Insulin increases the turnover rate of Na+-K+-ATPase in human fibroblasts   总被引:1,自引:0,他引:1  
Insulin stimulates K+ transport by theNa+-K+-ATPase in human fibroblasts. In othercell systems, this action represents an automatic response to increasedintracellular [Na+] or results from translocation oftransporters from an intracellular site to the plasma membrane. Here weevaluate whether these mechanisms are operative in human fibroblasts.Human fibroblasts expressed the 1 but not the2 and 3 isoforms ofNa+-K+-ATPase. Insulin increased the influx ofRb+, used to trace K+ entry, but did not modifythe total intracellular content of K+, Rb+, andNa+ over a 3-h incubation period. Ouabain increasedintracellular Na+ more rapidly in cells incubated withinsulin, but this increase followed insulin stimulation ofRb+ transport. Bumetanide did not prevent the increasedNa+ influx or stimulation ofNa+-K+-ATPase. Stimulation of theNa+-K+- ATPase by insulin did not produce anymeasurable change in membrane potential. Insulin did not affect theaffinity of the pump toward internal Na+ or the number ofmembrane-bound Na+-K+-ATPases, as assessed byouabain binding. By contrast, insulin slightly increased the affinityof Na+-K+-ATPase toward ouabain. Phorbol estersdid not mimic insulin action on Na+-K+-ATPaseand inhibited, rather than stimulated, Rb+ transport. Theseresults indicate that insulin increases the turnover rate ofNa+-K+-ATPases of human fibroblasts withoutaffecting their number on the plasma membrane or modifying theirdependence on intracellular [Na+].

The Na+-K+-ATPase and the ERK1/2 pathway appear to be linked in some fashion in a variety of cells. The Na+-K+-ATPase inhibitor ouabain can promote ERK1/2 activation. This activation involves Src, intracellular Ca2+ concentration ([Ca2+]i) elevation, reactive oxygen species (ROS) generation, and EGF receptor (EGFR) transactivation. In contrast, ERK1/2 can mediate changes in Na+-K+-ATPase activity and/or expression. Thus signaling between ERK1/2 and Na+-K+-ATPase can occur from either direction. Whether such bidirectionality can occur within the same cell has not been reported. In the present study, we have demonstrated that while ouabain (1 mM) produces only a small (50%) increase in ERK1/2 phosphorylation in freshly isolated rat salivary (parotid acinar) epithelial cells, it potentiates the phosphorylation of ERK1/2 by submaximal concentrations of carbachol, a muscarinic receptor ligand that initiates fluid secretion. Although ERK1/2 is only modestly phosphorylated when cells are exposed to 1 mM ouabain or 10–6 M carbachol, the combination of these agents promotes ERK1/2 phosphorylation to near-maximal levels achieved by a log order carbachol concentration. These effects of ouabain are distinct from Na+-K+-ATPase inhibition by lowering extracellular K+, which promotes a rapid and large increase in ERK1/2 phosphorylation. ERK1/2 potentiation by ouabain (EC50 100 µM) involves PKC, Src, and alterations in [Ca2+]i but not ROS generation or EGFR transactivation. In addition, inhibition of ERK1/2 reduces Na+-K+-ATPase activity (measured as stimulation of QO2 by carbachol and the cationophore nystatin). 研究了二化螟Chilo suppressalis线粒体Na+-K+-ATPase和Ca2-Mg2-ATPase的生物化学性质以及9种常用杀虫剂对这两种酶活性的影响。结果表明, 二化螟线粒体Na+-K+-ATPase和Ca2-Mg2-ATPase的最适反应条件为pH值7.4,温度37℃。 Na+-K+-ATPase的米氏常数(Km)为0.42 mmol/L,最大反应速度(Vmax)为302.47 nmol/(min·mg) 。Ca2-Mg2-ATPase的Km为0.40 mmol/L,Vmax为128.04 nmol/(min·mg)。药剂浓度为1×10-4 mol/L时,5种菊酯类杀虫剂对离体ATPase活性抑制的顺序为:溴氰菊酯>联苯菊酯>百树菊酯>三氟氯氰菊酯和氟硅菊酯;对二化螟Na+-K+-ATPase的抑制率分别为40.12%、39.69%、27.27%、19.49%和18.71%;对Ca2-Mg2-ATPase的抑制率分别为29.27%、23.78%、19.88%、11.64%和14.34%。硫丹对二化螟Na+-K+-ATPase和Ca2-Mg2-ATPase的抑制率均为17.46%。甲胺磷和呋喃丹对Ca2-Mg2-ATPase的抑制率分别为27.16%和17.42%,对Na+-K+-ATPase则几乎没有抑制作用。实验结果还表明, 在1.6×10-7~1×10-4 mol/L的浓度范围内,上述9种杀虫剂对二化螟ATPase活性的抑制率存在明显的剂量-效应关系。  相似文献

Ouabain, aspecific inhibitor ofNa+-K+-ATPase,was coupled to epoxy agarose via a 13-atom spacer to make an affinitycolumn that specifically bindsNa+-K+-ATPase.Na+-K+-ATPasefrom rat and dog kidney was bound to the column and was eluted as afunction of enzyme conformation, altered by adding specificcombinations of ligands.Na+-K+-ATPasefrom both sources bound to the column in the presence of Na + ATP + Mgand in solutions containing 30 mM K. No binding was observed in thepresence of Na or Na + ATP. These experiments suggest thatNa+-K+-ATPasebinds to the column under the same conditions that it binds tountethered ouabain.Na+-K+-ATPasealready bound to the column was competitively eluted with excess freeNa + ouabain or with Na + ATP. The latter eluted active enzyme. Forcomparable amounts of boundNa+-K+-ATPase,Na + ouabain and Na + ATP eluted more rat than dogNa+-K+-ATPase,consistent with the lower affinity of the ratNa+-K+-ATPasefor ouabain. The ouabain-affinity column was used to purify activeNa+-K+-ATPasefrom rat kidney microsomes and rat adrenal glomerulosa cells. Thespecific activity of the kidney enzyme was increased from ~2 to 15 µmolPi · mg1 · min1.Na+-K+-ATPasepurified from glomerulosa cells that were prelabeled with [32P]orthophosphatewas phosphorylated on the -subunit, suggesting that these cellscontain a kinase that phosphorylatesNa+-K+-ATPase.

We investigated the effect ofinhibiting Na+-K+-ATPase on the basolateral18-pS K+ channel in the cortical collecting duct (CCD) ofthe rat kidney. Inhibiting Na+-K+-ATPase withstrophanthidin decreased the activity of the 18-pS K+channel and increased the intracellular Ca2+ to 420 nM.Removal of extracellular Ca2+ abolished the effect ofstrophanthidin. When intracellular Ca2+ was raised with 5 µM ionomycin or A-23187 to 300, 400, and 500 nM, the activity of the18-pS K+ channel in cell-attached patches fell by 40, 85, and 96%, respectively. To explore the mechanism ofCa2+-induced inhibition, the effect of 400 nMCa2+ on channel activity was studied in the presence ofcalphostin C, an inhibitor of protein kinase C, or KN-93 and KN-62,inhibitors of calmodulin-dependent kinase II. Addition of calphostin Cor KN-93 or KN-62 failed to block the inhibitory effect of highconcentrations of Ca2+. This suggested that the inhibitoryeffect of high concentrations of Ca2+ was not mediated byprotein kinase C or calmodulin-dependent kinase II pathways. To examinethe possibility that the inhibitory effect of high concentrations ofCa2+ was mediated by the interaction of nitric oxide withsuperoxide, we investigated the effect of 400 nM Ca2+ onchannel activity in the presence of 4,5-dihydroxy-1,3-benzenedisulfonic acid (Tiron) orN-nitro-L-arginine methyl ester.Pretreatment of the tubules with 4,5-dihydroxy-1,3-benzenedisulfonicacid or N-nitro-L-arginine methylester completely abolished the inhibitory effect of 400 nMCa2+ on channel activity. Moreover, application of4,5-dihydroxy-1,3-benzenedisulfonic acid reversed the inhibitory effectof strophanthidin. We conclude that the effect of inhibitingNa+-K+-ATPase is mediated by intracellularCa2+ and the inhibitory effect of high concentrations ofCa2+ is the result of interaction of nitric oxide with superoxide.

Cardiotonic glycosides, like ouabain, inhibit Na+-K+-ATPase. Recent evidence suggests that low molar concentrations of ouabain alter cell growth. Studies were conducted to examine the effect of ouabain on Akt phosphorylation and rate of cell proliferation in opossum kidney (OK) proximal tubule cells. Cells exposed to 10 nM ouabain displayed increased Akt Ser473 phosphorylation, as evidenced by an increase in phospho-Akt Ser473 band density. Ouabain-stimulated Akt Ser473 phosphorylation was inhibited by pretreatment with phosphatidylinositol 3-kinase (PI3K) inhibitors (LY294002 and wortmannin), a PLC inhibitor (edelfosine), and an Akt inhibitor. Moreover, ouabain-mediated Akt Ser473 phosphorylation was suppressed by reduction of extracellular calcium (EGTA) or when intracellular calcium was buffered by BAPTA-AM. An inhibitor of calcium store release (TMB-8) and an inhibitor of calcium entry via store-operated calcium channels (SKF96365) also suppressed ouabain-mediated Akt Ser473 phosphorylation. In fura-2 AM-loaded cells, 10 nM ouabain increased capacitative calcium entry (CCE). Ouabain at 10 nM did not significantly alter baseline cytoplasmic calcium concentration in control cells. However, treatment with 10 nM ouabain caused a significantly higher ATP-mediated calcium store release. After 24 h, 10 nM ouabain increased the rate of cell proliferation. The Akt inhibitor, BAPTA-AM, SKF96365, and cyclopiazonic acid suppressed the increase in the rate of cell proliferation caused by 10 nM ouabain. Ouabain at 10 nM caused a detectable increase in 86Rb uptake but did not significantly alter Na+-K+-ATPase (ouabain-sensitive pNPPase) activity in crude membranes or cell sodium content. Taken together, the results point to a role for CCE and Akt phosphorylation, in response to low concentrations of ouabain, that increase the rate of cell proliferation without inhibiting Na+-K+-ATPase-mediated ion transport. Brain edema that forms during the early stages of stroke involves increased transport of Na+ and Cl across an intact blood-brain barrier (BBB). Our previous studies have shown that a luminal BBB Na+-K+-Cl cotransporter is stimulated by conditions present during ischemia and that inhibition of the cotransporter by intravenous bumetanide greatly reduces edema formation in the rat middle cerebral artery occlusion model of stroke. The present study focused on investigating the effects of hypoxia, which develops rapidly in the brain during ischemia, on the activity and expression of the BBB Na+-K+-Cl cotransporter, as well as on Na+-K+-ATPase activity, cell ATP content, and intracellular volume. Cerebral microvascular endothelial cells (CMECs) were assessed for Na+-K+-Cl cotransporter and Na+-K+-ATPase activities as bumetanide-sensitive and ouabain-sensitive 86Rb influxes, respectively. ATP content was assessed by luciferase assay and intracellular volume by [3H]-3-O-methyl-D-glucose and [14C]-sucrose equilibration. We found that 30-min exposure of CMECs to hypoxia ranging from 7.5% to 0.5% O2 (vs. 19% normoxic O2) significantly increased cotransporter activity as did 7.5% or 2% O2 for up to 2 h. This was not associated with reduction in Na+-K+-ATPase activity or ATP content. CMEC intracellular volume increased only after 4 to 5 h of hypoxia. Furthermore, glucose and pyruvate deprivation increased cotransporter activity under both normoxic and hypoxic conditions. Finally, we found that hypoxia increased phosphorylation but not abundance of the cotransporter protein. Extrusion of protons as a response to high-NaCl stress in intactmung bean roots was investigated at different external concentrationsof Ca2+ ions ([Ca2+]ex). The extrusion of protons was graduallyenhanced in the roots exposed to 100 mM NaCl, and high [Ca2+]exdiminished this enhancement of the extrusion. Vesicles of plasmalemmaand tonoplast were prepared from the roots and the H+-translocatingATPase (H+-ATPase) activities associated with the two typesof membrane and the H+-pyrophosphatase (H+-PPase) activity ofthe tonoplast were assayed. The plasmalemma ATPase was stimulatedin parallel with dramatic increases in the intracellular concentrationof Na+([Na+]in). High [Ca2+]ex prevented the increase in [Na+]inand diminished the stimulation of ATPase activity. The tonoplastATPase showed a rapid response to salt stress and was similarlystimulated even at high [Ca2+]M. The activities of both ATPaseswere, however, insensitive to concentrations of Na+ ions upto 100 HIM. By contrast, H+-PPase activity of the tonoplastwas severely inhibited with increasing [Na+]in under salt stressand recovered with high [Ca2+]ex. These findings suggest thathigh-NaCl stress increases the intracellular concentration ofNa+ ions in mung bean roots, which inhibits the tonoplast H+-PPase,and the activity of the plasmalemma H+-ATPase is thereby stimulatedand regulates the cytoplasmic pH. Oxidative stress during pathological conditionssuch as ischemia-reperfusion is known to promote the formationof hypochlorous acid (HOCl) in the heart and to result in depression ofcardiac sarcolemmal (SL)Na+-K+-ATPaseactivity. In this study, we examined the direct effects of HOCl on SLNa+-K+-ATPasefrom porcine heart. HOCl decreased SLNa+-K+-ATPaseactivity in a concentration- and time-dependent manner. Characterization ofNa+-K+-ATPaseactivity in the presence of different concentrations of MgATP revealeda decrease in the maximal velocity(Vmax) value, without a change in affinity for MgATP on treatment of SL membranes with 0.1 mM HOCl. TheVmax value ofNa+-K+-ATPase,when determined in the presence of different concentrations ofNa+, was also decreased, butaffinity for Na+ was increasedwhen treated with HOCl. Formation of acylphosphate by SLNa+-K+-ATPasewas not affected by HOCl. Scatchard plot analysis of[3H]ouabain bindingdata indicated no significant change in the affinity or maximum bindingcapacity value for ouabain binding following treatment of SL membraneswith HOCl. Western blot analysis ofNa+-K+-ATPasesubunits in HOCl-treated SL membranes showed a decrease (34 ± 9%of control) in the 1-subunitwithout any change in the 1- or2-subunits. These data suggestthat the HOCl-induced decrease in SLNa+-K+-ATPaseactivity may be due to a depression in the1-subunit of the enzyme.

Caloxin: a novel plasma membrane Ca2+ pump inhibitor   总被引:1,自引:0,他引:1  
Plasma membrane (PM) Ca2+ pump is aCa2+-Mg2+-ATPase that expels Ca2+from cells to help them maintain low concentrations of cytosolic Ca2+. There are no known extracellularly acting PMCa2+ pump inhibitors, as digoxin and ouabain are forNa+ pump. In analogy with digoxin, we define caloxins asextracellular PM Ca2+ pump inhibitors and describe caloxin2A1. Caloxin 2A1 is a peptide obtained by screening a random peptidephage display library for binding to the second extracellular domain(residues 401-413) sequence of PM Ca2+ pump isoform1b. Caloxin 2A1 inhibits Ca2+-Mg2+-ATPase inhuman erythrocyte leaky ghosts, but it does not affect basalMg2+-ATPase or Na+-K+-ATPase in theghosts or Ca2+-Mg2+-ATPase in the skeletalmuscle sarcoplasmic reticulum. Caloxin 2A1 also inhibitsCa2+-dependent formation of the 140-kDa acid-stableacylphosphate, which is a partial reaction of this enzyme. Consistentwith inhibition of the PM Ca2+ pump in vascularendothelium, caloxin 2A1 produces an endothelium-dependent relaxationthat is reversed byNG-nitro-L-arginine methyl ester.Thus caloxin 2A1 is a novel PM Ca2+ pump inhibitor selectedfor binding to an extracellular domain.

The effect of diabetes on sarcolemmal Na+-K+ pump function is important for our understanding of heart disease associated with diabetes and design of its treatment. We induced diabetes characterized by hyperglycemia but no other major metabolic disturbances in rabbits. Ventricular myocytes isolated from diabetic rabbits and controls were voltage clamped and internally perfused with the whole cell patch-clamp technique. Electrogenic Na+-K+ pump current (Ip, arising from the 3:2 Na+-to-K+ exchange ratio) was identified as the shift in holding current induced by Na+-K+ pump blockade with 100 µmol/l ouabain in most experiments. There was no effect of diabetes on Ip recorded when myocytes were perfused with pipette solutions containing 80 mmol/l Na+ to nearly saturate intracellular Na+-K+ pump sites. However, diabetes was associated with a significant decrease in Ip measured when pipette solutions contained 10 mmol/l Na+. The decrease was independent of membrane voltage but dependent on the intracellular concentration of K+. There was no effect of diabetes on the sensitivity of Ip to extracellular K+. Pump inhibition was abolished by restoration of euglycemia or by in vivo angiotensin II receptor blockade with losartan. Serum- and glucocorticoid-induced kinase 1 (SGK1) is thought to be an important regulator of Na+ reabsorption in the kidney. It has been proposed that SGK1 mediates the effects of aldosterone on transepithelial Na+ transport. Previous studies have shown that SGK1 increases Na+ transport and epithelial Na+ channel (ENaC) activity in the apical membrane of renal epithelial cells. SGK1 has also been implicated in the modulation of Na+-K+-ATPase activity, the transporter responsible for basolateral Na+ efflux, although this observation has not been confirmed in renal epithelial cells. We examined Na+-K+-ATPase function in an A6 renal epithelial cell line that expresses SGK1 under the control of a tetracycline-inducible promoter. The results showed that expression of a constitutively active mutant of SGK1 (SGK1TS425D) increased the transport activity of Na+-K+-ATPase 2.5-fold. The increase in activity was a direct consequence of activation of the pump itself. The onset of Na+-K+-ATPase activation was observed between 6 and 24 h after induction of SGK1 expression, a delay that is significantly longer than that required for activation of ENaC in the same cell line (1 h). SGK1 and aldosterone stimulated the Na+ pump synergistically, indicating that the pathways mediated by these molecules operate independently. This observation was confirmed by demonstrating that aldosterone, but not SGK1TS425D, induced an 2.5-fold increase in total protein and plasma membrane Na+-K+-ATPase 1-subunit abundance. Secretion of cerebrospinal fluid by the choroid plexus canbe inhibited by its cholinergic innervation. We demonstrated that carbachol inhibits the Na+-K+-ATPase in bovinechoroid tissue slices and investigated the mechanism. Many of theactions of cholinergic agents are mediated by nitric oxide (NO), whichplays important roles in fluid homeostasis. The inhibition ofNa+-K+-ATPase was blocked by the NO synthaseinhibitor [N-nitro-L-argininemethyl ester] and was quantitatively mimicked by the NO agonistssodium nitroprusside (SNP) and diethylenetriamine NO. Inhibition by SNPcorrelated with an increase in tissue cGMP and was abolished by1H-[1,2,4]oxadiazolo[4,3-a]quinoxalin-1-one, an inhibitor of soluble guanylate cyclase. Inhibition was mimicked bythe protein kinase G activator 8-bromo-cGMP and by okadaic acid, aninhibitor of protein phosphatases 1 and 2A. cGMP-dependent proteinkinase inhibitors Rp-8-pCPT-cGMP (0.5-5 µM) and KT-5823 (2.0 µM) did not block the effects of SNP, but higher concentrations ofthe more selective inhibitor (Rp-8-pCPT-cGMP) had a pharmacological inhibitory effect on Na+-K+-ATPase. The datasuggest that cholinergic regulation of theNa+-K+-ATPase is mediated by NO and involvesactivation of guanylate cyclase and elevation of cGMP.

Role of caveolae in signal-transducing function of cardiac Na+/K+-ATPase   总被引:2,自引:0,他引:2  
Ouabain binding toNa+/K+-ATPase activates Src/epidermal growthfactor receptor (EGFR) to initiate multiple signal pathways thatregulate growth. In cardiac myocytes and the intact heart, the earlyouabain-induced pathways that cause rapid activations of ERK1/2 alsoregulate intracellular Ca2+ concentration([Ca2+]i) and contractility. The goal of thisstudy was to explore the role of caveolae in these early signalingevents. Subunits of Na+/K+-ATPase were detectedby immunoblot analysis in caveolae isolated from cardiac myocytes,cardiac ventricles, kidney cell lines, and kidney outer medulla byestablished detergent-free procedures. Isolated rat cardiac caveolaecontained Src, EGFR, ERK1/2, and 20-30% of cellular contents of1- and 2-isoforms ofNa+/K+-ATPase, along with nearly all ofcellular caveolin-3. Immunofluorescence microscopy of adult cardiacmyocytes showed the presence of caveolin-3 and -isoforms inperipheral sarcolemma and T tubules and suggested their partialcolocalization. Exposure of contracting isolated rat hearts to apositive inotropic dose of ouabain and analysis of isolated cardiaccaveolae showed that ouabain caused 1) no change in totalcaveolar ERK1/2, but a two- to threefold increase in caveolarphosphorylated/activated ERK1/2; 2) no change in caveolar 1-isoform and caveolin-3; and 3) 50-60%increases in caveolar Src and 2-isoform. These findings,in conjunction with previous observations, show that components of thepathways that link Na+/K+-ATPase to ERK1/2 and[Ca2+]i are organized within cardiac caveolaemicrodomains. They also suggest that ouabain-induced recruitments ofSrc and 2-isoform to caveolae are involved in themanifestation of the positive inotropic effect of ouabain.

The cytoplasmic pH and the vacuolar pH in root-tip cells ofintact mung bean seedlings under high-NaCl stress were measuredby in vivo 31P-nuclear magnetic resonance (31P-NMR) spectroscopy.When roots were incubated with high levels (100 mM) of NaClat the control external concentration (0.5 mM) of Ca2+ ions,the vacuolar pH increased rapidly from 5.6 to 6.2 within 3 h,while the cytoplasmic pH only decreased by a mere 0.1 pH uniteven after a 24-h incubation under high-NaCl conditions. Theincrease in vacuolar pH induced by the high-NaCl stress wasdiminished by an increase in the external concentration of Ca2+ions from 0.5 mM to 5 mM. The intracellular concentration ofNa+ ions in the root-tip cells increased dramatically upon perfusionof the root cells with 100 mM NaCl, and high external levelsof Ca2+ ions also suppressed the in flow of Na+ ions into thecells. The vacuolar alkalization observed in salt-stressed rootsmay be related to the inhibition of an H+-translocating pyrophosphatasein the tonoplast, caused by the increase in the cytoplasmicconcentration of Na+ ions. It is suggested that, although thevacuolar pH increased markedly under salt stress, the cytoplasmicpH was tightly regulated by some unidentified mechanisms, suchas stimulation of the H+-translocating ATPase of the plasmalemma,in roots of mung bean under salt stress. Antibodies raised in rabbits against the purified erythrocyte membrane Ca2+ pumping ATPase were affinity-purified using an ATPase-Sepharose column. Addition of a few molecules of the purified antibody per molecule of ATPase was sufficient to inhibit the ATPase activity. Extensively washed ghosts or preincubated pure ATPase sometimes develop an appreciable Mg2+-ATPase activity. In such cases, the antibodies inhibited the Mg2+-ATPase as well as the Ca2+-ATPase. This is consistent with the hypothesis that a portion of the Mg2+-ATPase activity of ghosts is derived from the Ca2+-ATPase. When nitrophenylphosphatase activity was observed, both Mg2+ - and Ca2+-stimulated activities were observed. Only the Ca2+ activity was inhibited by the antibodies, confirming that this activity is due to the Ca2+ pump, and suggesting that the Mg2+-nitrophenylphosphatase is due to a separate enzyme. Amounts of antibody comparable to those which inhibited the Ca2+-ATPases had no effect on the Na+-K+-ATPase; 4-fold higher amounts of antibody significantly stimulated the Na+-K+-ATPase, but this effect of the antibody was not specific: Immunoglobulins from the nonimmune serum also significantly stimulated the Na+-K+-ATPase.In resealed erythrocyte membranes, antibodies incorporated into the ghosts inactivated the Ca2+-ATPase, while antibodies added to the outside had no significant effect.  相似文献

The effects of a new, potent, and selective inhibitor of the Na+/Ca2+ exchange, SEA-0400 (SEA), on steady-state outward (forward exchange), inward (reverse exchange), and Ca2+/Ca2+ transport exchange modes were studied in internally dialyzed squid giant axons from both the extra- and intracellular sides. Inhibition by SEA takes place preferentially from the intracellular side of the membrane. Its inhibition has the following characteristics: it increases synergic intracellular Na+ (Nai+) + intracellular H+ (Hi+) inactivation, is antagonized by ATP and intracellular alkalinization, and is enhanced by intracellular acidification even in the absence of Na+. Inhibition by SEA is still present even after 1 h of its removal from the experimental solutions, whereas removal of the cointeracting agents of inhibition, Nai+ and Hi+, even in the continuous presence of SEA, releases inhibition, indicating that SEA facilitates the reversible attachment of the natural Hi+ and Nai+ synergic inhibitors. On the basis of a recent model of squid Na+/Ca2+ exchange regulation (DiPolo R and Beaugé L. J Physiol 539: 791–803, 2002), we suggest that SEA acts on the Hi+ + Nai+ inactivation process and can interact with the Na+-free and Na+-bound protonized carrier. Despite extensive work in the field of glioblastoma research no significant increase in survival rates for this devastating disease has been achieved. It is known that disturbance of intracellular Ca2+ ([Ca2+]i) and intracellular pH (pHi) regulation could be involved in tumor formation. The sarco(endo)plasmic reticulum Ca2+-ATPase (SERCA) is a major regulator of [Ca2+]i. We have investigated the effect of inhibition of SERCA by thapsigargin (TG) on [Ca2+]i and pHi in human primary glioblastoma multiforme (GBM) cells and GBM cell lines, compared with normal human astrocytes, using the fluorescent indicators fura-2 and BCECF, respectively. Basal [Ca2+]i was higher in SK-MG-1 and U87 MG but not in human primary GBM cells compared with normal astrocytes. However, in tumor cells, TG evoked a much larger and faster [Ca2+]i increase than in normal astrocytes. This increase was prevented in nominally Ca2+-free buffer and by 2-APB, an inhibitor of store-operated Ca2+ channels. In addition, TG-activated Ca2+ influx, which was sensitive to 2-APB, was higher in all tumor cell lines and primary GBM cells compared with normal astrocytes. The pHi was also elevated in tumor cells compared with normal astrocytes. TG caused acidification of both normal and all GBM cells, but in the tumor cells, this acidification was followed by an amiloride- and 5-(N,N-hexamethylene)-amiloride-sensitive recovery, indicating involvement of a Na+/H+ exchanger. This report describes K+ efflux, K+ and Ca2+ uptake responses to endothelins (ET-1 and ET-3) in cultured endothelium derived from capillaries of human brain (HBEC). ET-1 dose dependently increased K+ efflux, K+ and Ca2+ uptake in these cells. ET-1 stimulated K+ efflux occurred prior to that of K+ uptake. ET-3 was ineffective. The main contributor to the ET-1 induced K+ uptake was ouabain but not bumetanide-sensitive (Na+-K+-ATPase and Na+-K+-Cl cotransport activity, respectively). All tested paradigms of ET-1 effects in HBEC were inhibited by selective antagonist of ETA but not ETB receptors and inhibitors of phospholipase C and receptor-operated Ca2+ channels. Activation of protein kinase C (PKC) decreased whereas inhibition of PKC increased the ET-1 stimulated K+ efflux, K+ and Ca2+ uptake in HBEC. The results indicate that ET-1 affects the HBEC ionic transport systems through activation of ETA receptors linked to PLC and modulated by intracellular Ca2+ mobilization and PKC.  相似文献

We examined protein kinase C (PKC)-dependentregulation ofNa+-K+-ATPasein frog mucociliary cells. Activation of PKC by12-O-tetradecanoylphorbol-13-acetate (TPA) or 1,2-dioctanoyl-sn-glycerol(diC8) either in intact cells or isolated membranes resulted in aspecific inhibition ofNa+-K+-ATPaseactivity by ~25-45%. The inhibitory effects in membranes exhibited time dependence and dose dependence [half-maximalinhibition concentration (IC50) = 0.5 ± 0.1 nM and 2.4 ± 0.2 µM, respectively, for TPA anddiC8] and were not influenced byCa2+. Analysis of the ouabaininhibition pattern revealed the presence of twoNa+-K+-ATPaseisoforms with IC50 values forcardiac glycoside of 2.6 ± 0.8 nM and 409 ± 65 nM,respectively. Most importantly, the isoform possessing a higheraffinity for ouabain was almost completely inhibited by TPA, whereasits counterpart was hardly sensitive to the PKC activator. The resultssuggest that, in frog mucociliary cells, PKC regulatesNa+-K+-ATPaseand that this action is related to the specificNa+-K+-ATPaseisoform.
