Catharanthus roseus(L.) G. Don cells acidified Mura-shige-Skoogmedium rapidly. Upon transfer to fresh medium, the medium pH(initially5.3) dropped below 4 within 2 d. This acidificationwas reversed under hypoxic conditions. The cells induced a similaracidification in a simple medium consisting of CaCl2, KCl, andglucose: medium pH dropped below 4 within 6 h. The acidificationwas accompanied by an influx of K+ at a H+(efflux)/K+ ratioof ca 0.6 as well as by an expansion of endogenous organic acidpool, in which malic and citric acids were the major components.Anoxia reversed all these processes: the direction of both K+and H+ fluxes reversed with a H+/K+ ratio of 1.70. Anoxia induceda cytoplasmic acidification from pH 7.6 (aerobic) to 7.4 asmeasured by 31P-NMR, accompanied by a rapid, long-lasting lactateaccumulation at expense of malic and citric acids. Evidencesuggested that accumulation of lactic acid was not a cause ofcytoplasmic acidification under anoxia, but a result of pH regulationby the biochemical pH-stat [Davies (1973) Symp. Soc. Exp. Unidirectional fluxes and the cytoplasmic and vacuolar contentsof potassium and sodium in root cells of intact barley seedlings(Hordeum vulgare L., cv. Villa) were determined by use of compartmentalanalysis. In addition, the net vacuolar accumulation Jcv andthe xylem transport øcx of K+ and Na+ were measured.Both of these data were needed for the evaluation of the effluxdata. Fluxes and compartmental contents of K+ and Na+ were comparableto data obtained with excised roots. The effect of the shoot-to-rootratio—as varied by partial excision of the seedlings seminalroots—on the fluxes and contents was investigated. Highershoot-to-root ratios induced an increase in xylem transport,in plasmalemma influx, and also in the cytoplasmic content ofK+ and Na+. With potassium the plasmalemma efflux was almostunaltered while the tonoplast fluxes and vacuolar content weredecreased (in presence of Na+). With sodium, on the other hand,the plasmalemma efflux and the tonoplast fluxes were also increasedin the plants having one root and a high shoot-to-root ratio.These changes occurred even under conditions of low humidity,when transpiration was low and guttation occurred. The latterwas also increased at the high shoot-to-root ratio. The observedchanges could be due to a relieved feedback control of ion fluxesby the shoot and mediated in part by a relatively higher supplyof photosynthates in the plants having one root In addition,hormonal signals were suggested to participate. In particulara possibly decreased level of cytokinins in the plants havingonly one root could contribute to the signal. The observed changesappear to be responses of the plant to an alteration that canoccur under natural conditions when the root system is damaged.  相似文献

Net fluxes of H+and Ca2+were measured in the mesophyll tissueof broad bean (Vicia faba L.) leaves and in protoplasts derivedfrom these cells. NaCl at 90 m M enhanced H+extrusion in bothprotoplasts and tissue, but in different ways. Proton extrusionwas inhibited by vanadate, suggesting the involvement of theplasma membrane H+-ATPase in cell responses to salinity. Therewas virtually no effect of NaCl on the net Ca2+flux in protoplasts,while in the tissue a large transient Ca2+efflux followed thesalt treatment. Salt-induced Ca2+efflux was essentially independentof external Ca2+concentrations in the range 0.1 to 10 m M. Also,Ca2+flux responses were ‘saturated’ above 50 m MNaCl. It is suggested that almost all the measured Ca2+fluxoriginates from Na+/Ca2+and H+/Ca2+ion exchange in the cellwall. This conclusion was supported by the results of modellingcation exchange in the cell wall. It has previously been shown that cotyledonary pricks inducedmodifications of ion levels (H+ and K+) in hypocotyl cells ofBidens pilosa. These modifications differed according to thelight quality: H + levels increased and K+ levels decreasedin white light (WL), whereas H+ levels decreased and K+ levelsincreased in blue light (BL). In this study, in order to determinethe mechanism responsible for these ionic modifications, plasmamembrane vesicles have been isolated and characterized fromhypocotyl cells. The effects of light quality and cotyledonarypricks on plasma-lemma ATPase activity (EC 3.6.1.3 [EC] ) were studied.Cotyledonary pricks induced, in WL, rapid (5 min) and transient(restoration in 60 min) inhibition of plasmalemma ATPase activity.Conversely, in BL, a rapid and transient stimulation was observed.These results suggest that, in Bidens pilosa, plasmalemma ATPaseis involved in 'short-term' ionic level modifications inducedby traumatisms. N-acetylchitooIigosaccharides (fragments of chitin) elicit defenseresponses, including phytoalexin production, in suspension-culturedrice cells. They induced rapid and transient membrane depolarizationaccompanied by a transient increase in net CP-efflux.The membrane depolarization was not affected by anaerobiosisor azide, suggesting that the major part of the depolarizationwas mediated by ion channels, not by energy-dependent ion pumps.Depolarization was partly inhibited in the presence of Ca2+-or Cl-channel blockers and highly inhibited by depletionof Ca2+ in the extracellular medium. A calcium ionophore, A23187 [GenBank] ,caused a transient depolarization but not an increase in Clefflux, while it did not inhibit the elic-itor-induced transientdepolarization and Cl efflux. These suggest that theinflux of Ca2+ from the extracellular space to the cytoplasmis necessary as an initial trigger but not sufficient for membranedepolarization, Cl efflux, and the following signalingprocesses. The application of D-glucose to solutions bathing excised maize,wheat, pea and bean roots causes a rapid depolarization of theelectrical potentials between the cut tops of the roots andthe bathing solutions. Similar effects are observed for theplasma membrane potentials of maize lateral roots. A flow cell apparatus was used to demonstrate qualitative andquantitative relations between glucose induced H+ influx andthe transient decrease in current through the root. The currentchanges appear to be due entirely to H+ fluxes. Current andH+ fluxes are strongly influenced by external pH, the optimumpH for glucose induced current change being about 4.0. A similarpH optimum was found for 3-O-methyl-D-glucopyranoside but 1-O-methyl--D-glucopyranosidedid not significantly affect the trans-root potential at anypH, suggesting a significant role for the anomeric hydroxylgroup of glucose. Compounds which depolarize the trans-root potential also inhibitthe glucose induced depolarization. Surface -SH groups are probablynot involved in the glucose/H+ cotransport. Eadie-Hofstee plots relating the depolarization of trans-rootpotential to the concentrations of D-glucose or 3-O-methyl-D-glucopyranosidehave shown that Km values increase with increasing monosaccharideconcentration and are very similar to reported values of 3-O-methyl-D-glucopyranosideuptake in maize root segments. Km values for a similar rangeof D-glucose concentrations do not vary significantly with pHor with membrane depolarization due to a 10-fold increase ofKCl concentration. However, Vmax is lowered by an increase inexternal pH or a decrease in trans-root potential. It appearsthat both proton and electrical gradients can affect glucoseinduced H+ influx. The auxin herbicide, 2, 4-dichlorophenoxyethanoic acid (0.01mM) stimulates the glucose induced depolarizations in a mannerconsistent with an increase in cytoplasmic pH. This is discussedin relation to the reported action of indole-3-acetic acid andfusicoccin on maize root tissue.  相似文献

H+ translocation driven by NO3, NO2 and N2O reductionswith endogenous substrates in cells of Rhodopseudomonas sphaeroidesforma sp. denitrificans was investigated by the oxidant pulsemethod. Upon injection of nitrogenous oxides to anaerobic cellsin darkness, an alkaline transient in the external medium wasobserved, followed by acidification. The alkaline transientwas enhanced by carbonyl cyanide m-chlorophenylhydrazone. When a viologen dye was used as an electron donor in the presenceof 1 mM Af-ethylmaleimide and 0.1 mM 2-n-heptyl-4-hydroxyquinoline-N-oxideto preclude respiration-linked H+ extrusion, addition of KNO3,KNO2 and N2O caused only a rapid alkalinization. The H+ consumptionstoichiometries, H+/2e ratios for NO3 reductionto NO2, NO2 reduction to 1/2 N2O and N2O reductionto N2 were –1.90, –3.18 and –2.04, respectively.These values agreed well with the fact that all reductions ofnitrogenous oxides in denitrification occur on the periplasmicside of the cytoplasmic membrane. When corrected for H+ consumption in the periplasm, the H+ extrusionstoichiometries, H+/2e ratios with endogenous substratesin the presence of K+/valinomycin for NO3 reduction toNO2, NO2 reduction to 1/2 N2O and N2O reductionto N2 were 4.05, 4.95 and 6.01, respectively. Reactive changes in free intracellularzinc cation concentration ([Zn2+]i) weremonitored, using the fluorescent probe Zinquin, in human lymphoma cells exposed to the DNA-damaging agent VP-16. Two-photon excitation microscopy showed that Zinquin-Zn2+ formscomplexes in cytoplasmic vesicles. [Zn2+]iincreased in both p53wt (wild type) and p53mut(mutant) cells after exposure to low drug doses. In p53mutcells noncompetent for DNA damage-induced apoptosis, elevated [Zn2+]i was maintained at higher drug doses,unlike competent p53wt cells that showed a collapse of thetransient before apoptosis. In p53wt cells, the[Zn2+]i rise paralleled an increase in p53and bax-to-bcl-2 ratio but preceded an increase in p21WAF1,active cell cycle arrest in G2, or nuclear fragmentation.Reducing [Zn2+]i, usingN,N,N',N'-tetrakis(2-pyridylmethyl)ethylenediamine, caused rapid apoptosis in both p53wt andp53mut cells, although cotreatment with VP-16 exacerbatedapoptosis only in p53wt cells. This may reflectchanged thresholds for proapoptotic caspase-3 activation incompetent cells. We conclude that the DNA damage-induced transient isp53-independent up to a damage threshold, beyond which competent cellsreduce [Zn2+]i before apoptosis.Early stress responses in p53wt cells take place in anenvironment of enhanced Zn2+ availability.

We have reported that ryanodine receptor (RyR) channels display three different responses to cytoplasmic free Ca2+ concentration ([Ca2+]) depending on their redox state (Marengo JJ, Hidalgo C, and Bull R. Biophys J 74: 1263–1277, 1998), with low, moderate, and high maximal fractional open times (Po). Activation by ATP of single RyR channels from rat brain cortex was tested in planar lipid bilayers with 10 or 0.1 µM cytoplasmic [Ca2+]. At 10 µM [Ca2+], low-Po channels presented lower apparent affinity to activation by ATP [[ATP] for half-maximal activation (KaATP) = 422 µM] than moderate-Po channels (KaATP = 82 µM). Oxidation of low-Po channels with thimerosal or 2,2'-dithiodipyridine (DTDP) gave rise to moderate-Po channels and decreased KaATP from 422 to 82 µM. At 0.1 µM cytoplasmic [Ca2+], ATP induced an almost negligible activation of low-Po channels. After oxidation to high-Po behavior, activation by ATP was markedly increased. Noise analysis of single-channel fluctuations of low-Po channels at 10 µM [Ca2+] plus ATP revealed the presence of subconductance states, suggesting a conduction mechanism that involves four independent subchannels. Intracellular K+ playsan important role in controlling the cytoplasmic ion homeostasis formaintaining cell volume and inhibiting apoptotic enzymes in thecytosol and nucleus. Cytoplasmic K+ concentration is mainlyregulated by K+ uptake viaNa+-K+-ATPase and K+ efflux throughK+ channels in the plasma membrane. Carbonyl cyanidep-trifluoromethoxyphenylhydrazone (FCCP), a protonophorethat dissipates the H+ gradient across the inner membraneof mitochondria, induces apoptosis in many cell types. In ratand human pulmonary artery smooth muscle cells (PASMC), FCCP opened thelarge-conductance, voltage- and Ca2+-sensitiveK+ (maxi-K) channels, increased K+ currentsthrough maxi-K channels [IK(Ca)], and inducedapoptosis. Tetraethylammonia (1 mM) and iberiotoxin (100 nM)decreased IK(Ca) by blocking the sarcolemmalmaxi-K channels and inhibited the FCCP-induced apoptosis inPASMC cultured in media containing serum and growth factors.Furthermore, inhibition of K+ efflux by raisingextracellular K+ concentration from 5 to 40 mM alsoattenuated PASMC apoptosis induced by FCCP and theK+ ionophore valinomycin. These results suggest thatFCCP-mediated apoptosis in PASMC is partially due to anincrease of maxi-K channel activity. The resultant K+ lossthrough opened maxi-K channels may serve as a trigger for cellshrinkage and caspase activation, which are major characteristics ofapoptosis in pulmonary vascular smooth muscle cells.

The extent by which salinity affects plant growth depends partlyon the ability of the plant to exclude NaCl. To study the uptakeof NaCl into excised roots of Zea mays L. cv. ‘Tanker’,two different techniques were applied. A root pressure probewas used to record steady state as well as transient valuesof root (xylem) pressure upon exposure of the root to mediacontaining NaCl and KCl as osmotic solutes. In treatments withNaCl, pressure/time responses of the root indicated a significantuptake of NaCl into the xylem. NaCl induced kinetics were completelyreversible when the NaCl solution was replaced by an isosmoticKCl solution. This indicated a passive movement of Na+-saltsacross the root cylinder. Root samples were taken at differenttimes of exposure to NaCl and prepared for X-ray microanalysis(EDX analysis). Radial profiles of ion concentrations (Na+,K+, Cl) were measured in cell vacuoles and xylem vesselsalong the root axis. Na+ appeared rapidly in mature xylem (earlymetaxylem) and living xylem (late metaxylem) before it was detectablein vacuoles of the root cortex. EDX results confirmed that thekinetics observed by the pressure probe technique correspondedmainly to an influx of Na+-salts into early metaxylem. In latemetaxylem, the uptake of Na+ was associated with a decline ofK+. The Na+/K+ exchange indicated a mechanism to reduce sodiumfrom the transpiration stream. Nicotine was used to induce an intracellular alkalinizationin Elodea densa leaves in order to study the regulation of theplasmalemma H+ pump activity by alkaline intracellular pH values.Nicotine was found to enter the cells rapidly in the unchargedform and to induce a significant intracellular pH increase,measured either directly as cell sap pH or as vacuolar and cytoplasmicpH by calculation from the distribution at equilibrium of labelledpH probes. The nicotine-induced alkalinization was associatedwith a progressive decrease in K+ uptake. A strong inhibitionof net H+ efflux was also evident in the presence of K+ in theexternal medium, whereas no nicotine effect on net H+ effluxwas detected in the absence of K+ (in spite of the larger accumulationof nicotine in the tissue) in agreement with a down-regulationof the activity of the K+-dependent plasmalemma H+-ATPase byalkaline intracellular pH values. The increase in vacuolar pHresulting from nicotine accumulation was small compared to thebase load calculated from the vacuolar buffer capacity and theintracellular dissociation of nicotine. Conversely, the nicotine-inducedincrease in cytoplasmic pH was considerably larger than expectedon the basis of the cytoplasmic buffer capacity and of the theoreticalaccumulation of nicotine in the experimental conditions adopted.A balance sheet between nicotine accumulation, intracellularalkalinization and malate system response was drawn up, andthe seeming discrepancies observed were discussed. Using excised low-salt roots of barley and Atriplex hortenslsthe transport of endogenous potassium through the xylem vesselswas studied It was enhanced by nitrate and additionally by sodiumions which apparently replaced vacuolar potassium which wasthen available in the symplasm of root cells for transport tothe shoot Vacuolar Na/K exchange also has been investigatedby measurements of longitudinal ion profiles in single rootsof both species. In Atriplex roots a change in the externalsolution from K+ to Na+ induced an exchange of vacuolar K+ forNa+, in particular in the subapical root tissues and led toincreased K+ transport and loss of K+ from the cortex. In inverseexperiments a change from Na+ to K+ did not induce an exchangeof vacuolar Na+; merely in meristematic tissues Na+—apparentlyfrom the cytoplasm—was extruded in exchange for K+. Inroots of barley seedlings without caryopsis, as in excised roots,a massive exchange of K+ for Na+ was observed in the continuouspresence of external 1.0 mM Na and 0.2 mM K. This exchange alsowas attributed to the vacuole and was most pronounced in theyoung subapical tissues. It did not occur, however, in the correspondingtissues in roots of fully intact barley seedlings. In these,the young tissues retained a relatively high K/Na ratio alsoin their vacuoles. Similarly, contrasting results were obtainedwith intact and excised roots of Zea mays L. Based on theseresults a scheme of the events that lead to selective cationuptake in intact barley roots is proposed. In this scheme acrucial factor of selectivity is sufficient phloem recirculationof K+ by the aid of which K+ rich cortical cells are formednear the root tip. When matured these cells are suggested tomaintain a high cytoplasmic K/Na ratio due to K+ dependent sodiumextrusion at the plasmalemma and due to recovery of vacuolarK+ by Na/K exchange across the tonoplast. Cytochrome c activates K+ channels before inducing apoptosis   总被引:10,自引:0,他引:10  
Cellshrinkage is an early prerequisite for apoptosis. Theapoptotic volume decrease is due primarily to loss ofcytoplasmic ions. Increased outward K+ currents have indeedbeen implicated in the early stage of apoptosis in many celltypes. We found that cytoplasmic dialysis of cytochrome c(cyt-c), a mitochondria-dependent apoptotic inducer,increases K+ currents before inducing nuclear condensationand breakage in pulmonary vascular smooth muscle cells. Thecyt-c-mediated increase in K+ currents tookplace rapidly and was not affected by treatment with a specificinhibitor of caspase-9. Cytoplasmic dialysis of recombinant (active)caspase-9 negligibly affected the K+ currents. Furthermore,treatment of the cells with staurosporine (ST), an apoptosisinducer that mediates translocation of cyt-c frommitochondria to the cytosol, also increased K+ currents,caused cell shrinkage, and induced apoptosis (determined byapoptotic nuclear morphology and TdT-UTP nick end labeling assay).The staurosporine-induced increase in K+ currents concurredto the volume decrease but preceded the activation of apoptosis(nuclear condensation and breakage). These results suggest that thecyt-c-induced activation of K+ channels and theresultant K+ loss play an important role in initiating theapoptotic volume decrease when cells undergo apoptosis.

The growth of garden orache, A triplex hortensis was studiedunder conditions of mild NaCl or Na2SO4 salinity. Growth, drymatter production and leaf size were substantially stimulatedat 10 mM and 50 mM Na+ salts. Increased growth, however, appearedto be due to a K+-sparing effect of Na+ rather than to salinityper se. The distribution of K+ and Na+ in the plant revealeda remarkable preference for K+ in the roots and the hypocotyl.In the shoot the K/Na ratio decreased strongly with leaf age.However, the inverse changes in K+ and Na+ content with leafage were dependent on the presence of bladder hairs, which removedalmost all of the Na+ from the young leaf lamina. Measurementsof net fluxes of K+ and Na+ into roots and shoots of growingAtriplex plants showed a higher K/Na selectivity of the netion flux to the root compared to the shoot. With increasingsalinity the selectivity ratio SK, Na* of net ion fluxes tothe roots and to the shoots was increased. The data suggestthat recirculation of K+ from leaves to roots is an importantlink in establishing the K/Na selectivity in A. hortensis plants.The importance of K+ recirculation and phloem transport forsalt tolerance is discussed. The effects of light/dark on cation fluxes in isolated guardcells of Commelina communis L. have been studied, using 86RbCland 22NaCl. Transfer to the dark has no effect on 86Rb influx,but produces a marked transient stimulation of 86Rb efflux,similar to that seen previously on adding ABA. The 86Rb effluxfalls on return to light only during the period of stimulatedflux; after the transient, return to light has no effect onefflux. The ability to produce this transient stimulation ontransfer to the dark is recovered in a subsequent light period.In general, in Na-loaded cells, the stimulated efflux is notseen. and the cells do not close in the dark. The results arenot consistent with a simple permeability or potential change,but suggest a specific ion excretion activated by the transferto the dark. Inlysinuric protein intolerance (LPI), impaired transport of cationicamino acids in kidney and intestine is due to mutations of theSLC7A7 gene. To assess the functional consequences of the LPI defect in nonepithelial cells, we have characterized cationic aminoacid (CAA) transport in human fibroblasts obtained from LPI patientsand a normal subject. In both cell types the bidirectional fluxes ofarginine are due to the additive contributions of two Na+-independent, transstimulated transport systems. One ofthese mechanisms, inhibited by N-ethylmaleimide (NEM) andsensitive to the membrane potential, is identifiable with systemy+. The NEM- and potential-insensitive component,suppressed by L-leucine only in the presence ofNa+, is mostly due to the activity of systemy+L. The inward and outward activities of the two systemsare comparable in control and LPI fibroblasts. Both cell types expressSLC7A1 (CAT1) and SLC7A2 (CAT2B and CAT2A) aswell as SLC7A6 (y+LAT2) and SLC7A7 (y+LAT1). Weconclude that LPI fibroblasts exhibit normal CAA transport throughsystem y+L, probably referable to the activity ofSLC7A6/y+LAT2.

Calcium ion is a key messenger in turgor regulation of internodalcells of Lamprothamnium succinctum in response to hypoosmotictreatment. An increase in the concentration of cytosolic freecalcium ion ([Ca2+]c) is prerequisite for the turgor regulation[Okazaki and Tazawa (1990) J. Membr. Biol. 114: 189], We examinedwhether or not a calcium-dependent protein kinase (CDPK) isinvolved in the Ca2+-mediated turgor regulation of Lamprothamniumcells. A 53-kDa CDPK which phosphorylated preferentially histoneH1 but poorly myelin basic protein or casein, was detected inthe cell extract of Lamprothamnium by an in-gel protein kinaseassay. This protein kinase was detected by Western blottingand was immunoprecipitated using an anti-Dunaliella tertiolectaCDPK antibody which can neutralize the Dunaliella CDPK activity[Yuasa et al. (1995) Plant Cell Physiol. 36: 699]. The 53-kDaCDPK was partially purified from Lamprothamnium and its activitywas shown to be inhibited by the antibody and K-252a, a proteinkinase inhibitor. Microinjection of the antibody into the cytosblof Lamprothamnium cells inhibited the decrease in turgor pressurein response to hypoosmotic treatment. However, a transient increasein [Ca2+]c, which was suggested by a transient reduction ofthe velocity of cytoplasmic streaming, was induced in antibody-injectedcells after hypoosmotic treatment. Turgor regulation upon hypoosmotictreatment was inhibited when the cells were treated with K-252a.These results imply that CDPK of Lamprothamnium functions ata down-stream position of Ca2+-mobilization in processing turgorregulation in response to hypoosmotic treatment. 2 These authors contributed equally to the work.  相似文献

Effect of Sudden Salt Stress on Ion Fluxes in Intact Wheat Suspension Cells   总被引:4,自引:0,他引:4  
Although salinity is one of the major problems limiting agriculturalproduction around the world, the underlying mechanisms of highNaCl perception and tolerance are still poorly understood. Theeffects of different bathing solutions and fusicoccin (FC),a known activator of plasma membrane ATPase, on plasma membranepotential (Em) and net fluxes of Na+, K+and H+were studied inwheat suspension cells (Triticum aestivum) in response to differentNaCl treatments. Emof cells in Murashige and Skoog (MS) mediumwas less negative than in cells exposed to a medium containing10 mM KCl + 0.1 m M CaCl2(KSM) and to a basic salt medium (BSM),containing 1 m M KCl and 0.1 m M CaCl2. Multiphasic Na+accumulationin cells was observed, peaking at 13 min after addition of 120m M NaCl to MS medium. This time scale was in good agreementwith net Na+flux changes measured non-invasively by moving ion-selectivemicroelectrodes (the MIFE system). When 120 m M NaCl was addedto all media studied, a quick rise of Na+influx was reversedwithin the first 20 min. In both 120 and 20 m M NaCl treatmentsin MS medium, net Na+efflux was observed, indicating that activeNa+transporters function in the plant cell response to saltstress. Lower external K+concentrations (KSM and BSM) and FCpre-treatment caused shifts in Na+fluxes towards net influxat 120 m M NaCl stress.
