Mcl-1 full-length (Mcl-11-350), a tightly regulated protein, plays an important role in protecting cells against apoptosis. Cleavage of Mcl-1 at Asp127 by caspase (Mcl-1C1) contributes to the regulation of Mcl-1 expression, but its pro-apoptotic function remains controversial. Here, we reported that Mcl-1128-350 expression induced caspase-dependent apoptosis. We demonstrated that Mcl-1128-350 but not Mcl-11-350 interacts with Bax. This interaction required an intact BH3 Mcl-1128-350 domain and leads to Bax activation and translocation to mitochondria. The silencing of Bax, but not of Bak, prevented Mcl-1128-350 induced apoptosis. In conclusion, Mcl-1128-350 exerts a pro-apoptotic function governed by its capacity to interact with Bax.

Red-black [TpiPr∗MoVO]2(μ-O)(μ-MoVIO4) (1, TpiPr∗ = hydrobis(3-isopropylpyrazolyl)(5-isopropylpyrazolyl)borate) has been isolated as a by-product in the synthesis of NEt4[TpiPrMo(CO)3] (TpiPr = hydrotris(3-isopropylpyrazolyl)borate) and characterized by spectroscopic and X-ray crystallographic techniques. The trinuclear, mixed-valence complex contains two distorted octahedral anti-TpiPr∗MoVO centers bridged by bent oxo (Mo-O-Mo av. 158.7°) and tetrahedral κO,κO′-molybdate ligands. The complex contains a six-membered, non-planar Mo3(μ-O)3 core and two 1,2-borotropically-shifted TpiPr∗ ligands (with the shifted pyrazolyl trans to MoV=O). Aerial decomposition of solid NEt4[TpiPrMo(CO)3] produces sky-blue, diamagnetic TpiPrMoO(iPrpz)(iPrpzH) (2, iPrpz- = 3-isopropylpyrazolate, iPrpzH = 3-isopropyl-2H-pyrazole). Molecules of 2 feature a tridentate fac-TpiPr ligand and mutually cis terminal oxo (MoO = 1.665(2) Å) and monodentate iPrpz and iPrpzH ligands. The latter are formed by B-N bond cleavage of TpiPr. The complex can also be synthesized by reacting NEt4[TpiPrMo(CO)3] with excess 3-isopropylpyrazole and dioxygen at 100 °C. Cleavage of the B-N bond(s) of TpiPr was also observed in the formation of TpiPrMoO(SPh)(iPrpzH) (3) as a by-product in the synthesis of TpiPrMoO2(SPh). In the monohydrate, 3 exhibits a distorted octahedral geometry defined by a tridentate fac-TpiPr ligand and mutually cis terminal oxo (MoO = 1.676(3) Å) and monodentate SPh and iPrpzH ligands. The pyrazole β-NH group is observed to participate in a hydrogen-bond to the lattice water molecule. The complex can be synthesized in high yield by reducing TpiPrMoO2(SPh) by HSPh or PPh3 in the presence of excess 3-isopropylpyrazole.  相似文献

The wild-type (dam+) and mutant (damh) forms of the bacteriophage T2 DNA adenine methylase have been partially purified; these enzymes methylate the sequence, 5/t' … G-A-Py … 3′ (Hattman et al., 1978a). However, in vitro methylation studies using phage λ DNA revealed the following: (1) T2 dam+ and damh enzymes differ in their ability to methylate λ DNA; under identical reaction conditions the T2 damh enzyme methylated λ DNA to a higher level than did the dam+ enzyme. However, the respective methylation sites are equally distributed on the l and r strands. (2) Methylation with T2 damh, but not T2 dam+ protected λ against P1 restriction. This was demonstrated by transfection of Escherichia coli (P1) spheroplasts and by cleavage with R·EcoP1. (3) T2 dam+ and damh were similarly capable of methylating G-A-T-C sequences on λ DNA; e.g. λ·dam3 DNA (contains no N6-methyladonine) methylated with either enzyme was made resistant to cleavage by R·DpnII. In contrast, only the T2 damh modified DNA was resistant to further methylation by M·EcoP1 (which methylates the sequence 5′ … A-G-A-C-Py … 3′; Hattman et al., 1978b). (4) λ·dam3 DNA was partially methylated to the same level with T2 dam+ or T2 damh; the two enzymes produced different patterns of G-A-C versus G-A-T methylation. We propose that the T2 dam+ enzyme methylates G-A-C sequences less efficiently than the T2 damh methylase; this property does not entirely account for the large difference in methylation levels produced by the two enzymes.  相似文献

Enhancer of rudimentary, e(r), encodes a small nuclear protein, ER, that has been implicated in the regulation of pyrimidine metabolism, DNA replication and cell proliferation. In Drosophila melanogaster, a new recessive Notch allele, Nnd-p, was isolated as a lethal in combination with an e(r) allele, e(r)p2. Both mutants are viable as single mutants. Nnd-p is caused by a P-element insertion in the 5′ UTR, 378-bp upstream of the start of translation. Together the molecular and genetic data argue that Nnd-p is a hypomorphic allele of N. The three viable notchoid alleles, Nnd-p, Nnd-1 and Nnd-3, are lethal in combination with e(r) alleles. Our present hypothesis is that e(r) is a positive regulator of the Notch signaling pathway and that the lethality of the N e(r) double mutants is caused by a reduction in the expression of the pathway. This is supported by the rescue of the lethality by a mutation in Hairless, a negative regulator of N, and by the synthetic lethality of dx e(r) double mutants. Further support for the hypothesis is a reduction in E(spl) expression in an e(r) mutant. Immunostaining localizes ER to the nucleus, suggesting a nuclear function for ER. A role in the Notch signaling pathway, suggests that e(r) may be expressed in the nervous system. This turns out to be the case, as immunostaining of ER shows that ER is localized to the developing CNS.  相似文献

The aim of this study was to characterize the role of CFTR during Cd2+-induced apoptosis. For this purpose primary cultures and cell lines originated from proximal tubules (PCT) of wild-type cftr+/+ and cftr?/? mice were used. In cftr+/+ cells, the application of Cd2+ (5 μM) stimulated within 8 min an ERK1/2-activated CFTR-like Cl? conductance sensitive to CFTRinh-172. Thereafter Cd2+ induced an apoptotic volume decrease (AVD) within 6 h followed by caspase-3 activation and apoptosis. The early increase in CFTR conductance was followed by the activation of volume-sensitive outwardly rectifying (VSOR) Cl? and TASK2 K+ conductances. By contrast, cftr?/? cells exposed to Cd2+ were unable to develop VSOR currents, caspase-3 activity, and AVD process and underwent necrosis. Moreover in cftr+/+ cells, Cd2+ enhanced reactive oxygen species (ROS) production and induced a 50% decrease in total glutathione content (major ROS scavenger in PCT). ROS generation and glutathione decrease depended on the presence of CFTR, since they did not occur in the presence of CFTRinh-172 or in cftr?/? cells. Additionally, Cd2+ exposure accelerates effluxes of fluorescent glutathione S-conjugate in cftr+/+ cells. Our data suggest that CFTR could modulate ROS levels to ensure apoptosis during Cd2+ exposure by modulating the intracellular content of glutathione.  相似文献

Deletions of the nitrogen fixation (nif) region of the Klebsiella genome were isolated by selecting for resistance to virulent phages whose resistance loci are adjacent to nif. The extent of the various deletions was monitored by assaying several different enzymes or gene products coded for by this segment of DNA. Three classes of deletion mutants were detected: (1) gluconate-6-phosphate dehydrogenase minus (gnd?), histidine minus but histidinol dehydrogenase plus (his?, his D+), nitrogenase plus (nif+), shikimate utilization plus (shu+); (2) gnd?, his D?, nif?, shu+; (3) gnd?, his D?, nif?, shu?. From these studies we conclude that the cluster of nif genes essential for nitrogenase activity is located on the genetic linkage map of Klebsiella between his and shu; the gene order in this region in thus phage-resistance locus (rfb?), gnd, his operon, nif, shu. Genetic analysis substantiates the finding that the nif cluster is located proximally to the operator end of the his operon.  相似文献

Tris(triazolyl)borate (Ttz) ligands are sterically similar to tris(pyrazolyl)borate (Tp) but complexes of Ttz show improved solubility in water and alcohols due to their propensity for forming hydrogen bonds. Recently developed bulky tris(triazolyl)borate ligands can produce four and five coordinate transition metal complexes and serve as models for enzyme active sites in an aqueous environment. Herein we report the synthesis of such complexes, i.e. (TtztBu,Me)ZnCl, (TtztBu,Me)ZnBr, (TtztBu,Me)NiCl, and (TtztBu,Me)CuCl, which were analyzed by X-ray crystallographic and spectroscopic methods [TtztBu,Me = tris(3-t-butyl-5-methyl-1,2,4-triazolyl)borate]. (TtztBu,Me)ZnCl crystallizes as two different polymorphs with cubic and monoclinic symmetry. Both polymorphs of (TtztBu,Me)ZnCl and (TtztBu,Me)ZnBr have tetrahedral zinc atoms whereas the geometries at the metal in (TtztBu,Me)NiCl and (TtztBu,Me)CuCl are distorted tetrahedral. All complexes are methanol soluble and they also dissolve in methanol/water mixtures with up to 60% water.  相似文献

The trpX mutation in Escherichia coli reduces trp operon attenuation in strains carrying wild-type tRNATrp. The trpX? phenotype is alleviated (attenuation is restored) in UGA-suppressor tRNATrp-carrying strains (Yanofsky &; Soll, 1977).The tRNA from various trpX? strains was characterized biochemically. Sequence analyses of wild-type tRNATrp and UGA suppressor tRNATrp, both derived from trpX? strains, reveal an unmodified A in the position (adjacent to the anticodon) normally occupied by the hypermodified base ms2i6A.In addition, several tRNAs from trpX? cells were characterized by RPC-5 column chromatography. We find that only tRNAs normally having ms2i6A exhibit altered elution profiles when compared to the homologous tRNAs from trpX? cells. Introduction of the UGA suppressor into trpX? cells does not restore normal Chromatographic behavior. These results suggest that the trpX gene product is necessary for the synthesis of ms2i6A. Thus, we propose that miaA (for the first gene involved in ms2i6A synthesis) replaces the trpX designation.The results reported here are discussed with regard to a model proposed by Lee &; Yanofsky (1977) in which efficient translation of the tandem trp codons in the leader sequence RNA is required for normal attenuation of the trp operon.  相似文献

The Ah locus regulates the induction of cytochrome P1-450 by foreign chemicals such as 3-methyl-cholanthrene and 2,3,7,8-tetrachlorodibenzo-p-dioxin. The induction process is controlled by the cytosolic Ah receptor. The cytosolic and nuclear Ah receptors were studied in the liver from inbred C57BL/6N (Ahb/Ahb) mice, inbred DBA/2N (Ahd/Ahd) mice and heterozygotes (Ahb/Ahd) and homozygotes (Ahd/Ahd) derived from the (C57BL/6N × DBA/2N)F1 × DBA/2N backcross. After [3H-1,6]-2,3,7,8-tetrachlorodibenzo-p-dioxin (3H-TCDD) is given in vivo, the receptor in Ahb/Ahb and Ahb/Ahd mice is detectable in the cytoplasm and nucleus; in Ahd/Ahd mice the receptor is not measurable in the cytosol, but is found in the nucleus at levels one fourth to one fifth of those in Ahb/Ahb mice. P1-450 (23S) mRNA content was estimated by Northern hybridization and by Rot analysis with a mouse P1-450 cloned cDNA. An excellent dose-response relationship (r = 0.99) was found between the amount of 3H-TCDD-Ah receptor complex appearing in the nucleus and the quantity of P1-450 mRNA induced in mice with all three possible Ah genotypes.  相似文献

O6-Alkylguanine derivatives sensitize tumor cells to chloroethylnitrosourea (CENU) chemotherapy by inactivation of O6-methylguanine-DNA methyltransferase (MGMT), which repairs CENU-induced O6-alkylguanines in DNA by accepting the alkyl group at a cysteine moiety. To test the biological significance of synthesized O6-fluorobenzylguanine derivatives, we measured their ability of inactivation of MGMT activity and their effects on the cytotoxicity of 1-(4-amino-2-methyl-5-pyrimidinyl)methyl-3-(2-chloroethyl)-3-nitrosourea hydrochloride (ACNU) in comparison with the effects of O6-benzylguanine and O6-phenylguanine. The O6-(4-and 3-fluorobenzyl)guanines considerably reduced the MGMT activity of HeLa S3 cell-free extract as did O6-benzylguanine. In contrast, O6-(2-fluorobenzyl)guanine and O6-phenylguanine had less of an effect on the activity. Two-hour pretreatment of O6-(4-and 3-fluorobenzyl)guanines potentiated ACNU cytotoxicity in HeLa S3 cells to a greater extent than did O6-(2-fluorobenzyl)guanine and O6-phenylguanine. The enhancement effects were consistent with the depletion of MGMT activity after the pretreatment of O6-fluorobenzylguanine derivatives. O6-Fluorobenzylguanines with a fluoro-substitution at the 4- or 3-position of the benzyl group were comparable to O6-benzylguanine and were powerful MGMT inactivators. The chemical features of the O6-benzyl group are a biologically important determinant in the reaction evolution with MGMT.  相似文献

Eight positional isomers of 61,6m-di-O-α-d-mannopyranosyl-cyclomaltooctaose (γCD) (m=2–5) and 6-O-α-(n-O-α-d-mannopyranosyl)-d-mannopyranosyl-γCD (n=2, 3, 4, and 6) in a mixture of products from γCD and d-mannose by condensation reaction of α-mannosidase from jack bean were isolated by HPLC. The structures of four isomers of 6-O-α-(n-O-α-d-mannopyranosyl)-d-mannopyranosyl-γCD were elucidated by NMR spectroscopy. On the other hand, four positional isomers of 61,6m-di-O-α-d-mannopyranosyl-γCD were determined by LC–MS analysis of degree of polymerization of the branched oligosaccharides produced by enzymatic degradation with bacterial saccharifying α-amylase (BSA), and combination of BSA and glucoamylase. Similarly cyclomaltodextrin glucanotransferase also digested these isomers.  相似文献

The ligand binding properties of folate chemotactic receptors on isolated membranes of Dictyostelium discoideum were analyzed. Three out of the four receptor states (BF, BS and BSS) were detected, showing rate constants and Kd values similar to those obtained for intact cells. Guanine nucleotides changed the proportioning of the receptor states as well as the rates of several conversions. (i) The transformation of BF into BS was inhibited by GDP but not by guanylyl imidodiphosphate (GuaPP[NH]P) or GTP. (ii) The number of BS sites was lowered by GTP and GuaPP[NH]P. (iii) The binding to BSS was lowered by GTP and GDP, but increased by GuaPP[NH]P. (iv) The rate of disappearance of BSS was increased by GTP, but not by GuaPP[NH]P. Effects of guanine nucleotides were not observed after treatment of the membrane preparations with 15 mg/ml bovine serum albumin. This treatment caused the detection of a binding type different from the types described previously. The affinity of this binding site was extremely high (Kd ≤ 0.2 nM for N10-methylfolic acid), while the dissociation was relatively slow (k−1 ≤ 3·10−4 s−1). It is proposed that bovine serum albumin uncouples the folate receptor from a guanine nucleotide regulatory (G) protein in an irreversible manner. A model is presented in which the four receptor states correspond to distinct interactions with a G protein and GDP or GTP.  相似文献

Two isomers (R,S,R,S- and R,R,S,S-) of five coordinate complex [Cu(L)Cl]+ have been separated and characterised. These two isomers have significantly different spectrochemical and electrochemical properties. Absorption maximum of R,S,R,S-[Cu(L)Cl]+ shifts to longer wavelength and its reduction potential shifts to more positive direction comparing those of R,R,S,S-[Cu(L)Cl]+. R,S,R,S-[Cu(L)Cl]+ is significantly distorted to trigonal-bipyramidal structure, whereas R,R,S,S-[Cu(L)Cl]+ retains almost square-planar geometry. The average bond distance of Cu-N in basal plane of R,S,R,S-[Cu(L)Cl]+ is longer by 0.024 Å than that of R,R,S,S-[Cu(L)Cl]+, whereas the bond distance of Cu-Cl in former is shorter by 0.200 Å than that in latter. The isolated square-planar complexes of R,R,S,S- and R,S,R,S-[Cu(L)](ClO4)2 are converted to the R,R,S,S- and R,S,R,S-[Cu(L)Cl]+ by the addition of Cl in nitromethane solution with the rate constants, k=1.70 (±0.02) and 8.31 (±0.07) M−1 s−1, respectively.  相似文献

Protease inhibitor (PI) polymorphism was observed in the laboratory opossum,Monodelphis domestica, by either one-dimensional acid polyacrylamide gel electrophoresis (PAGE; pH 4.6) or isoelectric focusing (pH 3.5-5.0) followed by immunoblotting with rabbit antiserum to human α1-antitrypsin; but acid PAGE produced superior resolution of the PI proteins. Family studies demonstrated an inheritance of nine codominant autosomal alleles,PI D ,PI E ,PI F ,PI G ,PI H ,PI I ,PI J ,PI K , andPI M , and a population study revealed frequencies of 0.411, 0.010, 0.341, 0.034, 0.023, 0.071, 0.035, 0.020, and 0.055, respectively.  相似文献

(i) The activity of purified NAD-specific isocitrate dehydrogenase from bovine heart was stimulated by free Ca2+ in the presence of ADP and subsaturating levels of magnesium isocitrate, but not in absence of ADP. However, Ca2+ was not absolutely required for ADP activation. This was particularly apparent when free Mg2+ was kept low (0.0024–0.020 mm) and the substrate magnesium dl-isocitrate ranged from 0.07–0.25 mm. When kinetic constants were determined at pH 7.4 under these conditions and in the absence of ethylene glycol bis(β-aminoethyl ether) N,N′-tetraacetate, Ca2+ had little or no effect on Km (app) for ADP; the stimulation of rate by Ca2+ was mainly due to increased V (app). With subsaturating ADP, there was an interdependence in the interaction of the enzyme with substrate and Ca2+. Thus, with ADP constant (0.30 mm) the values of Km (app) for magnesium dl-isocitrate declined from 0.35 mm at zero Ca2+ to 0.19 mm with saturating Ca2+ without affecting V; Km (app) for free Ca2+ declined with increasing magnesium isocitrate to a limiting Km of 0.3 μm. (ii) Ethylene glycol bis(β-aminoethyl ether)-N,N′-tetraacetate, frequently used as a calcium buffer, inhibited enzyme activity with and without ADP. (iii) The enzyme was not inhibited by the calmodulin inhibitors trifluoperazine and chlorpromazine. Inhibition by lanthanide ions of the isocitrate dehydrogenase was competitive with magnesium isocitrate and not with respect to Ca2+. The values of Kis (1.8 to 3.1 μm) for La3+, Yb3+, Gd3+, Eu3+, Tb3+, and Er3+ were about two orders of magnitude smaller than Km for magnesium dl-isocitrate.  相似文献

Reaction of [Mo(O)Cl(CNMe)4]+ with the linear tetraphos ligand meso and rac prP4 leads to a mixture of [Mo(O)Cl(κ4-meso-prP4)]+ and [Mo(O)Cl(CNMe)(κ3-rac-prP4)]+ which are identified by X-ray structural analysis and/or 31P NMR spectroscopy. In the meso κ4-product both of the phenyl groups of the central phosphorus atoms are oriented towards the oxo ligand whereas in the rac κ3-product one of these phenyl groups is oriented to the oxo and the other to the chloro ligand. The origin of the different coordination modes lies in the different steric demands of the oxo and chloro ligands. The influences of the steric interactions are enhanced by the fact that exchange of the fourth isonitrile is difficult. This hypothesis is supported by the preparation of the complex [Mo(O)Cl(CNMe)(dpepp)]PF6 whose isonitrile ligand is inert towards exchange by monophosphines, even under drastic conditions.  相似文献

The polymorphism of apolipoprotein E (Apo E) in man is controlled by two codominant alleles, Apo En and Apo Ed, at the Apo E-N/D locus and by two alleles, the dominant, Apo E4+, and the recessive, Apo E4o, at the Apo E4 locus.

Frequency distribution analysis of Apo E phenotypes demonstrated a highly significant association between both systems (P ~ 1%). The Apo E4-(+) variant was about twice as frequent in phenotype Apo E-N (30.1%) than in phenotype Apo E-ND (16.4%). The phenotypic combination Apo E-D/-E4(+) was not observed. The segregation of Apo E phenotypes in informative matings is consistent with a close linkage of both loci.

The results may be explained by different models. On the basis of the present data, these models cannot be distinguished by formal genetic criteria. (1) Haplotypes Apo En/E4+, Apo En/E4o, and Apo Ed/E4o determine the different phenotypes, and a linkage disequilibrium exists of Δ = .0147 between the E-N/D and E4 loci. (2) The fourth haplotype, Apo Ed/E4+, exists, but the gene E4+ is not expressed in coupling with Apo Ed. The four-haplotype model seems more attractive in view of Apo E-N/D polymorphism's quantitative character and of biochemical results, which show that phenotypes Apo E-N and Apo E-D differ in the apparent molecular weight (Mr) of the respective major Apo E polymorphic form. Hence, the Apo E-N/D locus may control structural genes involved in the posttranslational modification of Apo E. (3) Finally, there may exist only one Apo E structural gene locus but with mutations at two sites susceptible to posttranslational modification.

Effects of Δ8- and Δ9-tetrahydrocannabinol (Δ8- and Δ9-THC) on three experimentally induced seizure models, i.e., audiogenic seizure (AS) test, maximal electroshock seizure (MES) test and pentylenetetrazol (PTZ)-induced seizure test were determined in the audiogenic rat. Both tetrahydrocannabinols possess a dose-related anticonvulsant effect against AS, MES and PTZ-induced maximal seizure. Although anticonvulsant potencies do not significantly differ, Δ8THC is three times more neurotoxic than Δ9THC. In addition, both THC's are without effect on minimal seizure and lethality induced by PTZ. Furthermore, the low protective indexes (TD50/ED50) determined in this study suggest that Δ8 and Δ9 THC may have poor therapeutic potentials as antiepileptic drugs.  相似文献

Protochlorophyllide(PChlide)-a and its 8-vinylated analog, divinyl(DV)-PChlide-a, are common and essential intermediates in the biosynthesis of all naturally occurring chlorophyll (Chl) pigments. These porphyrinoid-type pigments have a single optically active (asymmetric) carbon atom at the 132-position, so their stereoisomers are (132R)- and (132S)-enantiomers. The former and latter are called (DV-)PChlide-a and (DV-)PChlide-a′, respectively. In this study, chiral-phase HPLC separation of enantiomeric (DV-)PChlides-a/a′ was demonstrated. The (132R)-enantiomeric PChlide-a was eluted more slowly than the corresponding (132S)-enantiomeric PChlide-a′ under the present HPLC conditions. On the other hand, the elution order of (132R)-DV-PChlide-a and (132S)-DV-PChlide-a′ was reverse to that of PChlides-a/a′. After the separation of each enantiomer by the chiral-phase HPLC, the stereoisomeric configuration at the 132-position was characterized by means of circular dichroism spectroscopy. The present chiral-phase HPLC method enables us to evaluate optical purities of (DV-)PChlide-a species. The salt elimination reaction between FeCl2(THF)1.5 and the sterically hindered thallium tert-butyl-tris(3-isopropylpyrazolyl)borate, Tl[t-BuTpi-Pr], affords the new air and moisture-sensitive, high-spin, four-coordinate complex Fe[t-BuTpi-Pr]Cl (1) in 52% yield. Compound 1 has been characterized by elemental analysis, paramagnetic 1H NMR, and X-ray crystal structure determination. Fe[t-BuTpi-Pr]Cl is monomeric, and the homoscorpionate ligand is κ3-coordinated. The Fe(II) ion lies in a trigonally distorted tetrahedral environment with average N-Fe-N and N-Fe-Cl angles of 89.4(2)° and 125.7(2)°, respectively. Upon reactions with methyllithium followed by carbonylation, the six-coordinate acetyl-dicarbonyl derivative Fe[t-BuTpi-Pr](CO)2{C(O)Me} was identified by IR spectroscopy. The reaction of 1 with ethylmagnesiumbromide furnished a mixture of mononuclear Grignard complexes Mg[t-BuTpi-Pr]X (X = Et, Cl, Br), resulting formally from a facile and quantitative replacement of the [Fe-Cl]+ fragment by [Mg-X]+.  相似文献
