Key message

Using DArT analysis, we demonstrated that all Solanum × michoacanum (+) S. tuberosum somatic hybrids contained all parental chromosomes. However, from 13.9 to 29.6 % of the markers from both parents were lost in the hybrids.

Abstract

Somatic hybrids are an interesting material for research of nucleus-cytoplasm interaction and sources of new nuclear and cytoplasmic combinations. Analyses of genomes of somatic hybrids are essential for studies on genome compatibility between species, its evolution and are important for their efficient exploitation. Diversity array technology (DArT) permits analysis of the composition of nuclear DNA of somatic hybrids. The nuclear genome compositions of 97 Solanum × michoacanum (+) S. tuberosum [mch (+) tbr] somatic hybrids from five fusion combinations and 11 autofused 4x mch were analyzed for the first time based on DArT markers. Out of 5358 DArT markers generated in a single assay, greater than 2000 markers were polymorphic between parents, of which more than 1500 have a known chromosomal location on potato genetic or physical map. DArT markers were distributed along the entire length of 12 chromosomes. We noticed elimination of markers of wild and tbr fusion components. The nuclear genome of individual somatic hybrids was diversified. Mch is a source of resistance to Phytophthora infestans. From 97 mch (+) tbr somatic hybrids, two hybrids and all 11 autofused 4x mch were resistant to P. infestans. The analysis of the structure of particular hybrids’ chromosomes indicated the presence of markers from both parental genomes as well as missing markers spread along the full length of the chromosome. Markers specific to chloroplast DNA and mitochondrial DNA were used for analysis of changes within the organellar genomes of somatic hybrids. Random and non-random segregations of organellar DNA were noted.

Repetitive DNA sequences have been widely used in cytogenetic analyses. The use of gene sequences with a low-copy-number, however, is little explored especially in plants. To date, the karyotype details in Brachiaria spp. are limited to the location of rDNA sites. The challenge lies in developing new probes based on incomplete sequencing data for the genus or complete sequencing of related species, since there are no model species with a sequenced genome in Brachiaria spp. The present study aimed at the physical location of conserved genes in chromosomes of Brachiaria ruziziensis, Brachiaria brizantha, and Brachiaria decumbens using RNAseq data, as well as sequences of Setaria italica and Sorghum bicolor through the fluorescent in situ hybridization technique. Five out of approximately 90 selected sequences generated clusters in the chromosomes of the species of Brachiaria studied. We identified genes in synteny with 5S and 45S rDNA sites, which contributed to the identification of chromosome pairs carrying these genes. In some cases, the species of Brachiaria evaluated had syntenic segments conserved across the chromosomes. The use of genomic sequencing data is essential for the enhancement of cytogenetic analyses.  相似文献

The Botrychium lunaria (Ophioglossaceae) complex worldwide includes the named species B. lunaria, B. crenulatum, B. tunux, and B. yaaxudakeit. These species have been distinguished from each other morphologically and genetically. This study further investigates the genetic diversity and geographic distribution of this complex, examining a large number of plants worldwide. Enzyme electrophoresis was used to examine allelic variation of 22 loci for 1574 plants of putative B. lunaria, B. crenulatum and B. tunux from North America, Eurasia, and New Zealand, and B. dusenii from the Falkland Islands. Variation in allelic composition assessed by genetic identity and cluster analysis using the programs PopGene and STRUCTURE as well as morphology and geography indicated that the complex is composed of six distinct entities; two of which warrant recognition as new species, B. neolunaria, endemic to North America, and B. nordicum, sister to the B. lunaria complex, from Iceland and Norway; and a new combination, B. lunaria var. melzeri , endemic to Greenland, Iceland, and Norway. The new taxa are described in this paper. Three entities within B. tunux are discussed but not proposed for recognition at this time. Botrychium lanceolatum, included in this study, is composed of three morphologically and genetically distinct entities warranting taxonomic recognition.  相似文献

Discussions concerning the composition of the genus Parendacustes Chop., in particular, its subgenus Minizacla Gor., are continued. Eleven new taxa of this subgenus are described: P. trusmadi sp. n., P. mulu sp. n., P. brevispina sp. n., P. modispina sp. n., P. longispina sp. n., P. forficula sabah subsp. n., P. doloduo sp. n., P. buton sp. n., P. pallescens sp. n., P. kendari sp. n., and P. lindu sp. n. The taxonomic composition of the subgenus Caucasoplinthus Davidian, 1995 is revised. The immunis and granulosus groups are downgraded to subgroups of the silphoides and fallax groups, respectively; the voriseki subgroup is transferred from the fallax to the fausti group. Plinthus attilai Davidian et Savitsky, sp. n. and P. ljudae Davidian et Savitsky, sp. n. are described from Turkey and Abkhazia, respectively. New data on the taxonomy, distribution, and ecology of some little-known species are presented. The following new synonymy is established: Molytes vittatus Motschulsky, 1839 (= Meleus irroratus Reitter, 1884, syn. n., = Meleus faldermanni Faust, 1884, syn. n., = Meleus difficilis Faust, 1884, syn. n.).  相似文献

The distribution of the species of Nepalomyia Hollis, 1964 over the principal zoogeographical regions is briefly summarized. A new species, Nepalomyia chalajaesp. n. from the North Caucasus, is described. The replacement name Nepalomyia igorinom. n. is proposed for the homonym Nepalomyia yangi Grootaert, 2013, nec Wang, Yang and Grootaert, 2007. The negative association between the I - 3 gene and increased sensitivity to bacterial spot is due to linkage drag (not pleiotropy) and may be remedied by reducing the introgression size.

Abstract

Fusarium wilt is one of the most serious diseases of tomato (Solanum lycopersicum L.) throughout the world. There are three races of the pathogen (races 1, 2 and 3), and the deployment of three single, dominant resistance genes corresponding to each of these has been the primary means of controlling the disease. The I-3 gene was introgressed from S. pennellii and confers resistance to race 3. Although I-3 provides effective control, it is negatively associated with several horticultural traits, including increased sensitivity to bacterial spot disease (Xanthomonas spp.). To test the hypothesis that this association is due to linkage with unfavorable alleles rather than to pleiotropy, we used a map-based approach to develop a collection of recombinant inbred lines varying for portions of I-3 introgression. Progeny of recombinants were evaluated for bacterial spot severity in the field for three seasons, and disease severities were compared between I-3 introgression haplotypes for each recombinant. Results indicated that increased sensitivity to bacterial spot is not associated with the I-3 gene, but rather with an upstream region of the introgression. A survey of public and private inbred lines and hybrids indicates that the majority of modern I-3 germplasm contains a similarly sized introgression for which the negative association with bacterial spot likely persists. In light of this, it is expected that the development and utilization of a reduced I-3 introgression will significantly improve breeding efforts for resistance to Fusarium wilt race 3.

Six new species are described from Vietnam, Apia simplexsp. n., Bisma angulatasp. n., Pitambara trypetoidessp. n., Sarmatoca cathemerinagen. et sp. n., Maracota soulieraegen. et sp. n., and Serida castaneasp. n., and Zeleja thoracalissp. n. from Malaysia. Two new genera, Maracotagen. n. (type species M. soulierae sp. n.) and Sarmatocagen. n. (type species S. cathemerina sp. n.), and a new subgenus Zelomachasubgen. n. (type species Zeleja thoracalis sp. n.) in the genus Zeleja are erected. The genus Binaluana Soulier-Perkins et Stroiński is downgraded to a subgenus of Zeleja Melichar. The genus Silvispina Wang et Soulier-Perkins with an uncertain systematic position is attributed to the subfamily Lophopinae and the tribe Lophopini. The placement of the genus Elasmoscelis Spinola (and, correspondingly, the tribe Elasmoscelini) in the subfamily Menoscinae, the genus Epiptyxis Gerstaecker in the tribe Menoscini, and the genus Jivatma Melichar in the tribe Lophopini is substantiated. New records of 11 species are given from Vietnam, some being first records for this country.  相似文献

A key to three species of Megahypurus from Thailand is given. New data on the distribution and taxonomy of Agonopterix species in Russia are given. The following new synonymy is established: Agonopterix anticella (Erschoff, 1877) = A. abjectella (Christoph, 1882), syn. n., = A. japonica Saito, 1980, syn. n.; A. pullella Hannemann, 1971 = A. dubatolovi Lvovsky, 1995, syn. n. A new species A. volgensissp. n. is described from the Middle Volga Region. Review of the six genera of the Silvestrichilis group (Dilta Strand, Haslundiella Janetschek, Silvestrichilis Wygodzinsky, Silvestrichiloides Mendes, Haslundichilis Wygodzinsky, and Himalayachilis Wygodzinsky) and of the 17 species of the southern Palaearctic genus Silvestrichilis was performed for the first time with a discussion of their phylogenetic relationships and evolutionary trends based on comparative analysis of the geographic distribution of the species and of the distribution of plesiomorphic and apomorphic states of their main morphological features. Haslundichilis quadrii Wygodzinsky, 1952 from northwestern India and H. lindbergi Wygodzinsky, 1962 from western Afghanistan are transferred to the genus Haslundiella, and new combinations Haslundiella quadrii (Wygodzinsky, 1952), comb. n. and Haslundiella lindbergi (Wygodzinsky, 1962), comb. n. are established. A new species, Silvestrichilis polinaesp. n., differs from all the congeners in the strongly dilated fore femur of the male and female, in the maxillary palpus chaetotaxy, and in the structure of the sensory field on the fore femur of the male.  相似文献
