 cells. In vascular smooth muscle cells, intracellular Ca2+ release was the major pathway for the ATP-induced [Ca2+]i increase; both removal of Mg2+ and replacement of Na+ with NMG did not affect the action of ATP. These data suggest that ATP4?-promoted pores are antagonized by Na+ and Mg2+ in dibutyryl cyclic AMP-differentiated NG108-15 cells.  相似文献
